Plaquettes et neutrophiles : acteurs clés dans le choc allergique dépendant des IgG / Platelets and neutrophils : key players in IgG-induced anaphylaxis

Le choc anaphylactique est une réaction allergique systémique qui survient en quelques minutes et pouvant être fatale. Mon travail de thèse s’articule autour de deux projets dont la finalité est de mieux comprendre le mécanisme physiopathologique. La première partie de ce travail consiste à étudier in vivo chez la souris les contributions des récepteurs Fc aux IgG (FcγRs), des cellules effectrices et des médiateurs contribuant dans un modèle d’anaphylaxie systémique passive induit par une sous-classe particulière d’IgG : des IgG1, des IgG2a ou des IgG2b monoclonales dirigées contre un même antigène. Cette étude a permis de démontrer que le FcγRIII, les neutrophiles et les monocytes/macrophages sont les acteurs majoritaires quelque soit la sous-classe d’IgG de souris ; en revanche, la participation des basophiles ainsi que la contribution relative des médiateurs histamine et PAF sont dépendantes de la sous-classe d’IgG utilisée. La deuxième partie de ce travail consiste à étudier plus particulièrement la population plaquettaire dans un modèle de souris humanisées. Contrairement à la souris, les plaquettes humaines expriment un FcγR, le FcγRIIA déjà identifié comme acteur clé de l’anaphylaxie. Un modèle de choc allergique induit par des IgG humaines dans des souris transgéniques pour le FcγRIIA m’a permis de tester l’hypothèse suivant laquelle les plaquettes participent à l’initiation et/ou à la propagation de la réaction. Ce modèle a permis de mettre en évidence une thrombocytopénie sévère, des complexes plaquettes-leucocytes circulants et de montrer que le transfert de plaquettes ou de leur surnageant restaure les signes cliniques du choc allergique. / Anaphylaxis is a systemic hyperacute allergic reaction that occurs within minutes and can be fatal. The aim of my PhD project is to investigate the physiopathological mechanisms underlying anaphylaxis induction. The first part of my work focused on the contribution of FcγRs, effector cells and mediators in passive murine models of systemic anaphylaxis induced by the different subclasses of mouse specific IgG ; directed against the same antigen: IgG1, IgG2a or IgG2b. This study demonstrated that FcγRIII, neutrophils and monocytes/macrophages are the key players of anaphylaxis induction whatever the mouse IgG subclasses used. On the contrary, basophil participation and the relative contribution of histamine and PAF are IgG subclass dependent. The second part of this work examined the role of platelets in anaphylaxis using a humanized mouse model. Opposing the murine situation, human platelets express an IgG receptor, FcγRIIA. This receptor has already been identified as a key player in anaphylaxis. Using aggregated human IgG to induce anaphylaxis in mice transgenic for FcγRIIA, we tested our hypothesis that platelets contribute to the initiation and/or the propagation of this reaction. Anaphylaxis in this model was accompanied by a severe thrombocytopenia, the presence of circulating platelet-leukocyte complexes and activated platelets. I further demonstrated that the transfer of platelets or their activated supernatent into resistant mice restored features of anaphylactic shock.

The investigated properties of composite belongs compressive strength, tensile strength in the flexural and tensile strength in the transverse, static modulus of elasticity in compressive strength, dynamic modulus of elasticity determined using NDT tests. The reference concrete mixture is certified as structural concrete of the class, which is commonly used on construction sites.

Sperm-­-egg membrane interaction and fusion is mediated by various molecules of the different protein network that are located on both egg and sperm membrane. So far, many proteins have been selected to be fusion candidates, some of them (Izumo1, CD9, Juno) were proven to be essential, whereas others were discovered to play an unsuspected new active role (CD46, tetraspanins). After the adhesion of sperm to an egg, Juno located on the oolema associates with monomeric Izumo1 on sperm membrane, which is results in Izumo1 dimerization following quick removal of Juno from the egg surface as described in mouse. It implies that additional receptor on the egg membrane is required to play a role in sperm-­-egg fusion. To find a human fusogenic receptor for IZUMO1 protein we used one-­-bead-­-one-­-compound (OBOC) assay, a random screening approach. A bead, fulfilling all the requirements when interacting with the human sperm, carried a peptide sequence showing homology with the conserved Ig domain of the human specific Fc receptor-­-like protein 3 (FcRL3). FC Rosengård bedriver flertalet sociala projekt. Ett av dessa projekt är Football for Life som i Sydafrika har som målsättning att stärka unga kvinnors självbild och rättigheter. Studien “Football for Life, så mycket mer än bara fotboll” ämnar att utvärdera denna verksamhet med hjälp av Teaching Personal and Social Responsibility modellen. Detta för att visualisera i vilken mån deltagarna i verksamheten erhåller ett ökat personligt- och socialt ansvarstagande. Studiens slutgiltiga målsättning är att stärka argumentet för att en liknande verksamhet ska initieras i Sverige genom att påvisa vilken inverkan Football for Life har haft för de deltagande tjejerna i Sydafrika. Tvärsnittsstudien är av kvantitativ karaktär där enkäter har varit den primära källan till empiri. Vidare har även relevant litteratur på ämnet Positive Youth Development bidragit till den slutgiltiga produkten. De huvudsakliga aspekter som undersökts i studien är deltagarnas upplevda effort, caring/helping, respect och self-direction kopplat till hur Football for Life har/har haft en inverkan på individen inom dessa områden. Enkäten distribuerades av personalen på plats i Sydafrika där en grupp individer som inte har deltagit i verksamheten agerade som kontrollgrupp. Dessa deltog i studien tillsammans med deltagare som i olika utsträckning varit en del av Football for Life’s verksamhet i olika utsträckning. På förhand förväntades de som haft ett djupare engagemang i Football for Life uppvisa större upplevt personligt- och socialt ansvarstagande jämfört med de som inte deltagit i verksamheten. Resultaten påvisar i viss mån att så inte var fallet, den största skillnaden mellan deltagarnas upplevda personliga- och sociala ansvarstagande återfanns mellan de som inte deltagit vid något tillfälle och de som deltagit ett fåtal gånger. Det återfanns dock skillnader mellan de som inte deltagit och de som haft en stor involvering i verksamheten, men inte i den utsträckning som kunnat tänkas på förhand. Vidare förs en diskussion som resonerar kring de resultat som framkom för att besvara bad resultatet kan bero på. Utöver detta behandlas även Teaching and Personal Responsibility modellens lämplighet vid utvärdering av ett program med anknytning till Positive Youth Development. / Football for Life in South Africa is one of many social project that is managed by the football club FC Rosengård. Football for Life is a Positive Youth Development program which aims to strengthen the young female participants regarding their confidence as well as to educate them regarding their personal rights in the South African society. The sex roles in South Africa is in many regards skew, especially seeing that women are barely allowed to participate in physical activities, which is one of FC Rosengård’s major driving forces for managing Football for Life. In order to ensure the quality of the activities, as well as to examine the possibilities of initiating a project of similar characteristics in Sweden, this essay was set of on the behalf of FC Rosengård. By studying the participants at scene in South Africa by utilising a questionnaire, their perceived personal- and social responsibility was investigated. The participants in the study was categorized depending on their involvement in Football for Life in order to visualise whether the project has a positive effect regarding the development of personal- and social responsibility.

Étude de la Structure-Fonction du Prosegment et du domaine CHRD de la PCSK9 humaine

L’excès des particules de LDL dans le sang constitue un facteur de risque majeur dans le développement des maladies cardiovasculaires. Dans ce contexte, nous étudions la protéine PCSK9 qui favorise directement ce facteur de risque. Cette protéine est sécrétée en majorité au niveau du foie par les hépatocytes et possède la capacité de reconnaître et de lier le récepteur LDLR. Le rôle premier de ce dernier est d’éliminer les particules de LDL circulant dans le plasma. Ainsi, lorsque la PCSK9 forme un complexe avec le LDLR et l’amène à la dégradation, la conséquence directe de la diminution des ces récepteurs est une accumulation malsaine des particules LDL dans le plasma. L’importante implication de la PCSK9 dans le métabolisme des lipides nous a menés vers des recherches de caractérisation de cette protéine ainsi que dans l’étude de son mode d’action. La PCSK9 est composée de trois domaines et notre intérêt s’est porté sur l’étude structure-fonction des deux domaines dont la fonction était inconnue, soit le domaine en N-terminal : le prodomaine et de son domaine en C-terminal : CHRD. Le premier article présenté dans cette thèse révèle l’importance d’une région acide (acide aminés 33-58) régulatrice de l’activité de la PCSK9 localisée en N-terminal du prodomaine ainsi que l’effet du pH acide, équivalent à celui des endosomes tardifs, qui accroît la capacité de la PCSK9 à induire la dégradation du LDLR. Le deuxième article dissèque davantage la structure de la PCSK9 et met en lumière la différence des prérequis structurels de la région ‘’Hinge’’ ainsi que du module M2, composant du domaine CHRD, dans la voie intracellulaire et la voie extracellulaire d’activité de la PCSK9. La mutation R434W localisée dans la région ‘’Hinge’’ résulte dans une inhibition totale de l’activité intracellulaire de la PCSK9 tandis que son activité extracellulaire est réduite à ~70%. Contrairement, la perte du module M2 du domaine CHRD est bien tolérée par la PCSK9 lors de son activité intracellulaire mais totalement inhibitrice pour son activité extracellulaire. Le troisième article se distingue en présentant une nouvelle stratégie d’inhibition de l’activité de la PCSK9 en utilisant une chimère composée de la fraction Fc de l’immunoglobuline IgG1 humaine couplée avec le prodomaine de la PCSK9. La protéine fusion Fcpro lie directement la PCSK9, crée un encombrement structurel qui résulte dans une régulation négative l’activité de la PCSK9. En résumé, nous présentons dans cette thèse, trois manuscrits qui apportent une contribution à la connaissance des composantes structurelles de la PCSK9 et leur implication dans le rôle de la protéine en tant que régulateur négatif du LDLR. / Hypercholesterolemia is one of the major risk factors leading to cardiovascular disease. In this context, we focused our study on a protein that directly influences hypercholesterolemia: PCSK9. Since 2003, the coding gene for PCSK9 has been identified as the third locus responsible for familial hypercholesterolemia (FH3). PCSK9 is a protein secreted mostly from the liver by hepatocytes and has the capacity to recognize, bind and direct to degradation the LDLR receptor. The latter is responsible for the elimination the LDL particles from the plasma. The direct consequence of the LDLR degradation induced by PCSK9 is the harmful accumulation of the bad cholesterol in the blood. Since PCSK9 activity has undesirable consequences on lipid metabolism homeostasis, we directed our research to characterize this protein to better understand its mechanism of action. Three domains compose PCSK9 structure and we focused on the ‘’structure-function study’’ of two domains, of which roles were still unknown: the prodomain located at the N-terminal extremity and the CHRD domain at the C-terminus of PCSK9. The first manuscript presented in this thesis brings to light the importance of the acidic N-terminal sequence of the prosegment (amino acids 33-58) and its effect on the activity of PCSK9. It also presents a novel mechanism for fine-tuning the activity of PCSK9, which is enhanced at acidic pHs close to those of late endosomes. The second manuscript dissects further the PCSK9 structure, revealing that the structural requirements of the hinge and the M2 module located in the CHRD domain are not the same for the intracellular and extracellular pathways of PCSK9-induced LDLR degradation. Although the R434W natural mutation in the hinge region is absolutely deleterious for the intracellular activity of PCSK9, it reduces by ~70% the extracellular one. In contrast, the loss of M2 module of the CHRD domain is tolerated for the intracellular activity of PCSK9 but not for the extracellular one. The third manuscript demonstrates for the first time that a chimera containing the prosegment (Fcpro) directly binds PCSK9 and effectively acts as a negative regulator (inhibitor) of its ability to induce LDLR degradation. Our work presents a new strategy to develop such inhibitors by interfering with the structure of PCSK9 and exploiting the properties of the PCSK9 prosegment and the advantage of its fusion to a humanized Fc of IgG1. In summary, the present research data sheds new light on the functional contribution of the prodomain and the CHRD domain of PCSK9.
