Modulation of GLO1 expression affects malignant properties of cells

The energy metabolism of most tumor cells relies on aerobic glycolysis (Warburg effect) characterized by an increased glycolytic flux that is accompanied by the increased formation of the cytotoxic metabolite methylglyoxal (MGO). Consequently, the rate of detoxification of this reactive glycolytic byproduct needs to be increased in order to prevent deleterious effects to the cells. This is brought about by an increased expression of glyoxalase 1 (GLO1) that is the rate-limiting enzyme of the MGO-detoxifying glyoxalase system. Here, we overexpressed GLO1 in HEK 293 cells and silenced it in MCF-7 cells using shRNA. Tumor-related properties of wild type and transformed cells were compared and key glycolytic enzyme activities assessed. Furthermore, the cells were subjected to hypoxic conditions to analyze the impact on cell proliferation and enzyme activities. Our results demonstrate that knockdown of GLO1 in the cancer cells significantly reduced tumor-associated properties such as migration and proliferation, whereas no functional alterations where found by overexpression of GLO1 in HEK 293 cells. In contrast, hypoxia caused inhibition of cell growth of all cells except of those overexpressing GLO1. Altogether, we conclude that GLO1 on one hand is crucial to maintaining tumor characteristics of malignant cells, and, on the other hand, supports malignant transformation of cells in a hypoxic environment when overexpressed.

Outcomes of stable and unstable patterns of subjective cognitive decline

Background: Subjective cognitive decline (SCD), i.e., the self-perceived feeling of worsening cognitive function, may be the first notable syndrome of preclinical Alzheimer’s disease and other dementias. However, not all individuals with SCD progress. Stability of SCD, i.e., repeated reports of SCD, could contribute to identify individuals at risk, as stable SCD may more likely reflect the continuous neurodegenerative process of Alzheimer’s and other dementias. Methods: Cox regression analyses were used to assess the association between stability of SCD and progression to MCI and dementia in data derived from the population-based Leipzig Longitudinal Study of the Aged (LEILA75+). Results: Of 453 cognitively unimpaired individuals with a mean age of 80.5 years (SD = 4.2), 139 (30.7 %) reported SCD at baseline. Over the study period (M = 4.8 years, SD = 2.2), 84 (18.5 %) individuals had stable SCD, 195 (43.1 %) unstable SCD and 174 (38.4 %) never reported SCD. Stable SCD was associated with increased risk of progression to MCI and dementia (unadjusted HR = 1.8, 95 % CI = 1.2–2.6; p < .01), whereas unstable SCD yielded a decreased progression risk (unadjusted HR = 0.5, 95 % CI = 0.4–0.7; p < .001) compared to no SCD. When adjusted for baseline cognitive functioning, progression risk in individuals with stable SCD was significantly increased in comparison to individuals with unstable SCD, but not compared to individuals without SCD. Conclusions: Our results, though preliminary, suggest that stable SCD, i.e., repeated reports of SCD, may yield an increased risk of progression to MCI and dementia compared to unstable SCD. Baseline cognitive scores, though within a normal range, seem to be a driver of progression in stable SCD. Future research is warranted to investigate whether stability could hold as a SCD research feature.

Evolution Of The Cicekdagi Basin, Central Anatolia, Turkey

&Ccedil / i&ccedil / ekdagi basin developed on the Central Anatolian Crystalline Complex (CACC) is a foreland basin developed as the southern integral part of the &Ccedil / ankiri Basin during the Late Paleocene to middle Oligocene. The basin has two compartments separated by the &Ccedil / i&ccedil / ekdagi High comprises two sedimentary cycles. The oldest cycle comprises Barakli, Koca&ccedil / ay and Bogazk&ouml / y formationsa and is exposed both in the northern and the southern sectors. They were deposited in marine conditions. The second cycle comprises incik and G&uuml / vendik formations and was deposited in continental settings. The first cycle comprises uniformly south-directed paleocurrent directions in both the northern and southern sectors whereas the second cycle deposits are represented by south-directed directions in the southern sector, and bimodal directions in the northern sector. In addition, the second cycle formations contain progressive unconformities and coarsening upwards sequences indicative of thrusting. Internal structures of the units and paleostress data indicate that the basin experienced over-all compression and local extension due to flexural bending. This gave way to inversion of some of the normal faults and uplift of the &Ccedil / i&ccedil / ekdagi High during the deposition of second cycle in the Late Eocene to middle Oligocene time which subsequently resulted in compartmentalization of the basin.

Neotectonics And Seismicity Of The Ankara Region: A Case Study In The Urus Area

Study area, the UruS province, is located 70 km WNW of city of Ankara. Major settlements in the study area are two counties, UruS and G&uuml / d&uuml / l / and there are a number of villages, such as, from W to E, Macun, Yogunpelit, Kabaca, &Uuml / regil, &Ouml / zk&ouml / y, Tahtaci&ouml / rencik, Kirkkavak, Kavak&ouml / z&uuml / , Kayi and Karaca&ouml / ren. The study area is 189 km2 in size and included in 1/25000-scaled topographic quadrangles of H28a3, a4, d1 and d2. The G&uuml / d&uuml / l- UruS section of the &Ccedil / eltik&ccedil / i morphotectonic depression (&Ccedil / eltik&ccedil / i Basin) drained by the Antecedent Kirmir River and its second-order drainage system was first mapped in detail in the present study, and faults determining northern margin of the &Ccedil / eltik&ccedil / i depression were named as the UruS fault set comprising the SW part of the &Ccedil / eltik&ccedil / i Fault Zone / and the mechanism of the master fault of the UruS fault set was determined as left lateral oblique-slip fault with reverse component by the morphologic markers such as the deformed drainage system and pressure ridges. This was also supported by the fault plane solutions of the 2000.08.22 UruS earthquake. Three fault plane solutions, of which two of them for the 2000.08.22 UruS earthquake, and one of them for the 2003.02.27 &Ccedil / amlidere earthquake, were done to determine nature of the source. Ground material underlying the city of Ankara were divided into three categories: (a) well-lithified basement rocks, (b) Pliocene fluvio-lacustrine v sedimentary sequence, and (c) unconsolidated terrace and alluvial sediments of Quaternary age. Quaternary unconsolidated sediments are densely populated in Ankara. These sediments are fine-grained and have a maximum thickness of 200 m or more. Inside these sediments, static ground water level is very close (as on average: 6 m) to ground surface. These conditions are quite suitable for liquefaction of these unconsolidated alluvial sediments. In addition, basement rocks are full of zone of weakness. Even if, the city of Ankara is characterized by the shallow focus and small earthquakes (M&amp / #8804 / 5), it is open to the risk of large earthquakes to be sourced from the North Anatolian Fault System and the Seyfe Fault Zone located 110 km and 80 km, respectively, owing the ground material conditions beneath the city of Ankara. This point has to be taken out in constructions and site selection solution.

SOCIAL STRATIFICATION AND CONSUMPTION PROFILES OF ANKARA: A CASE STUDY IN ANKARA RESIDENTIAL AREAS This dissertation attempts to develop a social stratification model considering its spatial dimension for the households in the city of Ankara. The spatiality of social class has rarely been in the agenda of scholars and has not been explored empirically. For this reason, the study aims to test the hypothesis that social segregation in Turkish cities is empirically measurable and manifests itself in the common patterns of behaviours and similar conditioning of existence in the urban space. The focus of attention of the thesis is based on the relationship between the material inequalities of different social strata within its territorial context. If a class becomes a social reality, this must be shown in the formation of common patterns of behaviour and attitude, and manifests itself in urban space. In short the scope of the study is a twofold: Thesis questions are: (1) how and upon what basis social groups and strata can be located in the economic and socio-cultural structure of the society. This part of the study deals with the objective &amp / #8216 / &amp / #8217 / set&amp / #8217 / &amp / #8217 / of criteria / thesis question (2) whether the same coherency can be coincided in the space. This part deals with the analysis of the spatial dimension of social &amp / #8216 / &amp / #8217 / class&amp / #8217 / &amp / #8217 / which means segregation. Thesis findings provide sufficient evidence that the differences stemmed from the material possessions and consumption patterns of the urban households cannot be understood by employing the conventional instruments as such rural/urban, traditional/modern as the division of axes. New conceptualisation urgently is needed and consumption studies offer prospective and highly potential issue.

Remote Sensing Study Of Surgu Fault Zone

The geometry, deformation mechanism and kinematics of the S&uuml / rg&uuml / Fault Zone is investigated by using remotely sensed data including Landsat TM and ASTER imagery combined with SRTM, and stereo-aerial photographs. They are used to extract information related to regional lineaments and tectono-morphological characteristics of the SFZ. Various image processing and enhancement techniques including contrast enhancement, PCA, DS and color composites are applied on the imagery and three different approaches including manual, semi automatic and automatic lineament extraction methods are followed. Then the lineaments obtained from ASTER and Landsat imagery using manual and automatic methods are overlaid to produce a final lineaments map. The results have indicated that, the total number and length of the lineaments obtained from automatic is more than other methods while the percentages of overlapping lineaments for the manual method is more than the automatic method which indicate that the lineaments from automatic method does not discriminate man made features which result more lineaments and less overlapping ratio with respect to final map. It is revealed from the detail analysis that, the SFZ displays characteristic deformation patterns of strike-slip faults, such as pressure ridges, linear fault controlled valleys, deflected stream courses, rotated blocks and juxtaposition of stratigraphical horizons in macroscopic scale. In addition to these, kinematic analyses carried out using fault slip data indicated that the S&uuml / rg&uuml / Fault Zone is dextral strike-slip fault zone with a reverse component of slip and cumulative displacement along the fault is more than 2 km.

La médecine intégrative est un domaine relativement récent qui offre une nouvelle approche holistique pour la médecine. Elle cherche à satisfaire les besoins du public en unifiant la médecine contemporaine basée sur la biomédecine et d'autre types de pratiques médicales, les médecines complémentaires et alternatives (MCA), qui ne sont pas encore scientifique­ment suffisamment vérifiées. Ce que nous appelons la médecine intégrative ici est apparue au début des années 1990. Son potentiel est prometteur, mais il y a cependant des besoins cruciaux de faire progresser les connaissances dans ce domaine et de reconnaître qu'il existe des problèmes méthodologiques qui ne sont pas pris en compte et qui constituent une entrave à son développement. Ce présent projet a pour objectif de fournir une base philosophique plus solide pour la médecine intégrative et, plus concrètement, d'examiner comment intégrer la vertébrothérapie japonaise dans la médecine. L'auteur pratique depuis longtemps la vertébrothérapie japonaise comme métier familial, dont le fondateur était son grand-père maternel. L'auteur se demande pourquoi cette thérapie ne se développe pas davantage et reste mal connue de la société en dépit de son efficacité. Comment peut-on vérifier sa rigueur, en quoi consiste cette thérapie, d'où vient-elle et qui l'a forgée ? Pour répondre à ces questions, l'auteur a commencé à faire des recherches en empruntant la méthodologie de la tradition de l'épistémologie française. Ainsi, pour cristalliser l'histoire scientifique de la vertébrothérapie, il recourt à la méthodolo­gie de Canguilhem et de Cavaillès ; pour former une épistémologie de cette thérapie, il fait appel à la méthodologie de Canguilhem, et pour la manière d'argumenter, à celle de Fagot-Largeault. / Integrative medicine is a relatively new field that offers a new, holistic approach to medicine to satisfy the needs of the public by unifying biomedical medicine and other medical practices; complementary and alternative medicine (CAM), which are not yet scientifically verified. What we call integrative medicine emerged in the early 1990s, and its potential i promising; however, there is crucial need to recognize that there exist methodological problems in this field. In this thesis I want to propose some reflections upon the consequences linked to the disregard for the theory of CAM. I would argue that the special character of today's dominant evaluation methodology -randomized controlled trial (RCT) and the ways in which excessive reliance on RCT distorted the essence of CAM and amplified the predicament. I equally want to propose some reflections upon the effect of the introduction of particular dogmas to CAM into scientific study. I would suggest that the ways in which researches might loose in tum scientific certainty, clarity, and objectivity if they make much of for instance the concept of qi or human sensory subjective diagnostics system in scientific study. Finally, would like to note advantages in introducing Japanese traditional medicine (JTM) into scientific study and the ways in which JTM has reduced influence of, and been freer from, the excessive theorization in Chinese traditional medicine (CTM).
