Ocorrência e caracterização de Giardia e Cryptosporidium em águas captadas para abastecimento público no município de Cajamar-SP e avaliação do risco / Occurrence and characterization of Giardia and Cryptosporidium in water collected for public supply in the municipality of Cajamar-SP and risk assessmentMarcel Oliveira Bataiero 18 April 2016 (has links)
Portanto, o caso é preocupante, tanto do ponto de vista do tratamento e abastecimento de água potável, quanto da degradação e contaminação do manancial, evidenciando a necessidade de se estabelecer medidas de intervenção direcionadas a promover a qualidade da água e garantir sua segurança / Introduction: The risk to human health caused by biological contamination of source water from public water supply is highlighted by the occurrence of outbreaks associated with the protozoa Giardia and Cryptosporidium, which have low infectious doses and high capacity of survival in the environment, besides being capable of withstanding traditional process for water disinfection (chlorination). Considering that there is a high risk of infection by these protozoa through drinking water treated with conventional methods uptaked from surface waters impacted by biological contamination, resulting in a possible increase in the incidence of diarrhea, this study aimed to verify the occurrence of these protozoa in waters collected for public supply in Cajamar-SP, characterize their pathogenicity and evaluate the risk associated with its consumption. Methods: Forty eight samples were collected in ribeirão dos Cristais, at uptake point of the water treatment plant, weekly, for 12 months (from 05/16/2013 to 05/21/2014). The detection and quantification of protozoa were carried out according to the United States Environmental Protection Agency Method 1623.1, and the extraction and characterization of species/genotypes of iardia and Cryptosporidium were performed through molecular methods and sequencing. The risk of infection by ingestion of Giardia and Cryptosporidium (oo)cysts present in the treated water were calculated using the Quantitative Microbial Risk Assessment tool, based on the concentration data of pathogens obtained by the Method 1623.1, removal efficiency of (oo) cysts in the conventional process of water treatment, dose-response model and rate of daily water intake for individuals under the age of 5 years and over 21 years. Results: Giardia cysts were detected in 83.3% of the samples (40/48), with concentrations ranging rom the detection limit (<0.1) to 8.6 cysts/L. Cryptosporidium oocysts were detected in 7.5% of the samples (18/48), with concentrations ranging from the detection limit (<0.1) to 2 oocysts/L. The species/genotypes found (Giardia intestinalis A and B and Cryptosporidium parvum and hominis) are characteristic of anthropogenic contamination and are often identified in epidemiological studies as being responsible for outbreaks. The estimated annual risk of infection by Giardia was 3,3x10-3 (95% CI 4,6x10-3) for children and 11,5x10-3 (95% CI 13,3x10-3) for adults, while the risk for Cryptosporidium was 1,1x10-3 (95% CI 1,7x10-3) for children and 3,9x10-3 (95% CI 5,0x10-3) for adults. The increase in the incidence of diarrhea was observed in the study scenario after an accident that resulted in an overflow of untreated sewage that reached the reservoir, coinciding with an increase in the detection of the (oo)cysts. Conclusion: The results showed that the vulnerability of the ribeirão dos Cristais to biological contamination may result in a high risk of infection and illness by Giardia and Cryptosporidium through the drinking water consumption. Therefore, the case is of concern, both from the point of view of the treatment and supply of drinking water, as well as to the degradation and contamination of the water source, highlighting the need to establish intervention measures aimed at promoting water quality and ensure its safety

Previous issue date: 2013-11-08 / Fundação de Apoio à Pesquisa - FUNAPE / Individual evaluation of the results of each one of the physical, chemical and biological
parameters monitored for quality control of water distributed to the population is an
activity hampered by the great diversity and quantity of data generated by the analysis.
Determinations of water quality indices (WQI) has been used to convert a series of data
of water quality in a single number, able to represent a simple and objective way, the
water quality as whole. Aimed at obtaining WQIs of the 224 municipalities in Goiás,
that have supply systems operated by state sanitation company (SANEAGO), this study
investigated data on physical-chemical and microbiological tests, performed according
to methods described in Standard Methods, in samples supply water collected during
two years (oct., 2011 to aug., 2013) and, by the Canadian method known as CWQI
(Canadian water Quality Index), determined the WQI of each one of 224 municipalities
evaluated, and also determined the WQI referent to geographical microregions and
mesoregions of Goiás, allowing the spatialization of results in maps of the state of
Goiás. The evaluation concluded that 97.8% of municipalities in Goiás, which water
supply systems are operated by SANEAGO, have WQI categorized as 'Excellent' or
'Good', compared with 2.2% of municipalities categorized as 'Regular' or ‘Bad’. The
spatial distribution of the results led to the conclusion that, at the end of the study
period, the mesoregion Center classified as 'Excellent' in terms of WQI, while
mesoregions East, Northwest, North and South are classified as 'Good'.

Desenvolvimento e aplicação de método para a determinação do 3-cloro-4-(diclorometil)-5-hidroxi-2[5H]-furanona (MX) em água potável / Development and application of method for the 3-chloro-4-(dichloromethyl)-2[5H]- furanone (MX)Rezemini, Andréa Lúcia 10 May 2002 (has links)
A by-product of chlorination in water is the 3-chloro-4-(dichloromethyl)-5-hydroxy-2[5H]-furanone (MX), a toxic compound which has been the subject of recent studies. Although MX has been found at low concentrations (ng/L), it accounts for approximately 50% of the total mutagenicity in drinking water. In this study, a method was developed to analyze MX at trace leveI in chlorinated waters. The MX extraction efficiency was evaluated using three different techniques: solid phase extraction (SPE), solid phase microextraction (SPME) and liquid-liquid extraction (LLE). The LLE technique provided higher recovery with value of 50% at concentrations ≤ 50 ng/L. A simple and fast procedure involving on-line based trimethylsilyl derivatization for the quantitative analysis of MX by gas chromatography with mass spectrometry (GCIMS) is proposed. Good precision and low limit of detection of the method allowed its application in real water samples. The reliable MX identification and quantification were obtained due to the use of the mass spectrometry. Since the instrumental limit of detection was equal to 3.0 µg/L, the samples had to be concentrated to 2000 times the original concentration. The optimized method was applied to water samples collected before and after chlorination process in water suppliers located in São Paulo City, Brazil. Concentrations of MX in those samples ranged from 3 to 22 ng/L which are similar to those detected in other countries.

Comparison of water quality between sources and between selected villages in the Waterberg District of Limpopo Province; South Africa: with special reference to chemical and microbial quality.Makgoka, Seretloane Japhtaline January 2005 (has links)
Thesis (MPH)--University of Limpopo, 2005 / Water and sanitation inadequacy is still an environmental health challenge in several
regions worldwide and a billion people lack access to safe water, while 2.4 billion people
have inadequate sanitation [2].
Assessment of water quality by its chemistry includes measures of elements and
molecules dissolved or suspended in water. Commonly measured chemical parameters
include arsenic, cadmium, calcium, chloride, fluoride, total hardness, nitrate, and
potassium [16]. Water quality can also be assessed by the presence of waterborne
microorganisms from human and animals’ faecal wastes. These wastes contain a wide
range of bacteria, viruses and protozoa that may be washed into drinking water supplies
[21].
Three villages were selected for water quality analysis, based on their critical situation
regarding access to water and sanitation: namely, Matlou, Sekuruwe and Taolome
villages, situated in the Mogalakwena Local Municipality within the Waterberg district of
the Limpopo Province, South Africa. A proposal was written to the Province of North
Holland (PNH) and was approved for funding to start with the implementation of those
projects, with 20% of each village’s budget allocated for water quality research [26].
This was a cross sectional, analytical study to investigate the chemical and microbial
quality of water in Matlou, Sekuruwe and Taolome villages. The study was also
conducted to explore methods used by household members to store and handle water in
storage tanks. Water samples were collected and analysed according to the standard
operating procedures (SOPs) of the Polokwane Municipality Wastewater Purification
Plant in Ladanna, Polokwane City of South Africa. The questionnaire used was adopted
from the one used for cholera outbreak in the Eastern Cape Province of South Africa.
Results show that water from all sources in all the villages had increased total hardness
concentration. Water from the borehole in Matlou village had increased number of total
coliform bacteria. There were increased total and faecal coliform bacteria in storage
tanks samples from Matlou village. Water samples from reservoirs in Sekuruwe and
Taolome villages did not test positive for any microbial contamination. Water from
xiv
informally connected yard taps in Sekuruwe village had increased total coliform bacteria,
while increased total and faecal coliforms were found in households’ storage tanks.
Water samples from communal taps in Taolome village had minimal number of total
coliform bacteria, while water from storage tanks had both increased total and faecal
coliform bacteria.
Matlou village was the only place with increased nitrate concentration at the households’
storage tanks. While all the villages had microbial contamination, Taolome village had
the least number of coliform bacteria in water samples from households’ storage tanks
as compared to Matlou and Sekuruwe villages.
It is concluded that water from sources supplied by the municipalities are safe to be
consumed by humans while water from informally connected taps and households’
storage tanks are not safe to be used without treatment.
It is recommended that a health and hygiene education package be prepared for all the
villages, so that handling of water from the main source into their storage tanks can be
improved. Secondly, it is recommended that water in all sources be treated for total
hardness and water in storage tanks in Matlou village be treated for nitrate. Thirdly, it is
recommended that water be accessed everyday of the week, so that people do not use
unsafe water supplies. / The Province of North Holland, Netherlands.

Une meilleure gestion patrimoniale des réseaux d'eau potable : le modèle de prévision du renouvellement à long terme OPTIMEAU / Better asset management of drinking water systems : OPTIMEAU the long-term forecasting model of renewalLarge, Aurore 27 June 2016 (has links)
In developed countries, drinking water is distributed to households. This comfort requires a long networkwith a high value. To optimize resources and performance, it is important to renew pipe sectionsat the best possible time. This thesis presents the short (1-3 years), medium and long term (> 30 years)models commonly employed. The short-term processes seem quite effective, but long term methods remainrough. The OPTIMEAU model proposes a long-term approach to get a prospective overview ofpipes and improve its management, while remaining connected to the technical practices at short-termlevel. This approach, based on survival statistical models, is tested using actual data of three Europeanwater services : SEDIF (Syndicat des Eaux d’Ile de France), Grand Lyon and eauservice Lausanne inSwitzerland. The originality of this model lies in the estimation of the past decommissioning age distribution, keystone in the construction of eleven indicators related to finance, achievements and futureperformance of water utilities.

Drinking water is produced from raw water and is either from groundwater or surface water. This thesis aims to find out if the cleaning process of raw water is sufficiently effective. This is important because consumers are otherwise at risk of waterborne infection caused by pathogens. There are three groups of pathogens; bacteria, virus and parasite. These have different characteristics which mean that they require different water treatment to be separated. In addition to normal operation, a number of scenarios were examined. This is to investigate how water treatment would do if they became a reality. The thesis has examined Borg´s waterworks operated by Norrköping Vatten AB. It was defined to cover the distance from water source to the consumer. In the work, the model Quantitative Microbial Risk Assessment (QMRA) was used to perform risk analysis by simulating the normal operation and different scenarios of the water purification process. Thus, knowledge can be obtained about the effectiveness of separation by bacteria, viruses and parasites. However, the QMRA-model is considered to contain some flaws and for that reason the Norwegian model called Good Disinfection Practice (GDP) was also used. GDP is a theoretical model which is based on formulas and tables. The model takes into account the raw water quality and also provides deductions for various measures that the water plant possesses to ensure a good supply of water. The results obtained with both models were similar and showed that the water treatment is sufficient for the bacteria, but not viruses and parasites. Both models were considered to be reliable but viruses and parasites are very difficult to analyze, which has resulted in uncertain literature values and hence in the results. The result also showed that neither viruses nor parasites exceeded the limit by so much that more hygienic barriers to the reduction of them are necessary. The conclusion which may be drawn from the fact that no parasites have been detected in the raw water is that the water treatment still might be sufficient. To determine the effects that an exclusion of various barriers may give, the normal operation was simulated and a purification step at a time was excluded. The result showed that the purification steps which are most important to maintain the treatment process are chemical precipitation followed by rapid filtration, slow filtration and disinfection with chlorine. If any of these cleaning steps were to fail, this introduces a large increase in the risk of waterborne disease. The results showed that the chemical precipitation step gave the greatest separation effect on the virus but also on the parasites. However, the slow filtration gave the largest separation of the parasites. Free chlorine had the greatest effect on bacteria. The investigated scenarios were assumed to be wastewater discharges, sewage discharges in relation to flood the nearby pastures, and sewage overflows due to heavy rainfall. The results of the simulated scenarios were the same when it was only bacteria that in all cases produced a result within the limits of the daily infection probability. Both viruses and parasites exceeded both values. However, there were few studies on these and thus literature values needed to be implemented in the QMRA-model. Hence, the uncertainty of the results was great. The QMRA-model also contained deficiencies in the simulation of the discharge of effluents, where the amount of virus was about 1000-10000 times too much. If this problem as well as more specific data for the investigated area, and more Swedish studies were available, a more credible simulation of the scenarios could be implemented.

Household access to water and willingness to pay in South Africa: evidence from the 2007 General Household SurveyKimbung,Ngum Julious January 2011 (has links)

This study assesses the present level of household water access and the willingness to pay in South Africa. Although the general literature informs that progress has been made in positing South Africa above the levels found in most African countries, there are some marked inequalities among the population groups and across the provinces, with some performing well and others poorly in this regard. The study looks at the extent to which households differ in terms of water access and willingness to pay according to the province of residence. The study focuses on household heads / male and female, through different social and demographic attributes, by taking account of variables such as age, education / attainment, geographic areas, and population group to name but a few. The data used in this study comes from the 2007 General Household Survey (GHS) conducted by Statistics South Africa. The scope is national and employs cross tabulation and logistic regression to establish relationships and the likelihood of living in a household with access to safe / drinking water in South Africa. Results presented in this study suggest that the difference is determined by socio- demographic characteristics of each household such as age, gender, population group, level of education, employment status income, dwelling unit, dwelling ownership, living quarters, household size and income. It throws more light as to what needs to be taken into account when considering demand and supply of and priorities for water intervention from the household perspective.

Ground water contamination from an abandoned landfill site in Delaware County, IndianaDay, Stephen Wayne 03 June 2011 (has links)
Groundwater contamination by landfill generated leachate is a problem that is increasingly addressed for proposed and existing landfill sites. This thesis examines groundwater contamination movement from the abandoned Delaware County Municipal landfill. The site is located in the crest of a highly permeable sand and gravel glacial esker which allows for rapid movement of ground water and any contaminants introduced into it.The landfill site was originally investigated in the late 1970's by Ed Lusch, a graduate student at the Ball State University Geology Department. That study showed some indications of ground water contamination movement to about 400 feet west of the site, in the direction of ground water flow. This indicated position of a contamination plume suggested that leachate, generated from the landfill site, had moved to that position since (or possibly before) the closing of the landfill in 1971.The present study, using a combination of surface resistivity methods, on-site test wells, and chemical analyses of ground water, attempted to determine the degree of contamination movement from the site since the original study and the extent to which the local aquifer had been affected. Results of this investigation revealed an apparent slow movement of leachate from the landfill westward towards the Mississinewa River, also in the direction of ground water flow. Surface resistivity methods of this study revealed the plume of contamination (indicated by resistivity "low" area) to now exist at approximately the same location as indicated in the earlier study, but to have expanded laterally. This investigation also found indications of the contamination plume well into the underlying fractured dolostone.Chemical analyses of nearby residential wells also revealed slightly elevated amounts of chloride, ammonia and specific conductivity in the ground water of the glacial esker south of the abandoned landfill site. The presence of three other dump sites, including a sludge dump, along the esker south of the landfill, offers questions as to the source of ground water contamination.Ball State UniversityMuncie, IN 47306

Biosensorsystem för övervakning av vattenkvalitetAlessandro, Martini January 2012 (has links)
Sweden's drinking water quality is considered to be high partly due to a high quality of the raw water and a well developed sewage infrastructure. Despite this, there is water contamination that could be prevented by installation of a sophisticated early warning system. Some of the major players in the production of drinking water have already invested in different types of early warning systems to ensure drinking water of high quality. There are various forms of early warning systems where automatic monitoring of E. Coli is an interesting alternative. Today's technology allows for this type of measurement, but it is often expensive and sometimes slow. This work aims to investigate whether the phenomenon of Extraordinary Optical Transmission (EOT) can be used as the technology of sensors for real-time measurement of indicators of fecal contaminated water. EOT is a transmissions peak which arises due to plasmon resonance. The study was conducted by performing verification measurements by spectrophotometry to detect the EOT. After this biomeasurements where done to show that the surface of the sample can be functionalized to provide the opportunity to choose which analytes should be detected. The verifying measurements showed that EOT was detected for one of the five samples where the hole size was 400 nm and the hole spacing was 600 nm. This is due to the high signal strength and to the fact that the phenomenon occurred within the measurement range of the instruments. Samples where designated with the name sPa where s indicates hole size in nm and a hole spacing in nm. The signal strength of the sample 140P600 and 200P600 was too low to determine whether EOT occurred. For samples 400P1000 and 600P1000 calculations showed that EOT occurred outside the measurement range, which meant that EOT could not be measured. Sample 400P600 was chosen to implement bio-measurements wherein the surface was functionalized with biotin Bovine serum albumin (B-BSA) to later bind with fluidMAG Streptavidin. Both B-BSA and streptavidin was bound as shown by the kinetic measurements.
