It is looked for, through the History, in a sociological perspective, to analyze the subject of the Popular Catholicism, trying to understand it starting from the general context Brazil, passing through Goiás, Goiânia, to place it in Trindade. The society, subject in study, presents secularazing, characteristics, however, this phenomenon is contested, because we can see that the Religious Phenomenon coexists, with the technician-scientific rational thought of the modernity. With regard to the reason of the permanence of the Religious Phenomenon, it is noticed that the crisis of the modernity generates insecurity in the people. The analysis of the Party of Trindade evidences that is in the religion that the people look for and find answers to their problems, besides the justification of been in a certain social context. The research, reveals, ratifying BOURDIEU'S thought (1998), on whom supported our theoretical ground, that religion accomplishes structural social functions, legitimary and of society. This fact is proven at the Party of Trindade. / Versa o presente estudo sobre a Festa do Divino Pai Eterno em Trindade como expressão do Catolicismo Popular em Goiás. Busca-se, através da História, numa perspectiva sociológica, analisar a questão do Catolicismo Popular, procurando entendê-lo a partir do contexto geral Brasil, perpassando por Goiás, Goiânia, para situá-lo em Trindade. A sociedade, objeto de estudo, apresenta características secularizadoras, no entanto, contesta-se este fenômeno, porque se vê que o Fenômeno Religioso coexiste, com o pensamento racional técnico-científico da modernidade... Com relação ao porquê da permanência do Fenômeno Religioso, percebe-se que a crise da modernidade gera insegurança nas pessoas. A análise da Festa de Trindade evidencia que é na religião que as pessoas buscam e encontram respostas aos seus problemas, além da justificativa de estarem situadas num determinado contexto social. A pesquisa, assim, revela, ratificando o pensamento de BOURDIEU (1998), em quem apoiamos nossa fundamentação teórica, que a religião cumpre funções sociais estruturadoras, legitimadoras e justificadoras da sociedade. Este fato é comprovado na Festa de Trindade.

In this study it was not intended to judge the function or know how it is performed by men. our intention was to understand how this man can re-signify his life after participating in the process of parturition and accompany this mother until discharge to return home. This is a ¿Bonding¿. Socio-cultural influences can be decisive when it comes to encouraging this parent to participate and to become more actively involved in their child birth. This decision is not always easy and in some situations can be recriminated and it can also stigmatize the man who wants to simply change a paradigm and be closer to this unique moment in the life of a couple. Our objectives were to seek answers to understand the re-signification of the father-baby bond after the father's participation in humanized childbirth or postpartum follow-up; To describe the perception of fatherhood experienced by this father after his participation in childbirth and / or postpartum in his relationship with mother and baby from the hospital environment; Unveil the prejudices and stigmas perceived by the man after his participation in childbirth or postpartum; To contextualize the influence of this experience on the family bond and on the construction of his paternal identity (father-man). In the scenario was the regional VI in Messejana and the Gonzaga Mota District Hospital and some neighborhood residences and localities. We spoke with nine participants who presented us with their narratives on the key subject: being a father today. We use Geertz's Interpretive Anthropology to shape the categories that emerged in the thematic analysis of Bardin with categories relevant to the male gender, parenthood, family, and parent-child relationship. We took advantage of the contextualized semantics of Bibeau & Corin to give more clarity to the narrated metaphors when interpreting our data. The study was approved by the ethics committee in research and protecting the informants and receiving authorization from the subjects to work their information after signing the term. We had a period of fifteen months to work with these subjects who were invited on an occasional basis upon their arrival in hospital to accompany the mother in childbirth. In order to arrive at the results we use interviews to approach and in the second moment a meeting in the homes where we worked with narratives until we reached the maximum of approximation of this reality lived by these men who perceived themselves as a father. The real gains that the historical changes presented were very evident when we began to interpret the lines. The participation of man at the moment of birth can bring changes in this local social universe, which is consistent with the principle defended by Kleinmann, where even suffering prejudice and stigma, if the suffering was experienced can modify the perception of the subject and help him to transform the local moral world in a positive way for the whole community. The influence of the public policies and the local project of the Gonzaguinha hospital were factors that shaped this new model of being a father directly interfering in the way of facing the paternity and its participation in the family relation. The interpretation of the subjectivity of the masculine universe and how it is perceived as a father who cares, was very explicit and shows the function of this man-father. / Ser pai, aparentemente, pode ser muito fácil. Neste estudo, não se pretendeu julgar a função ou saber como ela é executada pelos homens. A intenção foi compreender como o homem pode ressignificar sua vida após participar do processo de parturição e acompanhar a mulher-mãe até a alta para retornar à sua casa. Quis-se compreender o bonding. As influências sócio-culturais podem ser decisivas quando se trata de incentivar este pai a participar e a se envolver de forma mais ativa no nascimento de seu filho. Nem sempre essa decisão é fácil e, em algumas situações, pode recriminar e estigmatizar o homem que deseja, simplesmente, mudar um paradigma e estar mais próximo, neste momento único, da vida de um casal. Os objetivos deste estudo foram buscar respostas para compreender a ressignificação do vínculo pai-bebê após a participação do pai no parto humanizado ou acompanhamento pós-parto; descrever a percepção da paternidade vivida por este pai após sua participação no parto e/ou pós-parto no seu convívio com mãe e bebê desde o ambiente hospitalar; desvelar os preconceitos e os estigmas percebidos pelo homem após sua participação no parto ou pós-parto; contextualizar a influência desta experiência no vínculo familiar e na construção de sua identidade paterna (homem-pai). Participaram do cenário a Regional VI, em Messejana, o Hospital Distrital Gonzaga Mota e algumas residências e localidades do bairro. Conversou-se com nove participantes que apresentaram suas narrativas sobre o assunto-chave: ser pai hoje. Fez-se uso da Antropologia Interpretativa de Geertz para dar forma às categorias que emergiram na análise temática de Bardin com categorias pertinentes ao gênero masculino, paternidade, família e relação pai e filho. Dispôs-se de um período de quinze meses para trabalhar com esses sujeitos que foram convidados de forma ocasional em momento de sua chegada ao espaço hospital para acompanhar a mãe no parto. Para chegar aos resultados, foram usadas entrevistas para a aproximação e, no segundo momento, um encontro nos domicílios onde se trabalhou com narrativas até se chegar ao máximo de aproximação desta realidade vivida por estes homens que se perceberam como pai. Os ganhos reais que as mudanças históricas apresentaram ficaram bem evidentes quando iniciaram as interpretações das falas. A participação do homem no momento do nascimento pode, sim, trazer mudanças neste universo social local, o que condiz com o princípio defendido por Kleinmann, em que, mesmo sofrendo preconceito e estigma, se o sofrimento foi vivenciado, pode modificar a percepção do sujeito e ajudá-lo a transformar o mundo moral local de forma positiva para toda a comunidade. A influência das políticas públicas e do projeto local do hospital Gonzaguinha foram fatores que moldaram este novo modelo de ser pai, interferindo diretamente em sua forma de encarar a paternidade e a sua participação na relação familiar. A interpretação da subjetividade do universo masculino e como ele se percebe enquanto pai que cuida ficou bem explícita e mostrou a função deste homem-pai.

Cultura e estrutura em psicanálise

Cette thèse identifie dans la littérature psychanalytique actuelle la difusion d’un argument commun qui dit que le déclin de la fonction paternelle dans la contemporaneité – considéré comme une mutation culturelle – est corrélé à de nouveaux symptômes et formes cliniques. Selon cet argument, la pratique de la psychanalyse dans la contemporaneité trouve des difficultés particulières et sans précédent. Je recherche cette corrélation de façon critique, en soulignant ses aspects méthodologiques et épistémologiques. Une compréhension de la relation de la psychanalyse de Lacan avec la science est essentiel à cette but, parce qu’elle fournit les outils théoriques qui soutiennent cette critique et permet aussi la construction d'une topologie structurale : une méthode d'approche de structures localisées que se oppose à la façon comme les questions contemporains de la psychanalyse ont été développées. Je considère que l'hypothèse universaliste et totémique de la culture, ainsi que la rationalité diagnostic tripartite (névrose, psychose, perversion) qui elle soutient, sont le fondement insuffisant de la argumentation difusé dans la littérature. Ainsi, les solutions localisées sont proposées comme alternatives que, dans le contexte de la topologie structurale, sont créatrices ou émettrices d'éxistences, ce qui justifie qu’elles soient prises comme une troisième voie de la structure, ni empirique ni formelle, mais universalisant dans un sens que se oppose au universalisable généraliste et inductive. Enfin, on propose deux exemples d'utilisations de la topologie structurale dans des régions localisées de la théorie psychanalytique : la coupure interprétatif, soutenu par la bande de Moebius, et la relation entre la théorie et la technique psychanalytique, soutenu par la structure torique de l'Autre comme univers du discours incomplet. / Esta tese identifica na literatura psicanalítica atual a difusão de um argumento comum que afirma que o declínio da função paterna na contemporaneidade – considerado uma mutação cultural – é correlato a novos sintomas e formas clínicas. Segundo esse argumento, a prática da psicanálise na contemporaneidade encontraria dificuldades específicas e inéditas. Investigo essa correlação criticamente, enfatizando seus aspectos metodológicos e epistemológicos. A compreensão da relação da psicanálise de Lacan com a ciência é fundamental para esse propósito, pois tanto fornece as ferramentas teóricas que embasam essa crítica quanto permite a construção de uma topologia estrutural, um método de abordagem de estruturas localizadas que se opõe ao modo como as questões contemporâneas da psicanálise vem sendo desenvolvidas. Considero que a hipótese universalista e totêmica da cultura, assim como a racionalidade diagnóstica tripartida (neurose-psicose-perversão) por ela sustentada, são o fundamento insuficiente da argumentação difundida na literatura. Sendo assim, as soluções localizadas são propostas como alternativas que, dentro do contexto da topologia estrutural, são criadoras ou emissoras de existências, o que justifica que sejam tomadas como uma terceira via da estrutura, nem empírica nem formalista, mas universalizante num sentido que se opõe ao universalizável de cunho generalista e indutivo. Finalmente, são propostos exemplos de utilizações da topologia estrutural em regiões localizadas da teoria psicanalítica: o corte interpretativo, cujo suporte é a banda de Moebius, e a articulação entre a teoria e a técnica psicanalíticas, fundamentada pela estrutura tórica do Outro como universo de discurso incompleto. / This thesis identifies in the current psychoanalytic literature the diffusion of a common argument that states that the decline of the paternal function in the contemporaneity – considered as a cultural mutation – is correlative to new symptoms and clinical forms. According to this argument, the practice of psychoanalysis nowadays encounters specific and unprecedented difficulties. In this work, this correlation is critically investigated and its methodological and epistemological aspects are emphasized. Understanding the relationship of Lacan's psychoanalysis with science is essential for this purpose, since it provides the theoretical tools that support this criticism and allows the construction of a structural topology: a method that approaches localized structures in a way that is opposite to the way of how contemporary issues of psychoanalysis are being developed. I consider that the universalist and totemic hypothesis of culture, as well as the tripartite diagnostic rationality (neurosis, psychosis, perversion) that it sustains, are the insufficient foundation of the argument widespread in literature. Thus, within the structural topology context, the localized solutions are proposed as alternatives for the creation or emission of existences, which justifies them as a third way of the structure, neither empirical nor formalist, but universalizing in a sense that is opposite to the generalist and inductive universalizable. Finally, examples of uses of the structural topology in localized regions of the psychoanalytic theory are proposed: the interpretive cut, whose support is the Moebius band, and the relation between theory and psychoanalytic technique, sustained by the toric structure of the Other as incomplete universe of speech.

Studies on the Role of Vitronectin and Plasminogen-Activator Inhibitor-1 Complexes Beyond Inhibiting Proteases: Binding to the Extracellular Matrix, Cell Interactions and Pathogenesis

Plasminogen activator inhibitor-1 (PAI-1), a member of the serine protease inhibitor (serpin) superfamily of proteins, circulates in blood in a complex with vitronectin (VN). These two proteins are also found localized together in the extracellular matrix in many different pathophysiological conditions. Both of these proteins are involved with a number of physiologically important processes. Though PAI-1 is a well-known inhibitor of serine proteases, more emphasis is now geared towards its protease independent functions. VN, on the other hand, is a binding protein that exists in the circulation in a preferred monomeric conformation. However, in the extracellular matrix, VN exists as multimer with altered conformation. Though the exact reason for such conformational alterations and compartmentalization is unknown, there are a number of biomolecules, including PAI-1 that are proposed to cause such alterations. In last few years, sufficient experimental evidence has been gathered to confirm this protease- independent effect of PAI-1 by which it induces multimerization of VN in a concentration-dependent fashion. It has been observed also that PAI-1 remains associated with this multimeric complex for several hours. A major focus of this dissertation work was to extend our understanding of the mechanism of the interaction between these proteins and to explore the physiological relevance of the multimeric complexes formed by their interaction on cellular adhesion and migration. In our study, emphasis has been given to the presence of an appropriate microenvironment so that the role of the multimeric complexes could be investigated in a relevant biological setting. Our findings indicate the importance of the surrounding microenvironment in establishing the specific role of the VN/PAI-1 complex in cell-matrix interactions. In a previous study from our lab, it was found that vitronectin knock-out mice were more resistant to Candida infection compared to wild type C57Bl/6 mice. One of the goals of this dissertation work was to provide a mechanistic explanation for their increased survival of the vitronectin knock-out mice upon Candida infection. Another important aspect of this work was to establish biophysical methods for understanding the structural changes that happen in PAI-1 naturally or due to ligand binding.

The Jaded Garden:a cross-cultural comparison of nostalgic female characters by Pai Hsien-yung and Tennessee Williams

This study consist of a comparative analysis of the nostalgic female characters in Pai Hsien-yung's two short stories: "Wandering in a Garden, Waking from a Dream," and "A Celestial in Mundane Exile," and Tennessee Williams's two plays: The Glass Menagerie and A Streetcar Named Desire. Beginning with a brief discussion of the socio-historical background of Pai's Republican China and Williams's American South, a general analysis of previous scholarship on Pai and Williams's works follows. The analysis of the selected works focuses on the stylistic and symbolic features in Pai and Williams's characterizations, such as Pai's use of stream-of-consciousness, reference to the k'un opera Peony Pavilion, elaboration over descriptive details of the setting, symbolic use of clothing and accessories, and Williams's symbolic use of music genres: "Blues Piano" and the "Varsouviana Polka," and his use of rhythm and other poetic elements in his characters' speech, in the style of "personal lyricism." My study is based on a close-reading analysis of the selected works by Pai and Williams. Their humanistic approach to their respective declining aristocratic cultures and their sympathy for the nostalgic female characters' tragedies will be more apparent when the study focuses mostly on the texts themselves. Their similar belief in the universal values, such as compassion, sacrifice, and courage, has made their works comparable. In the discussion of themes, the idea of the humanistic role of literature articulated by William Faulkner in his Nobel Prize Speech is also used to connect Pai and Williams's sympathetic approach to their characters.

Empirical Correlates of the Personality Assessment Inventory (PAI) in an Outpatient Sample: A Replication and Extension

The Personality Assessment Inventory (PAI) has gained widespread favor since its publication. However, validation studies of its interpretive descriptors remained limited to date. As such, the primary goal of the current study aimed at validating the interpretive descriptors through the lens of empirical correlates using the PDSQ as the external criterion. It also served as a replication and extension to the 2018 study conducted by Rogers and colleagues. The final archival sample included 204 clients from the UNT Psychology Clinic who were administered PAI between May 2016 and December 2020. Overall, reliability and construct validity were strongly supported for PAI clinical scales. Further, the current study replicated large majority of the correlates identified by Rogers and colleagues, which boosted the confidence in reproducible interpretations based on empirical correlates. Importantly, investigation of item-level and gendered correlates provided crucial interpretive implications that were otherwise obscured. For example, item-level correlates refined interpretation by clarifying the nature of scale-level correlates, particularly those of moderate strength. On the other hand, notable gender differences were identified for certain scales, which led to drastic differences in patterns of gendered vs. non-gendered correlates. Finally, several important methodological considerations are proposed in hope to facilitate the empirical research concerning measurement validity, as well as combat the current replication crisis. The need to adopt more stringent standards for effect size, as well as the instability of correlates of moderate strength were discussed. Implications of clinical practice and future directions for research are also discussed.

Transcriptional Repression of the Plasminogen Activator Inhibitor Type 2 Gene

Plasminogen activator inhibitor type 2 (PAI-2) is a serine protease inhibitor traditionally regarded as a regulator of fibrinolysis and extracellular matrix degradation. More recently, PAI-2 has been implicated in diverse processes such as keratinocyte differentiation, cell death and viral pathogenesis. Although PAI-2s limited pattern of expression in vivo generates significant interest in this molecule, little is known about the underlying mechanisms controlling its cell specific regulation. In this thesis, the function that the previously identified PAI-2 gene silencer (Antalis et al., 1996) plays in the regulation of PAI-2 gene expression was investigated. The PAI-2 upstream silencer element 1 (PAUSE-1) is located approximately 1800bp upstream of the PAI-2 transcription initiation site. By employing electrophoretic mobility shift assays and transient transfection assays with mutant PAUSE-1 sequences, the sequence that defines PAUSE-1 was identified as TCT N3 AGA N3 T4. This element was shown to bind a number of protein complexes of similar electrophoretic mobility from various cultured cell lines. Transient transfection assays with the cervical adenocarcinoma, HeLa S3 and the macrophage-like, U937 cell lines, showed that PAUSE-1 repressed transcription by approximately 2.5 fold when cloned into the SV40 promoter or the minimal PAI-2 promoter. Ultraviolet (UV)-crosslinking analyses determined that the PAUSE-1 binding protein (BP) was approximately 67kDa. Examination of several similar DNA promoter sequences, such as the human IFNb and insulin promoters, suggested that PAUSE-1 might be an example of a universal silencer with the consensus sequence TCT Nx AGA, where x=4. The PAUSE-1 sequence shows significant homology to the binding sequence of the transcriptional regulators Ski, Smad3 and Smad4. EMSAs incorporating anti-Ski, -Smad3 and -Smad4 antibodies suggested that each are members of the PAUSE-1 BP complex in HeLa S3 cells. The PAUSE-1 BP complex has been purified by employing DNA affinity chromatography using streptavidin labelled magnetic beads. Approximately nine PAUSE-1 associated proteins from HeLa S3 extracts were visualised. Amino-terminal protein sequencing identified the first eight amino acids of the PAUSE-1 BP as EIQQRAAQ. The PAUSE-1 BP fails to show significant sequence similarity to any known protein and therefore potentially represents a novel DNA binding protein.
