Previous issue date: 2018-03-20 / A moradia na cidade manifesta-se como uma tem?tica para os estudos geogr?ficos, dentre outros motivos, por marcar a paisagem de diferentes formas. Nesse contexto, os condom?nios horizontais fechados aparecem como formas ?exclusivas? de morar, ampliando as possibilidades para a acumula??o de capital, no processo de produ??o do espa?o urbano. Em Campina Grande-PB, esse tipo de moradia configurou-se a partir dos anos 2000 como uma das estrat?gias utilizadas pelo mercado imobili?rio para uso do solo urbano voltado para a obten??o de lucro e sobrelucros. Este estudo tem por objetivo analisar a configura??o da produ??o/organiza??o do solo urbano de Campina Grande, considerando a a??o do mercado imobili?rio na estrutura??o dos condom?nios horizontais fechados. A base te?rica est? assentada nas contribui??es de David Harvey (1980, 2005, 2011, 2013), quando discute o processo de produ??o do espa?o urbano a partir da l?gica de acumula??o do capital e da renda da terra, e nas an?lises de Ribeiro (2015), que coloca a moradia como mercadoria situada na din?mica que envolve uso e troca, mercado e consumo. Essas contribui??es est?o amparadas pelas an?lises de base dial?tica, sendo a principal forma de abordagem a pesquisa qualitativa, que se deu a partir de procedimentos metodol?gicos como: an?lise do Plano Diretor Municipal de Campina Grande e de documentos encontrados no site da Secretaria de Planejamento da cidade, observa??es de campo, an?lise de an?ncios publicit?rios dos condom?nios e dados fornecidos pelo Instituto Brasileiro de Geografia e Estat?stica (IBGE), tamb?m alguns dados encontrados em trabalhos acad?micos j? desenvolvidos. Dessa forma, esses procedimentos nos permitem aprofundar o entendimento de como os condom?nios horizontais fechados se inserem na l?gica do mercado imobili?rio na cidade, utilizando estrategicamente as vantagens de localiza??o e monop?lio para ampliar as possibilidades de neg?cio da moradia e de uso do solo urbano no contexto social e espacial de Campina Grande. / The dwelling in the city manifests itself as a theme for geographic studies, among other reasons, for marking the landscape in different ways. In this context, the closed horizontal condominiums appear as a ?new? way of living, expanding the possibilities for the accumulation of capital in the process of production of the urban space. In Campina Grande-PB, this type of housing was configured from the 2000s as one of the strategies used by the real estate market to use urban land for profit and surpluses. This study aims to analyze the configuration of the urban land production/organization of Campina Grande considering the real estate market action in the structuring of the closed horizontal condominiums. The theoretical basis is based on the contributions of David Harvey (1980, 2005, 2011, 2013) when he discusses the process of production of urban space from the logic of accumulation of capital and income of the land and the analyzes of Ribeiro (2015) that places the dwelling as a commodity situated in the dynamics that involves use and exchange, market and consumption. These contributions are supported by dialectical analyzes and the main approach to qualitative research is the methodological procedures such as: analysis of the Municipal Master Plan of Campina Grande and documents found on the website of the City Planning Department, observations field analysis, commercial advertisements analysis of condominiums and data provided by Instituto Brasileiro de Geografia e Estat?stica (IBGE), also some data found in academic papers already developed. Thus, these procedures allow us to deepen the understanding of how closed horizontal condominiums fit into the logic of the real estate market in the city, strategically using the advantages of location and monopoly to expand the business possibilities of housing and urban land use in the context social and spatial development of Campina Grande.

Previous issue date: 2015-09-03 / Pesquisas recentes relacionadas ao desenvolvimento sustent?vel, mostram que a expans?o das constru??es nas cidades precisa ser melhor analisado, pois seu impacto ao meio ambiente e ? vida das pessoas ? preocupante e devem ser colocados como prioridade para as propostas de mudan?as. Portanto, faz-se necess?rio avaliar os empreendimentos existentes no meio urbano, relacionando-os as solu??es menos causadoras de tais impactos. Para isto, foi verificada a exist?ncia de condom?nios horizontais com propostas de marketing, destacando a ampla ?rea verde e uma infraestrutura que proporciona uma excelente qualidade de vida aos moradores. Assim, foi feito um levantamento dos condom?nios mais recentes, constru?dos na cidade de Natal e suas ?reas de expans?o, e que apresentam este tipo de publicidade, classificando-os desta forma como os condom?nios de alto padr?o. A sele??o dos condom?nios avaliados teve como crit?rios, o tempo de constru??o, os empreendimentos finalizados a partir de 2009, estando mais recente ?s novas tecnologias sustent?veis. Bem como a escolha de um condom?nio para fazer um contraponto, sendo ele, o primeiro condom?nio constru?do na cidade. Ao final, foram selecionados sete condom?nios para a avalia??o, sendo cada localizado em bairros de regi?es diferentes, possibilitando, desta forma, viabilizar a pesquisa. A avalia??o da qualidade ambiental dos condom?nios ? feita com base nos atributos desenvolvidos por Valaski (2008), os quais avaliam as edifica??es existentes, como tamb?m o condom?nio no aspecto geral. Os resultados obtidos na avalia??o geral mostraram que dois, dos sete empreendimentos, foram classificados positivamente em rela??o ? qualidade ambiental, entre eles o primeiro condom?nio implantado na cidade. Os demais mostraram que existe a necessidade de um maior estudo que anteceda a concep??o de um condom?nio horizontal. Em rela??o a avalia??o espec?fica das edifica??es foi poss?vel encontrar muitas casas portadoras de solu??es sustent?veis, tais como aquecimento solar de ?gua, e outras poucas, portadoras de energia alternativa por pain?is fotovoltaicos. O Armazenamento e uso de ?guas de chuvas mostrou-se um item crescente nas novas edifica??es, enquanto que, o reaproveitamento de ?guas servidas, juntamente com coberturas jardins e vegeta??es em fachadas n?o foram muito aplicadas. / Recent research related to sustainable development, show that the expansion
of the buildings in the cities needs to be better analyzed, because its impact on
the environment and to people's lives is worrying and must be made a priority
for the proposed changes. Therefore, it is necessary to evaluate the existing
facilities in the urban areas, linking them solutions less causing such impacts.
For this, it was verified the existence of condominiums with marketing
proposals, highlighting the large green area and an infrastructure that provides
an excellent quality of life for residents. Thus, a survey was made of the latest
apartments built in Natal and their areas of expansion, and presenting this type
of advertising, classifying them this way upscale condominiums. A selection of
assessed condominiums had as criteria, construction time, the projects
completed from 2009, being the latest to new sustainable technologies. And the
choice of a condominium to make a counterpoint, being the first condominium
built in the city. At the end of seven condominiums were selected for evaluation,
each being located in different neighborhoods regions, enabling, in this way,
enabling the search. The evaluation of the environmental quality of
condominiums is based on attributes developed by Valaski (2008), which
assesses the existing buildings, as well as the community in general
appearance. The results of the overall evaluation showed that two of the seven
projects were classified positively in relation to environmental quality, including
the first condominium deployed in the city. The other, showed that there is a
need for further study to precede the design of a horizontal condominium.
Regarding the specific assessment of the buildings could not find many homes
carriers sustainable solutions, such as solar water heating, and a few other,
bearers of alternative energy by photovoltaic panels. The storage and use of
rain water proved to be a growing item in new buildings, while the reuse of
wastewater, along with toppings gardens and vegetation in facades were not
very applied.

utilizados no tratamento de depress?o e ansiedade em v?rios est?gios da vida do
indiv?duo, inclusive durante a gravidez ou lacta??o. Nessa circunst?ncia, com o feto in
utero ou em amamenta??o, o mesmo ser? exposto a influ?ncia da hiperestimula??o
seroton?rgica capaz de desorganizar o desenvolvimento morfofuncional do SNC. A
modula??o do circuito seroton?rgico no c?rebro em desenvolvimento pode alterar a
organiza??o e forma??o de diferentes redes neurais espec?ficas e repercutir na
express?o comportamental do indiv?duo. Neste sentido, utilizamos o ISRS ? fluoxetina (dose: 10mg/kg)? no per?odo de desenvolvimento PND7PND21, para investigar poss?veis altera??es persistentes na
neuroqu?mica e no comportamento de ratos machos e f?meas durante as idades p?snascimento
de 45 (PND45) e 90 dias (PND90). Outros tr?s grupos experimentais foram
utilizados como controle da administra??o farmacol?gica, s?o eles: Naive (animais n?omanipulados),
Sham (animais residentes nas gaiolas de tratamento sem manipula??o)
e Veh (animais residentes nas gaiolas de tratamento que recebiam inje??o de ?gua
destilada). Nosso trabalho re?ne um conjunto de dados comportamentais
complementares e tamb?m in?ditos relacionados a avalia??o do comportamento
mnem?nico, da ansiedade e tipodepressivo
nos animais nessas diferentes idades e em
ambos os sexos. Aqui observamos aumento na ansiedade (avaliada no teste de campo
aberto), altera??es de mem?ria de curto prazo (atrav?s dos paradigmas de
reconhecimento de objetos e alterna??o espont?nea), bem como manifesta??o do
comportamento tipodepressivo
(usando os testes de prefer?ncia de sacarose e nata??o
for?ada) na prole em idade de 45 dias. Enquanto que na idade adulta (PND90) foi
observada (atrav?s dos paradigmas comportamentais usados na avalia??o da prole
juvenil) redu??o nos n?veis de ansiedade, manuten??o das altera??es de mem?ria de
curto prazo, bem como atenua??o do perfil tipodepressivo.
Em ambas idades foram
observadas redu??o no n?mero de neur?nios parvalbuminapositivos,
contudo somente animais em idade PND45 tratados com fluoxetina apresentaram redu??o significativa e
ainda quando comparados com os grupos Naive e Sham, mas n?o ao grupo Veh. Por
isso, mais estudos s?o necess?rios para investigar os efeitos persistentes da serotonina
sobre a matura??o, prolifera??o e migra??o de interneur?nios para as regi?es corticais. Os animais expostos ao ISRS apresentam maior tra?o de altera??es nos
comportamentos de ansiedade e tipodepressivo.
Nosso trabalho exp?e altera??es
mnem?nicas decorrentes da exposi??o previa a fluoxetina, sugerindo altera??es
persistentes no circuito corticol?mbico. Estes achados sugerem que a modula??o do
circuito seroton?rgico durante per?odos cr?ticos do desenvolvimento do sistema nervoso
central pode alterar a organiza??o dos diferentes circuitos neuroqu?micos e induzir
altera??es comportamentais, as quais podem repercutir para o surgimento de dist?rbios
neuropsiqui?tricos. / Selective serotonin reuptake inhibitors (SSRIs) are widely prescribed to treat
depression, anxiety and other disorders. The developmental exposure to selective
serotonin reuptake inhibitors (SSRIs) results in persistent behavioural impairment into
adulthood. In this way, serotonergic overexpression in early life may lead to structural
and functional changes in brain circuits that control cognitive and emotional behaviour. Here, we addressed the question of how postnatal (PND7PND21)
exposure to
fluoxetine affects memory, anxietyand
depressionlike
behaviours, as well as
neurochemical markers of interneurons and serotonergic cells in brain areas related to
these behaviours in juvenile (PND45) and adult (PND90) female and male rats. In a first
stage, we analysed both female and male rat?s performances in several behavioural
tasks and investigated the expression of serotonin (5HT)
in the dorsal and median
nucleus of raphe, and parvalbumin (PV) in PFCm and hippocampus at PND45. We found
that earlylife
exposure to fluoxetine increased anxietyand
depressionlike
behaviours
(more in female compared to male animals), and induced a working memory impairment
only in the juvenile male. Afterwards, we performed behavioural and neurochemical
analysis of male and female adult rats (PND90), where we found that fluoxetine affects
only anxietyrelated
male behaviour. Also, the memory impairment (more in male than
female) and depressivelike
profile (both sexes) remained in adult age. Moreover, the
exposure to fluoxetine affect PV immunoreactivity in the hippocampus in any sex at
PND45 and PND90; however, adult animals appear to recover neurochemical deficits
observed at the juvenile age. The results revealed developmental fluoxetine effects on
juvenile behaviour that can have implications for affective disorders and mnemonic
processes. These results revealed persistent changes a sex and agemanner
related to
developmental exposure to fluoxetine, where serotonergic modulation induce differential
profile of anxietyand
depressionlike
behaviour and mnemonic impairment on female
and male rats at juvenile and adult age. Also, suggest a sexdependent
compensatory
mechanism, which it is possibly related to serotonergic sinalisation. Circuits may involve
subcortical
and cortical information processing, including subcortical limbic and possible
(pre)frontal areas. Thus, our findings suggest that serotonergic modulation during critical
periods of SNC development may alter the organisation of excitatoryinhibitory
circuit and induce behavioural changes, which may have repercussions for the onset of
neuropsychiatric disorders.

Implementing and Analyzing Single Edge Nibble Transmission (SENT) Protocol for Automotive ApplicationsUllah, Naseem January 2014 (has links)
With advancement in automotive systems, it is not just the combination of mechanical devices like in old days. Almost all the systems of today's modern car are controlled electronically by a number of ECUs (Electronics Control Unit) with the combination of sensor modules. To exchange information between the ECU and sensor modules a number of communication standards are used. The most commonly used standards are CAN, LIN, and PWM etc. The data transmission between the ECU and sensor modules can be easily established with a PWM (Pulse Width Modulation) techniques in comparison with CAN or LIN. PWM provide a convenient solution in terms of cost and performance when the data-rate is up to 10-bits. While for higher resolution data rates its performance is not satisfied. Extra effort is needed to implement diagnostic information for the integrity of data. Also, the accuracy of PWM signal is dependent on the noise voltage and channel bandwidth. In 10-bit system a single bit is represented by 4mV which face serious problem in automotive system due to the noise voltage pulses which effect the resolution of the PWM. The alternative solution for safe and high data rate which is more than 10-bit resolution is to used CAN and LIN protocols. Both CAN and LIN have availability of diagnostic modes for an ensured data transmission. Also, their capabilities for interconnecting a number of nodes (sensors-modules) on the same network can significantly reduce the wiring cost. But in automotive a number of systems need to communicate through point-to-point link, and it seem to be too expensive to used CAN and LIN for point-to-point communication because of its development complexity and wiring cost for a standalone system. To overcome these issues and to provide an alternative low-cost solution the SAE (Society of Automotive Engineers) developed a 3-wire new digital point-to-point protocol called SENT. SENT (Single Edge Nibble Transmission) Protocol is now an international standard (SAE J2716). SENT is unidirectional point-to-point communication protocol, which can be used for high resolution data transmission between sensor module and ECU. The data are transmitted by sensor module in a series of pulses each pulse is 4-bit (one nibble) long and the data are measured between two falling edges by the receiving module. There are total of nine pulses which defined the SENT frame. The first pulse is called calibration pulse, it is used for compensating to recalibrate all the other pulses in case of transmitter clock deviation, this is the best feature of SENT and can be implemented in the decoder design. This thesis work focuses on the development of SENT protocol decoder and its signal robustness analysis in comparison with the conventional PWM signal. Our first goal is to developed SENT-Protocol decoder in software on the available microcontrollers is to check how much memory foot print is used and how much the processor overhead. Two platforms have been used for this purpose. First, two implementation designs prototype were made with fixed-point and floating-point development techniques on the 32-bit platform for SENT decoder. Secondly SENT-decoder were developed with 8-bit platform and compared with the two previous designs to check how much memory foot print is used and how much is the processor overhead. Finally, the signal integrity analysis has been performed for PWM and SENT signal using spice simulation. The purpose is to check the maximum data rate limit that the PWM signal support without creating any bit error in the transmitted signal. The same data rate is then used for SENT signal to be compared with PWM signal.

Adaptation of OSEck for an FPGA-Based Soft Processor PlatformStaf, Daniel January 2007 (has links)
Integrated systems become larger and more complicated every day while time to market is shortened. Due to this, there is a need for flexible hardware platforms that use programmable logic not only for custom hardware but also for realizing embedded processors. This thesis aims to select a suitable, FPGA targeted, soft processor core and adapt the real-time operating system OSEck to run on the selected target. A study of possibilities to integrate setup and configuration of OSEck into the processor’s IDE is also performed. Studies of OSEck and the two processor candidates MicroBlaze and Nios II have been performed. The processor study showed that MicroBlaze and Nios II have a very similar architecture and both are suitable to host OSEck. MicroBlaze was chosen as target processor mainly because of more available documentation regarding operating system integration. Performance and footprint was measured with OSEck on MicroBlaze. The performance figures indicate that MicroBlaze can not be expected to have the same processing power as hard processors but works well as a control processor. To achieve high application performance, custom hardware accelerators can be connected. Integration investigations and tests have been performed with the goal of making an interface that conforms to the normal MicroBlaze design flow. OSEck has been successfully adapted to run on MicroBlaze and integration in the development environment is possible although some steps have to be done manually. Alternative integration options are discussed.

Power Amplifier Linearization Implementation Using A Field Programmable Gate ArrayMenon, Abilash 01 January 2007 (has links) (PDF)
The emphasis on higher data rates, spectral efficiency and cost reduction has driven the field towards linear modulation techniques such as quadrature phase shift keying (QPSK), quadrature amplitude modulation (QAM), wideband code division multiple access (WCDMA), and orthogonal frequency division multiplexing (OFDM). The result is a complex signal with a non-constant envelope and a high peak-to-average power ratio. This characteristic makes these signals particularly sensitive to the intrinsic nonlinearity of the RF power amplifier (PA) in the transmitter. The nonlinearity will generate intermodulation (IMD) components, also referred to as out-of-band emission or spectral re-growth, which interfere with adjacent channels. Such distortion, or so called Adjacent Channel Interference (ACI), is strictly limited by FCC and ETSI regulations. Meanwhile, the nonlinearity also causes in-band distortion which degrades the bit error rate performance. Typically, the required linearity can be achieved either by reducing power efficiency or by using linearization techniques. For a Class-A PA, simply “backing off” the input power level can improve linearity; however, for high peak to average power ration (PAPR) signals, this normally reduces the power efficiency down to 10% while increasing heat dissipation up to 90%.When considering the vast number of base stations that wireless operators need to account for, increasing power consumption, or in other words, power back-off is not a viable tradeoff. Therefore, amplifier linearization has become an important technology and a desirable alternative to backing-off an amplifier in modern communications systems. In this work, a novel adaptive algorithm is presented for predistorter linearization of power amplifiers. This algorithm uses Pade-Chebyshev polynomials and a QR decomposition followed by back substitution to find the pre-distorter coefficients.This algorithm is implemented on a Field Programmable Gate Array (Stratix 1S80).The implementation provides improved linearization and also runs the algorithm fast enough so that the adaptive part can be done quickly. Yet another challenge was the integration of a transmitter, receiver and this adaptive algorithm into a single FPGA chip and its communication with a base station. The work thus presents a novel pre-distortion implementation technique using an FPGA and a soft processor (Nios 2) which provides significant intermodulation distortion suppression.

To perform real-time telemetry, a sensor network with ZigBee technology was set and some biological signals were captured in order to validate the proposed infrastructure. This network performs the capture and transmission of data collected to base stations where a treatment system embedded in an FPGA is instructed to receive data, perform the necessary calculations and send the information obtained through an Ethernet network to a central installed in a monitoring central station. The use of a dedicated processing device such as an FPGA, provides a much greater efficiency than is typically found in general purpose processors and allows the customization of the hardware. Besides, reduces the final cost of the system. It also presented the integration of Arthron and InteliCare. Arthron is tool that works with flow distribution. This integration allows more real experiences of telemedical procedures so that you can have in a single transmission, audio, video, and biological signals. The monitoring system installed in the central station is responsible for storage and display the received data. It will display data in tables and graphs in real time and allow a team of experts make decisions and guide patients and / or their caregivers to perform a certain procedure. / Este trabalho apresenta uma infraestrutura de monitoramento para pessoas em situação de internação domiciliar através da integração de redes de sensores sem fio e processamento de dados em sistemas embarcados, possibilitando um acompanhamento em tempo de execução (online) do quadro clínico de cada paciente. Para realizar a telemetria online, uma rede de sensores com tecnologia ZigBee, foi montada e alguns sinais biológicos foram captados de forma a validar a infraestrutura proposta. Esta rede realiza a captação e envio dos dados coletados até estações base onde um sistema de tratamento embarcado em um FPGA fica encarregado de receber os dados, realizar os cálculos necessários e enviar as informações obtidas, através de uma rede Ethernet, a um sistema central de supervisão instalado em uma central de monitoramento. A utilização de um dispositivo de processamento dedicado, como um FPGA, proporciona ao sistema uma eficiência muito maior do que normalmente é encontrada em processadores de uso geral além de permitir a customização do hardware reduzindo o custo final do sistema. É apresentada também a integração da infraestrutura InteliCare com a ferramenta de distribuição de fluxos Arthron. Esta integração permite tornar mais real a experiência de colaboração em procedimentos médicos de forma que é possível ter numa mesma transmissão, áudio, vídeo e sinais biológicos. O sistema de supervisão instalado na central de monitoramento é responsável por gerenciar o armazenamento e a visualização dos dados recebidos. Ele exibe os dados em tabelas e gráficos em tempo de execução e permitirá que uma equipe de especialistas tome decisões e oriente os pacientes e/ou seus cuidadores para realizar determinado procedimento.

soft-core Nios II and FPGA for a holter monitor that implements data compression, using the
PPM Algorithm, and simulate ECG signals through the implementation of the Fourier series.
Through a holter monitor, cardiologists can obtain ECG signals, serving as the basis for the
perception of symptoms and activities of patients. These signals are captured and recorded
by monitors in periods greater than or equal to 24 hours, requiring large storage size to store
them, therefore increasing cost of the monitor. Using the PPM algorithm, a monitor holter can
considerably reduce the size of the signals stored, thus reducing storage space and cost of
device, addition to allow rapid transmission of the data. Integrating the ECG signal simulator
to the device, is possible to generate samples of ECG via the embedded system, saving time
and eliminating difficulties in obtaining signals, compared with the capture of real ECG
signals by invasive and noninvasive methods. It enables the analysis and study of normal
and abnormal ECGs. An embedded system on programmable chip (SOPC) was prototyped
with a development kit containing peripherals and FPGA chip compatible with the Nios II.
Architecture soft-core was set to compact operating system and software modules have
been successfully developed, ported and validated on this platform. / Neste trabalho, é apresentado o desenvolvimento de um sistema embarcado com
prototipagem em FPGA contendo instanciação do processador soft-core Nios II (SOPC
System on a Programmable Chip), para um monitor holter que implementa compressão de
dados, utilizando o algoritmo PPM, e simula sinais ECG através da implementação das
Séries de Fourier. Através de um monitor holter, cardiologistas podem obter sinais ECG, que
servem de base para a percepção de sintomas e atividades em pacientes, captados e
armazenados pelos monitores em períodos maiores ou iguais a 24 horas, requisitando
grandes espaços de armazenamento, aumentando, assim, o custo deste monitor. Utilizando
o PPM, o dispositivo desenvolvido poderá reduzir consideravelmente a quantidade de dados
armazenados, reduzindo, portanto, o espaço de armazenamento e o custo do dispositivo,
permitindo ainda a rápida transmissão dos dados. Integrando o simulador de sinais ECG ao
dispositivo, possibilita-se a geração de amostras de sinais eletrocardiográficos através do
sistema embarcado, economizando tempo e eliminando dificuldades na obtenção de sinais,
em comparação com a captação real de sinais ECG através de métodos invasivos e nãoinvasivos.
O mesmo permite a análise e o estudo de sinais ECG normais e anormais. Um
sistema embarcado em chip programável (SOPC) foi prototipado com uma placa contendo
periféricos e uma pastilha FPGA dotada de compatibilidade com o Nios II; a arquitetura do
soft-core foi configurada em sistema operacional compacto e módulos de software foram
exitosamente desenvolvidos, portados e validados sobre essa plataforma.

O modelo atual de neurog?nese no hipocampo adulto assume que c?lulas tronco neurais (CTN) geram progenitores com potencial restrito ? gera??o de neur?nios ou astr?citos. Est?mulos ambientais e condi??es patol?gicas podem alterar a progress?o da linhagem, modulando a prolifera??o, diferencia??o, sobreviv?ncia e integra??o sin?ptica dos neur?nios gerados. Por exemplo, a Epilepsia do Lobo Temporal mesial (ELT), a forma mais comum de epilepsia em adultos, est? associada a altera??es na taxa de neurog?nese hipocampal adulta. Neste trabalho, n?s utilizamos dois modelos experimentais de ELT para avaliar os efeitos de um insulto epileptog?nico (i.e., status epilepticus, SE) sobre a linhagem e amadurecimento celular no giro denteado adulto. Atrav?s da t?cnica de fate-mapping utilizando animais Dcx-CreERT2/CAG-CAT-GFP, n?s acompanhamos o destino de c?lulas que apresentavam o promotor do gene doublecortin (DCX) ativado antes ou depois da inje??o intrahipocampal dos agentes convulsivantes ?cido ca?nico ou pilocarpina. Desta forma, pudemos avaliar o efeito destas drogas sobre progenitores e neur?nios imaturos DCX+ gerados antes ou ap?s o tratamento. Em ambos os modelos, foram observados um aumento de neurog?nese e altera??es no posicionamento e morfologia de c?lulas granulares, conforme descri??es pr?vias na literatura. Altera??es neuronais, tais como localiza??o ect?pica e presen?a de dendritos basais, foram observadas tanto em c?lulas geradas antes quanto ap?s a indu??o do SE, embora com frequ?ncias distintas. No entanto, apenas no hipocampo ipsilateral ? inje??o de ?cido ca?nico n?s observamos dispers?o da camada granular e morte neuronal em CA1 e CA3, apesar da atividade parox?stica epil?ptica ocorrer em ambos os hipocampos. Surpreendentemente, o aumento da neurog?nese em animais que receberam ?cido ca?nico foi restrito ao hipocampo contralateral, enquanto no lado ipsilateral foi observado um significativo aumento na gera??o de astr?citos a partir dos progenitores DCX+. Al?m disso, tamb?m observamos neste modelo a presen?a de c?lulas com morfologia e marcadores de CTNs, sugerindo que progenitores DCX+ poderiam regredir para estados mais primitivos na linhagem celular do hipocampo adulto. O aumento da astrogliog?nese no lado ipsilateral ? inje??o de ?cido ca?nico foi associado a uma degenera??o de interneur?nios parvalbumina (PV)+ no hipocampo, sugerindo que a atividade gaba?rgica poderia estar contribuindo para o redirecionamento da linhagem celular. Em conjunto, nossos dados indicam que a linhagem celular no giro denteado n?o ? unidirecional e irrevers?vel, e que o aumento da atividade el?trica neuronal induzida por ?cido ca?nico e pilocarpina t?m efeitos diferentes sobre a diferencia??o celular e destino fenot?pico dos progenitores e neur?nios nessa regi?o. Esses resultados imp?em a necessidade de revermos o modelo atual de neurog?nese hipocampal adulta e tamb?m indicam que diferentes modelos animais de epilepsia produzem altera??es celulares distintas no hipocampo adulto e, portanto, poderiam representar diferentes graus/est?gios da patologia. / The granular cells of the hippocampus are one of the few types of neurons generated in the central nervous system of adult mammals. The current model of neurogenesis in the adult hippocampus assumes that neural stem cells (NSCs) give rise to progenitors restricted to the generation of neurons or astrocytes. Environmental stimuli and pathological conditions can alter the lineage progression, modulating cell proliferation, differentiation, survival and synaptic integration of newly generated neurons. For example, mesial Temporal Lobe Epilepsy (TLE), the most common form of epilepsy in adults, is associated with changes in the rate of adult hippocampal neurogenesis. In this work, we used two experimental TLE models to evaluate the effects of an epileptogenic insult (i.e., status epilepticus, SE) on the cell lineage and neuronal maturation in the adult dentate gyrus. Using Dcx-CreERT2 / CAG-CAT-GFP animals, we fate mapped the fate of cells expressing the doublecortin gene (DCX) either before or after intrahippocampal injection of the convulsive agents kainic acid or pilocarpine. In this way, we could evaluate the effect of these drugs on DCX+ progenitors and immature neurons generated before or after treatment. In both models, we observed an increase in neurogenesis and changes in the positioning and morphology of granular cells, according to previous descriptions in the literature. Neuronal aberrations, such as ectopic localization and presence of basal dendrites, were observed both in cells generated before and after induction of SE, albeit at different frequencies. However, only in the hippocampus ipsilateral to the injection of kainic acid we observed granule cell dispersion and neuronal death in CA1 and CA3, although the paroxysmal epileptic activity occurred in both hippocampi. Surprisingly, the increase in neurogenesis in animals that received kainic acid was restricted to the contralateral hippocampus, whereas on the ipsilateral side a significant increase in astrocyte generation was observed within the DCX+ progenitor lineage. In addition, we also observed the presence of cells with NSC hallmarks, suggesting that DCX+ progenitors could regress to more primitive states in the adult hippocampal cell lineage. The increased astrogliogenesis on the ipsilateral side to the injection of kainic acid was associated with a degeneration of parvalbumin (PV)+ interneurons in the hippocampus, suggesting that GABAergic activity could be contributing to the rerouting of the DCX+ progenitor cell lineage. Taken together, our data indicates that the cell lineage in the dentate gyrus is neither unidirectional nor irreversible, and that the increased neuronal electrical activity induced by kainic acid and pilocarpine have different effects on cell differentiation, as well as on the fate of progenitors and neurons in that region. These results highlight the need to review the current model of adult hippocampal neurogenesis and also indicate that different animal models of epilepsy produce distinct cellular alterations in the adult hippocampus and could therefore represent different degrees / stages of the pathology.
