Effect of mixing elements on granule formation in hot melt twin screw granulation

Yes / Twin screw granulation (TSG) has been applied to wet granulation, although its application in melt granulation has been more limited. This work explores potential advantages of hot melt granulation using twin screw extrusion. Four main operating and formulation parameters were investigated: screw speed, number of mixing elements, temperature, and binder percentage. Combinations of these factors were then studied to determine their impact on the quantity and characteristics of granules within the desired size range of 125 - 1000 µm. A screening design of experiments (DOE) study was used with each factor set at three levels, to investigate individual factor effects and interactions. Two types of mixing elements were studied: kneading block (KB) and chaotic elements. The type and number of mixing elements were found to be paramount in contributing to the quantity and characteristics of granules formed. Results obtained agreed with previous findings in literature on the influence of different screw elements on the characteristics of granules formed by twin screw granulation. Additionally, the study revealed the unique impact which different mixer elements have on both granule production and characteristics. Depending on the specific need or use of granules in required applications, the granulation process can be effectively designed to meet the end product quality and outcome.

Data mining of fractured experimental data using neurofuzzy logic-discovering and integrating knowledge hidden in multiple formulation databases for a fluid-bed granulation process.

No / In the pharmaceutical field, current practice in gaining process understanding by data analysis or knowledge discovery has generally focused on dealing with single experimental databases. This limits the level of knowledge extracted in the situation where data from a number of sources, so called fractured data, contain interrelated information. This situation is particularly relevant for complex processes involving a number of operating variables, such as a fluid-bed granulation. This study investigated three data mining strategies to discover and integrate knowledge "hidden" in a number of small experimental databases for a fluid-bed granulation process using neurofuzzy logic technology. Results showed that more comprehensive domain knowledge was discovered from multiple databases via an appropriate data mining strategy. This study also demonstrated that the textual information excluded in individual databases was a critical parameter and often acted as the precondition for integrating knowledge extracted from different databases. Consequently generic knowledge of the domain was discovered, leading to an improved understanding of the granulation process.

No Brasil a esquistossomose é causada pela espécie Schistosoma mansoni e leva principalmente à formação de granulomas e fibroses, hipertensão portal e hepatoesplenomegalia. O praziquantel (PZQ) é o fármaco de primeira escolha para o tratamento e possui baixa solubilidade aquosa, sendo necessárias altas doses para ação terapêutica. A taxa de dissolução do PZQ é fator limitante para a sua biodisponibilidade, portanto, melhorar sua solubilidade é fundamental. O objetivo deste trabalho foi aumentar a solubilidade do PZQ pela obtenção de grânulos contendo dispersões sólidas deste fármaco pelo processo hot melt em leito fluidizado. Além disto, buscou-se estudar a fluidodinâmica e as variáveis do processo de granulação. Inicialmente, dispersões sólidas foram preparadas por simples fusão em polietilenoglicol (PEG) 4000 e PEG 6000 nas proporções 2:1; 1:1; 1:2; 1:5 e 1:10 e estas foram caracterizadas por teste de solubilidade, difração de raios-X, espectroscopia em infravermelho e análises térmicas (DSC e TG). Foram obtidas dispersões sólidas com uma solubilidade duas vezes maior do PZQ, uma menor cristalinidade, uma possível interação fármaco-carreador e sem degradação ou alteração nos principais grupos funcionais. Com estes resultados, a dispersão sólida 1:10 PZQ:PEG 6000 foi escolhida para a etapa da granulação. Os granulados foram obtidos utilizando o planejamento fatorial Box Behnken com as seguintes variáveis: % de dispersão sólida, proporção PZQ:PEG das dispersões e carga de lactose spray-dried (SD) (g). A caracterização dos grânulos foi realizada pelos testes: granulometria, densidade aparente, densidade compactada, fator de Hausner, índice de Carr, ângulo de repouso e doseamento do fármaco, sendo todas estas propriedades avaliadas pela técnica de superfície de resposta (ANOVA). A análise estatística demonstrou que as características dos granulados são, na maioria dos casos, dependentes da % de dispersão sólida e da proporção de PZQ:PEG, sendo satisfatórios e toleráveis os resultados do fluxo dos granulados. Os três melhores granulados foram escolhidos para as análises de microscopia eletrônica de varredura, espectroscopia em infravermelho, difração de raios-X, análise térmica e perfil de dissolução. Estas análises mostraram que nos grânulos houve diminuição da cristalinidade, possíveis interações entre PZQ, PEG e lactose, além do grande aumento no perfil de dissolução. Com estes resultados, confirmam-se as vantagens da granulação por hot melt em leito fluidizado e a importância do estudo de suas variáveis. / In Brazil, schistossomiasis is mainly attributed to the species Schistosoma mansoni, which causes granulomas and fibrosis, portal hypertension and hepatosplenomegaly. Praziquantel (PZQ) is the first choice in the treatment; however, it has low water solubility, thus requiring high doses for a proper therapeutical result. The PZQ dissolution rate is the limiting factor for its bioavailability; therefore, it is fundamental to improve of this drug solubility. The objective of this work was to increase the PZQ solubility with the production of granules containing its solid dispersions, prepared by hot melt process in fluidized bed. Moreover, the fluid dynamic and several variables of the process were studied. First, solid dispersions were prepared by mixing with melted polyethylene glycol (PEG) 4000 and PEG 6000 at the ratios of 2:1; 1:1; 1:2; 1:5 and 1:10. These were characterized by solubility test, x-ray powder diffraction, fourier transform infrared spectroscopy and thermal analyses (DSC and TG). Solid dispersions were obtained with PZQ solubility two times higher than the pure drug, lower crystallinity and without degradation or alteration in the main functional groups. With these results, the solid dispersion 1:10 PZQ:PEG 6000 was chosen for the studies of granulation. The granulates were obtained using the Box Behnken factorial design with the following independent variables: % of solid dispersion, ratio PZQ:PEG in the dispersions and amount of spray-dried lactose (g). The granules characterization was made by the tests of granulometry, bulk density, tapped density, Hausner ratio, Carr index, angle of repose and PZQ assay. In addition, all these properties were evaluated by response surface technique (ANOVA). The statistical analysis, in most cases, showed that the granules characteristics depend significantly on the % of solid dispersion and on the ratio PZQ:PEG. The results on the granules flow properties are satisfactory and acceptable for tableting. The three granulates with best properties were submitted to scanning electron microscopy, fourier transform infrared spectroscopy, x-ray powder diffraction, thermal analysis and dissolution test. These analyses showed that there were, in the granules, decrease of crystallinity, possible interaction among PZQ, PEG and lactose, and also, greater increase in the dissolution test. The advantages of fluid bed hot melt granulation and the importance of studying the process variables are confirmed by the results in this work.

Granulation de suspensions concentrées UO2/PuO2 : application à l'élaboration de compacts granulaires denses par pressage et à leu caractérisation structurale post frittage / Granulation of concentrated UO2/PuO2 suspensions : application to the shaping of granular compacts by pressing and post-sintering microstructural characterization

Le procédé actuel de fabrication des combustibles nucléaires MOX (UO2-PuO2) est réalisé exclusivement par voie sèche (broyage-tamisage des poudres, pressage et frittage). Afin d’améliorer ce procédé, des recherches sont menées sur le développement d’un procédé de fabrication du MOX par voie liquide. Ce procédé vise à réduire l’empoussièrement des boîtes à gants, améliorer l’homogénéité U/Pu et diminuer la quantité de défauts (fissures, éclats) des pastilles frittées, afin de minimiser le taux de pastilles rebutées. Dans cette optique, le proceed voie liquide étudié consiste à préparer une suspension aqueuse à partir des poudres brutes d’oxydes (mélange d’eau, d’additifs organiques et de poudres), puis à lui faire subir une granulation. Les granules obtenus sont ensuite pressés et frittés. Le procédé de granulation étudié est un procédé innovant de granulation cryogénique, consistant à atomiser la suspension dans de l’azote liquide puis à lyophiliser les granules gelés ainsi formés. L’étape clé du procédé est la préparation de suspension aqueuse de poudres UO2-PuO2, qui doit être dispersée, homogène, stable et suffisamment fluide pour l’étape de granulation. Une étude préliminaire a été réalisée avec des poudres simulantes, choisies pour leurs propriétés en suspension : TiO2 et Y2O3 pour simuler respectivement UO2 et PuO2. Une fois maîtrisé avec les poudres simulantes, ce procédé a été étudié avec UO2 et PuO2 pour déterminer ses conditions optimales de mise en oeuvre. La dispersion de suspensions d’UO2 et/ou de PuO2 a été étudiée par acoustophorométrie et rhéologie afin d’optimiser leur formulation, ainsi que l’étape de granulation cryogénique. Enfin, des pastilles d’UO2 et d’UO2-PuO2 ont été formées à partir des granules, puis leur frittage et leur microstructure ont été étudiés. / The current manufacturing process for MOX nuclear fuels (UO2-PuO2) is carried out by dry route exclusively (grinding, sieving, pressing and sintering). In order to improve this process, research is conducted to develop a liquid route MOX manufacturing process. It aims to reduce glove boxes dusting, increase U/Pu homogeneity and reduce the amount of defects (cracks, voids) in sintered pellets, in order to minimize scraps. In this scope, the liquid process studied consists in the preparation of an aqueous suspension from the raw oxide powders (mix of water, organic additives and powders), which is then granulated. The resulting granules are pressed into pellets and sintered. The granulation process studied is an innovative freeze granulation process that implies to spray the suspension in liquid nitrogen and then freeze-dry the frozen granules that are formed.The key step of the process is the preparation of aqueous suspension of UO2-PuO2 powders, which must be dispersed, homogeneous, stable and fluid enough for the granulation step. A preliminary study was carried out using surrogate powders, chosen for their properties in suspension: TiO2 and Y2O3 to surrogate UO2 and PuO2 respectively. Once mastered with surrogate powders, this process was studied with UO2 and PuO2 to determine its optimal working conditions. The dispersion of UO2 and/or PuO2 suspensions was studied by acoustophorometry and rheology in order to optimize their formulation, as well as the freeze granulation step. Finally, UO2 and UO2-PuO2 pellets were shaped from the granules, and their sintering and microstructure were studied.

Dry granulation process and compaction behavior of granulated powders / Granulation sèche par compactage à rouleaux et comportement en compression des granulés

Les solides divisés telles que les poudres pharmaceutiques nécessitent souvent des processus d'agrandissement de taille par agglomération pour améliorer leur comportement mécanique, notamment la coulabilité. Pour cette raison, le procédé de "granulation en voie sèche" est utilisé dans l'industrie pharmaceutique. Le procédé consiste à comprimer la poudre en la faisant passer entre deux rouleaux séparés par un entrefer, pour produire des plaquettes qui sont ensuite broyées en granulés et comprimés en compacts. Dans ce procédé, l'existence de différents modèles de compacteurs à rouleaux et de systèmes de broyage d’une part, et l'interaction entre les paramètres des procédés et des propriétés des produits (plaquettes, granulés et comprimés) d’autre part, rendent difficile la compréhension des phénomènes et des mécanismes sous-jacents. En particulier, le procédé entraîne une perte de résistance mécanique des comprimés formés à partir de granulés (comparativement à celles des comprimés de poudres non-granulés) et ce phénomène est encore mal compris. Ces aspects sont étudiés dans ce travail de thèse en menant des caractérisations expérimentales et des modélisations numériques permettant de mieux comprendre les modifications micro et macro structurales des poudres mises en forme par granulation sèche. Le but ultime est de progresser dans la compréhension des relations "propriétés des poudres - paramètres des procédés". Enfin, la compréhension des différences de comportement en compression de poudres granulées et non-granulées est menée à l’aide d’une modélisation du comportement dans le cadre de la mécanique des milieux continus poreux. / Particulate solids such as pharmaceutical powders often require size enlargement processes to improve the manufacturing properties like flowability. For that reason, dry granulation by roll compaction has been widely used in the pharmaceutical industry. The process consists of compressing powders between two counter-rotating rolls to produce ribbons that will be subsequently milled into granules. The obtained granules are tableted for oral dosage. In this process there are two main limitations: the existence of different designs of the roll compactors, milling systems and the interaction between process parameters and raw material properties are still a challenge and the roll-compaction process leads to an inferior tensile strength of tablets compared with direct compression. These aspects are investigated in this work. In the first part of this thesis, an analysis on the effect of different roll-compaction conditions and milling process parameters on ribbons, granules and tablet properties was performed, highlighting the role of the sealing system and the ribbon density distribution characteristics. In the second part, die compaction of roll-compacted powders, as the last stage of the process, is further investigated in terms of experimental analysis (effect of the granule size and composition and stress transmission measurements) and modelling the compaction behavior of granules.

Poudres et mélanges de poudres pour la fonctionnalisation et l’imprégnation de textiles à sec : formulation de poudres bifonctionnelles / Powder and powder mixture in textile dry impregnation and functionalization process

S’intéressant aux interactions entre la formulation, le procédé et le produit, ce travail original axé sur la fonctionnalisation ignifuge de textiles utilisés dans les transports et le bâtiment porte sur le développement de poudres non feu. La ligne conductrice de cette étude étant de comprendre comment les propriétés ignifuges du textile technique peuvent être influencées par les choix de formulation ou de procédé de mélange. L'identification des besoins, c.-à-d. la réalisation d’un produit fini ignifuge, la conception de mélanges de granulométrie définie qui s’écoulent facilement et qui possèdent une sensibilité à l’inflammation faible, a permis de proposer et d’examiner diverses formulations. En matière de méthodologie ce projet combine l’approche sécurité à l’ingénierie produit. La sélection finale des mélanges sera fonction des résultats de cette analyse technique globale. Le travail expérimental a permis de sélectionner le polymère et l’agent ignifuge à la concentration de 40 % pour l’obtention en mélangeur interne de particules bifonctionnelles répondant aux spécifications fixées. Contrairement aux mélanges à retournement ou haute vitesse, les grains préparés par granulation Hot-Melt ont des liaisons mécaniques renforcées. Ce résultat semble s’expliquer par la bonne affinité, le bon rapport en termes de taille de particules entre les composants et une morphologie favorable à la formation de ce type de structures. Ce mode de fabrication a également permis de lever les deux principaux verrous à savoir préparer une poudre bifonctionnelle à haute teneur en agent ignifuge et réaliser un mélange qui ne se démélange pendant l’étape d’imprégnation du procédé DPreg / This original work deals with the functionalization of textile fire retardant used in transport and the building focused on the development of fire retardant powders while studying the interactions of three criteria: the formulation, process and product. The main line of this study is the understanding and quantification of the flame retardant properties of the textile, which can be influenced by the choice of formulation or mixing process. The identification of needs, i.e. the realization of a finished fire retardant, the design of defined particle size mixtures, which flow easily and possess low sensitivity to inflammation, allowed to propose and consider various formulations. This project methodology is a good example of the combination between two approaches: the product engineering and a safety approach. The final selection of mixtures will be based on the results of a comprehensive technical analysis. The experimental work allows us to select the polymer and the flame retardant at a concentration of 40% in order to obtain in an internal mixer of bifunctional particles meet the specifications set out in this study. Unlike conventional mixtures by inversion or high speed, the grains prepared by Hot-Melt granulation have enhanced mechanical linkages. This result seems to be due to the good affinity and good particle size ratio between the components and a good morphology to the formation of such structures. This manufacturing method also overcomes two obstacles; the preparation of a bifunctional powder with high content of flame retardant and the realization of a mixture which does not demix under the action of the electric field of the DPreg impregnation process

Contribution de l'hypoxie à la cicatrisation cutanée anormale chez le cheval : méthodes physiques d'évaluation

La guérison des plaies cutanées appendiculaires chez le cheval, à la différence de celle des plaies corporelles, se complique régulièrement. Un retard de cicatrisation s’y observe et un tissu de granulation exubérant tend à s’y développer, le tout menant à une cicatrice pathologique hypertrophiée. La pathogénie exacte du tissu de granulation exubérant chez le cheval demeure inconnue à ce jour. Une hypoxie tissulaire pourrait favoriser son développement tout comme elle semble contribuer au développement de cicatrices cutanées pathologiques similaires observées chez l’Homme. L’objectif de cette étude était d’évaluer la perfusion vasculaire et la disponibilité locale en oxygène de plaies cutanées appendiculaires et corporelles en cours de cicatrisation normale et pathologique chez le cheval, à l’aide de la thermographie infrarouge et de la spectroscopie par réflectance dans le proche infrarouge. Six juments âgées de 3 à 4 ans ont été utilisées. Trois plaies cutanées ont été créées à l’aspect dorso-latéral du canon des membres thoraciques (plaies appendiculaires), et sur la paroi costale de l’un des hémithorax (plaies corporelles). Chez chaque jument, un canon a été aléatoirement bandé dans le but d’induire la formation de tissu de granulation exubérant dans les plaies s’y trouvant, tel que rapporté. La perfusion vasculaire et la disponibilité locale en oxygène ont été évaluées séquentiellement par thermographie infrarouge et spectroscopie par réflectance dans le proche infrarouge pour chaque plaie de chaque site (thorax; membre bandé; membre non bandé) au cours du processus de cicatrisation cutanée. Un modèle linéaire à doubles mesures répétées associé à une correction séquentielle de Bonferroni a révélé des différences significatives de perfusion vasculaire et de disponibilité locale en oxygène entre les plaies appendiculaires et corporelles. Ainsi la perfusion vasculaire et la disponibilité locale en oxygène étaient significativement plus élevées dans les plaies corporelles (P<0.05) et la perfusion vasculaire était significativement plus élevée dans les plaies appendiculaires non bandées que dans celles bandées (P<0.05). Nous avons récemment rapporté une plus grande occlusion de la micro-vascularisation au niveau des plaies appendiculaires chez le cheval. Nous rapportons maintenant que la perfusion vasculaire et la disponibilité locale en oxygène sont significativement inférieures dans les plaies appendiculaires, en particulier lorsqu’un tissu de granulation exubérant s’y développe. Compilés, ces résultats sous-tendent l’hypothèse que les plaies appendiculaires souffrent d’une altération de la perfusion vasculaire à l’origine possible d’une hypoxie tissulaire qui pourrait favoriser une cicatrisation cutanée anormale, telle la formation d’un tissu de granulation exubérant. / Cutaneous wound healing in horse limbs is often perturbed by the development of exuberant granulation tissue while body wounds tend to repair uneventfully. A delay in healing is usually observed in horse limb wounds, which tend to develop exuberant granulation tissue and excessive scarring. The events leading to exuberant granulation tissue formation in horses are not fully elucidated. Tissue hypoxia has been proposed as a major contributing factor as it appears to contribute to the development of similar abnormal skin scarring in human. The objective of this study was to investigate skin blood flow and tissue oxygen availability in normal / abnormal healing of full-thickness wounds created on the horse limb and body, using infrared thermography and near infrared reflectance spectroscopy. Six healthy, 3- to 4-year-old mares were used for the experiment. Three full-thickness cutaneous wounds were surgically created on the dorso-lateral surface of each metacarpal area (limb wounds) and on the lateral thoracic wall (body wounds). One randomly chosen distal forelimb was then left to heal by second intention without bandage, while the contralateral limb was bandaged postoperatively to induce the formation of exuberant granulation tissue and lead to excessive scarring. Thermal and spectroscopic data were collected from both anatomic sites (limb without bandage, bandaged limb, thoracic wall) at specific times following wounding. Mean changes in skin blood flow and tissue oxygen availability at specific wound sites over time were compared by use of repeated measures ANOVA with anatomical location and wound management as within-subject factors. A priori contrasts, submitted to Bonferroni sequential correction, were then used to compare pre-selected individual means. The statistical analysis revealed that there were significant differences in skin blood flow and tissue oxygen availability within wounds. Skin blood flow and tissue oxygen availability were significantly increased within body over limb wounds (P<0.05). Skin blood flow was significantly increased within unbandaged limb over bandaged limb wounds (P<0.05). We have recently reported a greater occlusion of microvessels in limb wounds in horses. We report here that skin blood flow and tissue oxygen availability are significantly inferior in limb wounds in horses, especially when exuberant granulation develops. Taken together, these results suggest that equine limb wounds may suffer from altered vascular perfusion and tissue hypoxia, which could favor abnormal skin healing and exuberant granulation tissue development.

Cette étude est dédiée à la compréhension du processus de granulation humide en mélangeurs à haut taux de cisaillement. Une étude systémique et méthodologique a été menée permettant l'investigation de l'influence des paramètres opératoires, de la technologie employée et des propriétés physico-chimiques des matières premières. Cette investigation est réalisé a travers des techniques de caractérisation morphologiques, rhéologiques et mécaniques. En reliant les courbes de couple enregistrés lors de la granulation humide à la cinétique de croissance des granules, aux caractérisations microscopiques et aux propriétés mécaniques des granules la prédiction du comportement lors de la granulation devient possible. La caractérisation des propriétés mécaniques des granules a été étudié à deux échelles: à l'échelle du milieu humide la consistance a été caractérisé par un rheometre à torque et à l'échelle de l'agglomérat sec la résistance mécanique a été caractérisé par des mesures de compression directe des grains individuelles. Cette approche permet d'avoir des informations complémentaires permettant de mieux décrire l'évolution des courbes de couple dépendantes de propriétés de la masse humide et la compétition entre les forces interfaciales et visqueuses conditionnant la qualité des grains secs résultés. Les paramètres investigués par cette approche sont l'effet du taux de remplissage du réacteur, l'effet de la vitesse d'agitation, de la présence et de la conception de l'émotteur, de la conception du réacteur employé, des propriétés physico-chimiques de la solution liante et des propriétés des mélanges binaires des poudres hydro-solubles / hydro-insolubles. / This study is dedicated to the understanding of the wet granulation process in high shear mixers. A systematic study has been carried out that allows the investigation of the influence of operating conditions, technology and physico-chemical properties of the starting materials. This investigation is achieved by morphological, rheological and mechanical characterization methods. By linking recorded torque curves during the granulation process to granule growth kinetics, microscope characterizations and to the end-granule properties granulation outcome prediction becomes possible. The characterization of the mechanical properties has been done at two scales: at the granule bed scale the bulk wet mass consistency has been determined on a mixer torque rheometer, at the granule scale single dry granule direct compression tests were carried out. This approach gives complementary information allowing better description of the torque curves directly related to the wet mass properties and the competition between static and viscous forces conditioning the dry end granule quality. The factors investigated in this study are: the effect of fill ratio, impeller speed, chopper presence and design, mixer design, binder physico-chemical properties and formulation properties for binary water-soluble / water insoluble powder mixtures.
