Sociální kapitál a rozvoj malých a středně velkých měst: případová studie měst Jihočeského kraje / Social capital and development of small and medium-sized towns: case study of towns in the South Bohemian Region

The main objective of thesis is to empirically assess the level of social capital in relation to the development of the city. First, the thesis discusses the theoretical framing of the concept of social capital; it also deals with the topic of regional development and its conditionality, including the issue of the development of small and medium-sized towns. The second part is devoted to a quantitative analysis of social capital and regional development in all Czech small and medium-sized towns. The last part presents detailed qualitative research on social capital, specifically the results of case studies observed at the level of comparable towns Týn nad Vltavou and Třeboň. In these case studies, key equalities and differences of social capital in connection with the formation of the city were identified. We also confirm the importance of social capital as one of many factors that determines the territorial development. Aristolochic acids (AA) are carcinogenic and nephrotoxic alkaloids from Aristolochia species. Aristolochic acid I (AAI), the major component of AA, causes the development of Aristolochic acid nephropathy (AAN) and Balkan endemic nephropathy (BEN). These two diseases cause total renal failure and urothelial malignancies. The fact that these diseases have not been developed in all persons, who have been exposed to their action, might be causd by different activities and protein levels of the enzymes metabolizing AAI. Thus, the identification of enzymes involved in the metabolism, and detailed knowledge of their expression and catalytic specifities is a major importance. Aristolochic acid I (AAI) can be metabolized by several types of reactions. Like most nitroaromatics, the main activation pathway of AAI is reduction of its nitro group to form a cyclic acylnitrenium ion, which can bind to the purine bases, thereby forming AAI-DNA adducts. The detoxication pathway of AAI is its oxidative demethylation by cytochromes P450 forming detoxication metabolite 8-hydroxyaristolochic acid Ia (AAIa). In the present thesis, using rat and human enzymes and as well as several mice models, the metabolism of AAI in vitro and in vivo was investigated. 1 Abstrakt Diploma thesis "Consequences of low unemployment for social work in the Rychnov nad Kneznou region" concerns about negative consequences of low unemployment on the citizens of the Rychnov nad Kneznou region, especially about impact on traffic and social infrastructure and criminality. The first chapter is given to unemployment overall and to statistics of unemployment in the Czech Republic. The second one brings statistic view of demography and regional unemployment in Rychnov nad Kneznou. The third chapter is focused on foreigners and their statistics in terms of the Czech Republic and the Rychnov nad Kneznou region. In the fourth chapter are described particular negative results caused by low unemployment in this region. The last final chapter contains the practical part where is done examination of citizens' opinions in three chosen microregions about this matters. This examination was done by survey and its result is statement that citizens in the center of happening does not think the situation is as bad as the citizens in other two microregions (which are quite isolated from the core of issues by distance) think it is.

The aim of this thesis is to elaborata a strategic plan of development for the town of Lomnice nad Lužnicí. The work begins with a literature review, which contains a list of findings related to strategic planning including strategy, a set of regional development strategy documents, funding sources and more. To obtain the information needed to draw up a strategic plan a method of documentary inquiry, survey and structured interview were used. In the analytical part of the documentary survey, it was found that in the area of regional development, the higher territorial units have detailed current strategic development documents. The primary points of development listed in these documents are consistent with each other. The practical part of this work is made up by a detailed outline of a strategic plan for the development of the Lomnice nad Lužnicí. This outline includes characteristics of Lomnice nad Lužnicí, SWOT analysis, proposed priority areas of development and also includes measures for individual priority areas and development activities.

O T. cruzi, é o agente causador da doença de Chagas. O estado redox NAD+/NADH intracelular é fundamental na manutenção do metabolismo celular. A GAPDH apresenta a função de proteção do telômero em mamíferos contra degradação, isto por causa de ligar se ao telômero. Aqui, mostramos que a GAPDH recombinante de T. cruzi (rTcGAPDH) interage com o DNA telomérico. A rTcGAPDH liga ao DNA de simples fita. Mostramos que a GAPDH liga ao DNA telomérico in vivo em células epimastigotas, onde a [NADH] é maior que [NAD+], mas a adição de NAD+ exógeno bloqueia esta interação. Corroborando a hipótese de que o equilíbrio NAD+/NADH determina a interação GAPDH-telômero, vimos que o tripomastigota tem maior [NAD+] intracelular que a [NADH] e a GAPDH não é capaz de ligar se ao DNA telomérico. Além disso, o NADH exógeno resgata a interação GAPDH-telómero nesta fase. É importante o equilíbrio NAD+/NADH desta interação em tripanosomas, sugerindo que a proteção do telômero do parasita pode ser regulada pelo estado metabólico das células. / The T. cruzi, is the causative agent of Chagas disease. The redox state of NAD+/NADH intracellular is critical in the maintenance of cellular metabolism. The GAPDH has the protection function of the telomere in mammals against degradation, because it is connecting to the telomere. Here we show the recombinant GAPDH of T. cruzi (rTcGAPDH) interacts with telomeric DNA. The rTcGAPDH binds to single-stranded DNA. We show GAPDH to bind to telomeric DNA in vivo epimastigotes cells, where [NADH] is greater than [NAD+], but the addition of exogenous NAD+ blocks this interaction. Corroborating the hypothesis that the NAD+/NADH balance determines the GAPDH-telomere interaction, we saw that the trypomastigote has higher [NAD+] that intracellular [NADH] and GAPDH is not able to connect to telomeric DNA. In addition, the exogenous NADH recovers the GAPDH-telomere interaction at this stage. It is important the NAD+/NADH balance this interaction in trypanosomes, suggesting that the protection of the telomere of the parasite can be regulated by the metabolic state of the cells.

Neste projeto, demonstramos a modulação da enzima NAD(P)H oxidase pela glicose, palmitato e interleucina - 1? através da análise da expressão protéica do componente p47PHOX e pela atividade dessa enzima via produção de superóxido e peróxido de hidrogênio. Demonstramos também a participação da enzima NAD(P)H oxidase no processo de secreção de insulina induzido pela glicose pois a inibição da enzima pelo DPI e oligonucleotídeo anti p47PHOX promoveu uma diminuição da secreção do hormônio. A partir desse dado passamos a avaliar o mecanismo de ação da enzima no processo secretório e demonstramos que a inibição dessa enzima promove uma inibição de genes essenciais no processo de secreção de insulina como GLUT-2 e glicocinase.Assim podemos concluir que a enzima NAD(P)H oxidase é modulada pela glicose, palmitato e interleucina 1? e que essa enzima participa do processo de secreção insulina modulando genes essenciais para o processo secretório como GLUT-2 e glicocinase. / The expression and activity of the componenents of NAD(P)H oxidase in pancreatic islets were described for the first time in our laboratory (OLIVEIRA, HR et ai, 2003). It was shown the gene and protein expression of the components of this enzyme in Seta cells and that enzyme activation is mediated by glucose. Glucose induced insulin secretion was followed by increase in EROS generation and this increase was in part mediated by NAD(P)H oxidase activation (the same mechanism observed in phagocytes). In this study, the modulation of NAD(P)H oxidase activity by glucose, palmitate and interleukin 1ß as investigated through protein expression of p47phox vity of this enzyme through superoxide and hydrogen peroxide production. To determinate the role of NAD(P)H oxidase in the process of glucoseinduced insulin secretion the enzyme was inhibited by DPI and oligonucleotide anti p47phox, in the both cases the enzyme inhibition produced a decrease on insulin secretion. In order to investigated NAD(P)H oxidase mechanism of action in insulin secretion, we shown that the inhibition enzyme by DPI reduced the GLUT-2 and glucokinase gene expression. We can concluded hat NAD(P)H oxidase was modulated by glucose, palmitate and interleukin 1ß and that enzyme participed in process of glucoseinduced insulin secretion through modulation of GLUT-2 and glucokinase gene expression.

Nouveaux analogues de substrats de déshydrogénases pour le développement d’interfaces enzymes/électrodes innovantes / New synthetic substrates used by dehydrogenases for the development of innovative enzyme/electrode interfaces

Les systèmes bioélectroniques tels que les biopiles enzymatiques nécessitant souvent l'utilisation des assemblages moléculaires complexes comprenant le cofacteur de l'enzyme, des agents de couplage et des médiateurs électrochimiques. Afin de les simplifier, nous avons remplacé ces différents partenaires par 13 analogues simples à synthétiser après identification par criblage in silico. Le noyau aromatique est couplé à un noyau aromatique et puis un médiateur électrochimique est couplé à celui-ci. Les produits sont des poudres de couleurs variées (rose, rouge). Le rendement de la première étape est de 83% avec une pureté d'environ 92%. Le rendement de la seconde étape est compris entre 45% et 65% avec une pureté de 97%. Ces analogues ont été caractérisés chimiquement (RMN, spectrométrie de masse) et électrochimiquement (voltammétrie cyclique et spectroélectrochimie). Les activités de deux enzymes, la formiate déshydrogénase (FDH) et l'alcool déshydrogénase de foie de cheval (HLADH), et d'un catalyseur organométallique, le [Cp*Rh(bpy)(H2O)]2+, ont été évaluées avec ces analogues. De faibles activités ont été observées en présence de l'HLADH avec 4 analogues et en présence de la FDH avec un seul analogue. Au contraire aux enzymes, la réduction d'un médiateur a pu été confirmée en présence du catalyseur [Cp*Rh(bpy)(H2O)]2+ par voltammétrie cyclique. La FDH native n'est pas adaptée à fonctionner avec ces nouveaux substrats solubles dans un LI, le [MMIm][Me2PO4]. Une FDH tolérante (N187S/T321S) au [MMIm][Me2PO4] précédemment obtenue par évolution dirigée a été donc étudiée en isolant les simples mutants N187S et T321S. Le double mutant N187S/T321S et le simple mutant N187S sont 4 fois plus actifs en solution aqueuse et en présence de LI. Des analyses par spectroscopie de fluorescence ont montré que la simple mutation N187S favorise la stabilité du dimère de FDH en modifiant le pKa de l'acide aminé E163. Celui-ci est impliqué dans la thermostabilité et la tolérance des FDHs aux LIs / Bioelectrical systems, such as enzymatic biofuel cells, often require a molecular construction complex comprising the enzyme cofactor, intermediary molecules and electrochemical mediators. In order to simplify them, we have replaced these different partners by 13 analogs that are simple to synthesize after identification by screening in silico. The nicotinamide ring is coupled to an aromatic moiety and an electrochemical mediator is then coupled to it as well, resulting in various colored powders (pink, red). The first step' s yield is around 83% with a purity of approximately 92%. The second step's yield is comprised between 45% and 65% with a purity of 97%. The analogs were characterized chemically (NMR, mass spectrometry) and electrochemically (cyclic voltammetry, spectroelectrochemistry). The activities of two enzymes, the formate dehydrogenase (FDH) and the horse liver alcohol dehydrogenase (HLADH), and an organometallic catalyst, [Cp*Rh(bpy)H2O]2+, were evaluated with these analogs. Weak activities were observed for 4 analogs using the HLADH and 1 analog using the FDH. Unlike the enzymes, the reduction of a conjugated mediator was confirmed with the catalyst [Cp*Rh(bpy)H2O]2+ using cyclic voltammetry. The wild type FDH is not adapted to function with these new substrates, which can be solubilized in an IL such as [MMIm][Me2PO4]. An FDH (N187S/T321S) shown to be tolerant to [MMIm][Me2PO4], and obtained previously by directed evolution, was studied by isolating the two single mutants, N187S and T321S. The double mutant N187S/T321S and the mutant N187S are 4 times more active in aqueous solution and in [MMIm][Me2PO4]. Fluorescence spectroscopy analyses showed that the single mutation N187S favorises FDH dimer stability by modifying the pKa of the amino acid E163. The latter is involved in FDH thermal stability and tolerance in ILs

The diploma thesis provides comprehensive insight into the individual types of services of tourism. The main goal is evaluation of the quality of provided tourism services in the selected locality Hluboka nad Vltavou. In the theoretical part the concept associated with areas of tourism, services, marketing mix, quality and instruments of strategic marketing environment. In the practical part it was made detailed analysis of accommodation services, catering services, cultural services, sports services, sport and relaxation services and complementary services. In the practical part are also evaluated results of a research, which main aim was find out the satisfaction of tourism participant with a quality, level and range of tourism services, which are provided in the locality. The visitor's opinion on opportunities of cultural and sport activities is also very important. Attention was focused on the local and foreign participants of tourism in this direction. Based on the gained information and results of marketing research, the last section contains appropriate suggestions.
