Avaliação da estrutura e do potencial fisiológico de sementes de crotalária por meio de recursos de análise de imagens / Evaluation of the structure and physiological potential of sun hemp seeds using image analysis

A utilização de procedimentos rápidos e confiáveis para análise de sementes é fundamental para a tomada de decisões nas diferentes etapas do processo produtivo e definição do destino dos lotes de sementes. Neste sentido, os objetivos desta pesquisa foram: a) avaliar a estrutura de sementes de crotalária, utilizando raios X para avaliação das partes da semente, identificação de possíveis danos causados por injúrias mecânicas, por percevejo e tecidos deteriorados nas sementes e sua relação com a germinação; b) verificar a possibilidade de utilização do sistema computadorizado de análise de plântulas (SVIS®) para detectar diferenças de vigor entre lotes de sementes de Crotalaria juncea, em comparação com as informações fornecidas por testes tradicionalmente utilizados para a espécie. Foram utilizadas seis lotes de sementes de crotalária do cultivar IAC-KR1, tanto para avaliação da estrutura de sementes quanto para a análise de imagens de plântulas. As análises relativas à morfologia interna das sementes foram realizadas em apenas uma época, no início da pesquisa, ao passo que aquelas relativas ao sistema computadorizado de imagens de plântulas e demais testes de vigor foram realizados em duas épocas, espaçadas de quatro meses. Inicialmente as sementes foram submetidas ao teste de raios X e ao teste de germinação. As plântulas normais, anormais e sementes mortas foram fotografadas e analisadas paralelamente às imagens radiográficas das sementes. Na segunda etapa da pesquisa, os seis lotes foram submetidos a testes de germinação e de vigor (primeira contagem de germinação, envelhecimento acelerado, emergência de plântulas em areia e condutividade elétrica). Para determinação do vigor das sementes por meio do SVIS®, as plântulas foram avaliadas aos três e quatro dias após a semeadura do teste de germinação, sendo obtidos dados referentes ao índice de uniformidade de desenvolvimento, vigor e comprimento de plântulas. Concluiu-se que a análise de imagens de raios X permite identificar danos mecânicos, por percevejo e tecidos deteriorados em sementes de crotalária e relacioná-las com a germinação; o SVIS® é uma alternativa viável para a avaliação do vigor de sementes de crotalária. / Fast and reliable procedures for analyzing seeds are fundamental for taking decisions during the different production stages and defining the destination of seed lots. Therefore, the objectives of this study were: a) to study the structure of sun hemp seeds with X-ray to evaluate the seed parts and identify possible damage caused by mechanical injuries, stink bugs and deteriorated tissues and their relation to germination; b) verify the possibility of using a computerized seedling analysis system (SVIS®) to detect differences in vigor between seed lots of Crotalaria juncea, and compare the results with those obtained from vigor tests used for the species. Six lots of the sun hemp cultivar IAC-KR1 were used to evaluate seed structure and also to analyze seedling images. The internal morphology seed was analyzed only at the beginning of the study whereas the computer analysis of seedling images and the other vigor tests were done at two different times, four months apart. The seeds were first submitted to X-ray and germination tests. Normal and abnormal seedlings and dead seeds were photographed and analyzed at the same time as the seed X-ray images. In the second stage of the study, the six lots were submitted to germination and vigor (first germination count, accelerated aging, seedling emergence in sand and electrical conductivity) tests. To determine the seed vigor using the SVIS®, the seedlings were evaluated three and four days after sowing for the germination test, when data on the uniformity development index, vigor and seedling length were obtained. It was concluded that the analysis of X-ray images is efficient for identifying mechanical damage, stink bug damage and deteriorated tissues and allowed to establish an association between their occurrence and germination; the SVIS® is a viable alternative for evaluating the vigor of sun hemp seeds.

Panneaux composites bio-sourcés destinés à l'isolation des bâtiments : caractérisation des ressources et procédé de production / Bio-based composite panels for building insulation : Characterization of resources and production process

L'objectif de cette thèse est de développer de nouveaux composites isolants biosourcés à partir de coproduits de l'agriculture exploitables dans le secteur du bâtiment. Les travaux s'organisent en trois principales étapes : La première, au travers d'un travail de formulation et de caractérisation, vise à évaluer les propriétés mécaniques, thermiques et hygriques de composites biosourcés obtenus par agglomération de granulats végétaux avec différents types de liants biosourcés et minéraux. Plusieurs agro-ressources sont envisagées (paille de blé, colza, anas, chènevotte et rafles de maïs). Une solution associant des granulats de chènevotte avec un liant biosourcé thermodurcissable est sélectionnée. La suite des travaux se focalise sur la recherche d'un procédé de mise en forme adapté à la fabrication de panneaux rigides isolants réalisés à partir de la formulation du composite de chènevotte. La fabrication de panneaux composites est testée sur différents outillages disponibles en laboratoire ainsi que sur des outils industriels permettant de tester les différentes technologies existantes. La possibilité de fabriquer le panneau rigide isolant de chènevotte sur une ligne de production industrielle entièrement automatisée est démontrée. Enfin, de nouvelles solutions permettant d'améliorer la réaction au feu des panneaux composites sont testées ce qui permet d'identifier des solutions adaptées au composite étudié. / The objective of this thesis is to develop new insulation composites from agricultural by-products for building applications. The research work is divided in three main steps : The first one focuses on the formulation of bio-based composites obtained by agglomeration of vegetal aggregates with different types of binders (bio-based or mineral binders), and on the characterization of their mechanical, thermal and hygric properties. Several bio-aggregates are envisaged (wheat straw, rape straw, flax shiv, hemp shiv and corn cob). A solution combining hemp shiv aggregates bonded with a bio-based thermosetting adhesive is selected. The next step aims to identify a suitable manufacturing process to produce rigid insulation panels using the previously determined composite formulation. Several manufacturing trials are conducted on different laboratory and industrial tools which allow to experiment existing manufacturing technologies. The possibility to produce the rigid insulation panel from hemp shiv at the industrial scale on a fully automatized production line is demonstrated. Finally, new solutions to improve the fire reaction of the insulation panels are tested which allows to identify suitable solutions for the developed composites.

Biopolymer Composite based on Natural and Derived Hemp Cellulose Fibres.

The aim of this research was to study the effect of pre-treatment and modification processes on the properties of hemp cellulose fibre for biopolymer composites application. Hemp fibres have been modified by various extraction, swelling, chemical and enzymatic treatments. The morphology and mechanical properties of the modified fibres have been measured. Biopolymer composites have been prepared using the modified fibres and matrices of cellulose acetate butyrate and cellulose solutions derived from hemp. The first fibre treatment employed was acetone extraction and mercerization. A low pressure acrylonitrile grafting initiated by azo-bis-isobutylonitrile was performed using alkali treated fibre. The AN grafted fibres had no transformation of crystalline structure as observed after mercerization. The mechanical properties performed by a single fibre test method were strongly influenced by the cellulose structure, lateral index of crystallinity, and fraction of grafting. Bioscouring of hemp using pectate lyase (EC 4.2.2.2), Scourzyme L, was performed. Greater enzyme concentration and a longer treatment improved the removal of the low methoxy pectin component. Removal of pectate caused no crystalline transformation in the fibres, except for a slight decline in the X-ray crystalline order index. Smooth surfaces and separated fibres were evidence of successful treatment. The shortening of fibre by grinding and ball-milling was introduced to achieve a desired fibre size. An increase in the milling duration gradual ly destroyed the crystalline structure of the cellulose fibres. An increase in solvent polarity, solvent-fibre ratio, agitation speed and drying rate resulted in the rearrangement of the ball-milled cellulose crystalline structure to a greater order. The thermal degradation behaviour of hemp fibres was investigated by using TGA. The greater activation energy of treated hemp fibre compared with untreated fibre represented an increase in purity and improvement of structural order. The all hemp cellulose composites were prepared by an introduction of fibres into 12% cellulose N-methyl-morpholine N-oxide (NMMO) solution and water-ethanol regeneration. A broadening of the scattering of the main crystalline plane, (002) and a depression of the maximum degradation temperature of the fibres were observed. These revealed a structural change in the fibres arising from the preparation. The mechanical properties of composites depended on size, surface area, crystallinity and the structural swelling of the fibres. Composites of cellulose acetate butyrate (CAB) and modified hemp fibres were prepared. Composites containing pectate lyase enzyme treated fibres showed better mechanical property improvement than untreated and alkali treated fibres respectively.

Genetic Analysis of Lignification and Secondary Wall Development in Bast Fibers of Industrial Hemp (Cannabis sativa)

Industrial hemp (Cannabis sativa) is a highly productive crop that is well suited to cultivation in Canada. To better understand the development of bast (phloem) fiber secondary walls and to facilitate reverse genetics screening for improved germplasm, I undertook two sets of microarray experiments. The first compared transcript expression in stem peels at three positions along the length of the stem. The second set of microarray experiments compared transcript expression in adjacent tissue layers along the radial axis of the stem. The transcripts that were enriched in fiber-producing tissues in both studies were consistent with a dynamic program of cell wall deposition. Detailed qRT-PCR analysis of specific lignification genes identified the best targets for reverse genetics. Finally, as a first step towards establishing a virally induced gene silencing (VIGS) system, I identified viruses that produced visual symptoms of infection, although qRT-PCR failed to confirm the infection / Plant Biology

Tyrimai atlikti 2012 – 2013 metais ASU Žemės ūkio inžinerijos ir saugos instituto laboratorijose bei ASU bandymų stotyje. Darbo tikslas – ištirti netradicinių energetinių augalų (drambliažolių, pluoštinių kanapių ir pluoštinių dilgėlių) ruošimo ir naudojimo biokurui technologinius-techninius parametrus bei nustatyti susmulkintų ir supresuotų augalų fizikines-mechanines savybes ir kokybinius rodiklius. Pateikto darbo literatūros apžvalgoje išanalizuota šių augalų nuėmimo ir paruošimo technologija biokurui bei įvertintos augalų naudojimo deginimui galimybės. Atlikus drambliažolių, pluoštinių kanapių ir pluoštinių dilgėlių eksperimentinius tyrimus buvo nustatyti biometriniai rodikliai, smulkinimo ir malimo parametrai, ištirtos pjaustinio ir miltų fizikinės-mechaninės savybės. Nustatyta frakcinė sudėtis pagal ES šalyse taikomą metodiką, naudojant sietų komplektą su skirtingų skersmens skylutėmis. Taip pat buvo ištirtos ir paruoštų granulių svarbiausios fizikinės-mechaninės savybės. / Investigations were carried out in 2012-2013 years in the laboratories of ASU Institute of Agricultural Engineering and Safety and ASU training farm. The aim – to investigate the technological-technical parameters of non-traditional energy plants (elephant grass, fibrous hemp and fibrous nettle) for the preparation and use of biofuels and to determine the chopped and pressed plant physical-mechanical properties and qualitative indicators. In the literature review of presented work was analyzed the plant harvesting and processing technology of biofuels and evaluates the use of plants burning capabilities. After the elephant grass, fibrous hemp and fibrous nettle experimental studies were established the biometric indicators, chopping and milling parameters, and were investigated the chaff and mill physical-mechanical properties. Fractional composition was determined according to the methodology of EU countries, while using the sieves with the different diameter holes. It was also been tested and most important physical-mechanical properties of prepared pellets.

L’utilisation de ressources lignocellulosiques renouvelables connaît à l’heure actuelle un indéniable regain d’intérêt pour l’élaboration de matériaux de construction. Partant de ce constat, un premier axe de travail a vu l’élaboration de bétons de végétaux constitués de granulats de végétaux lignocellulosiques et d’un liant pouzzolanique. Les broyats de tige de chanvre et de tournesol ont ainsi été sélectionnés pour leurs similarités et leur disponibilité dans la région Auvergne. En parallèle, un liant pouzzolanique constitué de 80% en masse de sable de ponce volcanique provenant d’une carrière locale, de 20% de chaux aérienne et d’un activateur a été formulé. Les résultats obtenus montrent la similitude des caractéristiques des granulats de chanvre et de tournesol ainsi que des performances mécaniques et thermiques des bétons légers (<500kg.m-3) constitués de ces végétaux. Les matériaux formulés satisfont aux critères fixés dans les règles professionnelles de la construction en chanvre. Dans un deuxième axe de travail, l’analyse bibliographique réalisée a permis d’identifier la qualité d’interface entre les particules végétales et le liant minéral comme un des principaux verrous scientifiques concernant les bétons de végétaux. Les problèmes de prise à coeur de ces matériaux mériteraient en effet d’être reliés avec ceux observés à l’interface liant/bois. Deux voies d’amélioration sont ainsi explorées dans une approche multi-échelles mêlant analyses physico-chimiques et essais mécaniques : le traitement préalable des particules végétales et l’adjuvantation spécifique du liant. La première stratégie a consisté à réaliser sur les granulats végétaux deux types de modifications : un recouvrement à l’huile de lin et un traitement en solution aqueuse de Ca(OH)2. Afin d’analyser à court terme l’interaction entre les particules ainsi modifiées et le liant frais, un dispositif de mesure spécifique dit de la plaque immergée est utilisé. Les résultats montrent une amélioration de l’interphase liant/végétal après le traitement des granulats en solution de Ca(OH)2. De façon surprenante, les performances mécaniques des bétons de végétaux chutent lorsque les granulats sont préalablement traités. Cette approche, en plus d’être contraignante d’un point de vue industriel, ne résout donc pas de façon satisfaisante les problèmes d’interfaces liant/bois. La seconde stratégie mise en place a nécessité l’intégration dans le liant d’un adjuvant rétenteur d’eau de type éther de cellulose dans des proportions variant de 0,5 à 1,5% par rapport à la masse des poudres. Les observations d’échantillons réalisées au MEB et couplées à une analyse EDX soulignent l’aptitude de l’éther de cellulose utilisé à améliorer les interfaces liant/bois. Une explication de l’action de ces molécules polymères sur les interfaces, basée sur la bibliographie et l’analyse du liant, est proposée. Cette approche a permis l’élaboration de bétons de végétaux immédiatement démoulables, à la compactabilité et la cohésion accrue et dont les propriétés mécaniques sont améliorées par rapport aux solutions en usage. Notons que cette amélioration n’est pas effectuée au détriment des propriétés thermiques. L’utilisation dans les bétons de végétaux d’un rétenteur d’eau ouvre dès lors des perspectives de développement : blocs préfabriqués, enduits légers, murs de béton projeté… / The use of renewable lignocellulosic resources experiences an undeniable resurgence of interest for building materials development. Based on this observation, a first line of work has seen the development of plant-based concretes constituted of lignocellulosic aggregates and a pozzolanic binder. Hemp and sunflower stem aggregates were then selected for their similarities and availability in the Auvergne region. In parallel, pozzolanic binder consisting of 80% by mass of volcanic pumice sand from a local quarry, 20% lime and an activator has been formulated. The results show the similarity of hemp and sunflower aggregates characteristics as well as mechanical and thermal properties of lightweight concretes (<500kg.m-3) made of these plants. Formulated materials meet the criteria appointed in French professional rules ofconstruction in hemp. In a second line of work, the literature review driven enabled to identify interface between the plant particles and the inorganic binder as one of the major scientific issues concerning concrete plants. Heart hardening problems of these materials indeed deserve to be linked with those observed at the interface binder/wood. Two ways of improvement are explored in a multi-scale approach combining physico-chemical analysis and mechanical testing : pre-treatment of vegetable particles and specific adjuvantation of the binder. The first strategy was to make on the plant aggregates two types of changes : a linseed oil recovery and a treatment in an aqueous solution of Ca(OH)2. In order to analyse the short-term interaction between the modified particles and the binder, a specific measurement device based on an immersed plate is used. The results show an improvement of the binder/plant interphase after aggregates treatment in Ca(OH)2 solution. Surprisingly, the plant concretes mechanicals performances fall when plants are pre-treated. This approach is finally restrictive for an industrial point of view and does not solve satisfactorily binder/wood interface problem. The second implemented strategy required the adding in the binder of a waterretaining agent (cellulose ether) in amounts ranging from 0.5 to 1.5% based on the weight of the powders. Observations of samples performed on a SEM microscope and coupled with an EDX analysis underline the ability of the cellulose ether (CE) to improve binder/wood interfaces. An explanation of the action of CE molecules on polymer interfaces based on the literature and analysis of the binder is proposed. This approach has allowed the development of plant concretes immediately demouldable. Designed concretes also show higher compactability and cohesiveness as well as improved mechanical properties compared to the solutions in use. It should be noticed that this improvement is not made at the expense of thermal properties. CE used in plant concretes offer development opportunities : prefabricated blocks, lightweight coatings…

Studiu klíčivosti a vzcházivosti semen technického konopí / Study of germination and emergence of industial hemp seeds

Nowadays hemp is listed among plants with a wide range of potentials. It can be used as a source of fibre, oil, food or medicine. The aim of this work was to investigate optimal conditions for hemp seed germination grown in laboratory as well as field conditions and to propose the application of acquired findings. Hemp seed was split into 2 size groups: above 3 mm and 2 - 3 mm. Divided seeds with the humidity level of 6, 9 and 12% were stored at the temperature of 0, 8 and 20? 1°C for half a year. In 2 months interval, there were evaluated: germination, energy of sprouting, speed of sprouting, further seed viability and laboratory emergence. From next influences were evaluated the influence of dryness, wetting, autotoxicity, exudates of perennial weeds, seed treatment with the infusion of field horsetail and the grown stimulator. The best germination and laboratory emergence had seeds stored at 0°C. Test of seed viability confirmed significant decline in live seeds stored over 4 months at the 0°C temperature. The best germination was achieved by seed size 2 - 3 mm. Hemp requires sufficient amount of water for germination. Leachates from rhizomes of couch grass inhibited germination of hemp by 10%. On the other hand, germination was significantly increased by the stimulator.

O delineamento experimental utilizado foi o de blocos casualizados, com 24 tratamentos e quatro repetições, dispostos em esquema fatorial 4x2x3, constituído de quatro sistemas de cobertura do solo: mucuna-preta, crotalária, milheto e pousio, dois cultivares de arroz: Primavera e IAC-202 e três níveis de N-uréia: 0, 50 e 100 kg ha-1. Para solo, realizaram-se avaliações químicas nas camadas de 0,0 a 0,10, 0,10 a 0,20 e 0,20 a 0,30 m, para pH, Ca, Mg, P, K, MO, H+Al e Al, N-total, e N mineral (NH4 + e NO- 3). Determinou-se a produtividade de fitomassa seca das plantas de cobertura e da vegetação espontânea (pousio); concentração de nutrientes nos adubos verdes, vegetação espontânea e na cultura do arroz à época do florescimento; ciclo e altura do arroz; número de panículas e colmos férteis m-2; número de espiguetas (total, chochas e granadas), rendimento de grãos no benefício, de inteiros e quebrados, além da produtividade de grãos. A sucessão crotalária-arroz foi a que promoveu maior produtividade de grãos, sendo o IAC-202 o cultivar mais produtivo. O milheto apresentou maior produtividade de fitomassa seca, sendo a melhor opção dentre as espécies de cobertura do solo avaliadas. As doses de N aumentaram linearmente a produtividade de grãos... / Nitrogen is the nutrient that usually more influences in the rice yield. It has a complex dynamic in the soil-plant system being it changed by soil management. This study was conducted at Experimental Station of UNESP/FEIS, located in Selvíria-MS country, in a dystroferric Red Latosol (Haplustox) with the objective of evaluating the productivity and amount of nutrients from mass of covering plants of soil and the effect of the isolated and combined use of covering plants and fallow besides urea as sources of nitrogen for the highlands rice in the implantation of the no till system during the agricultural year 2001/02. The used experimental design was a randomized blocks with 24 treatments and four repetitions, disposed in a factorial scheme 4x2x3, constituted of four systems of covering plants of the soil: velvet bean, sunn hemp, millet and spontaneous vegetation (fallow); two rice cultivars: Primavera and IAC-202; and three N doses: 0, 50 and 100 kg ha-1 (Urea). It was evaluated: Soil chemical analysis in the layers from 0.0 to 0.10, 0.10 to 0.20 and 0.20 to 0.30 m, for pH, Ca, Mg, P, K, OM, H+Al and Al, N-total, and N mineral (NH4 + and NO3 -); the productivity of dry matter of the covering plants and of the spontaneous vegetation (fallow); concentration of nutrients in the covering plants, spontaneous vegetation and in the rice crop at flowering; cycle and height of the rice; panicles number and stems fertile m-2; spikelets number, grain quality, besides grain yield. The succession sunn hemp - rice promoted larger grain yield, being IAC-202 the cultivar more productive. The millet presented larger productivity of dry matter, being the best option among the species of covering plants. The doses of N increased the grain yield lineally, but they did not interfere in the mass of 1000 grains and in grain quality. The increase of the doses of N decreased the number... Com o objetivo de avaliar a produtividade e quantidade de nutrientes da fitomassa de plantas de cobertura do solo e o efeito do uso isolado e combinado de adubos verdes e uréia como fontes de nitrogênio para o arroz de terras altas na implantação do sistema plantio direto, foi conduzido um experimento durante o ano agrícola 2001/02 na Fazenda Experimental da UNESP/FEIS, no município de Selvíria- MS, em área de Latossolo Vermelho Distroférrico. O delineamento experimental utilizado foi o de blocos casualizados, com 24 tratamentos e quatro repetições, dispostos em esquema fatorial 4x2x3, constituído de quatro sistemas de cobertura do solo: mucuna-preta, crotalária, milheto e pousio, dois cultivares de arroz: Primavera e IAC-202 e três níveis de N-uréia: 0, 50 e 100 kg ha-1. Para solo, realizaram-se avaliações químicas nas camadas de 0,0 a 0,10, 0,10 a 0,20 e 0,20 a 0,30 m, para pH, Ca, Mg, P, K, MO, H+Al e Al, N-total, e N mineral (NH4 + e NO- 3). Determinou-se a produtividade de fitomassa seca das plantas de cobertura e da vegetação espontânea (pousio); concentração de nutrientes nos adubos verdes, vegetação espontânea e na cultura do arroz à época do florescimento; ciclo e altura do arroz; número de panículas e colmos férteis m-2; número de espiguetas (total, chochas e granadas), rendimento de grãos no benefício, de inteiros e quebrados, além da produtividade de grãos. A sucessão crotalária-arroz foi a que promoveu maior produtividade de grãos, sendo o IAC-202 o cultivar mais produtivo. O milheto apresentou maior produtividade de fitomassa seca, sendo a melhor opção dentre as espécies de cobertura do solo avaliadas. As doses de N aumentaram linearmente a produtividade de grãos... It has a complex dynamic in the soil-plant system being it changed by soil management. This study was conducted at Experimental Station of UNESP/FEIS, located in Selvíria-MS country, in a dystroferric Red Latosol (Haplustox) with the objective of evaluating the productivity and amount of nutrients from mass of covering plants of soil and the effect of the isolated and combined use of covering plants and fallow besides urea as sources of nitrogen for the highlands rice in the implantation of the no till system during the agricultural year 2001/02. The used experimental design was a randomized blocks with 24 treatments and four repetitions, disposed in a factorial scheme 4x2x3, constituted of four systems of covering plants of the soil: velvet bean, sunn hemp, millet and spontaneous vegetation (fallow); two rice cultivars: Primavera and IAC-202; and three N doses: 0, 50 and 100 kg ha-1 (Urea). It was evaluated: Soil chemical analysis in the layers from 0.0 to 0.10, 0.10 to 0.20 and 0.20 to 0.30 m, for pH, Ca, Mg, P, K, OM, H+Al and Al, N-total, and N mineral (NH4 + and NO3 -); the productivity of dry matter of the covering plants and of the spontaneous vegetation (fallow); concentration of nutrients in the covering plants, spontaneous vegetation and in the rice crop at flowering; cycle and height of the rice; panicles number and stems fertile m-2; spikelets number, grain quality, besides grain yield. The succession sunn hemp - rice promoted larger grain yield, being IAC-202 the cultivar more productive. The millet presented larger productivity of dry matter, being the best option among the species of covering plants.
