Epitélio intestinal de juvenis de pacu (Piaractus Mesopotamicus, Holmberg 1887) e dourado (Salminus brasiliensis, Cuvier 1816) alimentados com dieta contendo colostro bovino liofilizado / Intestinal epithelium of pacu (Piaractus mesopotamicus, Holmberg 1887) and dourado (Salminus brasiliensis, Cuvier 1816) juveniles fed diet containing lyophilized bovine colostrum

O objetivo deste trabalho foi avaliar o efeito do colostro bovino liofilizado (CBL), utilizado como fonte parcial da dieta protéica, sobre as características histológicas do epitélio intestinal de juvenis de pacu (Piaractus mesopotamicus) e dourado (Salminus brasiliensis). Os juvenis foram distribuídos num delineamento experimental inteiramente casualizado em esquema fatorial 3x2. Foram utilizadas dietas com três níveis de inclusão de CBL (0%, 10% e 20%) e dois períodos experimentais (30 e 60 dias), oferecidas duas vezes ao dia até a saciedade aparente. Para o estudo histológico, o intestino foi dividido em três segmentos, S1, S2 e reto para pacu e S1, S2 e intestino posterior para dourado. Foram avaliadas a espessura da camada muscular; o volume parcial da mucosa absortiva (Vv); o número das células caliciformes contendo mucinas ácidas e neutras e totais, e os subtipos ácidas - sialomucinas e sufomucinas. Nos juvenis de pacu, a inclusão de 20% de CBL alterou a distribuição das células caliciformes contendo as mucinas ácidas, neutras e totais, os subtipos sialomucinas e sulfomucinas, e a espessura da camada muscular, enquanto o Vv foi afetado apenas pelo período experimental. Nos juvenis de dourados, efeito de período experimental foi observado para células caliciformes contendo mucinas ácidas, neutras e totais e os subtipos sialomucinas e sulfomucinas, espessura da camada muscular e Vv. A adição de 10% de CBL afetou apenas o Vv no segmento S1. Considerando os aspectos avaliados no presente estudo, a presença do colostro bovino liofilizado na dieta influenciou, no período estudado, as características histológicas entéricas de juvenis de pacu, enquanto que nos juvenis de dourado influencia desta secreção láctea não foi observada. / The aim of this study was to evaluate the effect of lyophilized bovine colostrum (LBC), used as partial source of protein in the diet, on the histological characteristics of the intestinal epithelium of juvenile pacu (Piaractus mesopotamicus) and juvenile dourado (Salminus brasiliensis). Juveniles were distributed in a completely randomized design in a factorial scheme 3x2. Three diets were used with different levels of LBC inclusion (0%, 10% and 20%) and two experimental periods (30 and 60 days) offered twice daily until apparent satiation. For the histological study, the intestine was divided into three segments, S1, S2 and rectum to the pacu and S1, S2 and posterior intestine to the dourado. The muscle layer thickness; the mucosal absorptive volume (Vv); the number of goblet cells containing acidic and neutral mucins and the acidic subtypes - sialomucin and sulphomucin were evaluated. In juvenile pacu, the inclusion of 20% of LBC changed the distribution of goblet cells containing acidic, neutral and total mucins, the subtypes sialomucins and sulphomucins, and the thickness of the muscle layer, while the Vv was affected only by the experimental period. In juvenile dourado, effect of experimental period was observed for goblet cells containing acidic, neutral and total mucins and subtypes sialomucins, sulphomucins, thickness of muscle layer and Vv. The addition of 10% of LBC affected only Vv in the segment S1. Considering the aspects studied, the presence of lyophilized bovine colostrum in the diet influenced, in the period studied, the enteric histological characteristics of juvenile pacu, while the juvenile dourado influence of this lacteal secretion was not observed.

Moreover, those species can display other behavioral adaptations, such as food flexibility. Thus, this study analized structure and composition of the ichthyofauna in accordance with the salinity gradients as well as ecology and trophic organization. Samples were collected in rainy and dry season, from January to Decembre 2012, in 12 points distributed into three areas defined according to the salinity gradient (Inferior, Intermediate and Upper). The fishes were captured using a beach seine. The low rainfall, high evaporation rate and the small input of freshwater in Tubarão River estuary favored a salinity between 30 and 45. The structure and composition of ichthyofauna showed spatial and temporal differences between the Tubarão River estuary areas, with salinity and substrate influencing the distribution of species. Atherinella brasiliensis, Lile piquitinga, Eucinostomus argenteus, Ulaema lefroyi, Lutjanus analis and Sphoeroides greeleyi are the most abundant distributed in all the estuarine areas. Among the 104 species caught at the estuary of Tubarão River, 18 were selected according to their abundance, for analysis of trophic ecology. However, only the spacial variation showed significant changes in fish diet. In general, the diet of the species was based mainly on Macroalgae, Polychaeta, Decapoda, Mollusca and Zooplankton. Among these items the representatives of Zooplankton are the main prey in the diet of small juveniles. Because of this preference for certain food items species were grouped into six different trophic guilds: Zooplanktivore, Omnivore, Herbivore-Macroalgae, Zoobenthivore- Hyperbenthos, Zoobenthivore-Epifauna and Zoobenthivore-Infauna. The largest overlap values occured in the Upper zone, due to a higher salinity restricting the establishment of various prey. The results presented in this study showed the direct relationship of the structure and composition of ichthyofauna and the trophic ecology with habitat heterogeneity and the variation in salinity. Thus, the data obtained in the study allows our understanding of the dynamics of community structure that hypersaline system and favors the development of approaches to conservation and sustainable use of biological diversity. / Nos sistemas estuarinos hipersalinos as espécies de peixes se movimentam entre os locais de maior oferta de recursos e entre os diferentes gradientes de salinidade, de acordo com a tolerância fisiológica que é uma adaptação comum para espécies de peixes estuarinos. Além disso, estas espécies podem exibir outras adaptações comportamentais, como a flexibilidade alimentar. Assim, o estudo analisou a estrutura e composição da ictiofauna de acordo com o gradiente de salinidade, bem como a ecologia e a organização trófica. As amostragens foram realizadas durante a chuva e a seca de 2012 em 12 pontos distribuídos em três zonas definidas de acordo com o gradiente de salinidade (Inferior, Intermediária e Superior). Os peixes foram capturados com a utilização de uma rede beach seine através de arrasto de praia. O baixo índice pluviométrico, a elevada taxa de evaporação e a pequena entrada de água doce presentes no estuário do Rio Tubarão favoreceram uma salinidade entre 30 e 45. A estrutura e composição da ictiofauna apresentaram diferenças espacial e temporal entre as zonas do estuário do rio Tubarão, com a salinidade e o substrato influenciando na distribuição das espécies Atherinella brasiliensis, Lile piquitinga, Eucinostomus argenteus, Ulaema lefroyi, Lutjanus analis and Sphoeroides greeleyi Dentre as 104 espécies capturadas no estuário do Rio Tubarão 18 foram selecionadas, de acordo com sua abundância, para análise da ecologia trófica. No geral a dieta das espécies foi baseada, principalmente, em Macroalgas, Polychaeta, Decapoda, Mollusca e Zooplâncton. Dentre esses itens os representantes do Zooplâncton são as presas principais na dieta dos pequenos juvenis. Devido a essa preferência por determinados itens alimentares as espécies foram agrupadas em seis guildas tróficas, distintas: Zooplanctívoro, Onívoro. HerbívoroMacroalga, Zoobentívoro-Hiperbentos, Zoobentívoro-Epifauna e Zoobentívoro-Infauna. Os maiores valores de sobreposição foram registrados na zona Superior, possivelmente pela elevada salinidade que restringe o estabelecimento de diversas presas. Em geral os resultados apresentados nesse estudo mostram a relação direta da estrutura e composição da ictiofauna e a ecologia trófica das espécies com a heterogeneidade do habitat e a variação de salinidade que possibilita proteção aos peixes juvenis e maior abundância de recursos tróficos. Assim, os dados obtidos no estudo possibilita a compreensão da dinâmica da estrutura da comunidade desse sistema hipersalino, bem como favorece o desenvolvimento de abordagens para a conservação e uso sustentável da diversidade biológica.

The influence of marsh edge and seagrass habitat on summer fish and macroinvertebrate recruitment to a northern Gulf of Mexico coastal system

Marshes and seagrass beds have been widely recognized as important habitat for estuarine species, but less has been done on how these habitats interact and function together, thereby limiting understanding of the variability of juvenile recruitment to coastal systems. Therefore, the objective of this study was to assess the interaction between fringing marsh and adjacent seagrass for the provision of habitat for juvenile nekton. Weekly seine net and benthic seagrass core sampling from June to November 2020 determine the relationship between nekton and marsh-edge and seagrass habitat. This study shows disparate results, in terms of the effects of proximity to marsh edge and seagrass biomass on nekton abundance and size, pointing to different selectivity of marsh edge versus seagrass by different species. In addition, there are no effects of proximity to marsh edge and seagrass biomass on community composition, but an interactive effect on community dispersion.

L’évaluation de l’impact des polluants dans l’environnement est une des préoccupations majeures qui s’inscrit dans la Directive Européenne Cadre Eau 2000. Les réglementations préconisées ont notamment pour objectif de parvenir au bon état chimique et écologique des masses d’eau. Dans ce contexte, notre étude a consisté à développer une approche multibiomarqueur sur des juvéniles de poisson afin d’évaluer les effets biologiques de la pollution chimique en milieu littoral. Des paramètres moléculaires de détoxification (EROD, GST) et une enzyme antioxydante (CAT) ont été utilisés en tant que «système d’alarme » susceptibles de détecter une perturbation avant l’apparition de signes pathologiques irréversibles. En parallèle, différents biomarqueurs physiologiques (croissance somatique et récente, rapport ARN/ADN, indices morphométrique et lipidique) ont été analysés en considérant que ceux-ci pourraient révéler les dommages induit par les polluants sur l’état de santé des juvéniles. La sensibilité et la pertinence des biomarqueurs moléculaires et physiologiques ont été testés expérimentalement sur des juvéniles : i) de bar exposés à une pollution aigüe de pétrole, ii) de bar et de turbot soumis à des mélanges de contaminants en concentrations environnementales en conditions contrôlée et semi-contrôlée (« caging »). Nos résultats montrent la capacité de l’EROD, et à un degré moindre de la GST, à détecter une exposition courte (2 et 4 jours) des organismes au pétrole et à refléter ses effets délétères sur leur état de santé. Cette relation entre biomarqueurs moléculaires et physiologiques a par contre été plus difficilement établie dans un contexte de pollution multiple. Les indices de croissance et de condition utilisés se sont avérés plus sensibles aux différents niveaux de contamination analysés (métaux et HAPs). Leur utilisation a permis d’évaluer la condition affaiblie des organismes mis en cage en milieu portuaire pendant 38 jours. Cette expérience de « caging » s’est révélée concluante, notamment pour les juvéniles de bar, sur lesquels aucun stress physiologique de la mise en cage n’a été détecté dans la station de référence. Les effets délétères des contaminants chimiques sur l’état de santé des juvéniles de turbot ont également été observés en condition contrôlée après exposition de 21 jours aux mêmes sédiments portuaires et à un sédiment estuarien. En complément de ces expériences, une étude de terrain a été réalisée sur des juvéniles de flet prélevés dans des estuaires le long de la côte française et belge. Une diminution des indices morphométrique et lipidique des juvéniles de flet, issu des trois estuaires anthropisés, a été observée en relation avec des bioconcentrations en métaux plus élevées que l’estuaire de référence. Les résultats issus de ces différentes études montrent la potentialité des indices de croissance et de condition à révéler les effets biologiques des contaminants chimiques sur les juvéniles de poissons marins. Cependant, leur spécificité vis à vis des polluants étant plus faible que les paramètres de détoxification, leur utilisation peut être limitée. Ces travaux montrent ainsi le besoin d’utiliser des biomarqueurs à différents niveaux d’organisation biologique dans les programmes de biosurveillance. / Evaluation of pollutant effects in environment is one of the major issues of the European Water Framework Directive 2000. Regulations have particularly the objective to reach to a good chemical and ecological status of water bodies. In this context, the aim of our study was to develop a multibiomarker approach on juvenile marine fish in order to evaluate the biological effects of chemical pollution in coastal areas. Molecular detoxification parameters (EROD, GST) and an antioxidant enzyme (CAT) were used as early warning tools of toxicity allowing the prevention of irreversible damages. In parallel, different physiological biomarkers (somatic and recent growth, RNA:DNA ratio, morphometric and lipidic indices) were analysed as reflecting damages on juveniles health. Sensitivity and relevance of molecular and physiological biomarkers were tested experimentally on juvenile : i) sea bass exposed to acute petroleum pollution, ii) sea bass and turbot submitted to a mix of contaminants in environmental concentrations during controlled and semi-controlled (caging) conditions. Our results show the ability of EROD, and in lower degree the GST, to detect short exposure (2 and 4 days) of organisms to petroleum and to reflect their deleterious effects on fish health. This relationship between molecular and physiological biomarkers was more difficultly established under multiple pollutions. Growth and condition indices were found to be more sensitive to the different levels of chemical contamination analysed (metal, PAHs). Their analyses allowed us to evaluate the weakened condition of organisms caged in the harbour area during 38 days. This caging experiment was relevant especially for juvenile sea bass in which no physiological stress was detected in the reference station. Deleterious effects of chemical contaminant on turbot juvenile health were also observed in controlled condition after 21 days exposure to the same harbour sediments and to an estuarine sediment. In complement to these experiments, a field study was realized on juvenile flounders sampled in some estuaries along the French and Belgium coast. A decrease of morphometric and lipidic indices were found in juvenile flounders from the three anthropogenic estuaries showing the highest metal bioconcentrations compared to the reference estuary. Results from these different studies showed the potentiality of growth and condition indices to reflect biological effects of chemical contaminants on juvenile marine fish. However, their use could be limited by their lower specificity to pollutant than parameters involved in detoxification. These works show therefore the need to use biomarkers at different level of biological organization in biomonitoring programs.

Monitoring plůdkových společenstev ve vybraných tocích na Moravě / Monitoring of 0+ juvenile fish communities in selected Moravian rivers

Monitoring of 0+ juvenile fish communities was realized in years 2012 and 2013 in 39 localities in 25 selected Moravian rivers. It was realized in collaboration with Povodí Moravy SOE research workers. The aim of this diploma thesis was to conduct a survey of 0+ juvenile fish communities and evaluate basic attributes of these ichthyocenosis (species richness, biodiversity, equitability, abundance and domiance). Secondary aim was the evalutation od hydromorphological conditions and water quality based on data provided by Povodí Moravy SOE. This monitoring was realized using the mobile FEG 1500 electrofishing device. The electrofishing crew moved upstream using continuous fishing strategy. Each locality was selected by highly experienced research workers of Povodí Moravy SOE. Caught fish were determined immediately and carefully retruned to the stream. In total there was present 28 0+ juvenile fish species. The most frequent was chub (Leuciscus cephalus, 23 localities), gudgeon (Gobio gobio, 19 localities) and roach (Rutilus rutilus, 19 localities). In 13 localities occured the fish fry of barbel (Barbus barbus) wich is positive surprise due to its sensitivity to water pollution. In contrast the massive presence of round goby (Neogobius melanostomus) in some localities was an unpleasant finding. 4 localities were without fish fry. This survey has observed increasing trend of biodiversity depending on improving of hydromorphological status while the abundance showed almost no diference. Saprobic index of 0+ juvenilie fish community increased with deteriorating of water quality which could mean that the fish fry is relatively reliable indicator of water quality. The results of this diploma thesis could be used in documenting of Moravian streams and in comparisson with another monitorings in these localities.

Les habitats côtiers à fonds meubles sont essentiels dans le cycle de vie de nombreuses espèces de poissons. Ces habitats ont pour caractéristique d’être peu étendus mais sont aussi très productifs et jouent à ce titre un rôle de nourricerie pour les juvéniles de poissons bentho-démersaux. Les fortes abondances de proies semblent être l’une des raisons principales de la présence des juvéniles au sein de ces habitats. Bien que déjà étudié, le caractère limitant de la nourriture disponible fait toujours l’objet de débats dans la communauté scientifique. Une des raisons principales est que l’étude des milieux côtiers est rendue complexe par la diversité des facteurs entrant en jeu dans le développement des jeunes stades de poissons. A partir du cas d’étude de la baie de Vilaine, une des nourriceries les plus productives du golfe de Gascogneles recherches menées dans cette thèse visent à définir le rôle du facteur alimentaire dans l’organisation de la nourricerie et dans sa capacité à soutenir le développement des juvéniles de poissons. Le fil conducteur de cette thèse est donc de déterminer si le facteur alimentaire limite la production de juvéniles. Les résultats montrent une forte exploitation de la production alimentaire ainsi qu'une superposition spatiale entre les densités de juvéniles de poissons et de leurs proies. La cohérence de ces résultats tend à soutenir l'hypothèse que la capacité d'accueil de la baie de Vilaine est atteinte et donc que le facteur alimentaire est limitant. Les implications de ce mécanisme de régulation sur la dynamique des populations nourricer / Soft sediment coastal habitats are essential in the life cycle of numerous fishes. These habitats are spatially-limited but very productive, and play a key role of nursery for the juveniles of benthic and demersal fishes. High abundance of prey seems to be one of the main reasons of the presence of juvenile fish within these habitats. Although widely studied, the limiting aspect of the feeding factor is still debated in the scientific community. One of the main reasons is that studying coastalhabitats is complex because numerous factors influence the development of the first life stages of fish. From the study case of the Bay of Vilaine, one of the most productive nurseries of the Bay of Biscay, research conducted in this thesis aims to define the role of the feeding factor in the organisation of the nursery and in its capacity to support the development of juvenile fish. The common thread of this thesis is thus to determine if the feeding factor limits the juvenile fish production. The results show a strong exploitation of the food production by the juvenile fish community and a spatial match between the densities of juvenile fish and their prey. The consistency of these results tends to support that the hypothesis that the carrying capacity of the Bay of Vilaine is reached, and that the feeding factor is limiting. The consequences of this regulation process on the dynamics of nursery-dependent fish populations are discussed.
