Corpo nu fotografado e autofotografado em ambientes digitais : discursividades e produção de subjetividade

A circulação do corpo nu fotografado e autofotografado em ambientes digitais (como em redes sociais, blogs, aplicativos de troca de mensagem) dispara à/a produção discursiva. Um encontro entre o sujeito e elementos que possibilitam a produção e o compartilhamento daquilo que passa a ser nomeado como “nude”, um encontro pensado enquanto produtor de subjetividade no contemporâneo. Com inspiração cartográfica, um percorrer rizomático por links foi sendo traçado, possibilitando a aproximação com textos, imagens, músicas, vídeos, páginas da internet, registradas em Diários de campo. Tais registros ofereceram elementos à pesquisa, os quais foram sendo trabalhados em composição com teorizações foucaultianas, além de outros autores que pensam a produção de discursos e de subjetividade. A identificação do enunciado “manda nude” constituiu-se no processo de análise dos materiais. Tal movimento incitou a abordagem uma série de aspectos, como o conjunto de práticas em um momento histórico, possibilitam que certas imagens sejam nomeadas como nude(s) e que verdades sejam produzidas a partir disso. No âmbito desta prática, foram traçadas discussões sobre formas e forças que operam no campo, produzindo e fazendo circular discursos sobre os nudes, perpassados, por exemplo, pela questão do humor e também pela constituição de um certo tipo de “expertise”. Nas considerações finais, a produção de subjetividade agenciada e a potência de atualização na interface do corpo com as tecnologias disponíveis para o seu registro foram retomadas. / The sharing of the naked body in photographed or self-photographed in digital environments (such as in social networks, blogs, messaging applications) it triggers the discursive production. NSFW contextual influences on perception : representations of the nude in public and private spaces

Submitted in partial fulfilment of the requirements for the Masters of Technology in Graphic Design Degree, Durban University of Technology, 2017. / The aim of this research is to investigate the influence of context on the artistic exploration of the nude figure, in the work of selected artists. Qualitative research methods were used. The research is practice-led, as my art making plays an essential part in guiding my research. An outline detailing the various influences and changes to artistic practice surrounding the nude – from ancient Greece to the development of feminist ideas - provides an historic context to the study. I look at major sociocultural and technological advances to examine the relationship between artistic representations of nudity and context. This includes a look at the terminology, naked and the nude, and how these shape understanding. Contextual influences are narrowed down to how they have affected the work of selected artists’ whose work utilizes the nude as a communication tool. The selected artists that I investigate are Jenny Saville (1970-), Erin M. Riley (1985-), Vinz (1979-), and myself. In my own work, issues of public and private space are interrogated by looking at aspects of social networking and online behaviour. This exploration focuses on issues relating to the overlap of these two spaces that where once seen as dichotomous. Through this research I showed that contemporary artists still make use of the historic subject matter. By including symbolism taken from contemporary sociocultural surroundings the nude’s communicative value is continuously updated. This supports the idea that context effects the artistic exploration of the nude. / M

The toxicity, pharmacokinetics, anti-inflammatory and anti-tumour properties of a methotrexate polymer

A major effort to develop anticancer drugs through both empiric screening and rational design of new compounds has been under way for over 30 years (Katzung, 2004). In recent years, research and development in the field of sitespecific drug therapy has progressed significantly. Safe and non-toxic formulations of cytotoxic drugs based on polymers with their improved sitespecific delivery and effective activation to biologically active cytotoxic compounds at the targeted tumours have become a promising approach to cancer therapy. Drug delivery systems based on polymer micelles, coated microand nanoparticles, liposomes and various pro-drug systems including watersoluble polymer–drug conjugates and immunoconjugates have been prepared and extensively studied as novel drug delivery systems designed for cancer chemotherapy. Amongst these drug delivery systems that enable specific drug delivery and release, water-soluble polymer–drug conjugates rank among the most promising, versatile and efficient systems. This dissertation reviews the preclinical testing and pharmacokinetic study of D85, a novel water-soluble macromolecular pro-drug that is a polymer with pHcontrolled methotrexate (MTX) release with potential for treatment of cancer in humans (Ulbrich&Subr, 2004). As MTX is also indicated in low doses for the treatment of chronic inflammatory conditions, the polymer was further tested in an acute inflammatory model to determine whether the polymer would be more effective than MTX in controlling inflammation. The objective of this study was to compare the potency and efficacy of D85 to MTX. D85, a MTX conjugated polymeric lead compound, was designed and synthesised as a potential anti-neoplastic and anti-inflammatory agent. It was initially tested in vitro on three different cancer cell lines where selective toxicity towards the cancer cell cultures compared to primary cell cultures and greater toxicity than MTX was observed. The initial in vitro tests showed very promising results with D85 demonstrating approximately 300 times greater cytotoxicity than MTX against a colon cancer cell line (COLO 320 DM). This high cytotoxic effect warranted further investigation in an in vivo colon cancer tumour model. An induced murine tumour model of COLO 320 DM was successfully developed in nude mice, and the anti-tumour efficacy of D85 tested in this model. The maximum tolerated dose of D85 was established by carrying out an in vivo dose ranging toxicity test in BALB/c mice. The anti-inflammatory effects of D85 were also determined using the carrageenan-induced paw oedema model in rats where carrageenan was injected into a footpad of a rat causing acute oedema, which was measured using a water displacement plethysmometer. D85 was found to exacerbate the inflammatory response. Finally, the pharmacokinetic parameters of MTX and D85 were assessed using a LC/MS/MS method specifically developed and validated to determine low concentrations of MTX in small volumes of plasma. This new method made use of online solid phase extraction and sample cleanup on 2μl injections of diluted plasma allowing an entire pharmacokinetics study to be completed on an individual rat. Cancer is one of the leading cause of death in modern world, so there is an emerging demand for better diagnostic tools and more specific less toxique therapeutics. Nanoparticles offers characteristics that could fullfill such perspectives. They can easily target tumor by ehanced permeation and retention effect (EPR). Nanoparticles can combine more than one imaging properties, so we can say that they are multimodal, some of them could combine diagnostic and therapeutic molecules in one nanoparticle, which is now highly popular topic of nanoparticles for theranostics . The aim of this thesis was to characterize new multimodal glycogen-based nanoparticle. Glycogen is an ideal structure for nanoparticle design. Glycogen is part of natural dendrimers group which are easily to modify. Glycogen's size is suitable for EPR effect. We have evaluated biological characteristics of five different types of modified glycogen. The in vitro experiments were carried on HepG2 cells. We have set time curve of cellular uptake of this glycogen probes, evaluated cytoplasmatic localization and for the first time we have carried MTT assay. This study is an analysis of how the artworks Övermålning, God Giving a Fuck and Lesbisk häxhora by Carolina Falkholt can be interpreted and received by the surrounding society. To do this, the study uses a semiotic image analysis and a reception history analysis. The study has its base in a feminist perspective as well as in art sociology, to further deepen the analysis with different perspectives and in connection to the society and societal norms. The result of this study shows that there are multiple ways to interpret the motifs of the artworks by Carolina Falkholt. They can be seen as a political statement to shed light on feminist struggles and the beauty of the nude female body and the vagina, but also as actively provocative art where the female body is violated as a result of being displayed in a public setting. However, the study also discusses how the works of Carolina Falkholt have previously been renowned and praised by art institutions, and how her work continues to find success, with added inquiries of her work in public settings, despite regulations. Therefore, the study concludes that the artworks by Carolina Falkholt is qualified to be seen as art that advances and favor Swedish culture, regardless of the negative feedback the art has received.

Methods and mechanisms to improve endothelial colony forming cell (ECFC) survival and promote ECFC vasculogenesis in three dimensional (3D) collagen matrices in vitro and in vivo

Indiana University-Purdue University Indianapolis (IUPUI) / Human cord blood (CB) derived circulating endothelial colony forming cells (ECFCs) display a hierarchy of clonogenic proliferative potential and possess de novo vessel forming ability upon implantation in immunodeficient mice. Since survival of ECFC post-implantation is a critical variable that limits in vivo vasculogenesis, we tested the hypothesis that activation of Notch signaling or co-implantation of ECFC with human platelet lysate (HPL) would enhance cultured ECFC vasculogenic abilities in vitro and in vivo. Co-implantation of ECFCs with Notch ligand Delta-like 1 (DL1) expressing OP9 stromal cells (OP9-DL1) decreased apoptosis of ECFC in vitro and increased vasculogenesis of ECFC in vivo. The co-culture of ECFC with HPL diminished apoptosis of ECFC by altering the expression of pro-survival molecules (pAkt, pBad and Bcl-xL) in vitro and increased vasculogenesis of human EC-derived vessels both in vitro and in vivo. Thus, activation of the Notch pathway by OP9-DL1 stromal cells or co-implantation of ECFC with HPL enhances vasculogenesis and augments blood vessel formation by diminishing apoptosis of the implanted ECFC. The results from this study will provide critical information for the development of a cell therapy for limb and organ re-vascularization that can be applied to recovery of ischemic tissues in human subjects.
