The architectural study deals with recovery and redevelopment of Hrušovany nad Jevišovkou castle, which is currently abandoned and derelict. Inside the castle are planned cultural, social, exhibitive and representative areas. One wing of Hrušovany’s castle will be used as a hotel. There are designed extra standard rooms with chateau interiors. In underground floors we will find a wellness and wine bar. The service of the castle is extended by spaces in new outbuildings. It is a greenhouse, a restaurant and a pavilion for social events. The castle and outbuildings create a courtyard suitable for open-air cultural events. One part of castle‘s complex is garden lapidarium and floral terraces. The design keeps historical and architectural value of this cultural heritage and offers its dignified use nowadays.

The diploma thesis focuses on design of a nursing home in Planá nad Lužnicí. The building is freely standing on a flat land, with three above-ground floors. The entrance to the building is solved from the northeast side of the local road, the main entrance is oriented on the northwest side. The vertical support structures are made of Heluz brick blocks and covered with a mineral wool thermal insulation system. Horizontal structures are designed as reinforced concrete slabs. The building is roofed with a single-layer flat roof.

The NAMPT-mediated NAD salvage pathway in cancer cell metabolism and its regulation by resveratrol

Nicotinamide adenine dinucleotide (NAD) is a key regulator of several metabolic and signaling pathways that are relevant in cancer cell survival. Cancer cells have an increased energy demand associated with an increased NAD turnover. Nicotinamide phosphoribosyltransferase (NAMPT), a key enzyme of the NAD salvage pathway, plays a crucial role in maintaining the intracellular NAD levels and in regulating the activity of NAD-dependent enzymes, such as sirtuins (SIRTs). The inhibition of NAMPT activity and the use of phytochemicals, such as resveratrol, represent novel therapeutic approaches in cancer therapy. Based on these facts, this thesis aimed to investigate (1) the chemotherapeutic potential and molecular mechanisms of FK866, a specific NAMPT inhibitor, and resveratrol on hepatocarcinoma cells and to find out whether there are differences compared to primary human hepatocytes; (2) to address the impact of NAMPT inhibition on the energy metabolism in cancer cells; and (3) to investigate the roles of NAMPT and SIRT1 in resveratrol´s mode of action and chemotherapeutic effects. This work demonstrates that FK866 and resveratrol possess potent chemotherapeutic effects in hepatocarcinoma cells which were absent in human hepatocytes. Hepatocarcinoma cells display a dysregulation in the AMP-activated kinase (AMPK)/mammalian target of rapamycin (mTOR) signaling as well as in the NAMPT-mediated NAD salvage pathway compared to human hepatocytes. FK866-induced NAMPT inhibition induces ATP depletion associated with AMPK activation and mTOR inhibition whereas resveratrol induces caspase3-mediated apoptosis that is not dependent on NAMPT and SIRT1 function. NAMPT and SIRT1 are differentially regulated by resveratrol in hepatocarcinoma cells and human hepatocytes. This work also reveals that resveratrol activates p53-induced cell cycle arrest in hepatocarcinoma cells which is partly mediated by SIRT1 inhibition. In summary, this thesis provides new insight into the role of the NAMPT-mediated NAD salvage pathway in energy metabolism and characterized FK866 and resveratrol as promising potential chemotherapeutic agents for treatment of hepatocellular carcinoma.

The libraries of Lichnowsky noble family are currently composed of the sections. The first one is the main family line library stored in Hradec nad Moravicí castle and the latter is the one originally from Chuchelná castle. This thesis focuses on 16-18th century prints as parts of the above mentioned libraries. The aim here is the content and provenance analysis of the library fund of Lichnowsky within the suggested periods of time. At the front, the attention is on signature lines and on the question, whether the Chuchelná library contains books that once belonged to private property of Lichnowsky family. In the following chapters, it is explained why the scale of the analysis is limited to the selected centuries. The answer also reflects the general topic of understanding castle libraries. The focus is also on the history of the noble family of Lichnowsky. After localization of the library itself, major parts of the thesis follow, which chapters comprising the content and provenance analysis of the libraries. The provenance chapter has two parts. The first one identifies members of Lichnowsky family behind provenance traces inside the books, whereas the major role is played by Robert Lichnowsky, largely responsible for operating and maintaining the family library. The purpose of this disertation is to capture the era of so-called normalisation during the 1970s and 1980s of the 20th century in Czechoslovakia. Even though the economic transformation in the 1990s is covered, the normalisation is in this case the primary investigated period. The main focus is on the largest producer and exporter of czechoslovak motorcycles - a company called JAWA with the main production plant located in Tynec nad Sazavou - Brodce. In addition to the motorcycle production in Tynec nad Sazavou the work looks at Czechoslovakia and its role as a power of the motorcycle sport during socialism and other aspects such as the role of motorcycles during the normalisation period in Czechoslovakia or the outline of the historical development of JAWA in the post-war years and its growth on the international scale before nationalization of the enterprise. From the mid-nineteenth century the North Bohemian industrial area, populated mostly by Germans, experienced an unprecedented urban and demographic growth. The onset of the industrial revolution meant a major breaking point, in particular because the following periods shaped it most notably.The characteristic trait of the cities is a precipitous urban development with building boom. Thereby an impressive scrum of mutually coexisting industrial complexes, palatial villas, spectacular public buildings, generous garden districts and small town housing projects originated, creating the peculiar local genius loci. The border areas have always been a busy crossroads of cultures, histories and ideologies, in which Czechs, Germans and Jews mingled and interacted with each other, and thus they reflect the complex ethnic relations in the territory of today's Czech Republic. The Thesis focuses on the architecture of Czech Germans which formed the natural counterpart to the architecture of the Czech majority. The aim is to set this "second stream" of culture into a wider Central European context and find and define the specifics of the buildings production of Czech Germans. The dissertation shows one of the possible portraits of the history of the Roman Catholic Church in Czechoslovakia between the years 1927-1963. On the example of Václav Boštík (1897-1963), the dean in Ústí nad Orlicí between the years 1934-1942 and 1945-1961, the thesis focuses on continuities and discontinuities in the history of the local Roman Catholic parish. The presented analysis combines the socio-historical, microhistorical and biographical approach, by means of which it shows pastoral and other strategies of dean Václav Boštík - an ecclesiastical administrator and statutory representative - in various situations during the first two-thirds of the twentieth century. Local events are interconnected with the context of Czech ecclesiastical history at the statewide and diocesan level as well as with a broker context of Church history. The subject undertaken in my dissertation work is the gallery painting collection from the aristocratic family 'Kolowrat', which is located within the family's palace in Rychnov nad Kněžnou. The Kolowrats established their gallery in the 16th century, the foundation of which was made to contain family member portraits. During this time portrait paintings comprised the main element and went on to create the portrait collection. The collection contains the oldest Bohemian aristocratic portraits from the Renaissance with portraits influenced by artists from the court of Emperor Rudolf II. It includes portraits from important artists as Karel Škréta, David Schultz or Philip Ch. Bentum and more. The portraits of family members document the history of the Kolowrat clan through the ages. At first this dissertation describes the history of the gallery and goes on to present select portraits from the beginning of the gallery until the 18th century. The introductory chapter fills a completely blank space concerning the history, organisation and spirituality of the Order of the Discalced Augustinians in the Czech context. The existing literature has so far only briefly outlined this topic; however, it cannot be explored with no knowledge of the 20th -century professional Italian research works or the preserved resources produced by the General Curia in Rome and the 18th century literature of the Order. The thesis also provides a general overview of the activities of the Czech mon­ asteries of the Discalced Augustinians in the period context, also reflecting the in­ fluence of the Order on the spiritual and cultural life of the laymen, an influence which, as regards the monastery in Lnare, extends into the early 20th century. The extant resources have helped to describe relations between the members of the Order and the outside world, be it the links to the local clergy or laymen, espe­ cially burghers and nobility or its dependants. The relationship between the famil­ ies who financed the foundation of the two latest monasteries (Lnare and Lysa nad Labem) and the monastic community is the focus of specific attention. The aim of this project is a reconstruction of a former kindergarten building in Žďár nad Sázavou to a community center with several functional units. The existing building was bulit in 1960s. The site is located on the border of the old and new buildings – on one side, there is a Stalingrad housing estate from the 1950s built from brick houses in the socialist realism style. On the other side of the site are built new family houses, which are quickly expanding. The smaller part of the object is still used, the rest is abandoned. The house is surrounded by a large garden. The garden is defined by a fence and trees in a way, which creates a barrier, so the building does not communicate with surroundings in any way. The aim of the project is to create a centre of Stalingrad connected with surroundings and creating a meeting place for communities living in the area by using building interventions.
