Einfluss von Mistelextrakt auf die Migration von kaninen Mammatumorzellen und die Genexpression und das Wachstum von humanen B-NHL-Zelllinien in Bezug auf den Einsatz der Misteltherapie bei Mensch und Tier

Für diese Arbeit wurden in-vitro-Studien an kaninen Mammatumorzellen und humanen follikulären B-NHL-Zellen durchgeführt. Es wurden erstmals die migratorischen Eigenschaften einer kaninen Mammatumorzelllinie (CMT-U-27, infiltrierendes duktuläres Karzinom mit Metastasen) und deren Beeinflussbarkeit untersucht. Die Experimente zeigten einen leichten promigratorischen Einfluss von Noradrenalin und EGF; mit dem Tumorpromotor PMA wurde eine sehr deutliche Stimulation der Migration erreicht. Es konnte gezeigt werden, dass Iscador® M in der Lage ist, die PMA-stimulierte Migration der CMT-U-27-Zellen zu hemmen, wobei die spontane, Matrix-induzierte Zellmigration unbeeinflusst blieb. Zwischen der migratorischen Aktivität von Tumorzellen und ihrer Fähigkeit Metastasen zu bilden, besteht ein Zusammenhang. Durch einen inhibierenden Effekt auf die Migration von Tumorzellen könnte eine Progression der Erkrankung durch Metastasenbildung verhindert oder zumindest verzögert werden. IL-6 dient als prognostischer Faktor bei lymphoproliferativen Erkrankungen der B-Zellen und kann sich negativ auf den Krankheitsverlauf bei B-Lymphompatienten auswirken. Unterstützend zu den guten in-vivo Erfahrungen in der Lukasklinik in Arlesheim, Schweiz, sollte in dieser Arbeit untersucht werden, ob Iscador® P bei follikulären B-NHL-Zelllinien (DoHH2, WSU-NHL, Sc-1) einen autokrinen IL-6-Loop auslösen kann. Dafür wurden die Zellen über definierte Zeiträume mit verschiedenen Iscador® P-Konzentrationen inkubiert. Es wurden Proben entnommen und durchflusszytometrische Messungen auf membrangebundenen IL-6R und gp130, Messungen der mRNA-Expression von IL-6, IL-6R und gp130 mittels Real-Time-RT-PCR und ELISA-Messungen zur Bestimmung von IL-6, löslichem IL-6R (sIL-6R) und löslichem gp130 (sgp130) durchgeführt. Es konnte kein Einfluss von Iscador® P auf die Oberflächen- oder Genexpression von IL-6 und den dazugehörigen Rezeptorkomponenten gp130 und IL-6R nachgewiesen werden. Zudem wird kein IL-6, sgp130 oder sIL-6R von den Zellen freigesetzt. Damit kann ein autokriner IL-6-Loop und ein durch Freisetzen von sIL-6R ermöglichtes Transsignalling bei den in dieser Arbeit untersuchten follikulären B-NHL-Zelllinien ausgeschlossen werden. In Wachstumskurven konnte gezeigt werden, dass Iscador® P den proliferativen Einfluss von IL-6, bei gleichzeitiger Inkubation, aufhebt. Genexpressionsmessungen der apoptoserelevanten Gene bcl-2, bax, bcl-xl und bad mittels Real-Time-RT-PCR zeigten bei den WSU-NHL-Zellen, dass die Apoptose über ein Verschieben des bcl-2/bax-Quotienten ausgelöst wird, welches mittels durchflusszytometrischer Analyse bestätigt werden konnte. Im Gegensatz dazu konnte bei Sc-1 Zellen keine Verschiebung des bcl﷓2/bax-Quotienten in Richtung Induktion der Apoptose beobachtet werden, sodass die bei dieser Zelllinie beobachtete Hemmung der IL-6 mediierten Proliferation durch Iscador® P, durch einen anderen, bislang noch nicht geklärten, Mechanismus ablaufen muss.

Regulation of Adrenal Steroidogenesis by Interleukin-6

Cortisol and dehydroepiandrosterone (DHEA) are steroids produced by the zona fasciculata (ZF) and reticularis (ZR), respectively, of the adrenal cortex. Both steroids are upregulated in response to adrenocorticotropic hormone (ACTH). Cortisol is a glucorticoid that is important in the regulation of inflammation and metabolism. DHEA is an adrenal androgen important in fetal growth and puberty but tends to decrease gradually after puberty in both men and women. DHEA has various effects on metabolism and immune function including inhibiting the effects of cortisol on some tissues. During the acute phase of stress, cortisol and DHEA rise due to an increase in ACTH released from the anterior pituitary. In contrast, during chronic stress, cortisol remains elevated but DHEA and ACTH levels decrease. Likewise, stress causes serum levels of IL-6 to increase. IL-6 increases cortisol release from the human and bovine adrenal cortex. IL-6 also decreases DHEA release from zona reticularis of the bovine adrenal gland. In humans the effect of IL-6 on DHEA production is still uncertain. To determine a possible mechanism of IL-6 on the zona fasciculata and reticularis, human H294R cells and bovine adrenal tissue were incubated in serum free medium containing IL-6, at various concentrations and incubation intervals. At the end of the incubation interval, mRNA or protein was extracted from the cells or tissue. Standard PCR, real time PCR, and western blot assays were used to determine the effects of IL-6 on the enzymes involved in cortisol and DHEA synthesis, steroidogenic factor-1 (SF-1), steroidogenic acute regulatory protein (StAR), and dosage sensitive sex reversal adrenal hypoplasia congenita critical region on the X chromosome, gene 1 (DAX-1). In human H295R cells and bovine zona fasciculata cells IL-6 caused an increase in SF-1, StAR, P450scc, 17α hydroxylase, 3β-hydroxysteroid dehydrogenase type 2 (3β HSD2), 21 hydroxylase, and 11β hydroxylase mRNA and protein. IL-6 caused DAX-1 mRNA and protein to decrease. These effects were manifest in a time dependent manner. Dose response treatments incubated for 60 min increased SF-1, StAR, P450scc, 17α hydroxylase, 3β HSD2, 21 hydroxylase, and 11β hydroxylase but there was not significant change between the different treatments of IL-6. The bovine zona reticularis stimulated with IL-6 showed a decrease in SF-1, StAR, P450scc, 17α hydroxylase, and 3β HSD2 with an increase in DAX-1 mRNA and protein. This response was manifest in a time dependent manner for both mRNA and protein, and the effect was dose-dependent for mRNA but not protein levels within the 60 min time period. These data provide a mechanism by which increased stress, physical or emotional, which increases IL-6 serum level, could increase cortisol and decrease DHEA. This would account for decreased immune function, increased blood pressure, and changes in metabolism.

Childhood Maltreatment is Associated with Adult Depression: Is Inflammation to Blame?

By 2030 major depression is predicted to be the leading cause of disease burden in the world; as such, it is critical to understand factors that contribute to the development of depression. The social signal transduction theory of depression hypothesizes that adversity and social threat upregulate pro-inflammatory biomarkers leading to depression. The current study examined whether pro-inflammatory biomarkers (interleukin-6, interleukin-8, c-reactive protein, and tumor necrosis factor alpha) mediate the association between various types of childhood maltreatment (physical abuse, sexual abuse, emotional abuse, physical neglect, emotional neglect) and adult depression symptoms in a sample of 740 adults (372 female; Mage= 51.6 years, SD = 13.6) who provided retrospective report of childhood maltreatment as part of the Midlife in the United States (MIDUS) Refresher Biomarker study. Additionally, it explored whether these relations differ for males versus females. A series of linear regression analyses were run in SPSS; separate models were run for each form of childhood maltreatment and for interleukin-6, interleukin-8, c-reactive protein, and tumor necrosis factor-alpha. The results showed that childhood maltreatment is a robust predictor of adulthood depression; however, this association did not differ between biological sexes. In addition, only interleukin-6 was shown to partially mediate the association between childhood maltreatment and adulthood depression. The role of endothelial cell reactive antibodies in peripheral arterial disease

No / Objectives It is hypothesised that endothelial cell reactive antibodies (ECRA) play a role in the progression of PAD through activation of endothelial cells and the release of inflammatory cytokines. We aimed to test this hypothesis by assessing levels of ECRA, E-selectin and IL-6 in patients with PAD of varying severity in a case controlled study. Design, materials, methods Patients were assessed clinically and with ankle¿brachial pressure indices. Patients with critical ischaemia (CI, n=30), stable claudicants (SC, n=30), and age-matched controls (AMC, n=20) were studied. Antibody, E-selectin and IL-6 levels were measured using ELISA. Results ECRA levels were significantly raised in the CI group over AMC. IL-6 levels were significantly elevated in both SC and CI over the control group and in CI over SC. There were no significant differences in E-selectin levels between the AMC, SC and CI. Conclusion Our findings support the hypothesis that autoantibodies play a role in promoting PAD by elevating IL-6. The absence of an elevation in E-selectin in this study may be due to its short half-life, and casts doubt on its value as a marker of inflammation in atherosclerosis.

An integrative approach to the effect of interleukin-6 on adaptation to restraint stress in rats

However, the relative contribution of centrally versus peripherally secreted IL- 6 remains unclear, especially under psychological stress conditions. We hypothesised that the HPA response to mild psychological stress is dependent on IL- 6, both centrally and peripherally. 120 male Wistar rats were divided into four groups, depending on whether they received an anti-IL-6 antibody (Ab) (2μg/ml/kg body weight) or a placebo (sterile saline) injection and whether or not they were subjected to 1 hour of restraint stress for 1, 2 or 3 days. Rats were euthanized 24 hours after stress exposure. Plasma corticosteroid (GC) levels remained significantly increased 24 hours after a single stress exposure (control placebo (CP) versus stress placebo (SP): p < 0.05). The undetectable plasma IL-6 levels evident across all groups may be explained by the short half-life of IL-6. Plasma IL-1β levels decreased when IL-6 was blocked in unstressed animals (CP versus CAb: p < 0.05), suggesting a role for IL-6 in the maintenance of IL-1β levels under tonic physiological conditions. At tissue level, pituitary gland mass increased significantly at time point 2, independently of stress when blocking IL-6 (CAb: p < 0.05). This suggests that when normal homeostasis is threatened, immediate adaption or at least compensation may occur. It was observed that GR, IL-1β, IL-1βR, IL-6, IL-6R and GABAARα1 showed no response to stress alone in the pituitary. It is therefore more likely that resistance to adaptation exists centrally. IL-1β and IL-1βR (p < 0.05) and GABAARα1 (p < 0.005) expression increased in the CAb group in the pituitary, again suggesting a role for IL-6 under control conditions. In terms of the adrenal, blocking IL-6 resulted in decreased glandular mass at time point 1, independent of stress (CAb and SAb: p < 0.005). The up-regulation in GR expression seen in CAb and SAb (p < 0.05) may be the effect of a compensatory mechanism to increase IL-6 dependent bioactivity of GCs. The fact that expression of IL-6, IL-6R, IL-1β and IL- 1βR consistently increased in the Ab groups, and mostly in the zona fasciculata and zona reticularis, suggests that lack of local direct negative cytokine feedback occurred in response to very low plasma IL-6 levels and that this contributes more than GCs in the down-regulation of inflammatory cytokine release. In conclusion, consistent effects of the Ab were apparent in the tissues investigated, even in control conditions, suggesting that IL-6 plays a role in the maintenance of basal homeostasis, including its regulation of the response to psychological stress. We found differential regulation in terms of cytokines and GCs when comparing peripheral versus central effects of stress and Ab, as well as the levels of cytokines in the blood compartment, compared to within tissues. / AFRIKAANSE OPSOMMING: Daar bestaan twee-rigting kommunikasie tussen HPA-as aktivering en interleukin-6 (IL-6), allhoewel die relatiewe bydrae van sentraal versus perifeer afgeskeide IL-6 nog onduidelik is, veral gedurende sielkundige strestoestande. IL-1β and IL-1βR (p <0.05) en GABAARα1 (p < 0.005) uitdrukking in die hipofise het toegeneem in die CAb groep, wat weereens ‘n rol vir IL-6 onder kontrole toestande uitwys. In terme van die bynier, het die blok van IL-6 ‘n afname in massa veroorsaak by tydpunt 1, wat weer onafhanklik van stres was (CAb en SAb: p < 0.005). Die opregulering in die CAb en SAb groepe (p < 0.05), kan wees as gevolg van ‘n kompensasie meganisme om IL-6 afhanklike GC aktiwiteit te verhoog. Die feit dat die uitdrukking van IL-6, IL-6R, IL-1β and IL-1βR in die Ab groepe deurlopend verhoog was, en meeste in die zona fasciculata en zona reticularis, stel voor dat daar ‘n tekort aan plaaslike, direkte sitokien negatiewe terugvoering was, as gevolg van die merkwaardige lae bloed IL-6 vlakke en dat dit meer bydra as GCs in die afregulering van inflammatoriese sitokien vrystelling.

Body composition and systematic low-grade inflammation in children : the PLAY study / Rachelle A. Pretorius

Background: Obesity-related diseases are arising as a major problem among children. inflammation has recently been identified to play an important role in the relationship between obesity.- as well as stunting-related diseases. Objectives: The aim of this study was to assess the association between serum tumour necrosis factor-alpha (TNF-α), interleukin-6 (IL-6) and C-reactive protein (CRP) concentrations and a variety of cardiometabolic and anthropometric indices of children in a township outside Potchefstroom, South Africa. Methods: Blood samples of 115 girls and 78 boys (mean age 15.6 ± 1.35) in the Physical Activity in the Young (PLAY) study were cross-sectionally analysed. Trained fieldworkers collected the demographic, Tanner growth stage and habitual physical activity information. Physiologists measured the children’s blood pressure. Anthropometric measurements were taken by. trained post-graduate students with level 1 or 2 qualifications in anthropometrics. A standard test battery was administered by trained postgraduate students in Human Movement Science to assess muscular strength. flexibility and endurance of the children. Blood samples were collected, centrifuged and stored frozen until further analyses. Results: Stunted girls had a significantly higher serum TNF-α concentration than the non-stunted girls (p=0.03). The factor analyses showed that the inflammatory. status clustered with the height for age-z-scores (HAZ) scores and the waist-hip-ratio (WHR). The HAZ-score of the over-fat boys (- 1.46) was significantly smaller than the lean boys (- 1.14, p=0.0 1). whereas the over-fat girls had a trend for a smaller HAZ-score (-1.07) than the lean girls (-0.89). No significant differences were found between the over-fat and the lean children-s inflammatory status. TNF-α and CRP levels tended to be higher in the over-fat children than in lean children. The girls' scrum IL-6 and CRP concentrations correlated significantly with their body mass index (BMI) and WHR (p<0.05 )and their TNF-α and IL-6 concentrations correlated significantly with their WHR (p<0.01 and p<0.05, respectively). Conclusion: In comparison to the non-stunted girls, stunted girls had a statistically significantly higher TNF-α concentration. Unusual fat distribution that is found in over-fat and stunted children may be associated with low-grade inflammation in children. Associação entre graus de mucosite e quantificação da interleucina 6 (IL- 6) e fator de necrose tumoral alfa (TNF-?) na saliva de pacientes submetidos a transplante de células-tronco hematopoiéticas (TCTH) / Association between degree of oral mucositis and quantification of interleukin 6 (IL-6) and tumor necrosis factor-alpha (TNF-?) in patients undergoing haematopoietic stem cell transplantation (HSCT)

A mucosite oral (MO) constitui uma condição dolorosa que se desenvolve entre 47% e 100% dos pacientes submetidos a transplante de células-tronco hematopoiéticas (TCTH), impactando enormemente em sua qualidade de vida. Investigar fatores preditivos para MO por meio de exames não invasivos faz-se necessário, visando melhorar a qualidade de vida dos pacientes. O objetivo do estudo foi investigar a relação dos regimes de condicionamento e dos níveis salivares de Interleucina 6 (IL- 6) e Fator de Necrose Tumoral alfa (TNF-?) com a MO, bem como investigar o impacto destes na qualidade de vida. Foram selecionados 82 pacientes submetidos a TCTH, que foram avaliados em 4 momentos diferentes: no início do condicionamento para o TCTH (M1), no dia da infusão das células (M2), após 12/20 dias do início do condicionamento para transplante autólogo e alogênico, respectivamente (M3), e após 30 dias ou na alta hospitalar para ambos (M4). Nestes momentos, foi avaliado clinicamente o grau de MO segundo critérios da Organização Mundial da Saúde (OMS), coletada saliva total e aplicados 3 questionários de avaliação de qualidade de vida em relação à MO e à saúde bucal: PROMS, OHIP-14 e OMQoL. As informações clínicas e laboratoriais foram correlacionadas através do STATA 13.0 com 5% de nível de significância. Verificou-se que a maior incidência e intensidade de MO, os piores índices de qualidade de vida e os maiores níveis de IL- 6 e TNF-? foram registrados no M3, porém não houve correlação entre as citocinas e graus de MO. Houve associação entre altos níveis salivares de IL-6 e maiores pontuações no PROMS. O regime de condicionamento mieloablativo (ML) foi relacionado à MO intensa (graus 3 e 4) e à maiores pontuações nos 3 questionários de qualidade de vida, e os escores dos questionários foram maiores conforme maior foi intensidade da MO (p<0,05). / Oral mucositis (OM) is a painful condition that develops between 47% and 100% of patients undergoing hematopoietic stem cell transplantation (HSCT), impacting greatly on their quality of life. Investigation of predictive factors for OM through noninvasive exams is necessary, in order to improve the quality of life of patients. The aim of the study was to investigate the relationship of conditioning regimens and salivary levels of Interleukin 6 (IL-6) and Tumor Necrosis Factor alpha (TNF-?) with OM, and to investigate their impact on quality of life. We selected 82 patients undergoing HSCT, which were assessed at four different moments: at the start of conditioning for HSCT (M1), on the cell infusion day (M2), after 12/20 days of the start of conditioning for autologous and allogeneic transplantation, respectively (M3), and after 30 days or at hospital discharge for both (M4). In these moments, it was clinically evaluated the degree of OM according to criteria of the World Health Organization (WHO), collected whole saliva and applied 3 questionnaires of assessment of quality of life related to OM and oral health: PROMS, OHIP-14 and OMQoL. Clinical and laboratorial data were correlated using STATA 13.0 in a 5% significance level. It was found that the highest incidence and intensity of OM, the worst indices of quality of life and higher IL-6 and TNF-? levels were found in M3, but there was no correlation between cytokines and levels of OM. There was an association between high levels of salivary IL-6 and higher scores in PROMS. The myeloablative conditioning regimen (ML) was related to intense OM (grades 3 and 4) and to highest scores in the 3 questionnaires of quality of life, and the scores of questionnaires were higher as higher was the intensity of OM (p<0,05).

Objetivo: determinar as concentrações séricas e peritoneais das interleucinas 1ß (IL-1ß) e 6 (IL-6) e da proteína amilóide A (SAA) em pacientes com endometriose pélvica. Métodos: foram avaliadas 97 pacientes submetidas à videolaparoscopia, 57 com endometriose (A), 27 com sintomas sugestivos porém sem endometriose (B) e 13 sem sintomas e doenças (C). Foram coletados no ato cirúrgico líquido peritoneal e sangue. As concentrações dos mediadores foram determinadas em pg/ml (IL-1 e 6) e ng/ml (SAA) por método imunoenzimático e leitura óptica. Resultados: (líquido peritoneal e sangue medianas) IL-1ß: A- 11,22 and 1,83; B- 15,62 and 1,16; C- 1,92 and 0,80; IL-6: A- 6,80 and 3,70; B- 8,60 and 3,90; C- 3,40 and 2,0; SAA- A- 310,30 and 14,01; B- 306,20 and 10,39; C- 53,4 e 9,5. Conclusão: as concentrações dos mediadores de inflamação avaliados estão elevadas no líquido peritoneal e no soro das mulheres com endometriose. Estas concentrações foram semelhantes às das pacientes com sintomas sugestivos porém sem a doença. A fase do ciclo menstrual, o tipo histológico envolvido e o local de acometimento da doença não influíram de forma significante nas concentrações séricas ou peritoneais de IL-1ß, IL-6 e SAA. / Objective: to assess peritoneal and seric interleukin-1ß (IL-1ß), 6 (IL-6), and protein amyloid A (SAA) concentrations in patients with pelvic endometriosis. Methods: 97 patients were submitted to video laparoscopic surgery, 57 with endometriosis (A), 27 with suggestive symptoms but no endometriosis (B), and 13 without symptoms and diseases (C). Peritoneal fluid and blood were collected during the procedure. Mediator s concentration was determined in pg/ml (IL-1ß and IL-6) and ng/ml (SAA) through immunoenzimatic test and optic measure. Results: (peritoneal fluid and blood -medians) IL-1ß: A- 11,22 and 1,83; B- 15,62 and 1,16; C- 1,92 and 0,80; IL-6: A- 6,80 and 3,70; B- 8,60 and 3,90; C- 3,40 and 2,0; SAA- A- 310,30 and 14,01; B- 306,20 and 10,39; C- 53,4 e 9,5. Conclusion: the inflammation mediators are increased in the peritoneal fluid and blood of women with pelvic endometriosis. The concentrations are similar to those found in patients with suggestive symptoms but no endometriosis. The place of the disease, the histological type and the day of the menstrual cycle didn t alter peritoneal or seric concentration of these cytokines.

Avaliação do efeito da interleucina-6 e da eritropoetina na lesão medular em ratos / Evaluation of the effect of interleukin-6 and erythropoietin in spinal cord injury in rats

Introdução: Este estudo teve como objetivo avaliar o efeito da interleucina 6 (IL-6) e eritropoetina (EPO) na lesão medular aguda experimental em ratos. Materiais e Métodos: A lesão foi induzida pelo equipamento padronizado NYU Impactor, com queda de um peso de 10 g à distância de 12,5 mm de altura. Foram utilizados 50 ratos da linhagem Wistar, divididos em cinco grupos de 10 animais. O grupo EPO, ratos tratados com EPO; grupo EPO/IL-6, animais tratados com EPO e IL-6; grupo IL-6, administração de IL-6; grupo placebo, solução placebo; grupo Sham, procedimento sham (apenas laminectomia, sem lesão medular). Todas as drogas e soro fisiológico foram administrados por via intraperitoneal, durante três semanas. Os animais foram acompanhados por 42 dias. A recuperação motora funcional foi monitorada pela escala de Basso, Beattie e Bresnahan (BBB) nos dias 2, 7, 14, 21, 28, 35 e 42. Exame de potencial evocado foi efetuado no 42o dia, sendo realizada análise histológica qualitativa e quantitativa após eutanásia. Resultados: Os resultados das avaliações da escala BBB mostraram recuperação funcional motora superior no grupo que recebeu EPO. A administração de IL-6 isolada não mostrou benefícios em relação ao grupo que recebeu solução placebo e a associação de IL-6 com EPO mostrou resultados inferiores ao grupo que recebeu apenas EPO. Conclusão: Concluímos que uso EPO após lesão medular contusa em ratos mostrou benefícios na recuperação motora. A associação de EPO e IL- 6 mostrou benefícios, porém com resultados inferiores aos da EPO isolada. O uso isolado de IL-6 não mostrou benefícios após lesão medular contusa experimental em ratos / Introduction: The aim of this study was to evaluate the effect of interleukin-6 (IL-6) and erythropoietin (EPO) in experimental acute spinal cord injury in rats. Methods: The injury was induced by a standardized equipment for spinal cord contusion injury, the NYU Impactor, which produced the lesion by means of a 10g weight drop on the animals\' spinal cord from a 12.5-mm height. Fifty Wistar rats were divided in five groups of ten animals: Group 1 rats treated with EPO; Group 2 animals treated with EPO and IL-6; Group 3, IL-6 administration; Group 4, placebo solution; Group 5, sham procedure (only laminectomy, without spinal cord injury). All drugs and placebo solution were administered intraperitoneally for three weeks. The animals were followed up for 42 days. The functional motor recovery was monitored by the scale of Basso, Beattie and Bresnahan (BBB) on days 2, 7, 14, 21, 28, 35 and 42. Evoked potential tests were performed on the 42nd day. Qualitative and quantitative histological analysis were performed after euthanasia. Results: The group receiving EPO demonstrated superior functional motor results in the BBB scale. IL-6 administration alone did not show benefits over the placebo group solution and the IL-6 combination with EPO showed results lower than those seen in the group that received only EPO. Conclusion: We conclude that using EPO after acute spinal cord injury in rats showed benefits in motor recovery. The association of EPO and IL-6 showed benefits, but with inferior results to the isolated EPO. Isolated use of IL-6 showed no benefit after experimental spinal cord injury in rats
