Anaemia, which is caused by iron deficiency, is a global nutritional disorder of utmost concern. It has been assumed that meat, which contains haem as well as non-haem iron, enhances non-haem iron absorption due to the presence of the “meat factor”. In the experiment reported here, 24 semi-anaemic, 3-week-old piglets were utilised as human nutrition models to assess the effects of dietary lactoferrin, meat and LSF (Low Molecular Weight Sarcoplasmic Fraction) on iron bioavailability during a 4-week feeding period. The parameters that were used as measurements of iron bioavailability were changes in haematological indices, haemoglobin iron repletion efficiency, intestinal morphology and mineral balances. Non-significant (p > 0.05) dietary effects were observed for growth performance and for all the haematological and some histological parameters (small intestine villi height, crypt depth and mucosal thickness). Haemoglobin iron repletion efficiency was highest for the control group and was not significantly different between the other 3 diets. The superiority of the control diet in this respect was not consistent with previous trials and can not be explained. Results suggested that increased retention of calcium, magnesium, phosphorous and manganese tended to inhibit iron absorption. However, the LSF and meat diets significantly (p = 0.003) increased the number of goblet cells/100 µm suggesting that mucin secretion was favoured by these two diets. Additionally, all immunological parameters were significantly (p < 0.05) improved by the LSF diet. As such, the LSF diet can be a potential immunobooster feed ingredient for weanling piglets. Overall, the level of LSF in the LSF diet was insufficient to exert a desirable enhancement of iron bioavailability and betterment of 3 iron status of the semi-anaemic piglets relative to the control group. However, the diet containing LSF was as effective as the meat diet with respect to these characteristics.

Liquid Organic Electrolytes: Blends of Low Molecular Weight Methoxyoligooxyethylene (MPEGs)/LiTFSI Salt

Blends containing methoxyoligooxyethyleneglycol (MPEGs) (MW 350 and 550) and bis(trifluoromethane)sulfonimide lithium (LiTFSI) salt were prepared by solution blending process using tetrahydrofuran (THF) as a solvent. The ionic conductivity of the blends of different compositions were determined at four temperatures i.e. 25°C, 40°C, 60°C and 70°C. A maximum ionic conductivity value of 3.9x10-3 S cm-1 at 25°C was obtained for the blends containing MPEG-350 at an ethylene oxide to lithium salt ratio of 1:10. The ionic conductivity increases with increasing temperature and shows that the ion transport is aided by the segmental motion of the MPEG chains. 7Li NMR spectroscopy was used to study the nature and dynamics of the salt clusters in the blends

O copolímero SBS é o aditivo mais empregado mundialmente na modificação de ligantes asfálticos, por melhorar diversas de suas características. Um dos principais problemas deste tipo de formulação é a separação de fases, que pode ser minimizada com a incorporação de aditivos. Um novo polietileno de alta densidade e baixo peso molecular oxidado (com denominação comercial TITAN 9686) foi utilizado nesta pesquisa, com o objetivo de testar a hipótese de que a adição de proporções adequadas deste aditivo possa melhorar as características reológicas do asfalto+SBS. O experimento laboratorial foi delineado com base na técnica de experimentos com misturas, englobando um total de nove formulações compostas com asfalto, SBS e TITAN. O teor de SBS variou entre 0 e 5% em peso e o de polietileno variou entre 0 e 2% em peso. Foi utilizado óleo aromático no teor de 4% em peso para todas as nove misturas. As misturas foram analisadas em três condições de envelhecimento: virgem, curto prazo (na estufa RTFO) e longo prazo (na estufa PAV). Foram realizados os seguintes ensaios reológicos: (i) de cisalhamento em regime oscilatório em diferentes condições de temperatura e frequência (para medida do módulo complexo e do ângulo de fase, usados na determinação do grau de desempenho, no cálculo dos parâmetros G*/sen(&#948;) e G*.sen(&#948;) e na construção de curvas-mestre), (ii) de viscosidade Brookfield, (iii) de fluência e recuperação sob múltiplas tensões (para medida do percentual de recuperação e da compliância não-recuperável), (iv) de varredura de amplitude linear (para cálculo do parâmetro a f e para ajuste do modelo de fadiga) e (v) de estabilidade à estocagem. Acerca das características escolhidas para mensurar a melhora ou piora das propriedades das misturas asfalto+SBS com a adição de TITAN, podem ser feitas as seguintes observações: (i) a estabilidade à estocagem é melhorada com a adição de TITAN, em particular no teores entre 1 e 2%; (ii) a sensibilidade ao envelhecimento tanto a curto quanto a longo prazos diminui com a adição de TITAN; (iii) a adição de TITAN aumenta a resistência à deformação permanente, à luz dos parâmetros Jnr e R e aumenta a sensibilidade dos ligantes asfálticos modificados com SBS a mudanças bruscas nos níveis de tensão aplicados; e (iv) a adição de TITAN contribui com o aumento da resistência à fadiga, à luz dos parâmetros Nf , a baixos níveis de deformação, e af , e a prejudica reduzindo o parâmetro Nf a altos níveis de deformação e aumentando a temperatura crítica de fadiga ou o parâmetro G*.sen(&#948;). Tendo por base estas evidências, é possível concluir que a adição de TITAN, em linhas gerais, melhorou a estabilidade dos ligantes asfálticos modificados com até 5,0% de SBS. Com base nas análises realizadas, é possível afirmar que teores da ordem de 1 a 2% de TITAN, para teores de SBS entre 3,0 e 5,0%, são recomendáveis, à luz das propriedades, índices e parâmetros analisados neste trabalho. / The SBS copolymer is the most widely used additive in asphalt binder modification, since it enhances many of the properties of the original material. A critical problem that arises from this type of modification is phase separation, which can be minimized by adding other modifiers to the formulation. A new oxidized, high-density polyethylene with low molecular weight (commercial designation of \"TITAN 9686\") was used in the study with the purpose of verifying the hypothesis that the addition of suitable contents of this modifier can improve the rheological properties of the SBS-modified binder. The laboratory matrix of formulations was based on the experiments with mixtures, and nine types of materials were prepared with asphalt binder, SBS and TITAN. The SBS contents ranged from 0 to 5% by weight, whereas the polyethylene contents ranged from 0 to 2% by weight. These formulations also contained 4% of aromatic oil by weight. Three aging conditions were selected: unaged, short-term aged in the rolling thin-film oven (RTFO) and long-term aged in the pressurized aging vessel (PAV). The following tests were carried out: (i) dynamic oscillatory shear at different temperatures and frequencies of loading for the determination of the complex modulus, the phase angles and the master curves, as well as the performance grades and the numerical values of the parameters G*/sin(&#948;) and G*.sen(&#948;); (ii) rotational viscosity; (iii) creep and recovery at multiple stress levels for the determination and the percent recovery and the nonrecoverable compliance; (iv) linear amplitude sweep, in order to calculate the parameter a f and fit the fatigue model to the data; and (v) storage stability. With respect to the characteristics used in the evaluation of the properties of the AC+SBS mixtures after the addition of TITAN, the following observations can be made: (i) storage stability is improved when TITAN is added to the formulation, especially at contents ranging from 1 to 2%; (ii) the sensitivity to the short-term and long-term aging processes decreased with the incorporation of TITAN; (iii) the changes in the R and the Jnr values indicate that the addition of TITAN makes the asphalt binder more resistant to rutting, and also increases the sensitivity of the SBS-modified binders to sudden increases in the stress level; and (iv) the presence of TITAN in the material leads to an increase in the fatigue resistance as based on the parameters Nf and af at low strain levels and the increases in the critical fatigue temperature and the parameter G*.sen(&#948;). With reference to these evidences, it is possible to conclude that the addition of TITAN generally improves the storage stability of the SBS-modified binders with no more than 5% of copolymer by weight. Based on the results of the present study, it is possible to say that percentages between 1 and 2% of TITAN by weight are recommended for SBS contents between 3 and 5% by weight.

Le microenvironnement des cellules tumorales présente plusieurs particularités comme l'hypoxie, l'acidification du milieu extracellulaire et l'hypersécrétion d'enzymes hydrolytiques. Ces hydrolases, comme la cathepsine D et l'héparanase, interviennent dans les étapes de la progression tumorale et notamment l'angiogenèse. Cette thèse s'intègre dans un projet dont la finalité est de concevoir un nano-objet moléculaire enzymo-sensible qui réagirait d'une manière spécifique aux enzymes hypersécrétées dans le microenvironnement tumoral pour assurer de façon simultanée, une fonction de détection et de ciblage des tumeurs. La première partie de nos travaux a été consacrée à la conception et à la validation d'un lien peptidique intégrable dans l'objet moléculaire, sensible aux formes de la cathepsine D actives du microenvironnement tumoral de cancers mammaires. Cet objectif a été réalisé suite à l'étude cinétique de l'hydrolyse de 5 peptides par la cathepsine D mature et la pro-cathepsine D dans les conditions de pH du microenvironnement tumoral. Nous avons également étudié l'effet de l'hypoxie et de l'acidification du milieu extracellulaire sur la sécrétion des formes actives de la cathepsine D par la lignée tumorale de cancer mammaire MCF-7. Dans une deuxième partie, nous avons travaillé sur l'élaboration d'héparines de bas poids moléculaire pouvant assurer la fonction thérapeutique de l'objet moléculaire grâce à leur activité anti-angiogénique. Nous avons mis au point une méthode innovante pour la dépolymérisation de l'héparine qui consiste en une hydrolyse radicalaire par le péroxyde d'hydrogène assistée par les ultrasons. Cette technique permet la production d'oligosaccharides d'héparines caractérisées par des poids moléculaires et des degrés de sulfatation contrôlés. En fonction des conditions de dépolymérisation par cette technique, les héparines de bas poids moléculaires produites peuvent être utilisées comme anticoagulant ou anti-angiogénique. / Tumor microenvironment is characterized by several particularities such as hypoxia, extracellular media acidification and the hyper-secretion of hydrolytic enzymes. These hydrolases, such as cathepsin D and heparanase, are involved in many steps of tumor progression like angiogenesis. This thesis is a part of a project that aims to develop a "smart" molecular nano-object that specifically reacts to hyper-secreted enzymes in the tumor microenvironment for the simultaneous detection and targeting of tumor. The first part of our work concerned the design and the validation of a peptide that is sensitive to active forms of cathepsin D which is a protease, unregulated in many tumors microenvironment such as breast cancers. This objective has been achieved following the kinetic study of the hydrolysis of 5 peptides by mature cathepsin D and procathepsin D in the pH conditions of the tumor microenvironment. On the other hand, we studied the effect of hypoxia and the acidification of the extracellular medium on the secretion of active forms of cathepsin D by the breast cancer cell line MCF-7. In a second part, we worked on the development of low molecular weight heparins that may provide therapeutic function of the molecular object through their anti-angiogenic activity. We have developed an innovative method for the depolymerization of heparin that consists on a radical hydrogen peroxide hydrolysis assisted by ultrasound. This technique allows the production of heparins oligosaccharides characterized by controlled molecular weight and degree of sulfatation. Depending on the depolymerization conditions by this technique, the produced low molecular weight heparins can be used as an anticoagulant or anti-angiogenic.

The behavior of the system consisted by mixture of two different molecular weight hyaluronates and surfactant was investigated. Mixtures were 17 kDa hyaluronate with 1,46 MDa, 73 kDa with 1,46 MDa, 300 kDa with 1,46 MDa, 806 kDa with 1,46 MDa and 1800 kDa with 1,46 MDa. These compounds were always mixed in the weight ratios 70:30, 50:50 and 30:70. As the surfactant cetrimonium bromide and TWEEN 20 were used. Interactions were studied in aqueous solution with different ionic strength. Sudan red was used as hydrophobic dye. In all experimental series with cetrimonium bromide was observed phenomenon of discontinuous separated phases, described as pearls. Samples containing pearls were tested on stability, were dried and rehydrated back, as were also heated in solution. Subsequently, the particle size was measured in the remaining sample after pearls were filtrated. Mixtures of hyaluronate were characterized by measuring the viscosity using rheology microrheology. It was found that these compounds are heterogeneous and each sample point is not the same viscosity.

The Role of Low-Molecular Weight Fungal Metabolites in Eutypa Dieback Grapevine Trunk Disease

Eutypa dieback, one of several grapevine trunk diseases (GTDs), is of serious concern to the grape industry globally. This disease is caused by the fungus Eutypa lata but it is often seen in consortia growth with Phaeoacremonium minimum and Phaeomoniella chlamydospora. It is vital to understand the mechanisms for how this disease functions to develop control measures to combat it. Brown rot fungi are able to use a complex of low molecular weight (LMW) metabolites to induce a Fenton reaction to deconstruct woody tissue. These metabolites are part of a chelator mediated Fenton (CMF) chemistry that produces reactive oxygen species that are capable of depolymerizing wood polymers. We propose that a mechanism similar to CMF chemistry may be occurring in grapevine trunk disease pathogens. This thesis investigates how LMW metabolites produced by the fungi contribute to the disease and decay progression in GTDs. Research on Mite control in the laboratory with abamectin was also investigated, as research in this area was required when mites infested our fungal cultures and suitable laboratory controls were not available. Research on the GTD fungi was initiated by first examining whether metabolites produced by the three fungi can function in a manner to promote reactions like the CMF system. We separated and identified specific metabolites that potentially could contribute to CMF chemistry. We found that all three GTD fungi were able to produced LMW metabolites that promoted CMF chemistry, and we hypothesized that this mechanism contributes to processes leading to tissue necrosis in grapevine trunk wood. To explore the development of effective control measures based on this newly discovered mechanism for pathogenesis, we also explored the use of antioxidant/chelator compounds, BHA and BHT, in the control of the consortia fungi. Biocontrol organisms, Bacillus subtilis and Trichoderma atroviride, that produce antioxidants were also tested as biocontrols against the fungi involved in Eutypa Dieback disease. We found that BHA was highly effective in inhibiting fungal growth for all three fungi at concentrations higher than 0.5mM, and both B. subtilis and T. atroviride proved to be effective biocontrol agents in inhibiting E. lata, P. minimum, and P. chlamydospora.

Nous avons mené une étude prospective randomisée dans le but de comparer l'effet de l'irrigation du cathéter de type PICC avec deux types d'anticoagulants: Héparine standard et Tinzaparine, une héparine de faible poids moléculaire. Notre étude s'adresse aux patients de la clinique externe d'oncologie de l'hôpital Maisonneuve-Rosemont. Entre début Mai 2005 et Mars 2008, nous avons recruté 131 patients dont 70 ont été randomisés. Parmi les 61 patients exclus, 23 n'ont pas rencontré les critères d'inclusion, 30 ont refusé de participer et 8 ne sont pas inclus pour d'autres raisons. Sur les 70, 36 sujets sont randomisés dans le groupe Héparine standard et 34 dans le groupe Tinzaparine. La population en intention de traiter comprend 65 sujets dont 32 dans le groupe Héparine standard et 33 dans le groupe Tinzaparine. Le médicament a été administré pendant un mombre maximal de 30 jours et les sujets ont été suivis pendant 90 jours. La thrombose veineuse associée au cathéter (TVAC) a été objectivée par une phlébographie ou une échographie-Doppler à la fin de la période de 30 jours suivant l'installation du cathéter. L'incidence de la TVAC sur 30 jours est de 14,39 par 1000 cathéter-jours (IC à 95%:[9,0;19,79]/1000 cathéter-jours ou 41,5% (27/65). L'incidence de la thrombose veineuse profonde (TVP) symptômatique du membre supérieur sur la période de suivi de 90 jours est de 0,41 par 1000 cathéter-jours (IC à 95%:[0,08;0,81]/1000 cathéter-jours ou 3% (2/65). Nous n'avons observé aucune différence entre les deux groupes par rapport à la fréquence de la TVAC ni de la TVP. Nous ne pouvons conclure à une différence dans l'efficacité de la Tinzaparine par rapport à l'Héparine standard dans la prévention de la TVAC. / Our prospective, randomized study aims at comparing the effectiveness of two types of heparin used for the instillation of peripherally inserted central catheter (PICC): Standard heparin and Tinzaparin, a low molecular weight heparin. Our population intent-to-treat included 65/70 patients, 32 of these received Standard heparin and 33 received Tinzaparin. The experimental intervention was administered for a maximum of 30 days and all the subjects were followed up for 90 days. The events were documented objectively with a venogram or Doppler ultrasonography by the end of the 30 days following the catheter installation. The incidence of catheter-related venous thrombosis (CRVT) during 30 days of instillation is 14,39/1000 catheter-days (CI 95%:[9,0;19,79]/1000 catheter-days or 41.5% (27/65). The interaction between three selected representatives of environmental pollutants - naphthalene, anthracene, and 2-aminoanthracene - and DNA was investigated using an electrochemical DNA biosensor based on a glassy carbon electrode (GCE) and low molecular weight DNA from salmon sperm (DNA/GCE). The interactions with DNA were monitored using square wave voltammetry (SWV) and electrochemical impedance spectroscopy (EIS). For naphthalene, there was no DNA damaging interaction observed. In the case of anthracene, the formation of an intercalation complex [DNA-anthracene] was observed. However, its formation does not cause DNA strand breaks. The formation of similar intercalation complex was observed for 2-aminoanthracene [DNA-2-aminoanthracene], where we suppose on the basis of the results obtained that the intercalation of 2-aminoanthracene into the DNA double helix induces a tension and subsequent formation of single-strand breaks, which cause that the fragments of DNA fall away from the electrode surface. The intercalative interaction of DNA with anthracene a 2-aminoanthracene was used in the development of electrochemical methods for determination of these compounds at the GCE and DNA/GCE.
