The Cecilian Music Society in Ústí nad Orlicí, East Bohemia

In the year 1903, the oldest existing Bohemian music society, the Tonkünstler-Wittwen-und-Waisen-Societät, founded in Prague in March 1803, finally ceased all its music activities; it continued its work just as a pension fund up to its final closing down in 1930. The Cecilská hudební jednota (Cecilian Music Society), founded in November 1803 in Ústí nad Orlicí/Wildenschwert, East Bohemia, was at that time still thriving. The ways in which the period social, political and cultural circumstances reflected in its work, are of special interest for anybody researching Bohemian music history.

Temperature sensitive Mycobacterium tuberculosis as a potential vaccine candidate

Mycobacterium tuberculosis remains one of the most common worldwide causes of illness and death due to an infectious disease. The emergence of multiple and extreme-drug resistant strains has increased the need to find an effective vaccine for tuberculosis. The goal of our research group is to engineer a temperature-sensitive (TS) M. tuberculosis strain that can be used as a tool in vaccine development. One approach to create TS M. tuberculosis involves the integration of the essential gene ligA encoding a TS NAD+ dependent DNA ligase, which was taken from the psychrophilic organism Pseudoalteromonas haloplanktis. The integration and functioning of ligA was demonstrated in the fast-growing organism Mycobacterium smegmatis. This strain had a TS phenotype with growth limited to below 37°C. The strain was found to have a stable TS phenotype and did not mutate to a temperature-resistant form at a detectable level. Following experiments with the fast growing M. smegmatis, the integration of the ligA gene was attempted in slow-growing M. tuberculosis. Merodiploids of M. tuberculosis containing both the psychrophilic and the WT ligA gene in its chromosome were obtained. The second approach used for the development of TS M. tuberculosis was the directed evolution of native M. tuberculosis essential genes. An advantage of this approach is that the gene encoding the essential protein will resemble the native M. tuberculosis gene and thus will closely match the native transcriptional and translational rates. A system to screen and select for TS essential genes engineered by directed evolution was designed, where the essential gene on the chromosome of E. coli was knocked out and this gene was supplied on a conditionally replicating plasmid. As a first step in developing this directed evolution approach, a family of conditionally replicating plasmids were created and tested in an essential gene knock-out strain of E. coli. / Graduate

Effect of Partial Poly (ADP-ribose) Glycohydrolase Gene Deletion on Cellular Responses to Genotoxic Stress

Polymers of ADP-ribose (PAR) are rapidly synthesized by poly(ADPribose) polymerases (PARPs) and rapidly degraded by poly(ADP-ribose) glycohydrolase (PARG) following genotoxic stress. Since PAR metabolism plays an important role in cell fate determination following genotoxic stress, enzymes involved in PAR metabolism potentially represent promising therapeutic targets for modulating diseases of inappropriate cell proliferation or death. PARP-1 has been well validated and several PARP-1 inhibitors are currently being evaluated in clinical trials for cancer and ischemia treatment. In contrast, the biological function of PARG is still poorly understood. Due to low abundance of protein levels in mammalian cells and its unique substrate, PARG potentially represents another attractive target for pathological conditions mentioned above. PARG-Δ2,3 cells derived from homozygous PARG-Δ2,3 mice with targeted disruption of exons 2 and 3 of the PARG gene are used in this dissertation. The nuclear isoform PARG60 in PARG-Δ2,3 cells lacks the putative regulatory domain A compared to the nuclear isoform PARG110 in wild type cells. We report in this dissertation that PARG-Δ2,3 cells accumulate less PAR in spite of more rapid depletion of NAD following treatment with N-methyl- N’- Nitro-N-Nitrosoguanidine (MNNG). The estimation of PARP and PARG activity in intact cells shows increased activity of both enzymes in PARG-Δ2,3 cells following MNNG treatment, indicating the important role of domain A in the regulation of PARG and PARP activity under these conditions. Following MNNG treatment, PARG-Δ2,3 cells show reduced formation of XRCC1 foci, decreased H2AX phosphorylation, decreased DNA break intermediates during repair, and increased cell death. The altered PAR metabolism and defective cellular responses related to DNA repair in PARG-Δ2,3 cells may contribute to increased sensitivity of these cells to MNNG. Studies presented in this dissertation clearly demonstrate the important role of PARG110 in PAR metabolism and cellular responses to genotoxic stress, and thus provide supportive data for the validation of PARG as a promising potential therapeutic target.

The issue of juvenile criminal justice The topic of my Master's degree thesis is "The issue of criminal juvenile justice". It is focused on the area of unlawful acts of youth and children under fifteen years old. I have decided to focus on this area as it of a great interest to me. My Master's degree thesis is devided into five chapters and further split into subsections. In my thesis I have analyzed possible issues of corrective measures and their effectiveness. The main focus of my thesis is juridical and minor part is also devoted to criminal aspects of youth deliquency. I have accompanied my thesis with appropriate jurisprudence and real life examples. In the introductory chapter I have provided basic facts of the youth criminal acts, focusing mainly in the criminal aspects of the characteristic conducts and behaviors. The following chapter is focused on the historic development of the criminal jurisdiction of the youth deliquency on the territory of the Czech republic with the applicable Law No. 218/2003 Coll. Furthermore, this chapter is providing additional details for issuance of the separate legal act on the youth deliquency and comparison of this act with simile legal acts in the other countries. of the master thesis The issue of criminal juvenile justice The thesis deals with problems of criminal justice over juveniles. The purpose of the study is to bring an objective view of actual legal regulation in The Juvenile Justice Act No. 218/2003 and to identify problematic aspects of the juvenile justice system. Legal regulation is based upon the principle not to punish juvenile delinquents in repressive way, but to eliminate negative influences. The reason for my research is to discover the main causes of children's criminality and to outline their possible solutions. I focus on two big areas in the study - problems of the juvenile criminal liability and the system of legal instruments as a reaction of society to delinquency. The thesis is divided into six chapters; each chapter is further divided into several more subchapters according to a logical succession of addressed problems. In the first chapter I define basic terms, which are important to understand these issues. These definitions come out of the Juvenile Justice Act. Resumé Irena Valentová Financial Markets Supervision Fragmented, and therefore insufficient, and ineffective regulation of and supervision over financial markets was blamed to be one of the main sources of the recent economic crisis, that manifested in 2008. The main theme of the present thesis consists of the changes in the institutional framework of financial markets supervision in the European Union introduced as a reaction to the recent economic crisis. The first part of the present thesis composed of chapters two, three, and four, deals with definition of some expressions used in the second part of the thesis, namely: financial markets, regulation and supervision of financial markets. The concept of financial market is explained, its components as well as structure described. Next, several reasons leading to need of effective regulation are brought and discussed and, finally, the concept of supervision as used in the context of financial markets is established. The thesis deals with soils of the middle Elbe floodplain. Basic information about the development of Elbe floodplain is focused on the middle Elbe region. The aim of the thesis is to capture the floodplain development in the area of interest, including the description and comparison of soils at different transects in surroundings of Kostelec nad Labem. For this purpose several transects from the historical Elbe canalisation documentation were chosen. Additional to this, small soil survey was carried out at identical locations in 2013. Obtained information was confronted and evaluated.

This thesis mainly deals with the procedures concerning the youth and children under fifteen years old, including the procedural status of subjects according to Act No. 218/2003 Coll. Youth on liability for unlawful acts of Juvenile Justice and amending certain acts (Act on Juvenile Justice), but the Act No. 40/2009 Coll., Penal Code. Introduction of this thesis is the historical development, both within Austria-Hungary or the Czech Republic after 2003. Its cross section is the interpretation of certain key concepts and principles that are essential to the interpretation and practical application of the Act itself. The chapter on the Institute diversions could not be omitted in this thesis. Using detours is still a new phenomenon for Czech criminal law and in the practice of law is still used less (unlike the penalties associated with imprisonment). It is mentioned both in the general characteristics of control over juveniles and children under fifteen years old, protection of private adolescents with an emphasis on the provision of information in cases, where the perpetrator or victim of the offense is a person under the age of fifteen years old. This thesis deals with archaeological tourism in the Ústí nad Labem region. Based on scientific literature, the general issues of archeology and tourism are analyzed. The result of this analysis points to the fact, that there are a lot of archaeological tourist destinations in this region. The most attractive ones are presented in this thesis in the form of a tour guide intended for the general public. An opinion poll was coducted as part of the thesis. Based on the poll we can assume that the general awareness of archeology and archaeological tourist destinations in the Ústí region is at an average level. It is safe to say that the public attitude towards archeology is generally positive. Czech castles are especially popular among the archaeological tourist destinations. The most convenient public presentation of archaeological sites is by using information boards with brief descriptions and vivid illustrations. The vast majority of interviewees indicated that they would welcome a higher degree of presentation of historical monuments in the Ústí nad Labem region - for example through a dedicated website. The general part applies generally to the institution of people's courts, their ideological grounds, the local people's courts statute and it's interpretation, the problems of chosen institutes are connected to the people's courts statute and to the activities of these courts.
