Inicialmente o método de adição de padrão interno foi aplicado no modo convencional na determinação de Ca e Mg em amostras biodiesel e urina por espectrometria de absorção atômica em chama de alta resolução e fonte contínua e na determinação de ureia em urina por espectroscopia Raman. Para fins comparativos, todas as amostras e materiais certificados foram também analisados pelos métodos de calibração externa, adição de padrão e padronização interna. Nitrato, Sr e Mn foram selecionados como padrões internos para ureia, Ca e Mg, respectivamente. Para avaliar a exatidão do método na determinação de Ca e Mg foram analisados nove materiais de referência certificados (CRM) e os resultados obtidos foram concordantes a 95% de confiança com os valores certificados (teste t). Para ureia a exatidão foi avaliada por meio de comparação dos resultados obtidos pelo método de adição de padrão interno e por um método de referência. Todos os resultados obtidos foram concordantes entre si (teste t pareado) ao nível de 95% de confiança. A exatidão do método também foi checada por meio de testes de adição e recuperação: Ca (93 – 119%), Mg (100 – 116%) e ureia (99 – 105%). A desvantagem do método de adição de padrão interno convencional foi o volume total de amostra utilizado, a quantidade de soluções analíticas empregadas e a baixa frequência analítica. No entanto, este problemas foram solucionados com o desenvolvimento de sistemas em fluxo e pelo uso de um gradiente de concentração. O método de adição de padrão interno por gradiente de concentração em fluxo foi avaliado na determinação de Na e K em amostras de biodiesel e urina por espectrometria de emissão atômica em chama e na determinação de etanol em gasolina por espectroscopia Raman. Lítio foi empregado como padrão interno para Na e K, e acetona foi utilizada como padrão interno para etanol. A exatidão do método na determinação de Na e K foi avaliada por meio da análise de cinco CRMs, e os resultados obtidos para Na foram concordantes a 95% de confiança com os valores certificados (teste t), para K os resultados apresentaram valores subestimados ao longo do trabalho (em torno de 80% de recuperação). Para determinação de etanol em gasolina a exatidão foi avaliada por meio da comparação dos resultados obtidos pelo método proposto e pelo método de referência, e todos os resultados obtidos foram concordantes entre si (teste t pareado) ao nível de 95% de confiança. A exatidão do método de adição de padrão interno por gradiente de concentração em fluxo também foi avaliada por meio de testes de adição e recuperação: Na (94 – 110%), K (73 – 86%) e etanol (99 – 104%). Os resultados obtidos utilizando o método de adição de padrão interno (convencional e em fluxo) em todas as aplicações apresentaram precisão e exatidão melhores ou comparáveis aos métodos de adição de padrão, padronização interna e calibração externa, mostrando ser uma estratégia de calibração eficiente para técnicas espectroscópicas. / The aim of this Doctoral Thesis was to evaluation of the internal standard addition method employing spectroscopic techniques. Initially, traditional standard addition method was applied for Ca and Mg determination in biodiesel and urine samples by high-resolution continuum source flame atomic absorption spectrometry and for urea determination in urine samples by Raman spectroscopy. For comparison purposes, all samples and certified materials were also analyzed by external calibration, standard addition and internal standardization methods. Nitrate, Sr and Mn were selected as internal standards for urea, Ca and Mg, respectively. In order to verify the accuracy for Ca and Mg determination were analyzed nine certified reference materials (CRM), the results were in agreement with certified values at the 95% confidence level (t-test). For urea, the accuracy was evaluated by comparison of the results obtained by internal standard addition and a reference method. All results were in agreement at 95% confidence level (paired t-test) with each other. The accuracy was also checked by addition and recovery tests: Ca (93 - 119%), Mg (100 - 116%) and urea (99 - 105%). The drawbacks of traditional internal standard addition were the total volume of sample used, the amount of analytical solutions employed and the lower sample throughput. However, these problems were solved with the development of flow systems and the use of a concentration gradient. The internal standard addition by flow gradient concentration was evaluated for Na and K determination in biodiesel and urine samples by flame emission atomic spectrometry and for ethanol determination in gasoline by Raman spectroscopy. Lithium was selected as internal standard for Na and K, and acetone was selected as internal standard for ethanol. The accuracy of the method for Na and K determination was evaluated by five CRMs analysis, and the results obtained for Na were in agreement with certified values at the 95% confidence level (t-test), for K the results presented underestimated values throughout the study (around 80% recovery). The accuracy for ethanol determination in gasoline was evaluated comparing the results obtained by the proposed method with the reference method. All results were in agreement at 95% confidence level (paired t-test) with each other. The accuracy of internal standard addition by flow gradient concertation method was also evaluated by addition and recovery tests: Na (94 - 110%), K (73 - 86%) e ethanol (99 - 104%). The results obtained by internal standard addition (traditional and flow modes) for all applications showed accuracy and precision better or comparable with traditional calibration methods (standard addition, internal standardization and external calibration), revealing to be an efficient calibration strategy for spectroscopic techniques. / FAPESP: 2012/23323-7

Quantitative Mass Spectrometric Investigations of Protein Biomarkers: Serum Thymidine Kinase 1 and Human Osteopontin

Mass spectrometry is being increasingly used in biomarker research mainly due to its ability to achieve high selectivity coupled with high sensitivity. This dissertation focuses on quantitative mass spectrometric investigations of two protein biomarkers i.e. serum thymidine kinase 1 (TK1) and human osteopontin (OPN). First part of this research was focused on developing a liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) method for measuring the activity of TK1 in serum by monitoring the conversion of a TK1 specific exogenous substrate, 3’-deoxy-3’-fluorothymidine (FLT), to its mono-phosphorylated form 3’-deoxy-3’-fluorothymidine monophosphate (FLT-MP). A method to quantify FLT-MP on LC-MS/MS was developed and validated. The method was linear over the range of 2.5-2000 ng/mL with a mean correlation coefficient of 0.9935. Using the developed method, serum TK1 activity was measured in serum from hepatocellular carcinoma (HCC) patients and age-matched controls under standardized conditions. A sub-population of the HCC patient samples showed an almost 20-fold enhanced TK1 activity compared to the controls. A method was developed and validated for quantifying human osteopontin from plasma using immunoaffinity isolations coupled with microflow liquid chromatography and tandem mass spectrometry (MFLC-MS/MS). A biologically relevant tryptic peptide ‘GDSVVYGLR’ which is unique to hOPN was identified and used as a signature peptide for this method. The method was validated over a range of 25-600 ng/mL. The performance of the method was compliant with USFDA validation guidance. In addition, a stable isotope labeled (SIL) peptide GDSVVYGLR* and an extended SIL peptide TYDGRGDSVV*YGLRSKSKKF’ were evaluated as internal standards (IS) to account for signature peptide digestion instability and variability. In the digestion variability studies, the use of extended SIL peptide as internal standard limited the total variability within ±30%. Alternatively, when SIL peptide was used as internal standard the variability ranged from -67.4% to +50.6 %. The applicability of the validated method was demonstrated by analyzing plasma samples obtained from 10 healthy individuals and 10 breast cancer patients. More than 9-fold increase in the mean plasma hOPN concentration was seen in 30% of the breast cancer patient samples (n=10) in comparison to the healthy volunteer samples. In a proof of concept investigation, a stable isotope labeled signature peptide was evaluated as an internal standard to compensate for immunocapture variability during quantification of human osteopontin (hOPN) by immunoaffinity coupled LC-MS/MS. Immunocapture variability was induced by varying the antibody amount per well. The immunocapture variability ranged from -80.9 % to +77.0 % when the IS was added after immunocapture and from -37.5% to +20.3% when the IS was added before immunocapture. The lower variability demonstrates the ability of SIL-IS peptide to compensate for variation during immunocapture.

Laser-Induced Breakdown Spectroscopy: Simultaneous Multi-Elemental Analysis and Geological Applications

Under high irradiation, a fourth state of matter named plasma can be obtained. Plasmas emit electromagnetic radiation that can be recorded in the form of spectra for spectroscopic elemental analysis. With the advent of lasers in the 1960s, spectroscopists realized that lasers could be used simultaneously as a source of energy and excitation to create plasmas. The use of a laser to ignite a plasma subsequently led to laser-induced breakdown spectroscopy (LIBS), an optical emission spectroscopy capable of analyzing samples in various states (solids, liquids, gases) with minimal sample preparation, rapid feedback, and endowed with in situ capability. In this dissertation, studies of LIBS for multi-elemental analysis and geological applications are reported. LIBS was applied to cosmetic powders for elemental analysis, screening and classification based on the raw material used. Principal component analysis (PCA) and internal standardization were used. The intensity ratios of Mg/Si and Fe/Si observed in talcum powder show that these two ratios could be used as indicators of the potential presence of asbestos. The feasibility of LIBS for the analysis of gasification slags was investigated and results compared with those of inductively-coupled plasma−optical emission spectrometry (ICP-OES). The limits of detection for Al, Ca, Fe, Si and V were determined. The matrix effect was studied using an internal standard and PLS-R. Apart from V, prediction results were closed to those of ICP-OES with accuracy within 10%. Elemental characterization of outcrop geological samples from the Marcellus Shale Formation was also carried out. The matrix effect was substantially reduced. The limits of detection obtained for Si, Al, Ti, Mg, Ca and C were determined. The relative errors of LIBS measurements are in the range of 1.7 to 12.6%. Gate delay and laser pulse energy, have been investigated in view of quantitative analysis of variation of trace elements in a high-pressure environment. Optimization of these parameters permits obtaining underwater plasma emission of calcium with quantitative results on the order of 30 ppm within a certain limit of increased pressure. Monitoring the variation of the trace elements can predict changes in the chemical composition in carbon sequestration reservoir.

Um estudo sistemático a respeito da utilização de padrão interno em determinações multielementares por espectrometria de absorção atômica (ETAAS) foi desenvolvido. O objetivo principal do presente trabalho foi verificar a possibilidade de melhorar a precisão e a exatidão dos resultados analíticos, que são obtidos na análise de fluidos biológicos. O pré-tratamento dessas amostras foi simplificado e reduzido a uma única etapa de diluição com surfactante (Triton X-100) e ácido (HNO3). Conseqüentemente, a complexidade da solução diluída de amostra, a ser introduzida no tubo de grafite, apresenta uma elevada quantidade de concomitantes que podem provocar interferências químicas. A seleção preliminar dos elementos a serem testados como padrão interno considerou a semelhança de parâmetros físico-químicos relacionados com o processo de atomização. Desta forma, Ag, Bi, In e Tl foram testados como padrão interno para a determinação simultânea de Cd/Pb em sangue e urina, enquanto Bi, Ge, In, Sb, Sn e Te foram os elementos selecionados para a determinação de Mn/Ni/Se em soro sangüíneo. A melhoria da qualidade dos resultados analíticos obtidos na determinação simultânea de Cd e Pb em sangue foi observada quando Ag foi utilizada como padrão interno, na presença de NH4H2PO4 como modificador químico. Verificou-se uma melhoria na exatidão dos resultados obtidos para Cd e Pb, após a correção com padrão interno. Por outro lado, os resultados obtidos na análise de urina não foram corrigidos por nenhum dos elementos testados. Os melhores resultados para a determinação simultânea de Mn, Ni e Se foram obtidos com a utilização de Bi, Sn e Te como padrão interno. Entretanto, verificou-se que a correção de todos os resultados não seria viável com o uso de um único padrão interno. O melhor desempenho nos testes realizados na presença de soro sangüíneo foi obtido com Bi, que melhorou discretamente a precisão dos resultados obtidos para Se. Desta forma, a padronização interna visando a determinação simultânea de Mn, Ni e Se não foi eficiente. A padronização interna em ETAAS, com a finalidade de melhorar a precisão e a exatidão dos resultados analíticos, é uma estratégia tão complexa, quanto os efeitos interferentes que se pretende corrigir: são necessários mais estudos para compreender melhor como a utilização de uma condição de compromisso afeta os processos de atomização, bem como mais informações a respeito das interferências físicas e químicas causadas por amostras complexas, analisadas por ETAAS após uma simples etapa de diluição. Deve-se considerar com especial atenção o modificador químico e as temperaturas das etapas de pirólise e de atomização empregadas, que são parâmetros críticos para o desempenho de um elemento como padrão interno. / A systematic study involving the use of internal standard for multielement determinations by electrothermal atomic absorption spectrometry was developed. The preliminary selection of the elements to be tested as internal standard considered the resemblance of physico-chemical parameters related with the atomization process. Thus, Ag, Bi, In and Tl were tested as internal standard for the simultaneous determination of Cd/Pb in blood and urine, and Bi, Ge, In, Sb, Sn and Te were the selected elements for the determination of Mn/Ni/Se in blood serum. The correction of the results obtained for the simultaneous determination of Cd and Pb in blood was achieved when Ag was used as internal standard, in presence of NH4H2PO4 as chemical modifier. An improvement for the accuracy of the results was observed for both analytes after their correction with the internal standard. On the other hand, the results obtained for the urine analysis were not corrected by using the tested elements. The best results for the simultaneous determination of Mn, Ni and Se were observed when Bi, Sn and Te were used as internal standard. However, the correction for the results for all analytes was not possible by using only one internal standard. The best performance in presence of the serum was obtained for Bi, which improves slightly the precision for the Se results. Thus, the internal standardization for the simultaneous determination of Mn, Ni and Se was not efficient. The internal standardization in ETAAS, aiming the improvement of precision and accuracy of the analytical results, is a strategy as complex as the interference effects to be corrected: more studies are required in order to better understand how the adoption of a compromised condition disturbs the atomization processes, as well as to get more information about the physical and chemical interference caused by complex samples, analyzed by ETAAS after a single dilution step. The chemical modifier and the selected temperatures for the pyrolysis and atomization steps are critical parameters for the performace of an internal standard and they should be carefully considered.

Laserablation mit induktiv gekoppelter Plasma-Massenspektrometrie für die medizinische Diagnostik

In dieser Arbeit wurde eine neue Markierungsstrategie von Antikörpern mit dem Markierungsreagenz MeCAT (Metal Coded Tag) unter physiologischen Reaktionsbedingungen, sowie deren Anwendung in einem 8-fach Multiplex-Immunoassay von in Formalin-fixierten und in Paraffin-eingebetteten Gewebeschnitten entwickelt. Für eine aussagekräftige LA-ICP-MS Detektion von MeCAT-modifizierten Antikörpern, wurde eine Standardisierung für biologische Proben auf NC-Membranen, basierend auf einer homogenen Aufbringung eines internen Standards und Kalibrierstandards durch einen kommerziell verfügbaren Tintenstrahldrucker entwickelt und mit der ICP-MS Analyse von Lösungen evaluiert. Die LA-ICP-MS wurde in zwei 8-fach Multiplex-Immunoassays von Tissue Micro Arrays vom Prostatakarzinom und in Maushirngewebeschnitten zur Einschätzung von neurogenerative Erkrankungen erfolgreich eingesetzt werden. Es konnte hierbei gezeigt werden, dass das Nachweisvermögen, der hier entwickelten Methode bereits ausreicht, um die gängigen klinischen Biomarker mit guter Ortsauflösung nachzuweisen. / In this work a new tagging strategy of antibodies with the tagging reagent MeCAT (Metal Coded Tag) was developed under physiological reaction conditions. Their application was proved in an 8-fold multiplex immunoassay of formalin-fixed and paraffin-embedded tissue sections. For a significant LA-ICP-MS detection of MeCAT tagged antibodies standardization for biological samples owere developed. The standardization based on a homogeneous deposition onto the NC membrane via conventional CD-ink-jet printer was validated in addition with the ICP-MS analysis of solutions. The internal standardization of LA-ICP-MS was successfully applied in two 8-fold multiplex immunoassays for Tissue Micro Arrays (TMA) of prostate cancer and for detection of biomarkers for neurodegenerative diseases in mouse brain tissue sections. In both examples it could be shown that the detection capability of the new tagging strategy in combination with the printing standardization allows the detection of the clinical biomarker with good spatial resolutions.
