Soares, Andrey Coatrini 16 February 2017 (has links)
The need of analytical devices to detect cancer at early stages has motivated research into nanomaterials where synergy is sought to achieve high sensitivity and selectivity in biosensors. In this work, 11-mercaptoundecanoic acid, the polymer chitosan and the protein concanavalin A (Con A) were used as a platform to immobilize the anti-CA 19-9 antibody using the self-assembled monolayer (SAM) and the layer-by-layer (LbL) techniques. The characterization with spectroscopic and gravimetric techniques allowed us to select the architectures with chitosan/Con A 2:1 (Sensor A), chitosan/Con A 1:1 (Sensor B) and 11 MUA (Sensor E) as optimized for immobilization of anti-CA 19-9 antibodies. Using impedance spectroscopy, the biosensors were capable of detecting low concentrations of CA 19-9 biomarker, with limit of detection in the range 0.17-0.69 U/mL, 0.31-0.91 U/mL and 0.56-0.91 U/mL for sensors A, B and E, respectively. These limits are sufficient to detect pancreatic cancer at early stages. The selectivity of the biosensors was inferred in a series of control experiments with cell samples SW-620 and HT-29, uric acid, ascorbic acid, glucose, mannose, serum and p24 antigen, indicating the absence of non-specific interference. With information visualization techniques, these samples could be easily distinguished in a visual map, and be classified according to their content of CA 19-9. Furthermore, the selectivity could be quantified through the silhouette coefficient, with values 0.853, 0.861 and 0.897 for sensors A, B and E, respectively. This biosensor specificity was confirmed with PM-IRRAS measurements by monitoring the amide I and II bands at 1566 cm-1 and 1650 cm-1. The specific interaction between antibody and antigen was modeled with a Langmuir-Freundlich isotherm. When the chitosan/Con A matrix was replaced by a SAM monolayer or if a larger biomarker was employed, adsorption was explained by a combination of two Langmuir-Freundlich processes. In conclusion, low cost biosensors may be effective for diagnostics and prognostics, and may be further implemented in the Brazilian national health system with technology transfer.

Hernández Bueno, Ania Yarasech 06 1900 (has links) (PDF)

Bonum non est in deo: On the Indistinction of the One and the Exclusion of the Good in Meister Eckhart

King, Evan 24 August 2012 (has links)
Meister Eckhart exhibits an unprecedented confidence in the transcendental way of thought in medieval philosophy. Eckhart, unlike his predecessors, identifies being as such (ens commune) and God, allowing the most primary determinations metaphysics – ‘being,’ ‘one,’ ‘true,’ ‘good,’ – to function as both metaphysical and theological first principles. Eckhart placed them at the head of his projected Tripartite Opus, a vast work of quaestiones and commentaries whose intelligibility, he insists, requires the prior foundation of a supposed series of a thousand axioms. The table of contents remains, the opus propositionum does not. This thesis argues that what enables Eckhart to pursue the direct application of the transcendentals to the divine also makes it unrealizable. His determination of unity is twofold: as (i) indivisibility, and the standard transcendental conception of unity as a negation of the given positive content of being (ens); as (ii) indistinction, comprehending both the negation of otherness which produces the indivisible and the otherness that is negated. There is an inherent tension between Peripatetic metaphysics and Procline henology. Consequently, the Good is devalued when the Procline One appears within the transcendental perspective. Metaphysics, theology and, a fortiori for Eckhart, ethics, take no consideration of Goodness. I show how this tension gives rise to Eckhart’s association of the divine essence with the Neoplatonic One, while the Peripatetic One and the transcendental “true” function as the explanans of the Trinitarian intellectual self-return. This, in turn, gives rise to the constitutive function of the imago dei, and every imago as such, within that self-relation. Ultimately, this produces a standpoint wherein every essence, only as idea, contains the divine uniform infinity.

Poiesis : an Eriugenian interpretation of Chaucer's Troilus and and Criseyde

Logan, Frank Daniel Hermitage January 1990 (has links)
This thesis deals with the interpretation of art, set against the background of the medieval Christian Neoplatonism of John Scotus Eriugena. For him, art and philosophy are regarded as the handmaidens of meaning. Therefore, although this thesis begins with a consideration of Eriugena's Periphyseon, it develops into a discussion on aesthetic theory, and ultimately into one on poetic theory. The object of this discussion is to account for meaning in Chaucer's Troilus and Criseyde according to Eriugenian poetics. / The essence of art will thus be pursued within the parameters of the Neoplatonic scala natura. In this way, the whole poetic interpretation of Chaucer's poem is grasped as a mirror of the ontological exitus-reditus pattern. In understanding the poem this way, this thesis comes to immediate terms with the medieval concept of the imago Dei, and understands the likeness of mankind to God to be primarily one made by virtue of language.
