Structural Studies of O-antigen polysaccharides, Synthesis of 13C-labelled Oligosaccharides and Conformational Analysis thereof, using NMR Spectroscopy

<p>In order to understand biological processes, to treat and diagnose diseases, find appropriate vaccines and to prevent the outbreak of epidemics, it is essential to obtain more knowledge about carbohydrate structures. This thesis deals with structure and conformation of carbohydrates, analysed by NMR spectroscopy and MD simulations.In the first two papers, the structures of O-antigen polysaccharides (PS) from two different <i>E. coli</i> bacteria were determined using NMR spectroscopy. The O-antigenic PS from <i>E. coli</i> O152 (paper I) consists of branched pentasaccharide repeating units, built up of three different carbohydrate residues and a phosphodiester, whilst the repeating unit of the O-antigen from <i>E. coli</i> O176 (paper II) is built up of a linear tetrasaccharide consisting of two different monosaccharides.</p><p>In papers III and IV, the conformational analysis of different disaccharides is described. Conformational analysis was performed using NMR spectroscopy and MD simulations (paper IV). In paper III four different glucobiosides were studied using coupling constants and Karplus-type relationships. By use of specific <sup>13</sup>C isotopically labelled derivatives, additional coupling constants were obtained and the number of possible torsion angles was reduced by half. In paper IV, we examine the conformations of two disaccharides that are part of an epitope of malignant cells. From NOE and T-ROE experiments, short proton-proton distances around the glycosidic linkage were estimated. Furthermore, interpretation of the extracted coupling constants using Kaplus relationships gave the values of the torsion angles. As in paper III, isotopically labelled compounds were synthesised in order to enhance the sensitivity of the analysis. Finally, MD simulations were performed and the results were compared with results from NMR data.</p>

Is it rational to buy eco-labelled food? : A study of the knowledge of, willingness to pay for and reasons to purchase eco-labelled seafood in Sweden

<p>Eco-labelling is a way to promote sustainable development. This is a quantified study about purchase behaviour regarding eco-labelled seafood in Sweden, based on interviews with a convenience sample of consumers in situ. The objectives were to establish how large the knowledge of eco-labelled seafood is, to investigate the reasons for acquiring the product and thus determine the prime driving force to do it. The willingness to pay for eco-labelled seafood was also investigated. The results show a low awareness where just about a quarter of the respondents knew that eco-labelled seafood existed. The willingness to pay was high; four out of five were ready to spend additional money on an eco-labelled product. Women displayed a higher willingness to pay than men, which supports previous research. Regarding the reasons to purchase the only options presented to the respondents were environmental concern, enhanced health or both alternatives in combination. Health as the single factor was the least preferred choice and the two other alternatives were favoured to an equal amount. A difference in purchase behaviour can be detected among respondents with awareness of ecolabelled seafood when compared to those without. Those aware stated a willingness to pay to a higher degree and also displayed more environmental concern and less health interest than those unaware of eco-labelled seafood.</p>

Eco-labelling of houses and the concept of sustainable housing : A comparative study of the UK and Sweden

<p>The aim with this study is to evaluate if standardization of sustainable housing is a practical solution to address the objectives/reach the goals of sustainable housing. The point of departure is that eco-labelling of houses is a way to put sustainable housing into practice. The Nordic Swan criteria for small houses and the UK Code for Sustainable Homes are two recently developed schemes for sustainable construction and design. These two labelling systems are chosen as case studies in order to compare and examine the practice of sustainable housing. How can the goals of sustainable housing be met in the form of eco- labelling? What are the benefits and obstacles/limitations for the cases to be effective and sustainable?  What can explain differences between the two cases? And is the function and organisation of the cases best explained by sustainable development ideas or ecological modernization?  It is suggested in the view of path dependency theory that the UK’s and Sweden’s decisions based on their expectations concerning development of the EU flower has made their respective eco-labelling of houses to have different organisational structure. The Code as a sector-specific label has the advantage, compared to the Swan, of being able to change its structure depending on the needs of pressure in the network of government, business and consumers.  The Code has several criteria concerning sustainable living besides criteria concerning environmental construction of the house and can therefore be considered as part of the sustainable development discourse.</p>

Design and Synthesis of 11C-Labelled Compound Libraries for the Molecular Imaging of EGFr, VEGFr-2, AT1 and AT2 Receptors : Transition-Metal Mediated Carbonylations Using [11C]Carbon Monoxide

This work deals with radiochemistry and new approaches to develop novel PET tracers labelled with the radionuclide 11C. Two methods for the synthesis of 11C-labelled acrylamides have been explored. First, [1-11C]-acrylic acid was obtained from a palladium(0)-mediated 11C-carboxylation of acetylene with [11C]carbon monoxide; this could be converted to the corresponding acyl chloride and then combined with benzylamine to form N-benzyl[carbonyl-11C]acrylamide. In the second method, the palladium(0)-mediated carbonylation of vinyl halides with [11C]carbon monoxide was explored. This latter method, yielded labelled acrylamides in a single step with retention of configuration at the C=C double bond, and required less amine compared to the acetylene method. The vinyl halide method was used to synthesize a library of 11C-labelled EGFr-inhibitors in 7-61% decay corrected radiochemical yield via a combinatorial approach. The compounds were designed to target either the active or the inactive form of EGFr, following computational docking studies. The rhodium(I)-mediated carbonylative cross-coupling of an azide and an amine was shown to be a very general reaction and was used to synthesize a library of dual VEGFr-2/PDGFrβ inhibitors that were 11C-labelled at the urea position in 38-78% dc rcy. The angiotensin II AT1 receptor antagonist eprosartan was 11C-labelled at one of the carboxyl groups in one step using a palladium(0)-mediated carboxylation. Autoradiography shows specific binding in rat kidney, lung and adrenal cortex, and organ distribution shows a high accumulation in the intestines, kidneys and liver. Specific binding in frozen sections of human adrenal incidentalomas warrants further investigations of this tracer. Three angiotensin II AT2 ligands were 11C-labelled at the amide group in a palladium(0)-mediated aminocarbonylation in 16-36% dc rcy. One of the compounds was evaluated using in vitro using autoradiography, and in vivo using organ distribution and animal PET. The compound was metabolized fast and excreted via urine. High radioactivity was also found in the liver, meaning that more metabolically stable compounds are desirable for future development.

Environmental Friendliness as a Marketing Strategy

People in Sweden have become more aware and interested in environmental issues. This has led to an increased demand for environmental friendly food products. Previously the price often determined if a customer bought the product or not. During the recent years internet has become a more widely used media than ever before. This has led to that inter-net is one of the important tools to gather information and knowledge, among other things environmental issues. With such aspects in mind the authors found it interesting to investigate if companies could use internet as a marketing tool for environmental friendly products. Therefore the purpose of this thesis is to: investigate if and how companies display environmental friendliness in their marketing strategy and how it is presented on their company webpage’s. In order to fulfil the purpose five general dealer’s home and webpage’s were examined. As a supplementing research one store for each of the five general dealers were visited and ex-amined. The used theoretical framework is focused on value and green marketing. How to create a specific value is often the core when formulating and creating a strategy for a company. Green marketing is such marketing that include environmental concerns in the marketing offers. The sales of ecological food has increased and according to SCB (2006) represented 6 Billions in revenue between 2004 and 2005. The literature indicates that green marketing have to change and become more like conventional marketing, in order to reach the larger consumer segments. As a supplement to this theory, a section with home and webpage de-sign and chromatics are added. The analysis and conclusion showed that there were some differences in how the re-searched companies used green marketing. Two of the companies used their webpage’s and displayed environmental awareness, while two had a clear focus on price. One company had a health focus. It was also found that there are differences between the messages and information on the home and webpage’s, compared to the findings in the stores. What also was found was that green marketing not only should target the already green consumers, but instead aim at the larger consumer segment. To do this green marketing should become more like conventional marketing and not only focus on the product, but also other value factors. The findings also suggest that the companies could use their home and webpage’s to a higher extent than they do today.

Diskurser om det "äldsta yrket" En komparativ studie om attityden till, och synen på prostitution i Sverige och Nederländerna.

Uppsatsen har till syfte att titta på hur verksamma inom stödorganisationer ser på sina klienter, samt vad de har för attityder till prostitution. Genom en diskursanalys lyfts diskursen om prostitution utifrån det nederländska och svenska samhället fram. Frågeställningar i uppsatsen är: Vilket syfte har organisationerna i arbetet med prostitution? Vilken syn på prostitution har de anställda på stödorganisationerna? Vad har organisationerna att erbjuda och hur arbetar de? Går det att finna skillnader eller likheter mellan synen på prostitution i de olika länderna? Uppsatsen har en beskrivande och tolkande design med ett kvalitativt diskursanalytiskt förhållningssätt. Frågeställningar rörande de anställdas värdegrund och förhållningssätt till sina klienter utgjorde grunden i uppsatsen. Intervjuer med sammanlagt tolv personer utfördes, sex stycken i Sverige och sex stycken i Nederländerna varav samtliga arbetar inom stödorganisationer för prostituerade. Textmaterial från organisationerna samlades även in vid intervjuerna. De två ländernas intervjuer och textmaterial bearbetades separat och jämfördes sedan. Resultatet visade på en rad olika subjektpositioner som karaktäriserar diskursen om prostitution. Den svenska diskursen präglas av radikalfeministiska värderingar, och den nederländska diskursen av liberalfeministiska värderingar. Mikroperspektivet, eller individperspektivet finns hos den svenska diskursen om prostitution, då det fokuseras mycket på den prostituerade individen. I det nederländska samhället ligger fokuset på makro eller samhällsnivå, där prostitutionsfrågan är en samhällsfråga och har mindre att göra med individen som prostituerar sig. / The aim of the essay is to look at how support organizations are working and dealing with prostitution clients and the attitude the employees of these organizations have toward their clients. Discourse analysis is used in the interpretation of the data collected. The questions of the study were: What is the purpose of the organizations, which work with prostitution? What are the views of the employees of these organizations towards prostitution? What do the organizations have to offer their clients? Are there differences or similarities in the Swedish and Dutch discourse on prostitution? Interviews with a total of twelve respondents were performed, six in Sweden and six in the. Text material from the organizations was also collected and used in the study. The interview results and the text material collected from the two countries were analyzed separately and then compared. The result showed different subject positions. The Swedish discourse showed influences from radical feministic values, and the Dutch discourse from liberal feministic values. A micro- or individual understanding of the question is reflected in the Swedish discourse of prostitution. In the Dutch material, the focus is on the macro- or the society understanding, where prostitution is a question of society and has less to do with the prostitutes as individuals.

Canadian consumers' functional food choices : labelling and reference-dependent effects

The growing interest among consumers in the link between diet and health makes functional food one of the fastest growing sectors in the global food industry, especially functional dairy products. Understanding consumer choices with respect to functional food is an important and relatively new research area. Given the credence nature of functional food attributes, labelling plays a key role in allowing consumers to make informed choices about foods with enhanced health attributes. In 2007, Canada launched a review of the regulatory system for health claims on functional foods, which included rules concerning the approval, labelling and verification of health claims. In 2010 two new health claims related to oat products and plant sterols were approved by Health Canada. An analysis of how consumers respond to health claim information is therefore timely. This thesis focuses on examining the effects of different types of labelling and verification of health claims on consumers stated preferences for a specific functional food product, Omega-3 milk. The analysis incorporates reference-dependent effects. This study improves the knowledge of Canadian consumer understanding of health claims and the impact of health claims on consumer choice. This research is one of the first studies to simultaneously examine the effects of different types of health claims (e.g. function claims, risk reduction claims and disease prevention claims) and other ways of signalling or implying health benefits (e.g. symbols) on Canadian consumers' functional food choices. This study contributes to the knowledge in this domain by providing a comparative analysis of different types of labelling strategies. The extant knowledge of labelling effects in the formats of risk reduction claims, disease prevention claims and symbols or imagery on functional foods is limited. One of the primary contributions of this study is addressing this gap in the literature. The theoretical framework of this thesis is based on random utility theory. A stated preference choice experiment is designed to examine consumers' response to Omega-3 milk under different labelling scenarios. Using data from an online survey of 740 Canadians conducted in summer 2009, discrete choice models, including Conditional Logit, Random Parameter Logit and Latent Class models, and Willingness-To-Pay (WTP) values are estimated. The results suggest that full labelling (function claims, risk reduction claims and disease prevention claims) is preferred over partial labelling (e.g. the use of a heart symbol to imply a health claim), but primarily for risk reduction claims. There is no significant difference between a function claim, such as "good for your heart" and partial labelling in the form of a red heart symbol. The results also suggest that consumers on average respond positively to verification of health claims by government and the third party agencies, however, the Latent Class models reveal considerable heterogeneity in consumer attitudes toward the source of verification. The influences of key-socio-demographic (e.g. income, education and health status) and attitudinal factors (e.g. attitude, trust and knowledge) provide further insights into consumer responses in the choice experiment to identify different consumer segments. Moreover, the results reveal reference-dependent effects where perceived losses of ingredient or price attributes have a greater influence on consumer choice than perceived gains. In terms of industry and public policy implications, this study suggests that food manufacturers in Canada would benefit from the ability to make more precise health claims. The implications derived from the Latent Class Models could help the Canadian functional food industry to identify target consumer segments with different characteristics for the purpose of developing marketing strategies. Furthermore, the results of this study suggest that Canadian consumers are receptive to both full labelling and partial labelling. It indicates that public policy makers need to pay attention to effectively regulating health claims for functional foods so as to balance the need for credible health claims to facilitate the development of the functional food sector with the imperative of protecting consumers from misleading health claims. Public policy makers should also be aware that the verification of health claims plays an important role in reducing consumers' uncertainty and making health claims more credible.
