Neurocorrelates of speech-motor planning and execution in adults and children who stutterBrown, Bryan T. 01 December 2015 (has links)
There is a rich literature demonstrating that adults who stutter (AWS) demonstrate atypical functional brain activity during speech production. These differences can be characterized by increased activity in the right inferior frontal gyrus and premotor regions and decreased activity in the left inferior frontal gyrus, premotor area, and bilaterally in the superior temporal gyrus. The process of speech production requires motor movements first be planned and then executed. However, few studies have examined activity related to speech-motor planning independently from speech-motor execution. Additionally, due to methodological limitations, few investigations have examined functional brain activity in children who stutter (CWS). We hypothesized that AWS and CWS would demonstrate atypical brain activity related to both speech-motor planning and execution. Using Near Infrared Spectroscopy (fNIRS), we measured the change in oxygenated hemoglobin concentration (HbO) during speech-motor planning (repetition of nonwords with three repeated or different syllables) and speech-motor execution (covert/overt naming). Results indicated that both AWS and CWS demonstrated cortical activity that was atypical during speech-motor planning processes in the right inferior frontal gyrus and atypical speech-motor execution processes in the left inferior frontal gyrus. Deactivations in the left inferior frontal gyrus may reflect inefficient feedforward mechanisms for speech production. Inefficient feedforward mechanisms will likely result in more variable movements, for which larger feedback correction signals will be necessary. Overactivations in the right inferior frontal gyrus may reflect this increased correction. Additionally, AWS demonstrated atypical speech-motor planning activity in the right middle frontal gyrus, potentially related to the production of prosody. These results are presented within a theoretical framework of two competing theories of stuttering.

Untersuchung der regionalen Gewebsoxygenierung anämischer Frühgeborener unter Transfusion von ErythrozytenkonzentratSeidel, Denise 28 August 2014 (has links)
In der Neonatologie ist die Verabreichung von Erythrozytenkonzentrat (EK) eine der häufigsten therapeutischen Maßnahmen. Ursachen dafür sind die Frühgeborenen-anämie und die iatrogen durch diagnostische Blutentnahmen hervorgerufene sekundäre Anämie. Obwohl ca. 80% der Frühgeborenen während des postnatalen stationären Aufenthaltes EK erhalten, gibt es noch immer keine einheitlichen Richtlinien zur Bluttransfusion bei Frühgeborenen.
In der vorliegenden Studie sollte der Effekt einer EK-Gabe mit Hilfe der Messung der cerebralen und peripheren Gewebsoxygenierung (crSO2 und prSO2) erfasst werden. Und es wurde der Annahme nachgegangen, dass in der Neonatologie Subgruppen existieren, welche unterschiedlich von einer EK-Transfusion profitieren.
Im Rahmen dieser Promotionsarbeit wurden Frühgeborene der neonatologischen Abteilung der Universitätsklinik und Poliklinik für Kinder- und Jugendmedizin Leipzig prospektiv analysiert. Es konnte gezeigt werden, dass bei allen untersuchten Patienten sowohl die cerebrale als auch die periphere Gewebsoxygenierung unter EK-Transfusion ansteigen und auch nach einem Intervall von 24 Stunden noch auf diesem gesteigerten Niveau konstant bleiben. Zudem konnte nachgewiesen werden, dass Kinder mit niedrigen crSO2-Ausgangswerten vor Transfusion mehr Sauerstoffsättigungsabfälle unter 80% (SaO2<80%) aufweisen. Nach Transfusion ist bei diesen Patienten zusätzlich ein stärkerer Rückgang der Anzahl der SaO2<80% zu beobachten.
Somit ist die Gewebsoxygenierung möglicherweise ein sinnvoller Parameter für die Indikation zur EK-Transfusion, welcher bei der Erarbeitung neuer Transfusionsricht-linien in der Neonatologie mit einbezogen werden sollte.

Sustainability of the soybean industry relies on the growth of new industries and the continued improvement of seeds for utilization. Grower adoption and growth of the edamame industry has been slow in part due to insufficient information on its potential profitability and marketability. As such, the first and second objectives of this thesis aimed at 1) determining production costs of hand-harvested fresh edamame enterprise and 2) exploring consumer willingness-to-pay (WTP) for fresh, local, organic, and "on-the-stalk" marketed edamame. Sucrose, raffinose, and stachyose sugars hold tremendous implications for utilization of soybean seeds in livestock, soyfood, and probiotics industries. Current sugar phenotyping methods using high-performance liquid chromatography (HPLC) are costly and inefficient. Therefore, the third objective of this study was to develop calibrations to predict sugar content using near-infrared reflectance spectroscopy (NIRS). Results showed that labor accounted for 72% of production costs for edamame pods, which largely limits its profit potential. Mean WTP for fresh and local edamame exceeded their frozen and non-local counterparts by 94 and 88 cents, respectively. In addition, mean WTP for organic edamame exceeded non-GMO edamame by 33 cents. Pro-environmental attitudes appeared to be a consistent driver of WTP these three attributes. Meanwhile, a 40-cent discount for "on-the-stalk" edamame compared to pods indicates convenience may also be a factor in edamame marketability. Calibration development for sucrose and stachyose was successful, with R2cal, R2cv, RMSEC, and RMSECV of 0.901, 0.869, 0.516, and 0.596, and 0.911, 0.891, 0.361, and 0.405, respectively. Meanwhile, on-the-stalk edamame shows poor marketing potential, likely due to preference for convenience of food preparation in the U.S. NIRS prediction models for sucrose and stachyose showed strong predictive accuracy and low error, suggesting potential for implementation. The prediction model for raffinose, however, remained poor.

En esta tesis se aborda el estudio de la grasa intramuscular como característica determinante de la calidad de la carne para ser utilizado en programas genéticos. El conejo se plantea no sólo por su interés como especie ganadera sino como modelo en otras especies. Este estudio se divide en tres experimentos, cada uno de ellos representa cada uno de los tres objetivos específicos de esta tesis:
1. Estudiar la variabilidad genética entre líneas de conejo de factores ligados directamente a la deposición de grasa tanto en músculo como en tejido adiposo, como son las enzimas que participan en la síntesis y degradación de la grasa y en la composición en ácidos grasos.
2. Puesta a punto de una calibración NIRS para estimar grasa intramuscular y la evaluación de su uso en programas de selección en conejo.
3. Selección divergente por grasa intramuscular. Estudio de las posibilidades de éxito de la selección por grasa intramuscular y conocer las relaciones genéticas entre caracteres examinando la respuesta correlacionada.
Supone el inicio de una nueva línea de investigación en metabolismo lipídico, que puede servir tanto a la producción de conejo como animal de carne como al uso del conejo como modelo experimental. Es la primera vez que se propone un experimento de selección divergente por grasa intramuscular. Este experimento permitirá conocer mejor las relaciones genéticas entre la grasa intramuscular, la grasa de la canal, así como las relaciones con otros caracteres productivos.
