Characterization of plasmids among the three species of Gluconobacter

The genus Gluconobacter consists of acetic acid bacteria which have the ability to generate acidic products from their substrates, particularly acetic acid from ethanol. For this reason, the gluconobacters live in acidic, sugary environments such as flowers, honey bees, fruits, cider, vinegar, wine and beer. The gluconobacters carry out a strictly respiratory type of metabolism using only oxygen as a terminal electron acceptor. They do not completely oxidize a substrate to carbon dioxide. Instead, they partially oxidize the substrate using membrane-bound dehydrogenases and excrete the product into the surrounding growth medium. It is these limited oxidations that make the gtuconobacters industrially useful. Although much is known about the physiology of the limited oxidations in the gluconobacters, little is known of their genetics, particularly, their plasmids. The overall purpose of this dissertation was to determine if Gluconobacter plasmids correlate with oxidative capability and/or antibiotic resistance. To achieve this goal, I first needed a way to screen strains of Gluconobacter for their ability to oxidize many different substrates. 'developed an assay that used an unusual artificial electron acceptor, tetranitroblue tetrazolium (TNBT) and then tested the ability of six strains to oxidize 13 chemical compounds. Although most strains were able to oxidize the 13 compounds tested, they accomplished this with varying extents of oxidation. Dietary Acculturation, Physical Activity and Body Image in Limited-Resource Latino Women in Northern Virginia

The purpose of this study was to collect exploratory data on dietary acculturation, physical activity, and body image in a limited resource Latina population in northern Virginia. Acculturation may be described as a process in which members of one cultural group adopt the beliefs and behaviors of another group. Acculturation has been associated with a variety changes in terms of diet, physical activity and body image. Most dietary acculturation research in the U.S. has focused on Mexican Americans, Cuban Americans, and Puerto Ricans; however this study was composed of mostly Central and South Americans. Eighty-five subjects were recruited from the Arlington County Expanded Food and Nutrition Education Program, Arlington County Women, Infants, and Children program, and the Fairfax County Parklawn Family Center. Demographic information, acculturation, fruit and vegetable intake, saturated fat avoidance, physical activity, and body image were assessed with written survey instruments. The results indicated that this Latina population with limited resources in northern Virginia was mainly from Bolivia and El Salvador, and was not highly acculturated. Almost half of the participants were overweight or obese. About 50% of the population met the 5-A-Day goal for fruit and vegetable intake and almost 95% of participants exhibited at least one form of saturated fat avoidance. Eighty-five percent of the population reported participating in 30 minutes or more leisure-time physical activity less than 3 times each week, though a similar percentage reported that physical activity was important for health. Sixty percent of respondents were on a weight loss diet. While there was a significant relationship between the number of servings of fruit consumed and acculturation, there was no significant relationship between acculturation and any other dietary, physical activity or body image factor measured. The results of this study provide a baseline for further research in the limited resource Latina population in northern Virginia. / Master of Science

Making Memories in 140 Characters or Less: Testing the Effectiveness of CSR Messages Disseminated by Major League Baseball Teams via Twitter on Recognition and Recall

Corporate Social Responsibility (CSR) in sport is an emerging area of interest among scholars, sport managers, sponsors, fans, and policy-makers (Breitbarth, Walzel, Anagostopoulos, and van Eekeran, 2015). Despite this increased scholarly attention, a gap in literature exists regarding to the effectiveness of the messages disseminated. This study investigated how sports organizations communicate CSR on Twitter by examining the effectiveness of CSR messages disseminated via Twitter by professional baseball teams. Using priming theory and Lang's (2000) Limited Capacity Model for Mediated Message Processing, this study tested the effects of priming and message sequencing on a persons' ability to recognize and recall these CSR messages. While no statistically significant relationships were found, observed results led to strong arguments about the effects of both priming and message sequencing on the effectiveness of CSR messages disseminated by Major League Baseball (MLB) teams in terms of recognition and recall. Additionally, results suggested that traditional media effects paradigms might not be transferable to social media. / Master of Arts

Error Analysis of Angle and Range Estimation in a Range Limited Antenna System Using Music and Modified Root Pisarenko Algorithms

The Range Limited Antenna (RLA) has the ability to track and pin-point RF devices operating within a certain accepted range. Using two antenna arrays, each made up of several elements, direction-finding algorithms such as MUSIC and Modified Root Pisarenko (MRP) are used to estimate the angles of arrival of incoming signals. Using the estimated angles of arrival, a range estimation algorithm is used to estimate the distance of the emitter from the antenna array. The aim of this thesis is to check the sensitivity of the range estimation and to form a statistical analysis for such conditions. Conditions of error are introduced into the system and several extensive Monte Carlo simulations are run for both MUSIC and MRP. Upon simulating physical errors in the antenna set-up, it is realized that the range estimation algorithm is sensitive to the slightest of phase shift differences, cable loss differences, mixer mismatches, and antenna mismatches. / Master of Science

Summary: The reason for choosing the topic of my thesis was primarily the popularity of a Limited Liability Company as a form of a corporation suitable especially for small and medium-sized undertakings and practical usability of the knowledge about this issue. During their life businessmen and other legal persons encounter the form of Limited Liability Company almost daily and they participate very often in its business with a prospect of profit. This is why the knowledge about the rights and duties as of a member of the Limited Liability Company is very important and helpful. The main object of this thesis is to focus on a broader description of members' legal position concerning their membership in the Limited Liability Company. Even though each right and duty is described in a separate chapter they need to be understood in their mutual coherence as a complex. My research begins with a general characteristic of a business corporation as a product of commercial law and a general description of the nature and specifics of the Limited Liability Company within the Czech legal order. Summary: Limited Liability Company is so popular form of a corporation suitable. I can say, this type of company is the most popular form of business organization. To have some knowledge about this issue could be very helpful and that is a reason of choosing this topic of my thesis. The regulation of duties of members of a limited liability company in the Commercial Code is only a little part of the whole limited liability company, but very I think, it is very important part. The regulation of duties members of a limited liability company is complex with their right and often should be view together. But on the other hand, sometimes the text of Commercial Code says nothing about the particular position of the member in the company and must be used theory. The most important of my thesis is to focus on the main duties of members of a limited liability company and the position of them. Every each duty of members I described in a separate chapter. I tried to explain every one duty in relation with other duties to explain mutual interconnection as a complex unit. The legal regulation of the Limited Liability Company should be often so much formal and complicated. Competencies of General Meeting of limited liability company The purpose of my thesis is to analyse competencies of a general meeting of a limited liability company, show different theoretical opinions and present my own ideas. I have chosen this topic because the limited liability company is the most common type of business entity in the Czech Republic and so I find this topic useful. The thesis is composed of Introduction, four chapters and Conclusion. Introduction defines aims and methods of this thesis. First general chapter is followed by three chapters, each of which dealing with different aspects of general meeting's competence. Chapter One is introductory and defines basic terminology used in the thesis. This chapter is subdivided into five parts. Personnel in medical status "limbo" decrease readiness and cost the Navy millions of personnel dollars each year. Estudou-se a influência do fosfato na multiplicação de Saccharomyces cerevisiae em uma fermentação contínua em mini-fermentador. Determinou-se a massa seca, a concentração dos substratos, a velocidade específica de consumo dos substratos, a velocidade específica de formação de gás carbônico, velocidade específica de consumo de oxigênio e o quociente respiratório bem como, o fósforo intracelular em diversas vazões específicas de alimentação, em cultivo contínuo de levedura de panificação, em condições de substratos limitantes. Controlando-se a quantidade de fosfato no meio de alimentação de tal modo que o fosfato residual no meio de fermentação mal pudesse ser detectado, o valor da vazão específica de alimentação crítica era aparentemente aumentado de 0,23 h-1 para 0,32 h-1. Isto sugere uma possível influência do fosfato nas funções anaeróbicas e aeróbicas da levedura de panificação. / The influence of phosphate in a continuous culture was studied using mini-fermentor on the Saccharomyces cerevisiae multiplication. Dry matter, substrate concentration, specific substrate comsumption, specific carbon dioxide release, specific oxygen uptake rates and respiration quotient , as well as phosphorous content of the cells were measured in dependence on the dilutionrate. In continuous culture glicose-limited, of baker\'s yeast if the supply of phosphorous were restricted to a extent that residual phosphate in the medium could hardly be observed, the value of critical dilution rate was apparently enhanced from 0,23 h-1 to 0,32 h-1. This observation suggests a possible mediation by phosphate between anaerobic and aerobic functions of the baker\'s yeast.
