New Concepts in Administration of Drugs in Tablet Form : Formulation and Evaluation of a Sublingual Tablet for Rapid Absorption, and Presentation of an Individualised Dose Administration System

<p>This thesis presents two new concepts in oral drug administration and the results of evaluation of some relevant formulation factors.</p><p>Investigation into improving the homogeneity of mixtures for tableting indicated that it may be possible to obtain interactive dry mixtures of micronised drugs containing drug proportions as low as 0.015% w/w. By studying the relationship between disintegration time and tensile strength, it was found that the microstructure surrounding the disintegrant particles may influence the disintegration process. Therefore, avoidance of excipients which are highly deformable or very soluble in water will result in more rapid disintegration. Further, it is possible to increase the bioadhesive properties of a non-bioadhesive carrier material by forming interactive mixtures containing a fine particulate bioadhesive material.</p><p>The new sublingual tablet concept presented is based on interactive mixtures consisting of a water-soluble carrier covered with fine drug particles and a bioadhesive component. With this approach, it is possible to obtain rapid dissolution in combination with bioadhesive retention of the drug in the oral cavity. Clinical data indicate that this allows rapid sublingual absorption while simultaneously avoiding intestinal absorption. </p><p>An individualised dose administration system is also presented. This system is based on the use of standardised units (microtablets), each containing a sub-therapeutic amount of the active ingredient. The required dose is fine-tuned by electronically counting out a specific number of these units using an automatic dose dispenser. A patient handling study supported the suggestion that the dosage of some medications can be more easily and safely individualised for each patient with this method than by using traditional methods of mixing different standard tablet strengths or dividing tablets.</p>

Control de qualitat en mètodes d'anàlisi qualitatius i la seva aplicació al control antidopatge

L'anàlisi qualitatiu ha estat sempre molt important en el control antidopatge i constitueix cada cop més una part molt significativa de l'activitat diària d'altres tipus de laboratoris. Tanmateix, no existeixen guies o documents específics per ajudar els laboratoris a implantar sistemes de qualitat quan es disposa d'aquest tipus de mètodes. L'objectiu general d'aquesta tesi ha estat el desenvolupament de les eines necessàries per assegurar la qualitat i fiabilitat dels resultats produïts per mètodes d'anàlisi qualitatius, treballant en tres aspectes fonamentals per a l'assegurament de la qualitat dels resultats als laboratoris d'anàlisi: la validació de mètodes, el control intern de la qualitat, i els exercicis d'intercomparació.S'ha elaborat un protocol de validació, s'ha implantat un sistema de control intern de la qualitat per a mètodes d'anàlisi qualitatius i, finalment, s'han elaborat protocols per a la preparació de mostres que seran utilitzades com a materials d'assaig en exercicis d'intercomparació o com a materials de referència, i per realitzar estudis d'homogeneïtat i d'estabilitat, que han estat aplicats a l'estudi de l'estabilitat de substàncies d'interès en control antidopatge. / Qualitative analysis has been very important in antidoping control and its becoming a significant part of the daily activity of other type of laboratories. However, there is a lack of guidance or specific documents to help laboratories to implement quality assurance systems when using these type of methods. The aim of this thesis has been the development of the necessary tools to ensure the quality and reliability of results released by qualitative analytical methods, working in the three main aspects of quality assurance in the analytical laboratory: method validation, internal quality control and intercomparison exercises.A protocol for method validation has been proposed and an internal quality control system for qualitative methodologies has been established. Finally, protocols for the preparation of samples to be used in intercomparison exercises or as reference materials, including homogeneity and stability studies, have been prepared. These protocols have been applied to study the stability of substances with especial interest in antiodoping control.

The problems of rape seed methyl ester (RME) production process by-products utilization were analyzed, statistical data for the capacities of biodiesel production and by-products generation were presented, the prospects of biodiesel production expansion, properties and environmental impact, main production technologies, Lithuanian and foreign countries experience were described, the main aims and tasks of work were defined. The investigation of glycerol – major by-product of RME production process – utilization was accomplished. One of the new possible utilization methods was researched, when combusting glycerol with highly sulphurous (~ 2.0 %) heavy fuel oil to reduce SO2 emissions, which limit value in the flue gas can not exceed 1700 mg/Nm3 (according to EU Directive 1999/32EC and LAND 43-2001 „Norms for the exhaust of pollutants from large fuel burning equipment“). The heavy fuel oil – glycerol emulsion was prepared in the pilot plant operating in the laboratory conditions, the most optimal ratio (1:1) for heavy fuel oil emulsification was estimated, the dispersivity investigation of heavy fuel oil - glycerol emulsion samples was performed. Financial institutions make use of a variety of delivery channels for servicing their customers. The primary channel utilised as a means of acquiring new customers and increasing market share is through the retail branch network. The 1990s saw the Internet explosion and with it a threat to branches. The relatively low cost associated with virtual delivery channels made it inevitable for financial institutions to direct their focus towards such new and more cost efficient technologies. By the beginning of the 21st century -and with increasing limitations identified in alternative virtual delivery channels, the financial industry returned to a more balanced view which may be seen as the revival of branch networks. The main purpose of this study is to provide a roadmap for financial institutions in managing their branch network. A three step methodology, representative of data mining and management science techniques, will be used to explain relative branch efficiency. The methodology consists of clustering analysis (CA), data envelopment analysis (DEA) and decision tree induction (DTI). CA is applied to data internal to the financial institution for increasing' the discriminatory power of DEA. DEA is used to calculate the relevant operating efficiencies of branches deemed homogeneous during CA. A color filter array interpolation method for digital cameras using alias cancellation

To reduce cost, many digital cameras use a single sensor array instead of using three arrays for the red, green and blue. Thus at each pixel location only the red, green or blue intensity value is available. And to generate a complete color image, the camera must estimate the missing two values at each pixel location .Color filter arrays are used to capture only one portion of the spectrum (Red, Green or Blue) at each location. Various arrangements of the Color Filter Array (CFA) are possible, but the Bayer array is the most commonly used arrangement and we will deal exclusively with the Bayer array in this thesis. Since each of the three colors channels are effectively downsampled, it leads to aliasing artifacts. This thesis will analyze the effects of aliasing in the frequency- domain and present a method to reduce the deterioration in image quality due to aliasing artifacts. Two reference algorithms, AH-POCS (Adams and Hamilton - Projection Onto Convex Sets) and Adaptive Homogeneity-Directed interpolation, are discussed in de- tail. Both algorithms use the assumption that there is high correlation in the high- frequency regions to reduce aliasing. AH-POCS uses alias cancellation technique to reduce aliasing in the red and blue images, while the Adaptive Homogeneity-Directed interpolation algorithm is an edge-directed algorithm. We present here an algorithm that combines these two techniques and provides a better result on average when compared to the reference algorithms.

As caracter?sticas avaliadas foram os pesos pr?-abate, peso 28, peso 35, peso e rendimentos da carca?a, peito, coxa+sobrecoxa e as caracter?sticas de qualidade da carne. As an?lises foram realizadas utilizando modelos de regress?o aleat?ria considerando efeito fixo de sexo e o efeito aleat?rio gen?tico aditivo direto como fun??es das rela??es triptofano:lisina da dieta, admitindo-se homogeneidade de vari?ncia residual. Os resultados para desempenho, caracter?sticas de carca?a e caracter?sticas de qualidade da carne indicam que o coeficiente de regress?o do intercepto (b0), foi maior que o coeficiente de regress?o aleat?rio (b1). A herdabilidade e a vari?ncia gen?tica aditiva foram influenciadas pelas diferentes rela??es triptofano:lisina da dieta. Two lines of cutting quails, LF1 and LF2, were used to perform the experiment. The feed provided in the growing period, from birth to 21 days, contained a 0.20% tryptophan:lysine ratio and in the final growth period, from 22 to 35 days of age, quails were separated into 5 treatments containing the Ratios of 0.15; 0.20; 0.25; 0.30; 0.35% tryptophan: lysine. The birds were weighed at 28 and 35 days of age and slaughtered at 35 days of age. The evaluated characteristics were the pre-slaughter weights, weight 28, weight 35, weight and yields of the carcass, breast, thigh + sobrecoxa and the quality traitsof the meat. The analyzes were performed using random regression models considering fixed sex effect and direct additive genetic random effect as functions of the tryptophan:lysine ratios of the diet, assuming homogeneity of residual variance. The results for performance, carcass traitsand meat quality traitsindicate that the regression coefficient of the intercept (b0) was higher than the random regression coefficient (b1). Heritability and additive genetic variance were influenced by the different tryptophan:lysine ratios of the diet. There was no genotype x environment interaction for the traitsof carcass yield, yield of breast and yield of thigh + overcoat. The quality traits of the meat CRA, b *, c * and H * (LF1) and CRA, PPC, L * and H * (LF2) presented genotype x environment interaction. It is possible to conclude that for yield traitsthe selection can be made in the lowest tryptophan: diet lysine ratios. For meat quality traitsthe sensitivity of the values indicates that the selection must be made in the tryptophan:lysine ratio in which the quails will be created.

Tendência histórica e variabilidade do brilho solar e modelagem da radiação solar em Santa Maria, RS / Historical trends and variability of sunshine and solar radiation modeling in Santa Maria, RS

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The objectives of this study were to test the homogeneity of monthly data of sunshine period from 1912 to 2010 hours of sunshine and the period from 1968 to 2010, recalculate the descriptive statistics of sunshine daily, monthly and seasonal Santa Maria, RS, characterize the secular trend and interdecadal and interannual variability of monthly sunshine hours and adjust and test the Ångström - Prescott and Penman equation to estimate the flux density of incident solar radiation level and in different time periods of the day for Santa Maria , RS. The data of sunshine duration and solar radiation were measured at the meteorological station of the Brazilian National Weather Center (8o DISME/INMET), located in the Experimental Department, Federal University of Santa Maria. The Run tests, and the Mann-Kendall and Pettitt tests were used for analysis of homogeneity and trends in the series respectively, and the power spectral density was used for detecting annual and decadal cicles related to El Nino Oscillation Southern and Pacific Decadal Oscillation. The time series of monthly and seasonal sunshine in Santa Maria, in the period 1912-2010 are homogeneous for all months and seasons, except for the months of December and autumn. In the time series of monthly and seasonal sunshine in Santa Maria, in the period 1912 to 2010 there is no trends to increase or decrease, ie the series are stationary. There are large interannual and interdecadal variability in the time series of sunshine in Santa Maria, RS, indicating a sign of El Nino Oscillation Southern and Pacific Decadal Oscillation climate in this element. The number of sunshine daily and average monthly number of sunshine during the periods of the day, in the range from 9 to 15 hours showed decreasing trends, especially in October and spring. There is interannual variability in the series of monthly average daily sunshine and the series of sunlight at different times of day. In La Niña years increased the mean daily sunshine and the different times of day in October, November and December. In El Niño years increased the mean daily sunshine and the different times of day during the months of January and February. The estimation of the global solar radiation by the Ångström equation modified by Prescott and Penman was not precise level and time in the different sub-periods of the day, it is suggested to be tested models of other estimates of the estimate of global radiation hourly. / Os objetivos deste trabalho foram testar a homogeneidade dos dados mensais de brilho solar do período de 1912 a 2010 e horários de brilho solar do período de 1968 a 2010, recalcular as estatísticas descritivas do brilho solar diário, mensal e sazonal em Santa Maria, RS, caracterizar a tendência secular e a variabilidade interanual e interdecadal do brilho solar mensal e horário e ajustar e testar a equação de Ångström - Prescott e Penman para estimativa da densidade de fluxo de radiação solar global incidente em nível horário e em diferentes períodos do dia para Santa Maria, RS. Os dados de brilho solar e de radiação solar analisados foram medidos na Estação Climatológica Principal do Instituto Nacional de Meteorologia (8º DISME/INMET), localizada no Campo Experimental do Departamento de Fitotecnia da Universidade Federal de Santa Maria. Foram usados os testes Run, Mann-Kendall e Pettitt para análise de homogeneidade e tendências respectivamente, e o método da função densidade espectral de potências para detectar ciclos anuais e interdecadais relacionados aos fenômenos El Niño Oscilação Sul e Oscilação Decadal do Pacífico. As séries históricas mensais e sazonais de brilho solar em Santa Maria, RS, no período de 1912 a 2010 são homogêneas para todos os meses e estações do ano, exceto para o mês de dezembro e outono. Nas séries históricas mensais e sazonais de brilho solar em Santa Maria, RS, no período de 1912 a 2010 não há tendência de aumento ou diminuição, ou seja, as séries são estacionárias. Há grande variabilidade interanual e interdecadal nas séries históricas de brilho solar em Santa Maria, RS, indicando sinal do fenômeno El Niño Oscilação Sul e da Oscilação Decadal do Pacífico neste elemento climático. A série de brilho solar médio mensal diário e a série de brilho solar nos períodos do dia, no intervalo das 9 às 15 horas apresentou tendências decrescentes, principalmente no mês de outubro e primavera. Existe variabilidade interanual na série de brilho solar média mensal diário e nas séries de brilho solar no diferentes períodos do dia. Em anos de La Niña aumenta a média de brilho solar diário e nos diferentes períodos do dia nos meses de outubro, novembro e dezembro. Em anos de El Niño aumenta a média de brilho solar diário e nos diferentes períodos do dia nos meses de janeiro e fevereiro. A estimativa da radiação solar global pela a equação de Ångström modificada por Prescott e Penman não foi precisa em nível horário e nos diferentes sub-períodos do dia, por isso sugere-se que sejam testados por outros modelos as estimativas da estimativa da radiação solar global horária.
