Caractérisation d'une nouvelle voie de biogenèse des petits ARN / Characterisation of a new small RNA biogenesis pathwayHallais, Marie 17 November 2010 (has links)
Les mécanismes régissant le trafic et la localisation des ARN à l'intérieur du noyau sont encore mal compris. La protéine PHAX (Phosphorylated adaptator for RNA export) est un facteur nécessaire à l'export nucléaire des précurseurs des snARN, ainsi qu'à l'adressage des précurseur des snoRNA coiffés vers les corps de Cajal (Ohno et al., 2000 ; Boulon et al., 2004 ). De manière remarquable, la liaison de PHAX à ces ARN est strictement dépendante de leur taille, et celle-ci est perdue au-delà d'une certaine longueur d'ARN (Ohno et al., 2002). Cependant, in vitro, PHAX est capable de s'associer aux ARN, mais il le fait sans spécificité de séquence ni de taille. L'étude des mécanismes impliqués dans la distinction des petits ARN non-codants des grands ARN comme les ARN messagers, est malgré tout essentielle car elle dirige ces ARN vers des voies de maturation différentes. Lors de ma thèse j'ai mis en place un projet qui aborde deux questions : (i) déterminer comment le facteur PHAX sélectionne les petits ARN, (ii) caractériser l'ensemble des ARN liés à PHAX dans le but de découvrir de nouveaux petits ARN non-codants. J'ai observé que PHAX forme un complexe avec le CBC, comme attendu, et avec une protéine non caractérisée à l'époque, et que nous avons nommée PBP1 (PHAX binding protein 1). J'ai pu déterminer que ces quatre protéines, CBC20-CBC80-PHAX-PBP1 forment un complexe, que j'ai appelé complexe CBCAP. In vivo, ce complexe est bien associé avec les précurseurs de snARN et snoARN. De façon très intéressante, le complexe CBCAP est aussi associé à de nombreux ARNs messagers de petite taille (inferieurs à 1500pb). De manière intéressante, les ARN liés par le CBCAP sont également parmi les plus abondants dans la cellule. L'expression de ces ARN est de plus affectée suite à la déplétion des protéines PHAX ou PBP1, et, de manière surprenante, ces protéines ont souvent des effets opposés sur la stabilité de ces ARN. PHAX est préférentiellement requis pour la production des snRNA, tandis que PBP1 est plutôt nécessaire à la production des snoRNA coiffés et des ARN messagers. Le complexe CBCAP pourrait donc sélectionner les ARN selon leur taille et il distinguerait plusieurs types d'ARN (codants ou non-codants; exportés ou retenus), af in qu'ils soient orientés vers la voie de maturation adéquate. De manière remarquable, la production de PBP1 est elle-même contrôlée par des signaux de croissance cellulaire. Ainsi, PBP1 permettrait de réguler l'expression des ARN les plus abondants dans la cellule, et jouerait ainsi un rôle clef dans le contrôle de la croissance cellulaire. Au cours de ma thèse, j'ai aussi observé par analyse tilling array, la co-précipitation avec PBP1 et de PHAX avec des petits ARN non annotés. J'initie la caractérisation de ces nouveaux ARN. / RNA localisation determines the fate of RNA. However inherent transport mechanisms still remain unclear. The protein PHAX is required for snRNA and snoRNA maturation as it both exports precursors of snRNA to the cytoplasm via the recruitment of the CRM1 export machinery, and transports precursors of capped snoRNA to the cajal bodies. Interestingly, PHAX associates with and transports mainly small RNA through a mechanism that is not specific of the RNA sequence. To determine how PHAX recognises specifically small RNA, we characterised the PHAX native complex. We purified a complex that we called CBCAP, which contains the CBC (cap binding complex), the protein that we called PBP1 (PHAX binding protein 1) and the transporter PHAX. As expected, this complex is associated with precursors of snRNA and snoRNA. Interestingly, we also co-purified with CBCAP small coding RNA (under 1500bp). Furthermore, expression of these RNA is affected by PBP1 and PHA X depletion. Surprisingly, these two proteins have an opposite effect on RNA stability. PHAX is required for snRNA expression, whereas PBP1 seems to control capped snoRNA and small mRNA expression. The complex CBCAP might thus be involved in the distinction of RNA size and seems to discriminate coding from non-coding RNA in order to address them to the appropriate processing pathway.

Unsettling the White Noise: Deconstructing the Nation-Building Project of CBC Radio One’s Canada ReadsBurns, EMILY 15 August 2012 (has links)
The Canadian Broadcasting Corporation’s Canada Reads program, based on the popular television show Survivor, welcomes five Canadian personalities to defend one Canadian book, per year, that they believe all Canadians should read. The program signifies a common discourse in Canada as a nation-state regarding its own lack of coherent and fixed identity, and can be understood as a nationalist project. I am working with Canada Reads as an existing archive, utilizing materials as both individual and interconnected entities in a larger and ongoing process of cultural production – and it is important to note that it is impossible to separate cultural production from cultural consumption. Each year offers a different set of insights that can be consumed in their own right, which is why this project is written in the present tense. Focusing on the first ten years of the Canada Reads competition, I argue that Canada Reads plays a specific and calculated role in the CBC’s goal of nation-building: one that obfuscates repressive national histories and legacies and instead promotes the transformative powers of literacy as that which can conquer historical and contemporary inequalities of all types. A pesquisa examina o modelo de comunicação pública adotado pela CBC/Radio-Canada e as principais discussões nacionais e internacionais sobre a missão da mídia de serviço público no século XXI em busca de subsídios à construção de um modelo de serviço público adequado ao Brasil, com sustentabilidade financeira, autonomia editorial e administrativa, gestão profissional e compromisso social. O modelo jurídico-institucional da CBC/Radio-Canada é semelhante ao da Empresa Brasil de Comunicação (EBC), criada em 2008 com a proposta de prestação de serviço público apesar de fortemente ligada ao governo federal. Tal ligação suscita dúvidas quanto à possibilidade de uma empresa de comunicação atrelada ao Estado oferecer conteúdos sem ingerência política, com uma pauta voltada ao interesse coletivo e com gestão profissional, livre de paternalismos. Por meio de revisão de literatura e de entrevistas estruturadas com oito especialistas canadenses em mídia pública, conclui-se, a partir da experiência da CBC/Radio-Canada, que o modelo estatal pode ser eficaz desde que haja vontade política e grande vigilância por parte da sociedade. Mesmo assim, é um modelo imperfeito e vulnerável às alterações do clima político-partidário. Recomenda-se o aperfeiçoamento do modelo por meio de um maior envolvimento da sociedade nos conselhos administrativos e curadores. Conclui-se, ainda, a partir dos novos paradigmas da mídia de serviço público, que, no novo cenário midiático de múltiplas ofertas e múltiplos canais, os investimentos do Estado em comunicação só se justificam se as empresas públicas apresentarem conteúdo distinto do oferecido pelas comercias. The CBC/Radio-Canada´s juridical and institutional model is similar to the one adopted by the Empresa Brasil de Comunicação (EBC), founded in 2008 to offer distinctive public service despite strong links with the federal government. Such liaison brings doubts whereas is possible for a media company connected to the State to offer programs without political interference, with public interest content and professional management, free of protégées. Through literature revision and structured interviews with eight Canadian public media specialists and from the CBC/Radio-Canada´s experience, this research arrived to the conclusion that the state model can be efficient if there are political will and strong society´s vigilance. Even so the model is imperfect and vulnerable to the political mood. We suggest the model improvement through stronger society´s participation on the Directors Board. It is also possible to conclude that, in a media environment of multiple channels and multiple offers, the States´ investment in communication is only justified if the public media provides distinguished program. A close partnership with the social movements and the transformation of the public broadcasting service in a democratic and plural forum open to the debate of subjects neglected by the mainstream companies is appointed as a viable alternative for the survival and empowerment of the public broadcasting service.

Previous issue date: 2014-12-04 / A presente dissertação, desenvolvida no Programa de Pós-graduação Profissional em Gestão e Avaliação da Educação Pública (Mestrado Profissional) da Universidade Federal de Juiz de Fora, versa sobre a implementação do Programa de Intervenção Pedagógica / Currículo Básico Comum (PIP/CBC), da Secretaria de Estado da Educação de Minas Gerais, como estratégia para melhoria da aprendizagem do aluno e da consequente elevação dos índices dos resultados das avaliações externas em uma escola estadual da região metropolitana de Belo Horizonte. O objetivo da investigação foi elucidar, no contexto escolar, a dinâmica de aplicação do PIP/CBC, apontando os entraves na consolidação deste e as dificuldades na elevação dos índices de desempenho alcançados nas avaliações externas com vistas à proposição de um Plano de Intervenção que possa auxiliar a escola na superação desses obstáculos. A pesquisa amparou-se na análise documental referente ao PPP da escola, estudo da legislação que trata do tema e entrevistas estruturadas com profissionais ligados ao projeto e à escola. Os dados coletados na pesquisa revelaram indícios de que o PIP/CBC realmente não alcançou os resultados esperados na escola estudada, considerando os objetivos propostos na sua implementação. Os principais elementos que impedem o sucesso do programa estão relacionados à falta de diálogo entre a escola e as diretrizes norteadoras do programa, ao acompanhamento ineficiente dos trabalhos desenvolvidos em sala de aula e aos gargalos apresentados na gestão escolar, especialmente na gestão pedagógica. Nesse cenário, apresentou-se um Plano de Intervenção, buscando ações que propiciem um diálogo maior com as diretrizes norteadoras do PIP/CBC e, consequentemente, a melhoria na aprendizagem dos alunos, bem como a elevação dos índices de proficiência nas avaliações externas. / This thesis focuses on the implementation of the Programme of Educational Intervention / Common Basic Curriculum (PIP / CBC), the State Department of Education of Minas Gerais, as a strategy for improving student learning and consequent elevation of indices of the results of external assessments. For this, concentrates the research outline in a state school in the metropolitan region of Belo Horizonte. The research objective was to elucidate, in the school context, the dynamics of implementing PIP / CBC, pointing obstacles in consolidating the same and the difficulties in raising the levels of proficiency attained in external evaluations, with a view to proposing an Intervention Plan that can help the school in overcoming these obstacles. The methodological approach is bolstered in document analysis, interpretation of legislation that addresses the theme and structured interviews with professionals working in the public state of Minas Gerais. The data collected in the survey revealed evidence that the PIP / CBC does not really achieve the expected results at the school , considering the objectives proposed in its implementation. The main elements of incumbent success of the program are related to lack of knowledge of the guiding principles of the program, to inefficient monitoring of work done in the classroom and the bottlenecks presented in school management, especially in educational management. In this scenario, he presented a plan of intervention, seeking actions that foster greater dialogue with the guiding principles of the PIP / CBC and consequently the improvement in student learning, as well as rising rates of proficiency in external evaluations.

Investigating the role of extended CBC complexes in RNA metabolism / Etude du complexe CBC et ses différents rôles dans la biogenèse des ARNBenbahouche, Nour el Houda 10 December 2015 (has links)
Le CBC intervient dans de nombreuses étapes du métabolisme des ARN, telle que l’épissage, la maturation de l’extrémité 3’, la dégradation, l’export et la traduction. Ainsi, le CBC constitue un complexe majeur qui peut orchestrer les différentes étapes de maturation des ARN. Récemment, nous avons identifié le complexe CBCAP, composé de CBC, ARS2 et PHAX. Nous avons montré que la protéine ARS2 stimule la formation des extrémités 3’ de plusieurs familles d’ARN dont les snARN. De plus, ARS2 stimule le recrutement de PHAX sur le CBC. Ainsi, nous proposons un modèle où CBC-ARS2 stimule la formation de l’extrémité 3’ des pré-snARN et recrute PHAX pour favoriser leur export. Une autre étude a identifié un autre complexe le CBCN, constitué de CBC, Ars2, et de ZC3H18-NEXT au lieu de PHAX. CBCN recrute l’exosome et stimule la dégradation de certains ARN, comme les PROMPTS et les transcrits «read-through » des snARN et des ARNm d’histone. Ainsi, PHAX et ZC3H18 destinent leur ARN cibles vers l’export ou la dégradation. Il a été montré que PHAX reconnait et lie spécialement les ARN de petite taille. D’une manière remarquable, nos données de CLIP-Seq et de RIP suivie par des analyses avec des puces « All genes » montrent que PHAX lie aussi d’autres familles d’ARN. En effet, PHAX lie les ARNm ainsi que des ARN non-codant avec une légère préférence pour les snARN (en comparaison avec ZC3H18). Afin de mieux comprendre le rôle de PHAX et ZC3H18, j’ai tout d’abord démontré si les deux protéines se lient simultanément au CBC. Pour ce faire, J’ai réalisé des tests de compétitions entre PHAX et ZC3H18, in vivo, et j’ai montré que la surexpression de ZC3H18 déplace PHAX du CBC et vice versa. Puis en utilisant la technique de « Tethering Assays » j’ai pu montrer que PHAX et ZC3H18 ont des effets opposés sur la biogénèse des ARNm. De plus PHAX semble avoir un effet positif sur la maturation des ARNm et ce, en empêchant ZC3H18 et l’exosome d’être recruter. Nous avons aussi montré que la déplétion de PHAX et ZC3H18 a des conséquences fonctionnelles sur le taux des formes matures des snARN. Dans le but de caractériser la protéine ZC3H18, j’ai réalisé un crible double-hybride et j’ai montré que ZC3H18 interagit avec plusieurs facteurs d’épissage. J’ai aussi identifié les domaines de ZC3H18 impliqués dans ses différentes interactions. D’une manière intéressante, l’interaction de ZC3H18 avec certains facteurs d’épissage peut être exclusive à son interaction avec NEXT. De plus, des expériences de protéomique réalisés sur un des facteurs d’épissage trouvé dans le crible, montrent qu’il co-purifie au sein d’un complexe qui pourrait faire le lien entre la coiffe et la machinerie d’épissage. En accord avec ces résultats, nos données de RNA-seq montrent que la déplétion de ZC3H18 engendre un défaut d’épissage pour des introns qui sont proches de la coiffe et ceci pour un nombre restreint de gènes. Ainsi, notre travail décode davantage le rôle de la coiffe dans les différentes étapes de maturation des ARN et suggère un modèle où la séquence des transcrits naissant stimule la formation d’un complexe spécifique à cet ARN parmi plusieurs autres. / The cap binding complex (CBC) plays a key role in a number of gene expression pathways and has been proposed to participate in the discrimination of RNA families. It also enhances many RNA processing steps, including transcription, splicing, 3’end formation, degradation, export and translation.Recently, we identified the CBCAP complex, composed of CBC, Ars2 and PHAX. We showed that Ars2 stimulates snRNA 3'-end processing as well as PHAX binding to the CBC, hence coupling snRNA maturation with their export. Other studies showed that the CBC and ARS2 can form another complex that contains ZC3H18-NEXT instead of PHAX. This complex, named CBCN, is a cofactor of the RNA exosome and is involved in the degradation of cryptic RNAs such as PROMPTs and read-through transcripts at histone and snRNA genes. Thus, PHAX and ZC3H18 target specific families of capped RNA toward either export or degradation. Previous studies proposed that PHAX binds specifically to small RNAs and discriminates them over other RNA species. Surprisingly, our CLIP-Seq and RIP-microarrays data showed that in contrast to expectations, PHAX was not specific for snRNAs. It also binds mRNAs as well as other non-coding RNAs and has a weak preference for snRNAs comparing to ZC3H18. To better understand the role of PHAX and ZC3H18, Ifirst determined whether PHAX and ZC3H18 can bind simultaneously to the CBC. Competitive LUMIER IPs indicated that binding of these proteins is mutually exclusive. I then used tethering assays and could show that PHAX and ZC3H18 have opposite effect on mRNA biogenesis. These data go against a model where binding of PHAX or ZC3H18 discriminate RNA families, and instead suggest promiscuous binding for these proteins. In addition, PHAX may exert a positive effect on mRNA processing by preventing binding of ZC3H18 and recruitment of the RNA exosome. Last but not least, our RT-QPCR data show that PHAX and ZC3H18 depletions have functional consequences on the level of mature snRNA, and this is due to a competition between both proteins which occur on those snRNA read-through transcripts.To further explore the role of ZC3H18, I performed a two-hybrid screen and identified several splicing factors. I could validate these interactions, identify the domains involved and show that binding of some of these factors is exclusive with that of NEXT. Importantly, proteomic experiments with one of these factors identified a complex that makes the link between the cap and the splicing machinery. In agreement, RNA-Seq analysis of ZC3H18 knock-down cells showed alterations in splicing of cap-proximal introns, for a small set of genes.Altogether, this work reveals how the multiple roles of the RNA cap are achieved at the biochemical level, and suggests that the nascent RNA sequence triggers formation of one among several mutually exclusive complexes.

The Canadian Broadcasting Corporation (CBC) has been a force on Canadian airwaves for nearly a century. Within that timeframe, kilometres of textual records and thousands of hours of audiovisual recordings have been produced. Those records are evidence of the CBC’s role in mirroring and developing Canada’s national consciousness. Yet, the CBC’s records are scattered throughout Canada in multiple archival institutions. This thesis analyzes the development of these archives, with special attention to the only repository the CBC links to on its “Resources and Archives” webpage: The CBC Digital Archives. With consideration of the challenges and opportunities presented by digital culture, this thesis aims to uncover the role of the CBC Digital Archives within CBC’s archival landscape and its wider broadcasting policies and mandate. / May 2017

Ingeniera Civil Industrial / El objetivo del presente trabajo de título fue diseñar y construir un modelo de intervención tecnológica que le permita a la Corporación de Desarrollo Tecnológico de Bienes de Capital (CBC) ofrecer a sus clientes PYMES servicios basados en las nuevas tecnologías de información (neo-TIC s).
La CBC es un organismo privado, sin fines de lucro, enfocado al apoyo del desarrollo tecnológico e industrial del país. Ofrece una plataforma de información relativa a proyectos de inversión en Chile mayores a USD 5 millones, en la que se identifican y difunden oportunidades de negocio para sus empresas asociadas.
Hoy en día, CBC ha empezado a incorporar entre sus clientes a pequeñas y medianas empresas (PYMES), entregándoles una Plataforma de Inteligencia de Mercado, con la idea de facilitarles el acceso a información que agregue valor a los productos y servicios que ofrecen. Sin embargo, el proceso se ha visto complicado debido a que las PYMES en su mayoría no cuentan con los recursos tecnológicos para procesar la información que se les entrega, lo que se ve reflejado en una baja utilización de esta herramienta.
La hipótesis de investigación utilizada para este trabajo es que con herramientas basadas en las nuevas tecnologías de información , o neo-TICs, CBC puede optimizar sus servicios ofrecidos actualmente en la plataforma, añadirles nuevas características y funcionalidades para mantener y generar ventajas competitivas y comparativas, apoyando a las PYMES a optimizar sus procesos de toma de decisiones.
En primera instancia realizó un análisis de la situación actual de la CBC en lo referente a su plataforma de información de mercado y su oferta hacia las PYMES. Luego se llevó a cabo un estudio de las principales neo-TICs existentes en el mercado y sus aplicaciones de negocios, en especial en esta clase de empresas, identificándose las siguientes tecnologías como las que podrían generar un mayor beneficio: CRM, páginas web y comercio electrónico, ofimática y networking.
Finalmente, se confeccionó un modelo de intervención tecnológica para CBC, el cual consiste en evaluar cada PYME por su nivel de tecnología y, según sus capacidades decidir cuáles son los elementos que tiene que instalar, desarrollar y/o aprender a utilizar, de los vistos anteriormente. Con esto se pretende que las empresas adquieran las herramientas necesarias para poder utilizar de forma eficiente la información que les entrega CBC, y por consiguiente, incrementar la usabilidad de la Plataforma de Inteligencia de Mercado.

ABSTRACT
Powerful themes of physical and emotional isolation are found in the Saskatchewan radio plays stored in the CBC files of the Saskatchewan Archives Recorded Sound Collection. Based on the examination of a selection of these plays in their audio format, this thesis will explore themes of isolation and how they are developed in contemporary Saskatchewan radio drama as organized into the following three categories: Timescape, Landscape, and Bodyscape. The Timescape chapter deals with themes of isolation and history plays. The Landscape chapter explores isolation and landscape within three radio plays by James Quandt. The Bodyscape chapter is dedicated to plays with themes of isolation, health, and healthcare. It also highlights the abundance of playwriting by women on healthcare topics. In doing so, the thesis addresses two larger questions: 1. To what extent are themes of isolation and the struggle to relieve it developed by Saskatchewan playwrights on the radio? And 2. If the most acute geographic and demographic isolation experienced by Saskatchewan residents was historic, i.e. during the history of European settlement, why are there still strong thematic currents of isolation appearing throughout Saskatchewan radio plays written after 1980? The development of themes of isolation in this selection of plays often reinforces the value of community and the dangers of isolation. The critical framework of the thesis relies most heavily on the thoughts of Northrop Frye, Margaret Atwood, Kelley Jo Burke, and Carol Gilligan. Most of the plays discussed were produced during the period between 1978-1988. These were the first ten years of local, full time radio drama production at CBC Saskatchewan in Regina. An annotated bibliography of the plays studied en route to this thesis follows the text.
