Artificial Ground Freezingin Clayey Soils : Laboratory and Field Studies of Deformations During Thawing at the Bothnia Line

Artificial ground freezing as a method to temporarily stabilize and create hydraulic sealing in urban as well as in rural areas has been used in a number of Swedish construction projects, particularly during the last decade. One problem with the freezing of soil and rock is that fine-grained clayey types of soils have showed a tendency to under certain circumstances, during the thawing process, create a pore water overpressure and to consolidate, despite a change in the external loading conditions. In certain cases, this condition can be a desired effect as the soil mass after a freeze- and thaw cycle acquires overconsolidated properties. The main objectives of this study are, to describe and review the knowledge and current state of practice of artificial ground freezing, to increase the understanding about the conceptual behaviour for prognosis of the vertical deformation concerning artificial ground freezing and to compare and discuss results from laboratory and field studies concerning vertical deformation during thawing process for Bothnia soil. The field studies and the laboratory tests in this research study have been performed with soil from the freezing of the Bothnia Line in the vicinity of Stranneberget. The Bothnia Line is the railway link between Nyland, north of Kramfors, and Umeå. This thesis relates to a part of the Bothnia Line. It deals with the behaviour of soil during thawing by means of temporary stabilization and hydraulic sealing of fine-grained soil through artificial freezing using brine as the cooling agent. However, the reason behind the problem consists of the final deformations due to the thawing process. The general conclusions of this study are; the Bothnia soil water content decreased in mean approximately 14 % after a freeze-thaw cycle, which approximately corresponds to; wth = 0.8w – 1.5 the decrease of the water content has no correlation to the depth below ground surface, in contrast, there is a strong correlation between the undisturbed soil water content and the magnitude of the decrease in water content the soil liquid limit decreases after a freeze-thaw cycle, simultaneously as the relative share of clay and fine silt grains decreases while the relative share of more coarse grains increases the coarser and denser soil created after a freeze-thaw cycle obtains an increased preconsolidation pressure and an increased undrained shear strength. / QC 20100721

Adipositas ist besonders in den Industrienationen ein zunehmendes gesellschaftliches und ökonomisches Problem. Dabei sind vor allem die kardiovaskulären und metabolischen Begleiterkrankungen von entscheidender Bedeutung. In den letzten Jahren konnte gezeigt werden, dass verschiedene Adipozyten- und Hepatozyten-sezernierte Proteine Mediatoren von Insulinresistenz und Dyslipidämie darstellen. Kürzlich wurde Angiopoietin-related growth factor (AGF) als ein neues, von der Leber produziertes Protein, das potentiell Insulinresistenz und Adipositas antagonisiert, vorgestellt. Im Mausmodell waren AGF-überexprimierende Tiere schlanker und insulinsensitiver verglichen zu Kontrolltieren. Zudem entwickelten AGF-knockout-Mäuse eine Adipositas, Insulinresistenz sowie eine Leber- und Skelettmuskelverfettung. Weiterhin fand sich in epidermalen Keratinozyten eine Hypervaskularisierung bei transgenen Mäusen mit AGF-Expression. Dies macht AGF möglicherweise zu einem Zielgen in der Behandlung moderner Zivilisationskrankheiten, wie z.B. dem Diabetes mellitus Typ 2 (DMT2). Bisherige Publikationen über AGF basieren zumeist auf Tiermodellen. Über die Regulation beim Menschen existieren dagegen bislang nur wenige Studien. In der vorliegenden Arbeit wurde AGF im Serum verschiedener Patientenpopulationen mit einem erhöhten kardiovaskulären Risikoprofil (Patienten mit chronischer Dialysepflicht, DMT2, Präeklampsie [PE]) mittels enzyme-linked immunosorbent assay quantifiziert und mit Kontrollpatienten verglichen. Die Ergebnisse zeigen, dass AGF bei Patienten mit DMT2 im Vergleich zu Nichtdiabetikern signifikant erhöht ist. Bei terminal-niereninsuffizienten Patienten dagegen fanden sich signifikant niedrigere AGF-Konzentrationen im Serum. Bei PE-Patientinnen waren signifikant höhere AGF-Spiegel nachweisbar verglichen zu gesunden schwangeren Kontrollen. Die vorgestellten Daten weisen darauf hin, dass erhöhte AGF-Spiegel bei DMT2 und PE eine physiologische Gegenregulation darstellen könnten, die der Insulinresistenz bei DMT2 bzw. antiangiogenetischen Faktoren bei PE entgegenwirkt. Alternativ wäre – ähnlich der Insulinresistenz – eine Resistenz von Patienten mit DMT2 bzw. PE gegen AGF möglich mit einer reflektorischen Erhöhung dieses Hepatozyten-exprimierten Faktors. Die genaue Rolle von AGF bei kardiovaskulären Risikopatienten muss in zukünftigen Arbeiten noch weiter aufgeklärt werden.

Analysis of Spleen-Induced Fimbria Production in Recombinant Attenuated Salmonella enterica Serovar Typhimurium Vaccine Strains

Salmonella enterica serovar Typhimurium genome encodes 13 fimbrial operons. Most of the fimbriae encoded by these operons are not produced under laboratory conditions but are likely to be synthesized in vivo. We used an in vivo expression technology (IVET) strategy to identify four fimbrial operons, agf, saf, sti, and stc that are expressed in the spleen. When any three of these operons were deleted, the strain retained wild-type virulence. However, when all four operons were deleted, the resulting strain was completely attenuated, indicating that these four fimbriae play functionally redundant roles critical for virulence. In mice, oral doses of as low as 1 x 10(5) CFU of the strain with four fimbrial operons deleted provided 100% protection against challenge with 1 x 10(9) CFU of wild-type S. Typhimurium. We also examined the possible effect of these fimbriae on the ability of a Salmonella vaccine strain to deliver a guest antigen. We modified one of our established attenuated vaccine strains, chi 9088, to delete three fimbrial operons while the fourth operon was constitutively expressed. Each derivative was modified to express the Streptococcus pneumoniae antigen PspA. Strains that constitutively expressed saf or stc elicited a strong Th1 response with significantly greater levels of anti-PspA serum IgG and greater protective efficacy than strains carrying saf or stc deletions. The isogenic strain in which all four operons were deleted generated the lowest anti-PspA levels and did not protect against challenge with virulent S. pneumoniae. Our results indicate that these fimbriae play important roles, as yet not understood, in Salmonella virulence and immunogenicity. IMPORTANCE Salmonella enterica is the leading cause of bacterial food-borne infection in the United States. S. Typhimurium is capable of producing up to 13 distinct surface structures called fimbriae that presumably mediate its adherence to surfaces. The roles of most of these fimbriae in disease are unknown. Identifying fimbriae produced during infection will provide important insights into how these bacterial structures contribute to disease and potentially induce protective immunity to Salmonella infection. We identified four fimbriae that are produced during infection. Deletion of all four of these fimbriae results in a significant reduction in virulence. We explored ways in which the expression of these fimbriae may be exploited for use in recombinant Salmonella vaccine strains and found that production of Saf and Stc fimbriae are important for generating a strong immune response against a vectored antigen. This work provides new insight into the role of fimbriae in disease and their potential for improving the efficacy of Salmonella-based vaccines.

The main objective of my thesis is to evaluate a suitability of the project of the centralisation of management of agriculture commodities transportation in AGF Logistics. The evaluation is based on the analysis of operational and economic data. The results of the application of theoretical models confirmed my hypothesis that the project of a centralisation is a suitable solution for performance optimisation. It is possible to compare the actual state of the project with expected goals based on acquired data and find out the opportunities and risks connected with the project.

QUALIDADE DE VIDA E EFICÁCIA ADAPTATIVA DE PESSOAS COM OBESIDADE / QUALITY OF LIFE AND ADAPTIVE EFFICACY OF PEOPLE WITH OBESITY

Since it is a hard to control pathology that favors the emergence of other health conditions, there is a pressing need to undertake research that might contribute to enhance treatment, as well as improve quality of life and adaptive efficacy. Therefore, this study aims at evaluating obese people s quality of life perception (QL) and adaptive efficacy by relating the results obtained from evaluation of their QL perception with the results from adaptive efficacy, as well as the results from the Global Functioning Evaluation (AGF). For that purpose, the WHOQOL-100 Portuguese-version questionnaire was employed to evaluate their quality of life perception, and a Preventive Diagnosis Interview Functional Adaptive Diagnostic Scale (FADS) was also undertaken as regards adaptive efficacy, to evaluate thirty obese persons (Body Mass Index BMI >=30 kg/m2), with an average age of 52.33, who use the services of anambulatory care center located in the Greater ABC region, in the State of Sao Paulo. The majority of participants have degree I of obesity 46.70%, 33.30% have degree II and 20.00% degree III. The majority started to gain weight at the start of pregnancy (43.30%). The second period during which subjects started to lose control of their body weight occurred between the ages of 40 and 50 (20.00%). In overall evaluation of QL, it has been observed that domain VI Spiritual/Religious/Personal Belief Aspects had the highest average score (16.17 - DP=2.95 [equivalent to 80.83% of the maximum score that could be obtained]), when compared to the remaining evaluated domains. On the other hand, domain I Physical/Fitness provided the owest average score (11.77 - P=2.78 58.83%). All participants are under adaptive nefficacy: Group 2 Light Inefficacy (26.7%), Group 3 oderate Inefficacy (33.3%) and group 4 Severe Inefficacy (30.0%). As regards the global functioning evaluation (GFE),36.67% are observed to fall in the interval between 51-60 points; 23.33% of participants are in the interval between 31-50 points; and only 23.33% obtained a score above 70 points. By correlating the results from the evaluations, strong, positive and significant relations have been found between the evaluation on quality of life perception, adaptive efficacy and global functioning. / A obesidade é uma doença crônica que vem acometendo, progressivamente, cada vez mais pessoas no mundo. Por ser uma patologia de difícil controle que favorece a eclosão de outros agravos à saúde, é premente a necessidade de realização de pesquisas que possam contribuir para o aperfeiçoamento dos tratamentos, bem como para a melhoria da qualidade de vida e eficácia adaptativa. Sendo assim, a presente pesquisa visou avaliar a percepção da qualidade de vida (QV), a eficácia adaptativa (EDAO) e o funcionamento global (AGF) de pessoas com obesidade, relacionando os resultados obtidos na avaliação da percepção da QV com os da eficácia adaptativa, bem como aos do funcionamento global (AGF). Para tanto, utilizou-se o questionário WHOQOL-100 versão em português para avaliação da percepção de qualidade de vida, a Entrevista Diagnóstica Preventiva Escala Diagnóstica Adaptativa Operacionalizada (EDAO) para a eficácia adaptativa e a Escala de Avaliação do Funcionamento Global (AGF) para o funcionamento global. Este estudo contou com a participação de trinta mulheres obesas (Índice de Massa Corporal IMC >=30 kg/m2), com idade média de 52,33 anos, que utilizavam os serviços de um ambulatório situado na região do Grande ABC, estado de SP. A maioria das participantes se encontrava no grau I de obesidade - 46,70%, situava-se no grau II 33,30% e 20,00% no grau III. O aumento de peso da maioria teve início nas gestações (43,30%), o segundo período onde ocorreu o início do descontrole do peso corporal foi entre os 40 aos 50 anos (20,00%). Na avaliação geral da QV, observou-se que no domínio VI Aspectos Espirituais/Religião/Crenças Pessoais foi encontrado o maior escore médio (16,17 - DP=2,95 [equivalente a 80,83% do escore máximo que poderia ser obtido]), comparando-o com os demais domínios avaliados. Em oposição, o domínio I Físico foi o que apresentou o menor escore médio (11,77 - DP=2,78 - 58,83%). Todas as participantes se encontravam em ineficácia adaptativa: Grupo 2 Ineficaz Leve (26,7%), no Grupo 3 Ineficaz Moderada (33,3%) e no Grupo 4 Ineficaz Severa (30,0%). Quanto à avaliação do funcionamento global (AGF), notou-se que 36,67% se situavam no intervalo entre 51-60 pontos. 23,33% das participantes no intervalo entre 31-50 pontos. Apenas 23,33% tiveram pontuação acima de 70 pontos. Relacionando os resultados das avaliações, foram encontradas correlações fortes, positivas e significativas entre a avaliação da percepção de qualidade de vida, da eficácia adaptativa e do funcionamento Global.
