Com a evolução e o passar dos séculos, foi se aprimorando lentamente, até chegar aos dias de hoje em que a atividade de panificação dispõe de equipamentos e técnicas para seu fabrico, de uma vasta quantidade e crescente variedade de pães. Com o crescimento da população e o aumento na demanda por alimentos prontos para o consumo - incluindo-se nestes os pães - os governos foram forçados a editar leis a fim de adequar sua fabricação a normas regulamentares, visando assegurar a melhor qualidade do produto e forçar a observância de requisitos mínimos de higiene e sanitização. No Brasil, dentre essas normas, sobressai a Resolução n. 216 da Agência Nacional de Vigilância Sanitária, de setembro de 2004, com o objetivo de padronizar os trabalhos de limpeza e higiene de máquinas e equipamentos, higiene pessoal, limpeza de reservatórios e o controle de pragas urbanas. A este projeto foi dado o nome de Boas Práticas de Fabricação (BPF). Esta Resolução padroniza os trabalhos e exige dos estabelecimentos produtores a elaboração de manuais de produção e os procedimentos operacionais padronizados, concernentes aos itens acima referidos. A presente pesquisa tem por objetivo descrever os trabalhos de implantação das BPFs em 25 panificadoras da região metropolitana do Recife. No escopo desta dissertação relaciona-se uma vasta revisão bibliográfica sobre o assunto e a descrição, pari passu, dos trabalhos de campo realizados entre os meses de agosto de 2005 e agosto de 2006. De posse dos dados coletados no trabalho de campo, é possível demonstrar como a implantação das Boas Práticas de Fabricação e o treinamento dos manipuladores de alimentos podem auxiliar a obter melhores resultados para os próprios estabelecimentos e a benefício da população. Resultados estes que transcendem o objetivo da qualidade dos produtos e contribuem para o aumento da produtividade

The second question was to assess the value in using the intervention against defaulting municipalities, by looking at whether the intervention was effective and what impact did it have on the defaulting municipalities.

Fire Tests Of Cut And Cover Tunnel Roof Segments At Positive Moment Region

The most important issue during a tunnel fire is safety of human life. The tunnel fire structural research and investigations have gained more importance in the last decade but studies show variable results depending on the concrete quality and tunnel design fire. For instance, a certain type of concrete with high moisture content can tend to explode in the first 10-15 minutes of fire with rapid increase of heat release rate. A sudden collapse of the tunnel roof during the fire is unacceptable. Especially in Netherlands, the possible sagging of cut and cover tunnel roof is undesired and prevention systems are applied. The main purpose of this research is to investigate fire response of the positive moment region of cut and cover tunnel roof through an experimental and analytical program without use of any protection. In this context a standard one cell rail road cut and cover tunnel has been designed for loads of backfill, lateral earth pressure and self weight. The typical concrete cover used in Turkish railroad tunnels is 6 centimeters. Four pairs of representative sample tunnel roof segments have been manufactured and only one segment out of each pair are tested under 2 hours extreme design tunnel fire in a furnace. Out of these four types, two types have been internally pre-stressed to simulate the internal loads at the positive moment region of the tunnel roof. Four pairs of sample segments are simply supported during the static load test and static load is applied at the mid-span to measure the difference in the post-fire structural performance. Compressive strength of concrete, tensile strength of reinforcing bars, electron microscope evaluation of concrete, moisture content of concrete are recorded during the test program. A finite element based solution is developed to simulate the results of static load tests. Post-fire structural performances of burnt segments are observed to be not much different than the unburnt segments.

Cette recherche se propose de mettre en exergue la place du péché dans les discours déclinés dans le duché de Bretagne du XIIe au début du XVIe siècle. Elle se fonde aussi bien sur les sources écrites (productions pastorales,textes normatifs de l’Église et l’État breton, oeuvres littéraires et moralisantes de la cour ducale) que sur l’iconographie, à travers les sculptures de l’époque romane et de la fin du Moyen Âge.Le péché y apparaît comme une clef de compréhension du monde et de son évolution : présenté comme une causalité historique majeure depuis le début de l’humanité, son instrumentalisation permet de représenter lesvices de l’altérité, liée à l’ethnie ou à la religion, d’appréhender les identités collectives de tout ordre (professionnelles, de classe, d’ordre, de genre), en même temps qu’il rappelle la place de l’individu dans la société. L’association constante des dominés avec les péchés du corps, ainsi que la mise en place d’une véritable géopolitique des princes « pécheurs » ennemis de la Bretagne (cupides, paresseux, homosexuels…), ne sont que deux exemples d’une utilisation socio-Politique des vices.Instrument de culpabilisation issu de la morale chrétienne, le recours massif au péché dans les discours dominants en Bretagne est d’abord lié à la réforme « grégorienne » du XIIe siècle, avant que les lettrés de la courducale des Montfort ne s’approprient ces discours pour légitimer le pouvoir de leur prince. Dans tous les cas, le péché s’impose comme un vecteur essentiel dans l’effort de normalisation morale de la société et du pouvoir au Moyen Âge / This research aims to highlight the place of sin in discourses in the Duchy of Brittany from the 12th to the early 16th century. The study is based on both written and iconographical sources (pastoral care production, normative documents of the Church and the State Breton, literary and moralistic works of the ducal court, sculptures from Romanesque period and late Middle Ages).Sin appears to be a key to understanding the world and its evolution : it is a major historical causality since the beginning of mankind ; it serves the representation of the ethnic or religious otherness ; it is an useful device to apprehend collective identities of any kind (professional, class, order, or gender identities) ; finally, it establishes the place of the individual in society. The recurring association of dominated classes with the sins of the flesh and the appearence of a ‘political geography’ of princes ‘sinners’ enemies of Britain (greedy, lazy, homosexual...) are two examples of a such socio-Political use of vices.As an instrument of guilt trip from Christian morality, the massive use of sin in the dominant discourses in Britain come up from the "Gregorian" reform of the twelfth century. Later, the scholars of the ducal court of Montfort begin to use these discourses to legitimize the power of their prince. In both cases, sin becomes a powerful agent in theeffort to moral normalization of society and power in the Middle Ages

SISTEMATIZAÇÃO DE FERRAMENTA DE APOIO PARA BOAS PRÁTICAS EM SERVIÇOS DE ALIMENTAÇÃO / SYSTEMIZATION OF A TOOL TO SUPPORT GOOD PRACTICES IN FOOD SERVICES

Concerning the importance and the necessity of fitting food services to actual lawmaking and knowing the difficulty found in it, the present paper aimed to systemize a Tool (Evaluation List, Plan of Action and Touching Collaborators) to support Good Practices. The present study was realized in twenty three, 23, downtown food services, in Santa Maria, Rio Grande do Sul, through a proportional stratified sample of eight restaurants, eleven snack bars and four bakeries. A List of Evaluation of Good Practices for Food Services (LEGPFS) was made, specific for RDC 21604 ANVISA. This list was applied for a professional technician and for the company, in March and October, respectively. After that, a simple Plan of Action to facilitate the process of fitting was suggested. Samples of swabs to verify Staphylococcus coagulase positive and total count of mesophyll aerobics microorganisms and were collected from manipulators and from surfaces after hygiene, before and after the use of the tool. All collaborators were invited to participate of a touching concerning microbiological analysis and formulary applied to participants, evaluated by the results of LEGPFS. The results showed a significant difference (p<0,05) after the application of LEGPFS such for the technician as to the company, according to the fitting percentage. It was observed a significant improve (p<0,05) in following lawmaking after the use of the tool. The touching was considered efficient and there was a fitting to the process of hands and surfaces hygiene. In conclusion, the tool elaborated was valid to support food services in fitting actual lawmaking. / Tendo em vista a importância e a necessidade de adequação dos serviços de alimentação à legislação vigente e sabendo-se da dificuldade encontrada, objetivou-se, com este trabalho, sistematizar uma Ferramenta (Lista de Avaliação, Plano de Ação e Sensibilização de colaboradores) de apoio às Boas Práticas. O presente estudo foi realizado em 23 serviços de alimentação do Bairro Centro, da cidade de Santa Maria-RS, a partir de uma amostra estratificada proporcional de 8 restaurantes, 11 lanchonetes e 4 padarias/ confeitarias. Elaborou-se uma Lista de Avaliação de Boas Práticas para Serviços de Alimentação (LABPSA), específica para RDC 216/04 - ANVISA. Ela foi aplicada por profissional técnico e pela empresa, em março e outubro de 2006, respectivamente. Após a aplicação da LABPSA, sugeriu-se um modelo simples de Plano de Ação para facilitar o processo de adequação. Realizaram-se coletas de swabs nas mãos dos manipuladores e nas superfícies, após a higienização, para verificação de Staphylococcus coagulase positiva e contagem total de microrganismos aeróbios mesófilos, antes e após o uso da Ferramenta. Todos os colaboradores foram convidados a participar de uma sensibilização, avaliada pelos resultados da LABPSA, das análises microbiológicas e de formulário aplicado aos participantes. Constatou-se diferença significativa (p< 0,05) após a aplicação da LABPSA tanto técnica quanto pela empresa, de acordo com o percentual de adequação. Houve também uma melhora significativa (p< 0,05) no atendimento à legislação após a utilização da ferramenta. A sensibilização foi considerada eficaz e houve adequação no processo de higienização das mãos e superfícies. Em suma, a ferramenta elaborada foi válida para apoiar os serviços de alimentação nas adequações exigidas pela legislação atual.

O objetivo é o de identificar os atores envolvidos, as discussões, diretas e transversais, que se apropriaram do tema durante o processo Constituinte mais recente da história do país. É também o de entender os meandros que levaram à redação final do que hoje conhecemos como artigo 216, o mais importante da Constituição da República no que tange à conceituação do patrimônio e a designação das formas de preservação e proteção. Para isso, foram utilizadas revisões bibliográficas, pesquisa em fontes como atas de audiência e reuniões no período Constituinte, e a consulta a jornais para estruturar a tese ora apresentada. Será visto que alguns conflitos tangenciaram a discussão, mas quando o tema foi diretamente debatido, a tendência foi a de conciliação como resultado das tratativas entre as partes envolvidas. A ANC, como ambiência política, será perscrutada, no que se refere ao patrimônio, para revelar quais mediadores, agentes e reivindicações destacaram-se e como suas ações refletiram num artigo abrangente, com um conceito amplo de patrimônio cultural. A demanda por reconhecimento de culturas, identidades e memórias será considerada, todavia a sua perspectiva econômica acerca da propriedade da terra, por parte de grupos indígenas e de negros, marginalizados nas políticas públicas de até então, foi encarada como assunto de menor relevo pela ANC, em uma nítida contenção de conflitos, por um lado, e atendimento a anseios de forças políticas incisivas, de outro. / This thesis addresses the issue of cultural patrimony during the Brazilian National Constituent Assembly of 1987 and 1988, with regard to its insertion in the body of the text of the 1988 Constitution. The aim is to identify the actors involved and the direct and transversal discussions that focused on this theme during the most recent constitutional process in the history of the country. It also aims to understand the intricacies that have led to the final text of what we know today as article 216, the most important one of the Constitution of the Republic concerning the conceptualization of patrimony and the designation of means for its preservation and protection. Therefore, the present work has been built upon bibliography review, consultation to newspapers and searches through minutes of hearings and meetings held during the constitutional period. It is possible to observe that some conflicts have appeared during the discussions, but once the issue was directly debated, there was a tendency to conciliation, as a result from negotiations among the actors involved. This is a comprehensive article, with a broad concept of cultural patrimony, which investigated the National Constituent Assembly (ANC, for its acronym in Portuguese) as a political ambience, on the issue of patrimony, in order to reveal the most remarkable mediators, actors and requests, as well as the reflections of their actions. The claims for the recognition of cultures, identities and memories have been considered by the Constituent Assembly. Nevertheless, the economic perspective towards land property by groups of indigenous and black people, marginalized by public policies until then, has been treated as a less relevant issue, on one side as a clear avoidance of conflicts, on the other as an answer to incisive political forces.

A Novel Route for Construction of Multipurpose Receptors through Chemical Modification of Glutathione Transferases

This thesis describes how the human Alpha class glutathione transferase (GST) A1-1 can be reprogrammed either to function as a multipurpose biosensor for detection of small molecule analytes, or as a handle providing for more efficient protein purification. A novel, user-friendly, and efficient method for site-specific introduction of functional groups into the active site of hGST A1-1 is the platform for these achievements. The designed thioester reagents are glutathione-based and they are able to label one single nucleophile (Y9) and leave the other 50 nucleophiles (in hGST A1-1) intact. The modification reaction was tested with five classes of GSTs (Alpha, Mu, Pi, Theta and Omega) and was found to be specific for the Alpha class isoenzymes. The reaction was further refined to target a single lysine residue, K216 in the hGST A1-1 mutant A216K, providing a stable amide bond between the protein and the labeling group. To further improve the labeling process, biotinylated reagents that could deliver the acyl group to Y9 (wt hGST A1-1) or K216 in the lysine mutant, while attached to streptavidin-coated agarose beads, were designed and synthesized. A focused library of eleven A216K/M208X mutants was made via random mutagenesis to provide an array of proteins with altered micro-environments in the hydrophobic binding site, where M208 is situated. Through the invented route for site-specific labeling, a fluorescent probe (coumarin) was introduced on K216 in all double mutants, with the purpose of developing a protein-based biosensor, akin to the olfactory system. The array of coumarin-labeled proteins responded differently to the addition of different analytes, and the responses were analyzed through pattern recognition of the fluorescence signals. The labeled proteins could also be site-specifically immobilized on a PEG-based biosensor chip via the single C112 on the surface of the protein, enabling development of surface-based biosensing systems. Also, a refined system for efficient detection and purification of GST-fusion proteins is presented. Through a screening process involving A216K and all produced A216K/M208X mutants, two candidates (A216K and A216K/M208F) were singled out as scaffolds for the next generation of fusion proteins. In addition to the features present in commercially available GST fusion constructs, the new mutants can be site-specifically labeled with a fluorophore in bacterial lysates providing quick and sensitive monitoring of expression and purification. Furthermore, the proteins could be labeled with a unique aldehyde moiety providing for a novel protein purification scheme.

Uma primeira aproximação é a expansão em harmônicos esféricos, onde os termos C20 e o C22 caracterizam a irregularidade do corpo. Usamos essa aproximação em superfícies de secção de Poincaré para estudar as regiões próximas ao planeta anão Haumea, onde foi observado um anel. A partir do mapeamento feito pela técnica de superfície de secção de Poincaré, foi possível identifi- car Famílias de órbitas periódicas e regiões estáveis. Duas Famílias de órbitas periódicas foram destacadas, a primeira uma Família de segundo tipo associada à ressonância 1:3 (Família ressonante) e a segunda uma Família de primeiro tipo (Família central). As simulações indicam que as partículas do anel podem estar em órbitas da Família central. Já a Família ressonante, não pode ser responsável pelo anel devido a excentricidade de suas órbitas e da sua posição. Para simular de forma mais realista a irregularidade de um pequeno corpo, é usada uma melhor aproximação para o cálculo do potencial gravitacional. O modelo de concentração de massa, ou modelo de mascons, é uma aproximação discreta da forma de um corpo, capaz de simular um potencial irregular, assimétrico e tridimensional. A esse modelo é aplicada a superfície de secção de Poincaré, com o objetivo de estudar a dinâmica da região próxima ao asteroide 4179 Toutatis. Quatro Famílias de órbitas periódicas são destacadas e estudadas. Uma Família é de primeiro tipo e as outras três são de segundo tipo associadas às ressonâncias 3:1, 2:1 e 2:3. Apesar do potencial gravitacional tridimensional ser adotado em uma ferramenta usualmente bidimensional, é possível analisar como um problema bidimensional quando a variação na terceira dimensão é baixa. Estudando em conjunto as superfícies de secção de Poincaré e a variação máxima na terceira dimensão, verifica-se a estabilidade ou não das trajetórias simuladas / In general, small bodies of the Solar system, e.g. asteroids and comets, have a very irregular shape. This feature affects significantly the gravitational potential around these irregular bodies, which hinders dynamical studies. A first approximation is an expansion in spherical harmonics, where C20 and C22 characterize the irregularity of the body. This approach is used on Poincaré surfaces of sections to study regions close to the dwarf planet Haumea. This regions are where the observed ring. By the technique of Poincaré surface of section, it was identified Families of periodic orbits and stable regions. Two Families of periodic orbits were studied, the first Family is a second type associated with the 1:3 resonance (resonant Family) and the second Family is a first type (central Family). During the simulations the ring particles can be in orbits of the central Family. But the resonant Family can not be responsible for the ring due the eccentricity and position of their orbits. In order to more realistically simulation of the irregularity of the body, a better approximation is necessary for the computation of the gravitational potential. The mass concentration model, or mascon model, is a discrete approximation of the shape of a body. This model simulates an irregular, asymmetric and three-dimensional potential. This model was applied in a Poincaré surfaces of section, mainly to study the dynamics of the region close to the asteroid 4179 Toutatis. Four Families of periodic orbits were studied. One of then is a first type and the others were the second type and associated with the resonances 3:1, 2:1 and 2:3. Although the three-dimensional gravitational potential is adopted in a usually two-dimensional tool, it is possible to analyze as a two-dimensional problem when the variation in the third dimension is low. By a analyzing of the Poincaré surfaces of section and a maximum variation in the free dimension together, the stability of the simulated trajectories is measured
