Protein partition and digesta flow in lactating Holsteins fed 2:1 and 1:2 soybean meal:fish meal

Attempts to improve upon the crude protein feeding system have strived to characterize feedstuffs with respect to ruminal protein degradability. In vitro an in situ procedures fall short of this goal by not accounting for ruminal turnover. Six lactating cows, fitted with ruminal and duodenal cannulae, were utilized for in vivo determination of protein degradability by employing a double-marker system. Treatments were corn silage-based diets supplemented with 2:1 soybean meal:fish meal nitrogen (SF) or 1:2 soybean meal:fish meal nitrogen (FS) at levels of 15. 9 and 16.0 percent protein. Ruminal cobalt ethylenediaminetetraacetic acid (CoEDTA) infusions marked liquid phase (LP) digesta and Yb-soaked hay was used as a solid phase (SP) marker. Duodenal digesta was separated into SP and LP at 3000xg. Least squares means of LP flows did not differ for total, precipitable protein, or microbial nitrogen, for SF and FS. Total solid phase flow of dry matter was higher for FS (9.06 kg/d) than for SF (7.97 kg/d), although intakes did not differ. Milk composition and yield did not differ for FS and SF. Average daily gain was 0.12 kg/d for SF and 1:26 kg/d for FS. Whole-tract digestibilties were not different although rumen digestibility of dry matter was 24.9% for FS and 35.7% for SF, and duodenal recovery of N was 93.8% for SF and 107.5% for FS. Inclusion of a higher level of fish meal in the diet increased the proportion of feed proteins delivered to the small intestine, increasing weight gain while having no effect on milk production. / Master of Science

<b>Production and Glucose Metabolism Responses Related to Late Gestational Muscle Reserves and Supplementation of Branched-Chain Volatile Fatty Acids in Transition Dairy Cattle</b>

<p dir="ltr">The periparturient period involves coordinated physiological adaptations as the dairy cow transitions from a non-lactating to lactating state. The ability of dairy cattle to adapt to the onset of lactation is impacted both by physiological and nutritional factors, and a poor transition can result in reduced productivity and welfare for the animal. Additionally, disease and disorder development are heightened in the transition period, with increased risk for involuntary culling occurring in early lactation. This study aimed to evaluate if the amount of late gestational muscle reserves and prepartum supplementation of branched-chain volatile fatty acids (BCVFA) impacts health and production parameters in multiparous, periparturient dairy cattle. Forty-eight multiparous Holstein dairy cattle were assigned to either a high or low muscle group (HM or LM, respectively) based on their <i>longissimus dorsi</i><i> </i>muscle depth 42 days before expected (BEC). After assignment to group, cattle were then randomly assigned to a control (CON; 80 g/d soyhull pellets as-fed basis) or BCVFA (40 g/d isobutyrate product, 20 g/d isovalerate product, 20 g/d 2-methybutyrate product, fed as calcium salt products on an as-fed basis) treatment, which was top-dressed in the prepartum period only. After parturition, treatment was no longer provided and cattle were fed a common lactating diet. Blood samples, ultrasound images, and feed intake were collected and recorded from 42 BEC through 28 days in milk (DIM), milk yield and composition data was collected from parturition until 28 DIM.</p><p dir="ltr">HM cattle began mobilizing muscle reserves prior to parturition, while LM cattle began to accrete muscle reserves prior to parturition. This difference in prepartum muscle utilization did not impact other body measurements (i.e. body weight or body condition score) between the groups of cows but did result in increased blood glucose concentrations prepartum for HM cows compared with LM. This increase in glucose concentrations is likely due to the increased supply of gluconeogenic precursors as a result of the degradation of muscle tissue. The difference in glucose concentration was not observed postpartum, neither was there a difference in tissue mobilization between the groups postpartum. HM cattle had greater DMI both pre- and postpartum, and produced greater yields of milk, milk fat, milk protein, and milk lactose postpartum compared to the LM group. Despite the increased milk yield, there was no difference in feed efficiency between the groups, as the HM cows consumed more feed. Prepartum supplementation of BCVFA did not impact body measurement changes throughout the entire transition period, but did increase pre- and postpartum DMI, likely due to increased fiber digestibility. The BCVFA treatment increased blood glucose concentrations both pre- and postpartum and reduced milk urea nitrogen concentrations postpartum, likely due to improved nitrogen efficiency. Results show that prepartum supplementation of BCVFA has an improved ruminal carryover effect into early lactation.</p><p dir="ltr">At 14 days BEC and 7 DIM, an intravenous glucose tolerance test (IVGTT) was performed on a sub-set of cows, to evaluate if insulin response could be a mechanism impacting the efficiency and production differences observed between muscle groups and BCVFA supplementation. BCVFA supplementation increased glucose area under the curve in the prepartum period only. No other differences were observed between muscle group or treatment in either the pre- or postpartum period. Because there were no major differences between the cows in response to an IVGTT, we cannot conclude that glucose metabolism is a mechanism to explain differences in production responses observed. IVGTT cannot measure peripheral tissue insulin sensitivity, which is a limitation of this assessment, so our conclusions cannot assess if muscle reserves or BCVFA treatment impact peripheral tissue insulin sensitivity response. These results highlight that the amount of muscle plays a key role in the production responses observed in early lactation and that providing a BCVFA supplement could increase DMI during a period of negative nutrient balance and improve rumen efficiency.</p>

Degradation kinetics of carbohydrate fraction of commercial concentrate feeds for weaned calves, heifers, lactating and dry dairy cattle / Variations in composition and disappearance of nutrients in dairy cattle feeds are dictated by ingredients, methods of processing, storage while milk production levels depend on the animal, environmental factors and largely on pools of available carbohydrates, proteins, vitamins and minerals in the concentrate feeds. There is a wide variety of concentrates for dairy cattle on the formal and informal markets and dairy farmers need to be astute in selecting feeds appropriate for specific production periods and animals to sustain their businesses. Composition of nutrients displayed on concentrate containers is however inadequate for in-depth assessment of products. This study determined nutrient composition, rumen dry matter disappearance and microbial colonization on residual substrate on commercial concentrate feeds and simulated total mixed rations for dairy calves, heifers, lactating and dry cows based on common feeding guidelines. Equivalent feeds for each herd group were obtained from three suppliers in the formal markets in Gauteng province of South Africa, making a total of twelve. Alternatives to replace antibiotics in broiler diets : effects on protein utilization and production performance

Thesis (MScAgric)--Stellenbosch University, 2008. / ENGLISH ABSTRACT: Different substances were evaluated and compared to an antibiotic, in terms of their effect on nitrogen - and amino acid digestibilities. Two digestibility trials and one performance trial were conducted. Trials one and two apparent nitrogen (AND)- and amino acid (AAD) digestibilities were determined from digesta collected at the terminal ileum (ileal digestibility method). In Trial 3 the substances were evaluated in terms of their potential to improve production performance. Broilers were fed a maizesoya based diet throughout the three trails. In the first trial, garlic and a commercial prebiotic (Bio-Mos®), were tested and compared in terms of AND and AAD, to an antibiotic (doxycyclin, Doxyvete-SOS). A starter and finisher diet were fed as either mash or pellets. The garlic was included at 8g/kg, 13g/kg and 18g/kg to the starter and finisher diets. Bio-Mos® was added at 1g/kg, 2g/kg and 3g/kg to the starter diet, and 0.5g/kg, 1g/kg and 1.5g/kg to the finisher diet. The doxycyclin was added at 0.3 g/kg. None of the treatments had any beneficial effects in terms of AND. Feeding a pellet seem to have some negative effects in terms of AND. In general most of the treatments did not show any improvement in AAD at any determination period (day 21, 28 or 35). At day 21 and day 35, the mash diet supplemented with 18g/kg garlic had a negative effect on AAD, when compared to the negative and positive control. It doesn’t seem that feeding either a mash or a pellet had an influence on the effects exerted by the different treatments. In the second trial the influence of Bio-Mos®, a blend of organic acids, probiotics and electrolytes (Acid-Pak 4-way®) and a medium-chain triglyceride (MCT) were evaluated and compared in terms of AAD and AND, to the effect of an antibiotic, doxycyclin. The starter and finisher diets were fed as a mash. Bio-Mos® was included at 1g/kg, 2g/kg, and 3g/kg in the starter diet, and at 0.5g/kg, 1g/kg, 1.5g/kg in the finisher diet, respectively. Acid-Pak 4-way® was included at 0.4g/kg, 1g/kg and 1.6g/kg for both the starter and finisher diets. Medium-chain triglycerides (MCT) were allocated at 3g/kg, 3.6g/kg, 4.2g/kg for the starter diet, and 2.1g/kg, 2.7g/kg and 3.4g/kg for the finisher diet. An antibiotic, doxycyclin, was included at 0.3 g/kg. With AND, no treatment had any significant effect for the entire experimental period. At day 21, the treatment supplemented with MCT (3.4g/kg) had the most significant beneficial effect on AAD, when compared to the negative- and positive controls, as it increased AAD for the majority of the amino acids. The treatment with Acid-Pak 4-way® (1g/kg) had the most significant negative effect on AAD when compared to the positive control. At day 28, the treatments with Bio-Mos® (0.5g/kg and 1.5g/kg) and Acid-Pak 4-way® (0.4g/kg) had the most significant beneficial effect on AAD when compared to the positive control. It increased AAD for more than half of the 17 amino acids evaluated. The treatment supplemented with MCT (2.7g/kg) has shown the most significant negative effect on AAD, when compared to the positive control. In the third trial the effect of Bio-Mos®, Acid-Pak 4-way® and MCT on production performance was evaluated, and compared to the effects of the presence or absence of doxycyclin. Body weight (BW), body weight gain (BWG), feed intake (FI) and feed conversion ratio (FCR) were measured. The starter and finisher diets were fed as a mash. Bio-Mos®, MCT and Acid-Pak 4-way® were included at 3.0g/kg, 4.2g/kg and 1.6g/kg, respectively in the starter and finisher diets. Birds were weighed (per pen) on arrival and on days 7, 14, 21, 28, 35. Feed intake (FI) per pen was measured at days 7, 14, 21, 28 and 35, and mortality was recorded daily. In terms of BWG, Acid-Pak 4-way® had a higher BWG, when compared to the negative control, Bio-Mos® and MCT. It can be concluded that Bio-Mos®, Acid-Pak 4-way®, as well as MCT can be a possible alternatives to antibiotic supplementation. These three treatments did not necessary prove to be more effective than antibiotics, but are definitely competitive alternatives. / AFRIKAANSE OPSOMMING: Verskillende behandelings is geëvalueer en vergelyk met ‘n antimikrobiese produk, in terme van hul uitwerking op stikstof - en aminosuur verteerbaarhede. Twee verteringstudies en produksieprestasie studie is uitgevoer. In die eerste twee studies is die skynbare stikstof (AND)- en aminosuur (AAD) verteringskoöeffisiënte bepaal deur gebruik te maak van digesta wat by die terminale ileum ingesamel is (ileale verteringsmetode). In die derde studie is die produksie prestasie van braaikuikens op ‘n gebalanseerde metaboliseerbare energie (AME) rantsoen, soos beïnvloed deur die verskillende behandelings, geëvalueer. In die eerste studie is knoffel en ‘n kommersiële prebiotikum (Bio-Mos®) geëvalueer en met ‘n antibiotikum (doksisiklien, Doxyveto-SOS) in terme van AND en AAD vergelyk. Beginner- en afrondingsrantsoene is as ‘n meel of pille gevoer. Die knoffel is teen 8g/kg, 13g/kg en 18g/kg in die rantsoen ingesluit. In die tweede studie is Bio-Mos®, ‘n organiese suur (Acid-Pak 4-way®) en ‘n medium-ketting trigliseried (MCT) geëvalueer en met ‘n antbiotikum, doksisiklien (Doxyveto-SOS) in terme van AND en AAD, vergelyk. Beginner- en afrondingsrantsoene is gevoer as ‘n meel. Bio-Mos® is teen 1g/kg, 2g/kg, and 3g/kg in die beginner rantsoen en teen 0.5g/kg, 1g/kg, 1.5g/kg in die afrondingsrantsoen, ingesluit. In die derde studie is die insluiting van Bio-Mos®, Acid-Pak 4-way® en MCT getoets om die invloed op braaikuiken produksie prestasie te evalueer, en te vergelyk met die invloed van die insluiting of afwesigheid van ‘n antibiotikum. Liggaamsmassa (BW), liggaamsmassa toename (BWG), voerinname (FI) en voeromsetverhouding (FCR) is gemeet. Beginner- en afrondings rantsoene is gevoer as ‘n meel. Bio-Mos®, MCT en Acid-Pak 4-way® is onderskeidelik teen 3.0g/kg, 4,2g/kg en 1.6g/kg in die rantsoen ingesluit. Die kuikens is met aankoms (per hok) geweeg, asook op dae 7, 14, 21, 28, 35. Voerinname per hok is gemeet op dae 7, 14, 21, 28 en 35. Mortaliteite is daagliks aangeteken. Die insluiting van Acid-Pak 4-way® het in vergelyking met die negatiewe kontrole, Bio-Mos® en MCT insluiting ‘n hoër BWG tot gevolg gehad. Die gevolgtrekking wat gemaak kan word is dat Bio-Mos®, Acid-Pak 4-way® en MCT gebruik kan word as ‘n moontlike alternatief vir antibiotika insluiting. Hierdie drie behandelings was nie noodwending meer effektief as die antibiotika nie, maar het wel bewys dat dit kompeterende alternatiewe is.

Effects of feeding omega-3 fatty acids and vitamin E on the chemical composition and microbial population of broiler meat

Thesis (MScAgric) -- University of Stellenbosch, 2000. / ENGLISH ABSTRACT: Lipids remain one of the most important nutrients required by broilers. The growing awareness that some Western societies have too high a dietary ratio of n-6/n-3 polyunsaturated fatty acids is of direct relevance to broiler nutrition and lipid metabolism. Meaningful quantities of n-3 polyunsaturates have been incorporated into major poultry tissues, so that the production of broiler meat with high n-3 polyunsaturates becomes advantageous for the broiler industry as they are perceived as having a 'healthier' lipid profile. Unfortunately, such broiler meat is rather susceptible to oxidative deterioration, and oxidation often determines shelf life of poultry meat products. The addition of a-tocopherol (vitamin E) to broiler diets is an effective means of improving the oxidative stability of broiler meat. Elevated a-tocopherol levels in broiler feeds increase tissue concentrations thereof resulting in improved stability of membranal structures which may be expected to increase the oxidative stability of broiler meat and meat products. Three investigations were done at Mariendahl Poultry Research Station in Stellenbosch. The broilers were kept in 1 x 0.4 x 0.5 m cages in a broiler rearing house. All the trials started with day-old chicks, except experiment 1 where 3-week old broilers were used. At the end of trials 2 and 3 the 6-week old broilers were slaughtered and the carcasses prepared for chemical analysis. Experiment 1: Metabolisabie energy of Canola acid oil and Famarol acid oil for broiler chickens. In trials with 21-day-old male broilers the true metabolisabie energy value, corrected for nitrogen retention (TMEn) was determined by the balance method for Canala acid oil (CAO) and Famarol acid oil (FAO). The trials were duplicated, each time using different samples of the two oils from the same source (experiment 1 and 2). Each of the two oils were blended in two ratios with a basal diet to form the test diets, viz. 100% Basal; 96% Basal: 4% Oil; 92% Basal: 8% Oil. In experiment 3, 50 % bran was added to the maize to form the basal diet. The balance trials lasted for 3 days after an adaptation period of 4 days. The TMEn values determined by regression for the broilers of CAO did not differ significantly (P>0.05) between experiments 1 and 2. However, the value for experiment 3 was significantly (P<0.05) higher than those for experiments 1 and 2. The TMEn values of FAO also did not differ significantly (P>0.05) between experiments 1 and 2, although the value for experiment 3 was significantly higher than that of experiment 1. The addition of 50 % bran to the basal diet in experiment 3 could have stimulated the digestive breakdown process and hence increase the secretion of digestive enzymes. This could lead to an increase in the utilisation of the test lipid and therefore an increase in the TMEn value. The TMEn values of CAO differed significantly (P<0.05) from those of FAO for all three the experiments (exp. 1:30.6 ± 0.399 MJ/kg for CAO vs. 25.9 ± 0.441 MJ/kg for FAO; expo 2: 31.0 ± 0.633 MJ/kg for CAO vs. 26.1 ± 0.668 MJ/kg for FAO: expo 3: 32.1 ± 0.867 MJ/kg for CAO vs. 27.1 MJ/kg for FAO).Experiment 2: of broilers. The dietary effects of various combinations of Canala acid oil (CAO, a high level of C18:3n-3 and MUFA) and Famarol Effects of various dietary n-6/n-3 fatty acid ratios on the perfonnance and body composition acid oil (FAO, a high level of 18:2n-6 and SFA) on tissue fatty acid composition were studied in broiler carcasses and abdominal fat pads. From day-old to six weeks, chicks were fed one of six diets containing 100% FAO, 80% FAO-20% CAO, 60% FAO-40% CAO, 40% FAO-60% CAO, 20% FAO-80% CAO, 100% CAO. There were no statistical differences (P>0.05) in average daily gain (1.71 ± 0.059 g) or feed conversion ratios (1.97 ± 0.051) among dietary groups. No statistical differences (P>0.05) were found in the chemical proximate composition of the carcasses for the moisture (66.20 ± 0.112 %), protein (17.63 ± 0.484 %), lipid (15.92 ± 1.507 %) and ash (0.95 ± 0.115 %) content among dietary groups. No statistical differences (P>0.05) were found in the chemical proximate composition of the abdominal fat pads for the moisture (28.77 ± 0.112 %), protein (3.03 ± 0.484 %), lipid (63.32 ± 9.789 %) and ash (0.45 ± 0.135 %) content among dietary groups. With the increase in dietary CAO levels, the percentages of C18:2n-6 and C20:4n-6 in the carcasses decreased respectively with 1.78 % from 20.88 % and 0.35 % from 1.05 %, whilst C18:3n-3 and longer chain n-3 fatty acids such as C20:5n-3 and C22:6n-3 increased respectively with 2.25 % from 1 %, 0.1 % from 0.1 % and 0.67 % from 0.2 %. The same tendency was seen in the abdominal fat pads where C18:2n-6 and C20:4n-6 decreased respectively with 1.55 % from 20.75 % and 0.98 % from 1.2 % with an increase in dietary CAO, whilst C18:3n-3, C20:5n-3 and C22:6n-3 increased respectively with 2.13 % from 1.15 %, 0.45 % from 0.03 % and 0.95 % from 0.05 %. The n-3/n-6 ratio in the carcasses and abdominal fat pads increased respectively with 0.16 % from 0.06 % and 0.19 % from 0.06 % with an increase in dietary CAO. These results clearly indicate that dietary CAO enriched with a-linolenic acid lower saturated fatty acids respectively in broiler carcasses and abdominal fat pads with 4.88 % from 31.6 % and 10.63% from 31.1 %, whilst increasing monounsaturated fatty acids with 3.87 % from 44.95 % and 7.25 % from 46.7 % respectively and polyunsaturated fatty acids with 1.02 % from 23.45 % and 2.38 % from 23.2 % respectively. Experiment 3: Effect of dietary vitamin E on the performance of broilers and oxidative stability, colour, microbiological stability, fatty acid composition and pH of broiler meat during refrigerated and frozen storage. Experiment 1 was carried out with 220 one-day-old broiler chicks to evaluate the effect of eleven concentrations of vitamin E (0, 20, 40, 60, 80, 100, 120, 140, 160, 180 and 200 mg a-tocopheryl acetate 1 kg diet) on their production performance and the oxidative stability of their frozen broiler carcasses. The diets with vitamin E levels 0 to 100 mg were fed from day-old to 42 days of age while the diets with vitamin E levels 120 to 200 mg were fed from 21 to 42 days of age. The oxidative stability, evaluated by thiobarbituric acid reactive substances (TBARS) values, was determined after 30, 90, 120 and 150 days of storage at -20°C. There were no statistical differences (P>0.05) in average daily gain (1.85 ± 0.111 g) or feed conversion ratios (2.29 ± 0.397) among dietary groups. TBARS values increased significantly (P<0.05) with increasing time of storage (basal diet: day 30 = 1.71 ± 0.51; day 150 = 4.89 ± 0.51), but decreased significantly (P<0.05) with increasing vitamin E levels (day 150: basal = 4.89 ± 0.51; 100 mg / kg = 1.09 ± 0.27). Experiment 2 was carried out with day-old broiler chicks to evaluate the effect of five concentrations of vitamin E (0, 40, 80, 120 and 160 mg atocopheryl acetate / diet) on their performance and the oxidative stability of their refrigerated carcasses. The experimental diets were fed from day-old to 42 days of age. The oxidative stability, evaluated by TBARS values, colour deterioration and microbiological stability were determined after 0, 4, 8, 10 and 12 days of storage at 4°C. Fatty acid analysis was done on the samples of days 0 and 12. There were no statistical differences (P>0.05) in average daily gain (1.88 ± 0.117 g) or feed conversion ratios (2.37 ± 0.467) among dietary groups. TBARS values increased significantly (P<0.05) with increasing time of storage, but decreased significantly (P<0.05) with increasing vitamin E levels. There were no statistical differences (P>0.05) in colour measurements for L* (44.97 ± 0.662), a* (5.23 ± 0.315) or b* (12.76 ± 0.321) values between treatments. Microbiological counts increased significantly (P<0.05) over time with vitamin E concentration showing no effect. There were no statistical differences (P>0.05) for any of the fatty acid groups measured (SFA: Day 0 = 26.1 ± 1.13%, Day 12 = 26.1 ± 1.17%; MUFA: Day 0 = 41.4 ± 1.46%, Day 12 = 40.2 ± 2.28%; PUFA: Day 0 = 32.4 ± 1.95%, Day 12 = 33.8 ± 2.52%) among dietary groups. Similarly, none of the fatty acids showed statistical significant (P>0.05) concentration changes over time. There were no statistical differences (P>0.05) in pH (6.01 ± 0.206) among dietary groups. / AFRIKAANSE OPSOMMING: Lipiede is steeds een van die mees belangrike voedingstowwe wat deur braakuikens benodig word. Die groeiende bewuswording dat sekere Westerse gemeenskappe 'n te hoë verhouding van n-6/n-3 poli-onversadigde vetsure in hul dieet het, is direk relevant vir braaikuikenvoeding en lipiedmetabolisme. Betekenisvolle hoeveelhede n-3 polionversadigde vetsure is geïnkorporeer in die belangrikste hoendersnitte, met die gevolg dat die produksie van braaikuikenvleis met hoë n-3 poli-onversadigde vetsure voordelig is vir die braaikuikenindustrie en geag word 'n meer "gesonde" beeld te hê. Ongelukkig is sodanige braaikuikenvleis redelik vatbaar vir oksidatiewe bederf, en oksidasie bepaal dikwels die rakleeftyd van hoendervleisprodukte. In al die proewe is dagoud kuikens gebruik, behalwe eksperiment 1 waar drieweek oue kuikens gebruik is. Aan die einde van proewe 2 en 3 is die ses-week oue braaikuikens geslag en die karkasse voorberei vir analise. Eksperiment 1: braaikuikens. Die ware metaboliseerbare energie waarde van Canola voergraadolie (CAO) en Famarol voergraadolie (FAO), Metaboliseerbare energie van Canola voergraadolie en Famarol voergraadolie vir gekorregeer vir stikstof retensie (WMEn), is by wyse van proewe op 21 dae oue braaikuikenhaantjies bepaal deur van die balansrnetode gebruik te maak. Die proewe is tweemaal herhaal vir verhoogde akkuraatheid, met die gebruik van verskillende monsters van die twee olies vanaf dieselfde bron. Die olies is in twee verhoudings met 'n basale diëet gemeng om die proef dieet te vorm, nl. 100% Basaal; 96% Basaal: 4% Olie en 92% Basaal: 8% Olie. Die balans proewe het 3 dae geduur na afloop van 'n aanpassingsperiode van 4 dae. Die WMEn waardes van CAO, bepaal deur middel van regressie analise, het nie betekenisvol verskil (P>0.05) tussen eksperimente 1 en 2 nie. Die waarde van eksperiment 3 was betekenisvol hoër (P<0.05) as die van die eerste twee eksperimente. Die WMEn waardes van FAO het ook nie betekenisvol verskil (P>0.05) tussen eksperimente 1 en 2 nie, maar die waarde vir eksperiment 3 was betekenisvol hoër as dié van eksperiment 1. Geen statistiese verskille (P>0.05) is gevind in die chemiese samestelling van die abdominale vetneerlegging vir vog (28.77 ± 0.112 %), proteien (3.03 ± 0.484 %), lipied (63.32 ± 9.789 %) en as (0.45 ± 0.135 %) inhoud onder die rantsoengroepe nie. Met die verhoging in die rantsoen CAO vlakke het die persentasie van C18:2n-6 en C20:4n-6 in die karkasse verminder met 1.78 % en 0.35 % respektiewelik, terwyl C18:3n-3 en langer ketting n-3 vetsure soos C20:5n-3 en C22:6n-3 respektiewelik met 2.25 %, 0.1 % en 0.67 % verhoog het. Dieselfde tendens is opgemerk in die abdominale vetneerlegging waar C18:2n-6 en C20:4n-6 afgeneem het met 1.55 % en 0.98 % respektiewelik met die verhoging van rantsoen CAO, terwyl C18:3n-3, C20:5n-3 en C22:6n-3 verhoog het met 2.13 %, 0.45 % en 0.95 % respektiewelik. Die n- 3/n-6 verhouding in die karkasse en abdominale vetneerlegging het verhoog met 0.16 % en 0.19 % respektiewelik met die verhoging van die rantsoen CAO. Die resultate toon onomwonde aan dat rantsoen CAO verryk met c-Iinoletensuur, verlaag versadigde vetsure in braaikuikenkarkasse en -adbdominale vetneerleggings met 4.88 % en 10.63% respektiewelik, terwyl die mono-onversadigde vetsure met 3.87 % en 7.25 % respektiewelik verhoog word en polionversadigde vetsure met 1.02 % en 2.38 % respektiewelik verhoog word. Eksperiment 3: Die invloed van vitamine E op die produksie van braaikuikens en die oksidatiewe stabiliteit, kleur, mikrobiologiese stabilitiet, vetsuursamestelling en pH van braaikuikenvleis gedurende verkoelde en bevrore berging. Eksperiment 1 is uitgevoer met 220 dagoud braaikuikens ten einde die effek van elf konsentrasies van vitamine E (0, 20, 40, 60, 80, 100, 120, 140, 160, 180 en 200 mg a-tokoferyl acetaat / kg voer) op hul produksieprestasie en die oksidatiewe stabiliteit van hul gevriesde braakuikenkarkasse te evalueer. Die diëte met vitamine E vlakke 0 tot 100 mg is vanaf dagoud tot 42-dae-ouderdom gevoer, terwyl die diëte met vitamine E vlakke van 120 tot 200mg gevoer is vanaf 21 tot 42- dae-ouderdom. Die oksidatiewe stabiliteit, soos geëvalueer deur tiobarbituriese suur reaktiewe stowwe (TBARS) waardes, is bepaal na 30, 90, 120 en 150 dae van berging teen -20°C. Daar was geen statistiese verskille (P>0.05) in die gemiddelde daaglikse toename (1.85 ± 0.111 g) of voeromsetverhoudings (2.29 ± 0.397) tussen die rantsoengroepe nie. TBARS waardes het betekenisvol toegeneem (P<0.05) met die verhoging in bergingsperiode, maar het betekenisvol afgeneem (P<0.05) met verhoogde vitamine E vlakke. Eksperiment 2 is uitgevoer met dagoud braaikuikens ten einde die effek van vyf konsentrasies van vitamine E (0, 40, 80, 120 and 160 mg a-tokoferyl acetaat / kg voer) op hul prestasie en die oksidatiewe stabiliteit van hul verkoelde karkasse te evalueer. Die eksperimentele diëte is gevoer vanaf dagoud tot 42- dae-ouderdom. Die oksidatiewe stabiliteit, geëvalueer deur middel van TBARS waardes, kleur afname en mikrobiologiese stabiliteit is bepaal na 0, 4, 8, 10 en 12 dae van berging teen 4°C. Vetsuuranalises is gedoen op die monsters van dae 0 en 12. Daar was geen statistiese verskille (P>0.05) in die gemiddelde daaglikse toename (1.88 ± 0.117 g) of voeromsetverhoudings (2.37 ± 0.467) tussen die rantsoengroepe nie. The digestibility and degradability of feeds and protein sources in Dohne merino sheep and boer goats

Thesis (MScAgric (Animal Sciences))--University of Stellenbosch, 2010. / ENGLISH ABSTRACT: The objective of this study was to evaluate Dohne Merino sheep and Boer goats in terms of the degradable parameters of a high-fibre diet, a low-fibre diet and two vegetable protein sources commonly used in South Africa. Differences between species were evaluated following the potential differences within species. The feedstuffs used were those for the following diets: low-fibre diet (LF); high-fibre diet (HF); sunflower meal (SFM) oilcake; and soybean meal (SBM) oilcake. In the first trial, the digestible characteristics of the HF and LF diets were determined by means of a digestibility study. A 6 × 6 Latin square design was used to determine whether Dohne Merino sheep or Boer goat wethers differ regarding the digestibility characteristics of low- and high-fibre diets. The diets were fed once daily at 1.24 kg to all the wethers, which had ad libitum access to fresh water. Each period consisted of 10 days of adaptation and seven days of faecal and urinary sampling. The results indicated that the intake and digestibility characteristics of nutrients did not differ between sheep and goats. However, the different diets differed in terms of the nutrient intake and digestibility range of sheep and goats. The second trial was an in sacco degradability trial to determine the dry matter (DM) and crude protein (CP) degradability of the LF, HF, SBM and SFM diets. Six Dohne Merino and six Boer goat wethers were fitted with rumen cannulae so that they could be used in the trial. All wethers received the same basal diet. The samples were incubated in the rumen in polyester Dacron bags, with the bags being removed at intervals of 0h, 3h, 9h, 12h, 24h, 48h, 72h, and 96h for the LF and HF diets. All the oilcake was removed at intervals of 0h, 2h, 4h, 8h, 12h, 16h, 24h, 36h and 48h. The sheep and goats were found not to differ from one another in terms of effective degradability of any of the feedstuffs concerned. However, within species differences were observed. To establish a fully integrated outcome of degradability, the study described in the current thesis was structured in such a way that the in vitro trial ran parallel with the in sacco trial, being performed with the aid of a Daisy Incubator (ANKOM Technology Corp., Fairport, NY). Such a procedure was only adopted in relation to the SFM and SBM diets in order to evaluate their in vitro data in relation to the in sacco data. The same oilcake was tested in the case of both trials, with the composite sample of rumen liquid of four sheep or goats, which was used in the in sacco trial, also being used in the in vitro study. In the study, DM disappearance values were determined and fitted to a single-compartment model by means of an iterative least-square procedure in order to determine the DM and CP degradability parameters. The DM used in vitro or in sacco was compared, using the actual values obtained after 8h incubation, due to only a limited amount of residue being left after incubation. In the study, the in vitro method overestimated the digestibility of SBM by 37% to 39% and the digestibility of SFM by 17% to 20% compared with that found to occur in the in sacco method. In vitro DM disappearance values for all SBM samples were found to be higher than those that were detected in the SFM samples. The percentage of in vitro true digestibility parameters was also calculated. No significant differences were found between species for effective degradability, though differences were observed within species between the two substrates concerned. In conclusion, the sheep and goats used in the study were not found to differ in terms of digestion parameters when they were compared on different types of roughage or protein sources. However, within species differences were, indeed, found to occur. Sheep and goats digested the SBM better than they did the SFM. / AFRIKAANSE OPSOMMING: Die doel van hierdie studie was om te bepaal of Dohne Merino skape verskil van Boerbokke in terme van degradeerbaarheidsparameters van ‘n hoë vesel-, ‘n lae veseldieët en twee plantaardige proteïenbronne wat algemeen in Suid-Afrika gebruik word. Die verskille tussen spesies is ge-evalueer en daarna die potensiële verskille binne spesies. Die volgende grondstowwe is geëvalueer: ‘n laevesel-dieët (LF), ‘n hoëvesel-dieët (HF), sonneblom-oliekoekmeel (SFM) en sojaboon-oliekoekmeel (SBM). In die eerste proef is die degradeerbaarheidsparameters van die HF dieët en die LF dieët met behulp van ‘n verteerbaarheidstudie bepaal. Om af te sluit het dit voorgekom dat skape en bokke nie verskil aan degradeerbaarheidswaardes wanneer daar ‘n vergelyking was tussen verskillende vesels- en proteϊenbronne nie, alhoewel verskille voorgekom het binne spesies. Skape en bokke het SBM effektief beter verteer as SFM.

The effect of fine particle removal from ground forage samples on In Saccro dry matter and neutral detergent fiber disappearance values

Thesis (MScAgric (Animal Sciences))--University of Stellenbosch, 2011. / ENGLISH ABSTRACT: In vitro and in situ methods using the in sacco technique have a wide application in ruminant nutrition as they allow the degradability and quality of forages and ruminant diets to be determined quicker and at a lower cost than in vivo methods. These trials make use of artificial fibre bags, made of polyester (dacron) or nylon which are available in variable pore sizes. Results from such degradability trials are of great value to feed formulation programs such as AMTS.cattle and CPM Dairy and the more accurate the results are obtained from such trials the more accurate feed formulation models are enabling the ruminant nutritionist to formulate the best diet possible to reach the genetic potential of ruminants. The accepted method for in sacco trials (NRC, 2001) requires that the feed samples are ground through a 2 mm screen. This usually results in a variety of particle sizes, including a significant amount of extremely fine particles. Research has, however, shown that these fine particles can potentially be washed out of the dacron bags that are used in in sacco degradability trials. This would result in an overestimation of the soluble and rapidly fermentable nutrient fractions. The objective of this study was to determine the effect of fine particle removal from ground forage samples on the chemical composition and in vitro dry matter (DM) and neutral detergent fibre (NDF) degradability of forages. Lucerne hay, oat hay and wheat straw samples were sourced from seven different locations in the Western Cape. Samples were milled through a 2 mm screen and then sieved through either 150 μm, 125 μm or 106 μm. All fractions were analysed for DM, crude protein (CP), NDF, fat and ash. Based on the NDF content of the original samples, four samples from each forage type were selected for in vitro trials to determine DM and NDF disappearance over time. Samples were incubated for 0, 6, 24 and 48 hours in an ANKOM Daisy II incubator. Significant variation occurred within forage types in terms of chemical composition. Fine particle removal had no effect on the NDF content of lucerne hay and wheat straw, but sieved oat hay fractions had a higher NDF content than the un-sieved samples. The NDF content was on average 635.9 for the sieved OH fractions, whereas the NDF content of the un-sieved samples was 606.8. The CP content of sieved oat hay (61.4 on average) and wheat straw fractions (47.7 on average) were lower than the un-sieved fractions (65.7 for OH and 55.4 for WS), whereas for lucerne hay, sieving had no effect on CP content. Dry matter and NDF disappearances were significantly higher for the un-sieved samples than for the sieved fractions for all three forage types at all incubation times, which indicates an overestimation of the soluble and readily digestible forage fractions. Compared to sieved samples, DMD values at 0 hours (washing only) of the un-sieved samples were, on average, 13.8% higher for lucerne hay, 27.3% for oat hay and 44.7% for wheat straw. At 48 h, the overestimation of lucerne DMD for the un-sieved samples was between 4.0% (compared to 106 μm sieve) and 7.3% (compared to 150 μm sieve). This over-estimation in the un-sieved samples was carried over to all four time points. No significant differences between the fractions (150, 125 and 106) were found within a forage type at all incubation times. The estimated degradation rates and the predicted digested proportions were also significantly higher for the un-sieved fractions compared to the sieved fractions. It was concluded that fine particle removal from forage samples would result in more accurate estimations of in sacco nutrient degradability. / AFRIKAANSE OPSOMMING: In sacco in vitro- en in situ-metodes word dikwels toegepas in die studie van herkouervoeding aangesien hierdie metodes vinniger, meer effektief en meer ekonomies is as in vivo-metodes. Hierdie studies maak gebruik van kunsveselsakkies, gemaak van poliëster (dacron) of nylon wat beskikbaar is in verskeie poriegroottes. Resultate van sulke verteringsproewe is belangrik vir toepassing in voerformuleringsprogramme soos AMTS.cattle en CPM Dairy. Die resultate wat deur hierdie studies verkry word, is belangrik vir akkurate voerformulering deur formuleringsmodelle en stel die herkouervoedingkundige in staat om die ideale voer te formuleer vir die manifestering van die dier se genetiese potensieaal. Sifting het geen invloed op die RP-inhoud van lusernhooi gehad nie, maar vir hawerhooi (61.4 gemiddeld) en koringstrooi (47.7 gemiddeld) was die RP-inhoud van die gesifte monsters betekenisvol laer as dié van die ongesifte monsters (65.7 vir hawerhooi en 55.4 vir koringstrooi). In vergelyking met die gesifte monsters, was die in vitro DM- en NDF-verteerbaarhede betekenisvol hoër vir die ongesifte monsters vir al drie ruvoertipes by alle inkubasietye. Hierdie resultate bevestig ‘n oorskatting van oplosbare en maklik verteerbare fraksies in gemaalde voermonsters. In vergelyking met die gesifte monsters, was die DMV-waardes van die ongesifte monsters by 0 ure (slegs gewas) gemiddeld 13.8% hoër vir lusernhooi, 27.3% vir hawerhooi en 44.7% vir koringstrooi. Na 48 h inkubasie was die oorskatting van lusern DMV vir die ongesifte monsters tussen 4.0% (vergeleke met die 106 μm sif) en 7.3% (vergeleke met 150 μm sif). Die oorskatting is oorgedra na al vier inkubasietye. Die resultate het geen noemenswaardige verskille tussen die fraksies (150 μm, 125 μm en 106 μm poriegroottes) van ‘n ruvoertipe by enige inkubasietyd aangedui nie. Die beraamde verteringstempo’s en verteerde fraksies was ook aansienlik hoër vir die ongesifte monsters in vergelyking met die gesifte monsters. Die gevolgtrekking is gemaak dat die verwydering van fyn partikels uit gemaalde ruvoermonsters die akkurate bepaling van in sacco verteerbaarheidswaardes verhoog.

A nutrição tem sido identificada como um fator chave que afeta diretamente o crescimento e o bom desempenho reprodutivo do animal quando utilizado como modelo experimental. Desta forma, é essencial uma dieta ideal acessível através da qual o zebrafish possa suprir todas suas necessidades nutricionais desde a fase larval até a sua fase adulta sem que haja necessidade de complementá-la com outros tipos de dietas, simplificando o cultivo destes animais. A dieta recomendada nos guidelines para a alimentação do Zebrafish criado em laboratório é composta por náuplios de Artemias (crustáceo rico em proteínas) em conjunto com uma ração seca rica em proteínas e sais minerais fundamentais para o bom desenvolvimento e reprodução desses animais. Este estudo teve como objetivo desenvolver e padronizar uma dieta ideal de alimentação simples, eficaz, saudável e de baixo custo a fim de obter o melhor desempenho possível nos experimentos desenvolvidos com o zebrafish. Foram utilizadas 4 diferentes modelos de dietas preparadas a partir de 3 formas de alimentação que consistiram em náuplios de Artemias, a ração padrão-ouro disponível no mercado europeu denominada Gemma Micro® desenvolvida pela Skretting e a ração elaborada para este estudo identificada como Ração Ideal. Os animais foram divididos em 4 grupos de acordo com a dieta recebida: Grupo1- alimentados com Ração Gemma(G), Grupo 2- alimentados com Ração Gemma + Artemia (G+A), Grupo 3- alimentados com Ração Ideal (I) e Grupo 4- alimentados com Ração Ideal+Artemia (I+A). Para avaliar o desempenho da Ração Ideal, foram analisados comparativamente entre os grupos os parâmetros de sobrevivência, crescimento, maturação sexual e fertilidade em duas gerações de animais (Geração 0 Geração 1). Após a análise e vários testes realizados dentro de um modelo de ração que se ajustasse ao perfil de referência, Gemma Micro® e que atendesse as necessidades do zebrafish passamos a dar início aos experimentos utilizando a ração que apresentou melhor desempenho que passou a ser denominada Ração Ideal. Em relação a sobrevivência dos animais, observamos que, na Geração 0 os animais dos grupos 3 e 4 apresentaram maiores taxas de sobrevivência em relação aos outros grupos, no entanto na geração 1 todos os grupos apresentaram taxas de sobrevivência semelhantes. Analisando o crescimento dos animais, na geração 0 os animais dos grupos 2 e 4 apresentaram crescimento semelhante e foram maiores que os animais dos grupos 1 e 3. Na geração 1, os animais do grupo 2 foram os que mais cresceram e o grupo 4 cresceu mais em relação ao grupo 3. Todos os animais estavam maduros sexualmente aos 90dpf e quantidade de ovos viáveis foram maior nos grupos 1 e 2 em ambas as gerações. A Ração Ideal forneceu um aporte calórico suficiente e semelhante á ração Gemma para a sobrevivência dos animais nas fases iniciais do desenvolvimento, apresentando taxas de sobrevivência semelhantes entre os grupos. O acréscimo da artêmia á ração ideal foi fundamental para que o crescimento dos animais alimentados com essa ração demonstrassem um crescimento semelhante á ração referência, demonstrando a deficiência proteica da ração ideal. Todas as dietas promoveram maturidade sexual na faixa etária esperada (a partir de 90dpf) no entanto parece que a fertilidade foi maior nos grupos 1 e 2, possivelmente pelo maior aporte proteico que esses animais receberam. Este estudo conseguiu desenvolver e padronizar uma dieta ideal de alimentação, eficaz, saudável e de baixo custo semelhante ao padrão-ouro utilizado em laboratórios europeus e americanos que, após alguns ajustes em sua formulação poderá ser comercializada e utilizada pelos laboratórios de pesquisa ao redor do país / Nutrition has been identified as a key factor that directly affects the growth and good reproductive performance of the animal when used as an experimental model. In this way, an ideal diet should be accessible through which the zebrafish can meet all their nutritional needs from the larval phase to adulthood without any complement with other types of diets, simplifying the management of these animals. The diet recommended in the Zebrafish feeding guidelines is composed of Artemias nauplii (crustacean rich in proteins) together with a dry diet rich in proteins and minerals that are essential for the proper development and reproduction of these animals. This study aimed to develop and standardize an ideal diet for simple, effective, healthy and low-cost feeding in order to obtain the best possible performance in the experiments developed with zebrafish. Four different diets were prepared from 3 feeding forms consisting of Artemias nauplii, the gold-standard feed available in the Europe, the Gemma Micro® developed by Skretting and the dry feed elaborated in this study identified as Ideal . The animals were divided into 4 groups according to the diet received: Group 1 fed with Gemma (G), Group 2 fed with Gemma + Artemia (G + A), Group 3 fed with Ideal dry feed (I) and Group 4- fed with Ideal dry feed+ Artemia (I + A). To evaluate the performance of the Ideal dry feed, the parameters of survival, growth, sexual maturation and fertility in two generations of animals (Generation 0 Generation 1) were comparatively analyzed between the groups. After several tests performed within a feed model that fits the reference profile, Gemma Micro® and meets the zebrafish needs, we initiated the experiments using the feed that presented the best performance, named Ideal. Regarding the animals survival, we observed that in the Generation 0 animals of groups 3 and 4 presented higher survival rates in relation to the other groups, however in generation 1 all groups had similar survival rates. Analyzing the growth of the animals, in generation 0 the animals of groups 2 and 4 presented similar growth and were larger than the animals of groups 1 and 3. In generation 1, the animals of group 2 were the ones that were bigger than other ones. Animals of group 4 were bigger than group 3. All animals were sexually mature at 90dpf and fertility was evaluated by viable egg count. The number of viable egg were higher in groups 1 and 2 in both generations. The Ideal feed provided a sufficient caloric intake similar to the Gemma for the survival of the animals in the early stages of development, once survival rates were similar between the groups. Adding artemia to the ideal feed was fundamental for animal development, once the growth of the animals fed with ideal plus artemia demonstrated a similar growth to the reference feed, demonstrating the protein deficiency of the ideal feed. All diets promoted sexual maturity at the expected age range (from 90dpf), however it appears that fertility was higher in groups 1 and 2, possibly due to the higher protein intake of these animals. This study was able to develop and standardize an ideal, effective, healthy and low-cost diet similar to the gold standard feed used in laboratories around the world. After some adjustments in its formulation, the ideal feed should be marketed and used in the research laboratories in the country

Tendo em vista o notável crescimento da aquicultura mundial e principalmente a brasileira, torna-se cada vez mais importante o desenvolvimento de tecnologias que favoreçam esta prática. No entanto, a intensificação da piscicultura faz aumentar as chances de ocorrência de epizootias nos sistemas de produção como resultado do maior estresse imposto aos animais (lotação, manejo, transporte etc). Este estudo determinou a eficácia na utilização de imunoestimulantes dietéticos (levamisol e vitamina E) e a farmacocinética do imunoestimulante levamisol em juvenis de surubim Pseudoplatystoma sp. O levamisol dietético não afetou as variáveis de desempenho, mas influenciou positivamente o sistema imunológico inespecífico do cachara, afetando a atividade da lisozima sérica, sendo o melhor resultado observado para a concentração de levamisol de 247 mg kg-1 de ração. No estudo com vitamina E, não foram registrados efeitos sobre as variáveis de desempenho; ainda, na dose dietética 166 mg kg-1 o acetato de DL-?-tocoferol promoveu aumento das globulinas séricas. O estudo farmacocinético revelou-se que o levamisol é eliminado do sangue do cachara em 12 horas, sendo a meia-vida de distribuição (t1/2?) de 0,10 horas, e para o intervalo de eliminação meia-vida (t1/2?) de 1,86 horas. / Continuous stress negatively influences the immune system of farmed fish, increasing diseases susceptibility. In such a context, preventive measures, such as the administration of immunostimulants, are better suited for diseases control than using drugs and chemicals as remedies. This study aimed to evaluate the effects of dietary immunostimulants (levamisole and vitamin E) as well as the pharmacokinetic study of levamisole in juvenile of Pseudoplatystoma sp. Dietary levamisole did not affect growth parameters. However, increased lysozyme activity and lysozyme concentration, the best effects were recorded for fish fed 247 mg levamisole per kg of feed. In the vitamin E assay, no differences in growth parameters were recorded. However, vitamin E levels positively influenced the innate immune system increasing serum globulins at dose of 166 mg kg-1 of DL-?-tocoferol acetate. Also, at pharmacokinetic study levamisole is shown to be completely eliminated from fish blood on a 12-h period, being the distribution half-life (t1/2?) of 0.10 hours, whilst for the elimination interval the half-life (t1/2?) was 1.86 hours.
