Nitroxydes chiraux à squelette imidazolidin-4-one comme catalyseurs d'oxydation énantiosélective d'alcools par O2 / L'auteur n'a pas fourni de titre en anglais

L'objectif de cette thèse était d'évaluer des nitroxydes chiraux à squelette imidazolidin-4-one comme catalyseurs d'oxydation énantiosélective d'alcools par le dioxygène. Pour cela, plusieurs nitroxydes avec différents substituants en position α ont été synthétisés afin de voir les effets possibles de chacun des groupements. Différents co-catalyseurs décrits dans la bibliographie ont été testés afin de faire ces réactions qui ont lieu à température ambiante et pression atmosphérique. Ces systèmes catalytiques peuvent se différencier selon s'ils utilisent ou pas de métal comme additif.La stratégie utilisée pour la synthèse des catalyseurs a été développée au laboratoire à partir de travaux sur des nitrones réalisés précédemment. Grâce à cette stratégie et à l'utilisation de différentes nitrones de départ, des analogues différemment substitués en position α peuvent être synthétisés rapidement en seulement deux étapes. En particulier, l'utilisation d'une nitrone chirale nous permettra l'obtention de catalyseurs énantiopurs qui pourront être utilisés pour les réactions d'oxydation énantiosélectives.Dans la première partie de notre étude, différents systèmes catalytiques ont été testés avec nos catalyseurs sur un substrat modèle : l'alcool benzylique. Les résultats obtenus nous ont aidé à déterminer : (1) la capacité des nitroxydes de type imidazolidin-4-one à agir en tant que catalyseurs pour les réactions d'oxydation aérobie ; (2) la différence de réactivité selon la substitution en position α ; (3) les systèmes catalytiques les plus efficaces avec nos catalyseurs ; (4) le type de substrats qui pourront être envisagés lors de l'oxydation énantiosélective.C'est ainsi la désymétrisation de diols conduisant à la formation d'atropoisomères qui a été envisagée. L'oxydation par le dioxygène de trois substrats a ainsi été testée avec plusieurs catalyseurs énantiopurs et un système catalytique à base de cuivre. Les résultats d'énantiosélectivité obtenus ont été variables selon le substrat et le catalyseur utilisés. Des résultats très encourageants ont été obtenus pour l'un des substrats, avec des excès enantiomériques d'environ 60 %.En parallèle avec les différentes réactions d'oxydation, des études de voltammétrie cyclique et de résonance paramagnétique électronique (RPE) ont aussi été réalisées. La voltammétrie cyclique nous a servi surtout pour expliquer la différence de réactivité de différents analogues selon leur substitution en position α. Avec la RPE, nous avons essayé de comprendre un peu plus l'organisation autour du cuivre des différents éléments qui font partie du système catalytique à base de cuivre utilisé pour les oxydations énantiosélectives.Grâce à tous ces travaux, nous avons démontré que les nitroxydes à squelette imidazolidin-4-one peuvent être utilisés en tant que catalyseurs pour l'oxydation aérobie d'alcools. Nous avons noté l'importance des substituants en position α qui jouent un rôle important sur la stabilité de l'espèce active, ce qui pourra faire varier l'efficacité de chaque composé. Les nitroxydes contenant le squelette imidazolidin-4-one ont aussi donné des bons résultats dans des réactions d'oxydation énantioséletive de diols benzyliques. / The aim of this thesis was to evaluate the use of chiral nitroxides containing an imidazolidin-4-one squeletton as enantioselective catalysts for the aerobic oxidation of alcohols. In this view, several nitroxides with different α substituents have been synthesized to investigate the influence of these groups. Several co-catalysts reported in the literature have been tested for the oxidation reactions that take place at room temperature and atmospheric pressure. These catalytic systems can be classified relatively to the presence or the absence of metal as additive.The strategy used for the catalyst synthesis has been developped in the laboratory from previous studies about nitrones. Using this strategy and different starting nitrones, several α-substituted analogues can be rapidly synthesized in only two steps. Particularly, the use of a chiral nitrone will permit the obtention of enantiopur catalysts that will be able used for the enantioselective oxidations.In the first part of our study, several catalytic systems have been tested with our catalysts with a reference substrate: benzylic alcohol. According to the results, we have been able to determine: (1) the capacity of imidazolidin-4-one nitroxides to behave as catalysts for the aerobic oxidations; (2) the difference of reactivity depending on the α substituents; (3) the more effective catalytic systems for our catalysts; (4) the scope of the reaction in order to determine the suitable substrates for enantioselective reactions.With all these results in hand, diol desymmetrization for the synthesis of atropoisomers has been considered. Dioxygen oxidation of three diols has been tested using different enantiopur catalysts with a copper-based catalytic system. The enantioselectivity obtained proved variable depending on the substrate and the catalyst used. Very encouraging results have been obtained for one of the substrate, with enantiomeric excess of about 60 %.At the same time as the oxidation reactions, cyclic voltammetry and electron paramagnetic resonance (EPR) studies have been carried out. Cyclic voltammetry has been useful to explain the difference of reactivity of the catalysts according the α substituents. Using the EPR, we have tried to understand the organisation around copper for the different species from the catalytic system used for the enantioselective oxidations.By means of this study, we have demonstrated that imidazolidin-4-one nitroxides can be used as catalysts for the aerobic oxidation of alcohols. The α substituents play an important role in the stability of the active species that can change the efficacity of each compound. Imidazolidin-4-one nitroxides have also given encouraging results in the enantioselective oxidation of benzylic diols.

Wheat is cultivated under many different environmental conditions, presenting high grain yield capacity, nutritional quality and high degree of adaptability. In the State of Rio Grande do Sul, Brazil, approximately 5.4 million hectares are considered hydromorphic soils and are mostly used for rice farming and cattle grazing, due to its low drainage capability. Annually, only one million hectares are cultivated with rice, leaving large amounts of land without cropping or cultivated with forage crops. Therefore, there is a strong challenge for breeding programs to develop genotypes capable of tolerating this environment, with a satisfactory cost/benefit ratio for farmers. In order to suppress the energy deficit provoked by hypoxic conditions, several species redirect their metabolic pathways in order to garantee the extra ATP production resulting from an increase in fermentation rates. In those species, a group of proteins that cathalyse glycolisis and phosphate sugar metabolism such as the enzyme alcohol dehydrogenase (ADH), is selectively synthesized. The objective of this work was to verify the possibility of selecting wheat genotypes tolerant to flooding stress, in the greenhouse, by using different periods of flooding conditions alterned with draining periods. Also, to estimate the degree of conservation of the ADH1 gene in some plant species. The results indicate that it is possible to detect genetic variability for the character flooding tolerance in greenhouse. The condition of eight days of flooding revealed that the genotypes BRS 120 and BRS 208 were the most tolerant and the genotypes CD 111 and BRS 194 were the most sensitive. The molecular phylogenetic analysis obtained for the alcohol dehydrogenase (ADH1), indicates that Poaceae members present a high degree of conservation. The closest species were Saccharum oficinarum and Zea mays, presenting higher nucleotide identity. The model species (Oryza sativa) presented a higher dissimilarity regarding the remaining species of the Poaceae, suggesting that this enzyme has acquired novel functions and or mutations that were selected under the flooding condition. / Entre os cereais, o trigo (Triticum aestivum L.) é a mais importante cultura, alcançando a terceira maior produção de grãos no mundo sobrepujado apenas pelo milho e arroz. O trigo é cultivado sob diversas condições ambientais, apresentando alta capacidade de produção de grãos, qualidade nutricional e alto grau de adaptabilidade. No Estado do Rio Grande do Sul, aproximadamente 5,4 milhões de hectares são considerados solos hidromórficos e são principalmente utilizados para cultivo de arroz e pastagem, devido a sua baixa capacidade de drenagem. Anualmente, apenas um milhão de hectares está sendo utilizado para o cultivo de arroz, deixando grandes quantidades de terras para o cultivo de culturas forrageiras. Portanto, existe um forte desafio para programas de melhoramento no desenvolvimento de genótipos capazes de tolerar esse ambiente, com uma boa relação custo/benefício para os agricultores. A fim de suprimir o déficit energético provocado pelas condições de hipoxia, várias espécies redirecionam suas vias metabólicas, a fim de garantir a produção extra ATP resultantes do aumento das taxas de fermentação. Nessas espécies, um grupo de proteínas que catalisam a glicólise e metabolismo de açúcares, como a enzima álcool desidrogenase (ADH), é sintetizada seletivamente. O objetivo deste trabalho foi verificar a possibilidade de seleção de genótipos de trigo tolerantes ao estresse por alagamento, em casa de vegetação, utilizando diferentes períodos de alagamento intercalados com drenagem. Além disso, estimar o grau de conservação do gene ADH1 em algumas espécies de plantas. Os resultados indicam que é possível a detecção de variabilidade genética para o caráter tolerância ao alagamento em casa de vegetação. A condição de oito dias de alagamento revelou que os genótipos BRS 120 e BRS 208 foram as mais tolerantes e os genótipos CD 111 e BRS 194 foram as mais sensíveis. A análise filogenética molecular obtida para o álcool desidrogenase (ADH1), indica que os membros Poaceae apresentaram um elevado grau de conservação. As espécies mais próximas foram Saccharum oficinarum e Zea mays, apresentando maior identidade de nucleotídeos. A espécie modelo para plantas monocotiledôneas (Oryza sativa) apresenta uma maior dissimilaridade quanto ao restante das espécies de Poaceae, sugerindo que esta enzima tenha adquirido níveis de funções ou mutações que foram selecionadas sob a condição alagamento.

Como consequência desse fato, os custos associados ao tratamento do alcoolismo, bem como das doenças associadas a ele também têm aumentado, onerando o sistema de saúde e se tornando um problema de saúde pública de grande relevância atualmente. Os mecanismos moleculares que desencadeiam muitas dessas doenças não estão completamente esclarecidos. O tecido hepático é o mais afetado pelo etanol e as mitocôndrias têm sido apontadas como alvos cruciais na toxicidade hepática induzida pelo álcool. Logo, o objetivo desse trabalho foi investigar como o consumo de etanol afeta o estado redox e o metabolismo mitocondriais no fígado. Ratos Wistar machos adultos jovens e de meia-idade receberam ad libitum solução alcoólica 25% (v/v) como única fonte de líquido. Os grupos controle receberam somente água. Ambos os grupos receberam ração ad libitum. Mitocôndrias hepáticas foram isoladas usando técnicas padrão. O consumo de ração e de líquidos foi significativamente menor em animais que ingeriram álcool, resultando em menor ganho de massa corpórea nos protocolos utilizados. As mitocôndrias dos animais que consumiram etanol apresentaram menores níveis de respiração em condição basal e quando energizadas com substratos respiratórios. A atividade e os níveis protéicos de citocromo c oxidase foi menor nos grupos tratados com etanol. Independente da duração do período de tratamento, mitocôndrias hepáticas de animais que ingeriram álcool foram menos susceptíveis à transição de permeabilidade mitocondrial induzida por cálcio, quando comparadas às mitocôndrias dos animais do grupo controle. Esse efeito foi revertido pela adição de oxidantes de nucleotídeos de piridina (acetoacetato, diamida ou tert butil-hidroperóxido) ou em mitocôndrias desacopladas. Também houve aumento em nucleotídeos de piridina na forma reduzida e aumento na razão NAD(P)H/NAD(P)+ em mitôndrias hepáticas de ratos consumidores de etanol. Em concordância a esses dados, houve aumento na capacidade de retenção de cálcio, processo que é dependente do estado redox intramitocondrial. Por outro lado, não houve diferença na produção de espécies reativas de oxigênio entre os grupos controle e tratados com álcool. A atividade de glutationa peroxidase e as quantidades de GSH e de GSSG também não sofreram alterações. Entretanto, houve redução nos níveis de DNA mitocondrial nos tratamentos agudos, porém com tendência para retornar aos níveis normais nos tratamentos crônicos, indicando uma resposta adaptativa à injúria induzida pelo etanol. Em conjunto, nossos resultados indicam que o consumo de etanol modula o estado redox mitocondrial e de sistemas antioxidantes, prevenindo a abertura do poro de transição de permeabilidade mitocondrial. A presença desse xenobiótico no fígado também altera significativamente os níveis de NADP reduzido, agente redutor final para o sistema glutationa redutase/peroxidase que detoxifica H2O2 na matriz mitocondrial. Além disso, a resposta adaptativa ao álcool observada no DNA mitocondrial pode contribuir para compreender melhor os mecanismos envolvidos no reparo de lesões a biomoléculas e os estágios iniciais de adaptação a esse xenobiótico, etapas que precedem a morte celular, hepatite alcoólica ou carcinogênese em tecido hepático exposto cronicamente ao etanol / Abstract: The number of people suffering from alcoholism has increased significantly over the last century. As a result, costs associated with treating the addiction itself as well as the associated pathologies have also increased, such that this is considered as public health issue. Furthermore, the molecular events leading to several of these diseases are not yet clearly understood. Hepatic tissue is the most affected by alcohol, and mitochondria have been suggested to be a crucial target in alcohol-induced liver toxicity. Thus, the aim of our study was to investigate how ethanol consumption affects the redox state and mitochondrial metabolism in the liver. Young adult and middle-aged male Wistar rats were given a 25 % (v/v) ethanol solution as the only source of drinking water. Control groups received water only. Liver mitochondria were isolated using standard techniques. Food and water intake was significantly lower in alcohol-drinking rats, resulting in lower weight gain during the treatment regimes. Mitochondria from the alcohol-drinking group had lower respiration under levels in basal condition, when energized by substrates feeding electrons into complexes I and IV. Cytochrome c oxidase activity and protein levels were lower in the alcohol group as well. Additionally, regardless of the length of the treatment, liver mitochondria from the alcohol-treated animals were more resistant to Ca2+-induced mitochondrial permeability transition (MPT), when compared to mitochondria from control animals. This effect was abrogated by oxidizing agents of pyridine nucleotides (acetoacetate, diamide or tert butylhydroperoxide) or in uncoupled mitochondria. We also found that liver mitochondria from the alcohol-drinking rats had a more reduced pyridine nucleotide pool and higher NAD(P)H/NAD(P)+ ratios. In addition, Nampt (an enzyme of the NAD+ synthetic pathway) protein levels did not differ after alcohol consumption. Accordingly, the calcium retention capacity of the isolated mitochondria, which is dependent upon intramitochondrial redox state, was higher in the alcohol group. On the other hand, levels of reactive oxygen species showed no differences between the control and alcohol groups, both in mitochondria and in splenic lymphocytes. Glutathione peroxidase activity and the amounts of GSH and GSSG were also not changed. However, mitochondrial DNA levels were decreased in the short term treatments, but tended to go back up to normal levels in the chronic treatments, indicating an adaptative response to ethanol-induced injury. Together, our results indicate that ethanol consumption modulates the mitochondrial redox state and the antioxidant systems, protecting against Ca2+-induced mitochondrial pore transition permeability opening. The presence of this xenobiotic can significantly change the levels of reduced NADP, the ultimate reducing agent in the gluthatione reductase/peroxidase system that detoxifies H2O2 in the mitochondrial matrix. In addition, the adaptative response to ethanol, seen in mitochondrial DNA, may contribute to further understand the mechanisms related to lesions in biomolecules and the initial steps that preceed cell death, alcoholic hepatitis or carcinogenic process in hepatic tissue exposed chronically to ethanol / Doutorado / Biologia Estrutural, Celular, Molecular e do Desenvolvimento / Doutor em Fisiopatologia Medica

Primeiramente, faz-se uma abordagem dos fatores mais gerais que explicam a formação, expansão e consolidação da agroindústria canavieira no Estado de São Paulo. Assim, busca-se atingir o objetivo de discutir a modernização tecnológica das usinas paulistas em relação aos principais centros produtores internacionais, principalmente visando os sistemas de preparo e extração de caldo de cana. No início da década de 1970, as usinas paulistas utilizavam uma tecnologia ultrapassada em relação aos outros produtores açucareiros do mercado internacional. Diante dessa situação, a Copersucar implementou um Programa de Modernização visando a elevar a produtividade das usinas paulistas para níveis internacionais definidos pelas usinas dos países que apresentavam tecnologias mais avançadas, principalmente África do Sul, Austrália e Havaí. Isso foi possível através de um programa pelo qual a Copersucar selecionou e implantou as tecnologias mais desenvolvidas e mais apropriadas ao parque sucroalcooleiro nacional. Os estudos realizados permitem concluir que o dinamismo do setor sucroalcooleiro, implementado pela Copersucar, possibilitou a consolidação dos objetivos do Programa de Modernização das usinas de açúcar e álcool no Estado de São Paulo. Isso ocorreu devido à superação dos gargalos tecnológicos com o desenvolvimento de novas tecnologias, o que propiciou uma elevação dos níveis de produtividade das usinas paulistas aos patamares das usinas internacionais que utilizavam tecnologias mais desenvolvidas / Abstract: This work presents the technological development of the sugarcane and alcohol industrial sector in the State of Sao Paulo. It is emphasized the most important points of the technological modernization of the plants regarding the main international producers centers. The sugarcane businessman from the State Sao Paulo realized that if they did not improved the quality of the product and they continued to produce sugar with an outdated technology, they would be eliminated of the world market. Ahead of this situation, Copersucar - The Brazilian Sugarcane Research Institute implemented a Modemization Prograrn aiming to elevate the productivity of the plants from Sao Paulo into international levels defined by the plants of the countries that introduced more advanced technologies, mostly Southern Africa, Australia and Hawaii. That was possible through a Program by which Copersucar selected and implanted the most developed and appropriated technologies to the Brazilian sugarcane industrial park. The studies done allow us to the conclusion that the dynamic of the sugar cane industry, summed to the Copersucar efforts gave the consolidation of the Sugar Cane Industries Modernization Program at the State of Sao Paulo. L’objectif de la présente étude est de démontrer comment Zola, dans L’Assommoir considère l’alcool et comment le naturalisme est un outil pour faire la leçon sans esthétique, ou plutôt en ayant développé une esthétique nouvelle, dure et brute. Nous avons également essayé d’analyser en quoi l’esthétique de l’alcool épouse ou nuance le message moralisateur de l’auteur dans cette oeuvre. Nous avons mis en relief les relations de cause à effet entre l’alcoolisme et la médiocrité humaine. L’idée directrice est que si la médiocrité peut toucher tous les hommes, quelle que soit leur classe sociale, elle touche plus souvent et plus durement la classe ouvrière, les laissés pour compte, en les conduisant ou en s’ajoutant en plus à la détresse sociale, financière, et la faim. Nous avons ainsi constaté en quoi l’alcoolisme est le principal facteur déclencheur et accélérateur de cette médiocrité, qu’elle brise les rêves et enchaine à la misère. / The purpose of this study is to demonstrate how Zola in L’Assommoir considers alcohol and how naturalism is a tool to lecture without aesthetics, or rather develops a new aesthetic, hard and brutal. We have also tried to analyse how the aesthetics of alcohol marries or nuances the moralizing message of the author in this work. We have highlighted the cause and effect relationship between alcohol and human mediocrity. The guiding line is that if mediocrity can affect all men, it affects the working class more often and harder, leaving them in or adding to a state of social and financial distress, even hunger. We have seen that alcoholism is the main trigger and accelerator of this mediocrity, that it breaks dreams and binds to misery.

Développement de stratifiés de papiers imprégnés à base de résine de tannin de mimosa et d'alcool furfurylique / Development of resin impregnated papers from mimosa tannin and furfuryl alcohol resin

Les papiers imprégnés de résine ont été largement utilisés dans la fabrication de stratifiés pour des panneaux de bois. Généralement, l'imprégnation du papier implique l’utilisation de resins mélamine-formaldéhyde, urée-formaldéhyde et melamine-formaldéhyde-urée. Cependant, ces matériaux sont coûteux lorsqu'ils sont utilisés en grande quantité et ils proviennent de ressources non renouvelables. Par conséquent, dans ce travail, nous avons préparé des papiers imprégnés de résines tannins furanique pour remplacer les résines mélamine-formaldéhyde et d'urée-formaldéhyde lors de l'imprégnation des papiers. Les résines tannins furaniques sont composées de deux matières naturelles principales: a) le tannin de mimosa qui est non-toxique, respectueux de l'environnement et relativement peu coûteux qui remplace la résine synthétique et b) de l'alcool furfurylique obtenu par réduction catalytique du furfural, un composé naturel obtenu par l'hydrolyse des sucres dérivés des déchets agricoles. Les tannins sont des composés phénoliques naturels extraits des écorces des arbres et qui ont été utilisés dans l'industrie depuis de nombreuses années comme adhésifs pour la production de panneaux de bois. Mais il n’y a pas jamais eu jusqu’à présent des recherches sur leurs utilisations dans la fabrication des stratifiés. Dans ce travail, nous avons étudié : 1) Le temps de gel entre des tannins de mimosa et des tannins de pin avec de l'alcool furfurylique, sans ajout de formaldéhyde de pH 1 à pH 11 2) L’analyse MALDI-TOF et RMN des tannins de mimosa avec de l'alcool furfurylique 3) La qualité de la surface du contreplaqué recouvert d’un papier imprégné de résine tannins de mimosa-alcool furfurylique 4) Les stratifiés de 10 plis à haute pression (HPL) ont été préparés avec de la résine tannins de mimosa-alcool furfurylique. Generally, paper impregnation involves melamine formaldehyde, melamine urea formaldehyde and urea formaldehyde resin. However, these materials are expensive when used in high quantity and derived from non renewable resources. Therefore, in this work we prepared tannin furanic resin impregnated paper to replace melamine formaldehyde and urea formaldehyde in the paper impregnation. Tannin furanic resin composed of two main natural materials: a) mimosa tannins which is non-toxic, environmentally friendly and relatively inexpensive to substitute the synthetic resin and b) furfuryl alcohol obtained by catalytic reduction of furfural, a natural compound obtained by the hydrolysis of sugars derived from agricultural waste. Tannins are naturally occurring phenolic compounds extracted from the bark trees that have been used industrially for many years as wood panel adhesives but no distance work have been done to study their potential in paper laminates. Les biocapteurs sont des moyens d'analyse en plein essor à la fois rapides, sélectifs et peu coûteux applicables à des domaines extrêmement variés (environnement, santé, agroalimentaire,…). Dans ce type d'outil, un élément sensible de nature biologique (anticorps, enzyme, microorganisme, ADN…) doté d'un pouvoir de reconnaissance pour un analyte ou un groupe d'analytes est associé à un transducteur pouvant être de type électrochimique, optique ou thermique. Dans ce travail, nous nous sommes intéressés au développement de trois biocapteurs utilisant des enzymes de la famille des oxydo-réductases (alcool oxydase, lactate oxydase et formaldéhyde déshydrogénase) pour la détection d'analytes d'intérêt dans le domaine de l'agroalimentaire ou de l'environnement (alcools, lactate et formaldéhyde). Les trois enzymes ont été immobilisées sur des microélectrodes interdigitées en or en vue d'une détection conductimétrique. Nous avons cherché à mieux comprendre le fonctionnement des biocapteurs et optimisé le procédé d'immobilisation des enzymes ainsi que différents paramètres de fabrication ou de mesure afin de maximiser les performances analytiques des outils développés (sélectivité, limite de détection, reproductibilité, stabilité). In this work, we developed biosensors based on three oxidoreductases (alcohol oxidase, lactate oxidase and formaldehyde dehydrogenase) for primary alcohols, lactate and formaldehyde analysis. The three enzymes were immobilized on interdigitated gold microelectrodes in view of conductometric detection. We tried to achieve a better understanding of biosensors processing and optimized enzyme immobilization as well as several fabrication or operational parameters to maximize analytical performances (selectivity, detection limit, reproducibility and stability). Les agents d'embolisation liquides utilisés dans le traitement endovasculaire ont de nombreuses limitations. Des polymères, tels que l’Onyx et les cyanoacrylates sont disponibles. L’alcool est un agent puissant, mais non radio-opaque. Les cyanoacrylates entrainent une réaction inflammatoire significative. Leur polymérisation rapide est responsable d’un comportement mal prévisible.Il existe une possibilité de traitement incomplet.L’Onyx est efficace pour le traitement des MAV.Une pénétration plus distale est obtenue avec l’alcool, mais associée à un risque de migration systémique. Nous avons évalué les propriétés d'un nouvel agent embolique (Alconyx) composé d'un mélange d'alcool et d'Onyx. Cet agent devrait cumuler les avantages respectifs de l'alcool et de l'onyx avec une visualisation adéquate sous fluoroscopie, une meilleure pénétration distale que l'Onyx seul et moins de toxicité systémique liée à la réduction de quantité d’alcool injecté. Divers mélanges ont été testés avec différentes concentrations d'Onyx 18 et d’alcool absolu. Alconyx 25 (75% Onyx 18; 25% d'éthanol) est la formulation la plus prometteuse. Nous avons démontré sa facilité d'injection in vivo, sa nature cohésive sans fragmentation ainsi que sa bonne visualisation sous fluoroscopie. En raison de sa moindre viscosité démontrée in vitro, Alconyx a été capable de pénétrer profondément dans le lit artériel.. L'occlusion proximale par Alconyx 25 devrait permettre d'améliorer le contact entre l'éthanol et la paroi vasculaire et donc augmenter son pouvoir sclérosant et limiter son passage systémique. Les propriétés occlusives d’Alconyx 25 sont similaires à celle de l’Onyx 18 sous haute pression in vitro. / Commercially available liquid embolization agents used in endovascular treatment have many limitations. Polymeric agents as Onyx and cyanoacrylate are available. Ethanol also is a potent sclero-embolic agent. Cyanoacrylates are effective liquid embolic agents, however, their rapid polymerization makes their behaviour unpredictable with possibility of incomplete treatment. These properties render their use challenging.Onyx is easy to use. However, in very small arterial niduses, Onyx, is unable to penetrate deeply. Deep penetration is obtained with ethanol, associated with risk of systemic migration.Poor visualization of ethanol under fluoroscopy is major drawback. Mixing Onyx with ethanol had never been described in the literature till now. In this work, various mixtures have been tested with different concentrations of Onyx 18 and absolute ethanol. Alconyx 25 (75% Onyx 18; 25% ethanol) seems to be a promising product. We proved its ease of injection in vivo and in vitro, its cohesive nature showing no fragmentation or interruption of the injected column as well as its good visualization under fluoroscopy. It was able to penetrate deeply in the arterial bed. The occlusive properties of Alconyx 25 were rated as good as Onyx 18 under high pressure in vitro. Further investigation is needed to better understand the behavior of ethanol in the suspension and its effect on tissues compared to Onyx diluted simply with an equivalent amount of DMSO. Studies on other commercially available concentrations of Onyx would certainly be interesting.

Comportement "in vitro" et "in vivo" de verres composites poreux : assimilation osseuse, explorations physiologiques et physico-chimiques / Behavior "in vitro" and "in vivo" of porous composite glasses : bone assimilation, physiological and physicochemical explorations

L'application des biomatériaux est de plus en plus élargie. Le progrès médical suggère l'utilisation des biomatériaux (verres bioactifs, apatites,..) en tant qu'implants selon le besoin de l'organisme. L'objectif de notre travail est de mettre en évidence l'influence biologique des molécules organiques (bisphosphonates, biopolymères et antibiotiques) incorporés dans des matrices de verres bioactifs. De même, notre étude vise à optimiser les meilleures techniques de synthèse et d'association des verres bioactifs à ces molécules. La détoxification des rats mâles de souche « Wistar » exposés au chlorure de nickel par une apatite synthétique a aussi fait l'objet de ce travail. Suite à une perte osseuse provoquée, nous avons démontré que l'utilisation des antibiotiques associés à des verres bioactifs en tant qu'implants osseux, chez des rattes ovariectomisées, permet d'éliminer certains effets indésirables par voie systémique. Ceci a été mis en évidence par l'évaluation des paramètres biochimiques et histologiques du foie et du rein. Aucune variation significative en comparaison avec ceux du témoin négatif n'a été révélée. L'étude in vitro a montré d'une part que l'introduction du Chitosan et surtout de l'antibiotique dans la matrice vitreuse font augmenter l'activité antibactérienne in vitro. Cette étude in vitro a montré d'autre part que la Ciprofloxacine induit un effet néfaste sur les cellules ostéoblastiques et endothéliales. Cet effet est local lorsqu'il s'agit des expérimentations in vivo. Ceci est mis en évidence lors des évaluations du statut oxydant. Les marqueurs du remodelage osseux, l'histologie de l'os et les paramètres physico-chimiques montrent l'effet retardateur de cet antibiotique sur la dissolution de l'implant et par conséquent sur son ossification. La synthèse par le procédé de sol-gel provoque une bioactivité plus importante que celle obtenue par fusion. La bioactivité des verres bioactifs étudiés diffère selon la molécule introduite. Celle-ci est réduite dans le cas de l'association du Clodronate et de Ciprofloxacine in vitro et in vivo. Alors que, le Polyvinyl Alcohol et surtout le Chitosan font modifier la cinétique de cette bioactivité in vivo. Concernant l'hydroxyapatite, nous avons essayé d'explorer son effet détoxifiant chez des rats reçevant le chlorure de nickel. Nos résultats ont montré que le nickel induit un stress oxydant au niveau du foie, du rein, de la rate et du culot érythrocytaire. Des troubles physiologiques ont été observés chez les rats exposés au nickel. Cependant, l'implantation de l'hydroxyapatite protège les rats intoxiqués par le nickel contre ses effets toxiques en diminuant l'état du stress. Le biomatériau utilisé s'avère efficace pour corriger l'équilibre ferrique et phosphocalcique, protéger les fonctions rénale et hépatique, abaisser le taux du nickel osseux et corriger l'anémie. / The application of biomaterials is increasingly widened. Medical progress suggest the use of biomaterials (bioactive glasses, apatites,..) as implants according to the need of the body. The aim of our work is to highlight the biological influence of organic molecules (bisphosphonates, biopolymers and antibiotics) incorporated into matrix of bioactive glasses. Similarly, our study aims to optimize the best synthesis and combination technique of bioactive glasses to these molecules. The detoxification of male rats strain "Wistar" exposed to nickel chloride by a synthetic apatite also has been the object of this work. Following the bone loss induced, we have demonstrated that the use of antibiotics associated with bioactive glass as bone implants, in ovariectomised rats, eliminates some adverse effects systemic. This has been highlighted by the evaluation of biochemical and histological parameters of liver and kidney. Any significant changes in comparison with those of the negative control was revealed. The in vitro study showed in the one hand that the introduction of Chitosan and especially of the antibiotic in the glass matrix can increase antibacterial activity. This in vitro study showed in the other hand that the Ciprofloxacin induces a negative effect on osteoblastic and endothelial cells. This effect is local when it has been an in vivo experiments. This is highlighted by the oxidative status evaluation. Markers of bone turnover, bone histology and physicochemical parameters show the retarding effect of this antibiotic on the dissolution of the implant and consequently on its bone formation. Synthesis by sol-gel method causes a more important bioactivity than melting. The bioactivity of elaborated bioactives glasses will differ depending on the molecule introduced. It is reduced in the case of combination of Clodronate and Ciprofloxacin in vitro and in vivo. While, Polyvinyl Alcohol and especially Chitosan modify the kinetic of the bioactivity in vivo. Concerning the hydroxyapatite, we tried to explore its detoxifying effect in rats receiving nickel chloride. Our results showed that nickel induces an oxidative stress in the liver, kidney, spleen and red cell pellet. Physiological disorders were observed in rats exposed to nickel. However, implantation of hydroxyapatite protects rats intoxicated by nickel against its toxic effects by decreasing the stress status. The used biomaterial is effective to correct ferric phosphate balance, protect kidney and liver function, reduce level of bone nickel and correct anemia.

L'alcool est une des principales causes de maladie du foie en Europe avec peu d'options thérapeutiques. Parmi les consommateurs d'alcool, seul certains patients vont évoluer vers des formes sévères d'atteinte hépatique et le microbiote intestinal a été identifié comme cofacteur de cette susceptibilité individuelle. Il interagit avec le foie notamment via la production de métabolites bactériens tels que les acides biliaires secondaires.L'objectif de ce travail a été dans un premier temps d'analyser le microbiote intestinal, au sein d’une cohorte de patients alcooliques à différents stades de la maladie alcoolique du foie, et d’étudier sa relation avec les acides biliaires. Dans un deuxième temps, ce projet avait pour but d’étudier s’il était possible d'améliorer l'atteinte hépatique liée à l'alcool, en modulant le microbiote intestinal, dans un modèle animal de maladie alcoolique du foie.Les patients avec une atteinte hépatique sévère liée à l'alcool ont un profil d’acides biliaires plus hydrophobe, et donc plus toxique, associé à une dysbiose et à des modifications des fonctions bactériennes. Ces modifications des fonctions bactériennes participent à la gravité de la maladie. La modulation du microbiote par la pectine, un prébiotique, permet de prévenir mais également de faire régresser les lésions hépatiques chez la souris. Les effets protecteurs de la pectine sont corrélés avec le métabolisme du tryptophane.Ce travail de thèse montre que le microbiote intestinal et ses métabolites sont une cible thérapeutique potentielle dans la maladie alcoolique du foie. Il ouvre la porte à la réalisation d'essais cliniques chez l'homme dans lesquels nous pourrions limiter la progression des lésions hépatiques liées à l'alcool en contrôlant le microbiote intestinal grâce à l’utilisation de la pectine ou du métabolisme du tryptophane. / Alcohol is one of the main causes of alcoholic liver disease in Europe with few therapeutic options. Among alcohol consumers, only a part of these patients will develop severe liver lesions. This individual susceptibility is driven by intestinal microbiota. Intestinal microbiota interacts with the liver through the production of bacterial metabolites including the secondary bile acids.The aim of my project, was firstly to study the relationship between the intestinal microbiota and the bile acids composition depending on the severity of alcoholic liver disease in a cohort of alcoholic patients. Secondly, I assessed the improvement of alcohol induced liver lesions by changing the intestinal microbiota in a mouse model of alcoholic liver disease.Patients with a severe form of alcoholic liver disease display a higher hydrophobic bile acid pool, more toxic, associated with a specific dysbiosis and changes in the bacterial functions. Changing the intestinal microbiota by using pectin, a prebiotic, prevents and reverts alcohol induced liver injury in mice. These protective effects of pectin involve changes in the tryptophan metabolism.In conclusion, these studies highlight that the intestinal microbiota and its metabolites are potential therapeutic targets for alcoholic liver disease. Moreover, pectin as an alimentary product could be proposed in the management of alcoholic liver disease in humans.
