Vu l’importance des molécules fluorées dans différents domaines et vu la réactivité engendrée par le 4,4,4-trifluorobut-2-ynoate d’éthyle 1 lors des travaux antérieurs réalisés au laboratoire, nous avons décidé d’étudier le comportement de dérivé fluoré 1 vis-à-vis d’hydrure tributylétain sans l’emploi de catalyseurs ou d’additifs. Le but est d’accéder à de nouveaux synthons fluorés par voies originales.Après une étude préliminaire pour effectuer cette réaction d’hydrostannation dans les meilleures conditions, nous avons réalisé pour la première fois la synthèse totalement régio- et stéréosélective des vinylétains (Z)-2α et (Z)-2β par un simple choix de solvant et sans l’utilisation d’aucun additif. Ces résultats inédits et très encourageants, nous ont conduis à étendre cette stratégie à d’autres hydrures comme l’hydrure de diphénylphosphine et l’hydrure de triphénylgermane..Les vinylétains (Z)-2α et (Z)-2β- de configurations bien définies sont des réactifs de choix pour élaborer de nouvelles structures fonctionnelles trifluorométhylées. En effet, le couplage cupro-catalysé de ces vinylétains en présence d’une quantité catalytique de CuI (10 moles%) avec une variété de bromures d’allyles, de propargyles, d’aryles, de benzyles et d’alcynyles conduisent à une grande variété de composés fluorés jamais décrits dans la littérature.Les méthodes de synthèse développées dans ce travail sont originales et permettent la synthèse d’une grande famille de composés fluorés avec un moindre coût. / The development of a simple method to obtain perfluoroalkylated building blocks for their subsequent utilization in the synthesis of Rf-containing compounds is therefore essential to organofluorine chemistry. Perfluoroalkylated vinyl metals constitute an important class of these building blocks.In order to prepare a new perfluoroalkylated bulding blocks, we investigated transition metal-catalyzed-free hydrostannylation of ethyl 4,4,4-trifluorobut-2-ynoate 1. The hydrostannylation took place smoothly in the absence of additive, providing regioselectively high yields of the corresponding α or β stannylated alkenoates depending on the nature of the solvent used. Indeed, we have demonstrated that the hydrostannylation of 1 in hexane provided the β-stannylated product with high regioselectivity (>95%) and excellent yield (>97%). Using methanol as solvent, total α-regioselectivity of the hydrostannylation of 1 was observed, providing α -tributylstannylacrylate as the sole regioisomer in a nearly quantitative yield.Theses new vinyltins reagents readily undergo copper (I) catalyst coupling reactions with various electrophiles as allyl, propargyl, benzyl and alkynyl bromides to provide good yields of the new corresponding acrylates esters bearing a β-trifluoromethyl group.This method provided a new efficient entry to this important class of compounds

La chimie des composés à base d'iode hypervalent (i.e., iodanes) connaît un fort intérêt depuis le début des années 1990 comme en témoigne le panel de composés iodés disponibles commercialement et le nombre de transformations chimiques que ces réactifs peuvent promouvoir. Au cours de la dernière décennie,les recherches se sont essentiellement concentrées sur le développement d’iodanes chiraux et leurs applications en synthèse asymétrique. La plupart de ces iodanes sont utilisés pour la création hautement sélective de liaison carbone-hétéroatome (C–O, C–N, C–F…). La création asymétrique de liaison carbone-carbone (C–C) médiée par des iodanes chiraux a cependant été très peu étudiée jusqu'à présent. Dans ce contexte, notre groupe de recherche a décrit la synthèse de nouveaux iodanes-λ5chiraux biaryliques et de type Salen ainsi que leurs applications à la désaromatisation oxygénante asymétrique de 2-alkyl phénols avec des excès énantiomériques (ee) supérieurs à 90%. Ces travaux de thèse sont consacrés au développement de nouveaux iodanes-λ3 chiraux porteurs de ligands carbonés transférables pour la création asymétrique sans métal de liaison C–C. Sont plus particulièrement décrits la synthèse de bis(alcynyl-iodanes-λ3) biphényliques atropopurs et leur utilisation dans l’alcynylation énantiosélective de β-cétoesters (jusqu’à 68% ee) et, pour la première fois, de naphtols (jusqu’à 84% ee). / The chemistry of hypervalent organoiodine compounds (i.e., iodanes) has unarguably experienced an impressive development since the early 1990s, as evidenced by both the diversity of iodane reagents that are commercially available today and the number of chemical transformations that those reagents can promote.Over the last decade, the elaboration of new chiral iodanes and their applications in asymmetric synthesis have attracted special interests and research efforts. Most ofthem are involved in highly selective carbon-heteroatom bond-forming reactions (C–O, C–N, C–F…). However, C–C bond-forming reactions in a stereoselective fashion with chiral iodane are yet underexploited. In this context, our research group reported the synthesis of new biarylic and Salen-type chiral λ5‐iodanes and their successful application to the asymmetric hydroxylative phenol dearomatization of 2alkylphenols, with enantiomeric excesses (ee) above 90%. This thesis work is dedicated to the development of new chiral λ3‐iodanes bearing transferable carbon-basedligands for metal‐free asymmetric C–C bond construction. In particular, the synthesis of chiral biphenylic bis(alkynyl-λ3-iodanes) and their successful application to the enantioselective alkynylation of β‐ketoesters (up to 68% ee) and 2‐substituted naphthols (up to 84% ee) are described.

Le projet de recherche est centré sur le développement de nouvelles transformations catalysées par de l'or en chimie des ions N-acyliminiums. L'objectif de la première partie de ce projet est de développer une séquence 5-exo-dig intramoléculaire hydroalkoxylation/aza-Ferrier-Petasis réarrangement catalysée par l’or. Une deuxième partie du projet est consacrée à l'élaboration de la première alcynylation catalytique d'ions cycliques N-acyliminium utilisant N,O-acétals. Ce type de réaction est potentiellement une transformation importante, donnant naissance à des dérivés d'amines propargyliques. Ces derniers peuvent faire l’objet d’autres réactions conduisant à des produits naturels azotés ainsi qu’à certains analogues. Compte tenu de nos objectifs, l’intérêt principal est l'utilisation d’alkynes TMS en accord avec des complexes d'or qui ont été coordines avec des contre-ions faiblement nucléophiles. Pour élargir le champ de la catalyse basée sur l'or, il est important d'étendre la gamme de substrats et les groupes fonctionnels qui peuvent être activées par des complexes d'or. Une dernière partie est consacrée à la catalyse séquentielle qui favorise des processus catalytiques à plusieurs étapes. De cette manière, deux transformations catalytiques et fondamentalement différentes sont réalisées dans un même récipient. Ce travail nous a permis l’accès rapide et efficace à une famille de composes à structure complexe avec bons rendements. Par conséquent, il s'agit d'un sujet innovant en synthèse organique moderne qui a permis d’ouvrir la voie à de nouveaux projets. / The research project is centred on the development of new gold catalyzed transformations in N-acyliminium ion chemistry. The objective of our first part of the project is to develop an enantioselective gold-catalyzed 5 exo-dig intramolecular hydroalkoxylation/aza-Ferrier-Petasis rearrangement sequence. This sequence could give an asymmetric and atom economic expeditious access to the structure of hydropyrrolizidines, which are known to be potential biologically active compounds. A second part of the project is devoted to the development of the first catalytic alkynylation of cyclic N-acyliminium ions using N,O-acetals. This type of reaction is potentially an important transformation, giving rise to propargylic amine derivatives amenable to further interesting synthetic manipulations en route to nitrogen-containing natural products and some analogs. Considering our objectives, key features is the use of TMS alkynes in conjunction with gold complexes that have been paired with poorly nucleophilic counter-ions. To broaden the scope of catalysis based on gold, it is important to extend the range of substrates and functional groups that can be activated by gold complexes. A final section is devoted to the sequential catalysis which designs the promotion of catalytic multistep processes. In this way, two fundamentally distinct chemical transformations are catalytically promoted in a single flask. This research area allows the rapid and efficient reach of complex molecular frameworks with improved yields and resource efficiency. Therefore, this is an exciting theme in modern organic synthesis that has recently stimulated the report of numerous excellent contributions.

Na primeira parte da tese foram abordadas diversas rotas sintéticas para a preparação dos fragmentos nordeste e sudoeste do alcaloide bisindólico Raputindol D, cuja síntese total nunca foi descrita. A proposta inicial era obter o fragmento nordeste em 13 etapas a partir do composto comercial 3-metil-4-nitrofenol, utilizando como etapas-chave uma reação de Diels-Alder, uma abertura redutiva de anel e uma contração de anel com iodo(III). Empregando uma reação de Diels-Alder regiosseletiva de um intermediário silil substituído, a construção de uma unidade tricíclica linear foi alcançada com a obtenção de um único regioisômero. Entretanto, todas as tentativas de abertura de anel do alqueno oxabicíclico resultaram apenas no regioisômero não desejado, apesar dos estudos prévios com compostos modelo terem revelado que essa proposta era viável. Assim, foi possível acessar um intermediário avançado em 13 etapas e 12% de rendimento global. O fragmento sudoeste foi obtido em 3 etapas a partir do 5-bromoindol comercial, em rendimento global de 47% e empregando como etapas principais uma reação de Sonogashira e uma reação de acoplamento com 1-hidróxibenziodoxolone. Na segunda parte da tese são apresentados os resultados referentes ao estudo da etapa-chave para a conexão dos fragmentos nordeste e sudoeste, através do desenvolvimento de uma nova metodologia de α-alquinilação eletrofílica de compostos carbonílicos aromáticos não-ativados com o iodo hipervalente TMS-EBX. Empregando tBuOK como base e TBAF como agente ativante, cetonas mono- e dialquiniladas foram obtidas em ótimos rendimentos. A utilização de aldeídos como substratos também se mostrou viável, o que permitiu acessar derivados de álcoois homopropargílicos em rendimentos moderados após a redução dos produtos com NaBH4 in situ. Finalmente, a aplicação da metodologia desenvolvida foi demonstrada através da preparação de um intermediário de alquinilação avançado. A atividade antiproliferativa desse composto foi investigada, mostrando-se apenas fracamente ativo com uma atividade mais pronunciada para células de carcinoma de ovário e leucemia. / In the first part of the thesis several synthetic routes for the preparation of the northeast and southwest fragments of the bisindolic alkaloid Raputindole D, whose total synthesis has never been described, were approached. The initial proposal was to obtain the northeast fragment in 13 steps from the commercial compound 3-methyl-4-nitrophenol, using as key steps a Diels-Alder reaction, a reductive ring opening and a ring contraction with iodine(III). Employing a regioselective Diels-Alder reaction of a silyl substituted intermediate, the construction of a linear tricyclic unit was achieved with the obtainment of a single regioisomer. However, all attempts to ring opening of the oxabicyclic alkene only resulted in the undesired regioisomer, although previous studies with model compounds revealed that this proposal was feasible. Thus, it was possible to access an advanced intermediary in 13 steps and 12% overall yield. The southwest fragment was obtained in 3 steps from commercial 5- bromoindole in 47% overall yield and employing as main steps a Sonogashira reaction and a coupling with 1-hydroxybenziodoxolone. In the second part of the thesis are presented the results regarding the study of the key step for the connection of the northeast and southwest fragments, through the development of a new methodology for the electrophilic α-alkynylation of non-activated aromatic carbonyl compounds with the hypervalent iodine TMS- EBX. Employing t-BuOK as a base and TBAF as an activating agent, mono- and dialkynylated ketones were obtained in good yields. The use of aldehydes as substrates also proved to be possible, allowing to access homopropargylic alcohols derivatives in moderate yields after reduction in situ using NaBH4. Finally, the application of the developed methodology was demonstrated by the preparation of an advanced alkynylation intermediate. The antiproliferative activity of this compound was investigated, showing only weakly active with a more pronounced activity for ovarian carcinoma and leukemia cells.

The synthesis of novel profluorescent nitroxide probes

A number of novel isoindoline nitroxides have been synthesised using a variety of synthetic techniques. Several carbon-carbon bond forming methodologies, including the first examples of Heck and Sonogashira coupling applied to the isoindoline nitroxide class, were utilised to give novel robust aromatic frameworks. Palladium-catalysed Heck coupling of brominated nitroxides and ester-substituted olefins generates novel nitroxides possessing extended conjugation. Hydrolysis of the nitroxide esters gave the corresponding carboxylic acids, which showed enhanced water solubility. Sonogashira coupling of an iodo-isoindoline nitroxide gave several novel alkynesubstituted nitroxides in high yield. Subsequent coupling of a deprotected ethynyl nitroxide with aromatic iodides gave acetylene-linked nitroxides and an acetylene linked nitroxide dimer. A butadiyne linked dinitroxide was successfully synthesised via Eglinton oxidative coupling of two ethynyl nitroxides. The synthesis of a novel water-soluble dicarboxy nitroxide was achieved by base hydrolysis of a dinitrile. Functional group interconversion furnished anhydride and imide substituted nitroxides from the diacid. Subjecting the imide to the Hofmann rearrangement gave an unexpected brominated amino-carboxy nitroxide. The dicarboxy nitroxide and the brominated amino-carboxy nitroxide were both shown to have a protective effect on Ataxia-Telangiectasia cells, indicating a possible role as antioxidants in the treatment of this disease. A fluorescein nitroxide was successfully synthesised through the condensation of the anhydride substituted nitroxide and resorcinol. After limited success using a variety of other techniques, Buchwald-Hartwig amination was able to furnish a rhodamine nitroxide, via a triflate-fluorescein nitroxide. The extended aromatic nitroxides possess suppressed fluorescence and we have described these systems as profluorescent. The profluorescent nitroxides were found to have significantly lower quantum yields than the non-radical analogues and displayed a substantial increase in fluorescence intensity upon radical trapping, making them useful probes for free radical reactions.

Vers la synthèse totale de la Thapsigargine inhibiteur de l’enzyme SERCA / Toward the total synthesis of thapsigargin, an inhibitor of SERCA enzyme

La thapsigargine (Tg) est une lactone sesquiterpène polyoxygénée appartenant à la famille des guaianolides, isolée de Thapsia (Apiacae), une plante poussant communément dans le basin méditerranéen. Son intérêt dans le traitement du cancer de la prostate est basé sur son potentiel inhibiteur de l’enzyme endo/sarcoplasmique calcium ATPase (SERCA). Ce phénomène conduit à une augmentation de la concentration du calcium dans le lumen du réticulum endoplasmique et engendre une apoptose cellulaire. Dans un premier temps, un modèle 8-desoxy-bicyclo[5.3.0]decadiénone, de structure proche du squelette de la Tg, a été synthétisé par le biais d’une réaction clé de Pauson-Khand allène-yne catalysée par un complexe de rhodium. L’approche directe développée est robuste avec de hautes sélectivités et rendements. Cette voie a ensuite été appliquée à la synthèse du produit naturel. A partir d’un époxyde optiquement enrichi, la partie Sud de la Tg (le motif lactonique) est tout d’abord mise en place. Les centres C6 et C8 sont ensuite construits respectivement par alcynylation/réduction asymétrique et par propargylation énantiosélective. Cette première approche a permis d’isoler un produit énantiopure possédant les centres chiraux C6, C7 et C8 ainsi que du motif lactonique en dix-sept étapes. Une seconde voie a été initiée permettant la mise en place plus rapide de la partie Sud de la molécule incluant les centres asymétriques C6, C7, C8 et C11 par le biais d’une réaction de dihydroxylation. Cette seconde voie a permis d’isoler un produit racémique possédant les centres chiraux C6, C7, C8 et C11 ainsi que du motif lactonique en dix étapes. Parallèlement, une étude méthodologique a été menée sur la réaction de Pauson-Khand allénol-yne intramoléculaire. Treize composés bicycliques ont été synthétisés en série acétal et NTs. / Thapsigargin (Tg) is a highly oxygenated sesquiterpene lactone belonging to the guaianolide family, isolated from the Mediterranean plant species Thapsia (Apiaceae). Its interest towards treatment of prostate cancer is based on the potency of this compound as an inhibitor of the endo/sarcoplasmic calcium ATPase (SERCA) inducing an increase in cytosolic calcium concentration and leading to apoptotic cell death. A straightforward approach to a highly functionalized 8-desoxy-bicyclo[5.3.0]decadienone model close to the Tg framework has been achieved through a key Rh(I) allenic Pauson-Khand reaction (APKR). The synthetic route developed therein is robust and the yields and selectivities are high. This approach was used in the synthesis of the natural compound. Starting from the same chiral epoxide, the bottom portion of the Tg (lactone core) is first built, then C6 and C8 carbons are functionalized respectively by asymmetric reduction and enantioselective propargylation. This first way allowed to synthesize a seventeen steps-enantiopure product with C6, C7, C8 chiral centers and the lactone core. A second way is performed to set up faster the bottom part of the molecule included C6, C7, C8 and C11 asymmetric centers through a dihydroxylation step. This second approach allowed to synthesize a ten steps-racemic product with C6, C7, C8, C11 chiral centers and the lactone core. Meanwhile, a methodological study was conducted on the intramolecular allenol-yne Pauson-Khand reaction. Thirteen bicyclic compounds were synthesized in acetal and NTs series.
