A Meeting of Land and Water

What is the opportunity afforded by a distinction in geography? Can a building respond to a threshold, or meeting point between two distinct geographical conditions? In particular, the duality of land and water and the moment when these two elements meet, could indicate and influence form and structure. What is a potential response to such a condition in geography, what are the mechanisms and means by which a building could respond to such a condition? This project seeks to highlight and celebrate the distinction between land and water, a meeting point where land ends and a river begins. The performance of PE investments is assessed by analyzing the risk-return relationship relative to simulated Public Market (PM) investments that mimic the cash flow patterns of PE investments. The model allows for a quantified analysis of the underlying inputs that outline the PE performance and risks, and accounts for survivorship bias. These inputs include the fund manager’s decisions regarding the selection, leverage, size, duration and timing of investment and divestments. / Este estudo apresenta um modelo de projeção de investimentos alternativos para estimar os valores futuros de investimentos de Private Equity (PE). O desempenho dos investimentos de PE é avaliado pelo risco-retorno em relação a investimentos de Mercados Públicos simulados de forma a imitar os padrões de fluxo de caixa dos investimentos de PE. O modelo permite uma análise quantificada dos inputs que caraterizam o desempenho e riscos de investimentos de PE, e tem em consideração rácios de sobrevivência (survivorship bias) destes investimentos. Estes inputs incluem decisões dos gestores do fundo em relação à seleção, alavancagem, tamanho, duração e timings dos investimentos e desinvestimentos.

At the beginning of the twentieth century, navigation was the most used means of transport, both for driving loads such as for locomotion. Border with Argentina, Itaqui, a border city of the state, at that time, used to use its port as defense and trade coming mainly from Argentina and Uruguay as well as European countries. The Public Market Itaqui appeared because of its location and the necessity to have an appropriate space for trade. This market lasted a few decades, but was beaten by several factors. Although the importance, recognized as the State Heritage, the building lost its value, and gave space for abandonment and disuse. Based in these aspects, this research aims to provide theoretical basis to the intervention area regain its original sense, in order to rehabilitate the environment and redeem their historical and architectural importance. These theoretical and practical researches, underlie and materialize these goals, along with issues related to the area. Thus, through the photographic survey, metric and analyze the current state of conservation, were drawn up maps of damage that were the basis for the development of guidelines for conservation and maintenance of the building, so that, the building perpetuates over time. From the data collected, was drawn up the damage map, in which was found the constant presence of biofilm, result of the lack of maintenance. Finally, methods were developed to serve as basis the rehabilitation and recovery guidelines in order to permit the future integrity of the building, perpetuating it over the time. / No inicio do século XX, a navegação era o meio de transporte mais utilizado, tanto para condução de cargas, como para locomoção. Itaqui, uma cidade fronteiriça do Estado do Rio Grande do Sul, faz divisa com a Argentina e utilizava, na época, o seu Porto como defesa e para as trocas de entrepostos comerciais vindos principalmente da Argentina e Uruguai e também de países europeus. Por sua localização e pela necessidade de se ter um espaço apropriado para o comércio, surgiu, nesse mesmo tempo, o Mercado Público de Itaqui. Mercado este que durou algumas décadas, mas foi vencido por diversos fatores. Embora fosse importante, reconhecido como Patrimônio Histórico do Estado, o prédio perdeu seu valor dando espaço para o abandono e desuso. Esta pesquisa, ao levar em consideração esses aspectos, tem a finalidade de dar embasamento teórico para que a área de intervenção readquira seu sentido original, com o objetivo de requalificar o entorno e resgatar sua importância histórica e arquitetônica. Os estudos teóricos e práticos fundamentam e concretizam essas metas, junto a temas relacionados à área. Com isso, através do levantamento fotográfico, métrico e da análise do estado atual de conservação, observou-se que muitas modificações com pouca significância foram feitas ao longo dos anos. Diante destes dados coletados, elaboraram-se os mapas de danos, na qual verifica-se a presença constante de Biofilme, resultado da inexistência de manutenção periódica. Por fim, desenvolveu-se métodos que servirão como base para as diretrizes de reabilitação e conservação de forma a permitir a integridade futura da edificação, perpetuando-a ao longo do tempo.

Our aim was to survey the plants sold for therapeutic purposes in public markets in the area studied, to select the plant species based on ethnopharmacological use and to analyze them for phytochemical composition and antibacterial and antiparasitic activities. Medicinal plant vendors were asked to fill out a questionnaire to determine the relative importance of the species and informant consensus factor. Ninety-one species were indicated as being therapeutically useful; they were distributed in 49 families and 89 genera. Thirteen species showed great versatility in their use. There were 291 indications of use in 13 disease categories. On the basis of these results, the plant species Operculina hamiltonii, Cephaelis ipecacuanha and Lippia gracilis were selected and evaluated as follows, according to their ethnopharmacological use. The infusion of O. hamiltonii (IOh) and decoction of C. ipecacuanha (DCi) were analyzed by HPLC-DAD, while the chemical composition of the essential oil of L. gracilis (OELG) was determined by gas chromatography coupled to mass spectrometry. IOh was tested in vitro for epimastigote and promastigote susceptibility and cytotoxicity. IOh, DCi and OELG were subjected to broth microdilution assays to determine the minimal inhibitory concentration against bacteria; solutions of the natural products were also tested at subinhibitory concentrations to evaluate them for antibiotic resistance-modifying activity. O. hamiltonii showed high leishmanicidal and trypanosomicidal activities, but also substantial cytotoxicity. O. hamiltonii and C. ipecacuanha did not exhibit satisfactory antibacterial activity, but assays for modulatory potential demonstrated interesting results with synergism observed using various combinations of antibiotics and natural products. OELG was able to modulate bacterial resistance and could be used as a coadjuvant therapy against multidrug-resistant microrganisms. / Esse trabalho bioprospectivo com abordagem etnodirigida levou em consideração o conhecimento dos vendedores de plantas medicinais em uma região do Nordeste brasileiro conhecida como Triângulo Crajubar e objetivou realizar um levantamento das plantas comercializadas para fins terapêuticos nos mercados públicos da área estudada, selecionar espécies vegetais a partir do seu uso etnofarmacológico e realizar análises químicas, microbiológicas e parasitológicas. Foram aplicados aos vendedores de plantas nos mercados públicos, formulários que permitiram o cálculo da importância relativa das espécies e do fator de consenso dos informantes. Foram indicadas por estes, 91 espécies úteis terapeuticamente, distribuídas em 49 famílias botânicas e 89 gêneros. Treze espécies apresentaram grande versatilidade quanto ao seu uso. Foram obtidas 291 indicações de uso em 13 categorias de doenças. Com base nesses resultados, foram selecionadas as espécies botânicas Operculina hamiltonii, Cephaelis ipecacuanha e Lippia gracilis, que foram avaliadas nas etapas subsequentes, segundo o seu uso etnofarmacológico. A infusão de Operculina hamiltonii (IOh) e o decocto de Cephaelis ipecacuanha (DCi) tiveram seus compostos fenólicos e flavonóides quantificados por HPLC-DAD enquanto o óleo essencial de Lippia gracilis (OELG) teve a sua composição química analisada por cromatografia gasosa acoplada a espectrofotometria de massas. A IOh foi submetida a ensaios de susceptibilidade epimastigota e promastigota in vitro e ensaios citotóxicos. Para a IOh, o DCi e o OELG foram determinadas as concentrações inibitórias mínimas em ensaio de microdilução em caldo; as soluções dos produtos naturais foram testadas em concentrações subinibitórias para avaliação da atividade moduladora da resistência aos antibióticos. Operculina hamiltonii apresentou uma elevada atividade leishmanicida e tripanossomicida, no entanto com citotoxicidade significativa. Operculina hamiltonii e a Cephaelis ipecacuanha não apresentaram atividade antibacteriana clinicamente satisfatória pelo método estudado, no entanto os testes de avaliação do potencial modulador demonstraram resultados relevantes com sinergismo observado em várias combinações dos antibióticos com os produtos naturais. O óleo essencial de Lippia gracilis teve a capacidade de modular a resistência bacteriana e pode ser utilizado como uma terapia coadjuvante contra microrganismos multirresistentes.

Transformation of Urban Public Space

The concept of my thesis is to employ architectural intervention in residual urban space as a catalyst for transformation. The goal is design of a building and environment that could be used for any combination of purposes, be used freely by all members of the community, be designed so that the art and architecture is interactive, and could be transformed by the users of the space. The project makes use of a residual urban space that would otherwise remain largely inaccessible. The project explores how the space could be designed to give a sense of ownership of it to the community, and how it could be designed to reunify areas of the city that have been severed by urban renewal. The site I have chosen is the Franklin Arterial in Portland, Maine, a four-lane divided surface highway surrounded by parking lots vacant lots, industrial sites and housing projects. Design interventions include a centrally located public market building, which would have a variety of uses throughout any given time period, a bicycle pedestrian path which reconnects the surrounding neighborhoods, and designated sites for art and performance throughout the area.

The Public Market System of New Orleans: Food Deserts, Food Security, and Food Politics

This study evaluates the public market system in New Orleans, Louisiana by focusing on the history of New Orleans public markets, the privatization of food, and the "greening" of the city with the creation of the Crescent City Farmers Market and other grass roots food activist efforts. Using qualitative methods, ethnographic fieldwork, participant observation and interviewing, issues of food access, food security, food production, food locality, quality, and affordability in New Orleans are explored. The history of public markets in New Orleans and the patterns of market proliferation, regulation, and privatization are significant in the landscape of cultural self-identification, community cohesion, neighborhood networks and economic and ecological development and sustainability. The city's various food shopping arenas and their locations become markers of history, status, rebellion, and of the "other," and become centers for issues of health, economy, politics, and food.

Antimicrobial sensibility profile in vitro of Staphylococcus aureus and Salmonella spp samples and the correlation between the simultaneous presence of faecal coliforms and Salmonella spp. were also performed. The study was carried out at six public/private markets and nine supermarkets of Recife, which were selected according to the administrative division of Local Authority Healthy General Office in six sanitary districts. A total of 24 samples of goat meat, 14 proceeded from public/private markets in natura and 10 chilled ones from supermarkets. Samples were processed using methodologies recommended by the Agriculture, Pecuary and Provision Ministry. Results demonstrated that 11 samples (45,83%) presented positive coagulasis staphylococcus (SCP) varying from 6.7 x 103 to 1.4 x 106 UFC/g, corresponding to 27 isolated from a total of 89. In total coliforms analyses, five samples (20.83%) presented satisfactory sanitary conditions and 19 (79.16%) had countings varying from 4.3 x 103 to 3.7 x 107, thermotolerants coliforms were detected in 15 (62.5%), which countings varied from 2 x 104 to 2.5 x 107 UFC/g and nine samples (37.5%) presented satisfactory sanitary conditions. Presence of Salmonella spp., was confirmed at seven samples (29.17%) in which eight sorovares were typed: four (50%) Salmonella enterica subsp houtenae (O:53), two (25%) S. Anatum, one (12.50%) S.Rubislaw and one (12, 50%) S. Derby. No correlation was observed among coliforms and Salmonella presure. Antimicrobial profile sensibility showed that S. aureus were sensitive to norfloxacin and vancomicin (100%), tetraciclin and sulpha + trimetoprim (96.30%) and oxacilin (87.50%) and 100 % of Salmonella spp. samples were sensitive norfloxacin, 75% sulpha + trimetoprim and 62.5% tetraciclin. It is concluded that goat meat sold at markets and supermarkets of Recife presented undesirable microbiological patterns emphasizing the importance of a strong supervision at the productive goat meat chain as well as a revision of the Resolution of Collective Management from the National Agency of Sanitary Vigilance, (RDC, n°12/2001) which does not establish microbiological patterns for the coliforms and Staphylococcus spp countings in chilled and in natura goat meat. / Objetivou-se com este estudo avaliar a qualidade microbiológica da carne caprina in natura e resfriada, comercializada em mercados públicos/privados e supermercados na Cidade do Recife, quanto a contagem de Staphylococcus aureus, coliformes totais e termotolerantes, e identificação e tipificação de Salmonella spp. Realizou-se, também, o perfil de sensibilidade antimicrobiana in vitro de amostras de Staphylococcus aureus e Salmonella spp e a correlação entre a presença simultânea de coliformes fecais e Salmonella spp. O estudo foi realizado em seis mercados públicos/privados e nove supermercados da Cidade do Recife, selecionados seguindo a divisão administrativa da Secretaria de Saúde do Município em seis Distritos Sanitários. Foram analisadas 24 amostras de carne caprina, sendo 14 in natura procedentes de mercados públicos/privados e 10 resfriadas de supermercados. As amostras foram processadas utilizando metodologias preconizadas pelo Ministério da Agricultura, Pecuária e Abastecimento. Os resultados demonstraram que 11 (45,83%) amostras apresentaram contagens de estafilococos coagulase positiva (SCP) variando de 6,7 x 103 a 1,4 x 106 UFC/g, correspondendo a 27 isolados de um total de 89. Para coliformes totais, cinco (20,83%) amostras encontravam-se em condições sanitárias satisfatórias e 19 (79,16%) apresentaram contagens variando de 4,3 x 103 a 3,7 x 107; e para termotolerantes 15 (62,5%) apresentaram contagens variando de 1,2 x 104 a 2,5 x 107 UFC/g e nove (37,5%) encontravam-se em condições sanitárias satisfatórias. Foram confirmadas em sete (29,17%) amostras a presença de Salmonella spp., nas quais foram tipificadas oito sorovares: quatro (50,00%) Salmonella enterica subsp houtenae (O:53), duas (25,00%) S. Anatum, uma (12,50%) S.Rubislaw, uma (12,50%) S. Derby. Não se observou correlação entre as bactérias estudadas. O perfil de sensibilidade antimicrobiana demonstrou que os antibióticos mais indicados para o tratamento de S. aureus foram norfloxacina e vancomicina (100%), tetraciclina e sulfa + trimetoprim (96,30%) e oxacilina (87,50%) e para Salmonella spp. 100% das amostras foram sensíveis à norfloxacina, 75,00% à sulfa + trimetoprim e 62,5% para tetraciclina. Conclui-se que a carne caprina comercializada nos mercados e supermercados da Cidade do Recife apresentou padrões microbiológicos indesejáveis ressaltando a importância da necessidade de uma maior fiscalização na cadeia produtiva da carne caprina bem como rever a Resolução da Diretoria Colegiada da Agência Nacional de Vigilância Sanitária (RDC, n°12/2001) que não estabelece padrões microbiológicas para a contagem de coliformes termotolerantes e Staphylococcus spp. em carne caprina in natura e resfriada.

The master´s thesis deals with the influence of non-residential spaces of medical attendance on the price of real estate. There are discussed some basic concepts, which are necessary for understanding, and commonly used methods for valuing property according to the purpose of valuation in the theoretical part. There are analysed these methods for particular houses in the practical part – method of valuation, comparative public notice, revenues method, annuity bond. There is investigated the role of non-residential spaces in these methods. The influence of non-residential space on price of real estate is compared by each method in conclusion.
