Elucidating the genetic basis of bast fibre production in flax (Linum usitatissimum L.)

Flax is often considered a total utilization crop because of the potential to extract value from two distinct products - seeds and stem fibres. However, very little genetic information is available on flax fibre genetics in comparison to oil improvement studies. In order to gain a detailed understanding of genetic control of the fibre concentration and search for the possibilities of developing dual purpose flax lines using both seed oil and stem fibre, the following studies were initiated: The first study evaluated the fibre and oil-related traits in a recombinant inbred population derived from a cross between a fibre flax variety Viking and an oilseed flax genotype E1747 over multiple locations under western Canadian field conditions. The study confirmed the presence of a significant genotype by environment interaction (p < 0.01) for fibre concentration indicating selection for this trait will be challenging. However, a lack of significant correlation between fibre and oilseed characteristics in field trials was encouraging and strengthened the hypothesis that breeding dual purpose flax types for western Canada is possible. The study also identified potential recombinant inbred lines (RILs) with enhanced fibre concentration as well as oil characteristics for use in future breeding endeavors. The second study established an anatomical basis for further research into flax fibre improvement by studying differences between the stem anatomy of 14 diverse flax genotypes in the field and under controlled environments such as a growth chamber. The results from the study supported the use of controlled environments for the purpose of quick screening of high fibre containing genotypes, especially at the green capsule stage of plant growth. The results also indicated that it was possible to select high fibre oilseed flax lines based on anatomical markers such as average area of single fibre cells, total fibre area and fibre to stem area ratio. In the third study, 17 simple sequence repeat (SSR) and 2 cleaved amplified polymorphic sequences (CAPS) molecular markers were used to assess the extent of genetic variability in the Viking × E1747 RIL population. CAPS markers LuFAD3A and LuFAD3B had the highest marker trait association (p < 0.0001) with linoleic and linolenic acid concentration. SSR markers such as CV8824, 5B6 and LU32 were found to be associated with plant height, oil concentration and protein concentration respectively using single marker analysis and step wise regression analysis. The molecular study confirmed the importance of Viking × E1747 mapping population in identifying genes/ markers related to both fibre and oilseed related traits in flax. In the fourth study, global transcript profiling using cDNA - based microarrays was performed to identify differentially expressed fibre related transcripts between Viking and E1747. The largest group of transcripts (7 %) found more abundant in Viking relative to E1747 fell under the functional group of cell wall development using gene ontology (GO) analysis. Transcripts such as callose synthases, expansins, cytochrome P450, fasciclin-like arabino galactan proteins and β-galactosidases were highly abundant in Viking relative to E1747. The transcripts more abundant in E1747 relative to Viking were UDP – glucose glucosyltransferase, auxin repressed protein, ubiquitin conjugating enzyme, peroxidases and lipid transfer proteins. Quantitative real time PCR results confirmed the suitability of the microarray platform to accurately discriminate transcript profiles between the two diverse flax types. In conclusion, this research has provided a number of new insights into flax fibre genetics. This information lays the foundation for further genetic studies on flax bast fibres and will complement research on developing dual purpose flax varieties.

Shear cracking in inverted-T straddle bents

Significant diagonal cracking in reinforced concrete inverted-T (IT) straddle bent caps has been reported throughout the State of Texas. Many of the distressed structures were recently constructed and have generally been in service for less than two decades. The unique nature of the problem prompted a closer look into the design and behavior of such structural components. A preliminary investigation highlighted outdated design requirements and a scarcity of experimental investigations pertaining to inverted-T bent caps. This research project (TxDOT Project 0-6416, Shear Cracking in Inverted-T Straddle Bents) aims to improve current understanding of the behavior of inverted-T caps, while providing updated design provisions. In order to develop strength and serviceability guidelines for inverted-T beams, an extensive experimental program was developed. This series of large scale tests was used to evaluate the strength and serviceability of IT deep beams in relation to the following parameters – shear span-to-depth (a/d) ratio, web reinforcement ratio, ledge height, ledge length, number of point loads, and member depth. This report focuses mainly on results from a first series of tests conducted within this experimental program. / text

Examining The Impact of Florida's Non-Economic Damage Cap on Elderly Populations

In this paper, I use data from the Florida Closed Claims Database to investigate how Florida’s 2003 non-economic damage cap legislation impacted elderly malpractice claimants. More specifically, I measure whether or not non-economic damage caps adversely impact claimants in counties with high elderly densities. To measure the effect of Florida’s non-economic damage caps, I look at multiple metrics that measure both elderly claimants’ monetary gains and their access to the justice system after the reform is passed. I find mildly conclusive evidence that counties with higher elderly density, and assumedly more elderly claimants, are more likely to settle cases before reaching a jury trial and are less likely to file a medical malpractice claim. Conversely, though, I find limited evidence supporting the idea that elderly claimants receive less monetary damage payments or drop cases more. Overall, then, my findings are not consistent with the view that non-economic damage caps significantly discriminate against elderly claimants.

A partir das experiências clínicas conduzidas nos CAPS, a psicanálise traz para a discussão a tensão existente nesse campo entre a clínica e a política, destacando a importância da articulação e do enlace entre os saberes para o exercício da clínica nesse espaço, porém poucas evidências desse diálogo são encontradas, principalmente no que diz respeito ao atendimento de crianças. Assim, a presente pesquisa pretende analisar como o discurso analítico na clínica com crianças pode se articular com os outros discursos presentes em um CAPS i e contribuir na direção do tratamento, à luz de recortes clínicos do caso de um menino de nove anos atendido por mim. Para tanto, examinou-se a especificidade da clínica psicanalítica com crianças; identificou-se o lugar da criança nas estruturas discursivas presentes nos laços sociais, tomando como a referência a teoria dos quatro discursos de J. Lacan, a fim de: analisar os discursos e práticas presentes no CAPS i; observar o tratamento dado ao sintoma e o manejo da transferência realizado pelo discurso do analista em meio aos demais discursos em um CAPS i; e, por fim, examinar a articulação entre os discursos na direção do tratamento de uma criança em um CAPS i. Esse estudo teve como referencial teórico a psicanálise e utilizou como método a construção de caso clínico. A realização dessa pesquisa torna-se relevante por possibilitar ampliar a discussão sobre crianças em sofrimento psíquico na saúde pública e seu tratamento em um CAPS i, deslocando o eixo discursivo do transtorno ou deficiência para o campo da estruturação subjetiva na interface dos campos da psicanálise, saúde mental e psiquiatria, fornecendo subsídios para a sustentação da teoria e da prática psicanalítica com crianças na saúde pública. / In the psychoanalytic work with children placed into Psychosocial Care Center for children and youth, CAPS i, the challenges of the psychoanalytic clinic itself, with the real of the psychosis and neurosis taking places in the severe cases of psychoanalytical, social and economic helplessness, associated to the difficulties of the job in a collective institution, convokes the psychoanalyst to question the functionnalty of clinical tools in this multidisciplinary field. On the bases of clinical experiences conducted in the CAPSs, the psychoanalysis brings to the discussion the conflict that exists between the clinic and the politic, pointing out the importance of the articulation between the knowledges that makes possible the exercise of the clinic in this space. However, there are a few evidences of this dialogue, especially in regards to the child’s assistance. Therefore, the present research intends to analyse how the analytic discourse about the child clinic can be articulated with others discourses presents in a CAPS i and contributes on the direction of the treatment, based on the clinical case of a nine years old boy which was my patient. To reach this goal, the specifies of the child analytic clinic was examined and the place of the children in the discourse’s structures in the social bond was identified, based on the Theory of four discourses of Jacques Lacan, intending to analyse the discourses and practice in CAPS i according to the Athos case; to analyse the treatment given to the symptom and to the management transfer in the analyst's discourse in between the others discourses in a CAPS i; and. finally, to exam the articulation between the discourses in the way to the treatment of a child in a CAPS i. This study is based on the psychoanalysis’s theory and will have as a method the construction of a clinical case. This research is relevant because it provides an ample discussion about children in public care who suffer psychology and their treatment in a CAPS i, shifting the focus of the discussion from the deficiency to the field of the subjective structuring in the different interfaces of psychoanalysis, mental health and psychiatry, hence, providing evidence to support the theory and psychiatric practice with children in public health.

Trata-se de um estudo qualitativo, com o referencial da Análise Institucional e inspiração na Etnografia, tendo como sujeitos participantes da pesquisa, quatro usuários com diagnóstico de psicose e filiação religiosa às igrejas pentecostais, seus familiares e equipe técnica do CAPS. Neste estudo, parte-se do entendimento da loucura como uma experiência disruptiva singular e plural, cuja processualidade, movimento incessante e potência instituinte, podem ser capturados, estagnados, assumindo o caráter de “doença/ sofrimento”. A complexidade que permeia a experiência da loucura, ao longo da história, aponta para o seu entrelaçamento com a vida, com a cultura e a produção de subjetividade. Neste entrelaçamento, o encontro com a religião aparece de modos distintos e contraditórios, tais como forma significativa de entendimento do mundo e da vida, espaço que promove a inclusão e suporte social, e pode atuar como agência terapêutica, que faz parte do itinerário terapêutico de pessoas em situação de dor e sofrimento. Parte-se do pressuposto de que a relação entre o CAPS - dispositivo estratégico substitutivo ao modelo manicomial, articulador de uma de rede de atenção integral em saúde mental, e a religião, se situa num campo de forças, enquanto modos de produzir saberes e fazeres sobre a loucura, sobre e com o louco. Esta tensão parece se acentuar na relação com determinadas religiões, como as pentecostais, que concebem a loucura como possessão demoníaca, o que mobiliza rituais de exorcismo, como fora percebido com os dados produzidos nesta pesquisa. Apesar da disputa entre modos de entender e lidar com a loucura, se constatou a existência de uma mesma lógica manicomial que perpassa a instituição religiosa e o CAPS, que produzem subjetividades manicomiais. Assim como se constatou que essas instituições, em alguns casos, se configuram como modos importantes de inclusão social e produção de novos sentidos para a experiência da loucura. / Salvador
