Genetic Characterization of Antimicrobial Activities of the Bacteria Burkholderia Contaminans MS14 and Pseudomonas Chlororaphis UFB2

Burkholderia contaminans MS14 shows excellent antimicrobial activities against a wide range of pathogens. Complete sequence analysis reveals that the MS14 genome harbors multiple gene loci that contribute to its antimicrobial activities and lacks key virulence features commonly found in pathogenic Burkholderia species. A mutagenesis study identified the genes required for MS14 antibacterial activities and gene expression profiling targeted a polyketide synthase (PKS) gene cluster. Site-specific mutagenesis confirmed the PKS gene cluster is directly related to MS14 antibacterial activities and the PKS gene product is predicted to be the MS14 antibacterial compound. Strain UFB2 isolated from Mississippi shows significant antifungal and antibacterial activities. UFB2 was classified to be Pseudomonas chlororaphis and its complete genome sequence was reported in this study. Green house trails showed P. chlororaphis strain UFB2 could efficiently reduce the disease severity of bacterial canker of tomato, by significantly inhibiting the growth of the pathogen Clavibacter michiganensis subsp. michiganensis. The research findings of B. contaminans MS14 and P. chlororaphis UFB2 have provided insights into the development of MS14 antibacterial compound for agricultural application and potential use of strain UFB2 as a biocontrol agent.

Coverage Path Planning And Control For Autonomous Mobile Robots

Coverage control has many applications such as security patrolling, land mine detectors, and automatic vacuum cleaners. This Thesis presents an analytical approach for generation of control inputs for a non-holonomic mobile robot in coverage control. Neural Network approach is used for complete coverage of a given area in the presence of stationary and dynamic obstacles. A complete coverage algorithm is used to determine the sequence of points. Once the sequences of points are determined a smooth trajectory characterized by fifth order polynomial having second order continuity is generated. And the slope of the curve at each point is calculated from which the control inputs are generated analytically. Optimal trajectory is generated using a method given in research literature and a qualitative analysis of the smooth trajectory is done. Cooperative sweeping of multirobots is achieved by dividing the area to be covered into smaller areas depending on the number of robots. Once the area is divided into sub areas, each robot is assigned a sub area for cooperative sweeping.

A transportation system designed to prioritize the mobility of automobiles cannot accommodate the growing number of road users. The Complete Streets policy plays a crucial part in transforming streets to accommodate multiple modes of transportation, especially active modes like biking and walking. Complete streets are referred to as streets designed for everyone and enable safety and mobility to all users. A strategy of complete streets transformation is to connect isolated complete street segments to form a complete network that improves active mobility and public transit ridership. This research assessed the impact of efficiently and equitably connecting and expanding the biking network using dedicated lanes on the safety and operation of the network in Atlanta, Georgia. These connections are aimed at increasing the multimodal use of the streets in midtown and downtown Atlanta and achieving the mobility and public health goals through the integration of various modes of travel. The evaluation was done by modeling a well-calibrated and validated network of Midtown and Downtown Atlanta in VISSIM using existing travel demand and traffic design conditions (i.e., the baseline or Scenario 0). A total of three different conditions: existing, proposed, and alternative conditions, were modeled to see the effectiveness of bike infrastructure design improvement and expansion. Three scenarios were then modeled as variations of modal demand of the different condition models. Scenarios modeled are based on input from the City and Community stakeholders. Using the trajectory data from microsimulation, the surrogate safety assessment model (SSAM) from FHWA was used to analyze the safety effect on the bike infrastructure improvement and expansion. Results of this study showed a positive impact of complete streets transformation on the streets of Midtown and Downtown Atlanta. These impacts are quantified in this thesis.

An earthquake response spectrum method for linear light secondary substructures

Yes / Earthquake response spectrum is the most popular tool in the seismic analysis and design of structures. In the case of combined primary-secondary (P-S) systems, the response of the supporting P substructure is generally evaluated without considering the S substructure, which in turn is only required to bear displacements and/or forces imposed by the P substructure (¿cascade¿ approach). In doing so, however, dynamic interaction between the P and S components is neglected, and the seismic-induced response of the S substructure may be heavily underestimated or overestimated. In this paper, a novel CQC (Complete Quadratic Combination) rule is proposed for the seismic response of linear light S substructures attached to linear P substructures. The proposed technique overcomes the drawbacks of the cascade approach by including the effects of dynamic interaction and different damping in the substructures directly in the cross-correlation coefficients. The computational effort is reduced by using the eigenproperties of the decoupled substructures and only one earthquake response spectrum for a reference value of the damping ratio.
