Evaluation of Different Techniques for Repair of Shear-span Corrosion-Damaged RC Beams

Deterioration of reinforced concrete structures due to reinforcement corrosion is a serious problem that faces concrete infrastructure worldwide. Effect of the rebar corrosion in the shear span on the structural behaviour is not fully addressed in the published literature. This study examined the effects of corrosion of the longitudinal reinforcement in the shear span on the structural behaviour of RC beams and the effectiveness of three rehabilitation schemes on the structural performance of such beams. The experimental program consisted of testing fifteen medium-scale reinforced concrete beams (150mm wide x 350mm deep x 2400mm long) under static load. Test variables included: span to depth ratio, the degree of corrosion and the anchorage end condition and repair schemes. Two span to depth (a/d) ratios were considered: a/d=3.4 with one-point loading and a/d=2.4 with two-point loading. Two anchorage end-conditions were used: bonded or un-bonded reinforcement in the an-chorage zone. Four degrees of corrosion were chosen to simulate minor (2.5% to 5% mass loss), medium (7.5% mass loss), and severe (15% mass loss) degrees of corrosion. Corrosion was induced in the longitudinal reinforcement in the shear-span using accelerated corrosion techniques based on Faradays’ law. Three different repair scenarios were applied. The first scenario included removing the deteriorated concrete, cleaning the corroded steel and patching with a new self-compacting concrete. The second scenario included U-wrapping the beams cross-section using Glass fiber reinforced cement-based composite (GFRCM), and Carbon fiber reinforced cement-based composite (CFRCM) without removing the deteriorated concrete. The third scenario included patch repair and confinement by wrapping with GFRCM or CFRCM. Following corrosion and repair, all specimens were loaded statically to failure. Test results showed no major effect of shear-span corrosion on the flexural behaviour for the beams with end anchorage whereas a noticeable effect on the flexural behaviour was observed for beams with no end anchorage regions. The corrosion degree and the shear span to depth ratio affected the mode of failure for the specimens with no end anchorages. The type of repair significantly affected the overall behaviour of the corroded specimens. An analytical model was proposed and used to predict the load-deflection response of the tested specimens. The program calculated the mid-span deflection for a given load as an integration of the deflection of a series of elements, with the deflection being based on the elongation of the steel reinforcement in each element. A modified bond stress-slip model was incorporated into the calculations to account for the change in bond strength caused by the corrosion and/or confinement that are provided by repairs. The predicted results were in reasonable agreement with the experimental results.

Habitat structure, degradation and management effects on coral reef fish communities

Coral reefs are among the most diverse and productive ecosystems on earth, and are critical to the survival of tropical marine ecosystems and sustenance of local human populations. However, coral reefs are quite vulnerable to disturbances, both natural and anthropogenic. This thesis looks at how coral reef communities have responded to climactic disturbances, particularly the 1997-98 El Niño Southern Oscillation (ENSO) and subsequent coral bleaching and mortality that affected much of the Indian Ocean, including the coastal waters of Tanzania, where the study was conducted. In particular, it investigates the effects of coral bleaching, habitat degradation and reef spatial arrangement on reef fish assemblages. Habitat structural complexity and spatial arrangement of reefs had an effect on reef fish communities. Fish communities showed patterns in distribution among habitats and between patch and continuous reefs. Fishes preferred live to bleached/dead or eroded coral, but trophic groups reacted differently to patch and continuous reefs. There were slight changes in fish abundance and significant changes in fish diversity on experimental, bleached branching Acropora coral plots over a period of one year. While fish abundance on one site increased shortly after a bleaching event, 6 years later fish abundance had decreased significantly. Conversely, coral reef communities in northern Tanzania had changed little over an 8-year period, with minor changes associated with the 1997-98 ENSO and the presence or absence of fisheries management. The coral reefs in the region were found to show high variability in community structure and responses of associated fish and invertebrate communities. The findings of this thesis indicate the importance of habitat structure and spatial arrangement of reefs, the detrimental effects of coral bleaching, and the possibility that some reefs and some (generalist) reef fish taxa may exhibit resilience to climate change. / At the time of the doctoral defense, the following papers were unpublished and had a status as follows: Paper 3: Manuscript. The conjunctive use of bonded repairs and crack growth retardation techniques

In an attempt to find a way of improving the damage tolerance of composite bonded repairs to metallic aircraft structures, the effect of using conventional crack growth retardation techniques in conjunction with bonded repairs was experimentally investigated. Hence, an experimental test program was set up to determine whether fatigue crack growth under bonded repairs is retarded further by giving the crack to be repaired a crack growth retardation treatment prior to repair patch application. In addition, it was set up to determine the influence of a bonded repair on the effectiveness of a crack growth retardation method. Centrally cracked aluminium plates were used. Stop drilling followed by cold hole expansion and the application of single overloads were selected as retardation treatments. Two patch materials were considered; boron/epoxy and Glare 2. Further test variables were the aluminium alloy and the plate thickness. Fatigue testing was carried out under constant amplitude loading and baseline results were determined first. In addition to optically monitoring the crack growth, local and global out-of-plane deformations were visualised with holographic interferometry and shadow moire??. Furthermore, the stress intensity factors under the repair patch were examined with strain gauges and measurement of the central crack opening displacement. Disbonds and fracture surfaces were studied after residual strength tests. The crack growth results obtained showed that retardation treatments decrease crack growth rates under a repair patch and that the effectiveness of a retardation treatment is increased by the patch. Although identical crack growth rates were observed under boron/epoxy and Glare 2 patches, the reinitiation period after the retardation treatment lasted longer when Glare 2 patches were applied. Analytical predictions of the extent of retardation based on existing models showed that the conjunctive effect of retardation treatments and bonded repairs was underestimated. A sustained reduction in crack growth rates was observed under bonded repairs with a prior overload retardation treatment. It was concluded that the damage tolerance of bonded repairs is increased by the application of a crack growth retardation treatment because the crack growth is retarded further. These findings indicate that the range of cracks in aircraft for which bonded repairs can be considered is expanded and that economic benefits can be obtained.

Bayesian geoacoustic inversion of seabed reflection data at the New England mud patch

This thesis presents Bayesian geoacoustic inversion of seabed reflection-coefficient data as part of the U.S. Office of Naval Research Seabed Characterization Experiment 2017 at the New England Mud Patch. First, a linearized, ray-based Bayesian inversion of acoustic arrival times is carried out for high-precision estimation of experiment geometry and uncertainties, representing an important first step to inferring seabed properties using geoacoustic inversion methods such as reflection inversion. The high-precision estimates for source-receiver ranges, source depths, receiver depths, and water depths at reflection points along the survey track are used to calculate grazing angles, with angle uncertainties computed using Monte Carlo methods. The experiment geometry uncertainties are obtained using analytic linearized estimates, and verified with nonlinear analysis. Second, a trans-dimensional (trans-D) Bayesian inversion of reflection-coefficient data is carried out for geoacoustic parameters and uncertainties of fine-grained/cohesive sediments. The trans-D inversion samples probabilistically over an unknown number of seabed interfaces and the parameters of a zeroth- or first-order autoregressive error model. The numerical method of parallel tempering reversible jump Markov-chain Monte Carlo sampling is employed. Spherical-wave reflection coefficient modelling is applied using plane-wave decomposition in the Sommerfeld integral. The inversion provides marginal posterior probability profiles for Buckingham's viscous grain-shearing parameters: porosity, grain-to-grain compressional modulus, material exponent, and compressional viscoelastic time constant as a function of depth in the sediment. These parameters are used to compute dispersion relationships for each layer in the model, providing marginal posterior probability profiles for compressional-wave velocity and attenuation at different frequencies, as well as density. The geoacoustic inversion results are compared to independent measurements of sediment properties. / Graduate

Nos ensaios funcionais in vitro, anéis de artéria mesentérica superior isolada de animais hipertensos e normotensos foram précontraídos com FEN (1μM) e o efeito de carvacrol (10-8-10-3M) foi observado. Em SHR, este monoterpeno induziu vasodilatação dependente de concentração (pD2=5,13 ± 0,05; Emáx=115,14 ± 5,46%; N=8) e, após a remoção do endotélio funcional, a potência da droga foi alterada significantemente (pD2=4,91 ± 0,05 N=9; p<0,01), sugerindo que a resposta vasodilatadora induzida por carvacrol, provavelmente, envolve uma via dependente e outra independente do endotélio vascular, porém, esta última parece ser a majoritária e, por isso, os ensaios seguintes foram realizados na ausência do endotélio vascular. Interessantemente, quando comparada com animais normotensos, a potência farmacológica de carvacrol foi reduzida significantemente (pD2=4,91 ± 0,05; N=9; p<0,05). Em anéis de ratos hipertensos, carvacrol reduziu o influxo de Ca2+ por canais Cav tipo-L, SOC e ROC, estes resultados foram semelhantes aos obtidos em ratos normotensos. Em ratos hipertensos, mas não em normotensos, a potência farmacológica do carvacrol em anéis pré-contraídos com FEN e na presença de diferentes inibidores de canais TRP (íon Gd3+, 10-5M; 2-APB, 10-6M ou 10-5M; BCTC, 2μM; 9-fenantrol, 10-5M; ou HC03003-1, 10-5M), foi reduzida em relação ao controle na ausência destes bloqueadores, sugerindo que os canais sensíveis à estes bloqueadores (TRPC1-7, TRPM2, M4 e TRPM8, TRPV1 e TRPA1), provavelmente, estão participando dos efeitos vasculares mediados por carvacrol e podem estar envolvidos no processo hipertensivo. Em estudos de patch-clamp em células de artéria mesentérica dispersas de ratos hipertensos, carvacrol (300μM) reduziu as correntes de entrada de Ba2+ por Cav tipo-L e este efeito foi semelhante em ratos normotensos. Além disso, em células de ratos hipertensos, o Mg2+ (2,5mM), bloqueador do TRPM6 e TRPM7, reduziu as densidades de ITRPM de entrada e saída, assim como carvacrol (100μM e 300μM), na ausência ou presença do 2-APB (100μM), bloqueador de TRPM7. A presença do 2-APB provocou inibição adicional nas densidades de ITRPM pelo carvacrol (100μM, mas não 300μM). Altas concentrações intracelulares de Mg2+ reduziram the magnitude of ITRPM7. Foi evidenciado que a ITRPM no controle é menor em ratos hipertensos que em normotensos. Estes dados obtidos e os relatados na literatura são sugestivos para provável inibição de ITRPM7 por carvacrol em células mesentéricas nativas. O efeito anti-hipertensivo do carvacrol foi avaliado por administração via orogástrica (50mg/kg/dia) durante 20 dias foi capaz de reduzir a pressão arterial média dos animais SHR tratados, no 20º dia do tratamento. O tratamento subcrônico com carvacrol não alterou os pesos cardíaco e corpóreo, nem a reatividade vascular. Em conclusão, esses dados sugerem que carvacrol possui atividade anti-hipertensiva em animais SHR, que pode ser devido ao seu efeito vasodilatador em anéis de artéria mesentérica superior isolada, provavelmente, por inibição do influxo de Ca2+ por Cav tipo-L, ROC, SOC e/ou canais TRPC1, 3 ou 6, além da inibição de correntes tipo-TRPM7 em miócitos mesentéricos.

Understanding wildlife distribution in the human-dominated landscape of Nepal:implications for conservation

In this thesis, I have first reviewed biodiversity status and its conservation in Nepal, which indicates the need of linking the gaps between research and conservation of rare and endangered flora and fauna. Using three mountain ungulates as model species (barking deer - Muntiacus muntjak, Himalayan goral - Naemorhedus goral and Himalayan serow - Capricornis thar), I have investigated effects of human disturbances on wildlife distribution in the human-dominated landscapes of western Nepal, spanning from the subtropical Bardia National Park to the mountainous Shey Phoksundo National Park. I have developed habitat suitability maps for these three ungulate species and recommended a conservation priority area for their conservation. A special emphasis was placed on the study of the distribution of Himalayan serow using different factors related to habitat fragmentation, hunting and patch characteristics and connectivity of forest in midhills landscape of Nepal. Finally, wildlife hunting pattern in the region was investigated in order to explore wildlife conservation issues from the social perspective.

O objetivo deste estudo foi investigar o papel das células dendríticas das placas de Peyer (PP) na determinação do padrão de resposta imune, Th1 e Th2, durante a infecção por via intragástrica de camundongos suscetíveis (BALB/c) e resistentes (C57BL/6) com a amostra virulenta de Y. pseudotuberculosis (YpIII pIB1 – Yp+) ou seu par isogênico, curado do plasmídeo de virulência (YpIII – Yp-). As DCs das PP foram obtidas no 1°, 3° e 5° dia pós-infecção, quantificadas e analisadas quanto às suas subpopulações, expressões de moléculas de superfície e capacidade imunoestimulatória por citometria de fluxo, e quanto à secreção de citocinas (IL-4, IL-10, IL-12 e TNF-α) por ELISA. Os linfócitos das PP também foram obtidos no mesmo período e tiveram suas sub-populações e o padrão de citocinas intracelulares Th1/Th2 (IL-2, IL-4, IL-10 e IFN-γ) analisado por citometria de fluxo. A infecção por Yp+ reduziu o número de DCs no 1° dia pós-infecção e aumentou, no período inicial, a expressão de B7.1 e B7.2 nos camundongos BALB/c. Nos camundongos C57BL/6 reduziu o número de DCs durante todo o período analisado, aumentou a expressão de B7.1 e B7.2 no período inicial e a expressão de ICAM-1. A infecção por ambas as amostras provocou redução da sub-população CD8α+ e da expressão de MHC II nas duas linhagens de animais, aumentou a sub-população CD11b+ nos animais suscetíveis e diminuiu nos animais resistentes. Studies using in vitro infection demonstrated that Y. enterocolitica can have as a target dendritic cells (DCs), affecting several of its functions, including their maturation and production of cytokines, and, consequently, contributing to the diminished activation of the T CD4+ cells. The aim of this study was to investigate the role of dendritic cell from Peyer’s patches (PP) in determining of immune response pattern, Th1 and Th2, during infection by the intragastric route in susceptible (BALB/c) and resistant (C57BL/6) mice with a virulent sample of Yersinia pseudotuberculosis (YpIII pIB1 – Yp+) or its isogenic pair, cured of the virulence plasmid (YpIII – Yp-). The PP DCs were obtained on the 1st, 3rd and 5th days postinfection, quantified and analyzed as far as their subpopulations, expressions of surface molecules and immunostimulatory capacity by flow cytometry, and the cytokines secretion (IL-4, IL-10, IL-12 and TNF-α) by ELISA. The PP lymphocytes were also obtained in the same period, and had their subpopulations and the pattern of intracellular Th1/Th2 cytokines (IL-2, IL-4, IL-10 and IFN-γ) analysed by flow cytometry. The infection by Yp+ reduced the number of DCs on the 1st day post-infection and increased, in the initial period, the expression of B7.1 and B7.2 in BALB/c. In C57BL/6 mice reduced the number of DCs throughout the study period, increased the expression of B7.1 and B7.2 in the initial period and the expression of ICAM-1. The infection by both samples reduced CD8α+ subpopulation and expression of MHC II in both animals, increased CD11b+ sub-population in susceptible animals and reduced the same sub-population in resistant animals.
