This thesis deals with supervision over the financial market and its legal framework. I chose the topic because I have always been interested in the issue concerning the financial market. This thesis enables readers to get familiarized with the current situation of the financial market supervision in the Czech Republic, European Union and three member states of European Union, i.e. United Kingdom, France and Slovakia. The Chapter One is an introduction to the theme. The second chapter deals with the basic terms used in the thesis as financial market, supervision, inspection etc. It is a theoretical part of the thesis. The third chapter examines relevant legislation of the European Union and it is dedicated to the EU financial supervision. A new structure for EU financial supervision was agreed in 2010 following the adoption of the de Larosière report in November 2008. At the beginning of 2011 the European Systemic Risk Board (ESRB) and three European Supervisory Authorities (ESAs) were established. The purpose of the new structure is to ensure effective surveilance of the EU financial market. The ESRB is responsible for monitoring and assessing potential threats to the stability of the financial system. Mecanismos fisiopatológicos do desequilíbrio redox em células neointimais vasculares / Pathophysiological mechanisms of redox inbalance in neointimal vascular cells

O crescimento de uma camada neoíntima é o marcador central do processo aterosclerótico, bem como do remodelamento vascular associado à reestenose após intervenções vasculares terapêuticas, tais como angioplastia ou colocação de stents. A célula neointimal é essencialmente uma célula com fenótipo muscular liso indiferenciado, cuja origem pode ser múltipla e com característica ação secretora de matriz extracelular. Dentre os fatores que coordenam a (des)diferenciação, proliferação e migração destas células, há evidências de que processos redox tenham papel preponderante, porém os mecanismos de tais processos não estão claros. A compreensão mais profunda de tais mecanismos redox tem sido dificultada pela falta de modelos de células neointimais cultivadas. Os objetivos específicos deste trabalho são: 1) Desenvolver um modelo de cultura de células neointimais de artéria ilíaca de coelho obtidas após lesão por catéter-balão. 2) Avaliar em tais células índices do estado redox e potenciais fontes enzimáticas de ERO, com ênfase no complexo NAD(P)H oxidase. 3)Avaliar marcadores de estresse do retículo endoplasmático e sua correlação com os índices do estado redox. 4) Estudar o efeito de estímulos proapoptóticos como deprivação de soro, estressores do RE e particularmente administração exógena de NO na viabilidade e estado redox de células neointimais. Os resultados obtidos demonstram que: é possível obter células neointimais em cultura de modo reproduzível e estável. Células provenientes da artéria lesada mantém um estado persistente de aumento do estresse oxidativo. O estresse oxidativo nessas células decorre de produção aumentada de radical superóxido e ativação do complexo da NADPH oxidase vascular. Diferentemente do observado em vasos lesados, os marcadores do estresse do reticulo endoplasmático não se apresentam alterados se comparados a células musculares lisas normais. Células neointimais têm resposta aumentada a agonistas da NADPH oxidase e estressores celulares. A exposição a óxido nitrico promove aumento da produção de superóxido, particularmente acentuado em células neointimais. As curvas de viabilidade celular indicam uma sensibilidade aumentada a estressores do RE, doadores de NO e particularmente a oxidantes exógenos. Em conjunto, estes resultados permitem concluir que o fenótipo neointimal é um fenótipo de estresse oxidativo acentuado e persistente, mesmo em condições de cultura celular, e que este estresse oxidativo decorre pelo menos em parte da ativação do complexo NADPH oxidase no contexto de uma adaptação à resposta celular integrada ao estresse. Estes dados indicam novas perspectivas no entendimento dos mecanismos envolvidos na fisiopatologia redox da neoíntima, podendo suscitar o desenho de intervenções terapêuticas racionais. / Formation of a neointimal layer is the hallmark of most vascular diseases, such as atherosclerosis and restenosis after angioplasty. Neointimal cells display an undifferentiated noncontractile smooth muscle phenotype with marked extracellular matrix secretion. Their origin can be multiple. Among factors that govern the (de)differentiation, proliferation and migration of neointimal cells, there is evidence for a key role of redox processes, but the underlying mechanisms are unclear. Advancing the knowledge about such redox mechanisms has been difficulted by the absence of a reproducible method of neointimal cell culture. The objectives of this work are: 1) To develop a model of neointimal cell culture, in which cells are harvested from rabbit iliac arteries 14 days after overdistention balloon injury. 2) To assess the redox status in such neointimal cells and the possible enzymatic source of reactive oxygen, with emphasis in the NAD(P)H oxidase complex. 3) To investigate the expression of endoplasmic reticulum stress markers and their corralation with redox status. 4) To investigate the effects of proapoptotic stimuli such as serum deprivation, endoplasmic reticulum stressors and particularly the exogenous administration of nitric oxide in viability and redox status of neointimal cells. Our results show that it is possible to harvest and cultivate neointimal cells after balloon injury. The neointimal cells in culture, even after several passages, exhibit increased indexes of oxidative stress. Oxidative stress in such cells is associated with increased activation of the vascular NAD(P)H oxidase complex. Contrarily to what was observed in healing arteries harvested from in vivo rabbits, markers of ER stress did not show any change when compared with primary smooth muscle cells kept in similar conditions. Oxidative stress response was increased after NADPH oxidase agonists; in particular, exposure to exogenous nitric oxide markedly increased superoxide radical production in neointimal cells. Cell viability curves showed increased sensitivity to ER stressors, NO donors and, particularly, exogenous oxidants. Therefore, the neointimal phenotype is a phenotype of intrinsic sustained oxidative stress even after several passages in culture. Such oxidative stress is due at least in part to activation of the NAD(P)H oxidase complex in the context of adaptation to an integrated stress response. This data provide new perspectives to understand redox mechanisms associated with neointimal pathophysiology and can lead to development of rational therapeutic interventions.

Une caractéristique essentielle du mécanisme catalytique des ALDH est l'importance de la flexibilité et de la dynamique conformationnelle dans le site actif, incluant les chaînes latérales de résidus, le substrat, et le cofacteur. Ma thèse a permis d'identifier des bases responsables de la reconnaissance du rétinal contrôlant la biosynthèse de l'acide rétinoïque dans les RALDH. J'ai ensuite pu suivre le basculement du cofacteur réduit en utilisant le FRET, ce qui m'a permis d'établir un modèle cinétique pour déterminer la constante de vitesse associée à cette étape. Enfin, les résultats obtenus pour identifier les bases moléculaires responsables de la dissociation tardive ou précoce du cofacteur réduit ont montré que le mode de stabilisation du cofacteur est à l'origine de cette différence entre ces deux familles d'enzymes / An essential feature of the catalytic mechanism of ALDH is the importance of flexibility and conformational dynamics in the active site, including the side chains of residues, substrate and cofactor. My thesis has identified bases responsible for the recognition of retinal controlling biosynthesis of retinoic acid in RALDH. I was then able to follow the flip of the cofactor reduced by using FRET, which allowed me to develop a kinetic model to determine the rate constant associated. Finally, the results obtained to identify the molecular basis responsible for late or early dissociation of the reduced cofactor showed that the mode of stabilization of the cofactor is the origin of this difference between these two families of enzymes

Modulação das vias de sinalização intracelulares pelo sistema NAD(P)H oxidase em melanoma humano / NAD(P)H oxidase modulates intracellular signaling pathways on human melanoma

Fundação de Amparo à Pesquisa do Estado do Rio de Janeiro / Evidências têm mostrado que as espécies reativas de oxigênio (ROS) geradas pela NAD(P)H oxidase são importantes moduladores de diversas funções celulares como migração, crescimento, proliferação e sobrevivência. Estudos recentes demonstraram o envolvimento da atividade da NAD(P)H oxidase no crescimento e sobrevivência de células de melanoma. Neste trabalho, investigamos o efeito da inibição da NAD(P)H oxidase por difenileneiodônio (DPI) sobre o crescimento das células de melanoma humano MV3 e observamos que este composto reduziu o crescimento destas células em aproximadamente 50%. A inibição da NAD(P)H oxidase induziu mudanças no formato celular, com arredondamento, diminuição do espraiamento e descolamento celular. Esta redução foi acompanhada por um rearranjo do citoesqueleto de actina, diminuição da fosforilação no resíduo Tyr397 da quinase de adesão focal (FAK) e redução na associação de FAK com actina e com a tirosina quinase c-Src. Isto indica que a inibição da geração de ROS está modulando negativamente vias de sinalização ativadas por integrinas, o que freqüentemente conduz a um tipo particular de morte celular conhecida por anoikis. Comprovando a ocorrência deste fenômeno, observamos que a inibição da atividade da NAD(P)H oxidase aumentou a apoptose das células de melanoma e induziu a ativação da caspase-3. Nossos resultados mostram ainda que a inibição da viabilidade celular por DPI foi revertida com o pré-tratamento das células MV3 com um inibidor de tirosina fosfatases (ortovanadato de sódio). Em resumo, este estudo mostra que a geração de ROS por NAD(P)H oxidase está envolvida nos mecanismos de sobrevivência em células de melanoma, uma vez que afetam as vias de sinalização dependentes de FAK-Src, através da inibição da atividade de proteína tirosina fosfatases. / NAD(P)H oxidase-derived reactive oxygen species (ROS) have emerged as critical mediators of several cell functions as diverse as migration, growth, proliferation and survival. Recent evidence has show that NAD(P)H oxidase activity is essential to melanoma proliferation and survival. We reported that NAD(P)H oxidase inhibitor diphenyleneiodonium (DPI) inhibited melanoma growth. NAD(P)H oxidase inhibition induced changes in cell shape with cell spreading decrease, rounding up and detachment. These phenomena were accompanied by rearrangement of actin network and a decrease in both focal adhesion (FAK) phosphorylation in Tyr397 residue and in FAK association to actin and c-Src, indicating that inhibition of ROS generation would down- modulate integrin-mediated signaling, what often results in a particular type of apoptotic cell death, known as anoikis. We observed that NAD(P)H oxidase inhibitor induced apoptosis in melanoma cells with activation of caspase-3. We results show that the effects promoted by NAD(P)H oxidase inhibition on melanoma growth were completely abolished by the pre-treatment of MV3 cells with the protein tyrosine phosphatases inhibitor sodium orthovanadate. In conclusion, our results strongly suggest that ROS generated by NAD(P)H oxidase complex transmit cell survival signals in melanoma cells through the FAK-Src pathway, probably inhibiting protein tyrosine phosphatases.

The aim of this thesis was to create a new communications strategy for the athletics meeting called Grand Prix Ústí nad Labem, thereby ensuring greater participation of the audience. The first part of this thesis is focused on the theoretical background with an emphasis on marketing communication and it's differences in the focus on sport and the development of effective communication using the tools of the communication mix. The second part of this work is devoted to the history and present of the athletics in Ústí nad Labem and the history of Grand Prix Ústí nad Labem. The third part of this thesis describes the current operation of the athletic meeting, it's organizational structure and financial aspects of the existing communication strategy. The fourth part is the actual research conducted through questionnaires. The results of the survey and additional outputs from SPSS helped to create the fifth part, the part which is designed to refocus the athletic meeting communication strategy -- billboards, tickets, TV commercials, to improve Internet and Internet forums communications, etc., which should help increase public participation in the actual Grand Prix meeting Ústí nad Labem.

The theoretical part is devoted to the definition of basic terms from the tourism and balneology branch. This section also includes basic theory of marketing and marketing research focusing on the specifics of marketing in spa industry and the marketing mix. The practical part deals with the characteristics of the Ústí Region, Klášterec nad Ohří and spa itself. To the subject of spa is devoted significant part of the work. It focuses on the history of spa, mineral springs and the current situation, which includes the characteristics of the services offered, the characteristics of clients, staff, competitors and marketing mix of the spa. The practical part also includes a research of customer satisfaction, its evaluation and recommendations which may be used in the spa for the future improvement of activities.

To work out this diploma thesis on the theme Marketing study of selected tourist attraction the author has chosen Castle Klášterec nad Ohří. The castle is the most important attraction in the town Klášterec nad Ohří and one of the most visited monuments in the Czech Republic. The castle is a tourist target of almost 20 thousands visitors every year. The task of this thesis is to analyze existing application of marketing tools of this attraction. To resolve this issue will be used the following analysis: analysis of marketing mix, environmental analysis, segmentation of visitors, turnout analysis of visitors and SWOT analysis. The aim of this thesis is, based on the data found out in the analyzes, to make up a concept of further development of Castle Klášterec nad Ohří for the next period which could lead to increase its attractiveness. The diploma thesis is devided into four chapters. The first chapter is devoted to basic concepts of tourism and marketing. Based on the quotes in this chapter, is made the analytical part which applies the knowledge to marketing activities of castle.

Porteur redox intervenant dans nombre de processus métaboliques, le NAD (nicotinamide adénine dinucléotide) est central pour les cellules vivantes. Outre son importance dans le métabolisme oxydoréductif, des données récentes suggèrent fortement d'autres rôles importants pour le NAD dans la signalisation cellulaire. Un système inductible d'enrichissement en NAD en surproduisant la quinolinate phosphoribosyltransférase (QPT) d'Escherichia coli chez Arabidopsis thaliana a permis de mettre en évidence l'implication du NAD dans les mécanismes spécifiques de défenses qui régissent les interactions plante-pathogène. Par ailleurs, une dérégulation de la synthèse de NAD sur l'étape enzymatique catalysée par la QPT endogène d'Arabidopsis thaliana souligne le rôle critique du NAD dans la balance C/N des plantes, en particulier en bouleversant l'assimilation de l'azote en conditions photorespiratoires. Ces travaux nous ouvrent à une nouvelle compréhension des mécanismes de signalisation impliquant le NAD dans les grandes fonctions métaboliques des plantes.

Pre-B Cell Colony-enhancing Factor (PBEF) Promotes Neutrophil Inflammatory Function through Enzymatic and Non-enzymatic Mechanisms

Pre-B Cell Colony-Enhancing Factor (PBEF) is a cytokine-like molecule that functions as a nicotinamide phosphoribosyl transferase (Nampt) in a salvage pathway of NAD biosynthesis. PBEF has well-characterized activity as an extracellular inflammatory mediator and has been proposed to signal through the insulin receptor (IR). As neutrophils are key effectors of the innate immune response to infection and injury, we hypothesized that PBEF promotes pro-inflammatory function in neutrophils and that these pro-inflammatory effects may occur through interactions with the neutrophil IR or through PBEF���s enzymatic Nampt activity. Our studies focused on two important facets of neutrophil inflammatory function: their ability to generate reactive oxygen species (ROS) and undergo constitutive apoptosis. We found that, although PBEF does not activate oxidative burst on its own, it primes for ROS generation through the NADPH oxidase. PBEF promotes membrane translocation of cytosolic NADPH oxidase subunits p40phox and p47phox, but not p67phox, induces p40phox phosphorylation and activates Rac. Priming, translocation and phosphorylation are dependent on activation of p38 and ERK mitogen activated protein kinases. PBEF priming of neutrophils occurs independent of its Nampt capacity or of interactions with IR. We next investigated the effects of PBEF on neutrophil constitutive apoptosis. Our lab previously established that extracellular PBEF delays neutrophil apoptosis. Accordingly, we next investigated the mechanism through which this delay was occurring. PBEF-induced delayed apoptosis was enhanced in the presence of Nampt substrates, and NAD alone could delay apoptosis to an extent comparable to PBEF. Delayed apoptosis was blocked by a Nampt inhibitor and was lacking when a mutated PBEF deficient in Nampt activity was utilized. The cell-surface NAD glycohydrolase, CD38, can convert NAD to cyclic ADP-ribose (cADPR). Blocking CD38 activity with a blocking antibody partially reversed the delay of apoptosis induced by PBEF in conjunction with its substrates, and delayed apoptosis could be achieved by addition of the CD38 product cADPR. Finally, we found that delayed apoptosis induced by PBEF did not involve IR. These results indicate that PBEF can prime for enhanced oxidative burst and delay apoptosis in neutrophils, and that these phenomena occur through distinct mechanisms.

The Role of FGF21 in Pancreatic Islet Metabolism

The endocrine-like factor FGF21 is a potent regulator of nutrient metabolism. Systemic FGF21 administration to obese animals improves glucose tolerance, lowers blood glucose and triglycerides, and decreases fasting insulin levels. Although FGF21 improves the survival and function of islet β-cells, the mechanisms are currently unknown. This thesis examines mechanisms of FGF21 in the regulation of pancreatic islet metabolism. Biochemistry studies showed FGF21 decreased Acetyl-CoA carboxylase (ACC) and Uncoupling protein-2 (UCP2) protein expression in mouse islets. Autofluorescence microscopy showed difference in NAD(P)H responses when challenged with TCA cycle intermediate citrate. FGF21-treated islets showed significant decreased mitochondrial energetics when acutely stimulated with high concentrations of glucose and palmitate. This decrease in energetics correlated with increased generation of NADPH. Importantly, insulin secretion was lowered but not abolished in this state. These data confirm that FGF21 alters pancreatic islets metabolism during high glucose and high fat loading and reduces insulin during nutrient stress.
