A busca por uma fonte de matéria prima renovável alternativa aos combustíveis fósseis tem impulsionado o desenvolvimento de novas fontes de biomassa para os biocombustíveis. Uma fonte de biomassa alternativa que vêm ganhando destaque são as microalgas, microorganismos fotossintéticos capazes de capturar o CO2 atmosférico e acumular altos teores de lipídeos em sua biomassa, tornando-os muito atrativos como matéria-prima para a síntese de biodiesel. Nesse sentido, muitos trabalhos têm sidos conduzidos para o desenvolvimento dos mais variados tipos de fotobiorreatores para o cultivo de microalgas. Assim, o presente trabalho propôs um fotobiorreator integrado (coluna de bolhas e tubular), internamente iluminado com luz de LED, para estudar os efeitos dos fatores: cor da iluminação, fotoperíodo, aeração e vazão de circulação de meio, no cultivo foto-autotrófico da microalga marinha Chlorella minutissima em duas fases, utilizando o planejamento estatístico de Taguchi (L9), com vistas à formação de biomassa e ao acúmulo de lipídeos nessa microalga, para a produção de biodiesel. Foi alcançada a máxima concentração de biomassa de 506 mg L -1 e a máxima produtividade volumétrica de biomassa de 62,5 mg L -1 com 20,2 % de lipídeos para o cultivo da Chlorella minutissima no fotobiorreator integrado internamente iluminado com a cor azul, em fotoperíodo de 24 h diárias de iluminação, provido com 0,5 vvm de aeração e 5,1 L min -1 de vazão de recirculação de meio de cultivo. O perfil dos ácidos graxos presentes no extrato lipídico da Chlorella minutissima apresentou predominância de ácidos graxos saturados, com destaque para o ácido palmítico (C 16 : 0), com 59,49 %, 57,28 % e 46,5 %, nos cultivos iluminados com as cores, branco, vermelho e azul, respectivamente. A partir dos lipídeos extraídos, atingiu-se 96,6 % de conversão em biodiesel utilizando etanol via catálise ácida com ácido sulfúrico (10 % m / m) durante 8 h de reação à 80 ?C. / The search for a renewable source alternative to fossil fuels has driven the development of new sources of biomass for biofuels. The fatty acids profile present in the lipid extract of Chlorella minutissima showed a predominance of saturated fatty acids, with emphasis on palmitic acid (C 16: 0), with 59.49%, 57.28% and 46.5%, to cultivations illuminated with white, red and blue, respectively. As microalgas têm sido objeto de diversos estudos visando-se à produção de biocombustíveis ou outros produtos de interesse dentro do conceito de biorrefinaria. Na produção de biocombustíveis, as microalgas têm sido utilizadas por apresentarem elevada produtividade em lipídeos e carboidratos. Para viabilização de seu uso como matéria prima, é fundamental o desenvolvimento de processos com elevada produção de biomassa. Os fatores que influenciam no crescimento e composição da biomassa microalgal incluem a seleção do biorreator, a concentração e natureza da fonte de nitrogênio e o tipo de cultivo com relação às fontes de carbono e energia (autotrófico, heterotrófico ou mixotrófico). Neste trabalho, a microalga Chlorella minutissima 26a foi cultivada em fotobiorreatores do tipo tanque de bolhas em modo batelada ou airlift de tubos concêntricos em modo contínuo. Em cultivo autotrófico, estes fotobiorreatores foram empregados para avaliação da influência da concentração de nitrato de sódio no meio de cultivo no crescimento microalgal, na composição química da biomassa e na produtividade em lipídeos, carboidratos, proteínas e biomassa seca. Foi analisado também o uso de lixiviado proveniente de aterro sanitário na composição do meio de cultivo em fotobiorreator airlift em processo mixotrófico contínuo, promovendo concomitante biorremediação do lixiviado empregado. Os resultados demonstraram que a concentração de nitrato no meio de cultivo influenciou o crescimento e a composição química da biomassa e, consequentemente, os valores de produtividade obtidos. Nos ensaios usando fotobiorreator de tanque de bolhas em batelada, a variação da concentração de nitrato de sódio no meio de 37,5 mg L-1 para 150 mg L-1 promoveu aumento da produtividade em lipídeos e carboidratos, com valores máximos de 105,2 ± 1,7 mg L-1 d-1 e 36,7 ± 0,6 mg L-1 d-1, respectivamente, obtidos com a maior concentração de nitrato avaliada. No cultivo usando o fotobiorreator airlift em processo contínuo, o aumento da concentração de nitrato de sódio de 75 mg L-1 para 225 mg L-1 resultou nos maiores valores de produtividade média no estado estacionário para biomassa, lipídeos, proteínas e carboidratos, os quais foram, respectivamente, de 188,6 ± 11.2 mg L-1 d-1, 92,8 ± 5,5 mg L-1 d-1, 37,7 ± 2,2 mg L-1 d-1 e 29,1 ± 1,7 mg L-1 d-1. Neste caso, quando a concentração de nitrato de sódio aumentou de 75 mg L-1 para 150 mg L-1, observou-se aumento no teor de lipídeos e redução no teor de carboidratos da biomassa, sem modificação composicional apreciável quando a concentração de nitrato de sódio foi elevada para 225 mg L-1. O emprego do lixiviado diluído como meio de cultivo da Chlorella minutissima 26a resultou em elevados valores de produtividade em lipídeos, carboidratos, proteínas e biomassa, correspondendo a valores máximos de 232,0 ± 7,6, 95,3 ± 5,2, 33,4 ± 2,1 e 69,2 ± 3,0 mg L-1 d-1, respectivamente, dependendo da concentração de lixiviado no meio. O cultivo microalgal em lixiviado diluído resultou ainda em remoção de até 92,8% da demanda química de oxigênio, 90,5% do carbono orgânico total e 100% do nitrato presente no meio. Além disso, metais presentes foram removidos do meio durante o cultivo, resultando na remoção de até 63%, 72%, 100% e 67% para Cr, Fe, Al e Ba, respectivamente. O fotobiorreator airlift de tubos concêntricos demonstrou grande potencial para o cultivo da microalga C. minutissima em processo contínuo, incluindo a possibilidade de uso de lixiviado diluído na composição do meio para cultivo mixotrófico com sua concomitante biorremediação. / Microalgae have been the subject of different studies aimed at the production of biofuels or other products of interest within the concept of biorefinery. In the production of biofuels, microalgae have been used because they present high productivity in lipids and carbohydrates. In order to enable its use as a raw material, the development of processes with high biomass production is fundamental. Variables influencing the growth and composition of microalgal biomass include the selection of the bioreactor, the concentration and kind of the nitrogen source and the type of cultivation with respect to carbon and energy sources (autotrophic, heterotrophic or mixotrophic). In this work, the microalgae Chlorella minutissima 26a was cultivated in photobioreactors of two different kinds: bubble tank in batch mode or airlift of concentric tubes in continuous mode. In autotrophic cultivation, these photobioreactors were used to evaluate the influence of sodium nitrate concentration in the culture medium on microalgal growth, chemical composition of biomass and productivity in lipids, carbohydrates, proteins and dry biomass. The use of landfill leachate was also analyzed as a component of the culture medium in continuous airlift photobioreactor in a mixotrophic process, promoting concomitant bioremediation of the used leachate. The results showed that the concentration of nitrate in the culture medium influenced the growth and chemical composition of the biomass and, consequently, the productivity values obtained. In the assays using batch bubble tank photobioreactor, the variation in the sodium nitrate concentration in the medium from 37.5 mg L-1 to 150 mg L-1 promoted an increase of the productivity in lipids and in carbohydrates, with maximum values of 105.2 ± 1.7 mg L-1 d-1 and 36.7 ± 0.6 mg L-1 d-1, respectively, obtained with the highest evaluated nitrate concentration. In the cultivation using continuous airlift photobioreactor, the increase in sodium nitrate concentration from 75 mg L-1 to 225 mg L-1 resulted in higher values of average productivity in steady-state for biomass, lipids, proteins and carbohydrates, which were, respectively, of 188.6 ± 11.2 mg L-1 d-1, 92.8 ± 5.5 mg L-1 d-1, 37.7 ± 2.2 mg L-1 d-1 and 29.1 ± 1.7 mg L-1 d-1. In this case, when the sodium nitrate concentration increased from 75 mg L-1 to 150 mg L-1, there was an increase in lipid and a reduction in the carbohydrate content of the biomass, with no appreciable compositional modification when the sodium nitrate concentration was increased to 225 mg L-1. The use of diluted leachate as a culture medium of Chlorella minutissima 26a resulted in high productivity values in lipids, carbohydrates, proteins and biomass, corresponding to maximum values of 232.0 ± 7.6, 95.3 ± 5.2, 33. 4 ± 2.1 and 69.2 ± 3.0 mg L-1 d-1, respectively, depending on the landfill leachate concentration in the medium. The microalgal cultivation in diluted leachate also resulted in the removal of up to 92.8% of the chemical oxygen demand, 90.5% of the total organic carbono and 100% of the nitrate present in the medium. In addition, present metals were uptake from the medium during cultivation, resulting in a removal of up to 63%, 72%, 100% and 67% for Cr, Fe, Al and Ba, respectively. The airlift photobioreactor of concentric tubes demonstrated great potential for the cultivation of C. minutissima microalgae in a continuous process, including the possibility of using landfill leachate diluted in the medium composition for mixotrophic cultivation with its concomitant bioremediation.

As microalgas são fontes sustentáveis de bioprodutos com alto valor agregado, que podem ser utilizados para diversos fins, tais como alimento, matéria-prima e energia. Devido a isso, as microalgas se condicionam como uma solução para suprir o aumento da população mundial, com a vantagem de serem obtidas sem uso de energia fóssil e terras agricultáveis. Entretanto, os custos associados à produção de microalgas ainda é muito alto, o que inviabiliza o seu uso em grande escala. A combinação do tratamento de esgoto e o crescimento de microalgas é uma promissora alternativa para reduzir os custos relacionados ao cultivo. No entanto, o esgoto afluente das ETEs brasileiras são altamente diluídos, o que torna o seu uso impraticável do ponto de vista técnico e econômico para o cultivo de microalgas. Nesse contexto, o presente trabalho propõe uma mistura de esgoto (esgoto sanitário e de suinocultura), visando aumentar a concentração de nutrientes (carbono, nitrogênio e fósforo), para o cultivo de Chlorella sorokiniana. Os resultados são promissores, com produção média de 1 g.L-1 de C. sorokiniana e remoção média de carbono inorgânico dissolvido (CID), ortofosfato (PO43-) e amônia (NH3) de 46 a 56%, 40 a 60% e 100%, respectivamente. Foi verificado que a remoção de NH3 por air stripping durante o cultivo afetou negativamente a produtividade de biomassa e remoção de nutrientes. Além disso, algumas propostas de separação foram estudadas, uma vez que essa etapa pode representar de 20 a 60% dos custos totais de produção de microalga. A alteração de pH seguida pela FAD apresentou altas eficiências (96,5 a 97,9%) nos pH de 12 a 13. A coagulação seguida pela FAD foi estudada utilizando coagulantes orgânicos e inorgânicos. As melhores doses foram de 10 mg·L-1 Zetag 8185; 75 mg·L-1 Tanfloc SG; 500 mg·L-1 Al2(SO4)3 e 1000 mg·L-1 FeCl3 que apresentaram uma eficiência máxima de 98,4; 94,5; 95,4; 96,7%; respectivamente. A sedimentação pela floculação alcalina com precipitados de fosfato de cálcio é eficiente, porém pode ser sensível à presença de compostos proteicos presentes no esgoto (albumina e matéria orgânica bacteriana). Entretanto, foram encontradas condições (pH 8 e 10) e concentrações (10 mg·L-1), nas quais a floculação alcalina foi efetiva. / Microalgae are sustainable sources of high-value bioproducts that can be used for a variety of purposes, such as food, raw materials and energy. For this reason, the microalgae are conditioned as a solution to supply the increase of the world population, with the advantage of being obtained without using fossil energy and arable land. However, the costs associated with the production of microalgae are still very high, which makes it impossible to use them in large-scale. The combination of wastewater treatment and microalgae cultivation is a viable option to reduce expenses related to cultivation. However, municipal wastewater from the centralized Brazilian sanitation system is highly diluted, and it is technically and economically impractical to use it for microalgae cultivation. In this context, the present work proposes a wastewater mixture (municipal and swine wastewater), aiming to increase the concentration of nutrients (carbon, nitrogen and phosphorus) for the cultivation of Chlorella sorokiniana. The results are promising, C. sorokiniana production reached around 1 g.L-1, with dissolved inorganic carbon (DIC), ortophosphate (PO43-) and ammonia (NH3) average removal from 46 to 56%, 40 to 60% and 100%, respectively. It was found that NH3 removal by air stripping during cultivation negatively affected the biomass productivity and nutrient removal. In addition, some harvesting method proposals have been studied, since this step represents 20 to 60% of the total costs of microalgae production. The pH modulation followed by DAF showed high efficiencies (96.5 to 97.9%) at pH 12 to 13. Coagulation followed by DAF was studied using organic and inorganic coagulants. The best doses were 10 mg·L-1 Zetag 8185; 75 mg·L-1 Tanfloc SG; 500 mg·L-1 Al2(SO4)3 and 1000 mg·L-1 FeCl3 which had a maximum efficiency of 98.4, 94.5, 95.4, 96.7%, respectively. Alkaline flocculation using calcium phosphate precipitates is an efficient harvesting method, but it can be sensitive to the presence of protein compounds in the wastewater (albumin and bacterial organic matter). However, it was found conditions (pH 8 e 10) and concentrations (10 mg·L-1) at which alkaline flocculation was effective.

O presente trabalho compreende a execução de 16 experimentos, realizados em duplicata, visando avaliar a influência de fatores, tais como iluminância, concentração de NaH2PO4.H2O, concentração de NaNO3, fotoperíodo e comprimento de onda, além da interação entre a iluminância e os demais fatores, na produção de biomassa e lipídios pela microalga marinha Chlorella minutíssima. Os experimentos estiveram dispostos em 2 arranjos ortogonais L8 de Taguchi, cada um dos quais recebeu dados obtidos em experimentos com diferentes comprimentos de onda (470 nm, além da luz branca), nos quais eram variados, em dois níveis, a iluminância (26 klx e 13 klx), as concentrações de NaH2PO4.H2O (5 mg/L e 15 mg/L) e NaNO3 (45 mg/L e 75 mg/L) e o fotoperíodo (12: 12 horas claro: escuro e 8 ciclos diários de 2: 1 horas claro: escuro). Os experimentos se processaram em reatores de vidro com formato de prisma de base quadrada com volume útil de 1,1 L, isolados da influência da luminosidade externa. Para a luz branca, o melhor resultado para a produção de biomassa foi de 1300 mg/L, ao passo que o melhor resultado para o teor lipídico foi de 22,75% em relação à biomassa seca. Para a luz de comprimento de onda equivalente a 470 nm, o melhor resultado para a produção de biomassa foi de 1270 mg/L, ao passo que o melhor resultado para o teor lipídico foi de 19,03% em relação à biomassa seca. / This work comprises the execution of 16 experiments, performed in duplicate, in order to evaluate the influence of factors such as illuminance, NaH2PO4.H2O concentration, NaNO3 concentration, photoperiod and wavelenght, in addition to the interaction between the illuminance and the others factors, in the production of biomass and lipids by the m arine microalgae Chlorella minutíssima. Nature changes continuously and is only seemingly at equilibrium. Environmental parameters like temperature, humidity or insolation may strongly fluctuate on scales ranging from seconds to millions of years. Being part of an ecosystem, species have to cope with these environmental changes. For ecologists, it is of special interest how individual responses to environmental changes affect the dynamics of an entire population – and, if this behavior is predictable. In this context, the demographic structure of a population plays a decisive role since it originates from processes of growth and mortality. These processes are fundamentally influenced by the environment. But, how exactly does the environment influence the behavior of populations? And what does the transient behavior look like? As a result from environmental influences on demography, so called cohorts form. They are age or size classes that are disproportionally represented in the demographic distribution of a population. For instance, if most old and young individuals die due to a cold spell, the population finally consists of mainly middle-aged individuals. Hence, the population got synchronized. Such a population tends to show regular fluctuations in numbers (denoted as oscillations) since the alternating phases of individual growth and population growth (due to reproduction) are now performed synchronously by the majority of the population.That is, one time the population growths, and the other time it declines due to mortality. Synchronous behavior is one of the most pervasive phenomena in nature. Gravitational synchrony in the solar system; fireflies flashing in unison; coordinate firing of pacemaker cells in the heart; electrons in a superconductor marching in lockstep. Whatever scale one looks at, in animate as well as inanimate systems, one is likely to encounter synchrony. In experiments with phytoplankton populations, I could show that this principle of synchrony (as used by physicists) could well-explain the oscillations observed in the experiments, too. The size of the fluctuations depended on the strength by which environmental parameters changed as well as on the demographic state of a population prior to this change. That is, two population living in different habitats can be equally influenced by an environmental change, however, the resulting population dynamics may be significantly different when both populations differed in their demographic state before. Moreover, specific mechanisms relevant for the dynamic behavior of populations, appear only when the environmental conditions change. In my experiments, the population density declined by 50% after ressource supply was doubled. This counter-intuitive behavior can be explained by increasing ressource consumption. The phytoplankton cells grew larger and enhanced their individual constitution. But at the same time, reproduction was delayed and the population density declined due to the losses by mortality. Environmental influences can also synchronize two or more populations over large distances, which is denoted as Moran effect. Assume two populations living on two distant islands. Although there is no exchange of individuals between them, both populations show a high similarity when comparing their time series. This is because the globally acting climate synchronizes the regionally acting weather on both island. Since the weather fluctuations influence the population dynamics, the Moran effect states that the synchrony between the environment equals the one between the populations. My experiments support this theory and also explain deviations arising when accounting for differences in the populations and the habitats they are living in. Moreover, model simulations and experiments astonishingly show that the synchrony between the populations can be higher than between the environment, when accounting for differences in the environmental fluctuations (“noise color”). / Die Natur unterliegt ständigen Veränderungen und befindet sich nur vermeintlich in einem Gleichgewicht. Umweltparameter wie Temperatur, Luftfeuchtigkeit oder Sonneneinstrahlung schwanken auf einer Zeitskala von Sekunden bis Jahrmillionen und beinhalten teils beträchtliche Unterschiede. Mit diesen Umweltveränderungen müssen sich Arten als Teil eines Ökosystems auseinandersetzen. Für Ökologen ist interessant, wie sich individuelle Reaktionen auf die Umweltveränderungen im dynamischen Verhalten einer ganzen Population bemerkbar machen und ob deren Verhalten vorhersagbar ist. Der Demografie einer Population kommt hierbei eine entscheidende Rolle zu, da sie das Resultat von Wachstums- und Sterbeprozessen darstellt. Eben jene Prozesse werden von der Umwelt maßgeblich beeinflusst. Doch wie genau beeinflussen Umweltveränderungen das Verhalten ganzer Populationen? Wie sieht das vorübergehende, transiente Verhalten aus? Als Resultat von Umwelteinflüssen bilden sich in Populationen sogenannte Kohorten, hinsichtlich der Zahl an Individuen überproportional stark vertretene Alters- oder Größenklassen. Sterben z.B. aufgrund eines außergewöhnlich harten Winters, die alten und jungen Individuen einer Population, so besteht diese anschließend hauptsächlich aus Individuen mittleren Alters. Sie wurde sozusagen synchronisiert. Eine solche Populationen neigt zu regelmäßigen Schwankungen (Oszillationen) in ihrer Dichte, da die sich abwechselnden Phasen der individuellen Entwicklung und der Reproduktion nun von einem Großteil der Individuen synchron durchschritten werden. D.h., mal wächst die Population und mal nimmt sie entsprechend der Sterblichkeit ab. In Experimenten mit Phytoplankton-Populationen konnte ich zeigen, dass dieses oszillierende Verhalten mit dem in der Physik gebräuchlichen Konzept der Synchronisation beschrieben werden kann. Synchrones Verhalten ist eines der verbreitetsten Phänomene in der Natur und kann z.B. in synchron schwingenden Brücken, als auch bei der Erzeugung von Lasern oder in Form von rhythmischem Applaus auf einem Konzert beobachtet werden. Wie stark die Schwankungen sind, hängt dabei sowohl von der Stärke der Umweltveränderung als auch vom demografischen Zustand der Population vor der Veränderung ab. Zwei Populationen, die sich in verschiedenen Habitaten aufhalten, können zwar gleich stark von einer Umweltveränderung beeinflusst werden. Die Reaktionen im anschließenden Verhalten können jedoch äußerst unterschiedlich ausfallen, wenn sich die Populationen zuvor in stark unterschiedlichen demografischen Zuständen befanden. Darüber hinaus treten bestimmte, für das Verhalten einer Population relevante Mechanismen überhaupt erst in Erscheinung, wenn sich die Umweltbedingungen ändern. So fiel in Experimenten beispielsweise die Populationsdichte um rund 50 Prozent ab nachdem sich die Ressourcenverfügbarkeit verdoppelte. Der Grund für dieses gegenintuitive Verhalten konnte mit der erhöhten Aufnahme von Ressourcen erklärt werden. Damit verbessert eine Algenzelle zwar die eigene Konstitution, jedoch verzögert sich dadurch die auch die Reproduktion und die Populationsdichte nimmt gemäß ihrer Verluste bzw. Sterblichkeit ab. Zwei oder mehr räumlich getrennte Populationen können darüber hinaus durch Umwelteinflüsse synchronisiert werden. Dies wird als Moran-Effekt bezeichnet. Angenommen auf zwei weit voneinander entfernten Inseln lebt jeweils eine Population. Zwischen beiden findet kein Austausch statt – und doch zeigt sich beim Vergleich ihrer Zeitreihen eine große Ähnlichkeit. Das überregionale Klima synchronisiert hierbei die lokalen Umwelteinflüsse. Diese wiederum bestimmen das Verhalten der jeweiligen Population. Der Moran-Effekt besagt nun, dass die Ähnlichkeit zwischen den Populationen jener zwischen den Umwelteinflüssen entspricht, oder geringer ist. Meine Ergebnisse bestätigen dies und zeigen darüber hinaus, dass sich die Populationen sogar ähnlicher sein können als die Umwelteinflüsse, wenn man von unterschiedlich stark schwankenden Einflüssen ausgeht.

Por estes motivos, novas tecnologias para geração de energia limpa vêm sendo criadas. Um exemplo destas são as células a combustível, que são dispositivos que convertem energia química em elétrica. Porém esta tecnologia apresenta algumas limitações, como deficiências na transferência de elétrons, baixa geração de potência e altos custos associados a utilização de catalizadores metálicos, os quais aumentam os custos de implantação e dificultam sua utilização em larga escala. Visando superar estas limitações uma variação desta tecnologia foi desenvolvida, a chamada célula a combustível fotossintética. Neste tipo de célula a combustível, microalgas como a Chlorella vulgaris são utilizadas no compartimento catódico substituindo catalizadores químicos melhorando sua sustentabilidade e reduzindo os custos de implantação. As utilizações de cátodos de microalgas também colaboram com o sequestro de carbono da atmosfera, o convertendo em oxigênio e biomassa rica em metabolitos de grande valor comercial como amido e lipídios. No presente estudo a microalga C. vulgaris foi utilizada no compartimento catódico de uma célula a combustível fotossintética a fim de se analisar sua eficiência na produção de energia, sua capacidade de sequestrar o dióxido de carbono da atmosfera e seu acúmulo de materiais de reserva como amido e lipídios totais. Na primeira parte deste estudo a microalga C. vulgaris foi utilizada como aceptora de elétrons em um compartimento catódico, onde durante 10 dias de experimento foram avaliados a quantidade de CO2 capturada pelas células de algas (7mg/L de CO2), a composição da biomassa, Amido (3%) e Lipídios (70%) e parâmetros eletroquímicos como a Eficiência Coulômbica (CE = 33,1%) e densidade de corrente máxima (Idmax = 147 mA cm²). Em seguida esta condição foi submetida a um planejamento fatorial completo 2², onde as variáveis independentes, tempo de iluminação e a concentração de nitrogênio foram testadas sobre a produção de eletricidade e acúmulo de amido e lipídios. Durante os experimentos foi constatado que a iluminação é o fator que mais influi na geração de energia, onde foram obtidos valores de densidade de corrente máxima Idmax = 178 mA/cm² com uma eficiência coulômbica de 42,5%, além de uma acumulação máxima de amido de 38% e 77% de lipídios, demonstrando que a utilização de um cátodo fotossintético para produção de energia é viável e eficiente na produção de metabólitos com elevado valor comercial. / The global population growth has caused a substantial increase in demand for energy, which in short-term may cause an energy crisis, since much of the energy consumed throughout the world comes from non-renewable sources like oil. Besides its exploration and use cause great damage to the environment, affecting diverse ecosystems and contributing to global warming. For those reasons, are being created new technologies for clean energy generation. An example of these technologies is Fuel Cells, which are devices that convert chemical energy into electric. However, this technology has some limitations, such as defects in electron transfer, low power output and high costs associated with the use of metal catalysts, which increase deployment costs and hamper its use on a large scale. Aiming to overcome these limitations, a variation of this technology was developed, the so-called photosynthetic fuel cell. In this type of fuel cell, microalgae such as Chlorella vulgaris are used in the cathode compartment replacing chemical catalysts improving their sustainability and reducing deployment costs. The uses of microalgae cathodes also collaborate with carbon capture from the atmosphere, turning it into oxygen and biomass rich in metabolites of great commercial value as starch and lipids. In the current study the microalgae C. vulgaris was used in the cathode compartment of a photosynthetic fuel cell in order to analyze its energy production efficiency, their ability to sequester atmospheric carbon dioxide and its accumulation of reserve materials such as starch and total lipids. The first part of this study microalga C. vulgaris was utilized as an electron acceptor in a cathode compartment, which were analyzed during 10 days of experiment the amount of CO2 captured by the algae cells (7mg/L-1 of CO2), the composition of the biomass starch (3%) and lipids (70%) and electrochemical parameter as coulombic efficiency (CE = 33.1%) and the maximum current density (mA Idmax = 147 cm²). Then this condition was subjected to a complete factorial design 2² where the independent variables, illumination time and the concentration of nitrogen were tested on the production of electricity and accumulation of starch and lipids. During the experiments it was evidenced that enlightenment is the factor that most affects the power generation, which were obtained maximum current density values Idmax = 178 mA/cm² with a coulombic efficiency of 42.5%, and a maximum accumulation of 38% of starch and 77% of lipid, demonstrating that the use of a photosynthetic cathode for energy generation is feasible, and also in the production of metabolites with a high commercial value.

Aplicación de Salvinia y Chlorella Vulgaris para el tratamiento de aguas residuales de una PTAR a nivel de laboratorio

El tratamiento de aguas residuales es un problema que aqueja a todo el mundo, dependiendo del lugar en mayor o menor medida, siendo Lambayeque el lugar de estudio de la presente tesis, se puede demostrar la evidente dificultad en el proceso ya mencionado. Es así como el documento en cuestión pretende dar una posible solución a la dificultad ya descrita, proponiendo el uso de Salvinia y Chlorella Vulgaris para mejorar la calidad de las aguas residuales en parámetros específicos como son el DBO, DQO, Ph, SST, contenido de cobre, contenido de zinc y coliformes fecales. Utilizando muestras de dos PTAR’s de la región, las cuales son la PTAR de Ferreñafe y la PTAR de Pimentel; se realizaron las aplicaciones de Salvinia, Chlorella Vulgaris y de Salvinia + Chlorella Vulgaris a cada muestra recolectada, obteniendo resultados favorables, concluyendo así que el tratamiento propuesto y ejecutado además de ser eco amigable y económico también es eficiente, confirmando la hipótesis planteada dentro de este escrito. / Wastewater treatment is a problem that afflicts everyone, depending on the place to a greater or lesser extent, being Lambayeque the place of study of this thesis, the evident difficulty in the aforementioned process can be demonstrated. Thus, the document in question intends to provide a possible solution to the difficulty already described, proposing the use of Salvinia and Chlorella Vulgaris to improve the quality of wastewater in specific parameters such as BOD, COD, pH, TSS, copper content, zinc content and fecal coliforms. Using samples from two WWTPs in the region, the Ferreñafe WWTP and the Pimentel WWTP, Salvinia, Chlorella Vulgaris and Salvinia + Chlorella Vulgaris were applied to each sample collected, obtaining favorable results, thus concluding that the proposed and implemented treatment is not only eco-friendly and economical but also efficient, confirming the hypothesis put forward in this paper.

Microalgae biorefinery : proposition of a fractionation process / Bioraffinage des microalgues : proposition d’un procédé de fractionnement

Le concept d’une algoraffinerie primaire traitant les principaux composants de microalgues (lipides, protéines, glucides et pigments) a été étudié. Une séquence d'opérations unitaires a été mis en œuvre afin d'obtenir des fractions enrichies de ces biomolécules tout en conservant leur integrité dans le procédé en aval. L'étude a été principalement centrée sur Chlorella vulgaris, une espèce connue pour sa paroi cellulaire rigide. La majorité de la fraction lipophile (lipides et pigments) a été récupérée en utilisant du dioxyde de carbone supercritique avec de l'éthanol en tant que co-solvant, sans opération unitaire de cassage cellulaire préalable. La fraction hydrophile (protéines et polysaccharides) a été récupérée dans la phase aqueuse après broyage à billes comme méthode de cassage cellulaire. Par la suite, la phase aqueuse a été séparée en trois fractions par un procédé d'ultrafiltration en deux étapes. Ainsi, les amidons, les pigments, les protéines et les sucres ont été séparés les uns des autres avec succès. Une analyse du cycle de vie sera nécessaire pour estimer le coût et la durabilité du procédé de fractionnement. / A primary algorefinery, concept that deals with the main components of microalgae (lipids, proteins, carbohydrates and pigments), has been studied. A sequence of unit operations has been implemented in order to obtain separated enriched fractions of these biomolecules by conserving their integrity in the downstream process. The study was mainly centred on Chlorella vulgaris, a species known for its rigid cell wall. Most of the lipophilic fraction (lipids and pigments) was recovered using supercritical carbon dioxide with ethanol as a co-solvent, without a preliminary unit operation of cell disruption. The hydrophilic fraction (proteins and polysaccharides) was recovered in the aqueous phase after bed milling as cell disruption method. Subsequently, the aqueous phase was fractionated into three fractions by means of a process of two-stage ultrafiltration. Thus, starches, pigments, proteins and sugars were successfully separated from each other. A life cycle assessment will be necessary to estimate the cost and the sustainability of the fractionation process

A la redécouverte des Chlorovirus : Contribution à l'étude des virus géants à ADN

Les virus du genre Chlorovirus (phycoDNAviridae) sont des virus à ADN double-brin de grande taille, qui infectent des algues vertes eucaryotes unicellulaires vivant en eau douce nommées les chlorelles (Trebouxiophyceae). A l'aide d'approches bioinformatiques, j'ai consacré mon travail de thèse à l'étude de la diversité génomique des chlorovirus et de leur histoire évolutive, ainsi qu'à l'étude transcriptomique de l'infection virale chez son hôte.Dans le premier volet de ma thèse, j'ai procédé à l'assemblage et à l'annotation de 50 nouveaux génomes de chlorovirus récemment séquencés (Roche-454). J'ai ainsi été en mesure de mieux caractériser les différences de structure du génome et de contenu en gène des différents chlorovirus en fonction de leurs hôtes. J'ai également mis en évidence l'existence d'un quatrième sous-groupe de chlorovirus, repoussant les limites connues de la diversité de ces virus. Enfin j'ai montré que les Chlorovirus ne suivent pas le même schéma évolutif des autres NCLDV et que l'origine de leurs gènes est encore inconnue, bien que probablement virale.Dans un autre projet, j'ai également étudié les variations de la transcription des gènes de la chlorelle (C. varabilis NC64A) induites par l'infection par un chlorovirus (PBCV-1) grâce au séquençage profond (Illumina) des ARNm polyadénylés présent dans la cellule saine, puis après infection. J'ai ainsi pu montrer que les différentes fonctions cellulaires sont impactées de façon préférentielle par l'infection. / Giant viruses in the genus Chlorovirus (Phycodnaviridae) infect eukaryotic green microalgae known as Chlorella (Trebouxiophyceae). Using bioinformatic approaches, I dedicated my thesis on the study of the genomic diversity and evolutionnary history of the chlorovirus at the genus level, and the transcriptomic of the viral infection.In the first part on my work, I conducted the assembly and annotation of 50 new chlorovirus genomes recently sequenced (Roche-454). I was able to refine the known differences between chloroviruses, both in genome structure and gene content terms. Clues for the existence of a fourth subgroup of chloroviruses were also found. I was also able to show that the chlorovirus does not follow the same evolutionnary pattern as the other NCLDV, and that the origin of their genes is still largely unknown, but presumably of viral origin.In a second project, I studied the variation in the transcription of chlorella's genes (C. variabilis NC64A) during the infection by a chlorovirus (PBCV-1) using the deep-sequencing (Illumina) of all polyadenylated messenger RNA in the healthy or infected cell. This way, I was able to show that the various cellular functions are preferentially impacted by the infection.
