Extending our understanding of Islamic banking through questioning assumptions and drawing unprecedented comparisons

This thesis challenges two key assumptions made in the current Islamic banking literature. Firstly, this thesis challenges and empirically invalidates the assumption that all Islamic banks are indistinguishable from their conventional counterparts and are thus equally unIslamic. To do so, this thesis uses the profit and loss sharing (PLS) criteria, which is central to the philosophy of Islamic banking and is the key principle differentiating Islamic from conventional banking, in theory and practice. By investigating variation in PLS levels between Islamic banks and comparing with conventional banks with and without Islamic windows, this thesis illustrates that the Islamic banking industry does not comprise a homogeneous group of banks that are all indistinguishable from their conventional counterparts. Rather, a typology of Islamic banks exists, comprising of three distinct groups of banks, each one following a different business model. While one group can genuinely be considered indistinguishable from conventional banks, another group shows clear evidence of pursuing PLS-oriented strategies in formulating its asset portfolio, differentiating itself from the purely debt-based intermediation model adopted by conventional banks. As such, empirical evidence shows that some Islamic banks are, in practice, operating closer to the PLS principle and can thus be considered more Islamic than others. Further investigation illustrates that the institutional environment matters for the provision of ideal PLS Islamic financing instruments. Secondly, this thesis overcomes two methodological issues to compare the corporate social performance (CSP) of Islamic and conventional banks. In doing so, this thesis challenges the second identified assumption from the literature, that religion-specific category of corporate social responsibility (CSR) is particular to Islamic banking, and invalidates it on conceptual, theoretical and empirical basis. A novel CSP Index based on the evidence-based disclosure criteria, comprising of 6 dimensions and 25 social performance indicators is constructed and complemented with three Social Performance Quantitative Indicators (SPQIs) to compare the CSP of Islamic and conventional banks. From this comparison, this thesis concludes that, contrary to the industry s claims and expectations held of it, Islamic banking does not offer an ethical alternative to conventional banking. Differences in the level and composition of CSP between the two industries are more subtle and require a nuanced approach to be studied.

Les plantes médicinales constituent une source inexhaustible de composés (des produits naturels - PN) utilisé en médecine pour la prévention et le traitement de diverses maladies. L'introduction de nouvelles technologies et méthodes dans le domaine de la chimie des produits naturels a permis le développement de méthodes ‘high throughput’ pour la détermination de la composition chimique des extraits de plantes, l'évaluation de leurs propriétés et l'exploration de leur potentiel en tant que candidats médicaments. Dernièrement, la métabolomique, une approche intégrée incorporant les avantages des technologies d'analyse moderne et la puissance de la bioinformatique s’est révélé un outil efficace dans la biologie des systèmes. En particulier, l'application de la métabolomique pour la découverte de nouveaux composés bioactifs constitue un domaine émergent dans la chimie des produits naturels. Dans ce contexte, le genre Acronychia de la famille des Rutaceae a été choisi sur la base de son usage en médecine traditionnelle pour ses propriétés antimicrobienne, antipyrétique, antispasmodique et anti-inflammatoire. Nombre de méthodes chromatographiques modernes, spectrométriques et spectroscopiques sont utilisées pour l'exploration de leur contenu en métabolites suivant trois axes principaux constituant les trois chapitres de cette thèse. En bref, le premier chapitre décrit l’étude phytochimique d’Acronychia pedunculata, l’identification des métabolites secondaires contenus dans cette espèce et l'évaluation de leurs propriétés biologiques. Le deuxième chapitre vise au développement de méthodes analytiques pour l'identification des dimères d’acétophénones (marqueurs chimiotaxonomiques du genre) et aux stratégies utilisées pour la déréplication de ces différents extraits et la caractérisation chimique des composés par UHPLC-HRMSn. Le troisième chapitre se concentre sur l'application de méthodologies métabolomique (RMN et LC-MS) pour l'analyse comparative (entre les différentes espèces, origines, organes), pour des études chimiotaxonomiques (entre les espèces) et pour la corrélation des composés contenus avec une activité pharmacologique. / Medicinal plants constitute an unfailing source of compounds (natural products – NPs) utilised in medicine for the prevention and treatment of various deceases. The introduction of new technologies and methods in the field of natural products chemistry enabled the development of high throughput methodologies for the chemical composition determination of plant extracts, evaluation of their properties and the exploration of their potentials as drug candidates. Lately, metabolomics, an integrated approach incorporating the advantages of modern analytical technologies and the power of bioinformatics has been proven an efficient tool in systems biology. In particular, the application of metabolomics for the discovery of new bioactive compounds constitutes an emerging field in natural products chemistry. In this context, Acronychia genus of Rutaceae family was selected based on its well-known traditional use as antimicrobial, antipyretic, antispasmodic and anti-inflammatory therapeutic agent. Modern chromatographic, spectrometric and spectroscopic methods were utilised for the exploration of their metabolite content following three basic axes constituting the three chapters of this thesis. Briefly, the first chapter describes the phytochemical investigation of Acronychia pedunculata, the identification of secondary metabolites contained in this species and evaluation of their biological properties. The second chapter refers to the development of analytical methods for the identification of acetophenones (chemotaxonomic markers of the genus) and to the dereplication strategies for the chemical characterisation of extracts by UHPLC-HRMSn. The third chapter focuses on the application of metabolomic methodologies (LC-MS & NMR) for comparative analysis (between different species, origins, organs), chemotaxonomic studies (between species) and compound-activity correlations.

ATP-Binding Cassette Efflux Transporters and Passive Membrane Permeability in Drug Absorption and Disposition

<p>Transport into and across the cells of the human body is a prerequisite for the pharmacological action of drugs. Passive membrane permeability and active transport mechanisms are major determinants of the intestinal absorption of drugs, as well as of the distribution to target tissues and the subsequent metabolism and excretion from the body. In this thesis, the role of ATP-binding cassette (ABC) transporters and passive permeability on drug absorption and disposition was investigated. Particular emphasis was placed on defining the molecular properties important for these transport mechanisms. </p><p>The influence of different transport pathways on predictions of intestinal drug absorption was investigated using experimental models of different complexity. Experimental models that include the paracellular pathway gave improved predictions of intestinal drug absorption, especially for incompletely absorbed drugs. Further, the inhibition of the ABC transporters breast cancer resistance protein (BCRP/ABCG2) and multidrug-resistance associated protein 2 (MRP2/ABCC2) was experimentally investigated using structurally diverse datasets that were representative of orally administered drugs. A large number of previously unknown inhibitors were identified among registered drugs, but their clinical relevance for drug-drug interactions and drug-induced toxicity remains to be determined. The majority of the inhibitors affected all three major ABC transporters BCRP, MRP2 and P-glycoprotein (P gp/ABCB1), and these multi-specific inhibitors were found to be enriched in highly lipophilic weak bases. </p><p>To summarize, the present work has led to an increased knowledge of the molecular features of importance for ABC transporter inhibition and passive membrane permeability. Previously unknown ABC transporter inhibitors were identified and predictive computational models were developed for the different drug transport mechanisms. These could be valuable tools to assist in the prioritization of experimental efforts in early drug discovery.</p>

Identifying Factors Influencing The Acceptance Of Processes: An Empirical Investigation Using The Structural Equation Modeling Approach

In this research, it was mainly aimed to develop an acceptance model for processes, namely the process acceptance model (PAM). For this purpose, a questionnaire, comprising 3-part and 81-question, was developed to collect quantitative and qualitative data from people having relationships with certain process-focused models and/or standards (CMMI, ISO 15504, ISO 9001, ISO 27001, AQAP-160, AQAP-2110, and/or AS 9100). To revise and refine the questionnaire, expert reviews were ensured, and a pilot study was conducted with 60 usable responses. After reviews, refinements and piloting, the questionnaire was deployed to collect data and in-total 368 usable responses were collected from the people. Here, collected data were screened concerning incorrectly entered data, missing data, outliers and normality, and reliability and validity of the questionnaire were ensured. Partial least squares structural equation modeling (PLS SEM) was applied to develop the PAM. In this context, exploratory and confirmatory factor analyses were applied, and the initial model was estimated and evaluated. The initial model was modified as required by PLS SEM, and confirmatory factor analysis was repeated, and the modified final model was estimated and evaluated. Consequently, the PAM, with 18 factors and their statistically significant relationships, was developed. Furthermore, descriptive statistics and t-tests were applied to discover some interesting, meaningful, and important points to be taken into account regarding the acceptance of processes. Moreover, collected quantitative data were analyzed, and three additional factors were discovered regarding the acceptance of processes. Besides, a checklist to test and/or promote the acceptance of processes was established.

Dans les domaines de l'analyse, du contrôle et de la mesure physique, le laser constitue un outil métrologique particulièrement puissant et polyvalent, capable d'apporter des réponses concrètes à des problématiques variées, y compris d'ordre sociétal. Parmi ces dernières, la contamination des sites et des sols par les métaux lourds est un enjeu de santé publique important qui requiert de disposer de moyens de mesure adaptés aux réglementations existantes et suffisamment souples d'utilisation. Technique rapide et ne nécessitant pas de préparation de l'échantillon, la spectroscopie sur plasma induit par laser (LIBS) présente des avantages très intéressants pour réaliser des mesures sur site de la teneur en métaux lourds à l'échelle de la dizaine de ppm; la conception d'un appareil portable à moyen terme est envisageable.<br />Dans cette thèse nous montrons d'abord que le régime femtoseconde ne présente pas d'avantages par rapport au régime nanoseconde standard pour notre problématique. Ensuite nous mettons en œuvre un traitement avancé des spectres LIBS par des méthodes chimiométriques dont les performances améliorent sensiblement les résultats des analyses qualitatives et quantitatives d'échantillons de sols.

Multivariate design of molecular docking experiments : An investigation of protein-ligand interactions

To be able to make informed descicions regarding the research of new drug molecules (ligands), it is crucial to have access to information regarding the chemical interaction between the drug and its biological target (protein). Computer-based methods have a given role in drug research today and, by using methods such as molecular docking, it is possible to investigate the way in which ligands and proteins interact. Despite the acceleration in computer power experienced in the last decades many problems persist in modelling these complicated interactions. The main objective of this thesis was to investigate and improve molecular modelling methods aimed to estimate protein-ligand binding. In order to do so, we have utilised chemometric tools, e.g. design of experiments (DoE) and principal component analysis (PCA), in the field of molecular modelling. More specifically, molecular docking was investigated as a tool for reproduction of ligand poses in protein 3D structures and for virtual screening. Adjustable parameters in two docking software were varied using DoE and parameter settings were identified which lead to improved results. In an additional study, we explored the nature of ligand-binding cavities in proteins since they are important factors in protein-ligand interactions, especially in the prediction of the function of newly found proteins. We developed a strategy, comprising a new set of descriptors and PCA, to map proteins based on their cavity physicochemical properties. Finally, we applied our developed strategies to design a set of glycopeptides which were used to study autoimmune arthritis. A combination of docking and statistical molecular design, synthesis and biological evaluation led to new binders for two different class II MHC proteins and recognition by a panel of T-cell hybridomas. New and interesting SAR conclusions could be drawn and the results will serve as a basis for selection of peptides to include in in vivo studies.

In silico tools in risk assessment : of industrial chemicals in general and non-dioxin-like PCBs in particular

Industrial chemicals in European Union produced or imported in volumes above 1 tonne annually, necessitate a registration within REACH. A common problem, concerning these chemicals, is deficient information and lack of data for assessing the hazards posed to human health and the environment. Animal studies for the type of toxicological information needed are both expensive and time consuming, and to that an ethical aspect is added. Alternative methods to animal testing are thereby requested. REACH have called for an increased use of in silico tools for non-testing data as structure-activity relationships (SARs), quantitative structure-activity relationships (QSARs), and read-across. The main objective of the studies underlying this thesis is related to explore and refine the use of in silico tools in a risk assessment context of industrial chemicals. In particular, try to relate properties of the molecular structure to the toxic effect of the chemical substance, by using principles and methods of computational chemistry. The initial study was a survey of all industrial chemicals; the Industrial chemical map was created. A part of this map was identified including chemicals of potential concern. Secondly, the environmental pollutants, polychlorinated biphenyls (PCBs) were examined and in particular the non-dioxin-like PCBs (NDL-PCBs). A set of 20 NDL-PCBs was selected to represent the 178 PCB congeners with three to seven chlorine substituents. The selection procedure was a combined process including statistical molecular design for a representative selection and expert judgements to be able to include congeners of specific interest. The 20 selected congeners were tested in vitro in as much as 17 different assays. The data from the screening process was turned into interpretable toxicity profiles with multivariate methods, used for investigation of potential classes of NDL-PCBs. It was shown that NDL-PCBs cannot be treated as one group of substances with similar mechanisms of action. Two groups of congeners were identified. A group including in general lower chlorinated congeners with a higher degree of ortho substitution showed a higher potency in more assays (including all neurotoxic assays). A second group included abundant congeners with a similar toxic profile that might contribute to a common toxic burden. To investigate the structure-activity pattern of PCBs effect on DAT in rat striatal synaptosomes, ten additional congeners were selected and tested in vitro. NDL-PCBs were shown to be potent inhibitors of DAT binding. The congeners with highest DAT inhibiting potency were tetra- and penta-chlorinated with 2-3 chlorine atoms in ortho-position. The model was not able to distinguish the congeners with activities in the lower μM range, which could be explained by a relatively unspecific response for the lower ortho chlorinated PCBs. / Den europeiska kemikalielagstiftningen REACH har fastställt att kemikalier som produceras eller importeras i en mängd över 1 ton per år, måste registreras och riskbedömmas. En uppskattad siffra är att detta gäller för 30 000 kemikalier. Problemet är dock att data och information ofta är otillräcklig för en riskbedömning. Till stor del har djurförsök använts för effektdata, men djurförsök är både kostsamt och tidskrävande, dessutom kommer den etiska aspekten in. REACH har därför efterfrågat en undersökning av möjligheten att använda in silico verktyg för att bidra med efterfrågad data och information. In silico har en ungefärlig betydelse av i datorn, och innebär beräkningsmodeller och metoder som används för att få information om kemikaliers egenskaper och toxicitet. Avhandlingens syfte är att utforska möjligheten och förfina användningen av in silico verktyg för att skapa information för riskbedömning av industrikemikalier. Avhandlingen beskriver kvantitativa modeller framtagna med kemometriska metoder för att prediktera, dvs förutsäga specifika kemikaliers toxiska effekt. I den första studien (I) undersöktes 56 072 organiska industrikemikalier. Med multivariata metoder skapades en karta över industrikemikalierna som beskrev dess kemiska och fysikaliska egenskaper. Kartan användes för jämförelser med kända och potentiella miljöfarliga kemikalier. De mest kända miljöföroreningarna visade sig ha liknande principal egenskaper och grupperade i kartan. Genom att specialstudera den delen av kartan skulle man kunna identifiera fler potentiellt farliga kemiska substanser. I studie två till fyra (II-IV) specialstuderades miljögiftet PCB. Tjugo PCBs valdes ut så att de strukturellt och fysiokemiskt representerade de 178 PCB kongenerna med tre till sju klorsubstituenter. Den toxikologiska effekten hos dessa 20 PCBs undersöktes i 17 olika in vitro assays. De toxikologiska profilerna för de 20 testade kongenerna fastställdes, dvs vilka som har liknande skadliga effekter och vilka som skiljer sig åt. De toxicologiska profilerna användes för klassificering av PCBs. Kvantitativa modeller utvecklades för prediktioner, dvs att förutbestämma effekter hos ännu icke testade PCBs, och för att få ytterligare kunskap om strukturella egenskaper som ger icke önskvärda effekter i människa och natur. Information som kan användas vid en framtida riskbedömning av icke-dioxinlika PCBs. Den sista studien (IV) är en struktur-aktivitets studie som undersöker de icke-dioxinlika PCBernas hämmande effekt av signalsubstansen dopamin i hjärnan.

Ανάπτυξη νέας τεχνικής επιφανειακής ενίσχυσης της σκέδασης Raman (SERS) για ποσοτικές μετρήσεις ενεργών ουσιών σε πολύ μικρές συγκεντρώσεις

Η φασματοσκοπία Raman θεωρείται αξιόπιστη μέθοδος χαρακτηρισμού της μοριακής δομής της ύλης. Τελευταία δε καταβάλλεται μεγάλη προσπάθεια να αναδειχθεί και ως μη επεμβατική τεχνική ανίχνευσης ουσιών φαρμακευτικού και βιολογικού ενδιαφέροντος. Επειδή όμως το φαινόμενο Raman είναι ασθενές και τα όρια ανίχνευσης των ουσιών αυτών ιδιαίτερα χαμηλά, η συμβατική φασματοσκοπία Raman ανταποκρίνεται με μεγάλη δυσκολία στις απαιτήσεις μιας μη επεμβατικής τεχνικής. Τα τελευταία χρόνια με την ανακάλυψη της επιφανειακής ενίσχυσης της σκέδασης Raman (Surface Enhanced Raman Scattering) και την ανάπτυξη της ομώνυμης τεχνικής SERS αυξήθηκε η πιθανότητα του φαινομένου κατά τάξεις μεγέθους. Στο πλαίσιο αυτό, έχουν αναφερθεί και μελέτες SERS με ανίχνευση σκέδασης Raman ακόμη και από ένα μόνο μόριο (single molecule detection). Προϋπόθεση όμως ανάπτυξης φαινομένου SERS είναι η γειτνίαση της εξεταζόμενης ουσίας με νανοσωματίδια αργύρου (Ag) ή χρυσού (Au). Τα νανοσωματίδια αυτά χρησιμοποιούνται κυρίως είτε υπό μορφή κολλοειδών διαλυμάτων είτε υπό την μορφή νανοδομημένων επιφανειών. Μια επιπλέον παράμετρος που πρέπει να συνυπολογισθεί ώστε η μέθοδος αυτή να χαρακτηριστεί αναλυτική είναι η δυνατότητα πραγματοποίησης με SERS ποσοτικών μετρήσεων. Στην παρούσα εργασία ως “υπόστρωμα” εμφάνισης του φαινομένου SERS χρησιμοποιήθηκαν κολλοειδή διαλύματα Au και Ag (~2 mL), στα οποία προστίθεντο 25-100 μL διαλύματος της προς ανίχνευσης ουσίας. Η χρησιμοποίηση της γεωμετρίας σκέδασης 90ο, της πλέον ενδεδειγμένης για λήψη φασμάτων Raman από υγρά και κολλοειδή διαλύματα νανομετρικής κλίμακας (10-100 nm), έδωσε τη δυνατότητα ανίχνευσης συγκεντρώσεων ουσιών-στόχων ακόμη και της τάξης των fg/mL. Επιπλέον, η χρησιμοποίηση μιας κυψελίδας με συνεχή ταλάντωση σε κάθετη ως προς το επίπεδο σκέδασης διεύθυνση (oscillating or shaking cell), επέτρεψε την πραγματοποίηση ποσοτικών μετρήσεων SERS σε συγκεντρώσεις της τάξης των (sub)-ng & (sub)-pg/mL. Η διάταξη αυτή φωταψίας δείγματος και συλλογής σκεδαζόμενης ακτινοβολίας εφαρμόστηκε σε αντικαρκινικά φάρμακα (Novantrone® & Doxorubicin), αντιβιοτικό (Ciproxin®) και βάσεις του DNA. Στην περίπτωση του αντιβιοτικού με ενεργή ουσία το Ciprofloxacin ελήφθησαν ποσοτικά αποτελέσματα σε διαλύματα τεχνητών δακρύων της τάξης των ng/mL, κάτω από την συνολική κλίμακα της μέσης ανασταλτικής πυκνότητας (minimum inhibitory concentration range) του φαρμάκου. Στόχος και φιλοδοξία είναι η τεχνική αυτή να αναδειχθεί ως μια ταχεία μη επεμβατική αναλυτική μέθοδος μέτρησης φαρμακευτικών ή/και βιολογικών ουσιών σε σωματικά υγρά (π.χ. σίελος, δάκρυα, …), για την μελέτη της φαρμακοκινητικής, την έγκαιρη διάγνωση ασθενειών καθώς και τη μελέτη ελεγχόμενης αποδέσμευσης ενεργών ουσιών από πολυμερικές μήτρες σε πολύ μικρές συγκεντρώσεις. Επιπλέον, έγινε προσπάθεια μελέτης/ανίχνευσης μορίων Ε18 primary rat cortical cells με την χρήση του SERS σε συνθήκες προσομοίωσης ηλεκτροχημικού εμφυτεύματος. Για τον λόγο αυτό σχεδιάστηκε και αναπτύχθηκε ένας μικρο-επωαστής (micro-incubator) που επιτρέπει αφ’ ενός μεν τη συντήρηση των κυττάρων σε περιβάλλον κατάλληλο για τη μελέτη τους (layer of Neurobasal-culture medium with a 95% air & 5% CO2 gas flow at 37oC), αφ’ ετέρου δε την προσαρμογή του σε χώρο υποδοχής δείγματος ενός φασματοφωτόμετρου μικρο-Raman για τη λήψη φασματοηλεκτροχημικών μετρήσεων Raman/SERS. Το κύριο σημείο ενδιαφέροντος στο οποίο εστιάσαμε στην παρούσα εργασία αναφέρεται στην διερεύνηση ανίχνευσης βασικών/θεμελιωδών μορίων των κυττάρων στo θρεπτικό υγρό του πειράματος για την ταχεία εκτίμηση ενδεχόμενης αποδέσμευσης DNA που θα αντικατόπτριζε την κατάσταση των κυττάρων. / Laser Raman spectroscopy plays an increasing important role in polymer science, biophysics/biochemistry and life science. Based on vibrational transitions, it has long been regarded as a valuable non destructive tool for the identification of chemical and biological samples as well as the elucidation of molecular structure, surface processes and interface reactions. Spontaneous Raman scattering is however an inherently weak process characterized by extremely small cross sections. Even so, the Raman signal can be highly enhanced when the analyte is placed on or near either to nano-rough noble-metal substrates or to nano-structured colloidal clusters of noble metals. This nano-enhanced scattering process is known as Surface Enhanced Raman Scattering, SERS. With SERS, extremely small amounts of substances can be detected; even single molecule detection has been reported. This constitutes a challenge of applying SERS to extremely low concentration measurements. Nevertheless, the quantitative evaluation by means of SERS was proved difficult, due largely to lack of nano-sized noble metal structures with analytically suitable stability and reproducibility. The commonly used SERS substrates are nanostructured colloids or roughened surfaces of mainly silver and gold undergoing visible or NIR excitation. A note is made of the fact that the Raman scattering geometry that is actually largely used is the backscattering geometry. For solids this is better achieved via a microscope objective, while for liquids via a macro lens. However, the best scattering geometry for Raman light collection from liquids and nanostructured colloids is the right angle scattering; that is, the scattered light is collected at 90o from the excitation light. Having all above in mind, in order to perform quantitative SERS measurements we have designed and developed an oscillating cell making use of the right angle Raman scattering geometry. Originally, the development of the oscillating or “shaking” cell allows large sample sectional surface monitoring, better mixing and homogeneity giving rise to SERS signal reproducibility. In addition, the application of the advantageous for nanocolloidal solutions right angle Raman scattering collection geometry enables SERS measurements at extremely low concentrations. We demonstrated the use this new surface enhanced Raman scattering excitation/collection configuration to monitor the level of the antitumor drugs mitoxantrone and doxorubicin as well as the antibiotic ciprofloxacin at very low concentrations performing fast SERS & SERRS measurements. We have also quantified the concentration of the active agents in aqueous solutions as well as in artificial tears by a partial least-squares (PLS) chemo metric regression algorithm. All above reveal the potential of this technique in the monitoring of the controlled release of active agents from polymeric matrices as well as in the quantitative analysis of drugs in corporal fluids. The only need is to add in a test tube containing 0.5 mL of a nanocolloidal noble metal solution3 about 25-100 μL of a sampling fluid. Finally, in an attempt to detect “living” cells by SERS, to identify DNA components released in a cell culture medium as a “touch mark” of cell death and to perform even single molecule detection, a home-made micro-incubator was designed to allow Raman measurements of neuron cells under the microscope. Commercial micro incubators feature higher optical path between coverslips than the required (<1-1,5 cm) for typical micro-Raman systems even bearing long working distance objectives. It was at that sense a prerequisite to design, construct and optimize a micro-incubator to fit a conventional micro-Raman configuration. The designed micro-incubator meets well-defined stable conditions (temperature, culture solution and atmosphere).

Atributos do vinho, risco percebido e intenção de recompra online : análise do efeito moderador da qualidade do site

O interesse por esse tema se deu, primeiramente, pelo fato de o consumo de vinhos no Brasil ter apresentado um substancial crescimento nos últimos anos, o que se permite supor tratar-se de um segmento de mercado em franca expansão e, ainda, pelo crescente uso da internet como um canal alternativo de compras deste produto, o qual apresenta complexidades peculiares, desde o processo de decisão de compra até o seu consumo. Com base na revisão da literatura, foi elaborado o modelo conceitual deste estudo, assim como o questionário utilizado para verificar se as relações teóricas observadas se confirmariam na amostra selecionada para este trabalho. A coleta de dados foi operacionalizada a partir do envio do questionário, por e-mail, à uma amostra composta por 5.200 possíveis respondentes, todos usuários de um dos maiores e-commerce de vinhos do mundo, tendo sido coletadas 376 respostas, durante o mês de setembro de 2017, as quais foram submetidas às análises multivariadas por meio da técnica da modelagem de equações estruturais, com o auxílio do pacote PLS-PM utilizado no software “R”. Os resultados demonstraram que os atributos do vinho não apresentaram significância estatística na relação com o risco percebido em compras de vinhos online. Por outro lado, foram encontradas evidências de que o risco percebido em compras de vinhos online impacta negativamente a intenção de recompra de vinhos online. Por fim, não foram encontradas evidências que permitissem afirmar o efeito moderador da qualidade do site nas relações observadas neste estudo. Os resultados desta pesquisa foram discutidos e, ao final, apresentaram-se as suas limitações e algumas sugestões para pesquisas futuras. / The purpose of this study was to analyze the effect of site quality on the relationship between wine attributes and perceived risk in online wine purchases, and to analyze its effect on the relationship between perceived risk in online wine purchases and the online wine repurchase intention. The interest in this theme came first because the consumption of wines in Brazil has shown a substantial growth in recent years, assuming that it is a rapidly expanding market segment and due to the increasing use of the internet as an alternative channel of purchases of this product, which presents peculiarities, from the purchase decision process to its consumption. Based on the literature review, the conceptual model of this study was elaborated, as well as the questionnaire used to verify if the theoretical relationships would be confirmed in the sample selected for this study. The data collection were done by sending the questionnaire, by e-mail, to a sample composed of 5,200 potential respondents, all users of one of the largest e-commerce wines in the world. Three hundred and seventy-six responses were collected in September 2017, which were subjected to multivariate analysis using the structural equation modeling technique in the PLS-PM package using software R. The results showed that the attributes of the wine did not present statistical significance in relation to the perceived risk in online wine purchases. On the other hand, there is evidence that the perceived risk in online wine purchases negatively affects the online wine repurchase intention. Finally, no evidence was found to support the moderating effect of site quality on the relationships observed in this study. The results of this research were discussed and, at the end, its limitations and some suggestions for future research are presented.
