The vase-occlusive phenomenon is associated with the inflammatory process unleashed by the polymerization of deoxygenated HbS, which favors the polymerization of erythrocytes in the microcirculation and the subsequent obstruction of endothelia light. Several evidences suggest that the reduction of the bioavailability of the nitric oxide (NO) can be a factor to favor the vase-occlusion and that the free radicals produced give rise to the lipidic peroxidation and the subsequent production of the abnormal quantities of malonaldehyde (MDA), which induce the permeability of the red cell membrane. Among the genetic factors that modulate the clinic of the AF, there is the HbF levels. The hydroxyurea (HU) is used in the treatment of SCD and apart from increasing the HbF levels it also seems to contribute as a nitric oxide donator. In this context, the present work aims to evaluate the oxidative stress by means of seric dosage of the MDA and to determinate the levels of NO and lactate desidogenase lactica (LDH) in patients with SCD by means within the Walter Cantídeo University Hospital (HUWC) and to correlate with the use of HU. This way, it was used 65 samples of peripheral blood of adult patients with AF with either HU or not, and a control group was composed by 20 donors of the blood bank with HbAA. The levels of NO, MDA and LDH were determined by biochemical methods. The participants of the research were selected in the Center of Hematology and Hemotherapy of Ceará State (HEMOCE). All the participants have signed a document stating free consent and approval (TCLE) and they were submitted to a previously elaborated questionnaire. The obtained data were expressed as averages ± pattern deviation and analyzed by using a statistics software SPSS15.0, using Pearson and Spearman correlation coefficients for the study of the correlations among MDA, NO, LDH, HbF and Hb. In order to compare the three groups through the levels of MDA, NO and LDH, the results were obtained by means of deviance analysis (ANODEV). It has been observed a majority of females and fallow race (group I) and black (group II), in which the most part of these patients have ages in the range of 20-35 (group I) and 20-40 (group II) and come from Ceará State. Most of them did not smoke, drink alcohol nor use vitamin supplements (C and E). The levels of MDA in the three groups are all different, i. e., the control group showed an average level of MDA higher than group II, and the later higher than that of the group one. It was not verified difference in the seric levels of NO in relation to the use of HU. In the three groups, it was observed that only the control group is different, showing an average much lower than those of the other two groups of patients regarding the levels of LDH. It was also observed a correlation inversely proportional between the two levels of NO and HbF for the group I, while in the group II there was no correlation. Regarding to the concentration of Hb, there was no correlation both in the group I and in the group II, with levels of NO. There was no significantly correlation among the levels of MDA, Hb and HbF for the studied groups. It was found a correlation inversely proportional between the levels of LDH and Hb for the group I. It was observed accentuated levels of MDA in patients of group II who have made two or more transfusions throughout the year, who had leg ulcers and who had three or more vase-occlusive crises form both the groups. It was found no relation of NO with the clinic variables. CONCLUSÃO: Our results have shown that patients who use HU are not protected against lipidic peroxidation, hemolysis and consume of available NO / A anemia falciforme (AF) se caracteriza por anemia hemolítica crônica e com fenômenos vaso-oclusivos, seguidos de lesões a órgãos alvos, responsáveis pela mortalidade associada a esta doença. O fenômeno vaso-oclusivo está associado ao processo inflamatório desencadeado pela polimerização da HbS desoxigenada, favorecendo a impactação das hemácias na microcirculação e subseqüente obstrução da luz do endotélio. Numerosas evidências sugerem que a redução da biodisponibilidade do óxido nítrico (NO) pode ser um fator que favoreça a vaso-oclusão e que os radicais livres formados induzem à peroxidação lipídica e à subseqüente produção de quantidades anormais de malonaldeído (MDA), que provoca alteração da permeabilidade da membrana eritrocitária. Dentre os fatores genéticos que modulam a clínica da AF consta os níveis de HbF. A hidroxiuréia (HU) é utilizada no tratamento da AF e além de aumentar os níveis de HbF parece contribuir como um doador de óxido nítrico. Nesse contexto, os objetivos do presente trabalho foram avaliar o estresse oxidativo por meio da dosagem sérica de MDA e determinar os níveis de NO e de lactato desidrogenase láctica (LDH) em pacientes com AF em acompanhamento ambulatorial no Hospital Universitário Walter Cantídeo (HUWC) e correlacionar com o uso da HU. Desta forma foram utilizadas 65 amostras de sangue periférico de pacientes adultos com AF em uso ou não de HU, e um grupo controle foi elaborado por 20 doadores do banco de sangue com HbAA. Os níveis de NO, MDA e LDH foram determinados por métodos bioquímicos. Os participantes da pesquisa foram selecionados junto ao centro de hematologia e hemoterapia do Ceará (HEMOCE). Todos os indivíduos assinaram o termo de consentimento livre e esclarecido (TCLE) e foram submetidos a um questionário pré-estruturado. Os dados obtidos foram expressos como médias  desvio padrão e analisados utilizando-se o programa estatístico SPSS15.0, utilizamos os coeficientes de correlação de Pearsaon e de Spearman para o estudo das correlações entre MDA, NO, LDH, HbF e Hb. Para comparar os três grupos através dos níveis de MDA, NO e LDH obtemos os resultados através da análise de deviance (ANODEV). verificamos um predomínio do sexo feminino, e das raças parda (grupo I) e negra (grupo II), sendo a grande maioria desses pacientes com idade variando de 20 a 35 anos (grupo I) e 20 a 40 anos (grupo II) e procedentes do estado de Ceará. Em sua maioria, não faziam uso de fumoetilismo e de suplementação de vitaminas (C e E). Os níveis de MDA nos três grupos são todos diferentes, isto é, o grupo controle apresentou um nível médio de MDA superior ao grupo II, e este por sua vez superior ao grupo I. Não encontramos diferença nos níveis séricos de NO em relação ao uso de HU. Nos três grupos observamos que apenas o grupo controle difere, apresentando uma média bem inferior a dos dois grupos de pacientes quanto aos níveis de LDH. Observamos uma correlação inversamente proporcional entre os de níveis NO e de HbF para o grupo I, enquanto que para o grupo II não houve correlação. Em relação à concentração da Hb não houve correlação tanto no grupo I quanto no grupo II, com os níveis de NO. Não houve correlação significativa entre os níveis de MDA e Hb e HbF para nenhum dos grupos estudados. Encontramos uma correlação inversamente proporcional entre os níveis de LDH e de Hb para o grupo I. Observou-se níveis elevados de MDA em pacientes do grupo II que realizaram duas ou mais transfusões no decorrer do ano, que apresentaram úlcera de perna maleolar e que tiveram três ou mais crises vaso-oclusivas para ambos os grupos. Não encontramos nenhuma relação do NO com as variáveis clínicas. CONCLUSÃO: Nossos resultados mostram que os paciente em uso de HU não estão protegidos contra a peroxidação lipídica, hemólise quanto ao consumo de NO disponível.

One hundred and twenty young male Wistar rats, mean weight 172.5±15g, were randomized in five equal groups (GS - Sham, GC – Control, GD – Dimethylsulfoxide, GA – Lipoic acid and GT/D – Ternatin/DMSO). Rats of each group were distributed into four subgroups (T-0, T-1, T-3 and T-6), each comprising six animals. All surgical procedures were performed under inhalatory ether anesthesia. Experimental rats received intraperitoneal (i.p.) injections of 3% aqueous solution of DMSO (10 ml/kg, GD), LA (10 mg/kg, GA) or TN/DMSO (12 mg/kg, suspended in 3% aqueous solution of DMSO), GT/D) 24, 12 and 4 hours before cord detorsion. Last dose was given 1 hour before ischemia induction or second Sham operation. Sham (GS) and Control (GC) rats received NaCl 0.9% 10 ml/Kg i.p. Thiobarbituric acid reactive substances (TBARS) and reduced glutathione (GSH) levels (µmol/g wet tissue) were assayed in testis. Total Antioxidant Power (TAP) (µM) was measured in blood plasma. Data were first analyzed by Kolmorogov-Smirnov test to assess differences in the general shapes of distribution. Comparisons between groups were made using the t test for normally distributed data and the Mann-Whitney U test for non-normally distributed data. P values of <0.05 were considered to indicate statistical significance. Comparisons between antioxidant-treated and saline-treated rats (Control group, GC) were made using Dunnett’s test. Spermatic cord torsion induced significant decrease in testis MDA levels during ischemia and reperfusion in DMSO (GD) and LA (GA) and in TN/DMSO (GT/D) treated rats during reperfusion, compared with respective controls. MDA levels were significantly increased in GC compared with Sham (GS) rats at the end of the ischemia and during reperfusion as well as one hour after detorsion (T-1) in GC rats compared with T-0, in the same group, indicating additional damage imposed by the afflux of oxygenated blood (reperfusion) to the ischemic tissues. GSH levels decreased significantly in saline-treated (GC) compared with Sham-operated rats and increased significantly in DMSO and LA treated rats at the end of ischemia and during reperfusion. The model utilized in this study did not induce systemic effects. TAP was significantly increased during reperfusion (T-1) in LA (GA) and TN/DMSO (GT/D) pretreated rats, compared with respective controls. The results of the present study reinforce the hypothesis that ischemia and reperfusion are free radicals generating processes. The different antioxidants studied here have demonstrated great efficacy in cell membrane protection by decreasing lipid peroxidation in all experiments. Increased reduced glutathione levels in DMSO and LA treated rats support the view that these substances can exert a protective effect against testis oxidative stress injury caused by ischemia/reperfusion in rats subjected to torsion of the spermatic cord lasting three hours. TN protective effects in reducing lipoperoxidation were more pronounced in ischemic tissues. / O efeito protetor do dimetilsulfóxido (DMSO), do ácido lipóico (LA) e da ternatina (TN), conhecidos seqüestradores de radicais livres e protetores da membrana celular foram avaliados utilizando um modelo experimental de isquemia/reperfusão do testículo. Cento e vinte ratos Wistar jovens com peso médio de 174 g foram distribuídos ao acaso em cinco grupos (GS - Simulado, GC – Controle, GD – Dimetilsulfóxido. GA – Ácido lipóico e GT/D – Ternatina/DMSO) numericamente iguais e posteriormente redistribuídos em quatro subgrupos (T-0, T-1, T-3 e T-6), com seis animais cada. Todos os procedimentos cirúrgicos foram realizados sob os efeitos da anestesia geral inalatória. Os animais receberam injeções intraperitoniais (i.p.) de solução aquosa de DMSO a 3% (10 ml/kg, GD), LA (10 mg/kg, GA) ou TN+DMSO (12 mg/kg, GT/D, em solução aquosa de DMSO a 3%) 24, 12 e 4 horas antes da destorção do cordão espermático. A última dose foi administrada uma hora antes da indução da isquemia ou da segunda operação simulada (Sham). Os ratos do grupo Simulado (GS) e do grupo Controle (GC) receberam de NaCl 0,9% (10 ml/Kg) i.p. Foram determinadas as concentrações de substâncias reativas ao ácido tiobarbitúrico (TBARS) e glutationa reduzida (GSH) no tecido (testículo), e a capacidade antioxidante total (CAP) do plasma. Para determinação do padrão de distribuição das amostras utilizou-se o teste de Kolmorogov-Smirnov. As comparações entre os grupos foram feitas utilizando-se o t teste ou o teste U de Mann-Whittney quando assim indicado. Para as comparações dos grupos tratados com antioxidantes ao grupo Controle (GC) utilizou-se o teste de Dunnett. Valores de p<0,05 foram considerados significantes. A torção do cordão espermático induziu uma redução significante das concentrações de MDA, no tempo máximo de isquemia e na reperfusão, em tratos pré-tratados com DMSO (GD) e LA (GA) e durante a reperfusão naqueles pré-tratados com TN/DMSO (GT/D), comparados com seus respectivos controles. Os níveis de MDA estavam significantemente elevados nos ratos do grupo GC comparados aos animais do grupo GS, no tempo máximo de isquemia e durante a reperfusão, bem como uma hora após a destorção (T-1) comparada ao T-0, demonstrando o dano adicional decorrente do afluxo de sangue oxigenado (reperfusão) ao tecido isquêmico. As concentrações de GSH diminuíram nos ratos pré-tratados com solução salina e aumentaram significativamente nos ratos pré-tratados com DMSO ou LA, no tempo máximo de isquemia e durante a reperfusão. O modelo isquêmico utilizado não foi capaz de gerar efeitos sistêmicos. A capacidade antioxidante total aumentou significantemente durante a reperfusão (T-1) nos ratos pré-tratados com LA (GA) e TN/DMSO (GT/D) comparados aos respectivos controles. Esses resultados fortalecem a hipótese de que a isquemia e reperfusão são processos geradores de radicais livres. Os diferentes antioxidantes estudados mostraram grande eficiência na proteção da membrana celular, reduzindo a peroxidação lipídica, em todos os experimentos. O aumento dos níveis de glutationa reduzida, nos ratos pré-tratados com DMSO e LA, mostra que estes antioxidantes exercem uma proteção eficaz contra o estresse oxidativo induzido pela isquemia/reperfusão em ratos submetidos à torção do cordão espermático por três horas. Os efeitos protetores da ternatina se manifestam com maior intensidade no tecido isquêmico, no controle da peroxidação lipídica.

Atualmente, somente o teste de germinação é usado para avaliar a qualidade fisiológica de sementes em bancos de germoplasma e outros métodos ainda são necessários para prever a longevidade de sementes. A espécie Astronium fraxinifolium Schott é uma árvore das regiões da Amazônia, Cerrado e Caatinga, ameaçada de extinção, que apresenta sementes não dormentes e que germinam rapidamente. O objetivo do presente trabalho foi estudar a longevidade de sementes de A. fraxinifolium, coletadas em diversos locais de duas regiões geográficas distintas e avaliar o transcriptoma. Sementes de dez acessos de A. fraxinifolium, sendo seis coletados no estado de Goiás (GO) e quatro em Minas Gerais (MG) foram avaliados. Foram realizados pré-testes para determinar a melhor temperatura para germinação e o tempo para iniciar a protrusão da radícula. Testes de envelhecimento, condutividade elétrica, teor de malonaldeído e análise do transcriptoma foram realizados. A temperatura ótima de germinação foi de 30° C e os primeiros eventos de protrusão radicular ocorreram após 20 horas de embebição. Sementes coletadas em Goiás apresentaram maior viabilidade e longevidade após os testes de envelhecimento para todas as condições testadas e os acessos GO 6 e MG 2 apresentaram, respectivamente, a maior e menor longevidade após o envelhecimento. O teste de condutividade elétrica não foi adequado para avaliar a longevidade dos acessos e a avaliação do teor de malonaldeído foi considerado um teste promissor para avaliar a longevidade de sementes de A. fraxinifolium. Foi realizada análise de RNASeq para avaliar os transcriptomas de eixos embrionários de sementes dos acessos GO 6 e MG 2, com sementes envelhecidas e não envelhecidas. Os genes expressos diferencialmente foram relacionados aos processos de transcrição de RNA, kinases, ubiquitinas, metabolismo de amido, microtúbulos, componentes de membrana, polimerase e transporte de carboidratos e zinco. / Seeds of different species stored under the same conditions differently lose their viability. Currently, the physiological quality of seeds stored in genebanks is evaluated only by the germination test and other methods are still needed to predict seed viability.The species Astronium fraxinifolium Schott is a tree in the Amazonian regions, Cerrado and Caatinga, which presents seeds that are non-dormant and which germinate rapidly. The goal of this work was to study the longevity of seeds A. fraxinifolium collected at several sites from two different geographic regions, and assess the transcriptome. For that, seeds from ten accessions of A. fraxinifolium were collected (six in the state of Goiás (GO) and four in Minas Gerais (MG) and evaluated. Pre-tests were performed to determine the optimal germination temperature and the onset of the radicle protrusion. Aging tests, electrical conductivity, malonaldehyde content and transcriptome analysis were performed. The optimal germination temperature was 30o C and the first radicle protrusion events occurred after 20 hours of imbibition. Seeds collected in Goiás displayed higher viability and longevity after aging in all tested conditions and seeds from accessions GO 6 and MG 2 showed, respectively, the highest and lowest longevity indexes after aging. The electrical conductivity test was not adequate to evaluate the longevity of the accessions and the assessment of the malonaldehyde content was considered a promising test to assess the longevity of A. fraxinifolium seeds. RNASeq analysis was performed to evaluate the transcripts of embryonic axes of seeds of the GO 6 and MG 2 accessions, with aged and unaged seeds. Differentially expressed genes were related to RNA transcription processes, kinases, ubiquitin, starch metabolism, microtubules, membrane components, polymerase, and transport of carbohydrates and zinc.

Efeitos da inalação da fumaça do cigarro no estresse oxidativo do sistema nervoso central de camundongos jovens / Effects of cigarette smoke in oxidative stress in the central nervous system of young mice

A fumaça do cigarro é composta por mais de 4.700 substâncias, muitas das quais tóxicas. O sistema nervoso central (SNC) possui poucas defesas antioxidantes, além de ser rico em lipídeos facilmente oxidáveis e de conter alto teor de metais de transição envolvidos na formação de radicais livres. Existem evidências de que a fumaça do cigarro causa alterações nas enzimas antioxidantes em roedores adultos, mas pouco se sabe sobre os efeitos do tabaco no SNC ainda em desenvolvimento. Assim, este trabalho tem como objetivo avaliar os possíveis efeitos da fumaça do cigarro no SNC de camundongos jovens. Camundongos BALB/c foram expostos a uma mistura de fumaça central e lateral do cigarro (Souza Cruz - Derby Vermelho) dentro de uma câmara de polipropileno acoplada a um sistema Venturi. Além da exposição aguda, realizada no 18º dia de vida, os animais foram expostos a partir do 5º dia de vida por 13 ou 35 dias, durante 2 horas por dia, 7 dias por semana. Os animais foram então eutanasiados por deslocamento cervical imediatamente ou três horas após a última exposição e foram realizadas as determinações das atividades enzimáticas da glutationa peroxidase, glutationa redutase, glutationa S-transferase, além da quantificação de malonaldeído (MDA) e 3-nitrotirosina no cerebelo, córtex frontal, estriado, hipocampo e hipotálamo. Nossos resultados indicam que o SNC em desenvolvimento é susceptível à fumaça do cigarro. Foram detectadas alterações nas enzimas antioxidantes em diferentes estruturas encefálicas imediatamente após a última exposição sugerindo diferença de sensibilidade das diferentes áreas à fumaça do cigarro. Contudo, essas perturbações não são persistentes, uma vez que a maior parte delas desapareceu três horas após a exposição. O cerebelo parece ser a estrutura mais resistente enquanto o córtex frontal e o estriado, as mais sensíveis. No córtex frontal as alterações enzimáticas são mais persistentes e houve aumento de MDA no grupo agudo e eutanásia 3 horas após a exposição, enquanto no cerebelo, estriado e hipocampo houve aumento de MDA no grupo agudo e eutanásia imediatamente após a exposição. Esses resultados sugerem uma resposta mais tardia do córtex frontal e uma possível adaptação dos tecidos ao xenobiótico. The central nervous system (CNS) has few antioxidant defenses, is rich in easily oxidable lipids and contains high levels of transition metals which are involved in the formation of free radicals. There are evidences that tobacco smoke causes changes in the antioxidant enzymes in adult rodents, but little is known about its effects during CNS development. Thus, the aim of this work was to evaluate the possible effects of cigarettes smoke in the CNS of young mice. BALB/c mice were exposed to a mixture of mainstream and sidestream smoke of cigarette (Souza Cruz Red Derby) in a polypropylene chamber attached to a Venturi system. Besides acute exposure, performed at the 18th day of life, the animals were exposed from the 5th day of life for 13 or 35 days, 2 hours a day, 7 days in a week. The animals were then euthanized by cervical dislocation, immediately or three hours after the last exposure. The determinations of enzymatic activities of glutathione peroxidase, glutathione reductase, glutathione S-transferase, and the quantification of malonaldehyde (MDA) and 3-nitrotyrosine in cerebellum, frontal cortex, striatum, hippocampus and hypothalamus were performed. Our results indicate that the CNS in development is susceptible to cigarettes smoke. Alterations in antioxidant enzymes in different brain structures were detected immediately after the last exposure suggesting differences in sensitivity of different areas to cigarette\'s smoke. However, these disturbances are not persistent, as most of them disappeared three hours after exposure. Cerebellum seems to be the more resistant structure, while frontal cortex and striatum the most sensitive. Enzyme changes in frontal cortex were more persistent and there was an increase of MDA only in the acute group and euthanasia 3 hours after exposure, whereas in cerebellum, striatum and hippocampus, MDA increased only in acute group and immediately euthanasia after exposure. These results suggest a delayed response of the frontal cortex and a possible adaptation of tissues to xenobiotics. It is important to point out that euthanasia performed immediately after chronic exposure to cigarette smoke not only reflect the effect of the last exposure, since the pattern found in the modification of enzymes and in the determination of MDA was different from that observed after acute exposure.

A fumaça do cigarro é composta por mais de 4.700 substâncias, muitas das quais tóxicas. O sistema nervoso central (SNC) possui poucas defesas antioxidantes, além de ser rico em lipídeos facilmente oxidáveis e de conter alto teor de metais de transição envolvidos na formação de radicais livres. Existem evidências de que a fumaça do cigarro causa alterações nas enzimas antioxidantes em roedores adultos, mas pouco se sabe sobre os efeitos do tabaco no SNC ainda em desenvolvimento. Assim, este trabalho tem como objetivo avaliar os possíveis efeitos da fumaça do cigarro no SNC de camundongos jovens. Camundongos BALB/c foram expostos a uma mistura de fumaça central e lateral do cigarro (Souza Cruz - Derby Vermelho) dentro de uma câmara de polipropileno acoplada a um sistema Venturi. Além da exposição aguda, realizada no 18º dia de vida, os animais foram expostos a partir do 5º dia de vida por 13 ou 35 dias, durante 2 horas por dia, 7 dias por semana. Os animais foram então eutanasiados por deslocamento cervical imediatamente ou três horas após a última exposição e foram realizadas as determinações das atividades enzimáticas da glutationa peroxidase, glutationa redutase, glutationa S-transferase, além da quantificação de malonaldeído (MDA) e 3-nitrotirosina no cerebelo, córtex frontal, estriado, hipocampo e hipotálamo. Nossos resultados indicam que o SNC em desenvolvimento é susceptível à fumaça do cigarro. Foram detectadas alterações nas enzimas antioxidantes em diferentes estruturas encefálicas imediatamente após a última exposição sugerindo diferença de sensibilidade das diferentes áreas à fumaça do cigarro. Contudo, essas perturbações não são persistentes, uma vez que a maior parte delas desapareceu três horas após a exposição. O cerebelo parece ser a estrutura mais resistente enquanto o córtex frontal e o estriado, as mais sensíveis. No córtex frontal as alterações enzimáticas são mais persistentes e houve aumento de MDA no grupo agudo e eutanásia 3 horas após a exposição, enquanto no cerebelo, estriado e hipocampo houve aumento de MDA no grupo agudo e eutanásia imediatamente após a exposição. Esses resultados sugerem uma resposta mais tardia do córtex frontal e uma possível adaptação dos tecidos ao xenobiótico.
