Sistema de estresse e uso de substâncias : efeito de polimorfismos no gene CRHR1 nas dependências de crack e nicotina

Os Transtornos por Uso de Substâncias (TUS) são propostos pela Associação Norte-Americana de Psiquiatria e englobam diversas dependências químicas, incluindo o tabagismo e a dependência de crack. O uso de crack tem aumentado consideravelmente nos últimos anos, segundo dados de levantamentos brasileiros e mundiais. Com mecanismo de ação semelhante ao da cocaína, porém com efeito mais rápido, o crack é uma droga que causa grave prejuízo a seus usuários e conduz à dependência em pouco tempo. O tabagismo também é uma condição bastante prevalente no mundo inteiro, sendo que os prejuízos relacionados a esse transtorno envolvem principalmente o desenvolvimento de doenças respiratórias e câncer. Diversos trabalhos têm buscado identificar fatores de suscetibilidade a esses transtornos, sendo que estudos de associação envolvendo genes do sistema de resposta a estresse vêm ganhando importância. O eixo Hipotálamo- Pituitária-Adrenal (HPA) é o sistema de resposta a estresse mais conhecido em humanos e alvo do presente trabalho, uma vez que o funcionamento dessa cascata de sinalização da produção de cortisol pode desempenhar um papel relevante no desenvolvimento de psicopatologias. Além disso, o eixo HPA pode ser considerado um mediador da relação entre situações adversas na infância e o surgimento de TUS. O objetivo desse trabalho, portanto, é investigar o papel de polimorfismos no gene CRHR1 (que codifica o receptor tipo 1 do hormônio liberador de corticotrofina – CRH, hormônio inicial do eixo HPA) sobre o desenvolvimento e características clínicas da dependência química. Para isso, foram realizados dois estudos, envolvendo amostras de mulheres. No primeiro artigo, a partir de três amostras – (1) 136 usuárias de crack, (2) 55 tabagistas e (3) 262 voluntárias saudáveis – foram identificadas diferenças de freqüências alélicas e genotípicas do SNP rs12944712; além disso, a análise de haplótipos envolvendo tal SNP e o rs110402 revelou maior frequência do haplótipo contendo o alelo G nos dois polimorfismos nos grupos de usuárias de crack e tabagistas. No segundo artigo, foi avaliado o efeito da interação geneambiente envolvendo os mesmos polimorfismos e situações adversas na infância – estimada pela escala Childhood Trauma Questionnaire (CTQ) –sobre características clínicas da amostra de usuárias de crack, como craving e gravidade da dependência – estimados, respectivamente, pelas escalas Cocaine Selective Severity Assessment (CSSA) e Addiction Severity Index (ASI6). A análise não revelou uma interação significativa. A presente dissertação, portanto, revela um efeito do gene CRHR1 sobre o desenvolvimento de TUS, embora esse efeito pareça não se manifestar sobre a gravidade. Mais estudos são necessários visando esclarecer o real efeito do gene e do eixo HPA como um todo sobre os fenótipos e comportamentos aditivos, bem como os mecanismos subjacentes a essa relação. / The Substance Use Disorders (SUD) are proposed by the American Psychiatric Association and comprise several addictions, including tobacco and crack cocaine smoking. The prevalence of crack cocaine dependence has grown considerably, according to survey data from Brazil and worldwide. With mechanism of action similar to cocaine but with faster effect, crack is a drug that causes severe impairments to its users and leads to dependence in a short time. Tobacco smoking is also a highly prevalent condition worldwide, and the impact related to this disorder includes cancer and respiratory diseases. Several studies have attempted the identification of susceptibility factors to these disorders, and association studies involving stress response system genes have been performed. The Hypothalamic-Pituitary-Adrenal (HPA) axis is the best known stress response system, and the signaling cascade production of cortisol may play a role in the development of psychopathology. In addition, HPA axis can be considered a mediator of the relationship between early life adversity and SUD. The aim of this study is to investigate the role of polymorphisms in CRHR1 gene (encoding the corticotrophin releasing factor – CRH – receptor type 1, an initial factor in HPA axis response) on the development and clinical traits of drug dependence. Two studies were conducted on women samples. In the first article, three groups were compared - (1) 136 crack smokers, (2) 55 tobacco smokers and (3) 262 healthy volunteers. Differences in allele and genotype frequencies of the SNP rs12944712 were identified; in addition, the haplotype analysis involving this SNP and rs110402 revealed a higher frequency of the haplotype containing G allele in both polymorphisms in the groups of nicotine dependents and crack smokers. In the second article, we evaluated the effect of a gene-environment interaction involving the same polymorphisms and childhood adversity - estimated by Childhood Trauma Questionnaire (CTQ) scale – on clinical characteristics of the sample of crack users, such as craving and severity of dependence - estimated respectively by the Cocaine Selective Severity Assessment (CSSA) and Addiction Severity Index (ASI6) scales. The analysis did not reveal a significant interaction. This dissertation thus shows an effect of CRHR1 genepolymorphism on the development of SUD, although this effect does not seem to impact disorder severity. More studies are needed in order to clarify the actual effect of the gene and of the HPA axis as a whole on addictive behaviors.

Hypothalamic pituitary adrenal axis dysregulation in obese pregnancy

There has been a global rise in obesity in the last three decades, and at present one in five women are obese at antenatal booking. Maternal obesity is associated with an increased risk of adverse pregnancy outcomes, including increased fetal size and prolonged pregnancy. In the longer-term, offspring of obese are at increased risk of premature death from a cardiovascular event in their adulthood. One mechanism that has been linked to these outcomes is fetal exposure to glucocorticoids in utero. During normal pregnancy, the maternal hypothalamic pituitary adrenal (HPA) axis undergoes major changes, resulting in exponentially increasing levels of the major circulating glucocorticoid cortisol, and other HPA axis hormones, such as corticotrophin releasing hormone (CRH). Cortisol and CRH are vital for normal fetal growth and length of gestation, but in excess they are associated with fetal growth restriction and preterm labour. In non-pregnant obesity, it is thought that the HPA axis is dysregulated, although evidence is inconclusive. Little is known about the effects of maternal obesity in pregnancy on the HPA axis. The work in this Thesis used clinical studies to test the hypothesis that the HPA axis is dysregulated in obese pregnant women with altered release, clearance and placental metabolism of cortisol. Associations with clinical outcomes related to fetal size and length of gestation were also studied. The HPA axis activity during pregnancy was investigated in a prospective case-control study cohort. Fasting serum cortisol levels were measured at 16, 28 and 36 weeks of gestation (obese n=276, lean n=135). In a subset (obese n=20, lean n=20), corticosteroid binding globulin (CBG), CRH, estrogens and progesterone were measured. Salivary cortisol was measured in samples collected at bedtime, waking and 30 minutes after waking at 16 weeks. Urinary glucocorticoid metabolites were measured at 19 weeks and 36 weeks (obese n=6, lean n=5) and non-pregnant (obese n=7, lean n=7) subjects. All circulating hormone levels rose similarly in obese and lean during pregnancy, but were significantly lower in obese women. The diurnal rhythm of cortisol was maintained. Urinary glucocorticoids increased with gestation in lean, but not in obese, indicating a lesser activation of the HPA axis in obese compared with lean pregnancy. These findings associated with increased birthweight and longer gestation in obese pregnancy, suggesting that decreased HPA axis activity may underlie these obese related adverse pregnancy outcomes. Whether or not lower glucocorticoids in obese pregnancies are maintained at delivery was investigated by measuring active glucocorticoids (cortisol and corticosterone) and their inactive versions (cortisone and 11- dehydrocorticosterone, respectively) from matched maternal and cord plasma samples (n=259, BMI 18 – 55 kg/m2). Active glucocorticoids were significantly higher in maternal than cord blood, and inactive versions were significantly higher in cord than maternal blood. Increased maternal BMI associated with lower maternal cortisol, corticosterone and 11-dehydrocorticosterone. Despite significant correlations between maternal and cord blood glucocorticoid levels, increased maternal BMI did not associate with lower cord blood glucocorticoids. This suggests that conditions at delivery may overcome any potential negative effects of low maternal glucocorticoids on the fetus in the short-term. However, it may not preclude the longer-term effects of fetal exposure to lower glucocorticoid levels during obese pregnancy, and offspring follow-up studies are required. Potential mechanisms leading to altered HPA axis activity in obese pregnancy were explored by studying the pulsatile release and placental metabolism of glucocorticoid hormones. Glucocorticoid pulsatility is thought to be important for transcriptional regulation of glucocorticoid responsive genes, and disruptions to pulsatility have been reported in some disease processes. Glucocorticoids were measured in 10-minute serum sampling between 08.00h-11.00h and 16.00h- 19.00h. Peripheral tissue cortisol was measured from 20-minute sampling of interstitial fluid, over 24-hours, at 16-24 weeks and 30-36 weeks (obese n=7, lean n=8), and non-pregnant controls (obese n=4, lean n=3). Total circulating serum cortisol levels were higher in pregnancy than non-pregnancy in lean and obese, and increased significantly with advancing gestation in lean but not in obese. Pulsatility of cortisol was demonstrated in interstitial fluid in both non-pregnancy and pregnancy. In obese pregnancy, interstitial fluid pulse frequency was lower with advancing gestation. This may be a novel mechanism underlying the observed decreased HPA axis activity in obese pregnancy. Placental cortisol metabolism and transport was studied using an ex vivo placental perfusion model, perfused with a deuterium-labelled cortisol tracer combined with computational modeling. The findings challenge the concept that maternal cortisol diffuses freely across the placenta, but confirmed that 11β- HSD2 acts as major ‘barrier’ to cortisol transfer to the fetus, protecting the fetus from the high maternal circulating cortisol levels. In addition we showed preliminary evidence of local cortisol production within the placenta. The model is able to predict maternal-fetal cortisol transfer and can now be used in future experimental design. In conclusion, in obese pregnancy, lower maternal cortisol and urinary clearance suggested reduced HPA axis activity. Altered glucocorticoid pulsatility may underlie this change. Future studies of placental cortisol metabolism in maternal obesity could be conducted using an ex vivo perfusion model. The lower HPA axis activity in obese pregnancy represents a novel pathway underlying increased fetal growth.

Orientador: Wílson Mello Júnior / Resumo: O carcinoma de bexiga urinária é o tipo de câncer mais prevalente no sistema urinário em seres humanos e apresenta altos índices de morbidade e mortalidade. Seu desenvolvimento está relacionado a diversos fatores de risco, como o hábito de fumar, a exposição a agentes químicos industriais e metais pesados e o estresse crônico. As experiências adversas no início da vida estão relacionadas à modulação de eixos hormonais importantes, como o hipotalâmicohipofisário-adrenal (HHA) e hipotalâmico-hipofisário-gonadal (HHG). Alterações na resposta do eixo HHA, componente central da resposta fisiológica ao estresse, levam a desordens fisiológicas, como aumento da proliferação celular, angiogênese e prejuízo da resposta imune, que podem ser responsáveis pelo aumento da susceptibilidade do indivíduo a uma série de doenças na vida adulta, incluindo o câncer. Paralelamente, alterações na fisiologia de hormônios esteroides como andrógenos, estrógenos e glicocorticoides, bem como seus receptores, também têm sido relacionadas ao desenvolvimento de enfermidades como o câncer de bexiga urinária. Em ratos, o comportamento materno funciona como um fator importante na modulação desses eixos hormonais, modificando suas respostas na vida adulta, e estudos recentes têm relacionado os cuidados recebidos por filhotes à incidência de tumores. Its development is related to several risk factors such as smoking, exposure to industrial chemicals and heavy metals and chronic stress. Adverse experiences early in life are related to modulation of important hormonal axes, as the hypothalamic-pituitary-adrenal (HPA) and hypothalamic-pituitary-gonadal (HPG). Changes in the response of the HPA axis, a central component of the physiological response to stress, lead to physiological disorders, such as increased cell proliferation, angiogenesis, and impaired immune response, which may be responsible for the increased susceptibility of the individual to a number of diseases in adult life, including cancer. Similarly, changes in the physiology of steroid hormones such as androgens, oestrogens and glucocorticoids and their receptors have also been related to the development of diseases such as bladder cancer. In rats, maternal behavior serves as an important factor in the modulation of hormonal axes, modifying their responses in adulthood, and recent studies have linked the care received by pups and the incidence of tumors. Padrão diurno de secreção de cortisol e manifestações psicológicas do estresse em profissionais de enfermagem. / Diurnal pattern of cortisol and psychological manifestations of stress among nursing

Introdução: A exposição frequente dos profissionais de enfermagem a estressores relacionados ao trabalho tem sido amplamente descrita na literatura. Entretanto, a magnitude e intensidade do estresse dependem não somente dos estressores, mas também da interação destes com a avaliação cognitiva da situação estressora, com os recursos de enfrentamento e com a reação psiconeuroendócrina do estresse. Embora diversos estudos tenham descrito as reações psicológicas do estresse e seu enfrentamento nos profissionais de enfermagem, pouco se sabe sobre as características da reação neuroendócrina. Isto é particularmente importante, dado que padrões atípicos de secreção diurna de cortisol, principal hormônio do estresse, estão associados ao aumento da susceptibilidade ao desenvolvimento de doenças cardiovasculares, imunológicas e transtornos mentais. Assim, questiona-se a frequência de padrões atípicos de cortisol e sua relação com manifestações psicológicas nestes profissionais. Objetivo: Analisar o padrão diurno de secreção de cortisol dos profissionais de enfermagem de unidades hospitalares. Método: Foram incluídos 56 profissionais de enfermagem randomicamente selecionados, alocados nas unidades ambulatório, clínica médica, clínica cirúrgica, centro cirúrgico, pronto socorro infantil e adulto, unidade de terapia intensiva adulto e pediátrica de um hospital universitário. Para avaliação do padrão diurno de secreção de cortisol foram coletadas amostras de saliva em dois dias úteis consecutivos de trabalho e, para as manifestações psicológicas, foram aplicados os instrumentos escala de estresse percebido (EEP), questionário de sofrimento mental (SRQ-20), inventário de depressão de Beck (IDB) e escala de estresse no trabalho (EET). Os dados foram armazenados e analisados utilizando o programa estatístico SPSS versão 14.0 e o nível de significância adotado foi de 5%. Resultados: Quanto ao padrão de secreção de cortisol, 42,5% dos profissionais de enfermagem apresentaram padrão atípico de secreção de cortisol, sendo que 19,5% eram técnicos de enfermagem. Quanto às variáveis psicológicas, 54,5% perceberam-se com alto nível de estresse (EEP), 51,2% referiram que o estresse estava relacionado ao trabalho (EET), 15,5% apresentaram distmia e depressão (IDB) e 56,8% apresentam sinais de sofrimento mental (SRQ-20). Não houve associação entre padrão de secreção de cortisol e as variáveis psicológicas. Conclusão: Mais de um terço da amostra de profissionais de enfermagem apresentou padrões atípicos de secreção de cortisol, além de relatarem elevados níveis de estresse, estresse relacionado ao trabalho e sofrimento mental. Estes dados sugerem que estes profissionais podem estar expostos a uma sobrecarga não apenas mental, mas biológica, estando expostos ao risco para o adoecimento. / Background: Frequent exposure of nurses to work-related stressors has been widely described in the literature. However, the magnitude and intensity of the stress depends not only on stressors, but also their interaction with the cognitive appraisal of stressful situation, the resources and coping with the psychoneuroendocrine stress response. Although several studies have described the psychological reactions of stress and coping with it in nursing, little is known about the characteristics of the neuroendocrine response of stress. This is particularly important since atypical patterns of diurnal cortisol secretion are associated with increased susceptibility to the development of cardiovascular, immunological diseases and mental disorders. Thus, we arised the question about the frequency of atypical patterns of cortisol and its relationship with psychological manifestations in nursing professionals. Objective: To analyze the diurnal pattern of cortisol secretion of nursing professionals at a hospital setting. Methods: Fifty six (n = 56) nursing professionals were randomly selected allocated to the outpatient clinic, medical clinic, surgical clinic, surgical center, pediatric unit, adult and pediatric emergency department and intensive care unit of a university hospital. To evaluate the diurnal pattern of cortisol secretion, saliva samples were collected on two consecutive working days. For the psychological manifestations of stress the following instruments were applied: the Perceived Stress Scale (PSS), the Self-report Questionnaire (SRQ-20), the Beck Depression Inventory (BDI) and the work-related stress scale (WSS). Data were stored and analyzed using SPSS version 14.0 and the level of significance was 5%. Results: Regarding the pattern of cortisol secretion 42.5% of nurses had atypical pattern of cortisol secretion and 19.2% were from nursing technical professional category. Regarding psychological variables, 54.5% perceived themselves at high stress level, 51.2% reported that stress were work-related 15.5% had depression and dysthymia and 56.8% showed signs of mental distress. There was no association between pattern of cortisol secretion and the psycological variables. Conclusion: More than one third of the sample exhibited atypical pattern of cortisol secretion as well as high levels of stress, work-related stress and mental distress. These data suggest that these workers may be exposed not just to psychological overload, but also to biological burden and could be exposed to a risk for the illness.

Em função da categoria dos estressores, vias neurais específicas são envolvidas e respostas distintas podem ser induzidas. A literatura tem reportado que o estrógeno (E 2 ) através de seus receptores de tipos (ER) e (ER) influencia a atividade do eixo hipotálamo hipófise adrenal (HPA). Além disso, há evidências de que o E2 exerça efeitos protetores em situação de choque hemorrágico. O objetivo deste trabalho foi verificar a participação dos receptores ER e ER na atividade do eixo HPA durante estresse hemorrágico. Foram utilizadas ratas Wistar ovariectomizadas que receberam injeções s.c. de DMSO (veículo), PPT (agonista ER) ou DPN (agonista ER), durante 3 dias. No segundo dia, as ratas foram canuladas para coleta seriada de sangue na manhã seguinte. Os animais receberam (controle) ou não (hemorrágicos) reposição imediata com salina. Os hormônios corticosterona (CORT), ocitocina (OT) e vasopressina (AVP) foram dosados por radioimunoensaio. Ao final do experimento, os ratos foram perfundidos e os cérebros processados para imuno-histoquímica de FOS, tirosina hidroxilase (TH) e hormônio liberador de corticotrofina (CRH). Nos animais tratados com veículo, a hemorragia gradual moderada aumentou a secreção de CORT, OT e AVP, a expressão de neurônios TH ativados na região A1C1 e de FOS no mpPVN. O PPT reduziu a secreção de CORT, na situação controle atuando no LC e mpPVN; e também após hemorragia atuando no LC, NTS, A1C1 e mpPVN. O DPN reduziu a secreção de CORT apenas após estresse hemorrágico atuando no LC, A1C1 e mpPVN. O PPT bloqueou o aumento da secreção de OT e aumentou a secreção de AVP, após hemorragia. O DPN, por sua vez, reduziu a concentração plasmática de OT e aumentou a concentração plasmática de AVP, independentemente da hemorragia. Em conclusão: o estrógeno pode exercer uma ação inibitória sobre a secreção basal de CORT somente através da ação do ER sobre o LC e mpPVN; a secreção de CORT aumenta em resposta à hemorragia gradual moderada e o estrógeno pode exercer um controle inibitório nessa resposta através de ER atuando sobre LC, NTS, A1C1 e mpPVN, bem como através de ER atuando sobre LC, A1C1 e mpPVN. / Depending on the stressors category, specific neural pathways are involved and different responses can be selected. It has been reported in the literature that estrogen (E2 ) can affect hypothalamus pituitary adrenal (HPA) axis activity through its receptors type (ER) and (ER). Moreover, there is evidence that E 2 has protecting properties after hemorrhagic shock. The aim of this work was to assess the participation of ER and ER on HPA axis activity during hemorrhagic stress. It was used ovariectomized Wistar rats that received s.c. injections of: DMSO (vehicle), PPT (ER agonist) or DPN (ER agonist), during 3 days. In the second day the rats were catheterized for serial blood collect in the next morning. Animals received (control) or not (hemorrhagic) immediate reposition with same volume of isotonic saline. The hormones corticosterone (CORT), oxytocin (OT) and vasopressin (AVP) were measured by radioimmunoassay. At the end of the experiment, animals were perfused and their brains were processed for immuno-histochemistry for FOS, tyrosine hydroxylase (TH) and corticotropin releasing hormone (CRH). In vehicle treated animals, the gradual hemorrhage enhanced CORT, OT and AVP secretion, TH activated neurons expression in A1C1 and FOS expression in mpPVN. PPT decreased plasma CORT in control situation acting on LC and mpPVN, and also after hemorrhage acting on LC, NTS, A1C1 and mpPVN. DPN reduced plasma CORT only after hemorrhagic stress acting on LC, A1C1 and mpPVN. PPT blocked the increase of OT secretion and increased AVP secretion, after hemorrhage. The agonist DPN reduced OT and increased AVP levels, despite hemorrhage. In conclusion: E2 can exert an inhibitory effect on CORT basal secretion only through ER action on LC and mpPVN; CORT secretion increases after gradual moderate hemorrhage and E2 inhibit this secretion through ER action on LC, NTS, A1C1 and mpPVN, as well through ER action on LC, A1C1 and mpPVN.

Spatial relationship based scene analysis and synthesis

In this thesis, we propose a new representation, which we name Interaction Bisector Surface (IBS), that can describe the general nature of spatial relationship. We show that the IBS can be applied in 3D scene analysis, retrieval and synthesis. Despite the fact that the spatial relationship between different objects plays a significant role in describing the context, few works have focused on elaborating a representation that can describe arbitrary interactions between different objects. Previous methods simply concatenate the individual state vectors to produce a joint space, or only use simple representations such as relative vectors or contacts to describe the context. Such representations do not contain detailed information of spatial relationships. They cannot describe complex interactions such as hooking and enclosure. The IBS is a data structure with rich information about the interaction. It provides the topological, geometric and correspondence features that can be used to classify and recognize interactions. The topological features are at the most abstract level and it can be used to recognize spatial relationships such as enclosure, hooking and surrounding. The geometric features encode the fine details of interactions. The correspondence feature describes which parts of the scene elements contribute to the interaction and is especially useful for recognizing character-object interactions. We show examples of successful classification and retrieval of different types of data including indoor static scenes and dynamic scenes which contain character-object interactions. We also conduct an exhaustive comparison which shows that our method outperforms existing approaches. We also propose a novel approach to automatically synthesizing new interactions from example scenes and new objects. Given an example scene composed of two objects, the open space between the objects is abstracted by the IBS. Then, an translation, rotation and scale equivariant feature called shape coverage feature, which encodes how the point in the open space is surrounded by the environment, is computed near the IBS and around the open space of the new objects. Finally, a novel scene is synthesized by conducting a partial matching of the open space around the new objects with the IBS. Using our approach, new scenes can be automatically synthesized from example scenes and new objects without relying on label information, which is especially useful when the data of scenes and objects come from multiple sources.

Effects of postnatal light environment on the development of the mouse stress system

The postnatal period is a critical time for development where external influences can help shape the long-term structure and function of the brain. Adverse experiences or stressors during the postnatal period, such as abuse or neglect, can have huge consequences on the long term function, health and susceptibility to disease. One environmental factor, whose importance is becoming increasingly more recognised for normal development, is light. Abnormal light during the first three weeks of life has been shown to have long term effects on the circadian system of rodents. On the other hand, the effects of abnormal light during the postnatal period on the stress system have yet been relatively unexplored. Therefore the aim of this thesis was to assess if altered postnatal light environment, such as that a preterm baby might be exposed to, has any long-term effects on the stress system. Mice were raised under constant light (LL), constant darkness (DD) or a normal 12:12hr light:dark cycle (LD) for the first three weeks of life from postnatal day (P)1 up until P21. From P21, all mice were then housed in LD conditions and the stress system was assessed by looking at several different levels of the HPA axis including neuropeptide expression in the brain, body and adrenal weight, and plasma corticosterone levels under both basal and stressed conditions. Learning and memory, anxiety-like behaviour and circadian output rhythms were also evaluated. Finally, mother-pup behaviour and maternal HPA axis were assessed to see if maternal care was changed by altered postnatal light. Both LL and DD rearing caused changes in the HPA axis of offspring with LL raised mice showing alterations in neuropeptide and glucocorticoid receptor expression in the brain. Postnatal DD resulted in a blunted corticosterone response to a stressor in females but had no effect in males. In terms of behaviour, LL raised mice had increased depressive-like behaviour. In contrast, postnatal light appears to have no effect on learning and memory or anxiety behaviour. When we looked at circadian output rhythms, we found that LL rearing appears to confer resilience to the rhythm disrupting effects of LL later on in life as seen by the maintenance of locomotor activity, body temperature and plasma corticosterone rhythms in LL. Maternal care and maternal stress systems appeared unaltered under the different postnatal light environments suggesting that the changes we see in the offspring are attributed to mechanisms other than alterations in maternal care.

Virtual five-axis flank milling of jet engine impellers

This thesis presents models and algorithms necessary to simulate the five-axis flank milling of jet-engine impellers in a virtual environment. The impellers are used in the compression stage of the engine and are costly, difficult to machine, and time-consuming to manufacture. To improve the productivity of the flank milling operations, a procedure to predict and optimize the cutting process is proposed. The contributions of the thesis include a novel cutter-workpiece engagement calculation algorithm, a five-axis flank milling cutting mechanics model, two methods of optimizing feed rates for impeller machining tool paths and a new five-axis chatter stability algorithm. A semi-discrete, solid-modeling-based method of obtaining cutter-workpiece engagement (CWE) maps for five-axis flank milling with tapered ball-end mills is developed. It is compared against a benchmark z-buffer CWE calculation method, and is found to generate more accurate maps. A cutting force prediction model for five-axis flank milling is developed. This model is able to incorporate five-axis motion, serrated, variable-pitch, tapered, helical ball-end mills and irregular cutter-workpiece engagement maps. Simulated cutting forces are compared against experimental data collected with a rotating dynamometer. Predicted X and Y forces and cutting torque are found to have a reasonable agreement with the measured values. Two offline methods of optimizing the linear and angular feeds for the five-axis flank milling of impellers are developed. Both offer a systematic means of finding the highest feed possible, while respecting multiple constraints on the process outputs. In the thesis, application of these algorithms is shown to reduce the machining time for an impeller roughing tool path. Finally, a chatter stability algorithm is introduced that can be used to predict the stability of five-axis flank milling operations with general cutter geometry and irregular cutter-workpiece engagement maps. Currently, the new algorithm gives chatter stability predictions suitable for high speed five-axis flank milling. However, for low-speed impeller machining, these predictions are not accurate, due to the process damping that occurs in the physical system.
