Comparative functional analysis of WOX genes during flower development in Petunia and Arabidopsis / Analyse comparative fonctionnelle des gènes WOX impliqués dans le développement de la fleur chez Petunia et Arabidopsis

Dans le domaine des plantes, la formation de la fleur a été un pas crucial dans la capacité des végétaux à coloniser une grande diversité de niches écologiques sur notre planète. Les deux espèces Petunia x hybrida et Arabidopsis thaliana représentent deux groupes majeurs des plantes à fleur. Nous avons montré que les gènes à homéodomaine d’une famille appelée WOX (Wuschel homeobOX) sont fortement impliqués dans le développement des organes dotés de polarité (dont les feuilles et des organes de la fleur : sépales, pétales, carpelles). Un double mutant (maw mawb), chez le Pétunia, développe des pétales en forme de filament, avec disparition du tube floral. De plus, nous avons découvert que ces mêmes gènes interagissent au niveau génétique avec d’autres gènes (appelés gènes à boite MADS) dans la formation des ovules, structures à partir desquelles les graines se forment. Nous avons aussi montré que des gènes de la même famille sont impliqués dans la formation d’autres structures chez le Pétunia : les trichomes ou poils aériens de surface. Ces derniers sont impliqués dans plusieurs taches, qui vont de la protection contre les pathogènes à celles contre les stress abiotiques. Grâce à des études de génétique fonctionnelle nous avons pu montrer un recrutement différentiel des gènes WOX ici étudiés, dépendant de l’organe et de l’espèce. Ces travaux de thèse montrent l’importance de cette famille génique pour les études d’evo-devo (Biologie Evolutionniste du Développement). Finalement, une analyse de RNA-Seq (séquençage du transcriptome), dévoile les réseaux génétiques contrôlés par ces gènes WOX. / In the Kingdom of Plants, the emergence of flowers was a crucial step in their ability to colonize a large variety of ecological niches on our planet. The two species Petunia x hybrida and Arabidopsis thaliana represent two major groups of flowering plants. In this work, we have shown that HOMEOBOX genes from the WOX family (Wuschel homeoboxes) are heavily involved in polar organ development (such as leaves and sepals, petals, and carpels at the flower level). The maw mawb double mutant in Petunia displays string-like petals, with consequent disappearance of the floral tube. Moreover, we found that these two genes genetically interact with genes from a different family (the MADS family) in ovule identity (ovules are the structures from which seeds develop). We have also shown that other genes from the WOX family are involved in development of a different kind of structures in Petunia: the trichomes. Trichomes are involved in different tasks, protecting the plant from pathogens or abiotic stress. Thanks to functional genetics studies, we have shown functional genetic recruitment of these WOX genes among different plant organs and among different species. This PhD thesis provides evidence for the importance of the WOX family in Evo-Devo studies. Eventually, we unravelled genetic networks controlled by MAW and MAWB trough RNA-Seq analysis.

Analysis of mass transfer in the emission of floral volatile organic compounds

<p>  </p> <p>Plants synthesize and release a variety of volatile organic compounds (VOCs) that are important for their reproduction, defense, and communication. These low-molecular-weight, lipophilic molecules also serve as practical products in industries such as food additives, fragrances, colorants, nutraceuticals, and pharmaceuticals. In addition, they have agricultural applications such as sustainable methods for pest control. Therefore, identifying the biological mechanisms involved in volatile emission could help researchers develop new ways to control the timing and release of volatiles, defend against pests, and engineer the production of these valuable chemicals.</p> <p>While progress has been made in understanding plant volatile biosynthesis, their release from the cell remains incomplete. For plant VOCs to be emitted into the environment, they must move from their site of biosynthesis through the cytosol, transverse the plasma membrane, hydrophilic cell wall, and sometimes cuticle to exit the cell. It was previously shown by mathematical modeling that to achieve observed emission rates solely by diffusion, VOCs would accumulate in the cellular membranes to levels that are likely detrimental to the membrane integrity and function. Hence, it was proposed that there are biological mechanisms involved to lower VOC concentrations in membranes. In this work, we focus on the aqueous cell wall, the thickest layer among the three subcellular barriers that should act as a barrier for the diffusion of VOCs. We hypothesize that the transport of VOCs across the cell wall is facilitated by lipid transfer proteins (LTPs) which enhance the solubility of hydrophobic volatiles in the aqueous environment, prevent their back partition into the plasma membrane after entering the cell wall, and hence enhance their net diffusion. To investigate if the presence of LTPs has influence on the total VOC efflux, we use reverse-genetic, biochemical, and mathematical modeling approaches. Out of three highly expressed LTPs identified in the petunia petal, only downregulation of <em>PhLTP3</em> expression led to a decrease in VOC emission in the corresponding transgenic plants. A facilitated diffusion model was built to quantify the VOC flux difference with the presence of LTPs in the cell wall. Modeling of the steady state system revealed the facilitation of VOC flux by LTPs is greatest when the VOC concentration gradient across the cell wall is shallow, which is a physiologically relevant condition. In addition, there exists an optimal protein dissociation constant value for maximal facilitated flux, indicating the balance between the binding and the unloading of VOC is critical. With the in vitro displacement assay, the binding constants of candidate PhLTPs with VOCs were obtained and were all found to be in the µM range, which is close to our model predicted optimal value. The results revealed that LTPs, specifically PhLTP3, play a role in the export of VOCs from the plasma membrane, across the cell wall, to the cuticle.</p> <p>In our earlier mathematical model, the emission of VOCs from the petunia flowers was modeled assuming negligible mass transfer resistance on the surface of the cuticle because of their high volatility. However, the resistance imposed by the surface boundary layer was not considered. To examine if surface convection influences VOC emission, a model system which utilized a model cuticular wax film containing 2-phenylethanol (2-PE) was built to imitate the VOC emission from plant cuticle. The convection mass transfer coefficient of 2-PE emission from a model cuticular wax film was obtained by experimental data fitting and calculated from the correlation that involves Sherwood number. The obtained values that were smaller than unity indicates that the surface boundary layer imposes a higher mass transfer resistance than the model cuticle for the emission of 2-PE in the range of wind velocities investigated. The examination of petunia flowers under air flow showed increases in total emission but no significant differences in total internal pools, which indicates an increase in biosynthesis. The emission changes of individual compounds were different and does not clearly correlate to any molecular properties of the compounds.  </p>

Genética de paisagens de espécies da planície costeira do Atlântico Sul

O entendimento da contribuição diferencial de processos neutros e adaptativos envolvidos na diferenciação genética entre populações, assim como sua relação com varáveis físicas e ambientais da área de distribuição das espécies, é fundamental para melhorar o conhecimento da história evolutiva, mas também para fazer um manejo e conservação mais adequados da diversidade genética das espécies. O surgimento da Planície Costeira do Atlântico Sul foi um processo relativamente recente, que conduziu a processos de colonização e expansão dos organismos para um ambiente costeiro. Os padrões de estrutura genética gerados em processos de colonização e expansão podem ser difíceis de interpretar devido ao fato de que podem apresentar sinais sobrepostos de efeito fundador em série, isolamento por distância e isolamento por ambiente quando envolvem gradientes ecológicos na área de estudo. No presente trabalho foram conduzidas caracterizações da diversidade e estrutura genética de dois taxa predominantemente costeiros co-distribuídos, Calibrachoa heterophylla e Petunia integrifolia ssp. depauperata, em toda a amplitude da distribuição. Também foram inferidas as dinâmicas de fluxo gênico entre populações e sua relação com variáveis topográficas e climáticas reconstruídas pelo meio de um levantamento exaustivo e modelamento para a área de estudo. Processos de diferenciação genética promovidos pelo regime diferencial de chuvas nos extremos da distribuição foram inferidos para as duas espécies. Também foram identificadas populações das duas espécies apresentando alto nível de mistura de identidade genética nas localidades ao redor da Lagoa dos Patos. Isso foi associado a alta instabilidade na história geomorfológica recente desta região e dinâmicas atuais do vento que favorecem a dispersão secundária de sementes a maiores distâncias. Adicionalmente foram identificados processos espécie-específicos que se relacionaram principalmente a fatores históricos de cada táxon. Em P. depauperata o efeito fundador relacionado a um processo único de colonização do ambiente costeiro determinou o nível superior de estrutura genética, enquanto que em C. heterophylla foi a história filogeográfica da espécie na qual a diferenciação intraespecífica é anterior à colonização da região costeira atual o fator preponderante. As diferenças de duração do ciclo de vida entre as espécies também influenciaram as dinâmicas contrastantes de fluxo gênico dos dois taxa, sugerindo que a colonização e adaptação local de C. heterophylla nas bordas da distribuição poderia ser condizente com um processo de monopolização. Em vista dos resultados obtidos neste trabalho, propõem-se o desenvolvimento de experimentos de transplante recíproco para confirmar o processo de adaptação local nas duas espécies e abordagens genômicas para identificar regiões do genoma responsáveis pelos processos de adaptação ao ambiente costeiro e de adaptação local nas margens da distribuição. / The understanding of differential contribution of neutral and adaptive processes to the genetic differentiation among populations, as well as its relationship to physical and environmental variables of species’ distribution area, is essential to improve the knowledge of species evolutionary history, but also to direct appropriate management and conservation policies for the genetic diversity. The emergence of the South Atlantic Coastal Plain was a relatively recent event that led to colonization and expansion processes to the coastal environment. Genetic structure patterns generated in colonization and expansion processes can be difficult to interpret because the overlapping signals, which can present the founder effect in series, isolation by distance, and isolation by environment in the presence of ecological gradients in the study area. In this work characterization diversity and genetic structure were conducted to two co-distributed and predominantly coastal taxa, Calibrachoa heterophylla and Petunia integrifolia ssp. depauperata alongside their complete geographical range. Moreover, we also inferred dynamic of gene flow among populations and investigated the relation between topographical and climatic variables reconstructed by means of an exhaustive survey and modeling for the study area and the gene flow. Shared genetic differentiation processes promoted by differential rainfall conditions at the distribution edges were inferred. In addition, we identified populations from both species with high level of mixed genetic membership in locations around the Patos Lagoon. This was associated with a high instability in recent geomorphological history of coastal region and current wind dynamics that favor the secondary seed dispersal over longer distances. Additionally, specific species processes were identified mainly related to historical factors of each taxon. In P. depauperata founder effects associated with unique colonization process to coastal environment determined the upper level of genetic structure, while in C. heterophylla the upper level of genetic structure was related to the phylogeographical history wherein the intra-specific differentiation preceded colonization to the current coastal region. The differences of the life span length of the species were also related to contrasting gene flow dynamics indicating that the colonization and local adaptation of C. heterophylla at the edges of the distribution could lead to monopolization process. In view of the results we propose the development of reciprocal transplant experiments to confirm the local adaptation process in both species and genomic approaches to identify regions of the genome responsible for the processes of adaptation to the coastal environment and local adaptation in distribution margins.

Shipping and nitrogen toning effects on postharvest shelf life of vegetative annuals

Vegetative annuals are currently popular in the ornamental horticulture industry. Many crops are newly domesticated species and little is known about how they perform during shipping or in the retail environment. Nine species and 21 cultivars were grown and underwent simulated shipping after harvest or nitrogen toning two weeks before harvest. Shipping was not found to affect the number of flowers on all but two cultivars post ship. Nitrogen toning affected vegetative growth of most Bracteantha bracteata (bracteantha) cultivars at harvest. All species had an effect due to toning postharvest. Bractenatha and Diascia ×hybrida (diascia) were chosen for further study due to their performance during these experiments. The effect of thidiazuron (TDZ) as a foliar spray and nitrogen toning on leaf yellowing and plant growth of bracteantha were evaluated. The two treatments were then combined to see how the two treatments worked together. It was found TDZ decreased leaf yellowing but its effects can be negated if the plants were not toned. Nitrogen toning reduced vegetative growth of the bracteantha without affecting the number of flowers on the plants. Diascia was found to have flower abscission in response to shipping. Further trials were conducted using 1- methylcyclopropene (1-MCP) an ethylene inhibitor. The effects of shipping duration and temperature were investigated. 1-MCP was found to hold flowers on treated plants longer postharvest than those not treated. Plants shipped for one day had no differences from the control but shipping for two days had a negative effect on plant quality. Postharvest shelf life was decreased when diascia was shipped at 24 °C when compared to cooler shipping temperatures. These results indicate shipping for no longer than one day and at less than 24 °C is recommended for diascia.

Efecto del tipo sustrato de cultivo, la fertilización y el agua de riego en la composición mineral y el desarrollo de Petunia x hybrida Vilm

Se ha realizado el estudio agronómico de Petunia para determinar opciones en el manejo de la planta referidas al sustrato, fertilizante y agua de riego a fin de producir plantines de calidad. El diagnóstico de la situación productiva indicó que los problemas existentes fueron: uso de agua de mala calidad, escaso empleo de fertilizantes y suelo orgánico como sustrato. Se comprobó la existencia de sectores dentro de esta zona de producción con características diferenciales en cuanto a la calidad de agua de riego por el aporte de salinidad de los acuíferos Puelche y Pampeano. Se observaron mayores valores en los parámetros de crecimiento cuando el agua empleada fue proveniente de la napa freática. Los 3 sustratos ensayados, poseyeron características químicas y físicas distintivas que condicionaron tanto el crecimiento y desarrollo como el contenido de macronutrientes en el tejido vegetal. Existieron ventajas comparativas del fertilizante granulado convencional respecto al de liberación lenta porque la respuesta de la planta dependió mayormente de la rápida disponibilidad de nutrientes, en los primeros 15 días tras el trasplante. El análisis de componentes principales combinado con el análisis en conglomerados jerárquicos se utilizó para sistematizar el estudio del efecto del sustrato, tipo y dosis de fertilizante y tipo de agua de riego en el desarrollo de petunia. Los indicadores de calidad que resultaron útiles en la mejora de la comercialización fueron el peso freso, peso seco, área foliar, flores/planta, el contenido de la clorofila y la intensidad de coloración de la flor. El tipo de fertilizante y agua de riego no afectaron a la calidad de la planta, a pesar de observarse incrementos de pH y de la CE al finalizar el ensayo. En etapas tempranas del desarrollo de la plántula, la presencia concreta de Na+ en el medio de crecimiento tuvo mayor incidencia y efectos más negativos que la propia salinidad. / Hashimoto, PN.
