Recent studies show that within the marine environment algae stand out as organisms with great biotechnological potential by having in its composition various bioactive molecules that have diverse and intense biological activities, among all algal groups in this study emphasize the of red algae, which the kind of study is red seaweed Hypnea musciformis (Wulfen). JV Lamouroux belongs to the order Gigartinales and Hypneaceae family, abundant on the coast of CearÃ. Biotechnological and scientific works are carried out with this species, in this sense, the main objective of this work is to identify and characterize proteins existing in this organism through a proteomics approach, is a new area in science that allows bioprospect a set of proteins in an organism by using analytical tools because under specific physiological conditions the biological functioning of the system can be reflected by the proteome analysis of the same, and thus the functional gene expression. Considering the lack of studies on proteomics in seaweed and in particular the carragenÃfita Hypnea musciformis, this paper aims to develop methods for separating proteins from protein extract of Hypnea musciformis and identify the proteins expressed by the organism. In this work we have established a methodology for separating proteins of this organism from a fractionated extract with ammonium sulfate and by mass spectrometry to identify proteins of some kelp, thus being of fundamental importance for the enrichment of macroalgae in the proteomics literature marine. The results of this work are consistent with our objective was the identification of three proteins by mass spectrometry using electrospray ionization type protein fraction of 20-40% and the identification of some proteins by Mascot / Atualmente vivemos em uma acirrada competiÃÃo na pesquisa de desenvolvimento de novos produtos biotecnolÃgicos, o ambiente marinho pode ser visto como uma fonte de materiais biotecnolÃgicos funcionais. Recentemente, estudos demostram que dentro do ambiente marinho as algas se destacam como organismos com grande potencial biotecnolÃgico por possuirem em sua composiÃÃo vÃrias molÃculas bioativas que apresentam diversas e intensas atividades biolÃgicas. Dentre todos os grupos algais, neste trabalho enfatizamos o das algas vermelhas, a qual a espÃcie de estudo à a alga marinha vermelha Hypnea musciformis (Wulfen). J. V. Teor de proteína no concentrado de vacas no terço inicial da lactação, mantidas em pasto de capim elefante / Protein levels in the concentrate for early lactation cows maintained in an elephant grass pasture

O objetivo com este estudo foi avaliar o efeito de teores crescentes de proteína no concentrado de vacas leiteiras no terço inicial da lactação mantidas em pastagens de capim elefante (Pennisetum purpureum), manejadas com altas taxas de lotação durante o período chuvoso, sobre as seguintes variáveis: 1) produção e composição de leite; 2) peso vivo e condição corporal dos animais; 3) consumo de matéria seca; 4) parâmetros sanguíneos; 5) parâmetros ruminais; 6) composição bromatológica e morfológica da pastagem; e 7) comportamento animal. Buscou-se também a caracterização da PB do capim e a adequação proteica da dieta, visando otimizar a eficiência de utilização do nitrogênio. Foram utilizadas 30 vacas (HPB × Jersey) distribuídas em 10 grupos homogêneos, cada um compondo um quadrado latino 3x3, de acordo com a composição racial, dias em lactação e produção de leite e receberam suplemento concentrado com três níveis de PB durante períodos de 21 dias. Os concentrados continham milho moído fino, minerais, vitaminas e os três níveis de proteína (T1: 7,8; T2: 15,4; T3: 20,5% PB) foram obtidos substituindo-se parte do milho por farelo de soja. O capim elefante foi adubado com 50 kg N ha-1 por pastejo e apresentou teor médio de PB de 15,5%. As produções de leite e de leite corrigido para 3,5% de gordura, os teores e produções de gordura, proteína e caseína no leite, o consumo de matéria seca (CMS) não foram influenciados (P>0,05) pelos teores de PB dos concentrados. A concentração de nitrogênio ureico no leite aumentou linearmente (P<0,01) com o aumento dos teores de PB dos concentrados. A excreção de N pela urina foi maior (P<0,01) para os animais recebendo os concentrados contendo farelo de soja (T2 e T3) em relação àqueles consumindo o concentrado contendo apenas milho (T1). Além disso, a concentração de N amoniacal no rúmen foi menor (P<0,05) para estes últimos animais, que apresentaram, ainda, perda de peso durante o período experimental. A excreção total de N no ambiente acompanhou o comportamento do teor de PB dos concentrados. O fornecimento de farelo de soja no concentrado de vacas em terço inicial de lactação, mantidas em pastagens tropicais manejadas intensivamente, não resulta em benefício produtivo, mas favorece a acréscimo do peso corporal dos animais. / The aim of this study was to evaluate the effects of increasing levels of protein in the concentrate for early lactation cows maintained in an elephant grass pasture (Pennisetum purpureum), which was managed under high stocking rate during the raining season. The following variables were evaluated: 1) milk yield and composition; 2) body weight and condition score; 3) dry matter intake; 4) blood parameters; 5) rumen parameters; 6) chemical and morphological composition of the pasture; and 7) animal behavior. Protein fractionation of the pasture, and diet protein balancing were also studied as a way to improve efficiency of nitrogen use. Thirty Holstein × Jersey cows, distributed in 10 homogenous groups according to breed composition, days on lactation, and milk production, were used. In each group a 3×3 Latin square experiment was designed. Treatments consisted of 3 levels of crude protein in the concentrate supplement (T1: 7.8; T2: 15.4; T3: 20.5% CP). Concentrates were composed of finely ground corn, and mineral and vitamin mixture. Protein levels were obtained by adding soybean meal. Elephant grass pasture was fertilized with 50 kg N ha-1 after every grazing period, and presented 15.5% CP in average. Milk yield, fat corrected milk (3.5% fat), milk fat (g day-1 and %), milk protein (g day-1 and %), milk casein (g day-1 and %), and dry matter intake were not affected (P>0.05) by the supplement protein level. Concentration of ureic nitrogen in the milk increased linearly (P<0.01) as protein supplementation was increased. Nitrogen excretion in the urine was higher (P<0.01) for cows receiving the concentrates with soybean meal (T2 and T3), in comparison with those fed supplements containing just corn (T1). Moreover, these later had lower N-NH3 concentration in the rumen (P<0.05), and a negative body weight gain. Total N excretion to the environment follows the increasing in the protein level supplementation. Feeding soybean meal to early lactation cows, maintained in an intensively managed tropical pasture, does not result in productive benefits; however, it promotes an increasing in body weight of the animals.

Utilization of canola seed fractions in ruminant feeds

Canola fibre-protein and can-sugar are the two by-products arising from a process for separating high quality protein fractions from canola meal. In the first trial chemical characteristics of fibre-protein and can-sugar were examined in comparison with commercial canola and soy meal. In the second trial in situ rumen degradability and kinetics of test feed was studied. Based on the findings of those two trials, available energy values were estimated based on NRC (2001) while protein contents potentially absorbable at small intestine were predicted using both NRC (2001) and DVE/OEB models. Subsequently a mixture of fibre-protein and can-sugar was used as an additive to dehydrated alfalfa pellet and two dairy cow trials were conducted to determine the palatability and examine effect on lactation performances of blended alfalfa pellet feeding in comparison with standard alfalfa pellet. Palatability difference was evaluated by Paterson -two choice alternating access method through a 7 day experimental period using 6 lactating Holstein cows. In the lactating performance trial, 6 cows were randomly assigned into two groups and two treatments were allocated over three experimental periods in a switchback design. <p>Can-sugar consisted of water soluble components (CP 15.6 %DM; SCP 96.2 %CP; NFC 99.9 %CHO) with non-protein nitrogen as the main CP fraction (NPN 96.2 %CP). Fibre-protein was a highly fibrous material (NDF: 55.6%; ADF: 46.3%; ADL: 24.1%) comparing to canola meal (NDF: 25.4%, ADF: 21.2%, ADL: 9.0%) due to presence of higher level of seed hulls in fibre-protein. Comparing to canola meal, fibre-protein contained 9% less CP and 1/4 of that consisted of undegradable ADIP. Rumen degradability of can-sugar was assumed as immediate and total as it was water soluble. Most of the ruminally undegradable nutrient components present in canola meal appeared to be concentrated into fibre protein during the manufacturing process and as a result fibre-protein has shown a consistently lower effective degradability of DM, OM, CP NDF and ADF comparing to both canola and soy meal. Available energy content in can-sugar was marginally higher than that of canola meal while fibre-protein contained only 2/3 that of canola meal. The predicted absorbable protein content at small intestine was about 1/2 that of canola meal. These results indicate that fibre-protein can be considered as a secondary source of protein in ruminant feed and a mixture of fibre-protein and can-sugar would nutritionally complement each other to formulate into a cheaper ingredient in ruminant ration. In the palatability study, there was no significant difference (P>0.05) in intake preference or finish time between the blended and standard alfalfa pellets. The results from the lactation study showed that there was no significant difference (P>0.05) in milk yield, dairy efficiency or milk composition between the blended and standard alfalfa pellets. The results from the two studies indicated that fibre-protein and can-sugar fractions could be used as an additive to alfalfa dehydrated pellet at 15% inclusion rate without compromising its palatability or the performance of dairy cows. <p>For future studies it is proposed to conduct feeding trials with varying levels of inclusions to alfalfa pellet to know the nutritional effect of fibre-protein and can-sugar while ascertain optimum inclusion rate.

Experiment was conducted to evaluate the productive potential and chemical composition of the dry mass of three cultivars of pearl millet (Pennisetum glaucum (L.) R. Brown) (ADR-7010, ADR- 500 and BRS-1501), submitted to sources of phosphorus (superphosphate and fused), nitrogen (0, 40, 80 and 160 kg ha-¹) in the form of urea in three cuts in the UFG / Goiânia-GO . The experimental design was completely factorial arrangement 2 x 3 x 4 with four replications. The parameters evaluated were dry matter production (DMP) of the whole plant, while the dry matter (DM), crude protein (CP), acid detergent fiber (ADF) and neutral detergent fiber (NDF) were determined for the whole plant and its fractions (leaf and stem). Only the whole plant was given the apparent conversion efficiency of nitrogen (NACS), apparent nitrogen recovery (NAR) and nitrogen accumulated in the plant (NA) and protein fractions (A, B1, B2, B3 and C). All tests were accomplished using the SAS program (2007). Statistical analysis revealed no significant interactions between the variables studied in this way, these were discussed in isolation. Among the evaluated cultivars the highest yield (P <0.05) was the cultivar ADR-7010 (1175 kg ha-1 DM). The increment of N increases in production. NA increased with the increase of N rates with maximum value of 42.70 kg. The NACS and RAN decreased with the addition of the nutrient, with a peak of 10.77 kg ha-1 and 33.44%. The DM content of whole plant and leaf differed (P <0.05) and stem fraction only differed among genotypes. The CP content of whole plant did not differ (P> 0.05), but increased with the succession of cuts. The leaf blade was no difference (P <0.05) reduction of CP in terms of cuts. The CP concentration of stem ranged from 8.90% to 14.04%. ADF whole plant increased due to the succession of cuts and leaf blade varied between the cultivars, the highest contents of ADF were found in the stem. The NDF in the whole plant did not differ between the N levels, however all values were below 60%. The greatest amount recorded in the leaf was 63.81%. The higher NDF were identified in the stem fraction (55.54% to 68.03%). The contents of HEM differ between courts and for all plant cultivars and their fractions. There was no significant difference (P> 0.05) for the nitrogen fractions among all treatments. The contents determined for fraction C were all lower than 10%. Cultivar ADR-7010 showed the best results as well as the source superphosphate. The sources of phosphorus and nitrogen fertilization influenced the parameters. / A produção de massa seca e aspectos qualitativos de milheto forrageiro ainda são pouco conhecidos, assim como informações específicas dessa gramínea manejada com fertilização fosfatada e nitrogenada. Foi conduzido experimento para avaliar o potencial produtivo e composição bromatológica da massa seca de três cultivares de milheto (Pennisetum glaucum (L.) R. Brown) (ADR-7010; ADR- 500 e BRS-1501), submetidos à fontes de fósforo (superfosfato simples e termofosfato), doses de nitrogênio (0; 40; 80 e 160 kg.ha-¹) sob a forma de uréia em três cortes na UFG/Goiânia-GO. O delineamento experimental foi inteiramente casualizado com arranjo fatorial 2 x 3 x 4, com quatro repetições. Os parâmetros avaliados foram produção de massa seca (PMS) da planta inteira, enquanto que os teores de matéria seca (MS), proteína bruta (PB), fibra em detergente ácido (FDA) e fibra em detergente neutro (FDN) foram determinados para a planta inteira e suas frações (lâmina foliar e colmo). Somente para a planta inteira foi determinada a eficiência de conversão aparente de nitrogênio (ECAN), recuperação aparente do nitrogênio (RAN) e nitrogênio acumulado na planta (NA), assim como as frações de proteínas (A, B1, B2, B3 e C). Todas as análises foram realizadas com auxílio do programa computacional SAS (2007). A análise estatística não revelou interações significativas entre as variáveis pesquisadas, dessa forma, as mesmas foram discutidas isoladas. Entre os cultivares avaliados a maior produção (P<0,05) foi do cultivar ADR-7010 (1.175 kg.ha-1 de MS). O incremento de N promoveu aumentos na produção. O NA aumentou em função da elevação das doses de N com valor máximo de 42,70 kg. A ECAN e RAN diminuíram com o acréscimo do nutriente, com valores máximos de 10,77 kg.ha-1 e 33,44%. Os teores de MS da planta inteira e lâmina foliar diferiram (P<0,05) e a fração colmo somente diferiu entre os cultivares. Os teores de PB da planta inteira não diferiram (P>0,05), porém, aumentaram com a sucessão dos cortes. Na lâmina foliar houve diferença (P<0,05) com redução do teor de PB em função dos cortes. Os teores de PB do colmo variaram de 8,90% a 14,04%. Os teores de FDA da planta inteira aumentaram devido à sucessão dos cortes e na lâmina foliar variaram entre os cultivares, os maiores conteúdos de FDA foram verificados no colmo. Os teores de FDN na planta inteira não diferiram entre as doses de N, entretanto todos os valores foram inferiores a 60%. O maior teor registrado na lâmina foliar foi de 63,81%. Os maiores valores de FDN foram identificados na fração colmo (55,54% a 68,03%). Os teores de HEM diferiram entre cortes e cultivares para planta inteira e suas frações. Não verificou-se diferença significativa (P>0,05) para as frações nitrogenadas entre todos os tratamentos. Os conteúdos determinados para a fração C, foram todos inferiores a 10%. O cultivar ADR-7010 apresentou os melhores resultados, assim como a fonte superfosfato simples. As fontes de fósforo e fertilização nitrogenada influenciaram os parâmetros avaliados.
