Among these, the viruses are a major barrier to the development and highly profitable production in viticulture. By in vitro techniques it is possible to produce plants and multiply matrices of high quality vine plant. Cryotherapy, based cryopreservation methods, it is a promising tool for obtaining high frequency regenerating plants free of viruses in a short time. The paper proposes the development of a protocol for efficient recovery viruses vine plants through cryotherapy methods. The experiments were conducted in Biotechnology EPAGRI Laboratory, Experimental Station of Lages. The selection of genotypes was through the presence of different viral species (GLRaV-1, GLRaV-3, GFKV; GFLV and GVA) by serological ELISA test. The study is divided into two complementary chapters with information that follows the chronological order. The first aims to assess the establishment and multiplication in vitro and ex vitro acclimatization of grape genotypes through nodal segments cultivated in five formulations of culture media without added growth regulators. In Chapter I, in addition to selecting the best saline formulation that will be used in Chapter II for the grape varieties, is apreserntada an updated review of the topics covered at work, with information available to the present. In the second chapter, cryotherapy different methods investigated for vitis were applied, with the objective of defining a standard protocol for the eradication of viruses in potential genotypes. In vitro propagation of the rootstock IAC 571-6 and cultivars Poloskei Muskotaly and Bordô through nodal segments provide high rates of regeneration and rooting, suggesting no need to use growth regulators that promote rooting or specific phase for rooting. Percentage of high survival were obtained in the acclimatization of IAC 571-6 rootstock and cvs. Poloskei Muskotaly and Bordô. Considering all variables, the formulation that provided the best growth and development of root and shoot better and in vitro multiplication of IAC 571-6 vine rootstock and cv. Poloskei Muskotaly was Roubelakis composition and for the cv. Bordô were to Roubelakis and Zlenko formulations. The results of cryotherapy experiments to cultivars in question reaffirm the specificity of genotypic responses to cryogenic procedures. Rates between 6% and 10% were achieved for regenerating rootstock IAC 571-6 through encapsulation methodologies, dehydration and vitrification, respectively. Propagules regenerating after freezing in liquid nitrogen are anomalous and after small seedling growth from corrosion of tissues and lack of evolution cresmimento. Oxidative damage observed in the investigated protocols may be unfeasible cryopreserved tissues, even if part of the exposed tissues are viable in apparent liquid nitrogen is low or no regeneration of tissues. Understanding the degree of sensitivity of materials to vitrificantes solutions is the first step towards optimization of vitrification protocols in this study dehydration for 15 minutes in ½ PVS2 solution followed by 30 minutes PVS2 showed the best results for the rootstock IAC 571-6 / Novos plantios de vinhedos esbarram na baixa produtividade e na má qualidade das uvas produzidas, devido à ação de fatores complexos que ainda não estão bem esclarecidos. Dentre esses, as viroses constituem um importante obstáculo para o desenvolvimento e a produção altamente rentável na viticultura. Por meio de técnicas in vitro é possível produzir e multiplicar plantas matrizes de videira de alta qualidade fitossanitária. A crioterapia, baseada em métodos de criopreservação, torna-se uma ferramenta promissora para obtenção de alta frequência de plantas regenerantes livres de viroses, em curto espaço de tempo. O trabalho propõe o desenvolvimento de um protocolo eficaz para recuperação de plantas de videira virosadas através de métodos de crioterapia. Os experimentos foram conduzidos no Laboratório de Biotecnologia da EPAGRI, Estação Experimental de Lages. A seleção de genótipos foi por meio da presença de diferentes espécies virais (GLRAV-1; GLRAV-3; GFKV; GFLV e GVA), através do teste sorológico ELISA. O estudo foi dividido em dois capítulos complementares, com informações que seguem a ordem cronológica. O capítulo I tem por objetivo avaliar o estabelecimento e multiplicação in vitro e a aclimatização ex vitro de genótipos de videira, por meio de segmentos nodais cultivados em cinco formulações de meios de cultura sem adição de reguladores de crescimento. Esse capítulo, além da seleção da melhor formulação salina que será utilizada no capítulo II para as cultivares de videira, é apresentada uma revisão atualizada dos temas abordados no trabalho, com informações disponíveis até o presente. No segundo capítulo, diferentes métodos de crioterapia investigados para Vitis foram aplicados, com o objetivo de definir um protocolo padrão para a erradicação de viroses em genótipos potenciais. A propagação in vitro do porta-enxerto IAC 571-6 e das cultivares Poloskei Muskotaly e Bordô por meio de segmentos nodais proporcionam elevadas taxas de regeneração e enraizamento, sugerindo que não há necessidade de utilização de reguladores de crescimento que promovam o enraizamento ou de fase específica para o enraizamento. Elevados percentuais de sobrevivências foram obtidas na aclimatização do porta-enxerto IAC 571-6 e das cvs. Poloskei Muskotaly e Bordô. Considerando todas as variáveis analisadas, a formulação que proporcionou o melhor crescimento e desenvolvimento da parte aérea e radicular e melhor multiplicação in vitro do porta-enxerto de videira IAC 571-6 e da cv. copa Poloskei Muskotaly foi a composição Roubelakis e, para a cv. Bordô foram às formulações Roubelakis e Zlenko. Os resultados dos experimentos de crioterapia para as cultivares em questão reafirmam a especificidade das respostas dos genótipos aos procedimentos criogênicos. Taxas entre 6 % a 10 % de regenerantes foram atingidas para o porta-enxerto IAC 571-6 por meio das metodologias de encapsulamento-desidratação e vitrificação, respectivamente. Propágulos regenerantes após o congelamento em nitrogênio líquido são anômalos e após pequeno crescimento da plântula apresentam oxidação dos tecidos e não possuem evolução de crescimento. Os danos oxidativos observados nos protocolos investigados podem ter inviabilizado os tecidos criopreservados, mesmo que parte dos tecidos expostos em nitrogênio líquido aparente viabilidade, ocorre baixa ou nula regeneração de tecidos. A compreensão do grau de sensibilidade de materiais para soluções vitrificantes é o primeiro passo para otimização dos protocolos de vitrificação, no presente estudo a desidratação por 15 minutos em solução ½ PVS2 seguido de 30 minutos em PVS2 apresentou os melhores resultados para o porta-enxerto IAC 571-6

Basic principles the work is based on are pragmatic pedagogy and project education. First part of the work is focusing on theory. The content of this work is dealing with the problem of media education. Second part of the diploma work is dealing with education to media literacy. Game and cooperation activities are emphasized which serve to train the interactive forms of communication. The endeavour is to develop the skills such as looking up, gaining, processing, evaluation and using of information which comes from media (newspapers, radio, television and internet). The component of this part is proposal of structure of practical projects for 6. up to 9. grade of the basic school.

Tais manifestações comunicam códigos simbólicos expressos por uma linguagem sociocultural extremamente rica semanticamente. Parto da hipótese de que uma linguagem supostamente transmitida pela doutrina espírita constitui uma das principais razões pela qual muitos sujeitos justificam sua adesão ao corpo de crenças, valores e preceitos morais baseados nas codificações feitas por Allan Kardec. E, por conseguinte, se sentem identificados com tal cosmovisão compartilhada, que prevê basicamente a possibilidade da vida além da morte, de uma continuidade para a trajetória do espírito através da reencarnação e da comunicação com o espírito dos mortos. A religião Espírita assim como qualquer outra religião é capaz de aferir orientações comportamentais responsáveis por guiar a conduta do indivíduo no cotidiano, fornecendo sentidos e significados imbuídos na construção da realidade dos sujeitos. Para um enquadramento analítico da manifestação mediúnica recorro ao discurso construído por meus interlocutores - os médiuns espíritas- acerca da experiência religiosa da mediunidade. A presente pesquisa visa discutir os possíveis sentidos elaborados em torno das práticas mediúnicas na religião Espírita, a partir dos relatos, das experiências e das memórias de vida de médiuns kardecistas da cidade de Juiz de Fora- MG. Para tanto, empreendo uma investigação de caráter qualitativo, elaborado com base em uma etnografia desenvolvida em centros espíritas Kardecistas da cidade, articulada à análise documental e bibliográfica acerca da mística espírita. / For the mediums, the mediumship phenomena conjuncture an immaterial dimension, intangible, ephemeral, subjective and particular, which is built and meant in the material plane of the objective reality of everyday life. Such manifestations communicate symbolic codes expressed by an extremely rich sociocultural language in its semantic ways. I start from the hyphotesis that a language supposedly transmitted by the spiritual doctrine is one of the main reasons why many subjects justify their adherence to the body of beliefs, values and moral principles based on the codifications made by Allan Kardec. And therefore, they feel identified with such shared worldview, which basically foresees the possibility of life beyond death, a continuity to the spirit’s path through reincarnation and the communication with the spirits of the dead. The Spiritualist religion as any other religion is able to assess behavioral guidelines responsible for guiding the conduct of the individual in its daily life, providing senses and meanings imbued with the construction of the reality of the subjects. For an analytical framework of mediumism manifestation, I resort to the discourse constructed by my interlocutors – the spiritual mediums - about the religious experience of mediumship. I consider important the question: what this mediumship experience means to them? This research aims to discuss the possible meanings developed around the mediumship practices in the Spiritualist religion, taken from the reports, experiences and living memories of Kardecists mediums from the city of Juiz de Fora – MG. To this end, I engage a qualitative research, prepared based on an ethnography which was developed in Kardecists spiritual centers of the city, linked to the documentary and literature analysis about the spiritualist mysticism.

Cette thèse propose une lecture historique du statut du musicien dans la cité – entendue comme une communauté politique organisée autour de la volonté du vivre-ensemble –, selon la conception qu’en avait les anciens Grecs. L’étude de la fonction sociale du musicien depuis la préhistoire a permis de dégager aujourd'hui un nouveau paradigme : celui de médiateur ou de médium, en prise avec un milieu éco-techno-symbolique qu’il contribue à transformer, en rendant visibles les êtres humains qui l’habitent et en particulier les personnes marginalisées, exclues de la société ou en situation de vulnérabilité. L’éclairage probant des neurosciences autour du phénomène musical vient renforcer cette idée que la musique, en tant que langage du corps, peut agir concrètement sur les individus, sans cependant réduire l’humain à des considérations purement neuronales qui expliqueraient la contemplation ou la création de l’œuvre d’art comme le prétend la neuroesthétique. / This doctoral thesis put forward an historical reading of the musician’s status in the city. The ancient Greeks had another approach of the city, known as a political community organised around the willingness of « the community life ». The study of the musician’s social status since prehistory has allowed nowadays to shed light on a new paradigm. The musician became a mediator or a medium in conjunction with an economical, technological and symbolic environment, he contributes to a transformation, which makes visible the human beings inside the environment, in particular those marginalised, banned from the society or in situation of vulnerability. The neurosciences highlight this matter by strengthening this phenomenon. Indeed, the music considered as a body language can genuinely operate on people without reducing the human beings to neuronal attentions which can explain the contemplation or the creation of the art work, as neuroesthetics claim.

The topic of master´s thesis is atmosferic gasification of biomass by the addition of oxygen and steam. The theoretical part is devoted to the description of biomass, process of gasification, kinds of gasification reactor and product gas. Further experiments are devised depending on the gasification medium and output temperature. In this case, the key aspect is the steam addition control. In the practical part, these experiments are performed in a fluidized bed reactor. Resulting values from samples of gas and tar are subsequently processed and evaluated.

This dissertation shows that people use media without understanding them. Person understands himself only as an object of oparation. Media are based on the essence of techniques - Gestell. This thesis introduces the concept of technical space which helps people to understand the scope of the media as a system. Cultural memory and its media have a productive power in constructing historical nar- ratives. Especially, in Central and Eastern Europe, memory is mostly created through bottom- up processes: The past is mediated in the public sphere and imagined in popular culture (Blacker, Etkind 2013: 10). The velvet revolution is followed by a memory boom in the Czech Republic, in which media acts as memory activists addressing tabooed memories. At a time where the last witnesses of the expulsion are passing away, the dying voice is taken up by media This thesis aims to explore the determinant patterns of the Czech narrative in post-1989 fiction. Particularly, it focuses on postmemorial representations. The thesis is based on an anal- ysis of five fictional works dealing with the expulsion of the Sudeten Germans. The expulsion is embedded in the century-long co-habitation with the Sudeten German depicted as a neigh- bour. However, the Sudeten Germans are only incorporated into a regional but not the national Czech identity. While the expulsion is reframed in an ethical-legal framework, on a narrative level strategies of individualisation and universalisation are employed to make the past acces- sible for those born later.

The Japanese way of tea has been explored with scientific rigor through the fields of anthropology, aesthetics, philosophy, religion, semiotics, and other academic disciplines. This thesis will examine the phenomenon of the "bowl of tea" (considered here as the axis mundi of a tea gathering) on three levels: as a gift, as the basis for the foundation of responsible and considerate relationships in the community, and as a medium of transformation into peacefulness. First, it will explore the nature of the gift in different social practices of exchange in general and in the way of tea in particular. Second, it will present research connecting the gift and the process of giving as a mode of hospitality in the tea gathering. Third, it will assess, through concepts of spatial semiotics, the transformative nature of the way of tea. It will conclude with a statement as to the potential of the way of tea experience to transform human experience and behaviour to others via the phenomenon of gift. La pratique de la médiation thérapeutique en groupe par le modelage de la terre avec des personnes autistes et psychotiques met en évidence les modalités de constitution et du travail du médium malléable.Ce dernier permet, à travers l’acte de modeler au sein du cadre dispositif, une mise en forme des traces sensori-motrices (ou traces sensorielles), préalables à la constitution de l’image du corps.Le travail du médium malléable rend aussi compte des modalités de constitution de l’enveloppe psychique, tant sur un plan individuel que groupal.Les formes constituées mettent en avant le recours à des actes symboliques, que nous répertorions, témoins de l’état de constitution de l’enveloppe psychique. De la bidimensionnalité (identification adhésive) à l’espace tridimensionnel (identification projective), le sujet émiette la matière, la morcelle, coupe, sépare, rassemble, assemble et construit.Le travail du médium malléable et le mode d’utilisation du médium réactualisent le lien à l’objet primaire. Ils permettent aussi de focaliser les effets transféro-contre-transférentiels. Degradace alifatických polyesterů - Vliv velikosti tělesa a jeho tvaru / Aliphatic polyesters degradation - Influence of the body size and habitus

In the theoretical part of this thesis are described the synthesis, properties and mechanism of biodegradation of aliphatic polymers - polylactic acid, polyglycolic acid and their copolymer PLGA. There are also discussed the possibilities of modifying the properties, such as block copolymer PEG-PLGA synthesis. Summary informations concerning the production and use of biodegradable polymers are also shortly described. The aim of the experimental part was to observe the effect of PLGA polymer matrix size and ionic strength of the aqueous medium on the polymer swelling and erosion. Samples with weight 150 mg and 1000 mg were placed into 37 řC citrate buffer with pH 6. Each of the used mediums had various concentration c [0; 0,125; 0,25; 0,5; 1]. Measurement was carried out in period of 28 days. Values of degree of swelling and of erosion were measured. The pulsion behavior of swelling in both sample sizes was confirmed. However, the sample size has a strong impact on the rate and extent of swelling. It was demonstrated that larger samples disintegrate faster which is explained by a higher rate of autocatalysis within the polymer matrix. The results demonstrated also the effect of ionic strength on erosion when isotonic solution suppressed erosion rate.
