Catabolisme de la proline et du GABA chez le colza : incidence de carences azotée et hydrique / Catabolism of proline and GABA in oilseed rape : impact of water and nitrogen deficiency

Dans le cadre du changement climatique et de l'évolution de la réglementation concernant les intrants azotés, la culture du colza risque d'être fortement pénalisée dans la mesure où c'est une culture qui nécessite d'importants apports azotés pour atteindre son potentiel de rendement. Par ailleurs, comme chez le colza un déficit hydrique induit l'accumulation de certains composés azotés, il est vraisemblable que cela conduise au détournement d'une quantité importante d'azote vers les organes végétatifs aux dépens des organes reproducteurs et donc du rendement. Chez le colza, la réponse métabolique au déficit hydrique se traduit par une très forte accumulation de proline et dans une moindre mesure une augmentation de la teneur en GABA (acide γ-aminobutyrique), deux acides aminés connus chez la plupart des plantes pour leur réponse à de nombreux stress abiotiques. L'objectif de cette thèse est de déterminer comment le métabolisme de ces deux molécules contribue à l'allocation de l'azote au cours du développement de la plante en situation normale comme en condition de stress hydrique et/ou azoté. Pour répondre à cette question nous avons fait le choix de caractériser deux voies enzymatiques majeures impliquées dans le catabolisme de la proline et du GABA : la proline déshydrogénase (ProDH) et la GABA-transaminase (GABA-T) et d'évaluer l'impact de carences hydriques et/ou azotées sur ces voies. Cette étude nécessitait d'identifier au préalable les gènes codant ces enzymes afin d'aborder une approche fonctionnelle. Les résultats montrent l'existence de multiples copies de gènes ProDH et GABA-T dans le génome du colza. L'analyse de leurs profils d'expression suggère que des processus de sub-fonctionnalisation sont en cours conduisant à l'expression spécifique, de certaines copies en réponse aux stress, et d'autres dans les processus développementaux. La comparaison des profils métaboliques avec les profils spécifiques des transcrits a permis d'élaborer des hypothèses sur le rôle de ces voies dans la gestion de l'azote. L'étude conjointe des métabolismes de la proline et du GABA suggère l'existence de régulations connexes entre les deux. Enfin, l'utilisation de plantules a permis - d'approfondir la régulation des gènes étudiés à des stades précoces de développement - et de mettre en évidence les effets délétères de l'inhibition de la GABA-T par une approche pharmacologique. En conclusion ces résultats apportent des précisions sur la régulation de ces deux enzymes et fournissent des éléments de réponse quant au rôle fonctionnel des catabolismes de la proline et du GABA dans les processus de gestion de l'eau et de l'azote chez le colza. Ces travaux constituent donc une première étape dans une démarche de validation de ces gènes comme candidats pour des programmes d'amélioration du colza visant à sélectionner des génotypes mieux adaptés aux conditions environnementales futures. / In the context of climate change and recent regulation concerning nitrogen inputs, the oilseed rape yields may be severely decreased because its crop requires significant nitrogen supply to reach high yield performance. Moreover, as water deficit induces the accumulation of some nitrogen compounds in oilseed rape, it is likely that this could lead to diversion of significant amounts of nitrogen to the vegetative organs at the expense of the reproductive ones and therefore of the yield. In oilseed rape, the metabolic response to water deficit results in a very high proline accumulation and, to a lesser extent, an increased content of GABA (γ-aminobutyric acid), both these amino acids known for their response to many environmental stresses in most species. The objective of the work presented here was to determine how the metabolism of proline and GABA contributes to the nitrogen allocation during plant development under optimal conditions and under water stress and/or nitrogen depletion. To answer this question, we have chosen to characterize two major enzymatic pathways involved in the catabolism of proline and GABA, proline dehydrogenase (ProDH) and GABA transaminase (GABA-T), and assess the impact of water and/or nitrogen deficiency on these pathways. This study has required to preliminary identify the genes encoding these enzymes in order to initiate a functional approach. The results show the presence of multiple copies of ProDH and GABA-T genes in the oilseed rape genome. Analysis of their expression profiles suggests that sub-functionalization processes are occurring, leading to the specific expression of some copies in response to stress, and some in developmental processes. Comparison of metabolic profiles with specific profiles of transcripts allows us to hypothesize about the role of these pathways in management of nitrogen. The combined study of proline and GABA metabolisms suggests the existence of relationships between them. Finally, the use of seedlings allows - further studying the regulation of genes in the early stages of development - and highlighting the deleterious effects of the inhibition of GABA-T by a pharmacological approach. In conclusion these results supply information on the regulation of these two enzymes and provide answers about the functional roles of proline and GABA catabolisms in the management processes of water and nitrogen in oilseed rape. These works constitute a first step in validation process of these genes as putative candidates for oilseed rape breeding programs to select genotypes better adapted to future environmental conditions.

Caracterização molecular e bioquímica da prolina desidrogenase de Trypanosoma cruzi, um possível alvo terapêutico. / Molecular and Biochemical Characterization of the proline dehydrogenase of T. cruzi, a possible therapeutical target .

Os nossos resultados demonstraram a atividade enzimática prolina desidrogenase (PRODH) do produto do gene anotado como codificante de uma prolina oxidase na base dados do genoma de Trypanosoma cruzi. A atividade da proteína codificada por esse gene foi avaliada inicialmente por complementação de uma linhagem de S. cerevisiae deficiente na expressão funcional da PRODH endógena, e posteriormente mediante a obtenção da enzima recombinante ativa expressa em um sistema bacteriano. Nos estudos feitos tanto com leveduras complementadas com o gene PRODH, quanto com as formas epimastigotas de T. cruzi, observamos que nesses organismos há uma correlação positiva entre o nível intracelular de prolina livre e a resistência ao estresse oxidativo. Através dos ensaios de RT-PCR quantitativo observamos que o RNAm do gene da PRODH é regulado ao longo do ciclo de vida do T. cruzi, com maior nível de expressão no estágio epimastigota intracelular, perfil apresentado também em experimentos de Western Blotting, o que é concordante com o papel fundamental desse aminoácido na diferenciação dessa forma para as forma tripomastigota. Nos diferentes ensaios de localização subcelular observou-se que a proteína PRODH está presente na mitocôndria. A seqüência da PRODH apresenta um sítio EF-hand de ligação a Ca2+. Mostramos experimentalmente a funcionalidade desse sítio e a sua função na regulação da atividade por Ca2+. Em ensaios de respiração mitocondrial, utilizando prolina como substrato, foi mostrado que a prolina fornece elétrons à cadeia respiratória através da PRODH, transferindo elétrons e gerando FADH2 no mesmo nível e com a mesma eficiência que o complexo II (succinato desidrogenase). O análogo de prolina T4C, inibidor do transporte de prolina, causa uma diminuição do conteúdo de prolina livre intracelular, o que foi relacionado com uma diminuição significativa da sobrevivência do parasito em combinação com o estresse nutricional e oxidativo, mostrando a participação do metabolismo de prolina na resistência a esses estresses. Todos esses dados sugerem ademais que o transportador de prolina do parasita pode ser um alvo para drogas terapêuticas de interesse quando combinada com outras drogas que agem via geração de espécies reativas de oxigênio. / In the present work, we demonstrated the proline dehydrogenase enzymatic activity (PRODH) for the protein encoded by a gene annotated as a proline oxidase in the T. cruzi genome data base. This activity was shown firstly through complementation of a S. cerevisiae lineage lacking its endogenous PRODH gene. The PRODH gene was also expressed in a bacterial system and the active recombinant protein was obtained. Experiments performed with both, complemented yeasts and T. cruzi epimastigotes, showed a correlation between the intracellular free proline levels and the oxidative stress resistance. Quantitative RT-PCR assay revealed that the PRODH gene is differentially expressed across T. cruzi life cycle, being the highest expression level shown by the intracellular epimastigote form, this result was confirmed by Western blotting. Both results are in accordance with the fact that proline is essential for the differentiation of the intracellular epimastigote into trypomastigote. Subcellular localization assays showed that PRODH is present preferentially in the mitochondria. In silico analyses of the PRODH peptidic sequence indicated the presence of an EF-hand domain, wich is, usually, involved in Ca2+ binding. In fact, our results confirm not only the ability of such domain of binding Ca2+ but also its function in the activity regulation. Mitochondrial respiration assays using proline as substrate showed that PRODH transfers electrons and generates FADH2, with an eficience comparable to that of the complex II (Succinate dehydrogenase). Experiments using the T4C, an analogue of proline that inhibits the proline uptake, caused the depletion of the intracellular free proline, which was followed by the significantly decrement of the cellular viability of the parasites under nutritional and oxidative stresses. Taken together, this data suggest that proline transporters are promising drug targets when combined with other drugs that act by generating reactive oxygen species.

Chiral thioureas, thiourea-phosphines and amines derived from biomass : synthesis and applications in asymmetric organocatalysis / Thiourées, thiourée-phosphines et amines chirales dérivées de la biomasse : synthèse et applications en organocatalyse asymétrique

Cette thèse porte sur la préparation de nouveaux catalyseurs chiraux à partir de composés naturels issus de la biomasse, et leurs applications en organocatalyse asymétrique. La première partie décrit la synthèse de thiourées énantiomériquement pures dérivées de l'isosorbide et l’isommanide. Ces thiourées ont été évaluées comme catalyseurs organiques dans des réactions asymétriques de Friedel-Crafts, d’alkylation, d’addition de type aza-Michael, d’hydroamination et de type Morita-Baylis-Hillman (MBH). Les meilleurs résultats en termes de réactivité et d’induction asymétrique, ont été obtenus dans le cas de la réaction de Friedel-Crafts. La deuxième partie est consacrée à la conception et la synthèse de thiourée-phosphines chirales à partir de la L-proline. Ces thiourée-phosphines sont capables de promouvoir la formation de liaisons C-N et C-S, selon un processus de substitution allylique énantiosélectif d’adduits modifiés de MBH. De très bons rendements (jusqu’à 98%) et énantiosélectivités (jusqu'à 93%) ont été obtenus. Dans la troisième partie, nous avons développé le premier exemple de cyclisation [4 + 2] entre un allénoate et un alcène tétrasubstitué, catalysée par une amine tertiaire. Nous avons montré que des composés de type 4H-pyranniques peuvent être sélectivement obtenus, avec d’excellents rendements, en utilisant le DABCO comme catalyseur. Dans le cas de catalyseur organique de type alcaloïde, le β-ICD conduit sélectivement aux composés 2H-pyranniques avec des excès énantiomériques jusqu’à 71 % ee. / The thesis focused on the preparation of new chiral catalysts derived from biomass and their applications in asymmetric organocatalysis. The first part described the synthesis of chiral thioureas derived from isosorbide and isomanide, naturally renewable resource, in moderate to good overall yields. These thioureas were evaluated in asymmetric reactions such as Friedel-Crafts alkylation, aza-Michael addition, hydroamination, Morita-Baylis-Hillman reaction. Good yields and enantioselectivities were obtained. The second part of our work went on the design and synthesis of chiral thiourea-phosphines from L-Proline. These thiourea-phosphines promoted C-N and C-S bond formation via the asymmetric allylic substitution of tert-butoxycarbonyloxy adducts. Good yields (up to 98 %) and enantioselectivities (up to 93 %) were observed. In the third part, we have developed the first example [4+2] annulation of allenoate and all-carbon tetrasubstituted alkenes catalyzed by an amine. In the case of DABCO catalyst, 4H-pyrans were isolated exclusively in good to excellent yield under mild reaction conditions. While employing β-ICD as catalyst, the enantioenriched 2H-pyran derivatives were obtained with enantiomeric excess up to 71 % ee.

Using Molecular Dynamics to Elucidate the Mechanism of Cyclophilin

Cyclophilins are ubiquitous enzymes that are involved in protein folding, signal transduction, viral proliferation, oncogenesis, and regulation of the immune system. Cyclophilin A is the prototype of the cyclophilin family. We use molecular dynamics to describe the catalytic mechanism of cyclophilin A in full atomistic detail by sampling critical points along the reaction coordinate, and use accelerated molecular dynamics to sample cis-trans interconversions. At these critical points, we analyze the conformational space sampled by the active site, flexibility of the enzyme backbone, and modulation of binding interactions.We use Kramer’s rate theory to determine how diffusion and free energy contribute to lowering the activation energy of prolyl isomerization. We also find preferential binding modes of several cyclophiln A inhibitors, and compare the conformational space sampled by inhibited cyclophilin A to the conformational space sampled during wild-type interactions. We also analyze the mechanism of the next family member cyclophilin B in order to probe differences in enzyme dynamics and intermolecular interactions that could possibly be exploited in isoform-specific drug design. Our results indicate that cyclophilin proceeds by a conformational selection binding mechanism that manipulates substrate sterics, electrostatic interactions, and multiple reaction timescales in order to speed up reaction rate. Conformational space sampled by cyclophilin when inhibited and when undergoing wild-type interactions share significant similarity. Cyclophilins A and B do have notable differences in enzyme dynamics, due to variation in intramolecular interactions that arise from variation in primary structures. This work demonstrates how computational methods can be used to clarify catalytic mechanisms.

The tautomerase superfamily is composed of a group of proteins characterized by two key features: the N-terminal proline and a beta-alpha-beta-motif. This superfamily has been divided into five families represented by 4-oxalocrotonate tautomerase (4-OT), 5-(carboxymethyl)-2-hydroxymuconate isomerase (CHMI), cis-3-chloroacrylic acid dehalogenase (cis-CaaD), malonate semialdehyde decarboxylase (MSAD), and macrophage migration inhibitory factor (MIF). Cg10062 is a homologue of cis-CaaD, but has several distinct biochemical properties from cis-CaaD. For example, Cg10062 can be irreversibly inhibited by (R)- or (S)-oxirane-2-carboxylate, whereas cis-CaaD can only be irreversibly inhibited by (R)-oxirane-2-carboxylate. FG41MSAD is a homologue of MSAD, with comparable decarboxylase activity but missing Arg-73 known to be crucial for the MSAD activity. In order to understand the unique biochemical characteristics of Cg10062 and FG41MSAD, we have solved five crystal structures. These crystal structures have established a solid structural basis for understanding the mechanisms of their activities. The eukaryotic protein phosphatases are composed of a group of proteins that are responsible for reversible phosphorylation. The eukaryotic protein phosphatases have been divided into three families, the phosphoprotein phosphatase (PPP) family, the protein phosphatase Mg2+- or Mn2+-dependent (PPM) family and the protein Tyr phosphatase (PTP) family. PDP1 is a member of PPM family. PDP1 is also an important component of the large pyruvate dehydrogenase complex (PDC) which catalyzes the decarboxylation of pyruvate to yield acetyl-CoA with the accompanying reduction of NAD+. In order to understand the mechanism in which it dephosphorylates its target protein we have solved the structure of the catalytic domain of PDP1. Analysis of these structures in the light of their evolutionary contexts enables us to appreciate the unity and diversity of the biological systems at the chemical level and help us solve interesting problems, such as the possible physiological functions for some members within the tautomerase superfamily. / text

Drėgmės trūkumo poveikis žirnių morfofiziologiniams rodikliams / The influence of moisture lack on pear morphofizioligical indices

Magistrantūros studijų baigiamajame darbe pateikiami žirnių fotosintezės pigmentų, prolino, sacharidų bei santykinės drėgmės pokytis lapuose esant drėgmės deficitui. Tyrimų objektas – `Ilgiai` veislės sėjamasis žirnis (Pisum sativum L.). Darbo metodai. Tyrimai atlikti Aleksandro Stulginskio universiteto Agrobiotechnologijos laboratorijoje 2012-2013 metais. Žirniai pasėti į 0,16 m x 0,23 m (aukštis x diametras) vegetacinius indus su substratu (pH − 6,31, P2O5 − 633,94 mg kg-1, K2O – 912 mg kg-1). Eksperimentas vykdytas 6 pakartojimais po 5 augalus inde. Augalai auginti programuojamoje auginimo kameroje esant 20/18 °C (diena/naktis) temperatūrai, 16/8 val. (diena/naktis) fotoperiodui, 50 µmol m-2 s-1 apšviestumui. Augalai auginti programuojamoje auginimo kameroje aukščiau nurodytomis sąlygomis bei esant drėgmės deficitui substrate. Sausros tyrimas pradėtas po sėjos praėjus trims savaitėms. Darbo rezultatai. Esant drėgmės deficitui, patikimai didžiausi chlorofilo a ir b bei karotenoidų kiekiai žirniuose (atitinkamai 2,501 ir 0,821, bei 1,059 mg l-1) nustatyti praėjus 8 dienoms po laistymo. Esmingai didžiausi prolino ir sacharidų kiekiai (atitinkamai 46,543 μM g-1 ir 0,155 g kg-1) žirniuose, esant drėgmės deficitui, nustatytas praėjus 12-ai dienų. Dirvožemio drėgnis (SWC) esmingai kito visą tyrimų laikotarpį. Mažiausias dirvožemio drėgnis (15,12 proc.) nustatytas paskutinę tyrimų dieną, praėjus 16-ai dienų po paskutinio laistymo. The object of researches – ‘Ilgiai’ variety of sowing peas (Pisum sativum L.) Methods. Research carried out at Aleksandras Stulginskis University in Laboratory of Agrobiotechnology 2012–2013. Peas were sown in 0.16 m x 0.23 m (height x diameter) vegetative vessels with the substratum (pH – 6.31, P2O5 – 633.94 mg kg-1, K2O – 912 mg kg-1). The experiment was carried out in 6 replicates of 5 plants in container. Plants were cultivated at programmable climatic chamber at 20/18 °C (day/night) temperature, 16/8 hours (day/night) photoperiod, 50 µmol m-2 s-1 illumination. Plants were grown at programmable climatic chamber by conditions which are set out above and with deficit of moisture in substrate. The investigation of drought launched after three weeks of sowing. Results. In the moisture deficit, reliably the largest chlorophyll a and b and carotenoids amount in peas (respectively 2.501 and 0.821, and 1.059 mg l-1) were determined after 8 days after watering. Essentially the maximum proline and saccharins contents (by 46.543 μM g-1 and 0.155 g kg1) in peas at the moisture deficit were set after 12 days. Os genes de Prolina racemase (PRACs) são enzimas intracelulares ou secretadas de T. cruzi (TcPRAC); essas enzimas estão envolvidas no metabolismo, diferenciação e virulência. Os genes PRAC foram identificados e caracterizados molecularmente em isolados representantes de toda a diversidade interespecífica de T. cruzi, T. cruzi marinkellei, T. dionisii, T. erneyi, T. rangeli, T. conorhini e T. lewisi. Além dessas espécies de tripanossomas restritas a mamíferos; homólogos de PRAC foram encontrados em tripanossomas de cobra (T. serpentis), crocodilo (T. grayi) e anuro (T. sp. 339). Análises filogenéticas e de sintenia entre homólogos de PRAC suportaram uma historia evolutiva totalmente congruente com as relações evolutivas previamente descritas dentro do gênero Trypanosoma. / Proline racemaces (PRACs) are intracellular or secreted enzymes of Trypanosoma cruzi (TcPRAC), implicated in metabolism, differentiation, virulence and the induction of nonspecific polyclonal B-lymphocyte in the host. We identified and molecularly characterized PRAC genes from isolates representing all intra-specific diversity (TcI-TcVI and Tcbat), T. cruzi marinkellei, T. dionisii, T. erneyi, T. rangeli (isolates of lineages A-E), T. conorhini, and T. lewisi. In addition to these trypanosome species restricted to mammals, PRAC homologs were found in trypanosomes from snake (T. serpentis), crocodile (T. grayi) and anuran (T. sp 339). Phylogenetic and synteny analysis of PRAC homologs supported an evolutionary history totally congruent with the evolutionary relationships within the genus Trypanosoma.

Produção e caracterização do biossurfatante produzido pela bactéria marinha Brevibacterium luteolum / Production and characterization of a biosurfactant produced by marine bacterium Brevibacterium luteolum

Os biossurfatantes (BS) são produtos de origem microbiana com propriedades tensoativas e emulsificantes. Estes compostos são candidatos para substituir os surfatantes sintéticos em aplicações industriais devido à sua menor toxicidade, maior biodegradabilidade, maior diversidade química e maior eficiência e efetividade em condições físicas extremas de salinidade, pressão e temperatura. O uso comercial e industrial dos BS ainda não é sustentável devido a seu alto custo de produção relacionado principalmente ao baixo rendimento. A utilização de substratos de baixo custo e ferramentas estatísticas para melhorar o rendimento de produção dos BS são duas das principais estratégias para tratar este problema. O objetivo do trabalho foi estudar a produção e recuperação do BS produzido por B. luteolum, visando o melhoramento na sua produção através do uso do planejamento fatorial e caracterizar a estrutura química do BS. A partir dos resultados, determinou-se que a adsorção em resina foi mais efetiva para a recuperação do BS comparada com a precipitação ácida. A produção do BS foi melhorada através de um planejamento fatorial 23 usando como fatores as concentrações da fonte de carbono (vaselina), a fonte de nitrogênio (nitrato de amônio) e a água do mar artificial e como resposta a tensão superficial da solução 0,1% de BS. A maior produção de BS foi obtida com 4% de fonte de carbono, 2% de fonte de nitrogênio e 20% de água do mar artificial gerando tensão superficial de 27 mNm-1. O BS foi caracterizado como uma mistura de lipopeptídeos com ácidos graxos cujo comprimento da cadeia variou entre 10-18 unidades de carbono e um conteúdo de proteína total de 5%. Três estruturas químicas foram sugeridas para os compostos ativos: dois prolina-lipídeos com os ácidos graxos C16:0 e C18:0 respectivamente e um lipopeptídeo com uma sequência peptídica Phe-Al-X-X-Pro-Pro-Thr (X=Leu/Ile) ligada a uma cadeia de ácido graxo C16:0. Não observou-se atividade antimicrobiana contra as cepas de S. aureus, E. coli, S. enteritidis, L. monocytogenes e S. mutans nas faixas de concentrações de BS testadas. O uso de vaselina como substrato para a produção do BS sugere que a bactéria e o BS podem ser explorados para aplicações como a biorremediação e a recuperação melhorada de petróleo (EOR). / Biosurfactants (BS) are microbial-derived molecules showing tensoactive and emulsification properties. These compounds are candidates to replace synthetic surfactants for industrial applications due to their less toxicity, greater biodegradation capacity, greater chemical diversity and greater efficiency and effectiveness under extreme physical conditions of salinity, pressure and temperature. Commercial and industrial use of BS is not sustainable due their high production cost mainly related to low production yields. The use of low cost substrates and statistical tools to enhance the production yield of biosurfactants are two of the main strategies to deal with that problem. The objective of this work was to study the production and recovery of the BS produced by B. luteolum, aiming to enhance its production through a factorial experimental design, and to characterize the chemical structure of the BS. It was found that resin adsorption was more effective than acid precipitation to recover the BS. The production of BS was enhanced through a factorial experimental design 23 using the concentrations of the carbon source (mineral oil), the nitrogen source (ammonium nitrate) and artificial seawater as the factors and the surface tension of a solution 0,1% of BS as the response. The value of factors that enhanced the production of BS were 4% of carbon source, 2% of nitrogen source and 20% of artificial sea water showing a surface tension of 27mNm-1. The BS was characterized as a mix of lipopeptides with fatty acid chains varying between 10-18 carbon units and a total protein content of 5%. Three chemical structures were proposed for the active compounds: two proline-lipids with the fatty acid chains C16:0 e C18:0 respectively and a lipopeptide with a peptide sequence Phe-Al-X-X-Pro-Pro-Thr (X=Leu/Ile) linked to a fatty acid chain C16:0. BS did not show antimicrobial activity against S. aureus, E. coli, S. enteritidis, L. monocytogenes and S. mutans at concentration range tested. The use of mineral oil as a substrate for the production of the BS suggests that the bacteria and the BS can be explore for applications as bioremediation and enhanced oil recovery (EOR).

Caracterização molecular e bioquímica das enzimas envolvidas na biossíntese de prolina em Trypanosoma cruzi. / Molecular and biochemical characterization of the enzymes involved in proline biosynthesis in Trypanosoma cruzi.

Alguns organismos podem biossintetizar L-prolina (L-Pro) principalmente via L-glutamato (L-Glu). A Δ1-pirrolina-5-carboxilato (P5C) sintase (P5CS) converte o L-Glu ao intermediário da via, o P5C que uma vez formado é reduzido a L-Pro via P5C redutase (P5CR). Nesse trabalho, verificou-se a possível ocorrência dessa via em Tritryps. Em Trypanosoma cruzi toda a via é operativa, em Trypanosoma brucei está ausente, enquanto que em Leishmania amazonensis acontece parcialmente. A biossíntese de L-Pro em T. cruzi é abordada com mais detalhes. Ambas as enzimas da via foram expressas e tiveram sua localização determinada no citosol, compartimento distinto das enzimas do catabolismo de L-Pro. Ainda realizou-se a caracterização bioquímica da TcP5CR, que curiosamente sofre inibição incompetitiva pelo NADPH, e por isso é candidata a regular esse metabolismo. Por fim, evidenciou-se a saída do P5C da mitocôndria, possibilitando a existência de um mecanismo de transidrogenação via a interconversão de L-Pro em P5C e vice versa. / Some organisms biosynthesize L-proline (L-Pro) mainly from L-glutamate (L-Glu). The Δ1-pyrroline-5-carboxylate (P5C) synthase (P5CS) converts L-Glu into the intermediate P5C, which in turn, is reduced to L-Pro by a P5C reductase (P5CR). In this study, we verified the possible occurrence of this pathway in Tritryps. In Trypanosoma cruzi the entire route is operative. In Trypanosoma brucei, is absent, while Leishmania amazonensis this pathway is partially operative. In T. cruzi, both enzymes were expressed and had their location determined in the cytosol, differently from those of the catabolism of L-Pro. In addition, the biochemical characterization of TcP5CR showed an uncompetitive inhibition by NADPH, and therefore it is a candidate to be a regulation point of the pathway. Finally, it was observed the exit of PC5 from the mitochondria, a condition for the existence of a transhydrogenation mechanism via the interconversion between Pro and P5C.

Origem, evolução e relações filogenéticas de homólogos de prolina racemase em espécies de Trypanosoma. / Origin, evolution and phylogenetic relationships of proline racemase homologs from species of Trypanosoma.

Os genes de Prolina racemase (PRACs) são enzimas intracelulares ou secretadas de T. cruzi (TcPRAC); essas enzimas estão envolvidas no metabolismo, diferenciação e virulência. Os genes PRAC foram identificados e caracterizados molecularmente em isolados representantes de toda a diversidade interespecífica de T. cruzi, T. cruzi marinkellei, T. dionisii, T. erneyi, T. rangeli, T. conorhini e T. lewisi. Além dessas espécies de tripanossomas restritas a mamíferos; homólogos de PRAC foram encontrados em tripanossomas de cobra (T. serpentis), crocodilo (T. grayi) e anuro (T. sp. 339). Análises filogenéticas e de sintenia entre homólogos de PRAC suportaram uma historia evolutiva totalmente congruente com as relações evolutivas previamente descritas dentro do gênero Trypanosoma. / Proline racemaces (PRACs) are intracellular or secreted enzymes of Trypanosoma cruzi (TcPRAC), implicated in metabolism, differentiation, virulence and the induction of nonspecific polyclonal B-lymphocyte in the host. We identified and molecularly characterized PRAC genes from isolates representing all intra-specific diversity (TcI-TcVI and Tcbat), T. cruzi marinkellei, T. dionisii, T. erneyi, T. rangeli (isolates of lineages A-E), T. conorhini, and T. lewisi. In addition to these trypanosome species restricted to mammals, PRAC homologs were found in trypanosomes from snake (T. serpentis), crocodile (T. grayi) and anuran (T. sp 339). Phylogenetic and synteny analysis of PRAC homologs supported an evolutionary history totally congruent with the evolutionary relationships within the genus Trypanosoma.
