Bioacumulação e biomagnificação de metais pesados em teias tróficas de estuários do sul-sudeste do Brasil / Bioaccumulation and biomagnification of heavy metals in trophic webs of estuaries of the southern and southeastern of Brazil

A região costeira do Brasil é historicamente impactada por atividades antrópicas, que liberam contaminantes, como os metais pesados. Nos estuários de Paranaguá (PR), Cananéia e Santos (SP), tais atividades estão presentes em diferentes níveis. Neste estudo foram investigadas as concentrações de arsênio, cádmio, chumbo, cobre, cromo, níquel, selênio, zinco e mercúrio, além de isótopos estáveis de carbono e nitrogênio, em sedimentos, fragmentos vegetais e organismos marinhos, como invertebrados bentônicos, peixes bentívoros e mamíferos marinhos, coletados nos três estuários. As amostragens foram realizadas em 2015, sendo as análises de isótopos realizadas por EA-IRMS e as análises de metais pesados realizadas por ICP OES-VGA e ICP-MS. As razões isotópicas permitiram a distinção entre fontes de matéria orgânica e nível trófico. Exemplares da ictiofauna e de cetáceos analisados não apresentaram distinção entre os níveis tróficos 3 e 4, indicando uma dieta generalista para estes animais e potencial compartilhamento de nicho trófico. Os metais apresentaram maiores concentrações nos sedimentos do estuário de Santos, coincidindo com o local com maior intensidade de atividades antrópicas. Comparando os estuários há maior acumulação de: Cr, Cu, Zn e Hg em Paranaguá, As e Pb em Cananéia, e Cd, Ni e Se em Santos, resultados justificados por atividades antrópicas, fontes naturais e características geoquímicas de cada região. A tendência de biomagnificação de Se foi observada nas teias tróficas de todos os estuários, e de Zn e Hg nas teias de Paranaguá e Cananéia. Entretanto, As, Cr, Cu, Ni e Pb demostraram tendência a biodiluição nas teias tróficas estudadas, com maior bioacumulação em invertebrados bentônicos. Portanto, este estudo traz um panorama atual da distribuição espacial e trófica de metais pesados nos estuários, colaborando com o monitoramento e ações de gestão costeira. / The coastal region of Brazil is historically impacted by anthropic activities, which release contaminants such as heavy metals. In the estuaries of Paranaguá (PR), Cananéia and Santos (SP), these activities are present in different levels. In the present study, concentrations of arsenic, cadmium, lead, copper, chromium, nickel, selenium, zinc and mercury were investigated, as well as stable carbon and nitrogen isotopes in sediments, plant fragments and marine organisms such as benthic invertebrates, benthivorous fish and marine mammals, collected in the three estuaries. Samples were collected in 2015, with isotope analysis performed by EA-IRMS and heavy metal analysis performed by ICP OES-VGA and ICP-MS. Isotopic ratios allowed the distinction between organic matter sources and trophic level. The specimens of the ichthyofauna and cetaceans analyzed did not present distinction between trophic levels 3 and 4, indicating a generalist diet for these animals and a potential sharing of trophic niche. The metals have presented higher concentrations in the sediments from Santos estuary, what coincides with the place with greater intensity of anthropic activities. Comparing the estuaries, there is more bioaccumulation of Cr, Cu, Zn and Hg in Paranaguá, As and Pb in Cananéia, and Cd, Ni and Se in Santos; results which are justified by anthropic activities, natural sources and geochemical characteristics of each region. A biomagnification tendency of Se was observed in the trophic webs of all estuaries, and Zn and Hg in the Paranaguá\'s and Cananéia\'s webs. However, As, Cr, Cu, Ni and Pb displayed a tendency to biodilution in the studied trophic webs; with a higher bioaccumulation in benthic invertebrates. Therefore this study provides a current overview of the spatial and trophic distribution of heavy metals in the estuaries. Thus, it is collaborating with the monitoring and actions of coastal management.

Reconstruction et étude de la variabilité du régime alimentaire des sociétés préhispaniques de la basse vallée de Santa, Pérou

Ce projet de recherche porte principalement sur la reconstruction du régime alimentaire à l’aide d’une approche paléochimique. Des analyses isotopiques du carbone de l’azote ont été réalisées sur le collagène des os sur un échantillon de trente-huit individus provenant de trois sites différents de la basse vallée de Santa, sur la côte nord du Pérou : El Castillo, Guadalupito et Huaca China. Parmi les individus sélectionnés, certains sont affiliés aux groupes des Gallinazo, des Mochica, des Tanguche ou des Chimú couvrant ainsi la Période intermédiaire ancienne, l’Horizon moyen et la Période intermédiaire récente. L’approche isotopique a été utilisée afin de caractériser dans un premier temps, le régime alimentaire de la population globale en la comparant à d’autres groupes préhispaniques de l’Aire andine. Les résultats obtenus sur trente-deux individus suggèrent une alimentation variée incluant le maïs ainsi que des ressources marines, illustrant que ces groupes agriculteurs exploitaient les deux types d’écosystèmes à leur portée. Ces résultats sont supportés par l’analyse du carbone sur l’apatite pour cinq individus. Le second but fut d’étudier la variabilité du régime alimentaire entre les différents individus de l’échantillon, en fonction de la période d’occupation, du site de provenance, de l’affiliation culturelle, de l’âge et du sexe et du statut social. La petite taille de l’échantillon a limité considérablement la discussion de ces différents paramètres. Il a toutefois été possible de remarquer un accès aux ressources riches en protéines plus avantageux pour les hommes et la présence de possibles différences dans l’alimentation chez les individus juvéniles. Finalement, la confrontation des données funéraires à la reconstruction du régime alimentaire a été faite pour cinq cas particuliers de pratiques funéraires provenant du site El Castillo, premier centre régional de la civilisation Mochica dans la basse vallée de Santa. Dans tous les cas, le régime alimentaire suggéré par les compositions isotopiques supporte les interprétations des statuts sociaux de ces individus faites à partir des contextes funéraires. / This research focuses on the reconstruction of diet using a chemical approach. Carbon and nitrogen isotopes were analyzed on a bone collagen sample of thirty-eight individuals from three different sites of the Lower Santa valley, on the north coast of Peru : El Castillo, Guadalupito and Huaca China. Among those individuals selected, some are affiliated with groups of Gallinazo, Moche, Tanguche and Chimu, dated to the Ancient Intermediate Period, Middle Horizon and Recent Intermediate Period. The isotopic approach has been used to characterize in a first step, the diet of the entire population by comparing with some other prehispanic groups from the Andean area. The results of thirty-two individuals suggest a mixed diet including maize and marines resources, showing that these agriculturist groups exploited their surrounding ecosystems. These results are supported by the carbon analysis on bone apatite for five individuals. The second aim was to study the variability of the diet between individuals in the sample, depending on the time period, site, cultural identity, age and sex and social status. The small sample size has considerably limited the discussion of these parameters. However, it was possible to note a more preferential access to high proteins resources for men and some possible differences in diet among juveniles. Finally, the comparison between dietary reconstruction and funerary patterns was made for five cases of burial pratices at El Castillo site, the first Moche regional center in the Lower Santa valley. In all cases, diet suggested by isotopic compositions is consistent with interpretations of the funerary context about the individual status.

In addition to presenting ecological importance, since these enzymes are involved in biological control, through the process of mycoparasitism. The gender Trichoderma comprises a group of filamentous fungi, saprophytic soil, found on decomposing organic matter in the rhizosphere, and some plants. Considering the importance of β-1.3 glucanases proposing in this study to evaluate the production of β-1.3 - glucanases by T. asperellum using different sources of carbon and nitrogen. The enzyme was produced using cell wall of Rizoctonia solani (PCRS), chitin, chitosan, starch, cellulose, sucrose, maltose, lactose, celobiose and glucose. The best production of enzymes was obtained in the middle containing PCRS, with two bands of activity of polyacrylamide gel were found. Furthermore, the effect of varying concentrations of ammonium sulfate (2, 4, 6, 8, 20, 40 and 60 mM) in the production of β-1.3-glucanase was evaluated during the growth of the fungus in PCRS. A great increase in activity of the enzyme was detected in the presence of nitrogen source. We can see that there was a considerable increase in the expression of the enzyme of low molecular weight at this culture conditions. / As b-1,3-glucanases apresentam diversas funções na célula fúngica como metabólica, na hidrólise de polissacarídeos para eventual assimilação celular; função morfogenética, na hidrólise ou modificações da parede das células, que inclui o crescimento e extensão da parede, alterando sua estrutura e sua composição e autólise. Além de apresentar importância ecológica estas enzimas estão envolvidas no controle biológico, através do processo de micoparasitismo. O gênero Trichoderma compreende um grupo de fungos filamentosos saprófitas de solo, encontrados sobre matéria orgânica em decomposição e na rizosfera de algumas plantas. Considerando a importância das b-1,3-glucanases propõe-se neste trabalho a avaliação da produção de b-1,3-glucanases por T. asperellum utilizando diferentes fontes de carbono e nitrogênio. A enzima foi produzida utilizando parede celular de Rizoctonia solani (PCRS), quitina, quitosana, amido, celulose, sacarose, maltose, lactose, celobiose e glicose. A melhor produção de enzimas foi obtida n o meio contendo PCRS, sendo detectadas duas bandas de atividade em gel de poliacrilamida. Além disto, o efeito de concentrações variadas de (NH4) 2SO4 (2, 4, 6, 8, 20, 40 e 60 mM) na produção de b-1,3-glucanases foi avaliado durante o crescimento do fungo em PCRS. Um aumento considerável na atividade da enzima foi detectado na presença desta fonte de nitrogênio. Pode - se observar que houve um aumento considerável na expressão da enzima de menor massa molecular nestas condições de cultivo.

The influence of biological relationships on the morphology of the human skeleton is a current theme in bioarchaeology and forensic research. Whether it is the discovery of kinship relationships in an anonymous cemetery or the question of individual identification based on familial similarity, research is limited by a number of factors. Above all, there is the shortage of osteological assemblages with genealogical documentation. Most such samples also include only a small number of individuals, and thus the number of different degrees of their relatedness is limited. Related to this is the lack or absence of methods suitable for either the exact expression of the degree of morphological similarity between individuals or the statistical evaluation of results. However, for this dissertation an opportunity arose to analyze several osteological assemblages with genealogical documentation from the 17th to 20th centuries, comprising almost one hundred individuals. This represents one of the largest such units in the world to date. The individual samples also include several cases of consanguineous marriages with subsequent generations. Storage of Pine Tree Substrate Influences Plant Growth, Nitrification, and Substrate Properties

Pine tree substrate (PTS) is a relatively new substrate for container crop production. There are no detailed studies that elucidate how storage time impacts PTS chemical, physical, and biological aspects. The objective of this research was to determine how PTS storage time influenced PTS chemical and physical properties, nitrification, and plant growth. Pine tree substrate was manufactured by hammer-milling chips of loblolly pine trees (Pinus taeda L.) through two screen sizes, 4.76 mm (PTS) and 15.9 mm amended with peat (PTSP). PTS and PTSP were amended with lime at five rates. A peat-perlite mix (PL) served as a control treatment. Prepared substrates were placed in storage bags and stored in an open shed in Blacksburg, Virginia. Subsamples were taken at 1, 42, 84, 168, 270, and 365 days. At each subsampling day, twelve 1-L containers were filled with each substrate. Six containers were left fallow and six were planted with marigold (Tagetes erecta L. "Inca Gold") seedlings. Substrate was also collected from select treatments for Most Probable Number assays to estimate density of nitrifying microorganisms, and for chemical and physical property analyses. Pour-through extracts were collected from fallow containers at 0, 2, and 4 weeks, and from marigold containers at harvest for determination of pH, electrical conductivity, ammonium-N and nitrate-N. At harvest, marigold height, width, and dry weight were measured. At least 1 kg·m-3 lime for PTS, and 2 to 4 kg·m-3 lime for PTSP were needed to maintain pH values ≥ 5.5 for 365 days. Bound acidity of unlimed PTS increased but cation exchange capacity for unlimed PTS and PTSP decreased over 365 days. Carbon to nitrogen ratio and bulk density values were unchanged over time in all treatments. There were minor changes in particle size distribution for limed PTS and unlimed and limed PTSP. Marigold growth in PTS and PTSP was ≥ PL in all limed treatments, except at day 1. Nitrite-oxidizing microorganisms were present and nitrification occurred in PTS and PTSP at all subsampling days. Pine tree substrate is relatively stable in storage, but pH decreases, and lime addition may be necessary to offset this decrease. / Ph. D.

Two-Dimensional Conjugated Metal-Organic Frameworks for Electrocatalysis

A highly effective electrocatalyst is the central component of advanced electrochemical energy conversion. Recently, two-dimensional conjugated metal-organic frameworks (2D c-MOFs) have emerged as a class of promising electrocatalysts due to their advantages including 2D layered structure with high in-plane conjugation, intrinsic electrical conductivity, permanent pores, large surface area, chemical stability, and structural diversity. In this review, we summarize the recent advances of 2D c-MOF electrocatalysts for electrochemical energy conversion. Firstly, we introduce the chemical design principles and synthetic strategies of the reported 2D c-MOFs, as well as the functional design for the electrocatalysis. Subsequently, we present the representative 2D c-MOF electrocatalysts in various electrochemical reactions, such as hydrogen/oxygen evolution, and reduction reactions of oxygen, carbon dioxide and nitrogen. We highlight the strategies for the structural design and property tuning of 2D c-MOF electrocatalysts to boost the catalytic performance, and offer our perspectives in regard to the challenges to be overcome.

Applying isotope geochemistry to identify mechanisms regulating the aquatic-terrestrial carbon and nitrogen dynamics across scales in a moraine landscape

In dieser Studie wurden stabile Isotopenverhältnisse genutzt um die Mechanismen der aquatisch-terrestrischen C – und N-Dynamiken über verschiedene Skalenebenen hinweg in der Moränenlandschaft von Nordostdeutschland zu identifizieren; einer Landschaft, die stark landwirtschaftlich genutzt wird und in der es eine Vielzahl von kleinen Wasserkörpern (Sölle) gibt. Auf der regionalen Landschaftsskala spiegeln d13C-Isotopenkarten des org. Materials in Oberböden und von Pflanzenblättern eines 38.2 km2 großen Gebietes den Eintrag von org. Material von C3-Pflanzen, deren Wassernutzungseffizienz im org. Material des Bodens eingeprägt wurde, sowie den Eintrag von Mais (C4-Pflanze), wider. Die d15N-Isotopenkarte des org. Materials in Böden weist verschiedene Düngepraktiken hin. Auf der regionalen Sollskala deuten die d13C- und d15N-Isotopenwerte von Oberflächensedimenten von 51 Söllen auf kürzliche Einträge des org. Materials und Bewirtschaftungseffekte im Einzugsgebiet hin. Tiefere Sedimente sind durch die Ablagerung org. Materials von terrestrischen Pflanzen sowie dessen Umsetzungsgrad geprägt in Abhängigkeit von der Wasserführung. Auf der Transekt-Skala, d.h. entlang von Transekten von Erosions- zu Depositionsgebieten im Einzugsgebiet eines Solls, beeinflussen Erosion, Pflanzenproduktion, mikrobielle Umsetzung und Gülledüngung verschiedene Fraktionen des org. Materials. Auf der Aggregat-Skalenebene sind die Art und der Anteil spezifischer organo-mineral Assoziationen entlang des Transekts variabel. Bodenpartikel vom Feld und hereinwachsende Makrophyten sind die Quellen des org. Materials in Sedimenten. Diese Studie hat erfolgreich stabile Isotopenverhältnisse zur Identifikation von Mechanismen der C- und N-Dynamik auf individuellen Skalenebenen angewendet. Kleine Inlandwasserkörper sind Schlüsselelemente für die C- und N-Dynamik in landwirtschaftlich genutzten Moränenlandschaften. / In moraine landscapes, carbon (C) and nitrogen (N) dynamics can vary greatly across landscape structures and soil types especially when small water bodies are interspersed in the landscape. This study used stable isotope ratios to identify the mechanisms regulating the aquatic-terrestrial C and N dynamics across different scales in the young moraine landscape of NE Germany – a landscape intensively used for agriculture and interspersed with countless of small water bodies, the so-called kettle holes. At the regional landscape scale, d13C isoscapes of topsoil bulk soil organic matter (SOM) and plant leaves collected from a 38.2 km2 area revealed long-term inputs OM from C3 crops, which imprinted their water use efficiency status onto the soil, as well as short-term inputs from corn. The d15N SOM isoscape identified fertilization-induced impacts on the N dynamics of agricultural fields and grasslands. At the regional kettle hole scale, d13C and d15N of surface sediments of 51 kettle holes reflected recent OM inputs and management practices in the catchment. Deeper sediments recorded the degree to which the OM has been processed within the kettle hole depending on the water-logging period. At the transect scale, erosion, plant productivity, microbial decomposition and slurry fertilization affected OM fractions in topsoil along transects spanning erosional to depositional areas in the catchment of one arable kettle hole. At the aggregate scale, the pathway and magnitude of OM-mineral associations changed along the transect. OM in sediments was derived from clay- and silt-sized particles from the field, together with emergent macrophyte contributions. By means of stable isotopes techniques, different mechanisms were identified at the individual scales. Consideration of the spatial heterogeneity of all scales is essential to understand landscape C and N dynamics. Small inland water bodies are key constituents of C and N dynamics in moraine agricultural landscapes.

L'objectif de cette thèse est l'étude du régime alimentaire de poissons côtiers commerciaux et la compréhension de l'utilité des isotopes stables du carbone (δ13C) et de l'azote (δ15N) dans l'étude des niches trophiques et de l'occupation des habitats côtiers par les poissons dans le sud-ouest de l'océan Indien. Dans la Baie de La Possession, les espèces représentatives des récifs artificiels immergés en 2003 (Luljanus kasmira, Priacanthus hamrur et Selar crumenophthalmus) ingèrent surtout des proies pélagiques et partitionnent leurs niches trophiques afin de limiter la compétition. Les valeurs de δ15N sont positivement corrélées avec la taille des poissons, traduisant une contribution croissante des larves de poissons au régime alimentaire. Les isotopes stables de la matière organique particulaire (MOP) ont été analysés à microéchelle (< 10 km) dans la Baie de La Possession, à mésoéchelle (10aines de km) autour de La Réunion et, pour les muscles de poissons à macroéchelle (l00aines de km) le long,du plateau .des Mascareignes.et du cana1 du Mozambique. À microéchelle, le δ15N de la MOP ne varie pas; à mésoechelle, Il existe un enrichissement significatif en 15N dans le sud-ouest de l'île; à macroéchelle, les muscles des poissons s'enrichissent progressivement en 15N vers le nord. À microéchelle, la MOP est enrichie en 13C à faible profondeur; à mésoéchelle, les stations de l'est sont enrichies en 13C ; à macroéchelle, aucune tendance n'est observée. Ainsi, l'utilité des isotopes stables du carbone pour l'étude de l'utilisation des habitats côtiers est limitée à méso- et macroéchelle et les isotopes stables de l'azote constituent un outil plus pertinent à macroéchelle / The aim of this thesis was to study the feeding habits of coastal fish species of commercial interest andto use stable isotopes of carbon (δ13C) and nitrogen (δ15N) to investigate the trophic niches of fish and their use of coastal habitats in the south-west Indian Ocean. In the Bay of La Possession, the species among the most abundant around the artificial reefs immersed in 2003 (Luljanus kasmira, Priacanthus hamrur and Selar crumenophthalmus) feed essentially on pelagic prey and partition their trophic niche to limit interspecific competition. There was also a positive correlation between fish size and muscle δ15N values due to fish larvae contributing more to the diet of fish with age. The stable isotopie composition of the particulate organic matter (POM) was measured at the microscale (< 10 km) in the Bay of La Possession, the mesoscale (10's of km) around Reunion Island and, for the fish muscles, at the macroscale 100’s of km) along the Mascarene plateau and the Mozambique channel. At microscale, POM δ15N values do not show any spatial variability; at mesoscale, POM is 15N-enriched in the south-west of Reunion Island; at macroscale, fish muscles are gradually 15N-enriched towards the north. At microscale, POM shows a 13C-enrichment on the shallowest station; at mesoscale, stations on the east coast of the island are 13C-enriched compared to the west; at macroscale, POMδ13C values are not significantly correlated with latitude. Thus, the stable isotopes of carbon do not seem to be appropriate for the study of fish coastal habitat use at the meso- and macroscale, and the stable isotopes of nitrogen represent a better indicator of fish habitat use at the macroscale.
