Characterisation and improvement of whiskey yeast

Thesis (MSc)--Stellenbosch University, 2003. / ENGLISH ABSTRACT: Scotch whiskey is of two main types, namely Scotch malt whiskey, made from malted barley alone, or Scotch grain whiskey, made from cereals, such as wheat or maize, together with malted barley. In both processes, the enzymes from the barley are responsible for starch conversion and should always be derived entirely from the malted barley. No exogenous enzymes are allowed to be added to any mashing. The enzymes involved in the conversion process to fermentable sugars, are the aand p-amylases, limit dextrinase and p-glucosidase. Maize, on the other hand, contains no enzyme activity, therefore enzymes need to be added when producing whiskey from maize alone. In other whiskey-producing countries where maize is freely available and cheaper than barley, the use of exogenous enzymes are allowed in the mashing process and is crucial for the formation of fermentable sugars from complex carbohydrates. The cost of the enzymes, however, can push the production cost of whiskey to higher levels. Saccharomyces cerevisiae does not have any amylolytic activity, but is an excellent fermenter and produces favourable organoleptic notes, which makes it very suitable for producing potable spirit. Efforts have been made to genetically improve industrial strains, relying on classical genetic techniques followed by the selection of broad traits, such as ethanol tolerance, absence of off-flavours and carbohydrate/starch utilisation. No strain has thus far been selected for total starch degradation during the fermentation of whiskey mash. Over the last decade, considerable progress has been made in the development of genetically improved strains for the distilling, wine, brewing and baking industries. The expression of heterologous genes introduced a new dimension in approaches to the genetic improvement of industrial strains. It would therefore be cost-effective to use a yeast strain that can produce active and sufficient enzymes to ferment raw starch efficiently to alcohol without lowering the quality of the end product. No such strain has been developed to date, but the continuous improvement of starch-utilising strains has made this goal more achievable. Two a-amylase genes, namely LKA 1 and LKA2, were previously isolated from Lipomyces kanonenkoae. In this study, we selected 4 strains on the basis of criteria that are important for whiskey-specific strains. The selected strains were transformed with LKA 1, as well as with a combination of LKA 1 and LKA2 genes. The wine yeast VIN13 was included in the transformation of LKA1 and LKA2 because of its rapid fermentation rate. The genes were integrated into the genomes of the yeast strains and were stable after many generations. Assays showed that a significant increase in enzyme activity was induced in the whiskey strains, compared to the untransformed strains. The strains also showed good fermentation ability in whiskey fermentations, although optimum alcohol production was still not achieved. / AFRIKAANSE OPSOMMING: Skotse whiskey bestaan uit 2 tipes, nl. mout whiskey, gemaak slegs van mout d.w.s. gars wat die mout proses ondergaan het, en graan whiskey wat gemaak word van gewasse soos mielies of koring, waarby mout gevoeg word. Die ensieme afkomstig van die mout is verantwoordelik vir die omsetting van stysel na fermenteerbare suikers en geen eksogene ensieme mag by die gars- of graanmengsel gevoeg word nie. Die ensieme wat betrokke is by die omsetting van stysel, is die a- en ~- arnitases, limiet dekstrinase en ~-glukosidase. Mielies bevat geen ensiemaktiwiteit nie, dus moet ensieme by die proses gevoeg word indien slegs mielies vir die vervaardiging van whiskey gebruik word. In whiskey produserende lande waar mielies vryelik beskikbaar is en goedkoper is as gars, word eksogene ensieme by die graanmengsel gevoeg vir die vrystelling van fermenteerbare suikers vanaf komplekse koolhidrate. Die hoë koste van die ensieme kan egter die produksiekoste van whiskey verhoog. Saccharomyces cerevisiae besit geen amilolitiese aktiwiteit nie, maar is 'n uitstekende fermenteerder en produseer gewensde organoleptiese geure. Om hierdie redes is S. cerevisiae baie geskik vir die produksie van drinkbare etanol. Navorsingspogings om industriële rasse geneties m.b.v. klassieke genetiese metodes te verbeter, kom wydverspreid in die literatuur voor. Dit sluit in die seleksie van rasse met 'n verskeidenheid van eienskappe soos etanol toleransie, die afwesigheid van afgeur produksie en koolhidraat/stysel benutting. Geen ras is egter tot op hede geselekteer vir totale stysel afbraak gedurende fermentasie nie. Groot vordering is gedurende die laaste dekade gemaak in die ontwikkeling van genetiese verbeterde rasse vir die wyn- stokery- en brouers industrieë. Die uitdruk van heterogene gene in gisrasse gee 'n nuwe dimensie aan die genetiese verbetering van industriële rasse. Die gebruik van 'n gisras wat aktiewe en genoegsame ensieme produseer om rou stysel te fermenteer, sonder om die kwalitiet van die eindproduk nadelig te beïnvloed, kan die produksiekoste van whiskey aansienlik verminder. Geen gisras met hierdie eienskap is tot op hede ontwikkel nie, maar die voortdurende verbetering van rasse om stysel af te breek maak hierdie doel meer bereikbaar. Twee a-amilase gene, nl. LKA 1 en LKA2 is voorheen uit Lipomyces kononenkoae geïsoleer. In hierdie studie is 4 gisrasse geselekteer op grond van die kriteria wat nodig is vir whiskey giste. Die geselekteerde rasse is getransformeer met LKA 1 sowel as 'n kombinasie van LKA 1 en LKA2 gene. Die wyngis VIN13 is ingesluit by die transformasie met die LKA1 en LKA2 gene, omrede VIN13 bekend is as 'n vinnige fermenteerder. Die gene is geïntegreer in die genoom van die verskillende gisrasse en is stabiel na vele generasies. Die getransformeerde rasse het 'n betekenisvolle verhoging in ensiemaktiwiteit teenoor die nie-getransformeerde rasse getoon. AI die transformante het ook goeie fermentasie vermoë getoon in whiskey fermentasie proewe. Optimum alkoholproduksie is egter nie verkry nie.

Nitrogen utilisation of selected non-Saccharomyces yeasts and the impact on volatile compound production

Thesis (MSc)--Stellenbosch University, 2015. / ENGLISH ABSTRACT: During fermentation, nitrogenous compounds serve as nutrients for the yeasts, which enable their growth, functioning and maintenance of the yeasts cells. From a winemaking perspective, a certain amount of nitrogen is required for the yeasts in order to avoid sluggish or stuck fermentation. Moreover, nitrogen metabolism leads to the production of aroma compounds such as higher alcohols, fatty acids and esters which contribute positively to overall sensory characteristics of wine. Nitrogen metabolism (uptake of ammonium and amino acids) have been extensively studied in Saccharomyces cerevisiae. Nonetheless, the fairly great variances observed between strains in terms of preference for certain nitrogen sources and metabolism thereof are not so well understood. Additionally, these mechanisms nitrogen metabolism of non- Saccharomyces yeasts are even vaguer and simply assumed to be globally similar to those of S. cerevisiae. This study aimed to investigate the uptake of nitrogen compounds (ammonium and individual amino acids) by selected non-Saccharomyces yeasts (Lachancea thermotolerans IWBT Y1240, Torulaspora delbrueckii Biodiva TD291, Pichia kluyveri FrootZen, Metschnikowia pulcherrima IWBT Y1123 and Metschnikowia pulcherrima Flavia) to assess the impact of fermentation kinetics and the production of aroma compounds during sequential fermentations with S. cerevisiae under different initial YAN concentrations, with 300 mg/L, 150 mg/L and 75 mg/L, respectively). Fermentations were performed in a synthetic grape juice medium with pure and sequential fermentations. The data showed that the assimilation of nitrogen compounds were species specific. For example, L. thermotolerans preferred alpha amino nitrogen above ammonia, where the opposite hold true for T. delbrueckii. Notable differences could also be identified for the uptake of certain single amino acids. Irrespective of the initial YAN concentrations during sequential fermentations, the yeasts only assimilated about half of the initial YAN. The non-Saccharomyces yeasts did not influence fermentation performance during sequential fermentations. However, a low initial YAN (75 mg/L) had a strong influence on the fermentation kinetics and aroma compound production. The higher uptake (compare to S. cerevisiae) of specific single amino acids by non-Saccharomyces yeasts (especially L. thermotolerans), can be tentatively correlated with certain aroma compounds produced at the end of fermentation. The results also revealed that agitation could impact overall fermentation performance and aroma compound production. This study contributes to an improved understanding of how different initial nitrogen concentrations affect growth, fermentation performances and aroma compound production of wine-related yeasts under fermentative conditions. Moreover, the uptake of single amino acids by selected non-Saccharomyces yeasts had also been identified, which is a good starting point to better understand non- Saccharomyces yeasts nitrogen requirements which may be used for the optimization of nitrogen source addition, during alcoholic fermentation, when used in mixed fermentations in order to ensure a complete alcoholic fermentation. To the best of our knowledge, the uptake of single amino acids and YAN consumption by selected non-Saccharomyces yeasts under fermentation conditions tested, have never been studied before. / AFRIKAANSE OPSOMMING: Tydens wynfermentasies dien talle stikstof komponente as voedingstowwe vir wyngis wat hul groei, funksie en onderhoud bevorder. Van `n wynmaak perspektief word daar `n sekere hoeveelheid stikstof benodig deur die wyngis om te verhoed dat slepende of onvolledige fermentasies plaasvind. Verder lei stikstofmetabolisme na die produksie van aroma verbindings, soos hoër alkohole, vlugtige vetsure en esters wat positief bydra tot die sensoriese karaktereienskappe van wyn. Die stikstofmetabolisme (opneem ammonium en aminosure) is deeglik nagevors in die wyngis Saccharomyces cerevisiae, maar die klein variasies waargeneem tussen die gisras in terme van die voorkeur van sekere stikstof komponente is egter nog onduidelik. Daarbenewens is die stikstofmetabolisme nog meer onbekend in nie- Saccharomyces wyngis en word dit oor die algemeen aanvaar dat die werking van die stikstofmetabolisme dieselfde is as in S. cerevisiae. Die studie dra by om beter te verstaan van hoe verskillende aanvangsstikstof die groei, fermentasie kinetika en aroma produksie beïnvloed onder fermentasie kondisies. Die opneem van sekere aminosure deur nie-Saccharomyces gis word ook beskryf, wat `n goeie beginpunt is om beter te vertaan wat die stikstof vereistes vir die geselekteerde wyngis is, wat gebruik kan word vir die optimisering van stikstofaanvullings, sodat die risiko van probleemfermentasies verlaag sal word. So ver as wat ons kennis strek is die opneem van aminosure en die gebruik van assimileerbare stikstof deur nie-Saccharomyces wyngis onder fermentasie kondisies nog nie ondersoek nie.

ALCOHOL CONSUMPTION IN A VILLAGE IN NORTH INDIA

Culture plays a significant role in defining how alcohol is consumed and in determining the attitudes of the nonconsuming population toward the consumer. While the mechanics of the relationship between alcohol consumption and culture have been extensively investigated in the industrialized world, there is a paucity of studies concerning developing nations. This study contributes to available data by examining in detail alcohol consumption within one North Indian caste group, the Rajputs. The Rajputs have played a unique role in Indian history, first as conquering invaders and later as martial rulers sworn to defend Hindu India. In the subject village, which is over 80 percent Rajput, they are the principal landholders and control the power structure. A survey of a variety of jatis within the village also confirmed their reputation as heavy drinkers. Investigation of the Rajput in relation to other jatis revealed significant differences in childrearing practices, family structure, marriage, religiosity and sex role differentiation between the two groups. This was seen to result in a different basic personality being evident in Rajputs and non-Rajputs. The Rajputs' ongoing ties to their marital past were found to dictate that Rajput males express their martiality by repressing a broad range of intense emotions, including fear, loneliness and despair, and that they restrict the expression of intimacy in personal relationships. Family life, childrearing, marriage and sexual relationships were all subordinated to this overriding concern. Alcohol plays a crucial role in this cultural scheme in that it is viewed as a culturally-sanctioned "escape valve," its consumption resulting in "disinhibiting" behavior not usually associated with the stoic Rajput personality. Evidence of this assigned role was seen in the high associations of alcohol consumption with the release of violence and sexual passions and its function as a social lubricant providing the means for the individual Rajput to function in social situations in which he feels uncomfortable. The sociocultural roles assigned to the consumption of alcohol in Rajput culture were found to bear a striking resemblance to those found in North American culture, specifically in the United States.

SILDENAFIL ATTENUATES ETHANOL-INDUCED CARDIOMYOCYTE INJURY AND PRESERVES CARDIAC FUNCTION THROUGH PROTEIN KINASE G-DEPENDENT SIGNALING

Background: Ethanol is a cardiotoxic substance that damages the heart by increasing apoptosis, free radical formation and calcium overloading. Consequently, there is an increase in cell death leaving fewer functioning myocytes leading to heart failure. Sildenafil is a phosphodiesterase type-5 (PDE-5) inhibitor approved for treatment of erectile dysfunction. Studies from our lab have demonstrated that PDE-5 inhibition reduces myocardial infarct size and attenuates post-ischemic cardiac dysfunction in both ischemia-reperfusion and permanent coronary artery ligation models. Therefore, in the present study, we hypothesized that treatment with sildenafil will prevent cardiotoxicity associated with acute alcohol exposure by reducing myocyte apoptosis and preserving cardiac function through PKG signaling. Methods and Results: Adult cardiomyocytes were isolated and treated with 100 mM of 100% ethanol ± 10 µM sildenafil. At 24 hours necrosis was assessed via trypan blue exclusion assay, JC-1 staining assessed mitochondrial membrane potential and ROS production was measured by DCF fluorescence. At 48 hours apoptosis was assessed by TUNEL assay. Ethanol increased the rate of necrotic and apoptotic cell death. This was attenuated by co-treatment with sildenafil. Ethanol disrupted the mitochondrial membrane potential and increased ROS production. Sildenafil preserved mitochondrial membrane potential and attenuated ROS production. Treatment of myocytes with 5-HD, a mitochondrial K+atp channel antagonist, blocked the protective effect of sildenafil. Knockdown of PKG using adenoviral siRNA blocked the protective effect of sildenafil, while overexpression of PKG1α conferred protection against ethanol cytotoxicity. To further demonstrate the effect of sildenafil ethanol-cardiotoxicity in vivo, mice were treated with ethanol (3 g/kg/day) with or without sildenafil (0.7 mg/kg) by i.p. injection for three consecutive days. After treatment, the animals were sacrificed and the hearts removed and perfused on a Langendorff system to measure function. After functional analysis, apoptosis and PKG activity was measured in the heart samples. Ethanol decreased the rate-force product and increased myocardial apoptosis. Sildenafil preserved cardiac function and significantly reduced apoptosis. Sildenafil treated myocardium also showed an increase in PKG activity. Conclusion: Sildenafil attenuates the toxic effect of ethanol by reducing apoptosis and maintaining the mitochondrial integrity in cardiomyocytes. Sildenafil also preserved cardiac function in ethanol-treated mice. Protein kinase G-dependent signaling plays a critical role in attenuating cardiotoxic effect of ethanol.

"It's like a Gordian Knot" : how older men in sobriety experience their emotions in therapy, using Interpretative Phenomenological Analysis

Older men are an under-researched population in Counselling Psychology. This thesis explores how older men in sobriety experience their emotions in therapy and aims to understand the meaning of these experiences from a psychologically gendered subject perspective. Semi-structured interviews were carried out with six older men (aged 62 and above) and interview transcripts were analysed using Interpretative Phenomenological Analysis (IPA), a qualitative methodology that focuses on the lived experience and the meaning people give to these experiences. The analysis highlighted significant challenges for older men when attempting to unlock, process or discuss emotions in therapy. The analysis also revealed positive transformational effects by those who were able to transcend the confines of gendered constructs with concerted emotional investments. The two master themes are: (1) CONTROL/REGULATION OF EMOTIONS including subthemes: Challenges to unlocking emotion; Importance of therapeutic emotional containment; Role of Alcoholics Anonymous (AA) in the emotion sharing experience and (2) TRANSFORMATION/ALLEGIANCE TO SELF, including subthemes: Selfactualisation; Going into the heart of emotions as an emotional-spiritual journey. Participants expressed challenges to unlocking their emotions in therapy,which were mainly experienced as controlled or suppressed. Aging, masculinity and helpseeking theories, as well as addiction and recovery literature, are discussed in light of the findings along with suggestions for future research and implications in Counselling Psychology.

Identificar e analisar a representação social dos enfermeiros de Hospital Geral frente ao paciente alcoolista e à etiologia para o alcoolismo. Os sujeitos do estudo foram 20 enfermeiros de três Hospitais Gerais de uma cidade do interior de São Paulo (Brasil). Os dados foram coletados por meio de entrevistas autogravadas que foram transcritas e analisadas pelo método da Análise de Conteúdo, tendo como categoria para análise o referencial teórico da representação social. Da análise dos dados, emergiram duas categorias empíricas: 1-Representação social dos enfermeiros frente ao paciente alcoolista e 2- Representação da etiologia para o alcoolismo Os resultados apontaram que os enfermeiros concebem o paciente alcoolista como uma pessoa doente, portanto, o alcoolismo como uma doença. Em relação ao trabalhar com esse tipo de paciente, alegaram que é de difícil manejo, tendo em vista que os profissionais não estão preparados para atuar com este tipo de paciente e que sua assistência é permeada pelo estigma e preconceito. Na representação desses enfermeiros, a etiologia do alcoolismo está relacionada às questões sociais, psicológicas, emocionais e biológicas. Concluiu-se que a representação dos sujeitos do estudo frente ao paciente alcoolista e a etiologia para o alcoolismo estão atreladas mais ao senso comum do que ao conhecimento técnico-científico, o que em última análise remete à necessidade de melhor preparo e capacitação dos profissionais enfermeiros sobre a temática álcool e alcoolismo, uma vez que o pouco conhecimento da temática pode contribuir para uma assistência fragmentada e pautada nas questões de ordem moral que permeiam o senso comum. / Identify and analyzed the social representation of General Hospital nurses front of the alcoholic patient and the etiology in order to alcoholism. The subjects of the study were 20 nurses from three General Hospital in a country town of São Paulo (Brazil).The data were collected through interviews self recorded that were transcribed and analyzed by The content Analysis Method, having as category for analysis the theoretical reference of social representation. From analysis of empirical data emerged two categories: 1 - Social representation of nurses front of alcoholic patient. 2- Representation of the etiology in order to alcoholism. The results pointed out that the nurses conceive the alcoholic patient as a sick person, therefore, the alcoholism as a disease. In relation to working with this type of patient, claimed that it is unwieldy, in order that professionals are not prepared to work with this type of patient and that its assistance is permeated by the stigma and prejudice. In the representation of these nurses, the etiology of alcoholism is related to social, psychological, emotional and biological issues. It was concluded that the representation of subjects of the study front of the alcoholic patient and the etiology in order to alcoholism are more tied to common sense than to scientific-technical knowledge, wich ultimately refers to the needs for better preparation and professional training of nurses on the thematic alcoholism, in that the few knowledge about the theme can contribute to a fragmented assistance and guided in matters of moral order that pervade common sense.

Esse estudo tem por objetivo validar o Michigan Alcoholism Screening Test - Geriatric Version (MAST-G) e identificar os problemas relacionados ao uso de bebida alcoólica entre os idosos usuários da Unidade Saúde da Família (USF) do município de São Carlos (SP). O desenho metodológico do estudo é do tipo descritivo de abordagem quantitativa. Os dados foram coletados por meio de um questionário contendo as informações sociodemográficas e o MAST-G, seguindo as etapas de tradução e adaptação transcultural. A amostra foi constituída por 111 pessoas com idade igual ou superior a 60 anos cadastrados na USF do município de São Carlos. Do resultado, a idade média foi de 70 anos, sendo 45% do sexo masculino e 55% do sexo feminino, escolaridade média de 3 anos e 92% residem com a família. O MAST-G apresentou um bom índice de confiabilidade, com Alfa de Cronbach ? = 0,7873 e por meio da curva de ROC mostrou uma boa especificidade e sensibilidade no valor de corte de 5 respostas positivas, corroborando a literatura internacional. Pode-se concluir que o instrumento é de fácil aplicação e pouco intimidativo, além de ser possível averiguar diversas questões acerca do comportamento do beber do idoso e assim, possibilitando um atendimento especializado, pontual para que o idoso tenha uma assistência de qualidade. / This study aims to validate the Michigan Alcoholism Screening Test - Geriatric Version (MAST-G) and to identify the pattern of consumption and alcohol use among elderly users of Family Health Units (USF) in the municipality of São Carlos (SP), Brazil. The methodological design of the study is a descriptive quantitative approach. Data were collected using an instrument that contains sociodemographic information and the MAST-G, following the steps of translation and cultural adaptation. The sample consisted of 111 people aged over 60 years who were enrolled in the USF of São Carlos. The result, the average age was 70 years, 45% male and 55% female, average schooling for 3 years and 92% living with family. The MAST-G had a good level of reliability, with Cronbach\'s alpha ? = 0.7873 and shows a good specificity and sensitivity in cut-off of 5 positive answers, as observed by the ROC curve, in good agreement with the literature. It could be concluded that the instrument is easy to be applied and less intimidating, besides being able to ascertain other questions about the behavior of the life of the elderly so it can allow a specialized service, timely so that the elderly have a better quality of assistance.

A preocupação com o uso do álcool entre universitários é evidente em várias partes do mundo. Diversos estudos mostram que o uso e abuso dessa substância vêm aumentando em ritmo acelerado. Nesse contexto, o presente estudo tem como objetivo identificar o padrão de consumo de álcool e suas conseqüências entre universitários da área de saúde de uma Faculdade privada do interior do estado de São Paulo. Trata-se de um estudo descritivo e exploratório com uma amostra de 1007 estudantes universitários. Para a coleta de dados, utilizou-se um questionário auto-aplicável, contendo três partes: a primeira referente aos dados sóciodemográficos, a segunda refere-se ao padrão de consumo de álcool e a terceira refere-se aos problemas vivenciados por essa população após beber. Entre os resultados estão que a maioria dos estudantes pertence ao sexo feminino 84%, estado civil solteiro 85,1%, com idade entre 18 e 25 anos 79%, provenientes de outras cidades 62,6% e 60,0% referiram não trabalhar. A maioria dos estudantes pertence ao curso de enfermagem 24,2% e em relação ao ano do curso, 30,3% são calouros. Quanto à moradia, 75,1% referiram morar com a família. No que se refere ao tipo de religião 64% afirmaram ser católicos e 67% referem que a religião é muito importante em suas vidas. Em relação ao consumo de álcool, 64% fazem uso de baixo risco, incluindo os abstêmios 11%, enquanto que 20% são bebedores de risco moderado e 5% bebedores de alto risco. A análise de associação demonstrou que o consumo de álcool é maior entre o gênero masculino, na faixa etária entre 18 e 25 anos, entre os casados, entre aqueles que tiram notas baixas em relação à média, entre os que moram sem a família, e entre aqueles que não pertencem a nenhum tipo de religião. O aparecimento de náuseas, vômito e ressaca, o fato de dirigir após beber ou dirigir bebendo, perder aulas por estar passando mal, ?matar aulas? após beber demais, apresentar problemas com a lei ou com a administração da faculdade por beber, tirar notas baixas, a freqüência que fuma, ser criticado por beber, brigar após beber ou ainda apresentar qualquer tipo de comportamento negativo por beber, foi encontrado com maior freqüência entre os universitários que apresentaram um maior consumo de álcool. A bebida de maior consumo foi a cerveja. Esses resultados nos evidenciam a importância do planejamento de estratégias de cunho preventivo no âmbito universitário, na tentativa de detectar precocemente aqueles com potencial para o abuso e possíveis problemas relacionados ao consumo dessa substância. / The worry about the alcohol use among college students is evident in several parts of the world. Many studies show that the use and abuse of this substance is increasing in accelerated rate. In this context, this study has as purpose identifying the standard of alcohol consumption and its consequences among health area college students from a Sao Paulo State private college. It?s a descriptive and explorative study with a 1007 college students? sample. For the data collect, it was used a self-applicable questionnaire that contains three parts: the first regarding the social demography data, the second refers to the standard of alcohol consumption and the third refers to the problems lived by this population after drinking. Among the results the most part of the students belongs to the female sex 84%, single 85,1%, age between 18 and 25 years old 79%; other cities provenance 62,6% and 60% referred that they don?t work. The most part of the students belongs to the Nursing course 24,2% and in relation to the course year, 30,3% are undergraduates. As for the residence, 75,1% referred to live with the family. According to the kind of religion 64% are Catholics and 67% referred that the religion is very important in their lives. In relation to the alcohol consumption, 64% are low risk users, including the teetotaler 11%, while 20% are moderate risk drinkers and 5% high risk drinkers. The association analysis showed that the alcohol consumption is larger among male gender, age group between 18 and 25 years old, married people, those who get low grades in relation to the average, who live without the family, and those who don\'t\' belong to any kind of religion. The nausea, vomit and hangover appearance, the fact of drive after drinking or to drive drinking, to lose classes after drinking too much, to have problems with the law or with the college administration for drinking, to get low grades, the often that smokes, being criticized for drink, to fight after drinking or to show any kind of negative behavior for drink, it was found with larger frequency among the college students who showed a larger alcohol consumption. The most consummated beverage was the beer. These results show us the importance of preventive strategies planning in the college scope at the attempt to detect prematurely those students with potential for the abuse and possible problems related to the consumption of this substance.

Uso da padronização interna em espectrometria de absorção atômica em forno de grafite na determinação de 'SE' e 'NI' em bebidas não alcoólicas /

Orientador: José Anchieta Gomes Neto / Resumo: Foram feitos estudos sistemáticos envolvendo padronização interna (PI) em determinações de elementos-traço em bebidas não alcoólicas por espectrometria de absorção atômica em forno de grafite (GFAAS). A influência da composição química das amostras, da concentração do padrão interno, do volume injetado de amostra, da diluição e do desgaste da superfície grafítica ilustraram a potencialidade de PI na melhoria da precisão e da exatidão analítica ou mesmo na minimização de efeitos de amostragem e de matriz. Arsênio e Ge foram avaliados como padrões internos na determinação de Se em água mineral gaseificada, água de coco e leite. Ambos melhoraram o desempenho analítico, seja no aspecto da precisão e exatidão como na minimização dos efeitos de matriz e amostragem. Nesse estudo foram utilizados como modificadores químicos Pd(NO3)2 + Mg(NO3)2 , W/Rh e W associado a co-injeção de Pd(NO3)2. Os desvios padrões relativos (%RSD) obtidos por meio da PI foram 0,5 a 3,5% (As) e de 1,0 a 3,1% (Ge), e na ausência da PI foram de 1,0 a 6,5%. A exatidão do método proposto foi avaliada por meio de teste de adição e recuperação de analito e os valores de recuperação (n=3) obtidos foram 94-109% (As) e 98-104% (Ge) com a PI e de 70-100% sem a PI. As concentrações de Se nessas amostras variaram de 5,0-25 æg L-1. No caso das amostras de água mineral gaseificada houve um aumento de 15% no tempo de vida útil do tubo e um acréscimo na confiabilidade dos resultados, apesar da superfície grafítica deteriorada. Cobalto foi avaliado como padrão interno na determinação de Ni em refrigerante e apresentou potencial para minimizar problemas de amostragem causados por gases dissolvidos. O modificador químico utilizado foi Mg(NO3)2. Os desvios padrões relativos obtidos foram de 0,5 a 3,4% e de 1,0 a 7,0% na presença e na ausência da PI, respectivamente... The influence of samples matrices, analyte concentration, injected sample volume, sample dilution and graphite surface degradation showed the potentiality of IS on the improvement of precision and accuracy of results, or in the minimization of matrix and sampling effects. Arsenic and Ge were evaluated as internal standards for the determination of Se in sparkling waters, coconut water and milk. Different chemical modifiers Pd(NO3)2 + Mg(NO3)2 , W/Rh e W + co-injection of Pd(NO3)2 were evaluated. Tolerância ao estresse e características fermentativas de leveduras Dekkera bruxellensis isoladas da fermentação alcoólica / Stress tolerance and fermentative characteristics of Dekkera bruxellensis yeasts isolated from the alcoholic fermentation

A espécie Dekkera bruxellensis tem sido detectada como a principal levedura contaminante em diversos processos fermentativos, dentre eles o de produção de etanol combustível, apresentando uma surpreendente capacidade de crescimento e adaptação naqueles substratos. No entanto, pouco se conhece sobre suas características de crescimento em condições de estresse e comportamento fermentativo. Desta forma, este trabalho objetivou avaliar a tolerância ao estresse e as características fermentativas exibidas por três linhagens de D. bruxellensis isoladas da fermentação alcoólica, além do efeito da contaminação em mosto de caldo de cana sobre os parâmetros fermentativos, buscando informações que possam contribuir para o manejo da fermentação alcoólica quando contaminada com esta levedura. Foram realizados testes de caracterização em meio YEPD sólido e líquido em condições estressantes para as três linhagens de D. bruxellensis e uma linhagem de S. cerevisiae (PE-02). O efeito do tratamento ácido associado ao etanol sobre a viabilidade das quatro linhagens em situação de agitação e sem agitação foi também avaliado. Em seguida, testes fermentativos em meio sintético (sem reciclo celular) e em meio de caldo de cana (com reciclo celular e utilizando-se ou não tratamento ácido) foram conduzidos para verificar as características fermentativas das linhagens de D. bruxellensis em comparação com S. cerevisiae, simulando-se uma contaminação por 103 células/mL da linhagem CCA155 em meio de caldo de cana. As três linhagens de D. bruxellensis apresentaram crescimento invasivo em meio YEPD sólido, possivelmente um mecanismo de sobrevivência da levedura em condições estressantes. Observou-se uma variação na resposta das linhagens às situações de estresse (baixo pH e alta concentração de etanol). Em condições não estressantes, a linhagem PE-02 apresentou melhor desenvolvimento, no entanto, em situações de estresse de pH, concentrações de açúcar e etanol, as linhagens de D. bruxellensis desenvolveram-se melhor. O controle eficiente do crescimento destas leveduras poderia ser obtido com um tratamento combinado de baixo pH (1,5) e etanol (9%), porém houve também prejuízo significativo à levedura S. cerevisiae, embora em menor extensão. Em sistema de batelada sem reciclo celular em meio sintético, verificou-se que a agitação influenciou significativamente a produção de etanol e ácidos por D. bruxellensis. O teor alcoólico foi maior quando se utilizou glicose como fonte de carbono ao invés de sacarose. Em sistema de batelada com reciclo celular em meio de caldo de cana, foram obtidos melhores resultados quanto à produção de etanol, menor teor de açúcar redutor total residual e maior eficiência fermentativa quando se utilizou o tratamento ácido do fermento (pH 1,5), assim como melhor controle do crescimento da linhagem CCA155 quando em cultura mista com S. cerevisiae (PE-02). O tratamento ácido utilizado teve efeito não só sobre o crescimento da levedura contaminante, mas também beneficiou a levedura do processo, resultando assim na minimização do efeito da contaminante sobre a fermentação conduzida em meio de caldo de cana sob dez ciclos fermentativos de 12 horas. / The species Dekkera bruxellensis has been considered as the main contaminant yeast in several fermentative processes, including the fuel alcohol production, showing a surprising growth capacity and adaptation in those substrates. However, a little is known about their growth characteristics in stressing conditions and fermentative profile. In this context, this work aimed to evaluate the stress tolerance and fermentative characteristics exhibited by three strains of D. bruxellensis isolated from the alcoholic fermentation, besides the effect of the contamination in sugar cane juice over the fermentative parameters, searching for information that could contribute to the management of the alcoholic fermentation when the medium is contaminated with this yeast. Characterization tests in YEPD medium (solid and liquid) under stressing conditions for the D. bruxellensis and S. cerevisiae (PE-02) strains were carried out. The effect of the acid treatment associated with ethanol over the cell viability of the four strains in shaken and non-shaken flasks was also evaluated. Following, fermentative tests in synthetic medium (without cell recycle) and in sugar cane juice (with cell recycle and with/without acid treatment) were carried out to verify the fermentative characteristics of the strains of D. bruxellensis comparing to S. cerevisiae, simulating a contamination by 103 cells/mL of the strain CCA155 in sugar cane juice. All the strains of D. bruxellensis showed invasiveness in YEPD agar medium, probably a survival mechanism in stressful conditions. A variation in the response of the strains to the stressing conditions (low pH and high ethanol concentration) was observed. In nonstressing situations, the strain PE-02 showed better growth; however, in stressing conditions of pH, ethanol and sugar concentrations, the strains of D. bruxellensis had better growth performance. The effective control of their growth could be obtained with a combined treatment of low pH (1.5) and ethanol (9%), however, a significant harmful effect to the S. cerevisiae strain was also verified, but in a lower extension. In batch system without cell recycle in synthetic medium, it was verified the influence of agitation over the ethanol and acid production by D. bruxellensis. The alcohol content was significantly higher when glucose was utilized instead of sucrose. In batch system with cell recycle in sugar cane juice, the best results for ethanol production, lower residual total reducing sugar and higher fermentative efficiency were obtained with the acid treatment of the ferment at pH 1.5, as well as a better growth control of the strain CCA155, in mixed culture with S. cerevisiae (PE-02). The acid treatment utilized in this work had effect not only over the growth of the contaminant yeast, but also benefited the yeast S. cerevisiae, resulting in the minimization of the effect of the contaminant over the fermentation developed in sugar cane juice medium in 12-hour ten fermentative cycles.
