HSP90 are ATP-dependent chaperones responsible for the activation and stabilization of more than 200 proteins. Most of these proteins are connected to cancer and need HSP90 to execute their activities. Thus, HSP90 inhibitors may indirect inhibit plenty oncogenic proteins. According to the literature, there are N-terminal and C-terminal domains, nevertheless, C-terminal inhibitors present more advantages. Then, the advancement of C-terminal inhibitors represents an applicable alternative and an intense field of study. Novobiocin is the first compound identified as C-terminal domain inhibitor. Nowadays, Novobiocin analogues have been synthesized, besides its structure-activity relationship elucidated in order to develop more potents analogues. For that reason, the goal of this project is to obtain 2 analogue series of Novobiocin. The scheme of these series was based on maintaining the coumaniric part presented on Novobiocin: isosteric replacement of the amide group that binds coumarin to the 4-hydroxy-3-(3-methylbut-2-yl)-benzamide, by the [1,2,3]-triazole; and exploration of methyl groups (AN1) and hydroxyl (AN2) in substitution of subunit noviose. AN1 series (76a-g) and AN2 (77a-g) were synthesized from the followed reactions: 7-hydroxy-coumarine o-methylation (83); 7-methoxy-coumarine 3-bromation (82); Sonogashira cross-coupling from 3-bromo-7-R-coumarine (81a-b); 7-R-3-((trimethylsilyl)ethynyl-coumarine trimethylsilyl deprotection (88a-b); and lastly, synthesis of the triazole ring, through cicloaddition reaction, 1,3-dipolar catalysed by copper (CuAAc), using 3-ethynyl-7-R-coumarine (80a-b) and aromatic azides. It is worth taking into consideration that 3-bromo-7-R-coumarine (81b) was obtained from the key compound, 3-bromo-7-R-methoxy-coumarine (81a) by o-demethylation reaction. 12 compounds were obtained (76a-g) and 8 of them (76a-b, 77d) were nicely purified and then, characterized by spectroscopic techniques (IR, 1H and 13C NMR). The ?one-pot? synthesis of 7-methoxy-3-(1H-1,2,3-triazole-4-phenyl)-coumarine 76b from 88a compound (Yield=86%) was more efficient than the one realized in two steps (global yield=47,2%). Results are potential and satisfactory, moreover there is no citation in any literature about biological activities of the synthesized compounds. Continuing this work, the pure compounds (76a-b, 77d) will be tested on breast cancer cell lines SkBr3 and MCF-7. / O c?ncer ? um termo gen?rico que se refere a um conjunto de mais de 100 tipos de doen?as, as quais se assemelham pelo crescimento celular desordenado. As HSP90 s?o chaperonas ATP-dependente respons?veis pela ativa??o e estabiliza??o de mais de 200 prote?nas. Muitas destas prote?nas s?o relacionadas ao c?ncer e necessitam da HSP90 para exercerem suas atividades. Assim, inibidores da HSP90 s?o promissores, pois possibilitam inibir de maneira indireta v?rias prote?nas oncog?nicas simultaneamente. S?o descritos inibidores dos dom?nios N-terminal e C-terminal, sendo estes mais vantajoso que aqueles. O Novobiocin foi o primeiro composto identificado como inibidor do dom?nio C-terminal da HSP90. Atualmente, diversos an?logos ao Novobiocin tem sido sintetizados, afim de se estabelecer a rela??o estrutura-atividade, objetivando desenvolver an?logos mais potentes. Assim, o objetivo deste trabalho foi obter duas s?ries an?logas ao Novobiocin. O planejamento das s?ries foi baseado na manuten??o do n?cleo cumar?nico presente no Novobiocin; troca isost?rica do grupo amida, que liga a cumarina ao anel 4-hidroxi-3-(3-metillbut-2-il)-benzamida, pelo anel [1,2,3]-triazol; e explora??o de grupos metoxila (AN1) e hidroxila (AN2) em substitui??o ? subunidade noviose. As s?ries AN1 (76a-g) e AN2 (77a-g) foram sintetizadas a partir das rea??es de: O-metila??o da 7-hidroxi-cumarina (83); broma??o da posi??o 3 da 7-met?xi-cumarina (82); rea??o de acoplamento cruzado de Sonogashira, a partir da 3-bromo-7-R-cumarina (81a-b); desprote??o do grupamento trimetilsilila da 7-R-3-((trimetilsilil)etenil)-cumarina (88a-b); e por fim, s?ntese do anel triaz?lico, atrav?s da rea??o de cicloadi??o 1,3-dipolar catalisada por cobre (CuAAc), utilizando 3-etenil-7-R-cumarina (80a-b) e azidas arom?ticas. Vale ressaltar que, a 3-bromo-7-hidr?xi-cumarina (81b) foi obtida a partir do composto chave, 3-bromo-7-met?xi-cumarina (81a) via rea??o de O-demetila??o. Obteve-se 12 compostos (76a-g e 77a-g) sendo que 8 (76a-b, 77d) foram, satisfatoriamente, purificados e, ent?o, caracterizados por t?cnicas espectrosc?picas (IV, RMN H e C). Tamb?m foi constatado que a s?ntese ?one-pot? de 7-metoxi-3-(1H-1,2,3-triazol-4-fenil)-cumarina 76b partir do composto 88a (R=86%) foi mais eficiente que a realizada em duas etapas, desilina??o/CuAAc (Rglobal= 47,2%). Os resultados s?o satisfat?rios e promissores, al?m disso n?o h? na literatura descri??o das atividades biol?gica dos compostos sintetizados. Em continua??o a este trabalho, os compostos puros (76a-b, 77d) ser?o devidamente ensaiados frente a c?lulas das linhagens de c?ncer de mama SkBr3 e MCF-7.

The best condition observed was under reflux for 30 hours and stoichiometric ratio 2: 1: 1 for Mg+2, Al+3 and GLYP, as indicated by the characteristic bands of LDH intercalated with glyphosate and no other bands present, which could be assigned to other intercalated ions or to free glyphosate. Under those preparation conditions the LDH interlamellar spacing was 0.62 nm, compatible with the intercalation of glyphosate. The controlled release of glyphosate was studied using ion exchange reactions of Layered Double Hydroxides containing glyphosate with an aqueous medium containing carbonate, chloride or nitrate anions. In those cases the glyphosate was quantified spectrophotometrically by Ruhemann's purple method (?max = 570 nm), and the initial releasing rates of 0.65 mg/ml/h (carbonate), 0.14 mg/ml/h (nitrate) and 0.08 mg/ml/h (chloride) were measured. Regarding the XRD patterns of the ion-exchanged products, it was observed that the structure of HDL was maintained, but with the reduction of the interlayer spacing in all cases, having a sufficient size for the anion of the involved ion exchange, but not enough for the presence of GLYP, demonstrating so that the exchange was successful. / Neste trabalho foram preparados por coprecipita??o e caracterizados por DRX de p? e FTIR compostos do tipo hidrotalcita, conhecidos como Hidr?xidos Duplos Lamelares (HDL), intercalados com o ?nion glifosato (GLYP), e foram estudadas as melhores condi??es de prepara??o e a raz?o estequiom?trica Mg:Al:GLYP. A melhor condi??o foi sob refluxo por 30 horas e raz?o estequiom?trica 2:1:1 para Mg+2, Al+3, e GLYP, a partir da observa??o das bandas caracter?sticas no espectro de FTIR de um HDL intercalado com glifosato, sem outras bandas presentes que pudessem ser atribu?das a outros ?ons intercalados ou ao glifosato livre. Nessas condi??es de preparo o espa?amento interlamelar do HDL foi de 0,62 nm, compat?vel com a intercala??o do glifosato. A libera??o controlada do glifosato foi estudada atrav?s de rea??es de troca i?nica dos Hidr?xidos Duplos Lamelares contendo glifosato com um meio aquoso contendo os ?nions carbonato, cloreto ou nitrato. Nestes casos o glifosato foi quantificado espectrofotometricamente pelo m?todo do p?rpura de Ruhemann (?max = 570 nm), e foram obtidas as taxas de libera??o iniciais de 0,65 mg/ml/h para carbonato, 0,08 mg/ml/h para cloreto e 0,14 mg/ml/h para nitrato. Nos difratogramas de raios-X dos produtos das trocas i?nicas, em todos os casos foram observadas a manuten??o da estrutura do HDL e a diminui??o do espa?o interlamelar, de 0,62 nm para valores entre 0,33 e 0,35 nm, tamanho suficiente para o ?nion da troca i?nica envolvido, mas n?o suficiente ainda para a presen?a de GLYP, demonstrando assim que a troca foi bem sucedida.

Metab??litos secund??rios com potencial farmacol??gico, incluindo fisalinas que exibem atividades anti-inflamat??rias, imunomoduladoras e antiparasit??rias, foram identificados nesta esp??cie. Poucos estudos investigaram os locais de armazenamento de metab??litos secund??rios na planta e o efeito dos bioativos na linfoprolifera????o causada pelo v??rus linfotr??pico de c??lulas T humanas (HTLV-1). Objetivos: Caracterizar as estruturas anat??micas vegetais, o perfil fitoqu??mico dos extratos e avaliar o efeito da Physalis angulata em linf??citos de indiv??duos infectados pelo HTLV-1 Material e m??todos: Foram empregadas t??cnicas de microscopia eletr??nica e ??ptica convencional para a caracteriza????o anat??mica dos ??rg??os da P. angulata (folha, raiz, caule e frutos). Extratos metan??licos de folhas, ra??zes, caule e frutos foram qualitativamente caracterizados pela presen??a de esteroides, terpenoides, taninos, alcaloides, saponinas, flavonoides, antraquinonas, cumarinas, teor de compostos fen??licos totais e conte??dos de flavonoides. A capacidade antioxidante desses extratos foi determinada pela atividade de sequestro de radicais livres do DPPH. Para avaliar o efeito do extrato da P. angulata e Fisalina F sobre a linfoprolifera????o, c??lulas de tr??s indiv??duos infectados pelo HTLV-1 e tr??s controles n??o infectados foram cultivadas em presen??a de dilui????es seriadas do extrato e da Fisalina F por 5 dias. As c??lulas foram pulsadas por 18h com 1??Ci 3H- Timidina. Al??m disso, foi realizado a an??lise do ciclo celular, na presen??a ou aus??ncia da Fisalina F e do extrato. Resultados: Os est??matos anisoc??ticos localizados na face abaxial foram mais abundantes que os est??matos na face adaxial. Tricomas foram evidenciados mais abundantes ao longo das nervuras do pec??olo, caules, al??m da margem nas s??palas e p??talas e dispersos no ov??rio. Esteroides e terpenoides estiveram presentes nas folhas, caules e frutos de P. angulata. Saponinas eram exclusivas nos frutos. A triagem fitoqu??mica n??o detectou flavonoides, antraquinonas e alcaloides em todas os extratos metan??licos das partes testadas da planta. As maiores capacidades antioxidantes foram identificadas nos extratos foliares e frut??feros, possivelmente devido ?? presen??a de compostos fen??licos nesses ??rg??os. O IC50 encontrado para Fisalina F foi 3,0 ??M e o IC50 do extrato etan??lico do caule de P. angulata foi de 7,0 ??g. A Fisalina F e o extrato da P. angulata inibiram a prolifera????o espont??nea de c??lulas infectadas de HTLV-1 e reduziram o n??mero de c??lulas em processo de mitose. Conclus??o: Este estudo descreveu as caracter??sticas anat??micas e bioqu??micas de P angulata e identificou ??rg??os do indiv??duo abundantes em antioxidantes (folhas e frutos) e esteroides (possivelmente fisalinas; folhas). Fisalina F e extrato de P. angulata apresentaram um efeito inibit??rio sobre c??lulas de indiv??duos infectados pelo HTLV-1, diminuindo a propor????o de c??lulas em divis??o celular.

A µCT Investigation of the Electrical Breakdown Mechanisms in Mica/Epoxy Machine Insulation / Undersökning av de elektriska nebrytningsmekanismerna i glimmer/epoxi-isolation

Mica/Epoxy machine insulation is a composite material of the mineral mica, fibre-glass mesh and epoxy resin. The material is used in motors and generators rated for high voltage, where the material must withstand multiple stresses such as elevated temperatures, mechanical and electrical stresses. This is a novel investigation that focuses on the breakdown mechanisms related to electrical stress of the insulation. These include partial discharges and electrical treeing. The micro Computed Tomography (μCT) uses a series of X-ray images to visualise the interior. This provides a method for non-invasively examination of the insulation for any signs of material degradation caused by high electrical fields. The following thesis presents an initial study on how this material system interacts with the μCT, the production of 16 samples capable of being electrically aged and scanned in the μCT, with minimal interference, sample preparation and an analysis of the images produced from the μCT. The samples produced are electrically aged at different voltages after which they are scanned with the μCT. The images produced are then analysed for signs of material degradation using different software. Some of the samples did have a complete electrical breakdown and an attempt of correlating these breakdowns to the amount of folds in the insulation is done. It is found that samples with an aluminium core are suitable for μCT imaging. The visualisation of defects such as voids, resin rich parts or folds in insulation is also possible. The detection of material degradation due to electrical aging is not clear enough to draw any conclusions although some possible signs are found. For samples that had an electrical breakdown the result is much clearer. It shows the breakdown channels path inside the insulation, moving in between the mica tape layers. The samples with a complete breakdown also had tendencies that the breakdown channels followed the resin rich part in the sample. Even if no signs of electrical treeing can be seen in the samples during aging, the breakdown channels visualised still show what path the final treeing did take and how it did so by moving around the mica tape.

The geology of the Miami Mica field (Urungwe District)

[Preface] The detailed geological survey of the country described in this bulletin commenced in 1953 and was completed in 1956 with the mapping of 1,480 square miles of country. Information is supplied on 227 mines which produced mica alone or both mica and beryl, and 134 mines which produced only beryl. In addition, wolfram, gold, graphite, kyanite and rutile deposits and mines are described. Mineral outputs are given up to end of 1959. The field mapping and writing of the bulletin is entirely the work of Mr. Wiles. Mapping of the Field has established that, with a few exceptions, a metamorphic control has operated in the development of economic sheet mica and has led to the conclusion that very many of these pegmatites are composite bodies which have derived their material from two sources: one metamorphic and the other igneous. This conclusion was arrived at after a detailed petrographic investigation of the metamorphic rocks. The map at the end of this bulletin was drawn by Mr.A. H. Barrie and the diagrams are largely the work of Mr. D. O. L. Levy. Chemical analyses are by Messrs. A. J. Radford and E. Golding and were made in the Geological Survey Laboratory. Some of the photographs are by H. J. Cotterel of the Geological Survey while the remainder were taken by the Planning and Technical Services of the Federal Ministry of Home Affairs (formerly the Federal Information Department). It gives me pleasure to acknowledge the assistance and facilities offered the Department by mining men and residents in the district.

Neste trabalho oferecemos a discussão transdisciplinar da construção dos modos de pensamento complexos emergentes no fim do segundo milênio ocidental e uma introdução às contribuições das concepções de investigação acopladas às questões pertinentes em educação, cultura e ciência, nos sentidos amplos das palavras. Still, emphasizing the contribution to biogeochemistry and soil science, we present a new method for the investigation of proton exchange patterns, in the equilibrium and out-of-equilibrium, of complex systems present in terrestrial biospherical ecosystems, in this case the mycelium of the basidiomycete fungus Trametes hirsuta, through the improvement of the application of potentiometry (using a combined glass electrode) as an intermediate technology. Lastly, the conceptual contribution of the research in chemistry toward ecology/transdisciplinary (Chapter 1) and biogeochemistry (Chapter 2) assume the development of culture, fundamental premise in education, as the main goal.

To elaborate teaching activities involving astronomical concepts, such as distance, brightness, image manipulation, colors as well as to explain the nature of different objects showing their images in different filters, among others. In the total, we have elaborated four activities that were applied to students of the 6th year of elementary school, high school and undergraduate ones for the Engineering and Pedagogy courses. All activities and the results of their evaluation with students are detailed discussed and analyzed; a teacher guide is also provided. Our results show that in the four activities, the students have a significant learning supporting the use of such methodology. Also, we elaborated two memory games, based on Java platform, with the images of some astronomical objects. The activities are in The Portuguese language, but they can easily be adapted for any other language. This research also made the translation of Aladin to Portuguese. / Este trabalho contempla a an?lise de um estudo de aplica??es, em sala de aula, de estrat?gias e t?cnicas de facilita??o, que utilizem o software Aladin (http://aladin.u-strasbg.fr/) na forma??o de conceitos de Astronomia e possibilidade de desenvolvimento cognitivo, por meio da aprendizagem, consolidando o aprendizado atrav?s do uso de computadores. Conceitos de dist?ncia, brilho, cor, exist?ncia de diferentes tipos de objetos astron?micos, manipula??o de imagens e dados astron?micos, entre outros aspectos, foram tratados em sala de aula e fizeram parte deste trabalho. A pesquisa foi aplicada em diversas etapas do ciclo de aprendizagem escolar, mais especificamente: 6? ano do ensino fundamental, 2? ano do ensino m?dio, alunos de gradua??o em Engenharia e, por fim, uma turma de alunas de Pedagogia, quase todos professores atuantes e, portanto, divulgadores dos conceitos para eles passados. Todas as atividades utilizaram o software Aladin e uma o aplicativo de Observat?rio Virtual, denominado VO-Stat. Como produto, foi elaborado material did?tico com o conte?do das atividades assim como um roteiro voltado para os professores realizarem a aplica??o da atividade para o ensino fundamental, um jogo da mem?ria virtual que trata de Astronomia. Due to the high singularity in terms of genres and endemic species as well as the potential use of these plants for various purposes, studies on the in vitro propagation and ex situ conservation of these species must be established. Thus, the aim of this study was to understand the anatomical, physiological and molecular changes associated with the propagation and conservation in vitro Discocactus zehntneri subsp boomianus and Stephanocereus leutezlburgii, species endemic cactus of Bahia with ornamental potential. The regulators applied not exceeded the control in the induction of shoots/explant in both species and for S. luetzelburgii combination of cytokinins and auxins hiperidricos increased the percentage of shoots that did not survive after acclimatization. Normal shoots of this species have 100% survival in ex vitro conditions. The callus formation was enhanced by adding auxin to the nutrient medium for the two species analyzed and calluses with potential for in vitro morphogenesis were obtained. The proteomic analysis revealed the marker proteins differential expression of somatic embryogenesis in two strains of S. luetetzelburgii calluses. The protocol for cryopreservation D. zehntneri shoots was not effective, however the seed storage either in liquid nitrogen conditions as for ultrafreezer maintained physiological quality after storage for 360 and 180 days, respectively. Osmotic agents were effective in inhibiting in vitro growth of the species for a year without loss of viability. / A fam?lia Cactaceae apresenta cerca de 100 g?neros e 1500 esp?cies predominantemente americanas e distribu?das por uma ampla diversidade de climas e ecossistemas. Devido ? alta singularidade em termos de g?neros e esp?cies end?micas bem como o uso potencial destas plantas para diversas finalidades, estudos acerca da propaga??o in vitro e conserva??o ex situ dessas esp?cies devem ser estabelecidos. Desta forma, o objetivo geral deste trabalho foi compreender as mudan?as anat?micas, fisiol?gicas e moleculares associadas ? propaga??o e conserva??o in vitro de Discocactus zehntneri subsp boomianus e Stephanocereus leutezlburgii, esp?cies de cact?ceas end?micas da Bahia com potencial ornamental. Os reguladores aplicados n?o superaram o controle na indu??o de brotos/explante em ambas as esp?cies e para S. luetzelburgii a combina??o de citocininas e auxinas aumentaram o percentual de brotos hiperidricos que n?o sobreviveram ap?s a aclimatiza??o. Brotos normais dessa esp?cie apresentaram 100% de sobreviv?ncia em condi??es ex vitro. A calog?nese foi potencializada pela adi??o de auxinas ao meio nutritivo para as duas esp?cies analisadas e calos com potencial para morfog?nese in vitro foram obtidos. A an?lise prote?mica revelou a express?o diferencial de prote?nas marcadoras da embriog?nese som?tica em duas linhagens de calos de S. luetetzelburgii. O protocolo para criopreserva??o de brotos de D. zehntneri n?o foi eficiente, no entanto a armazenamento de sementes tanto em condi??es de nitrog?nio l?quido quanto em ultrafreezer mantiveram a qualidade fisiol?gica ap?s o armazenamento por 360 e 180 dias, respectivamente. Os agentes osm?ticos foram eficientes em inibir o crescimento in vitro da esp?cie por um ano sem perda na viabilidade.

This master's thesis focuses on composite systems based on varnish combined with different filler ratios of micronized mica. The aim is to analyse impact of the filler on dielectric properties of the varnish, i.e. relative permittivity and loss factor as functions of frequency as well as charging and discharging currents as functions of time. Dielectric mixture formulas are applied on relative permittivity values. Calculated and measured values are compared.

INTEGRATION OF CERAMIC-METAL VERTICALLY ALIGNED NANOCOMPOSITE THIN FILMS ON FLEXIBLE MICA SUBSTRATES

<p>  </p> <p>Integration of functional thin films on flexible substrates has piqued interests owing to the needs of flexible devices. Selecting a suitable flexible substrate is crucial for such integration. Recently, muscovite mica has been developed as a flexible platform for functional thin film epitaxy growth. Mica can be easily peeled off due to the weak van der Waals interaction between different layers of mica, along with other advantages including cheap, high elasticity and thermal stability, biocompatible, <em>etc</em>. On the other hand, vertically aligned nanocomposites (VANs) have been attractive because of their unique anisotropic structures, which can achieve physical property anisotropy, easy tunability, out-of-plane strain engineering as well as combined multifunctionality. However, limited work on the integration of nanocomposite thin films on mica with tunable physical properties has been reported due to growth challenges. </p> <p>In this dissertation, different ceramic-metal VAN systems integrated on mica substrates towards different functionalities using pulsed laser deposition (PLD) have been demonstrated. The first chapter is on the integration of BaTiO3-Au nanocomposite system on mica. Tunable optical properties have been achieved by controlling the geometries of the Au nanostructures between nanoparticles and nanopillars by varying the growth temperature. The laser energy was also found to play a role in terms of the Au pillar dimension. The second chapter is on the integration of BaZrO3-Co VAN system on mica towards flexible spintronics. Tunable, anisotropic ferromagnetic property has been realized by controlling the aspect ratio of the Co pillars. The third chapter is on integration of BaTiO3-Fe VAN system on mica towards multiferroics. Different buffer layers have been tried out to facilitate the growth of VAN structure. Room temperature ferroelectric and anisotropic ferromagnetic properties of the films have been confirmed. The last chapter is focused on multiphase nitride-metal nanocomposite design and integration, with films showing unique optical and magnetic properties. The reliability and stability of the physical properties of the films have been verified though bending tests. The growth mechanism and criteria of ceramic-metal nanocomposite on mica have also been discussed. These demonstrations all pave a new way towards the integration and design of multifunctional nanocomposites towards flexible nanodevices.</p>
