IMPORTANCIA DE LOS MICROORGANISMOS EN LA PRODUCCIÓN DE LANGOSTINOS MARINOS EN SISTEMAS DE BIOFLÓCULOS Y SIN RENOVACIÓN DE AGUA

Llario Sempere, Ferran 30 October 2018 (has links)
Microorganisms are essential for the intensive production of marine shrimps, since they are responsible for maintaining the water with an optimum quality for them. Also, these microorganisms minimize or eliminate water renewal. Among all the microorganisms present in intensive culture systems, without water renewal, microalgae and bacteria play the most important role and are the most abundant ones. In this thesis we analyze the microalgae dynamics using HPLC/CHEMTAX methodology in two intensive culture systems of marine shrimp, a shrimp nursery and a biofloc system. We also test the role of the bacteria Bacillus amyloliquefaciens as a probiotic and its capacity to improve the maturation process of the biofloc system. The results showed that the HPLC/CHEMTAX methodology is a fast and efficient way to analyze phytoplankton and periphyton in intensive culture systems of shrimp, provided that autotrophic processes are predominant. This technique allowed to detect prasinophytes and prymnesiophytes for the first time in the periphyton that grows on the polyvinyl chloride (PVC) tanks. Also, it has allowed to study the influence of trophic state and light exposition on microalgae dynamic in the biofloc systems. The application of Bacillus amyloliquefaciens in biofloc systems strengthens the immune system of shrimp and adds its probiotic effects to that produced by bioflocs. These benefits happen even with doses of 103 cfu/mL, lower than recommended doses for other probiotic bacteria. Other researches indicated that Bacillus amyloliquefaciens might have other effects such as participating in the biofloc system formation, improving water quality and shrimp growth; however, the application of spores in the culture water did not show any statistically significant effect on the system dynamic, water quality or zootechnic parameters. Plant colonization by GFP-labeled Bacillus amyloliquefaciens FZB42 and transcriptomic profiling of its response to plant root exudates

Fan, Ben 03 February 2011 (has links)
In this work colonization of three different plants genera, maize, Arabidopsis, and Lemna, by GFP-labeled Bacillus amyloliquefaciens FZB42 in a gnotobiotic system was firtly studied using confocal laser scanning microscopy and electron microscopy. It was shown that FZB42 is able to colonize all these three plants with a specific pattern. Root hairs and the junctions where lateral roots occurred were a preferred area of FZB42 on both maize and Arabidopsis seedlings. On Arabidopsis, tips of primary roots were another favored site of FZB42; while, on maize, the concavities in root surfaces were preferred. FZB42 cells were also able to colonize Lemna, preferably accumulating along the grooves between epidermis cells on roots and the concaved intercellular space on fronds. Secondly, microarray experiments were performed concerning the transcriptomic response of FZB42 to maize root exudates. A total of 302 genes representing 8.2% of FZB42 transcriptome were significantly altered in transcription by the presence of root exudates, the majority of them (260) were up-regulated in expression. The induced genes with known function were mainly involved in nutrition utilization, chemotaxis and motility, and antibiotic production. The transcriptome of seven FZB42 mutants, defective in five sigma factor genes (sigB, sigD, sigM, sigV, and sigX) and two global transcriptional regulator genes (degU and abrB), were also investigated through microarray experiments. A vast number of genes were indentified to be controlled by the protein factors respectively. Possible mechanisms were proposed of how these protein factors are involved in the response to root exudates. Finally, by northern blot existence of six out of 20 small RNA (sRNA) candidates was identified, which were significantly altered in expression by root exudates. This suggests that sRNA may play a hitherto unrecognized role in plant-microbe interaction.

Comprehensive proteomic study of Bacillus amyloliquefaciens strain FZB42 and its response to plant root exudates

Kierul, Kinga 19 August 2013 (has links)
Bacillus amyloliquefaciens strain FZB42 is a free-living bacterium that competitively colonizes plant roots and stimulates plant growth by many different modes of action. The molecular basis of singular beneficial effects that this Plant Growth-Promoting Rhizobacteria (PGPR) exert on their hosts have been studied. To decipher the molecular cross-talk of B. amyloliquefaciens and its’ host plants as a whole system, an extensive proteomic approach was performed. Reference maps of the extracellular and cytosolic protein fractions were established. The highest number of secreted proteins was observed during stationary growth phase. Identified extracellular proteins belong to different functional classes, with the most prominent classes involved in carbohydrate degradation and transportation of molecules across the cell wall. Cytosolic extracts obtained from cultures grown in 1C and minimal media subjected to the 2 Dimensional Electrophoresis (2 DE), revealed 461 and 245 different protein entries, respectively. Created reference maps were subsequently used to identify proteins and processes involved in the interaction with plants, prior to exposure of bacteria to maize (Zea mays L.) root exudates. The proteomics of two strains lacking expression of genes coding for global transcriptional regulators (degU, abrB) and four sigma factors (sigB, sigM, sigV, and sigX) were also inves-tigated, in order to analyse their involvement in bacterial responses to root exudates. In summary, this is the first study presenting comprehensive proteomics of Gram-positive PGPR, evaluating at the same time changes in protein expression caused by addition of root exudates at the extracellular and cytosolic level.

Regulation der Phytaseexpression in Bacillus amyloliquefaciens

Makarewicz, Oliwia 17 October 2006 (has links)
Several Bacillus strains secrete phytase, an enzyme catalysing dephosphorylation of myo-inositol hexakisphosphate (phytate). The monocistronic phyC (phytase) gene from environmental Bacillus amyloliquefaciens FZB45 was identified as a member of the phosphate-starvation inducible PhoPR regulon. The transcriptional start was determined downstream of a sigmaA-like promoter region located 27 bp upstream of the translation start codon. Inspection of the phyC promoter sequence revealed an unusual structure, since the -35 and -10 region are separated by a window of 21 bp. In vitro transcription analysis established that PhoP-P is necessary to initiate the transcription from phyC promoter. PhoP binding boxes occurring in most B. subtilis promoters activated by PhoP consist of at least four TTAACA-like sequences repeated at intervals of 5-6 bps. The upstream region of the B. amyloliquefaciens FZB45 phyC gene deviates from this general architecture in that there is only one appropriate binding site for the dimeric PhoP protein, which consists of two motives centered at -47 and -35 and separated by five base pairs. A single PhoP binding site is located at -13 to -8, nearly matching the -10 consensus. Functionality of the three PhoP binding boxes was demonstrated by DNaseI footprinting suggesting that a pair of dimeric PhoP molecules cover the -35 region directing the RNA-polymerase to the –10 region. Binding of PhoP at a single PhoP binding site covering the -10 consensus repress the transcription. It seems to be a unique feature of the phyC promoter structure and has not been reported for any other member of the PhoPR regulon, previously. Furthermore an inhibitory effect via AbrB, a global regulator, could be verified. The binding regions could be determine upstream of –147 and downstream of +29, but have to be specify in further experiments. This work presents for the first time the models for the regulation of phyC in Bacillus via PhoP and AbrB.

Effect of knock out of yxkO gene on environmental stress adaptation in genus Bacillus

Tkadlec, Jan January 2011 (has links)
We have previously characterized a Bacillus subtilis mutant defective in growth and osmoadaptation under limited K+ concentrations. In this mutant, the yxkO gene encoding a putative ribokinase is disrupted. This gene is supposed to belong to the sigma B operon and its expression is induced after osmotic, heat and ethanol shock. In comparison to the wild type, this mutation causes pleiotropic changes in host phenotype. In addition to its osmosensitivity, the mutant differs in cell shape, motility and ability to produce endospores. Our goal was to focus on manifestations of the mutation in the yxkO gene in other bacteria of the genus Bacillus. Using plasmid pMUTIN4 we have prepared mutants with disruptions of this gene derived from Bacillus amyloliquefaciens and Bacillus subtilis subsp. spizizenii strains differing in the yxkO surroundings and in the level of laboratory domestication. As in the previous study (with laboratory strain Bacillus subtilis 168) we demonstrate impaired ability of the mutant strain derived from Bacillus amyloliquefaciens to grow in potassium limitation and osmotic shock. We have studied this phenomenon at the level of the growth dynamics of the bacterial culture. We have also detected an increased sensitivity of the strain derived from Bacillus amyloliquefaciens to...

Environmentally friendly approach to postharvest quality maintenance of mango (Mangifera indica L.) cv. ‘Tommy Atkins’&‘Kent’

Van Deventer, Francois Johannes 14 February 2012 (has links)
The mango (Mangifera indica L.) is an appealing subtropical fresh fruit with a pleasant flavor and taste, high nutritional value, beneficial medicinal properties and various processing options. However, as is the case with most subtropical fruit, it is a sensitive commodity, prone to losses postharvestly. The South African mango industry is highly dependent on a hot water and cold prochloraz dip treatment, to control postharvest anthracnose and soft brown rot on fruit destined for export. However, negatve public perceptions of synthetic fungicides and its use on fresh produce for disease control has been increasing in major export markets such as the European Union. This growing concern from a public point of view is forcing industry to consider more environmentally acceptable methods to maintain quality of mangoes during extended export periods. ‘Tommy Atkins’ and ‘Kent’ mangoes either uninoculated or artificially inoculated with Colletotrichum gloesporioiedes, Botryosphaeria parva or sterile agar, were used to evaluate softer, greener alternatives, in this study. Fruit were subjected to either a hot, Bacillus amyloliquefaciens (PPCB004) containing dip treatment for two minutes or a 24 hour 1-methylcyclopropene (1-MCP) gas treatment at 16 ºC or no treatment. Fruit were then stored at 10 ºC under either 5% O2 and 5% CO2 (CA-1) or 3% O2 and 8% CO2 (CA-2) controlled atmospheres (CA) for 18 days and allowed to ripen for five days at 25 ºC. Similarly, uninoculated or artificially inoculated fruit subjected to B. amyloliquefaciens, 1-MCP or a combination of the two treatments was stored at 10 ºC for 18 days under conventional storage. ‘Tommy Atkins’ fruit were packed into bags made from four different film types, untreated or after being subjected to a cold B. amyloliquefaciens dip treatment and stored for 23 days at 10 ºC. Overall, ‘Kent’ fruit were more susceptible to anthracnose and SBR after artificial inoculation. In vivo inoculated ‘Tommy Atkins’ fruit, storage under CA-1 gave the best control of soft brown rot whilst CA-2 storage gave the best control of anthracnose. For quality retention no definite conclusion could be made for both cultivars after CA storage or the combination of 1-MCP pre-treatment and CA storage. The combination of 1-MCP pre-treatment and B. amyloliquefaciens maintained the quality of ‘Kent’ mangoes under conventional storage the best. Anthracnose severity on both cultivars was reduced with 1-MCP treated fruit combined with the biocontrol pre-treatment. Modified atmosphere packaging in this study was found to be ineffective in maintaining quality of mangoes.

Stavropoulou, Archontia 04 August 2005 (has links)
To find out the mode of tolerance increasing action against salinity of the plant root colonizing PGPR Bacillus subtilis, bacterial metabolites of the strains FZB24 and FZB41 were studied in the test plant tomato under the influence of high salinity. Because the culture filtrate with the whole range of produced metabolites by B. subtilis showed to a certain extent a tolerance increasing action at dilution of 0,1 % in axenic plant growth tests after 7 days treatment of seedlings and subsequent cultivation under salt stress, it has been fractionated with adsorber resin and HPLC. These fractions, as well as fractions from the culture filtrate after 19 h fermentation were tested also by seedlings in axenic culture. Fractions with different proteins and peptides, which were produced by B. subtilis, showed partly activities also depending of concentration with regard to the growth stimulation and at the same time tolerance increase against salt stress. Following also a peptide extract from B. subtilis was examined in the axenic plant test system, showing similarly a visible action depending of concentration. In the same test system there were tested further auxin precursors and auxin itself, which are known metabolites of B. subtilis, on seedlings both by root treatment and leaf treatment. Additionally was studied the action of auxins on the water content of the seedlings under salt stress, as well as on the adventitious root formation of hypokotyl segments from etiolated seedlings, in presence and absence of salinity. Finally it was studied the uptake and transport of auxins in segments of stems from etiolated seedlings in presence and absence of salinity. Lastly it was tested the action of auxins on plant growth and water content in a hydroponic cultivation under greenhouse conditions and salt stress. The results show that particularly auxin precursors and partly auxin as metabolites of B. subtilis can induce an increase in the salt stress tolerance of the test plant, although the action on the salt tolerance was differentiated and variable in its extent. The existing effect firstly of the auxin precursors is discussed as obviously main mechanism for the plant growth stimulating and at the same time tolerance increasing action of the rhizobacterium against salinity by root colonization and interaction with the plant metabolism.
