Estudo do metabolismo oxidativo de neutrófilos humanos expostos a produtos de alga marinha e fungo endofí­tico provenientes da Antártica / Study of the oxidative metabolism of human neutrophils exposed to compounds of marine algae and endophytic fungi from Antarctica

Neutrófilos são a primeira linha de defesa do sistema imunológico, eles produzem substâncias microbicidas, tais como espécies reativas de oxigênio (ERO), são capazes de eliminar patógenos e possuem papel importante em processos inflamatórios fisiológicos e patológicos. No entanto, os efeitos benéficos e nocivos mediados por esta célula dependem, em grande parte, do equilíbrio redox, que se estabelece entre a produção de ERO e a ação de antioxidantes. A quebra deste equilíbrio leva ao estresse oxidativo, capaz de causar danos pelas ERO sobre as estruturas biológicas. Sendo assim, a regulação das funções dos neutrófilos é um importante alvo terapêutico. O objetivo deste trabalho foi avaliar a ação de produtos naturais - extrato bruto, frações e subfrações - oriundas da Antártica na regulação do metabolismo oxidativo de neutrófilos humanos. Para este propósito, o extrato bruto da alga Palmaria decipiens, nove (9) frações do fungo endofítico Aspergillus unguis e nove (9) subfrações deste foram estudadas. As amostras foram avaliadas quanto à atividade scavenger, utilizando-se o 2,2-difenil-?-picrilhidrazil; à citotoxidade aos neutrófilos, por análise da viabilidade celular com o ensaio de exclusão do Azul de Trypan; e ao efeito sobre a produção de ERO pelos neutrófilos, medida por quimiluminescência dependente de luminol, utilizando-se forbol-12-miristato-13-acetato como estímulo para os neutrófilos. Os resultados mostraram que: 1) nenhuma das amostras apresentou atividade scavenger; 2) a viabilidade celular manteve-se igual ou maior que 90% quando neutrófilos foram expostos às frações FR5, FR6*, FR8, e às subfrações contendo 100% Acetato de Etila, 10% Acetato de Etila:Hexano, 20% Acetato de Etila:Hexano e 40% Acetato de Etila:Hexano; 3) dentre as frações que mostraram viabilidade celular maior que 90%, a inibição da produção de ERO pelos neutrófilos foi observada com a FR5 (51%), a FR6* (20%), a subfração 100% Hexano (73%), a subfração 20% Acetato de Etila/Hexano (42%) e a subfração 40% Acetato de Etila/Hexano (38%). Os resultados mostram que algumas das frações e subfrações do fungo endofítico Aspergillus unguis apresentaram inibição da produção de ERO pelos neutrófilos humanos entre (20 a 73%). Esta inibição não é por atividade scavenger de radiciais de oxigênio, sugerindo que este efeito regulador sobre o neutrófilo possa resultar da atividade dos componentes nestas frações sobre outras vias metabólicas desta célula. A partir destes resultados, faz-se necessário identificar a composição destas frações e seus efeitos sobre vias metabólicas e sobre outras funções efetoras dos neutrófilos. A contribuição deste estudo é a procura por moléculas bioativas, capazes de regular parcialmente as respostas do neutrófilo, para restabelecer o equilíbrio funcional desta célula em estados patológicos, diminuindo seus efeitos nocivos sem prejuízo do seu papel crucial para a homeostase. / Neutrophils are the first line of defense of the immune system, they produce microbicidal substances, such as reactive oxygen species (ROS), able to eliminate pathogens and play important roles in physiological and pathological inflammatory processes. However, the beneficial and harmful effects mediated by this cell depend on the redox balance, which is established between the production of ROS and the action of antioxidants. The imbalance leads to oxidative stress capable of causing damage by ROS on biological structures. Thus, the regulation of neutrophil functions is an important therapeutic target. The aim of this work was to evaluate the action of natural products - crude extract, fractions and subfractions - originating from Antarctica environment in the regulation of the oxidative metabolism of human neutrophils. For this purpose, the crude extract of the Palmaria decipiens algae, nine (9) fractions of the endophytic fungus Aspergillus unguis and nine (9) subfractions from this fungus were studied. The samples were evaluated for scavenger activity using 2,2-diphenyl-?-picrylhydrazyl; cytotoxicity to neutrophils, by cellular viability analysis with the Trypan Blue exclusion assay; and to the effect on neutrophil production of ROS, as measured by luminol-dependent chemiluminescence, using phorbol 12-myristate 13-acetate as a stimulus for neutrophils. The results showed that: 1) none of the samples had scavenger activity; 2) the cell viability remained equal to or greater than 90% when neutrophils were exposed to fractions FR5, FR6*, FR8, and subfractions containing 100% Ethyl Acetate, 10% Ethyl Acetate:Hexane, 20% Ethyl Acetate:Hexane and 40% Ethyl Acetate:Hexane; 3) among the fractions that showed cellular viability greater than 90%, the inhibition of ROS production by neutrophils was observed with FR5 (51%), FR6* (20%), subfraction 100% Hexane (73%), the subfraction 20% Ethyl acetate / Hexane (42%) and subfraction 40% Ethyl acetate / Hexane (38%). The results show that some of the fractions and subfractions of the endophytic fungus Aspergillus unguis showed inhibition of ROS production by human neutrophils between (20 to 73%). This inhibition is not by scavenger activity of oxygen radicals, suggesting that this regulatory effect on the neutrophil may result from the activity of the components in these fractions on other metabolic pathways of this cell. From these results, it is necessary to identify the composition of these fractions and their effects on metabolic pathways and on other effector functions of neutrophils. The contribution of this study is the search for bioactive molecules, able to partially regulate neutrophil responses, to restore the functional balance of this cell in pathological states, reducing its harmful effects without prejudice to its crucial role for homeostasis.

Caracterização funcional da isoforma A do fator de início de tradução de eucariotos 5A (eIF5A) /

Orientador: Augusto Ducati Luchessi / Resumo: O fator de início de tradução de eucariotos 5A (eIF5A) é uma proteína altamente conservada em archaeas e eucariotos. Caracteriza-se por ser a única proteína conhecida a apresentar o resíduo de aminoácido hipusina, produzido a partir de uma modificação pós-traducional denominada hipusinação, a qual é essencial para sua atividade. Atualmente eIF5A é correlacionada com o início, elongação e término da tradução, além disso, sua função têm sido associada com diferentes processos celulares, fisiológicos e patológicos. Em humanos, é predito que eIF5A seja gerada a partir de cinco variantes transcricionais (A, B, C, D e X5). As variantes B, C, D e X5 codificam a proteína canônica isoforma B de 17 kDa, a qual vêm sendo estudada nas últimas décadas. Entretanto, é previsto que a variante A codifique uma isoforma alternativa (isoforma A) de 20 kDa, que tem uma sequência adicional de 30 aminoácidos na extremidade N-terminal. Neste contexto, o presente estudo teve como objetivo caracterizar eventos moleculares inéditos envolvendo a isoforma A de eIF5A1 humana. Neste estudo, mostramos que a isoforma A é produzida em células HeLa, a qual foi mostrada ser passível de hipusinação. Análises de bioinformática mostraram que a isoforma A possui alta probabilidade de direcionamento para mitocôndria. Assim, técnicas de fracionamento subcelular mostraram que a isoforma A co-purifica com a mitocôndria, diferentemente da isoforma B. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The eukaryotic translation initiation factor 5A (eIF5A) is a highly conserved protein in archaeas and eukaryotes. It is characterized by being the only protein known to present the amino acid residue hypusine, produced from a post-translational modification called hypusination, which is essential for its activity. Currently eIF5A is correlated with the beginning, elongation and termination of translation, and its function has been associated with different cellular, physiological and pathological processes. In humans, eIF5A is predicted to be generated from five transcriptional variants (A, B, C, D and X5). Variants B, C, D and X5 encode the 17 kDa canonical protein B, which has been studied in recent decades. However, variant A is expected to encode an alternative 20 kDa isoform (A isoform), which has an additional 30 amino acid sequence in the N-terminal region. In this context, the present study aimed to characterize unpublished molecular events involving the human eIF5A1 isoform A. In this study, we showed that in HeLa cells, isoform A was detected, which was shown to be subject to hypusination. Bioinformatics analysis showed that isoform A has a high probability of targeting mitochondria. Avaliação da técnica de biopsia pulmonar transtoracica em ovinos clinicamente sadios e os efeitos do procedimento sobre o metabolismo oxidativo

Foram utilizados 20 ovinos clinicamente sadios e destinados ao abate. Inicialmente, foram coletadas amostras de sangue da veia jugular em tubos de ensaio contendo heparina. Após a colheita, foi realizado o exame clínico seguido da biopsia pulmonar com agulha semi-automática no 7º espaço intercostal direito cerca de 5 cm acima do olécrano. Avaliou-se o número de tentativas para a realização do procedimento e o tamanho dos fragmentos. As amostras obtidas foram analisadas histologicamente. Uma nova colheita de sangue foi realizada 30 minutos após a biopsia, e as amostras levadas ao laboratório para a detecção indireta dos radicais livres [superóxido dismutase (SOD), glutationa total (GSH-t) e peroxidase (GSH-Px) e substâncias reativas ao ácido tiobarbitúrico (TBARS)]. Após o abate foi realizado o exame pos mortem para avaliação de complicações da técnica e das eventuais lesões provocadas. Entre todos os animais submetidos à biopsia apenas dois demonstraram resistência à técnica, sendo obtidos fragmentos pulmonares que mediram entre 4 mm a 7 mm de comprimento, com média de 1,8 1 tentativas por fragmento. As principais alterações relacionadas à técnica foram: tosse, dispnéia inspiratória, elevação das freqüências cardíaca e respiratória e aumento do ruído broncobronquiolar. Ao exame pos mortem observou-se hemorragia dos músculos intercostais e pleura visceral de tamanho variado em todos os cordeiros, além de feridas lineares de até 14 mm nos dois animais que não reagiram bem ao procedimento. Das 20 tentativas de execução da técnica, 18 obtiveram sucesso, enquanto que em duas o fígado foi equivocadamente puncionado. As amostras de tecido pulmonar foram consideradas representativas... / It was evaluated the safety and effectiveness of the percutaneous transthoracic lung biopsy technique and its effects of the procedure on oxidative metabolism in sheep. It was used 20 clinically healthy sheep. Initially, blood samples were collected from the jugular vein in test tubes containing heparin. After collecting the blood, the clinical examination was carried out followed by lung biopsy with a semi-automatic needle biopsy. The number of attempts and size of the fragments were measured and the samples were analyzed histologically. New blood samples were collected 30 minutes after the biopsy. All blood samples were taken to the laboratory for the indirect detection of free radicals [superoxide dismutase (SOD), total glutathione (GSH-t), glutathione peroxidase (GSH-Px) and thiobarbituric acid reactive substances (TBARS)]. After the slaughter, a post mortem examination assessed the technical complications and possible injuries. Among all animals underwent biopsy only two did not tolerate the technique. It were obtained Lung fragments from 4 mm to 7 mm and it were required on average 1,8 attempts. The main changes related to the procedure were: cough, labored breathing, increased heart and breathing rate and increased broncobronquiolar noise. From 20 attempts to implement the technique, 18 were successful, while in two of them the liver was mistakenly punctured. Samples of lung tissue were representative. It was possible to visualize intact structures, including bronchioles and alveoli. The activity of SOD and GSH-Px decreased thirty minutes after the biopsy. Atividade funcional de polimorfonucleares do sangue de cabritos neonatos, induzida por Escherichia coli e Staphylococcus aureus \"in vitro\": influência etária e do manejo colostral / Functional activity of polymorphonuclear cells from the blood of goats neonates induced by Escherichia coli and Staphylococcus aureus "in vitro": influence of age and colostrum management.

O objetivo do presente estudo foi avaliar a atividade funcional dos polimorfonucleares (PMNs) do sangue de cabritos neonatos "in vitro" - metabolismo oxidativo e a fagocitose induzidos por S.aureus e E.coli, por meio da citometria de fluxo. Foram utilizados 32 cabritos da raça Saanen, acompanhados quanto ao estado de saúde do nascimento até os 15 dias de idade, distribuídos em quatro grupos: G1- colostro de cabra "in natura"; G2- colostro de cabra aquecido a 56ºC por 60 minutos; G3- colostro de vaca "in natura" e G4 - leite de cabra; sendo avaliados nos seguintes tempos: t0 (antes da ingestão do colostro); t1 (24-48h p.n.); t2 (72-96h p.n); t3 (168-192h p.n) e t4 (336-360h p.n). A avaliação estatística foi realizada pela análise de variância (ANOVA) para a comparação entre os grupos e intragrupos para amostras de pequeno tamanho, de acordo com as variáveis do estudo, com nível de significância de 5%. O metabolismo oxidativo basal não diferiu nos grupos e com a idade. O metabolismo oxidativo aumentou com o estímulo por S. aureus após a ingestão de colostro tratado pelo calor (G2), não apresentando diferenças nos demais grupos. O estímulo com E. coli o aumentou após a ingestão de colostro tratado (G2) e após a ingestão de leite de cabra (G4) a partir de t3, não diferindo nos grupos que receberam colostro de cabra e vaca "in natura" (G1 e G3 respectivamente). A comparação entre metabolismo oxidativo basal e induzido para o S. aureus foi maior antes e após a mamada do colostro nos grupos colostro de cabra "in natura" e sem colostro (G1 e G4); e, após a ingestão do colostro nos grupos que o receberam tratado pelo calor e de vaca respectivamente (G2 e G3). Para a E. coli a resposta foi maior após a ingestão de colostro tratado pelo calor (G2); antes e após a mamada de colostro de vaca (G3) ou leite (G4); não houve diferença entre o metabolismo basal e induzido para a E. coli no grupo que recebeu colostro de cabra "in natura" (G1). A fagocitose induzida pelo S.aureus não diferiu em nenhum dos grupos; o percentual de fagocitose aumentou após a ingestão de colostro de cabra "in natura" (G1) no momento t1. A fagocitose induzida pela E.coli foi maior após a ingestão de colostro de cabra nos dois grupos. O percentual de fagocitose foi maior após a ingestão do colostro tratado pelo calor e não diferiu nos demais grupos. Os resultados permitiram concluir que a ingestão do colostro não interferiu no metabolismo oxidativo basal; o desencadeamento da explosão respiratória de PMNs induzido pelas bactérias foi maior após a ingestão de colostro de cabra aquecido a 56ºC por 60 minutos; houve indícios de aumento da fagocitose para S. aureus e um aumento da intensidade de fluorescência e do percentual de fagocitose para E.coli nos grupos que receberam colostro de cabra (G1 e G2); não ficou esclarecida a influência da idade na atividade funcional de PMNs do sangue de cabritos neonatos "in vitro"; os resultados do presente estudo subsidiam propostas de melhoria do manejo da produção caprina baseada na utilização de colostro de cabra tratado pelo calor. / The aim of this study was to evaluate "in vitro" functional activities of neonates goats blood polymorphonuclear leukocytes (PMNs) respiratory burst and phagocytosis induced by S. aureus and E. coli, by flow cytometry. We used 32 Saanen goats, accompanied on the health status from birth to 15 days of age divided into four groups: G1-goat colostrum \"in nature\"; G2- goat colostrum heated to 56 ° C for 60 minutes; G3-cow colostrum \"in nature\" and G4 - goat milk, being evaluated at the following times: t0 (before intake of colostrum), t1 (24-48h after birth), t2 (72-96h a.b), t3 (168-192h a.b), t4 (336-360h a.b). Statistical analysis was performed by analysis of variance (ANOVA) for comparison between groups and within groups for samples of small size, according to the study variables, with 5% of significance. The respiratory burst did not differ in basal and age groups. The stimulation of respiratory burst in S. aureus increased significantly after ingestion of colostrum treated by heat (G2) and it was not different in the other groups. Stimulation with E. coli increased after colostrum treated by heat intake (G2) and after ingestion of goat\'s milk (G4) from t3; no difference in the groups that received colostrum from goats and cows \"in nature\" (G1 and G3 respectively). The comparison of basal and induced respiratory burst for S. aureus was higher before and after the intake of colostrum or goat milk in the groups colostrum \"in nature\" and without colostrum (G1 and G4) and, after ingestion of colostrum in the groups treated with heat and bovine colostrum, respectively (G2 and G3). For E. coli, response was higher after ingestion of colostrum treated by heat (G2), before and after feeding cow colostrum (G3) or goat milk (G4), there was no difference between basal metabolism and induced to E. coli in the group that received goat colostrum \"in nature\" (G1). Phagocytosis induced by S. aureus did not differ in any group, the percentage of phagocytosis increased after ingestion of goat colostrum \"in nature\" (G1) at time t1. Phagocytosis induced by E. coli was increased after colostrum intake in both groups that received goat colostrum. The percentage of phagocytosis was increased after ingestion of colostrum but it did not differ in treated and other groups. The results showed that ingestion of colostrum did not affect the basal respiratory burst; triggering the respiratory burst of PMNs induced by bacteria was greater after eating goat colostrum heated to 56 ° C for 60 minutes, there was evidence of increased phagocytosis of S. aureus and there was an increase in fluorescence intensity and percentage of phagocytosis of E.coli in the groups receiving goat colostrum (G1 and G2); it was not clarified the influence of age on functional activity of PMNs from the blood of goats neonates "in vitro"; and the results of this study subsidize proposals for improving the management of goat production based on the use of goats colostrum treated by heat.

Avaliação da resposta imune inata respiratória em bezerros sadios durante o segundo trimestre de vida / Evaluation of respiratory innate immune response in healthy calves during the second trimester

A idade entre o terceiro e o sexto mês de vida é um período peculiar para o estabelecimento da imunidade própria dos bezerros. Nesse intervalo, há susceptibilidade e índice de letalidade à broncopneumonias maiores, provavelmente devido à imaturidade do sistema imunológico pulmonar, quer seja por uma insuficiente resposta, nos primeiros momentos, quer seja por uma resposta citotóxica exagerada, no momento subsequente. A par disso, este trabalho teve o intuito de verificar o momento em que ocorre a maturidade do sistema imunológico, como se comportam os perfis Th1 e Th2 e a existência de uma resposta citotóxica exagerada, durante esta fase de estabelecimento da imunidade ativa dos bezerros. Para tal, estudaram-se as funções de fagocitose e metabolismo oxidativo de leucócitos sanguíneos e broncoalveolares, as classes de imunoglobulinas e citocinas incriminadas nos padrões de resposta linfocitária Th1 e Th2, em 10 bezerros da raça holandesa, sadios, avaliados em sete momentos experimentais, com intervalo quinzenal, entre o terceiro e o sexto mês de vida. Os dados foram submetidos à análise estatística, pela comparação entre as médias ou medianas, confrontadas pelo teste de Anova e Tukey, quando paramétricas, e pelo teste de Kruskal Wallis e Dunn, quando não paramétricas, considerando nível de significância P≤ 0,05 e tendência P≤ 0,10. Os dados que apresentaram dinâmicas semelhantes entre si foram submetidos ao teste de correlação de Pearson. Na região broncoalveolar, observaram-se um aumento progressivo das funções dos macrófagos alveolares, equilíbrio de secreção dos isotipos IgG1 e IgG2 e predominância de citocinas compatíveis com padrão de resposta Th1, até os 150 dias de vida dos bezerros. Aos 165 dias de vida, ocorreram diminuição da função celular, aumento dos títulos de IgG2 e aumento da citocina regulatória IL-10. Aos 180 dias, retornou-se o equilíbrio entre secreção de IgG1 e IgG2, diminuiram os teores de IL-10 e ocorreu tendência a aumento de IL-12, TNF-α e metabolismo oxidativo de macrófagos alveolares, o que permitiu concluir que a resposta imune tem característica própria, nesta faixa etária, e não se torna matura até os seis meses de vida. Apesar de os fagócitos pulmonares já estarem eficientes, a partir dos 135 dias de vida, tornam-se hiperresponsivos aos 150 dias de vida, momento em que gera consequentemente uma resposta regulatória e/ou humoral aos 165 dias de vida, para que aos 180 dias de vida, o equilíbrio entre os perfis Th1 e Th2 seja atingido / The age between the third and sixth month of life is a peculiar period for the establishment of active immunity of calves. There is a greater susceptibility and lethality by bronchopneumonia, probably due to the immaturity of the pulmonary immune system, whether by an insufficient response , in the first moments , whether by an exaggerated cytotoxic response in the subsequent time. So, this work aimed to verify when the maturity of the immune system occurs, how the Th1 and Th2 profiles behave and if there is an exaggerated cytotoxic response during this active phase for immunity of the calves without maternal interference. For this purpose we studied the functions of phagocytosis and oxidative metabolism of blood and bronchoalveolar leukocytes, classes of immunoglobulins and cytokines incriminated in lymphocyte response patterns Th1 and Th2, in 10 holstein healthy calves. They were sampled every fifteen days, during the third until sixth month of life. Data were statistically analyzed by comparing the means or medians, confronted by ANOVA test and Tukey, when the data were parametric, and by Kruskal Wallis and Dunn's test, when the data were nonparametric, level of significance p ≤ 0.05 and trend p ≤ 0.10. The data that showed similar dynamics between them were subjected to Pearson correlation test. In bronchoalveolar region, until 150 days of age, the alveolar macrophages functions increased progressively, the IgG1 and IgG2 isotypes secretion showed a balance, and the cytokines profile were compatible with Th1 response. At 165 days of age, there was a decrease of cellular function, an increased of IgG2 titers and the IL-10 secretion, a regulatory cytokine, increased. At 180 days of life, we observed a balance of IgG1 and IgG2 secretion, a decreased of IL-10 levels and a tendency to increase IL-12, TNF-α and alveolar macrophage oxidative metabolism. These results indicated that the calves have an active immune response with particularities for this age group and it does not become mature until six months of life. Despite of the macrophages alveolar are already efficiency from the 135 days of age, they become more reactive at 150 days. After this moment, a regulatory and/or humoral response begins at 165 days of life, as the balance of Th1 and Th2 profiles are reached at 180 days of calves life

O sistema imune inato é organizado por processos complexos que envolvem os diversos tipos celulares que culminam na eliminação de uma variedade de microorganismos a fim de proteger o organismo de infecções. Os neutrófilos são células importantes neste sistema, sendo capazes de migrar rapidamente ao foco da infecção. Estas células reconhecem e fagocitam partículas estranhas e atuam no sentido de elimina-los através da produção de espécies reativas de oxigênio (EROs), proteínas antimicrobianas e enzimas proteolíticas dos grânulos, que são liberadas no fagossomo e no espaço extracelular. Entretanto, em algumas doenças inflamatórias e autoimunes têm sido encontradas lesões nos tecidos envolvendo a produção excessiva de EROs pelos neutrófilos, desencadeada por imunocomplexos (ICs) via receptores Fc gama (Fc) e de complemento (CR). Assim, a modulação da ativação destes receptores e da produção de EROs é importante na manutenção da homeostasia. Nas últimas décadas, flavonóides são considerados como metabólitos secundários promissores mostrando atividade antioxidante e imunomodulatória. Neste trabalho foi avaliado o efeito modulatório dos flavonóides galangina, kaempferol, quercetina e miricetina em algumas funções efetoras de neutrófilos humanos, como o metabolismo oxidativo estimulado via receptores de membrana Fc e/ou CR, fagocitose e atividade microbicida, além do efeito citotóxico, antioxidante e inibitório dessas substâncias sobre a atividade das enzimas mieloperoxidase (MPO) e NADPH oxidase. Os flavonóides analisados mostraram um efeito modulatório negativo no metabolismo oxidativo dos neutrófilos humanos na seguinte ordem: galangina> kaempferol> quercetina > miricetina. Embora a galangina tenha exercido maior atividade, sua ação antioxidante e inibitória na atividade das enzimas MPO e NADPH oxidase foi a menos efetiva dentre os flavonóides avaliados. Além disso, esta substância inibiu de maneira mais acentuada a resposta celular mediada por ambos os receptores Fc e CR quando comparado a estimulação destes receptores individualmente. As substâncias contendo o grupo catecol, quercetina e miricetina mostraram elevada eficiência na inibição da atividade das enzimas NADPH oxidase e MPO, e na redução do radical livre 2,2-difenil-1-picrilhidrazil (DPPH). De maneira geral, os flavonóides não interferiram no processo fagocítico e no mecanismo microbicida, e não induziram toxicidade e morte celular por apoptose ou necrose. Os resultados apresentados neste trabalho mostram uma possível aplicação destes flavonóides como fármacos de origem natural em doenças inflamatórias que apresentem acúmulo de neutrófilos, sendo uma ferramenta promissora para o tratamento efetivo destas doenças. / The innate immune system is composed of complex biological processes involving a variety of cell types that eliminate invading microorganisms to protect the host against infection. Neutrophils are key effector cells of this system, being able to migrate rapidly to infection sites. These cells recognize and engulf foreign particles by phagocytosis, and act to destroy them through production and release of reactive oxygen species (ROS), antimicrobial proteins and proteolytic granule enzymes, which are delivered to the phagossomes and to the extracellular environment. However, tissue damage related to excessive neutrophil ROS production triggered by immune complexes (ICs) via Fc-gamma (FcR) and complement receptors (CR) has been found in some inflammatory and autoimmune diseases. So, modulation of both the activation of these receptors and ROS generation are important to the maintenance of body homeostasis. In the last decades, flavonoids have been considered promising plant secondary metabolites with antioxidant and immunomodulatory activity. This study evaluated the modulator effect of four flavonoids (galangin, kaempferol, quercetin and myricetin) in some human neutrophil effector functions, such as the oxidative metabolism stimulated via FcR and CR classes of membrane receptors, phagocytosis and microbicidal activity, as well as the cytotoxic, antioxidant and inhibitory effect of these compounds in the MPO and NADPH oxidase activity. The flavonoids studied showed a negative modulator effect in the human neutrophils oxidative metabolism in the following ranking order: galangin > kaempferol > quercetin > myricetin. Although galangin had shown the highest activity, it was the least effective antioxidant and inhibitor of MPO and NADPH oxidase activity among the flavonoids tested herein. Furthermore, this compound inhibited more the cellular response mediated by both Fc and CR receptors than by each one of them individually. The flavonoids bearing the catechol group, quercetin and myricetin, had a high efficiency to inhibit NADPH oxidase and MPO activity, as well as to reduce the 2,2-diphenyl-1-picrylhidrazil (DPPH) free radical. In general, the flavonoids did not interfere in the phagocytic process and microbicidal mechanism, and also did not induce toxicity and cell death by apoptosis or necrosis. The group of results presented here provides a possible application of these flavonoids as drugs of natural source in inflammatory diseases with excessive neutrophil infiltration, being a promising tool for an effective treatment of these illnesses.

Adaptação mitocondrial induzida pelo exercício físico aeróbio: desvendando novos mecanismos moleculares / Aerobic exercise-induced mitochondrial adaptation: unraveling novel molecular mechanisms

O aumento da capacidade oxidativa é considerado o fator central dos seus benefícios à saúde induzidos pelo exercício físico aeróbio (EFA). A musculatura esquelética é um dos tecidos mais envolvidos na realização de exercícios físicos, com capacidade notável de adaptação metabólica e estrutural frente ao estímulo mecânico. Os músculos esqueléticos são ricos em mitocôndrias e altamente dependentes da fosforilação oxidativa para a produção energia. Assim, o aumento da capacidade aeróbia induzido pelo EFA ocorre grande parte em função de adaptações mitocondriais. Inúmeros estudos demonstram a capacidade do EFA em induzir biogênese mitocondrial, onde o coativador de transcrição PGC-1?1 atua coordenando a expressão de genes nucleares e mitocondriais no contexto do EFA. No entanto, animais com deleção de PGC- 1?1 no músculo esquelético ainda apresentam remodelamento mitocondrial importante após período de treinamento físico aeróbio, evidenciando a existência de mecanismos ainda desconhecidos. Embora a formação de novas mitocôndrias seja fundamental, a manutenção de mitocôndrias saudáveis por meio de mecanismos de controle de qualidade parece ser de igual ou maior importância para uma adaptação mitocondrial adequada. Um desses mecanismos de controle de qualidade mitocondrial envolve a remoção de mitocôndrias danificadas/envelhecidas via autofagia mitocondrial (mitofagia). Contudo, os mecanismos envolvidos na mitofagia induzida pelo EFA são pouco conhecidos. Considerando o papel da adaptação mitocondrial sobre os efeitos benéficos do EFA, realizamos um estudo exploratório para buscar novos mecanismos envolvidos neste processo. Para isso, utilizamos uma abordagem proteômica direcionada à fração mitocondrial muscular de camundongos submetidos a uma única sessão de EFA. Num primeiro estudo, utilizamos os resultados de proteômica para procurar por proteínas envolvidas na ativação da mitofagia durante o EFA. A partir desse estudo, verificamos que uma sessão de EFA de fato induz sinais de mitofagia na musculatura esquelética. Além disso, propomos que as proteínas Phb2 e Mief2 podem acumular em mitocôndrias danificadas durante o EFA e colaborar para o recrutamento da maquinaria autofágica para a organela, auxiliando no controle de qualidade e adaptação mitocondrial induzidos pelo EFA. Em segundo estudo, tivemos como objetivo identificar nos resultados de proteômica possíveis reguladores de transcrição gênica envolvidos na adaptação mitocondrial induzida pelo EFA. Dessa maneira, identificamos que a proteína mitocondrial Spryd4, cuja função não havia sido estudada até então, parece aumentar na fração mitocondrial muscular durante o EFA. Observamos ainda que a expressão gênica muscular de Spryd4 diminui em camundongos idosos ou com distrofia muscular, aumenta em animais saudáveis após treinamento físico aeróbio e também parece aumentar em humanos treinados. In vitro, observamos que a atenuação da expressão de Spryd4 em miotubos primários promove disfunção mitocondrial, associada à diminuição da expressão de genes de complexos mitocondriais e envolvidos no transporte e metabolismo de lipídeos, além de promover atrofia de miotubos. Num contexto geral, a análise do proteoma mitocondrial muscular após uma sessão de EFA nos permitiu identificar proteínas que parecem estar envolvidas em adaptações mitocondriais, em especial, em mecanismos de mitofagia e controle do fluxo de substratos energéticos / Increased oxidative capacity induced by regular aerobic exercise (AE) is considered a major factor in health. Skeletal muscle is one of the most compromised tissues during exercise and has remarkable metabolic and structural plasticity upon mechanical stimuli. Muscles are rich in mitochondria and heavily reliant on oxidative phosphorylation for energy production. Thus, increased aerobic capacity induced by regular AE occurs largely due to mitochondrial adaptations. Many studies have shown that AE is able to induce mitochondrial biogenesis, and the transcription coactivator PGC-1?1 is known to coordinated gene expression both in nuclei and mitochondria. However, muscle-specific PGC-1?1 knockout mice still display major mitochondrial remodeling after AE training, supporting the existence of unknown mechanisms of AE-induced mitochondrial adaptations. Although making new mitochondria is crucial, the maintenance of a healthy pool of this organelle through mechanisms of quality control seems to be of equal or greater importance during mitochondrial adaptation. One mechanism for mitochondrial quality control comprises the removal of damaged/aged mitochondria via autophagy (mitophagy). Nonetheless, the mechanisms of AE-induced mitophagy are poorly understood. Given the importance of mitochondrial adaptation to the health benefits of AE, we conducted an exploratory study to uncover new mechanisms underlying this process. For this, we performed a proteomic analysis in the skeletal muscle mitochondria-enriched fractions of mice submitted to a single bout of AE. In a first study, we have used these proteomics data to seek for proteins that might be involved in AE-induced mitophagy. On this matter, we confirmed that a single bout of AE in mice increases skeletal muscle mitophagy signaling. Additionally, we suggest that Phb2 and Mief2 accumulate in damaged mitochondria in skeletal muscle during AE and might assist in the recruitment of the autophagic machinery to the organelle, thus aiding to mitochondrial quality control and AE-induced mitochondrial adaptation. In a second study, we have used the same proteomics data to identify transcriptional regulators that might have a role in AE-induced mitochondrial adaptation. Thereby, we have found that the mitochondrial protein Spryd4, whose function was so far unknown, seems to increase in the skeletal muscle mitochondria-enriched fractions following AE. Here we show that skeletal muscle Spryd4 gene expression decreases in aged and dystrophic mice, increases in healthy trained animals and also seems to increase in trained humans. In vitro, we have seen that Spryd4 loss of function in primary myotubes promotes mitochondrial dysfunction, decreases expression of genes involved in mitochondrial complex function, as in fatty acid oxidation and transport. Indeed, Spryd4 loss of function promoted myotube atrophy. Taken together, by analyzing the skeletal muscle mitochondrial proteome after a single bout of AE in mice, we have identified proteins that might participate in AE-induced mitophagy and substrate metabolism, and thus in skeletal muscle adaptation to AE

O objetivo foi avaliar a atividade funcional dos polimorfonucleares (PMN) do sangue de bezerros neonatos \"in vitro\" - metabolismo oxidativo induzida por: S. aureus, E. coli e PMA e fagocitose para S. aureus e E. coli, em citômetro de fluxo. Quinze animais avaliados nos tempos: t0 (antes da ingestão de colostro/leite); t1(até 48 hs. p.n); t2 (48 - 96 hs. p.n.); t3 (96 - 144 hs. p.n.); t4 (144-192 hs. p.n.); t5 (192-240 hs.p.n.) receberam quatro litros (grupo 1 - G1), dois litros de colostro ( grupo 2 - G2) e somente leite (grupo 3 - G3). O metabolismo oxidativo basal não diferiu em nenhum dos grupos e idades. Entre os grupos este foi menor em G1 do que em G3 (t3). Induzido pela S. aureus foi maior entre t1 e t3 (grupo 1), entre t2 e t3 (grupo 2) e sem diferenças (grupo 3). Entre os grupos G1 foi maior que G3 (t1). Induzido pela E.coli foi maior entre t1 e t3 (grupo 1), maior em t2 (grupo 2) e sem diferenças (grupo 3). Entre os grupos G1 foi maior que G2 e G3 (t1). O PMA estimulou a explosão respiratória em G1, G2 e G3 entre t0 e t5. Entre os grupos G1 foi menor para do que G2 e G3. A comparação entre metabolismo basal e induzido para G1 foi maior para PMA e S. aureus entre t0 e t5 e para E. coli entre t1 e t5; para G2 foi maior para o PMA entre t0 e t5, para S. aureus entre t0 e t3 e para a E. coli entre t2 e t4; para G3 foi maior para o PMA e S.aureus entre t0 e t5 e para a E. coli entre t3 e t5. A fagocitose induzida pela S. aureus foi maior nos tempos t1, t2 e t4 (grupo1) e o percentual de fagocitose foi maior após t0; para o grupo 2, foi maior em t2 e t3 e o percentual foi maior após t0; para o grupo 3, não diferiram entre os tempos. Entre os grupos G1 foi maior que G2 e G3 (t4) e o percentual foi maior para G1 e G2 (t3) do que G3 e maior para G1 do que G2 (t5). A fagocitose induzida pela E. coli e o percentual de fagocitose não diferiram em nenhum grupo. Entre os grupos G1 foi maior do que G3 (t0) e o percentual foi maior para G1 do que G2 (t2). Conclusão: o colostro não interferiu no metabolismo oxidativo basal e estimulado pelo PMA, a S. aureus estimulou o metabolismo oxidativo e a fagocitose antes e após a ingestão de colostro. A E. coli não induziu o metabolismo oxidativo (G3) e não foi eficiente para estimular a fagocitose. Não ficou esclarecida a influência da idade sobre a atividade dos PMNs. O fornecimento de quatro litros de colostro melhora resposta funcional dos PMNs para as bactérias. / The aim of this study was to evaluate \"in vitro\" functional activities of neonatal calves blood polymorphonuclear leukocytes (PMNs) - respiratory burst and phagocytosis induced by S.aureus, E.coli and PMA, measured by flow cytometry. Fifteen calves were tested at times: t0 (before milk/colostrum ingestion); t1 (until 48 hours post partum); t2 (48-96 hrs p.p.); t3 (96 - 144 hrs. p.p.); t4 (144-192 hrs. p.p.); t5 (192-240 hrs. p.p.) and the groups intakes were 4 liters of colostrum (group 1), 2 liters of colostrum (group 2) and only milk (group 3). The basal respiratory burst did not differ among groups but it was lower in G1 than G3 (t3). The burst induced by S.aureus was higher between t1 and t3 (G1) and t2 and t3 (G2) and showed no difference (G3). The G1 values were higher than G3 (t1). E.coli induced higher values between t1 and t3 (G1), higher on t2 (G2) and showed no difference (G3). Among groups G1 was higher than G2 and G3 (t1). The PMA induced the respiratory burst in G1, G2 and G3 between t0 and t5. Among groups G1 was lower in G2 than G3. The induced basal metabolism was compared and G1 was higher to PMA and S.aureus between t0 and t5 and to E.coli between t1 and t5. The G2 showed higher values to PMA between t0 and t5, to S.aureus between t0 and t3 and E.coli between t2 and t4. The G3 showed higher values in PMA and S.aureus between t0 and t5 and E.coli between t3 and t5. The S.aureus induced phagocytosis was higher on t1, t2 and t4 (G1) and the phagocystosis percentage was higher after t0. G2 groups showed higher values on t2 and t3 and higher percentage after t0. G3 had no difference on times. G1 showed higher values than G2 and G3 (t4) and G1 and G2 (t3) had higher percentage than G3 with G1 higher than G2 (t5). The E.coli induced phagocytosis and its percentage did not differ among groups. G1 was higher than G3 (t0) and G1 showed higher phagocytosis percentage than G2 (t2). Conclusion: the colostrum did not affect either, the basal respiratory burst and the one stimulated by PMA. S. aureus stimulated the respiratory burst and phagocytosis before and after the colostrum intake. E.coli did not induce the respiratory burst (G3) and did not stimulate the phagocytosis. The age influence in PMNs activity was not totally clarified. The intake of 4 litters of colostrum increases the PMNs activity against the species of bacteria studied.

Dinâmica da resposta imune inata do sistema respiratório de bezerros / Dynamic of the innate immune response of the respiratory system in calves

As influências etárias do sistema imune de bezerros são descritas na primeira fase de vida desses animais e a hipótese de também ocorrerem ariações nos principais mecanismos de resposta inata do pulmão pode identificar períodos de maior suscetibilidade às principais doenças respiratórias que acometem os bezerros nesse período. Com a finalidade de minimizar os prejuízos econômicos associados às doenças respiratórias em bezerros, este estudo objetivou avaliar a dinâmica imunológica inata do sistema respiratório de bezerros sadios nos três primeiros meses de vida, no qual nove bezerros sadios foram acompanhados por três meses e submetidos a oito avaliações imunológicas. O material recuperado do lavado broncoalveolar colhido por broncoscopia foi submetido à avaliação funcional dos macrófagos alveolares utilizando as provas de fagocitose (SaPI e E.coli), burst oxidativo, quantificação de imunoglobulinas e expressão de CD14. Os dados foram avaliados pelo teste ANOVA oneway (unstacked) (paramétricos) e pelo teste Mann-Whitney (não paramétricos). Verificaramse alterações funcionais de fagócitos CD14+, que apesar de se manterem constantes em seus valores relativos durante todo o período, apresentou intensidade de fagocitose elevada pontual na terceira semana de vida e um aumento da fagocitose por mononucleares CD14+ aos 45 dias de idade com diminuição da intensidade da fagocitose por essas mesmas células a partir dessa idade. Conclui-se que a partir de 45 dias de vida os animais começam a montar uma resposta imune própria, porém pontual e que até os 90 dias não atingem a estabilidade necessária para atestar a conclusão do processo de maturação da resposta inata local. / The influences of age in calves\' immune system are described in their first phase of life. The hypothesis that variations occur in the main mechanisms of lung innate response can help to identify periods of greater susceptibility to the respiratory diseases that affect calves in the first stage of their life. With the purpose of minimizing the economic losses associated with respiratory disease in calves, this study aimed to evaluate the innate immune dynamics of the respiratory system of healthy calves in the first three months of life. Nine healthy calves were monitored for three months and eight immunologic evaluations were performed. Bronchoalveolar lavage samples were recovered by bronchoscopy. Then, the alveolar macrophages in samples were identified by protein expression of CD14 and undergone functional evaluation of phagocytosis (SAPI and E.coli) and oxidative burst. Immunoglobulin were also quantified in samples. Data was assessed by one-way ANOVA (unstacked) (parametric) and the Mann-Whitney test nonparametric). Functional alterations in phagocytes CD14 + were observed, and although their relative values were kept throughout the period, higher intensity of phagocytosis in the third week and increased phagocytosis by macrophages CD14 + at 45 days of life was observed. Decreased intensity of phagocytosis was observed after this age. It is concluded that from 45 days of life on, calves began to maintain their immune response, but until 90 days of life they did not achieve the stability to conclude the maturation of local innate response.

Os neutrófilos representam a primeira linha de defesa do hospedeiro, atuando na contenção e eliminação de patógenos. Contudo, alterações na vida média, no excessivo recrutamento e ativação dos neutrófilos estão associados a danos teciduais e a doenças inflamatórias e autoimunes. A modulação das funções efetoras dos neutrófilos pode auxiliar no tratamento de tais patologias. Neste sentido, os produtos naturais constituem uma importante fonte de novas substâncias imunomoduladoras. Um estudo recente demonstrou que, a inibição do metabolismo oxidativo de neutrófilos pelo extrato etanólico bruto das folhas de Baccharis dracunculifolia (EEBBd) correlaciona-se com a proporção entre ácido cafeico (CaA) e outros compostos fenólicos contidos nesta amostra. Para dar prosseguimento à investigação do potencial imunomodulador do EEBBd e do CaA, os objetivos do presente estudo foram avaliar o efeito modulador desses produtos naturais: (i) em três funções efetoras de neutrófilos humanos - fagocitose, atividade microbicida e metabolismo oxidativo estimulado por agentes independentes de receptores (forbol-12-miristato-13-acetato; PMA) e dependentes apenas de receptores Fcgama (imunocomplexos não-opsonizados; IC) ou de receptores Fcgama associados a receptores do complemento (imunocomplexos opsonizados com complemento; IC-SHN); (ii) na atividade da mieloperoxidase (MPO); (iii) na expressão de receptores de membrana; (iv) e na captura (scavenger) de H2O2 e HOCl. O CaA foi mais efetivo do que o EEBBd em inibir a atividade da MPO e em capturar H2O2 e HOCl. A análise in silico revelou que o CaA bloqueia a entrada do sítio ativo da MPO através da interação com os resíduos Gln-91, His-95 e Arg-239; os dois últimos resíduos são essenciais para clivar o H2O2 e para estabilizar o sítio ativo, respectivamente. A eficiência dos agentes utilizados para estimular o metabolismo oxidativo, medido por quimioluminescência dependente de lucigenina e de luminol, ocorreu na seguinte ordem: PMA > IC-SHN > IC. Embora o PMA tenha sido o agente mais efetivo em estimular o metabolismo oxidativo, ambas as amostras (EEBBd e CaA) inibiram com maior intensidade esta função celular estimulada por PMA do que a mesma função estimulada por IC-SHN e IC. Além disso, ambas, EEBBd e CaA, não alteraram os níveis de expressão dos receptores TLR2, TLR4, CD16, CD32, e CD11b/CD18. Nas maiores concentrações avaliadas, EEBBd (50 ug/mL) e CaA (90 ug/mL) não foram citotóxicos para os neutrófilos. O EEBBd inibiu intensamente a capacidade fagocítica e reduziu discretamente a capacidade microbicida dos neutrófilos frente à Candida albicans. Portanto, o CaA contribui para ação inibitória do EEBBd no metabolismo oxidativo e na atividade da MPO, mas não na capacidade fagocítica e microbicida de neutrófilos. Por fim, o efeito imunomodulador do CaA e do EEBBd não é mediado por alterações na viabilidade celular ou na expressão de receptores de membrana em neutrófilos. O conjunto de resultados obtidos pode auxiliar na elucidação do mecanismo de ação destes produtos naturais sobre as funções efetoras de neutrófilos, bem como no desenvolvimento de novos fármacos para o tratamento de doenças inflamatórias mediadas pela ativação exacerbada de neutrófilos. / Neutrophils represent the first line of host defense that acts in the containment and clearance of pathogens. However, changes in life span and excessive recruitment and activation of neutrophils are associated with tissue damage and inflammatory and autoimmune diseases. Modulation of the effector functions of neutrophils can help to treat these pathologies. In this sense, natural products constitute an important source of novel immunomodulating compounds. A recent study has demonstrated that the neutrophil oxidative metabolism inhibition by the crude ethanol extract of Baccharis dracunculifolia (EEBBd) leaves correlates with the ratio of caffeic acid (CaA) to other phenolic compounds that exist in it. To continue investigating the immunomodulating potential of EEBBd and CaA, the present study aimed to examine whether these natural products modulate: (i) three effector functions of human neutrophils phagocytosis, microbial killing, and oxidative metabolism elicited by a receptor-independent stimulus (phorbol-12-myristate-13-acetate; PMA) and by receptor-dependent stimuli that bind Fcgama receptors alone (non-opsonized immune complexes; IC) or in combination with complement receptors (complement-opsonized immune complexes; IC-SHN); (ii) myeloperoxidase (MPO) activity; (iii) expression of membrane receptors; (iv) H2O2 e HOCl scavenging. CaA was more effective than EEBBd in inhibiting MPO activity and scavenging H2O2 and HOCl. The in silico analysis revealed that CaA blocks the entrance of the active site of MPO through the interaction with Gln-91, His-95, and Arg-239; the two last residues are essential to cleave H2O2 and stabilize the active site, respectively. The agents used to stimulate the oxidative metabolism, as measured by the lucigenin- and luminol-dependent chemiluminescence assays, acted in the following ranking order of efficiency: PMA > IC-SHN > IC. Although PMA was the most efficient agent at stimulating the neutrophil oxidative metabolism, both samples (EEBBd and CaA) suppressed the PMA-induced oxidative metabolism more effectively than they suppressed the same cell function elicited by IC-SHN and IC. Both EEBBd and CaA did not alter the levels of TLR2, TLR4, CD16, CD32, and CD11b/CD18 receptors expression. At the highest concentrations tested, EEBBd (50 ug/mL) and CaA (90 ug/mL) were not toxic towards neutrophils. EEBBd strongly diminished the phagocytic capacity and slightly reduced the Candida albicans killing ability of neutrophils. In conclusion, CaA contributes to the EEBBd inhibitory action on the oxidative metabolism and MPO activity but not on the phagocytic capacity and microbial killing ability of neutrophils. Furthermore, the immunomodulating effect of CaA and EEBBd is not mediated by alterations in either cell viability or expression of membrane receptors in neutrophils. Together, the results can help to unravel the mechanism of action of these natural products on the effector functions of neutrophils, and to develop new drugs to treat inflammatory diseases mediated by neutrophil overactivation.
