Effect of fatty acids on hyphal growth in the pathogenic yeast Candida albicans

Shareck, Julie 09 1900 (has links)
The yeast Candida albicans is an inhabitant of the oral cavity, the gastrointestinal and genitourinary tracts of humans. Generally encountered as a commensal, it is also an opportunistic pathogen that causes a spectrum of infections, ranging from superficial mycoses (thrush, vulvovaginitis) to severe and life-threatening systemic infections. A striking feature of C. albicans is its ability to grow in different morphological forms, including budding yeasts, pseudohyphae, and hyphae. Environmental cues that mimic host conditions (elevated temperature, neutral or alkaline pH, and serum) induce the yeast-to-hypha transition. Morphogenesis is considered to be an attribute of pathogenesis, as mutants locked as yeasts or filamentous forms are avirulent. Given that the yeast-to-hypha transition is a virulence factor, it may also constitute a target for the development of antifungal drugs. Indeed, evidence has shown that impairing morphogenesis is a means to treat systemic candidiasis. Concurrently, a number of molecules have been reported to modulate morphogenesis in C. albicans. For instance, several fatty acids, including conjugated linoleic acid (CLA), inhibited the yeast-to-hypha transition. By interfering with an important attribute of C. albicans pathogenesis, CLA may harbor antifungal properties. However, before assessing its therapeutic potential in a clinical context, it is mandatory to address CLA’s mode of action. The present study aims to further characterize the hypha-inhibiting properties of fatty acids and CLA and to elucidate the mechanism by which these molecules inhibit the yeast-to-hypha transition in C. albicans. Gene expression analyses were performed to gain insight into the transcriptional response of cells to CLA on a genome-wide scale and to probe the fatty acid’s mode of action. CLA downregulated the expression of hypha-specific genes and blocked the induction of genes encoding regulators of hyphal growth, including that of RAS1, which encodes the small GTPase Ras1p. A membrane-associated signaling protein, Ras1p plays a major role in morphogenesis. Quantitative PCR analyses showed that CLA prevented the increase in RAS1 mRNA levels which occurred at the onset of the yeast-to-hypha transition. Unexpectedly, CLA reduced the steady-state levels of Ras1p. Additionally, CLA caused the delocalization of GFP-Ras1p from the plasma membrane. These findings indicate that CLA treatment results in suboptimal Ras1p cellular concentrations and localization, which impedes Ras1p signaling and inhibits the yeast-to-hypha transition. CLA may indirectly affect Ras1p localization by altering the structure of the plasma membrane. These studies have provided the mechanism underlying CLA’s hypha-inhibiting properties and may serve as the rationale to examine CLA’s therapeutic potential in the context of a Candida infection. There is a general lack of clinical evidence demonstrating that impairing morphogenesis is a sound approach to treat candidiasis. To remedy this situation, the therapeutic potential of molecules that modulate morphogenesis, such as CLA, should be clinically assessed.

Lipotoxicidade na leptospirose humana: aspectos prognósticos e potencial efeito benéfico da administração de albumina / Lipotoxicity in human leptospirosis: prognostic aspects and potental beneficial effect of albumin administration

Caroline de Azevedo Martins 27 April 2011 (has links)

In order to evaluate the proteeting aetion of some substanceson ram spermatozoa, semen samples of 25 rams were colleeted and submitted to cryopreservation after treatment with cholesterol, desmosterol, oleic-linoleic acid or Q-Iactalbumin. Before the processing and after the procedures of cooling, freezing-thawing and incubation, semen was evaluated as to kínetcs by subjective and computerized analyses (CASA), morphology, simultaneously to plasma membrane integrity (PMI), acrosome integrity (ACI) and mitochondrial membrane potential (MMP) by isothiocyanate-conjugated Pisum sativum (FITCPSA), propidium iodide (PI) and JC-1 combination and as to sperm capacitation and acrosome reaction determined by chlortetracycline (CTC) assay. The influence resulted from the additives in a few parameters was not enough to suggest the superiority of any treatments. The cryopreservation process, especially the freezing-thawing, promotes damages to kinetics, PMI, ACI, MMP and induees accelerated eapacitation and acrosome reaction in spermatozoa. Differenees among groups of rams were observed regarding capacitation status in fresh semen as well as the sensibility of their semen to cryoinjury and cryocapacitation. Higher and more accelerated capacitation-like ehanges were observed in groups of rams presenting lower plasmatic membrane cryoresistance after thawing. The segregation of rams in different levels of PMI influenced the behavior of several other parameters. It was concluded that cholesterol, desmosterol, oleic-Iinoleic acid and Q-Iactalbumin do not protect the ram semen against the cryoinjuries as well as freezing-thawing is more harmful than cooling. Important differenees among groups of individuais were noticed as for the spermatozoa resistance to the damages provoked by cryopreservation such as the eryocapacitation susceptibility.

Avaliação do desempenho em confinamento, do metabolismo ruminal e do perfil de ácidos graxos da carne em novilhos Nelore, utilizando milho com alto teor de óleo nas dietas de terminação / Evaluation of feedlot performance, ruminal metabolism and fatty acids profile of Nellore cattle using high oil corn on finishing diets

José Luiz Domingues 07 December 2006 (has links)
Beef cattle researchers, over than animal performance and economics are looking for nutritional characteristics of the products from ruminants. Inclusion of high oil corn (HOC) on diets increases weight gain and carcass quality compared to common corn grain. This study looks for data from animal performance on diets using high oil corn, evaluating live weight gain, carcass composition, diets digestibility, dry matter degradability, effects on ciliated protozoa number and quality, muscle quality characteristics, meat composition and fatty acids profile on intramuscular fat. Where used 48 Nellore steers, with initial average live weight of 435 kg in six treatments, using two corn grain varieties (common and high oil corn) and three levels of corn grain in concentrate diets (25%, 40% and 55%). Animals were in feedlot pens for 84 days, after an adaptation period to diets and installations. The Longissimus dorsi was used for evaluating meat characteristics and fatty acids profile on intramuscular fat. There was no effect of diets on daily gain, intake, conversion, or carcass parameters. Meat quality was not also affected by treatments. Diets with high oil corn altered protozoa population and ruminal degradation of NDF. The total biohidrogenated fatty acids in intramuscular fat were increased by treatment with high oil corn and also rumenic acid. Rumenic acid was the major conjugated linoleic acid (CLA) detected in this tissue, with concentrations increasing from 0,43% to 0,49%, representing an average increase of 12,7% caused for HOC diets.

Estudo das mudanças quali e quantitativas da participação de ácidos graxos no sebo de pacientes com acne inflamatória, graus II ou III, em uso sistêmico de limeciclina e/ou suplementação oral à base de ácidos linoleico, gamalinolênico / Study of qualitative and quantitative changes of fatty acids in the sebum of patients with inflammatory acne, grade II or III, under systemic use of lymecycline and/or oral supplementation based on linoleic, gamma-linolenic, and oleic acids using gas chromatography

Adilson da Costa 14 February 2012 (has links)
Objectives: To study the possible quantitative and qualitative changes of fatty acids found in the sebum of patients with acne vulgaris grade II or III, through the daily use of lymecycline and/or oral supplementation based on linoleic, gamma-linolenic, and oleic acids. Methods: Fortyfive male research subjects, ages 12 to 40, presenting acne vulgaris grade II or III, were submitted to a 90-day use of: Group 1, 300mg of lymecycline per day; Group 2, 540mg of gamma-linolenic acid, 1,200mg of linoleic acid, and e 510mg of oleic acid per day; Group 3: Groups A+B. Every 30 days, a sample of the sebum of their forehead was collected for chromatographic analysis of the fatty acids contained therein. Results: Forty research subjects (88.9%) concluded the study. They ingested all dosages of the products, without presenting side effects in any of the Groups. The number of comedones, pustules, and cysts reduced with time (p<0.001, for all), in all the 3 Groups. With respect to the total number of lesions, Group B´s patients presented a larger quantity of lesions, as compared to those of Group A (p=0.033) and Group C (p=0.030). Seven fatty acids presented pattern changes during the study. In general, the changes that became more evident were: 1) the squalene concentration in the sebum did not diminish in any Group under study; 2) C12:0, C14:0, and C16:1 had their respective concentrations increased in the 3 Groups over the time of the treatments; 3) there is an indication for increasing of C18:1n9t+C18:1n9c when both of therapies are associated; 4) lymecycline and/or the ingestion of linoleic acid did not increase the participation of linoleic acid in the sebum; and 5) gama-linolenic acid had its concentration increased during the first 60-day and diminished during the last 30-day therapies period using lymecycline and/or with its own oral supplementation. Conclusions: The daily administration of lymecycline and/or gamma-linolenic, linoleic, and oleic acids may alter the behavior of some fatty acids present in the sebum of patients with acne vulgaris grade II or III. Gamma-linolenic, linoleic, and oleic acids, under systemic use, are not considered therapeutic agents in the treatment of acne; more studies need to be carried out in order to infer the existence of a possible clinical benefit of their isolated use and/or association in the treatment of this dermatosis

Haubold, Susanne 23 August 2021 (has links)
