Controle alternativo da hérnia das crucíferas causada por Plasmodiophora brassicae em brócolis através de compostos orgânicos

No presente trabalho foram avaliados diferentes fatores que estão diretamente relacionados a este assunto. Desta maneira, foi demonstrado que sob condições controladas, a combinação de dois compostos orgânicos, CBL (Composto Bioativo Líquido) + CBS (Composto Bioativo Sólido) foi a melhor estratégia de controle da hérnia das crucíferas, com o CBL promovendo a supressividade da doença e o CBS contribuindo para o aumento de peso de parte aérea. Entretanto, sob condições de campo de produção comercial altamente infestado, 108 esporos/g de solo, estes compostos não reduziram o índice da doença. A combinação CBL + CBS associada com 400g de calcário/m2 de solo, sob a mesma densidade de inóculo, reduziu em 98% o índice da doença. Não houve redução significativa neste índice quando o calcário foi aplicado isoladamente ou em combinação com o NPK. Utilizando os mesmos adubos em sistemas de produção orgânico, integrado e convencional, sob plantios consecutivos, foi verificado que a redução do índice da doença promovida pelo CBL + CBS foi maior no sistema de produção orgânico, e que o histórico de produção afetou significativamente o índice da doença, o qual foi maior em solos sob sistema convencional. Mesmo em plantios consecutivos, onde a fonte de inóculo tende a aumentar a cada novo plantio, os tratamentos CBL e CBL + CBS reduziram o peso da hérnia fresca ao final de quatro plantios consecutivos, em relação ao controle, nos três sistemas de produção. A estrutura da comunidade bacteriana foi estudada e os perfis de PCR-DGGE obtidos com o uso de primers universais foram avaliados por meio de dendrogramas que mostraram diferenças na comunidade microbiana do solo com a adição de compostos orgânicos, quando comparada com fertilização química. A adição do composto orgânico CBL ao solo aumentou o índice de riqueza dos grupos específicos de Alphaproteobacteria, Betaproteobacteria e, principalmente, Actinobacteria e Pseudomonas, que são amplamente utilizados como agentes de controle biológico. O índice da hérnia das crucíferas e a diversidade bacteriana foram negativamente correlacionados para o domínio Eubacteria, as classes Alpharoteobacteria e Betaproteobacteria, e o grupo Pseudomonas. _______________________________________________________________________________ ABSTRACT / The clubroot caused by Plasmodiophora bassicae Woronin is one of the most difficult controls of plant diseases and the use of organic compounds is presented as an alternative for controlling because of the enrichment of the soil, contributing to improve their properties chemical, physical and biological resulting in suppression of soilborne diseases. In the present study were evaluated different factors that are directly related to this matter. Thus, it was demonstrated that under conditions of greenhouse, the combination of two organic compounds, CBL (bioactive compound liquid) + CBS (bioactive compound solid), was the best strategie for the control of the clubroot, with CBL promoting the suppressive effect of the clubroot and the CBS contributing to increase plant top weight. However under conditions of commercial production field highly infested, 108 spores/ g of soil, these compounds did not reduce the index of disease. The combination CBL+CBS associated with 400g of limestone/m2 of soil, under the same density of inoculum, reduced in 98% the disease index. There was no significant reduction in this index with the application of lime alone or in combination with NPK. Using the same fertilizer in organic production systems, integrated and conventional, under consecutive crops, has been found that the reduction of the index of the disease promoted by CBL + CBS was higher in organic production system. The historical of production affected the index of disease in which was higher in soils under the conventional system. Even in consecutive plantations where the source of inoculum tends to increase with each new planting, the treatments CBL and CBL + CBS reduced the fresh weight of galls at the end of four consecutive crops in relation to control in three production systems. The structure of the bacterial community was evaluated and the PCR-DGGE profiles obtained with the use of universal primers were evaluated by dendrograms that showed differences in soil microbial community with the addition of organic compounds compared with mineral fertilizers, corresponding to a higher richness of bacterial community. The addition of organic compound CBL increased the richness index of the specific groups of Alphaproteobacteria, Betaproteobacteria, Actinobacteria and Pseudomonas mainly that are widely used as biological control agents. The disease index of clubroot and bacterial diversity were negatively correlated for the domain Bacteria, the class Betaproteobacteria and Alpharoteobacteria, and Pseudomonas group.

Adubação fosfatada e potássica para brócolis e couve-flor em latossolo com alto teor desses nutrientes /

Na UNESP, Campus de Jaboticabal - SP, no período de 1-2 a 20-05-2010, quatro experimentos foram realizados com o objetivo de avaliar o efeito das doses 0, 80, 160, 240 e 320 kg ha-1 de P2O5, na forma de superfosfato triplo e 0, 50, 100, 150 e 200 kg ha-1 de K2O, na forma de cloreto de potássio, nas produtividades da couve-flor 'Verona' e do brócolis 'BRO 68', cultivados em Latossolo Vermelho Eutroférrico, com alto teor de P e K. Cada experimento foi instalado em delineamento de blocos ao acaso, com cinco tratamentos e quatro repetições. Para as duas brássicas avaliadas, foram constatados efeitos significativos das doses de P e K no teor de P e K no solo, diâmetro e massa da inflorescência e produtividade, com aumentos dos valores destas características mediante aplicação de P e K em relação à dose zero. Máximas produtividades de couve-flor (35.970 t ha-1) e de brócolis (11.425 t ha-1) foram obtidas com 245 e 320 kg ha-1 de P2O5, respectivamente e 12.476 kg ha-1 (Brócolis) com 160 kg ha-1 de K2O, e efeito linear para couve-flor com elevação das doses de potássio. Conclui-se que o brócolis e a couve-flor respondem ao fornecimento de fósforo e potássio mesmo em Latossolo com alto teor de P e K, e que as duas brássicas demandam quantidades diferentes de fósforo e potássio na adubação de plantio, a fim de maximizarem o diâmetro e a massa das inflorescências e suas produtividades / Abstract: Cauliflower and broccoli are two important vegetables, but there is little information about crop response to phosphorus and potassium fertilization when these nutrients are high in the soil. At UNESP, Jaboticabal - SP, Brazil, in the period of 2-1 5-20 2010, four experiments were carried out to evaluate the effect of doses 0, 80, 160, 240 and 320 kg ha-1 of P2O5 in the form of triple superphosphate and 0, 50, 100, 150 and 200 kg ha-1 of K2O in the form of potassium chloride, the yield of cauliflower 'Verona' and broccoli 'BRO 68', grown in dystrophic Eutrudox, with high levels of P and K. Each experiment was conducted in a randomized block design, with five treatments and four replications. For both species evaluated, were found significant effects of doses of P and K in P and K in the soil mass and diameter of the inflorescence and productivity, with increased values of these characteristics by applying P and K in relation to the zero dose. Para testar as hipóteses de que os efeitos promovidos pela adubação mineral sobre a produção, ganho de matéria seca, acúmulo e eficiência de utilização de nutrientes em cultivos sucessivos e características da solução do solo, podem ser igualados pela adubação verde (isolada ou em conjunto com adubo mineral), foram realizados experimentos. Os objetivos froam avaliar a resposta de crescimento, produção, ganho de matéria seca, acumulo e eficiência de utilização dos nutrientes nos cultivos sucessivos de brócolis e braquiária, manejados com mucuna cinza e adubação mineral aplicadas isoladamente ou em conjunto; avaliar as mudanças da acidez ativa, condutividade elétrica e teor de N, P, K, Ca, Mg e S da solução de solo com os diferentes tratamentos de adubação na primeira cultura; assim como, avaliar o efeito residual dos tratamentos sobre a produção de matéria seca e acúmulo de macronutrientes na braquiária e nas características da solução do solo no segundo cultivo. Similar efeito na produção, ganho de matéria seca, acumulo de nutrientes, eficiência de recuperação (ER) e eficiência de utilização (EU) em brócolis obteve-se com adubação verde (5 o 10 t/ha de mucuna cinza) e o controle na primeira cultura. Menor crescimento, produção, matéria seca e ER de macronutrientes obteve-se com adubação verde (5 o 10 t/ha de mucuna cinza) em comparação com adubação mineral na primeira culutra. Adubação verde em conjunto com adubo mineral promove crescimento de plantas, matéria seca, acúmulo, ER e EU de nutrientes semelhante ao obtido adubação mineral na alta dose (100% ou 200% da dose recomendada). Adubação verde (isolada ou em conjunto com adubação mineral) promovem efeito residual na segunda e demostram efeitos a mediano prazo. Maior condutividade elétrica e teor de N, K, Ca, Mg e S na solução do solo ocorre na etapa de crescimento inicial das culturas, maiores valores em solos adubados com fertilizante mineral na primeira cultura e maiores valores em solos adubados com adubação verde (isolado ou combinado) na segunda cultura. / Green manure can be an viable agronomically, economical, social and environmental alternative to partially or totally replace synthetic fertilizers. To test the hypotheses that the effects promoted by mineral fertilization on yield, dry matter gain, accumulation and nutrient utilization efficiency in successive crops and soil solution characteristics can be obtained by green manure (alone or combined), experiments were carried out with objectives to evaluate the response of growth, yield, dry matter gain, accumulation and utilization efficiency of macronutrients in the successive broccoli and brachiaria crops, managed with velvet-bean and mineral fertilization applied alone or in combination; to evaluate the changes of the active acidity, electrical conductivity and N, P, K, Ca, Mg and S contents of the soil solution with the different treatments of fertilization in the first crop; as well as to evaluate the residual effect of the treatments on dry matter production, accumulation and utilization efficiency of macronutrient and soil solution characteristics in the second crop (brachiaria grass). The effect on yield, dry matter gain, nutrient accumulation, recovery efficiency (RE) and efficiency of use (UE) in broccoli was obtained with green manure (5 to 10 t/ha of velvet-bean) and control in first crops. Lower growth, yield, dry matter and RE of macronutrients were obtained with green fertilization (5 or 10 t/ha of velvet-bean) compared to mineral fertilization in the first crop. Green manure in conjunction with mineral fertilizer promotes growth of plants, dry matter, accumulation, RE and UE of nutrients was similar to that obtained mineral fertilization at high dose (100% or 200% of the recommended dose). Green manure (alone or combined) promote residual effect in the second crop and demostred medium- term effects. Higher electrical conductivity and N, K, Ca, Mg and S content in the soil solution occurs in the initial growth stage of the crops, higher values in soils fertilized with mineral fertiilization in the first crop and higher values in soils fertilized with green manure (alone or combined) in the second culture.

O romanesco é uma hortaliça herbácea, com anatomia muito parecida à de brócolis e couve-flor, sendo mais tenra que a couve-flor. Foram realizados dois experimentos. No primeiro, objetivou-se avaliar a efetividade do uso de atmosfera modificada passiva na manutenção da qualidade pós-colheita do romanesco através das avaliações físico-químicas dos mesmos, mantidos sob refrigeração. No segundo, faz-se uma comparação entre as brássicas: romanesco, couve-de-folha, couve-flor, brócolis ramoso e brócolis de inflorescência única. No primeiro experimento foram realizados os seguintes tratamentos: T1: armazenamento em bandeja de poliestireno expandido sem filme; T2: armazenamento em bandeja de poliestireno expandido recoberta com filme de policloreto de vinila (PVC); T3: armazenamento em filme plástico de polietileno de baixa densidade (PEBD) de 0,6µ; T4: armazenamento em filme plástico de polietileno de baixa densidade (PEBD) de 0,8µ. Após a realização dos tratamentos os romanescos foram armazenados em câmara fria a temperatura de 5 ± 1 °C e UR entre 80 ± 5 % durante 16 dias, sendo avaliados de quatro em quatro dias. Utilizaram-se seis repetições por tratamento. Neste experimento foram realizadas as seguintes avaliações físico-químicas: teores de macronutrientes das folhas e inflorescências, perda de massa, sólidos solúveis, potencial hidrogeniônico (pH), acidez titulável, ácido ascórbico, açúcares redutores, clorofila a e b, antocianina e carotenoides. No segundo experimento foram avaliadas as características físico-químicas: número de folhas por planta; massa da matéria fresca das folhas; massa da matéria fresca da inflorescência; diâmetro transversal da inflorescência, circunferência da inflorescência, massa do ramo sem caule, sólidos solúveis, potencial hidrogeniônico (pH), acidez titulável, ácido ascórbico (exceto couve-de-folha), clorofila a e b, antocianina, carotenoides, açúcares redutores, proteína bruta, cinza, umidade, lipídeos e fibra bruta. Foi realizada análise de variância, teste de Tukey considerando-se um nível de significância p˂0,05, média e desvio padrão. Observou-se que para as características sólidos solúveis, pH, acidez titulável e ácido ascórbico, a utilização de filmes plásticos foi importante para a conservação e manutenção das características do romanesco, a partir do 12° dia de armazenamento, enquanto que para açúcares redutores, clorofila a e b, antocianina e carotenoides, foi importante em todos os tratamentos e dias de armazenamento. Para a perda de massa, a utilização de filmes plásticos foi essencial para a conservação do romanesco, possibilitando a viabilidade comercial do produto final. Para o número de folhas por planta; massa da matéria fresca das folhas; massa da matéria fresca da inflorescência; diâmetro transversal da inflorescência, circunferência da inflorescência e massa do ramo sem caule, as médias variaram, com exceção do número de folhas por planta e massa da matéria fresca das folhas do romanesco, quando comparado as demais brássicas. Para sólidos solúveis, pH, acidez titulável, pigmentos, açúcares redutores, proteína bruta, cinza, umidade, lipídeos e fibra bruta, foram observadas diferenças significativas entre as brássicas avaliadas. / The brassicas are vegetables of important economic value, as well as good source of minerals, vitamins and substances with anticarcinogenic properties. The romanesco is an herbaceous vegetable, with anatomy very similar to that of broccoli and cauliflower, being tenderer than cauliflower. Two experiments were carried out. In the first one, the objective was to evaluate the effectiveness of the use of passive modified atmosphere in the maintenance of post-harvest quality of the romanesco through the physical-chemical evaluations of the same, kept under refrigeration. In the second, a comparison is made between the brassicas: romanesco, cabbage leaf, cauliflower, branchy broccoli, and single inflorescence broccoli. In the first experiment the following treatments were performed: T1: tray storage of expanded polystyrene without film; T2: tray storage of expanded polystyrene coated with polyvinyl chloride (PVC) film; T3: storage of low density polyethylene (LDPE) film of 0.6μ; T4: low density polyethylene plastic film (LDPE) of 0.8μ. After the treatments, the romanescos were stored in a cold room at 5 ± 1 ° C and RH at 80 ± 5% for 16 days and evaluated every four days. Six replicates were used per treatment. In this experiment the following physical and chemical evaluations were performed: macronutrient contents of leaves and inflorescences, loss of mass, soluble solids, hydrogenation potential (pH), titratable acidity, ascorbic acid, reducing sugars, chlorophyll a and b, anthocyanin and carotenoids. In the second experiment the physical-chemical characteristics were evaluated: number of leaves per plant; mass of fresh leaf matter; mass of fresh inflorescence matter; (pH), titratable acidity, ascorbic acid (except leaf kale), chlorophyll a and b, anthocyanin, carotenoids, reducing sugars, crude protein , ash, moisture, lipids and crude fiber. A variance analysis was performed, Tukey's test considering a level of significance p˂0.05, mean and standard deviation. It was observed that for the soluble solid characteristics, pH, titratable acidity and ascorbic acid, the use of plastic films was important for the preservation and maintenance of the characteristics of the romanesco, from the 12 day of storage, whereas for reducing sugars, chlorophyll a and b, anthocyanin and carotenoids, was important in all treatments and storage days. For the loss of mass, the use of plastic films was essential for the preservation of the romanesco, allowing the commercial viability of the final product. For the number of leaves per plant; mass of fresh leaf matter; mass of fresh inflorescence matter; the transverse diameter of the inflorescence, the circumference of the inflorescence and the mass of the stem less branch, the averages varied, except for the number of leaves per plant and fresh matter mass of the leaves of the romanesco, when compared to the other brassicas. For soluble solids, pH, titratable acidity, pigments, reducing sugars, crude protein, ash, moisture, lipids and crude fiber, significant differences were observed among the evaluated brassicas.

Enfezamento do brócolis: identificação molecular de fitoplasmas, potenciais insetos vetores e hospedeiros alternativos, e análise epidemiológica da doença / Broccolo stunt: identification of phytoplasmas, potential insect vectors and alternative hosts and epidemiology of the disease

O brócolis (Brassica oleraceae var. italica) é uma das hortaliças mais importantes do país, cujo volume de comercialização na CEAGESP é de aproximadamente 13 mil toneladas por ano. Recentemente, uma nova doença tem causado perdas relevantes para as culturas instaladas na maior região produtora do Estado de São Paulo. Os sintomas característicos da doença são expressos pelo enfezamento da planta e necrose dos vasos de floema. Devido ao fato destes sintomas indicarem a presença de fitoplasmas nas culturas de repolho e couve-flor, localizadas na mesma região geográfica onde foi observada esta nova doença, levantou-se a suspeita de que estes mesmos agentes patogênicos pudessem estar associados com as plantas doentes de brócolis. Assim, o DNA total de plantas de brócolis sintomáticas foi analisado por PCR com primers específicos para a região 16S rDNA de fitoplasmas. Os resultados revelaram que estes patógenos estavam associados com as plantas doentes. Através das técnicas de RFLP do sequenciamento de nucleotídeos desta mesma região genômica, os fitoplasmas foram identificados como pertencentes aos grupos 16SrI, 16SrIII e 16SrXIII. Através de análise de RFLP, fitoplasmas também foram identificados em diversas espécies de plantas daninhas e em cigarrinhas da família Cicadellidae coletadas em áreas adjacentes a campos de produção de brócolis. Fitoplasmas do grupo 16SrIII foram identificados em plantas daninhas das espécies Agetarum conyzoides (mentrasto), Crotalaria lanceolata (crotalária), Lepidium virginicum (mentruz), Nicandra physalodes (juá-de-capote), Paulicourea marcgravii (erva-de-rato), Ricinus communis (mamona), Sida rhombifolia (guanxuma), Sonchus oleraceae (serralha amarela), Bidens pilosa (picão preto), Erigeron bonariensis (buva), Emilia sonchifolia (falsa serralha), Leonorus sibiricus (rubim), enquanto que fitoplasmas do grupo 16SrVII foram encontrados as últimas quatro espécies citadas. Com relação aos insetos, fitoplasmas foram detectados em indivíduos das subfamílias Deltocephalinae, Agalliinae e Typhlocybinae. Dentro da subfamília Deltocephalinae, a cigarrinha Balclutha hebe portava fitoplasma do grupo 16SrI, enquanto que cigarrinhas das espécies Atanus nitidus, Planicephalus flavicosta e Schapytopius fuliginosus abrigavam fitoplasmas do grupo 16SrIII. Nos tecidos de duas cigarrinhas da subfamília Agalliinae e uma da Typhlocybinae, as quais não foram identificadas quanto a espécie, foram encontrados fitoplasmas do grupo 16SrIII. As análises epidemiológicas revelaram um padrão espacial agregado de plantas doentes e a ocorrência de um maior progresso da doença nos bordos dos campos de cultivo de brócolis, que estão localizados nas proximidades de áreas com a presença de plantas daninhas. / Broccoli (Brassica oleraceae var. italica) is one of the most important vegetables in Brazil, whose trading volume in CEAGESP is approximately 13 000 tons per year. Recently, a new disease has caused significant losses in this crop cultivated in the largest producing region of the São Paulo State. The characteristic symptoms of the disease are expressed by plant stunting and necrosis of phloem vessels. Because these symptoms indicate the presence of phytoplasmas in cabbage and cauliflower crops, grown in the same geographical region, it was suspected that the same pathogens could be associated with the affected broccoli plants. Therefore, the total DNA from symptomatic plants of broccoli was analyzed by PCR with specific primers for the 16S rDNA of phytoplasmas. Through the techniques of RFLP and nucleotide sequencing of the same genomic region, the phytoplasmas were identified as belonging to the groups 16SrI, 16SrIII and 16SrXIII. Through RFLP analysis, phytoplasmas were also identified in several species of weeds and leafhoppers in the family Cicadellidae collected in adjacent areas of broccoli fields. Phytoplasmas belonging of the 16SrIII group were identified in the weeds belonging to the species Agetarum conyzoides, Crotalaria lanceolata, Lepidium virginicum, Nicandra physalodes, Paulicourea marcgravii, Ricinus communis, Sida rhombifolia, Sonchus oleraceae, Bidens pilosa, Erigeron bonariensis, Emilia sonchifolia, Leonorus sibiricus, while phytoplasmas of the 16SrVII group were found in the last four mentioned species. In respect to insects, phytoplasmas were detected in individuals from subfamilies Deltocephalinae, Agalliinae and Typhlocybinae. Within the subfamily Deltocephalinae, the leafhopper Balclutha hebe carried phytoplasmas of the 16SrI group, while that of the species Atanus nitidus, Planicephalus flavicosta e Schapytopius fuliginosus harbored phytoplasmas of the 16SrIII group. In the tissues of two leafhoppers of the subfamily Agalliinae and one of the Typhlocybinae, which were not identified as specie, were found phytoplasmas of the 16SrIII group. The epidemiological analysis revelead an aggregated pattern of the diseased plants and a higher progress of the diseased in the border of the broccoli fields, whitch were located nearby areas where the presence of weeds was abundant.

Avaliação da multiplicação de escherichia coli e staphyloccocus aureus em frutas e vegetais expostos a diferentes temperaturas e modelagem preditiva nos alimentos de maior risco

Este estudo teve como objetivo inicial avaliar a multiplicação de Staphylococcus aureus e Escherichia coli em frutas e vegetais expostos a diferentes temperaturas. Para identificar as frutas e vegetais frequentemente servidos em buffet, foram visitados restaurantes comerciais (n=50), onde os principais alimentos encontrados foram: cenoura ralada, brócolis, pepino, repolho verde, tomate, melancia e mamão. Amostras desses vegetais foram adquiridas em supermercado local e processadas ou preparadas conforme modo de consumo, sendo então contaminadas artificialmente com um pool de S. aureus e E. coli, separadamente, e expostos a 10, 20 e 30 °C. Os resultados desses experimentos demonstraram que não houve multiplicação dessas bactérias nas frutas e vegetais expostos a 10 °C durante 6 h. A 20 e 30 °C, S. aureus demonstrou multiplicação mais rápida no brócolis, onde a fase estacionária iniciou em menos de 2 h, possivelmente por este ser o único alimento cozido nesse estudo. Observou-se também que a 30 °C, E. coli se multiplicou em menos de 2 h nos seguintes alimentos: mamão, pepino, melancia e brócolis. Já no tomate, S. aureus não se multiplicou em nenhuma temperatura avaliada. No entanto, a população final de E. coli no tomate atingiu 9,7 log, em 24 h, a 30 °C, apesar do baixo pH (4,21). Por esse motivo e porque o tomate foi o vegetal mais frequentemente servido nos restaurantes comerciais avaliados, foi utilizado o modelo primário de Baranyi para modelar os parâmetros cinéticos de multiplicação e o modelo secundário de Ratkowsky para modelar a taxa de multiplicação e o tempo de fase lag em função da temperatura de E. coli no tomate, exposto a temperaturas de 10 a 37 °C. Os resultados obtidos indicaram que a fase lag da E. coli no tomate foi de 2,13 h e 2,46 h quando exposto a 37 e 30 °C, enquanto que a 20 e 10 °C, as fases lag foram de 15,6 h e 42,5 h, respectivamente. O modelo secundário foi integrado em uma simulação com dados nacionais de temperaturas reais coletados em planilhas de serviços de alimentação de 225 restaurantes de três regiões do Brasil (Sul, Sudeste e Norte/Nordeste). Aplicando o modelo gerado, foi observado que E. coli é capaz de se multiplicar em tomate em 1,58 h a 29,3 °C, temperatura mais crítica encontrada na cadeia de distribuição do tomate. Em seguida, realizou-se outro estudo no intuito de avaliar o comportamento de S. aureus em brócolis tratados termicamente, visto que este micro-organismo obteve um grande potencial de multiplicação nesse alimento. Também foram desenvolvidos modelos primário e secundário para avaliar a multiplicação do S. aureus em brócolis expostos a temperaturas de 10 a 37 °C. Nesse alimento, a fase lag de S. aureus foi de 1,4 h quando o vegetal foi exposto a 30 e 37 °C; enquanto que a 20 e 10 °C as fases lag foram de 5,3 h e 160 h, respectivamente. O modelo secundário foi capaz de descrever a influência da temperatura (de 10 a 37 °C) sobre a taxa de multiplicação e a fase lag de S. aureus em brócolis. Os resultados demonstraram que as frutas e vegetais avaliados podem ser distribuídas sob temperaturas de refrigeração de 10 °C ou menos e não devem ser mantidas mais de 2 h em temperaturas próximas de 30 a 37 oC, a fim de evitar a multiplicação bacteriana. Tais parâmetros podem contribuir na gestão de segurança dos alimentos em serviços de alimentação, prevenindo Doenças Transmitidas por Alimentos. / This study aimed to evaluate the multiplication of Staphylococcus aureus and Escherichia coli on fruits and vegetables exposed to different temperatures. The results of these experiments demonstrated that these microorganisms did not grow on fruits and vegetables exposed to 10 °C during 6 h. The secondary model was integrated in a simulation with national real temperature data collected in food service of 225 restaurants in three regions of Brazil (Southern, Southeast and North / Northeast). The lag phase of broccoli was 1.4 h when the bacteria was exposed at 30 and 37 °C; while at 20 °C and 10 °C the lag phases were 5.3 h and 160 h, respectively. Secondary models were able to describe the influence of temperature (10-37 °C) on the growth rate and lag phase of S. aureus on broccoli. The results demonstrated that the evaluated fruits and vegetables can be distributed under refrigeration temperatures of 10 °C or less and should not be maintained for longer than 2 h at temperatures close to 30 to 37 °C in order to avoid bacterial multiplication. Such parameters can contribute to the management of food safety in food services, preventing Foodborne Diseases.

However, the profitable crop production can be limited by occurrence of diseases such as Alternaria black spot, caused by Alternaria brassicicola and/or Alternaria brassicae, and black rot, caused by Xanthomonas campestris pv. campestris. In that context, the present study had as objectives to evaluate the intensity of Alternaria black spot and of black rot diseases on brassica under organic cultivation in the states of Pernambuco and Santa Catarina, and the spore production longevity of A. brassicicola in leaf debris of broccolis. The intensity of the diseases was evaluated from November 2001 to February 2002, in 103 fields under organic farming systems, including broccoli, Chinese cabbage, cauliflower, kale and cabbage. High prevalence of the diseases was registered in both states, except on Chinese cabbage in Santa Catarina. Prevalence of Alternaria black spot was 100% on broccoli fields in Pernambuco, as well as on cauliflower in the two states, while the black rot reached that level on broccoli and cauliflower fields in Santa Catarina. In the average of the different brassica species, the diseases were more prevalent in Pernambuco than in Santa Catarina. However, when considered severity averages of each disease, no significant differences were verified between the two states, although the climatic conditions have been sharply different. The levels ofdisease severity were low, considering that 98% of the fields presented severity lower than 5%. The Alternaria black spot severity varied among the brassica species in Pernambuco, being higher on Chinese cabbage and lower on kale. In Santa Catarina no significant differences were verified among the brássicas species. In relation to the black rot, only in Santa Catarina there was difference in the disease severity among the brassicas, being registered the lowest level on Chinese cabbage. No significant correlations were verified between severity levels of Alternaria black spot and black rot, neither between disease severity and total number of plants or plant age. The spore production longevity of A. brassicicola was investigated in leaves debris of broccoli, considering different depths of debris incorporation, periods of the year, and systems of soil handling, in two experiments carried out in Urussanga city, State of Santa Catarina, Brazil. Infected leaves debris of broccoli were conditioned in bags of polyethylene and distributed in portions in the field, on the soil surface and at depths of 5 and 10 cm. Periodically, the debris in the bags were collected and conidia concentrations were quantified for longevity of spore production andextinction rate. In both experiments there was higher spore production longevity in leaves debris in the period I, characterized by lighter temperatures, higher air relative humidity and lower pluvial precipitation than in the period II. Depth of debris incorporation influenced directly on A. brassicicola spores production, being lower when incorporation was made at 10 cm depth. In relation to soil handling systems, when the inoculum source was deposited on the soil surface there was not difference in the rate of spore production extinction between the soils under conventional and organic handling. However, at depths of 5 and 10 cm, the rate was significantly higher in the soil under conventional handling. The management of Alternaria black spot on broccoli at the area under study can be accomplished by integrated control, including the incorporation of infected leaves debris in the soil, at least 10 cm depth, along with crop rotation, for no less than a 60-day interval in subsequent brassica plantings. / O cultivo de brássicas tem destacada importância na olericultura brasileira. No entanto, a produção dessas olerícolas pode ser limitada pela ocorrência de doenças, dentre as quais se destacam a alternariose, causada por Alternaria brassicicola e/ou Alternaria brassicae, e a podridão negra, causada por Xanthomonas campestris pv. campestris. Nesse contexto, o presente estudo teve como objetivos realizar o levantamento da intensidade da alternariose e da podridão negra em cultivos orgânicos de brássicas nos Estados de Pernambuco e Santa Catarina, bem como avaliar a longevidade da esporulação de A. brassicicola em restos foliares de brócolis. Os levantamentos foram realizados no período de novembro de 2001 a fevereiro de 2002, num total de 103 cultivos orgânicos, incluindo brócolis, couve-chinesa, couve-flor, couve-manteiga e repolho. Foram registradas elevadas prevalências das doenças nos dois Estados, com exceção em couve-chinesa em Santa Catarina. A prevalência da alternariose foi de 100% nos cultivos de brócolis em Pernambuco, bem como em couve-flor nos dois Estados, enquanto a podridão negra atingiu esse nível nos cultivos de brócolis e couve-flor em Santa Catarina. Na média das diferentes espécies de brássicas, as doenças foram mais prevalentes em Pernambuco que Santa Catarina. Entretanto,quando consideradas as médias de severidade de cada doença no conjunto das brássicas, não foram constatadas diferenças significativas entre os dois Estados, embora as condições climáticas tenham sido nitidamente distintas. Os níveis de severidade das doenças foram baixos, tendo em vista que 98% dos cultivos apresentaram severidade inferior a 5%. A severidade da alternariose variou entre as espécies de brássicas em Pernambuco, com maior severidade registrada em couve-chinesa e menor em couve-manteiga, enquanto em Santa Catarina não foram constatadas diferenças significativas entre as brássicas. Em relação à podridão negra, somente em Santa Catarina houve diferença na severidade entre as brássicas, sendo registrados os menores níveis em couve-chinesa. Não foram constatadas correlações significativas entre os níveis de severidade da alternariose e da podridão negra, bem como da severidade destas com o número total de plantas e a idade das plantas nos cultivos. Foi também investigada a longevidade da esporulação de A. brassicicola em restos foliares de brócolis, considerando diferentes profundidades de incorporação dos restos, períodos do ano e sistemas de manejo do solo, em dois experimentos realizados em Urussanga - SC. Restosfoliares de brócolis infectados pelo patógeno foram acondicionadas em sacolas de polietilenoperfuradas e distribuídos em parcelas no campo, na superfície do solo e nas profundidades de 5 e 10 cm. Nos dois experimentos foi verificada uma maior longevidade da esporulação nos restos foliares no período I, caracterizado por temperaturas mais amenas, maior umidade relativa do ar e menor precipitação pluvial que no período II. A profundidade de incorporação dos restos influiu diretamente na esporulação de A. brassicicola, que foi inferior quando a incorporação foi efetuada a 10 cm de profundidade. Em relação aos sistemas de manejo, quando a fonte de inóculo foi depositada na superfície do solo não houve diferença na taxa de extinção da esporulação entre os solos sob manejo convencional e orgânico. No entanto, nas profundidades de 5 e 10 cm, essa taxa foi significativamente superior no solo sob manejo convencional. O manejo da alternariose em brócolis na região do estudo pode ser realizado pela combinação de vários métodos de controle, inclusive pela incorporação dos restos foliares infectados no solo, à profundidade mínima de 10 cm, combinado com rotação de culturas, visando um intervalo mínimo de 60 dias entre cultivos de brássicas.

Efeito do processamento na concentração de substâncias bioativas em brócolis e couve-flor

O brócolis e a couve-flor são vegetais que pertencem a família Brassicaceae e contém um complexo de substancias bioativas, como os compostos fenólicos, os carotenoides, os flavonoides, as clorofilas, vitaminas e minerais. O consumo destes vegetais está associado com a redução do risco de câncer, doenças cardiovasculares, catarata e outros distúrbios funcionais relacionados com a idade, por apresentar compostos bioativos. O descongelamento e o processamento são métodos que modificam as condições físicas e químicas e podem afetar a quantidade de compostos bioativos nos alimentos. Deste modo, este estudo teve como objetivo avaliar o efeito de diferentes condições de descongelamento (temperatura ambiente, temperatura refrigerada e micro-ondas) sobre o conteúdo de substâncias bioativas em brócolis (Brassica oleracea var. avenger); também avaliar o efeito do processamento (ebulição, vapor, micro-ondas e sous vide) na estabilidade de compostos fenólicos, flavonoides, carotenoides, clorofila, vitamina A e atividade antioxidante em inflorescências de brócolis e couve-flor cultivados em sistema orgânico. Os resultados mostraram que o descongelamento em micro-ondas reteve maiores conteúdos de vários compostos bioativos analisados, entre eles a clorofila, os compostos fenólicos e a quercetina. Observou-se que para ambas as hortaliças (brócolis e couve-flor) o processamento contribuiu, de alguma maneira, para o aumento do conteúdo de compostos antioxidantes, devido ao rompimento do tecido vegetal pelo aquecimento, mostrando uma maior capacidade antioxidante no processamento em sous vide para o brócolis e a couve-flor orgânica. / Broccoli and cauliflower are vegetables that belong to the Brassicaceae family and contains a complex of bioactive substances, such as phenolic compounds, carotenoids, flavonoids, chlorophylls, vitamins and minerals. The consumption of these vegetables is associated with reduce of the risk of cancer, cardiovascular disease, cataracts, and other functional disorders related to age, for presenting bioactive compounds. Thawing and processing are methods that modify the physical and chemical conditions and can affect the amount of bioactive compounds in foods. Thus, this study aimed to evaluate the effect of different thawing conditions (room temperature, refrigerated temperature and microwave) on the content of bioactive compounds in broccoli (Brassica oleracea var. avenger); also evaluate the effect of processing (boiling, steaming, microwave and sous vide) on the stability of phenolic compounds, flavonoids, carotenoids, chlorophyll, vitamin A and antioxidant activity in inflorescences of broccoli and cauliflower grown in organic system. The results showed that thawing in microwave retained higher contents of various bioactive compounds analyzed, including chlorophylls, phenolic compounds and quercetin. It was observed that both vegetables (broccoli and cauliflower) processing contributed in some way to increase the content of antioxidant compounds, due to disruption of the plant tissue by heating, showing a higher antioxidant capacity in sous vide process for organic broccoli and cauliflower.

Avaliação da multiplicação de escherichia coli e staphyloccocus aureus em frutas e vegetais expostos a diferentes temperaturas e modelagem preditiva nos alimentos de maior risco

Este estudo teve como objetivo inicial avaliar a multiplicação de Staphylococcus aureus e Escherichia coli em frutas e vegetais expostos a diferentes temperaturas. Para identificar as frutas e vegetais frequentemente servidos em buffet, foram visitados restaurantes comerciais (n=50), onde os principais alimentos encontrados foram: cenoura ralada, brócolis, pepino, repolho verde, tomate, melancia e mamão. Amostras desses vegetais foram adquiridas em supermercado local e processadas ou preparadas conforme modo de consumo, sendo então contaminadas artificialmente com um pool de S. aureus e E. coli, separadamente, e expostos a 10, 20 e 30 °C. Os resultados desses experimentos demonstraram que não houve multiplicação dessas bactérias nas frutas e vegetais expostos a 10 °C durante 6 h. A 20 e 30 °C, S. aureus demonstrou multiplicação mais rápida no brócolis, onde a fase estacionária iniciou em menos de 2 h, possivelmente por este ser o único alimento cozido nesse estudo. Observou-se também que a 30 °C, E. coli se multiplicou em menos de 2 h nos seguintes alimentos: mamão, pepino, melancia e brócolis. Já no tomate, S. aureus não se multiplicou em nenhuma temperatura avaliada. No entanto, a população final de E. coli no tomate atingiu 9,7 log, em 24 h, a 30 °C, apesar do baixo pH (4,21). Por esse motivo e porque o tomate foi o vegetal mais frequentemente servido nos restaurantes comerciais avaliados, foi utilizado o modelo primário de Baranyi para modelar os parâmetros cinéticos de multiplicação e o modelo secundário de Ratkowsky para modelar a taxa de multiplicação e o tempo de fase lag em função da temperatura de E. coli no tomate, exposto a temperaturas de 10 a 37 °C. Os resultados obtidos indicaram que a fase lag da E. coli no tomate foi de 2,13 h e 2,46 h quando exposto a 37 e 30 °C, enquanto que a 20 e 10 °C, as fases lag foram de 15,6 h e 42,5 h, respectivamente. O modelo secundário foi integrado em uma simulação com dados nacionais de temperaturas reais coletados em planilhas de serviços de alimentação de 225 restaurantes de três regiões do Brasil (Sul, Sudeste e Norte/Nordeste). Aplicando o modelo gerado, foi observado que E. coli é capaz de se multiplicar em tomate em 1,58 h a 29,3 °C, temperatura mais crítica encontrada na cadeia de distribuição do tomate. Em seguida, realizou-se outro estudo no intuito de avaliar o comportamento de S. aureus em brócolis tratados termicamente, visto que este micro-organismo obteve um grande potencial de multiplicação nesse alimento. Também foram desenvolvidos modelos primário e secundário para avaliar a multiplicação do S. aureus em brócolis expostos a temperaturas de 10 a 37 °C. Nesse alimento, a fase lag de S. aureus foi de 1,4 h quando o vegetal foi exposto a 30 e 37 °C; enquanto que a 20 e 10 °C as fases lag foram de 5,3 h e 160 h, respectivamente. O modelo secundário foi capaz de descrever a influência da temperatura (de 10 a 37 °C) sobre a taxa de multiplicação e a fase lag de S. aureus em brócolis. Os resultados demonstraram que as frutas e vegetais avaliados podem ser distribuídas sob temperaturas de refrigeração de 10 °C ou menos e não devem ser mantidas mais de 2 h em temperaturas próximas de 30 a 37 oC, a fim de evitar a multiplicação bacteriana. Tais parâmetros podem contribuir na gestão de segurança dos alimentos em serviços de alimentação, prevenindo Doenças Transmitidas por Alimentos. / This study aimed to evaluate the multiplication of Staphylococcus aureus and Escherichia coli on fruits and vegetables exposed to different temperatures. To identify the fruits and vegetables most frequently served in buffet, commercial restaurants (n=50) were visited, where the main foods found were: grated carrots, broccoli, cucumber, green cabbage, tomato, watermelon and papaya. Samples of these vegetables were purchased from the local supermarket and processed or prepared according to the mode of consumption and were then artificially contaminated with a pool of S. aureus and E. coli separately and exposed at 10, 20 and 30 ° C. The results of these experiments demonstrated that these microorganisms did not grow on fruits and vegetables exposed to 10 °C during 6 h. At 20 and 30 ° C, S. aureus showed faster multiplication on broccoli, where the stationary phase started in less than 2 h, possibly because this was the only food cooked in that study. For this reason, and because tomato was the most frequently served food in the evaluated commercial restaurants, the Baranyi primary model was used to model the kinetic parameters of multiplication and the Ratkowsky secondary model to model the multiplication rate and lag phase time as a function of the temperature of E. coli on tomato, which was exposed to temperatures of 10 to 37 °C.The results indicated that the lag phase of E. coli on tomato was 2.13 h and 2.46 h when exposed at 37 and 30 °C, respectively; while at 20 °C and 10 ° C the lag phases were 15.6 h and 42.5 h, in that order. The secondary model was integrated in a simulation with national real temperature data collected in food service of 225 restaurants in three regions of Brazil (Southern, Southeast and North / Northeast). The results demonstrated that the evaluated fruits and vegetables can be distributed under refrigeration temperatures of 10 °C or less and should not be maintained for longer than 2 h at temperatures close to 30 to 37 °C in order to avoid bacterial multiplication. Such parameters can contribute to the management of food safety in food services, preventing Foodborne Diseases.
