Activation and allosteric modulation of the [alpha]1 glycine receptor

The glycine receptor (GlyR) is a ligand-gated ion channel and member of the nicotinic acetylcholine receptor superfamily. Glycine and the partial agonist taurine are both believed to be the endogenous ligands of the receptor. Partial agonists have lower efficacies than full agonists, eliciting submaximal responses even at saturating concentrations. Recent evidence suggests that efficacy at these receptors is determined by conformational changes that occur early in the process of receptor activation. We previously identified a mutation of the aspartate-97 residue to arginine (D97R), which produces a spontaneously active mutant with behavior that mimics the effects of saturating glycine concentrations on wildtype (WT) GlyR. This D97 residue is hypothesized to form an electrostatic interaction with arginine-119 on an adjacent subunit to stabilize a closed channel closed state. We found that the disruption of this bond converts taurine into a full agonist and greatly increases the efficacies of other [beta]-amino acid partial agonists. Our findings suggest that the determination of efficacy in the GlyR involves the disruption of an inter-subunit electrostatic interaction soon after binding. We next investigated whether the taurine efficacy could be enhanced by ethanol, a well-studied positive allosteric modulator of receptor function. Whole-cell recordings of WT GlyRs demonstrated that alcohol could potentiate the effect of low concentrations of taurine, but did not increase the efficacy of a saturating concentration. Therefore we sought to understand the mechanism by which alcohol enhances the GlyR, because ethanol's actions at inhibitory receptors in the brain are thought to produce many of the physiological effects associated with its use. We examined the effects of 3 [mu]M glycine ± 50 or 200 mM ethanol on outside-out patches expressing WT [alpha]1 GlyR, to determine the effects of alcohol at the single-channel level. Alcohol enhanced GlyR function in a very specific manner. It had minimal effects on open and closed dwell times. Instead, ethanol potentiated GlyR function almost exclusively by increasing burst durations and increasing the number of channel openings per burst, without affecting the percentage of open time within bursts. Kinetic modeling suggests that ethanol increases burst durations by decreasing the rate of glycine unbinding. / text

COMPARATIVE ANALYSIS OF SOYBEAN (GLYCINE MAX) ACCESSIONS USING INTER SIMPLE SEQUENCE REPEAT (ISSR) AND RANDOM AMPLIFIED POLYMORPHIC DNA (RAPD) MARKERS

Soybean (Glycine max) is an important crop in the world in terms of total production and usage. It is also among the least diverse species. The main objectives of the present study were 1) to determine differences between ISSR and RAPD marker systems in detecting genetic variation in soybeans and 2) to identify and characterize accession- diagnostic molecular markers in G. max accessions. Genomic DNAs from 108 G. max accessions from 11 different gene pools were analyzed using several ISSR and RAPD primers. The levels of polymorphic loci detected with the two marker systems were in general moderate and similar.. Overall, 82% of genetic distance values were above 0.40 based on ISSR analysis. However, RAPD data revealed that the accessions from different countries are closely related with 64% genetic distance values below 0.40. The dendrograms constructed with ISSR data revealed that the South Korean accessions formed an out-group while the RAPD analysis showed that accessions from Sweden were separate from the other 10 gene pools. One variety-diagnostic marker generated with ISSR 5 primer was identified in the accession Kao Chien Tao from China. This marker was cloned, and sequenced. Although RAPD and ISSR marker systems detected similar levels of genetic variability, they target different regions of the soybean genome, resulting in different clustering of the 11 gene pools indicating different genetic relatedness among them. This finding demonstrates the usefulness of both marker systems in assessing diversity and relatedness among Glycine max gene pools.

Para simular a produtividade da cultura foi usado o modelo AquaCrop versão 5.0 da FAO, calibrado com dados do ano de 2016 e validado com os de 2014, usando parâmetros de clima, solo, cultura e manejo coletados em duas campanhas experimentais realizadas entre os meses de junho e outubro nos anos de 2014 e 2016 em Palmas, TO. O desempenho do modelo foi avaliado utilizando os indicadores estatísticos: erro de previsão (Ep), coeficiente de determinação (R2), raiz quadrada do erro médio (RMSE), erro médio absoluto (EMA), eficiência de Nash e Sutcliffe (NSE), e o índice de concordância de Willmontt´s (d). A calibração e validação da produtividade da cultura de soja estimada pelo modelo AquaCrop, apresentaram resultados satisfatórios, ilustrando a robustez e a aplicabilidade geral do modelo. O modelo AquaCrop subestima a produtividade de grãos de soja, para condições de estresse hídrico severo durante todo o ciclo de cultivo. Após a calibração e validação, o AquaCrop foi utilizado como ferramenta de simulação de produtividade da cultura da soja para o cenário atual (2016) e de mudanças climáticas a médio (2045/2046; 2055/2056) e longo prazo (2075/2076; 2064/2095), alimentado por dados de dois modelos climáticos (HadGEM2-ES e MIROC5) e considerando as RCP 4.5 e 8.5. Em seguida, calculou-se a pegada hídrica (verde, azul e cinza) de soja atual dos principais municípios produtores, de cada estado que compõem a região do Matopiba. Posteriormente, avaliaram-se os efeitos das possíveis mudanças climáticas sob a produtividade e pegada hídrica da soja, considerando as variações climáticas com foco na temperatura, precipitação e CO2. Os modelos climáticos projetaram aumento da produtividade em ambas as RCP consideradas, porém mais acentuado sob a RCP 8.5, em decorrência do aumento da temperatura e concentração de CO2 e a precipitação, que mesmo sofrendo redução nos totais pluviométricos ao longo do tempo, ainda atendendo a necessidade hídrica da soja. As PHsoja atuais da região do Matopiba, variaram de 2036,60 m³t-1 a 2584,12 m³t-1, valores similares aos encontradas na literatura. Sob cenários de mudanças climáticas, a PHsoja decresce ao longos os anos. A PHsoja futura decresce, especialmente a componente verde, devido ao aumento menos acentuado da evapotranspiração, resultando em maior rendimento final. As PHverde diminuem ao longos dos anos, as PHazul aumenta na mesma proporção e as PHcinza apresentam comportamento praticamente continuo. Os resultados deste estudo podem ser usados para quantificar a produtividade futura da soja, a demanda de água e a sua utilização, bem como obter informações úteis para a gestão dos recursos hídricos na região de estudo. / This study evaluated the current conditions and effects of climate change on the productivity and water footprint of soybean cultivated in Matopiba region. To simulate crop productivity, the FAO AquaCrop version 5.0 model was used, calibrated with data from 2016 and validated with 2014, using climate, soil, crop and management parameters collected in two experimental campaigns conducted between the months of June and October in the years 2014 and 2016 in Palmas, TO. The performance of the model was evaluated using the statistical indicators: prediction error (Ep), coefficient of determination (R2), square root mean error (RMSE), mean absolute error (EMA), Nash efficiency and Sutcliffe (NSE) and Willmontt's agreement index (d). Calibration and validation of soybean crop productivity estimated by the AquaCrop model presented satisfactory results, illustrating the robustness and general applicability of the model. The AquaCrop model underestimates soybean grain yield for severe water stress conditions throughout the growing cycle. After calibration and validation, AquaCrop was used as a simulation tool for soybean crop productivity for the current scenario (2016) and medium-term (2045/2046; 2055/2056) and long-term (2075/2076; 2064/2095), fed by data from two climatic models (HadGEM2-ES and MIROC5) and considering RCPs 4.5 and 8.5. Then, the water footprint (green, blue and gray) of the current soybean of the main producing municipalities of each state that compose the Matopiba region was calculated. Subsequently, the effects of possible climatic changes under soybean productivity and water footprint, considering the climatic variations with focus on temperature, precipitation and CO2, were evaluated. The climatic models projected increase of productivity in both RCP considered, but more accentuated under RCP 8.5, due to the increase in temperature and concentration of CO2 and precipitation, that even undergoing a reduction in rainfall totals over time, still taking into account water requirement of soybeans. The current PHsoja of the Matopiba region, ranged from 2036.60 m³t-1 to 2584.12 m³t-1, values similar to those found in the literature. Under scenarios of climate change, the PHsoja decreases over the years. The future PHsoja decreases, especially the green component, due to the less accentuated increase of the evapotranspiration, resulting in greater final yield. PHverde decreases over the years, PHazul increases in the same proportion and PHcinza show practically continuous behavior. The results of this study can be used to quantify future soybean yield, water demand and utilization, as well as to obtain useful information for the management of water resources in the study region.

Dentre os muitos desafios encontrados pelo melhoramento genético, um dos principais está relacionado com a variação ambiental, que muitas vezes dificulta a seleção e recomendação de genótipos superiores. Sendo assim, faz-se necessário que os ensaios experimentais sejam conduzidos em vários ambientes, por vários anos para que possa ser analisado o comportamento quanto a adaptabilidade e estabilidade, visando futuras recomendações. Neste contexto, o objetivo do presente estudo consistiu em avaliar a desempenho agronômico, a adaptabilidade e estabilidade de genótipos de soja precoces. Os experimentos foram instalados em dois locais, a saber: localidade de Jaboticabal nos anos agrícolas 2012/2013, 2013/2014, 2015/2016 e 2016/2017 e localidade Piracicaba nos anos agrícolas 2014/2015 e 2015/2016. O delineamento utilizado foi de blocos ao acaso com três repetições. Foram avaliados os caracteres agronômicos número de dias para a maturidade e produtividade de grãos. Para estimar a adaptabilidade e estabilidade dos genótipos avaliados, foi utilizada a metodologia de Eberhart e Russell (1966). Desta maneira, foi possível detectar os genótipos mais responsivos e adaptados às regiões de cultivo. Para o caráter produtividade de grãos, merecem destaque os genótipos 4, 6,11, 26, 27, 29 e 34, sendo linhagens pertencentes ao programa de melhoramento de soja da UNESP Jaboticabal, todos com médias superiores à média geral e portadores de boa estabilidade fenotípica para o caráter. Para o caráter número de dias de maturidade, destacaram-se os genótipos 4, 5, 7, 9, 13, 15, 18, 22 e 35, sendo também linhagens pertencentes ao programa de melhoramento de soja da UNESP Jaboticabal, todos com médias inferiores à média geral, portanto portadores de maior precocidade e com boa estabilidade fenotípica para o caráter. Dessa forma, por meio do presente estudo, foi possível identificar genótipos de soja superiores e que podem ser indicados para áreas de reforma de canaviais para o Estado de São Paulo. / Soy is the main oilseed crop in the world. In this context, Brazil stands out as the world's second largest producer of grain. Among the many challenges encountered by genetic improvement, one of the main challenges is related to environmental variation, which often hinders the selection and recommendation of higher genotypes. Thus, it is necessary that the experimental tests be conducted in several environments, for several years so that behavior can be analyzed for adaptability and stability, aiming at future recommendations. In this context, the objective of the present study was to evaluate the agronomic performance, adaptability and stability of early soybean genotypes. The experiments were installed in two locations, namely: Jaboticabal locality in the agricultural years 2012/2013, 2013/2014, 2015/2016 and 2016/2017 and locality Piracicaba in the agricultural years 2014/2015 and 2015/2016. The experimental design was a randomized block design with three replications. The agronomic characters were evaluated for the number of days for grain maturity and yield. To estimate the adaptability and stability of the evaluated genotypes, the methodology of Eberhart and Russell (1966) was used. In this way, it was possible to detect the genotypes more responsive and adapted to the growing regions. For grain yield, genotypes 4, 6, 11, 26, 27, 29 and 34 deserve special mention, being strains belonging to the soybean breeding program of UNESP Jaboticabal, all with averages above the general average and good stability phenotypic character. For the number of days of maturity, the genotypes 4, 5, 7, 9, 13, 15, 18, 22 and 35 were highlighted, being also lineages belonging to the soybean breeding program of UNESP Jaboticabal, all with lower averages to the general average, therefore bearers of greater precocity and with good phenotypic stability to the character. Thus, through the present study, it was possible to identify superior soybean genotypes that can be indicated for sugar cane reforestation areas for the State of São Paulo. / CNPq: 132711/2016-8

Para o teste de condutividade elétrica se estabelecer como parte do programa de controle de qualidade nas empresas produtoras de sementes são necessárias informações que correlacionem níveis de condutividade elétrica com o potencial de desempenho da semente. Desta forma o presente trabalho teve por objetivo o estudo da utilização do teste de condutividade elétrica como alternativa para avaliação de vigor de sementes de soja verificando sua relação com diferentes testes de vigor e assim sugerir valores de condutividade elétrica para determinação de valores ou faixa de valores que indicam o potencial de desempenho de um lote de sementes em campo. Para tanto, foram utilizadas sementes de soja de quatro cultivares - BRS 388 RR, BRS 1010 IPRO, BRS 1001 IPRO e BRS 1007 IPRO, cada um representado por 11 lotes divididos em quatro repetições. As sementes foram avaliadas por meio dos testes de: condutividade elétrica, germinação, tetrazólio, envelhecimento acelerado, primeira contagem de germinação, classificação do vigor de plântulas, emergência de plântulas em campo e emergência de plântulas em areia. Determinou-se também o teor de água das sementes. O teste de condutividade elétrica apresentou correlação significativa e negativa (p<0,01) para todos os testes avaliados. A análise de regressão linear possibilitou a separação dos lotes de sementes de vigor muito alto: CE < 70 μS.cm-1.g-1; alto: CE entre 71 a 90 μS.cm-1.g-1; médio: CE entre 91 a 110 μS.cm-1.g-1 e baixo: CE > 111 μS.cm-1.g-1. / In order to evaluate the vigor of seed lots, fast and efficient vigor tests are necessary, among them the electrical conductivity test, because it is able to detect the seed deterioration still in its initial phase, in view of the disorganization of the cellular membranes. For the electrical conductivity test to establish as part of the quality control program in the seed producing companies, information is required that correlate levels of electrical conductivity with the potential of seed performance. In this way the present work had the objective of the study of the use of the electrical conductivity test as an alternative to evaluate the vigor of soybean seeds, verifying its relation with different vigor tests and thus to suggest values of electrical conductivity for determination of values or range of values which indicate the potential performance of a field seed batch. For this, soybean seeds of four cultivars - BRS 388 RR, BRS 1010 IPRO, BRS 1001 IPRO and BRS 1007 IPRO were used, each represented by 11 lots divided into four replicates. The seeds were evaluated through the following tests: electric conductivity, germination, tetrazolium, accelerated aging, first germination count, seedling vigor classification, emergence of seedlings in the field and emergence of seedlings in sand. The water content of the seeds was also determined. The electrical conductivity test showed significant and negative correlation (p <0.01) for all tests evaluated. The linear regression analysis allowed the separation of seed lots of very high vigor: CE <70 μS.cm-1.g-1; high: CE between 71 to 90 μS.cm-1.g-1; mean: CE between 91 to 110 μS.cm-1.g-1 and low: CE> 111 μS.cm-1.g-1. / 1596391

Um dos métodos adotados para a aplicação de nematicida químico nas lavouras de soja é a aplicação via tratamento de sementes, porém os estudos de controle com este método, na grande maioria dos casos, apontam período de proteção de até 30 dias após a emergência das plantas, e na maioria das vezes tem trazido resultados insatisfatórios. A aplicação de nematicida químico, no sulco de plantio, ou associação do tratamento de sementes com a aplicação no sulco de plantio, pode melhorar os resultados de eficácia e eficiência, todavia esta técnica é pouco estudada no Brasil para a cultura de soja, consequentemente são poucos trabalhos que comprovem essas características nos produtos disponíveis atualmente. Também, o uso de um condicionador de solo poderá contribuir com um melhor desenvolvimento radicular, da planta e na eficácia e eficiência do produto nematicida químico. O objetivo deste estudo foi avaliar a eficácia e eficiência do nematicidas químicos tiodicarbe, cadusafós e do condicionador de solo Maskio, usados em tratamento de sementes e/ou aplicados no de sulco de plantio, no controle dos nematoides de galha (Meloidogyne incognita), de cisto da soja (Heterodera glycines) e das lesões radiculares (Pratylenchus brachyurus) na cultura de soja. Foram instalados ensaios a campo em áreas naturalmente infestadas pelos nematoides, e em vasos onde foram avaliados o desenvolvimento e produtividade da cultura e a redução na população de nematoides. Nenhum dos tratamentos testados causou sintomas visuais de intoxicação na cultura. Isoladamente, o tratamento de sementes com tiodicarbe (600 mL 100 kg sementes-1) é tão eficaz na redução da população total de P. brachyurus nas raízes da soja, quanto o tratamento de sulco com cadusafós (4,0 L ha-1) ou cadusafós + Maskio (4,0 L ha-1 + 2,0 L ha-1). Porém nenhum tratamento aumentou significativamente a produtividade da cultura. Para que haja incremento significativo em produtividade foi necessário associar a aplicação de tiodicarbe na semente e no sulco de plantio para áreas infestadas por P. brachyurus. Para áreas com M. incognita os tratamentos, de semente ou de sulco, podem ser eficazes no controle e incrementar em produtividade a cultura da soja se utilizados em área com alta infestação de nematoides de galha. A aplicação de tiodicarbe nas sementes foi tão eficaz na redução de cistos de Heterodera glycines quanto a aplicação no sulco de plantio de tiodicarbe, cadusafós, Maskio e suas associações. One of the methods adopted for the application of chemical nematicide in soybean crops is the application through seed treatment, but the studies in the control of this method, in the great majority of cases, indicate protection period up to 30 days after the emergence of the plants, in the most of the time it has brought unsatisfactory results. The application of chemical nematicide, in the planting groove, or association of the treatment of seeds with the application in the planting groove, can improve the results of efficacy and efficiency, however this technique is little studied in Brazil for the soybean crop, consequently are few works that prove the effectiveness and efficiency of the currently available products. Also, the use of a soil conditioner may contribute to a better root development of the plant and to the effectiveness and efficiency of the chemical nematicide product. The objective of this study was to evaluate the efficacy and efficiency of the chemical nematicides thiodicarb, cadusafos and the Maskio soil conditioner, used in seed treatment and / or applied to the planting groove, in the control of root-knot nematode (Meloidogyne incognita), soybean cist nematode (Heterodera glycines) and root-lesion nematode (Pratylenchus brachyurus) on soybean crop. Field trials were carried out in areas naturally infested by nematodes, and in pots where the development and productivity of the crop and the reduction in the nematode population were evaluated. None of the treatments tested causes visual symptoms of intoxication in the culture. Seed treatment with thiodicarb (600 mL 100 kg seed-1) is as effective in reducing the total population of P. brachyurus in soybean roots as in grouper treatment with cadusafos (4.0 L ha-1) or cadusafos + Maskio (4.0 L ha-1 + 2.0 L ha-1). However, no treatment significantly increases the productivity of the crop. In order to have a significant increase in productivity it is necessary to associate the application of thiodicarb in the seed and in the planting groove to areas infested by P. brachyurus. For areas with M. incognita, seed or planting groove treatments may be effective in controlling and increasing soybean productivity if used in an area with high root-knot nematode infestation. The application of thiodicarb in the seeds is as effective in the reduction of Heterodera glycines cysts as the application in the planting groove of thiodicarb, cadusafos, Maskio and their associations. The association of thiodicarb in seeds and a planting groove treatment improves the control efficacy of H. glycines. Leaving 27% efficacy for isolated seed treatment, to 75% efficacy for treatment with thiodicarb in seeds associated with the use of the same molecule in the planting groove.
