Biophysical studies of cholesterol in unsaturated phospholipid model membranes

Indiana University-Purdue University Indianapolis (IUPUI) / Cellular membranes contain a staggering diversity of lipids. The lipids are heterogeneously distr ibuted to create regions, or domains, whose physical properties differ from the bulk membrane and play an essential role in modulating the function of resident proteins. Many basic questions pertaining to the formation of these lateral assemblies remain. T his research employs model membranes of well - defined composition to focus on the potential role of polyunsaturated fatty acids (PUFAs) and their interaction with cholesterol (chol) in restructuring the membrane environment. Omega - 3 (n - 3) PUFAs are the main bioactive components of fish oil, whose consumption alleviates a variety of health problems by a molecular mechanism that is unclear. We hypothesize that the incorporation of PUFAs into membrane lipids and the effect they have on molecular organization may be, in part, responsible. Chol is a major constituent in the plasma membrane of mammals. It determines the arrangement and collective properties of neighboring lipids, driving the formation of domains via differential affinity for different lipids . T he m olecular organization of 1 -[ 2 H 31 ]palmitoyl -2- eicosapentaenoylphosphatidylcholine (PEPC - d 31 ) and 1 -[ 2 H 31 ]palmitoyl -2- docosahexaenoylphosphatidylcholine (PDPC -d 31 ) in membran es with sphingomyelin (SM) and chol (1:1:1 mol) was compared by solid - state 2 H NMR spectroscopy. Eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids are the two major n - 3 PUFAs found in fish oil, while PEPC -d 31 and PDPC -d 31 are phospholipids containing the respective PUFAs at the sn - 2 position and a perdeuterated palmitic acid a t the sn - 1 position . Analysis of s pectra recorded as a function of temperature indicate s that in both cases, formation of PUFA - rich (less ordered) and SM - rich (more ordered) domains occurred. A surprisingly substantial proportion of PUFA was found to infil trate the more ordered domain. There was almost twice as much DHA (65%) as EPA (30%) . The implication is that n - 3 PUFA s can incorporate into lipid rafts, which are domains enriched in SM and chol in the plasma membrane, and potentially disrupt the activity of signaling proteins that reside therein. DHA, furthermore, may be the more potent component of fish oil. PUFA - chol interactions were also examined through affinity measurements. A novel method utilizing electron paramagnetic resonance (EPR) was develope d, to monitor the partitioning of a spin - labeled analog of chol , 3β - doxyl - 5α - cholestane (chlstn), between large unilamellar vesicles (LUVs) and met hyl - β - cyclodextrin (mβCD). The EPR spectra for chlstn in the two environments are distinguishable due to the substantial differences in tumbling rates , allowing the population distribution ratio to be determined by spectral simulation. Advantages of this approach include speed of implementation and a vo idance of potential artifact s associated with physical separation of LUV and mβCD . Additionally, in a check of the method, t he relative partition coefficients between lipids measured for the spin label analog agree with values obtained for chol by isothermal titration calorimetry (ITC). Results from LUV with different composition confirmed a hierarchy of decreased sterol affinity for phospholipids with increasing acyl chain unsaturation , PDPC possessing half the affinity of the corresponding monounsaturated phospholipid. Taken together, the results of these studies on model membranes demonstrate the potential for PUFA - driven alteration of the architecture of biomembranes, a mechanism through which human health may be impacted.

Tunable High-Field/ High-Frequency ESR and High-Field Magnetization on Single-Molecule Clusters

In this work, low dimensional iron group clusters have been studied by application of high magnetic fields. The magnetization has been probed with an MPMS as function of temperature and field. The combination with pulse field measurements up to 52\,T allowed determination of the magnetic exchange coupling parameters, and to probing the effective spin of the ground state. The main focus was on tunable high-field/high-frequency (tHF) ESR in static fields < 17 T and pulse field ESR up to 36 T. This magnetic resonance method has been used for the characterization of the local magnetic properties: The detailed analysis of the field dependence of dedicated spin states allowed to determine the magnetic anisotropy and g-factors. The results were analyzed in the framework of the appropriate effective spin Hamiltonians in terms of magnetization fits and ESR spectrum simulations.

<b>Molecular investigation of the multi-phase photochemistry of environmental aquatic systems</b>

<p dir="ltr">The chemical constituents of terrestrial and atmospheric waters originate from biomass burning, fertilizer runoff, and anthropogenic activity, among other sources, and their multi-phase chemistry is complex. Sunlight plays an essential role in aquatic chemistry. Photosensitizers in terrestrial and atmospheric waters absorb light to form highly reactive species such as triplet excited carbon (<sup>3</sup>C*), hydroxyl radical (•OH), and singlet oxygen (<sup>1</sup>O<sub>2</sub>), driving the photochemical transformations of dissolved organic matter (DOM) in the aqueous phase. Of note, these reactive species transform DOM compounds that do not undergo direct photolysis. DOM frequently undergoes a change in optical properties following photochemical processing, with implications for air quality, water quality, and human and animal health. The presence of inorganic minerals, such as the fertilizer compound struvite, in terrestrial or atmospheric waters introduces further complexity and impacts the photochemical processes that occur. Simplified proxy systems are created in the laboratory to simulate aquatic photochemical processes and evaluate the formation and/or photodegradation of photoproducts. These mixtures typically consist of a representative organic carbon (OC) compound and a photosensitizer, along with struvite or another inorganic mineral.</p>

Detailing radio frequency controlled hyperthermia and its application in ultrahigh field magnetic resonance

Die vorliegende Arbeit untersucht die grundsätzliche Machbarkeit, Radiofrequenzimpulse (RF) der Ultrahochfeld (UHF) Magnetresonanztomographie (MRT) (B0≥7.0T) für therapeutische Verfahren wie die RF Hyperthermie oder die lokalisierte Freigabe von Wirkstoffträgern und Markern zu nutzen. Im Rahmen der Arbeit wurde ein 8-Kanal Sened/Empfangsapplikator entwickelt, der bei einer Protonenfrequenz von 298MHz operiert. Mit diesem weltweit ersten System konnte in der Arbeit experimentell bewiesen werden, dass die entwickelte Hardware sowohl zielgerichtete lokalisierte RF Erwärmung als auch MR Bildgebung und MR Thermometrie (MRTh) realisiert. Mit den zusätzlichen Freiheitsgraden (Phase, Amplitude) eines mehrkanaligen Sendesystems konnte aufgezeigt werden, dass der Ort der thermischen Dosierung gezielt verändert bzw. festgelegt werden kann. In realitätsnahen Temperatursimulationen mit numerischen Modellen des Menschen, wird in der Arbeit aufgezeigt, dass mittels des entwickelten Hybridaufbaus eine kontrollierte und lokalisierte thermische Dosierung im Zentrum des menschlichen Kopfes erzeugt werden kann. Nach der erfolgreichen Durchführung dieser Machbarkeitsstudie wurden in theoretischen Überlegungen, numerischen Simulationen und in ersten grundlegenden experimentellen Versuchen die elektromagnetischen Gegebenheiten von MRT und lokal induzierter RF Hyperthermie für Frequenzen größer als 298MHz untersucht. In einem Frequenzbereich bis zu 1.44GHz konnte der Energiefokus mit Hilfe spezialisierter RF Antennenkonfigurationen entscheidend weiter verkleinert werden, sodass Temperaturkegeldurchmesser von wenigen Millimetern erreicht wurden. Gleichzeitig konnte gezeigt werden, dass die vorgestellten Konzepte ausreichende Signalstärke der zirkular polarisierten Spinanregungsfelder bei akzeptabler oberflächlicher Energieabsorption erzeugen, um eine potentielle Machbarkeit von in vivo MRT bei B0=33.8T oder in vivo Elektronenspinresonanz (ESR) im L-Band zu demonstrieren. / The presented work details the basic feasibility of using radiofrequency (RF) fields generated by ultrahigh field (UHF) magnetic resonance (MR) (B0≥7.0T) systems for therapeutic applications such as RF hyperthermia and targeted drug delivery. A truly hybrid 8-channel transmit/receive applicator operating at the 7.0T proton MR frequency of 298MHz has been developed. Experimental verification conducted in this work demonstrated that the hybrid applicator supports targeted RF heating, MR imaging and MR thermometry (MRTh). The approach offers extra degrees of freedom (RF phase, RF amplitude) that afford deliberate changes in the location and thermal dose of targeted RF induced heating. High spatial and temporal MR temperature mapping can be achieved due to intrinsic signal-to-noise ratio (SNR) gain of UHF MR together with the enhanced parallel imaging performance inherent to the multi-channel receive architecture used. Temperature simulations in human voxel models revealed that the proposed hybrid setup is capable to deposit a controlled and localized RF induced thermal dose in the center of the human brain. After demonstrating basic feasibility, theoretical considerations and proof-of-principle experiments were conducted for RF frequencies of up to 1.44GHz to explore electrodynamic constraints for MRI and targeted RF heating applications for a frequency range larger than 298MHz. For this frequency regime a significant reduction in the effective area of energy absorption was observed when using dedicated RF antenna arrays proposed and developed in this work. Based upon this initial experience it is safe to conclude that the presented concepts generate sufficient signal strength for the circular polarized spin excitation fields with acceptable specific absorption rate (SAR) on the surface, to render in vivo MRI at B0=33.8T or in vivo electron paramagnetic resonance (EPR) at L-Band feasible.

A Tailor-Made Approach for Thin Films and Monolayer Assemblies of bis(oxamato) and bis(oxamidato) Transition Metal Complexes

The present work is dealing with the synthesis and characterization of mono- to trinuclear Cu(II)- and Ni(II)-containing bis(oxamato) and bis(oxamidato) complexes, respectively. It will be derived to which extent the spin density distribution of mononuclear complexes, determined by electron paramagnetic resonance studies experimentally and calculated by quantum mechanical calculations, can be regarded as a measure of the magnitude of magnetic superexchange interactions of corresponding trinuclear complexes. The usability of tailor-made trinuclear complexes for their deposition in form of thin film by spin-coating is described as well as the magneto-optical characterization of these thin films. It is shown, that the tailor-made functionalization of mono- to trinuclear bis(oxamidato) complexes with long alkyl chains is suited to allow these complexes to assembly in monolayers on, e.g., HOPG(0001) (highly-oriented pyrolytic graphite), whereas the functionalization of the alkyl chains with S atoms allows the generation of self-assembled monolayers on metallic gold. Furthermore it is shown, that the functionalization of mono- and trinuclear Cu(II)-containing bis(oxamato) complexes with ferrocenediyle fragments allows to modify the magnetic properties by making use of the oxidation process Fe(II)/Fe(III). / Die vorliegende Arbeit befasst sich mit der Darstellung und Charakterisierung ein- bis dreikerniger Cu(II)- bzw.
