Hipertensão arterial em SHR : modificações homeodinamicas da inervação renal

Dentre os mecanismos renais envolvidos no desenvolvimento pressórico são descritos àqueles relacionados a alterações hemodinâmicas e na manipulação tubular renal hidro-eletrolítica. No estudo fisiopatológico da hipertensão arterial primária humana têm sido utilizados modelos experimentais análogos dentre os quais destaca-se a linhagem geneticamente hipertensa de Kyoto (SHR). A gênese da hipertensão arterial nestes animais ainda não está inteiramente entendida. Tem sido observado nestes animais um envolvimento etiopatogênico mutifatorial dentre os quais destacam-se as modificações na atividade neural central e periférica,distúrbios neuro-endócrinos. e eletrolíticos, acentuada atividade mitogênica da musculatura vascular e significativas alterações funcionais nos rins. Algumas destas alterações antecedem a elevação pressórica. Sabendo-se que a inervação simpática eferente e sensorial aferente renal participam da modulação hemodinâmica, da secreção de renina e do controle da excreção urinária de sódio, o presente trabalho teve como objetivo geral investigar a participação neural (aferente e eferente) renal nestes aspectos etiopatogênicos da hipertensão arterial em SHR. Especificamente, para avaliar a contribuição da atividade nervosa renal sobre a excreção urinária de sódio, investigamos a manipulação tubular deste íon em diferentes segmentos do néfron (proximal e pós-proximal) em SHR antes e após a elevação pressórica (4 e 12 semanas) comparando os resultados a ratos Wistar Kyoto (WKy). Nossos resultados demonstraram elevada reabsorção tubular (proximal e pós-proximal) de sódio em SHR acompanhada de significativa redução na natriurese. Esta menor excreção renal de sódio foi atenuada pela denervação renal bilateral. Estes resultados sugerem aumento da atividade simpática neural associada ou não a menor atividade aferente sensorial (ARNA) neste modelo. Estudos prévios indicam que ratos espontaneamente hipertensos apresentam diminuição da ARNA Uma vez que a ARNA modula a atividade simpática eferente renal (ERSNA) e, conseqüentemente a natriurese (reflexo renorenal) e, que a resposta aferente renal a estímulos mecano- (MR) e quimioreceptores (CR) e a SP está deprimida em SHR adultos, avaliamos se existe modificações na expressão e localização de neuropeptídeos envolvidos na mediação MR e CR, no gânglio da raiz dorsal (GRD) e na pelve renal (PR), destes animais. O presente estudo avaliou em ORD e na PR a expressão e distribuição sub-celular de CGRP, SP e seu receptor 'NK IND. 1R' (por western blot, microscopia confocal a laser e eletrônica de transmissão) em SHR (de 7 e 14 semanas) e em WKy. Nossos resultados mostram, pela primeira vez, a localizaçao nuclear de CORP, SP e seu receptor 'NK IND. 1R' ('NK IND. 1R')em neurônios de GRD. Os resuhados também indicam em WKy, a presença de 'NK IND. 1R' associados à heterocromatina em 30% das sub-populações de neurônios presentes nos ORD. Adicionalmente, a presença nuclear de 'NK IND. 1R' foi também evidenciada em células de Schwann e satélites. A transposição nuclear e os sítios aceptores destes neuropeptídeos junto à heterocromatina ainda deverão ser futuramente investigados. Em SHR, observamos aumento na expressão nuclear de CGRP e SP, associado a menor expressão de NK}. Tal redução de 'NK IND. 1R' ocorreu na superficie de neurônios do ORD e na pelve renal. O presente estudo indica que alterações na expressão e localização de neuropeptídeos e 'NK IND. 1R', podem estar relacionados à atenuada ARNA com conseqüente elevação da ERSNA, retenção renal de sódio e hipertensão arterial em SHR / Abstract: The kidneys play a pivotal role in the pathogenesis of essential hypertension by a primary defect in renal hemodynamics and/or tubule hydro-saline handling that resuhs in the retention of fluid and electrolytes. In the physiopathological studies of human essential hypertension similar experimental models have been used including spontaneously hypertensive rats (SHR). In this lineage the genesis of hypertension is not totally understand. In this animal have been observed multifactorial etiopathogenesis that include changes at central and peripheral neural activity, neuroendocrine and electrolytic alterations, rise in the mitogenic activity muscle vascular cells and kidney functional alterations. Some that alterations, preceding the pressoric rise. The efferent renal sympathetic neural activity (ERSNA) and the sensorial afferent renal neural activity (ARNA) are involved in the homeodynamic modulation, renin secretion and in the controlof urinary sodium excretion, thus, this work has a general objective of investigate the neural (afferent and efferent) renal participation in that ethiopathogenic aspects of arterial hypertension in SHR. Specifically, to analyze the renal nervous activity contribution in the sodium urinary excretion, we investigate the manipulation of this ion at the different nephon segments (proximal and post-proximal) in SHR before and afier arterial blood pressor rise (4 and 7 weeks) compared to resuhs obtained to Wistar Kyoto (WKy) rats. Our results show an increased tubular (proximal and post-proximal) sodium reabsorption in SHR accompanied to significant reduction in natriuresis. This decreased sodium renal excretion was attenuated by bilateral renal denervation. These results suggests enhancement in the neural sympathetic activity associated or not, to the lesser ARNA in this genetic hypertensive model. Previous studies indicated that SHR presents reduced ARNA. The ARNA modulate ERSNA and, consequently the natriuresis (renorenal reflexes) and the afferent renal responses to mechano- (MR) and chemoreceptors (CR) and to Substance P (SP) are depressed in adult SHR, thus we evaluated the existence of modifications in the expression and localization of neuropeptides involved in the mediation of MR. and CR, in the dorsal root ganglia (DRG) and renal pelvis (RP), on these animals.The present study verified in DRG ('T IND. 10' to 'L IND. 1') and in RP the subcellular expression and distribution of CGRP, SP and 'NK IND. 1R' receptor (by western blot, confocallaser scanning and transmission microscopy) in SHR (7 and 14-old-weeks) compared to WKY. Our results show, by the first time, the nuclear location of CGRP, SP and 'NK IND. 1R' receptors ('NK IND. 1R') in DRG neurons. The results a1so indicate in WKY, the occurrence of 'NK IND. 1R' associated to heterochromatin in neurons sub-populations of DRG. Additionally, the nuclear presence of 'NK IND. 1R' was also showed in satellite and Schwann cells. The nuclear translocation and acceptors sites at the chromatin will be investigated in the future. In SHR we observed elevated nuclear expression of CGRP, SP e NKIR, associated to minor cellular membrane expression of 'NK IND. 1R' This reduction was also observed in the RP. Em aves, os tipos de neurônios quimiossensíveis intermediadores da resposta ventilatória não são conhecidos, assim como sua localização. Por outro lado, em mamíferos, sabe-se que estes quimiorreceptores compreendem uma grande população neuronal e estão distribuídos no sistema nervoso central. Dentre os tipos de quimiorreceptores centrais, em mamíferos, destacam-se os neurônios serotoninérgicos (5-HT) contidos em elevada concentração na rafe e que possuem projeções às principais regiões envolvidas na regulação da respiração. A partir do exposto, objetivou-se verificar a participação da serotonina no controle respiratório de frangos. Foram utilizados pintos carijós machos e fêmeas, em dois conjuntos experimentos com abordagem distintas que propuseram reduzir o conteúdo de serotonina liberado no espaço sináptico em resposta à hipercapnia e à hipóxia. O primeiro com deleção química dos neurônios 5-HT, mediante a injeção de 1µL de anti-SERT-SAP (0,1µM) no quarto ventrículo (4V), os grupos controle receberam injeções de PBS e IgG-SAP. O segundo experimento por inibição da biossíntese de serotonina pela p-clorofenilalanina (PCPA) em 100mg/kg i.p. (animais controles receberam salina 0,9% ou solução veículo). In birds, the types of chemosensitive neurons mediating the ventilatory response are not known, as is their location. On the other hand, in mammals, it is known that these chemoreceptors comprise a large neuronal population and are distributed in the central nervous system. Among the types of central chemoreceptors in mammals, serotoninergic (5-HT) neurons are found in high concentration in the raphe and have projections to the main regions involved in the regulation of respiration. From the above, we aimed to verify the participation of serotonin in respiratory control of broilers. Male and female carybush chicks were used in two sets with distinct approaches that proposed to reduce the serotonin content released in the synaptic space in response to hypercapnia and hypoxia. The first with chemical deletion of 5-HT neurons, by injecting 1μL of anti-SERT-SAP (0.1μM) into the fourth ventricle (4V), the control groups received injections of PBS and IgG-SAP. The second experiment by inhibition of serotonin biosynthesis by p-chlorophenylalanine (PCPA) at 100mg/ kg i.p. (control animals received 0.9% saline or vehicle solution). As estruturas de quimiorrecepção e mecanorrecepção são reconhecidas como os principais sentidos usados pelos crustáceos decápodas para identificação de partículas alimentares. O Macrobrachium rosenbergii é uma espécie que passa por uma fase planctônica e outra bentônica, faz grandes migrações a favor ou contra a corrente conforme a fase do ciclo de vida ou estágio fisiológico e muda o hábito alimentar de carnívoro à onívoro. Portanto, deve apresentar mecanismos de percepção dos estímulos do meio, que se modificam ao longo do desenvolvimento. Assim, a hipótese levantada nessa pesquisa foi que M. rosenbergii apresenta estruturas sensitivas na superfície do corpo e apêndices, que se modificam desde a eclosão da larva até a fase adulta. O objetivo deste trabalho foi pesquisar a ocorrência de estruturas sensitivas ao longo do desenvolvimento ontogenético de M. rosenbergii. A pesquisa ocorreu no setor de carcinicultura do CAUNESP. As larvas e pós-larvas foram coletadas de larvicultura sob sistema fechado dinâmico segundo Valenti (1998) e os juvenis e adultos dos sistemas de cultivo do setor. Os animais foram fixados com Karnovsky e dissecados. De cada animal, retiraram-se as antênulas, as antenas, as maxilas, as placas mandibulares, os três maxilípedes e os olhos para possibilitar a observação dos apêndices e as estruturas sensitivas. Os apêndices e olhos foram metalizados e fotodocumentados em microscópio eletrônico de varredura. Os olhos também foram analisados com técnicas de microscopia eletrônica de transmissão e técnica de rotina para análises histológicas segundo Behmer (2003). Chemoreceprion and mechanoreption are known as the main senses used by the decapod crustaceans to indentify food particles. The Macrobrachium rosenbergii is a species that goes through a planktonic and a benthonic phase performs great migrations against or within the current depending on the life cycle phase or physiological stage and changes its feeding haits from carnivorous to omnivorous. Thus, it should present perception mechanisms of environment stimuli that are modified during development. Therefore, the hypotheisis brought up in this research was that the M. rosenbergii presents sensitive structures on the surface of the body and appendices that change from since the hatching of the larva up to the adult phase. The object of this paper was to research the occurrence of the sensitive structures during the entogenetic development of the M. rosenbergii. The research was performed at the carciniculture sector of the CAUNESP. The larvae and post larvae were collected from larvae culture under a closed dynamic system according to Valenti (1998) and the yong and adult larvae from the cultivating systems of the sector. The animals were set with Karnovsky and dissected. From each animal the antennules, the antennas, the maxillas, the mouthpiece plates, the three maxilipedes and the sensitve structures. A dispnéia na insuficiência cardíaca(IC) é complexa, com possível envolvimento de quimioreceptores periféricos(QP) e centrais(QC). Avaliamos a resposta de QP e QC no exercício submáximo em 15 pcts com IC e 7 ind. normais em testes ergoespirométricos de caminhada de 6 min: hipóxia isocápnica(HPX),hipercapnia hiperóxica(HPC) e ar ambiente. HPX aumentou ventilação (VE) com resposta aguda(RVA), freq. cardíaca(FC) e volume de O2 consumido;reduziu o espaço morto,distância caminhada(DC) e pressão arterial sistêmica(PAS). A HPC aumentou VE acima da HPX com RVA.Os QP têm ação maior sobre FC e PAS do que QC, apesar da maior ativação simpática.QP possuem estimulo rápido sobre VE,porém menor do que QC. / Heart failure(HF) dyspnea is complex with potential enrolment of central(CC) and peripheric chemoreceptors(PC).We investigated CC and CP behavior through submaximal exercise in 15 HF patients and 7 normal subjects in treadmill 6-minute cardiopulmonary walking tests:isocapnic hypoxia(HPO), hypercapnia hyperoxic(HCP) and room air.HPO increased:ventilation(VE) with acute ventilatory response(AVR), heart rate (HR) and O2 uptake and reduced dead space, distance walked (DW) and systemic blood pressure(SBP).The HPC improved VE above HPO level with AVR. PC have greater action on HR and SBP than CC,despite their largest sympathetic activation. PC have faster impulse on VE although be lowest than CC

Estruturas sensoriais tarsais de opiliões (Arachnida, Opiliones): morfologia funcional, evolução e uso em sistemática / Tarsal sensory structures in harvestmen (Arachnida, Opiliones): functional morphology, evolution and their use in systematics

Opiliões (Arachnida, Opiliones) são especialmente dependentes da quimiorecepção de contato, além de serem muito dependentes de altos níveis de umidade e de temperaturas amenas. No entanto, o conhecimento acerca das estruturas que detectam esses estímulos é muito limitado em opiliões, quando comparado ao que se sabe sobre outros aracnídeos. Além disso, são raros os estudos investigando a morfologia interna dessas estruturas - um tipo de informação essencial para a determinação de suas funções- e o potencial das sensilla para a sistemática. Neste trabalho, nós investigamos as sensilla tarsais de todos os pares de perna do opilião Heteromitobates discolor (Laniatores, Gonyleptidae), buscando refinar o conhecimento das sensilla quimioreceptoras e investigar a localização dos receptores de temperatura e umidade, através de microscopia eletrônica de varredura (MEV) e transmissão. Para determinar a abrangência dos resultados obtidos com H. discolor e testar o uso de algumas estruturas para a sistemática do grupo, realizamos uma amostragem com MEV em espécies das quatro subordens de Opiliones (Cyphophthalmi, Eupnoi, Dyspnoi e Laniatores), com foco em Laniatores (subordem com 2/3 das espécies do grupo). Na primeira parte, fornecemos a primeira evidência morfológica de receptores olfativos em Laniatores (em H. discolor), mostrando que há abundância e diversidade de sensilla olfativas. Além disso, fornecemos evidência (com MEV) de cerdas olfativas em 17 famílias de Laniatores, o que sugere que a olfação é mais importante para os Laniatores do que previamente considerado. Na segunda parte, fornecemos a primeira evidência morfológica de detectores de umidade e temperatura em opiliões (em H. discolor), discutindo os mecanismos de funcionamento dessas estruturas e uma associação morfológica entre elas, localizada na parte mais distal das pernas I e II. Por fim, mostramos que essas sensilla candidatas a detectores de umidade e temperatura são extremamente conservadas em Laniatores (28 famílias), e que existem estruturas comparáveis em espécies de Cyphophtalmi, Eupnoi e Dyspnoi. Com uma análise de reconstrução de estado ancestral em uma filogenia de Opiliones compilada da literatura, mostramos que as variações na morfologia externa dessas estruturas fornecem informação filogenética em vários níveis de relacionamento em Opiliones. Esse estudo contribui para o conhecimento de aspectos básicos da anatomia celular de sensilla em Opiliones, refinando o conhecimento sobre a função das sensilla tarsais e forncendo uma base para fomentar o uso de sensilla para a sistemática do grupo / Harvestmen (Arachnida, Opiliones) are especially dependent on contact chemoreception and are dependent on high humidity levels and amenable temperatures. However, knowledge on the sensory structures (sensilla) that detect such stimuli is limited in harvestmen when compared with other arachnid orders. Besides, there are few studies investigating the internal morphology of these structures -which is important for inferring function - and the potential of sensilla for systematics. To refine the knowledge on chemoreceptive sensilla and investigate the identity of hygro- and thermoreceptors, we investigated the tarsal sensilla of all leg pairs of the species Heteromitobates discolor (Laniatores, Gonyleptidae), using scanning electron microscopy (SEM) and transmission electron microscopy. To determine scope of the results obtained with H. discolor and to test the use of some structures for systematics, we surveyed (with SEM) species in all four suborders of Opiliones (Cyphophthalmi, Eupnoi, Dyspnoi and Laniatores), with focus in Laniatores (suborder with 2/3 of harvestmen species). In the first part, we provide the first morphological evidence of olfactory receptors in a species of Laniatores (H. discolor), showing that olfactory sensilla are abundant and diverse. Also, we show evidence (with SEM) of olfactory sensilla on 17 families of Laniatores, which suggests that olfaction is more important for Laniatores than previously considered. In the second part, we provide the first morphological evidence of hygro- and thermoreceptors in harvestmen (in H. discolor) discussing the functioning mechanisms of these structures and their morphological inter-association on the distal-most part of leg pairs I and II. Finally, we show that these putative hygro- thermoreceptive sensilla are widespread in species of Laniatores (28 families) and that comparable structures occur on species of the suborders Cyphophthalmi, Eupnoi and Dyspnoi. With an ancestral state reconstruction on a compiled phylogeny of Opiliones, we show that morphological variations on these sensilla are informative on several levels of phylogenetic relationships in Opiliones. This study constributes for the basic knowledge on the cellular anatomy of Opiliones sensilla, refining the function of tarsal sensillar types and providing a base for their use in systematics

No presente trabalho, avaliamos o envolvimento da região Kölliker Fuse (KF) e núcleo parabraquial lateral (NPBL) nas respostas cardiorrespiratórias induzidas pela ativação dos quimiorreceptores centrais e periféricos em ratos não anestesiados. A injeção bilateral de muscimol (200 pmol/100 nl) no KF reduziu a ventilação basal (978 ± 100, vs. salina: 1436 ± 155 ml/kg/min). Injeção de muscimol no KF reduziu a hiperventilação (1827 ± 61, vs. salina: 3179 ± 325 ml/kg/min) e a taquicardia (380 ± 9, vs. salina: 423 ± 12 bpm), produzidos pela hipóxia (8% O2 - 10 min). Muscimol no KF reduziu a hiperventilação (1488 ± 277, vs. salina: 3539 ± 374 ml/kg/min) produzida por hipercapnia (7% CO2 - 10 min). Injeção de muscimol no NPBL promoveu um aumento de PAM (D = 119 ± 2, vs. salina: 104 ± 2 mmHg), mas não foi capaz de alterar a hiperventilação produzida por hipóxia e hipercapnia. Nossos experimentos mostram a participação da região KF, e não do NPBL, no controle do respiratório durante a ativação do quimiorreflexo central e periférico. / Here we evaluated the involvement of Kölliker-Fuse region (KF) and lateral parabrachial nucleus (LPBN) in the cardiorespiratory responses elicited by chemoreceptor activation in conscious rats. Bilateral injection of muscimol (200 pmol/100 nl) into the KF decreased resting ventilation (978 ± 100, vs. saline: 1436 ± 155 ml/kg/min). No. of bitstreams: 1 DoutCLB.pdf: 812696 bytes, checksum: b0eb5a5003d3b08cb3457ce3764679d6 (MD5) Previous issue date: 2004-01-03 / Universidade Federal de Minas Gerais / In the present study we determined the location and orientation of the CO2 and/or pH chemoreceptors and analyzed the cardio-respiratory responses and the aerial respiration in jeju (Hoplerythrinus unitaeniatus) during graded hypercarbia (1, 2.5, 5, 10 e 20% de CO2) and acid medium (pH ~ 7, 6.0, 5.8, 5.6, 5.3 e 4.7) (1, 2, 3, 4 e 5 mM de HCl). The chemoreceptors orientation was determined by means of a series of injections of different hypercarbic solutions (5, 10 e 20% de CO2) in the caudal vein (internal) and buccal cavity (external). To distinguish the orientation of the CO2 and H+ chemoreceptors, acid solutions (HCl) at same pH of the internal (pH 5.1, 4.9, and 3.7) and external (pH 5.6, 5.3, and 4.7) solutions were applied, and significant responses in relation to the control values were not recorded. In another group of fish previously tested acid solutions (30, 40, and 50 mM HCl) were injected. This injections caused a decrease in the extra-cellular blood pH to levels similar to those found during the injections of internal and external hypercarbic solutions. The location of the chemoreceptors was determined by bilateral denervation of branches of the cranial nerves IX (glossopharyngeal) and X (vagus) to the gills. The heart rate, arterial blood pressure, respiratory frequency and ventilation amplitude were recorded for intact and denervated fish. The data indicated that the CO2/pH chemoreceptors eliciting the cardio-respiratory responses to hypercarbia are exclusively branchial and are externally oriented. Jeju respond to hypercarbia of 5% CO2 with bradycardia, hypertension and increases in the ventilatory frequency and ventilation amplitude. Concentrations higher than 5% caused anesthetic effect. The cardiac responses are mediated by CO2/pH chemoreceptors distributed in all gill arches, while the respiratory responses are mediated by chemoreceptors located in the 1st gill arch. Furthermore, hypercarbia also induced aerial respiration in intact fish and in those with the 1st gill arch denervated. The denervation of the four gill arches did not abolish completely the aerial respiration, suggesting that chemoreceptors involved in this response are located in extra-branchial sites. The changes in the internal and external H+ concentrations or in the water did not affect the cardiorespiratory responses and aerial respiration frequency, unless if accompanied by hypercarbia. / O presente estudo determinou a localização e orientação dos quimiorreceptores de CO2 e/ou pH, analisou as respostas cardio-respiratórias e a freqüência de respiração aérea em jeju (Hoplerythrinus unitaeniatus) durante hipercarbia gradual (1, 2.5, 5, 10 e 20% de CO2) e meio ácido (pH ~ 7, 6.0, 5.8, 5.6, 5.3 e 4.7) (1, 2, 3, 4 e 5 mM de HCl). A orientação dos quimiorreceptores foi determinada pela aplicação de uma série de injeções com diferentes soluções hipercárbicas (5, 10 e 20% de CO2) na veia caudal (interna) e na cavidade bucal (externa). Para distinguir a orientação dos quimiorreceptores de CO2 e do H+, foram aplicadas soluções ácidas (HCl) de mesmo pH encontrado nas soluções hipercárbicas internas (pH 5.1, 4.9 e 3.7) e externas (pH 5.6, 5.3 e 4.7), não havendo resposta significativa em relação à injeção controle. Em outro grupo de animais foram injetadas soluções ácidas (30, 40 e 50 mM de HCl), previamente testadas, que diminuíram o pH extracelular sanguíneo a valores aproximados aos encontrados durante a aplicação das soluções hipercárbicas internas e externas. A localização foi determinada por denervações bilaterais do IX (glossofaríngeo) e X (vago) nervos cranianos dos arcos branquiais. Durante estes procedimentos foram registradas a freqüência cardíaca, pressão arterial, freqüência e amplitude ventilatória. Os resultados indicaram que a localização dos quimiorreceptores de CO2/pH no jeju que controlam as respostas cardio-respiratórias são exclusivamente branquiais e os receptores estão orientados externamente. O jeju responde a hipercarbia de 5% de CO2 com uma bradicardia, hipertensão e aumento na freqüência e na amplitude ventilatória; e concentrações acima de 5% apresentam um efeito anestésico. As respostas cardíacas são mediadas por quimiorreceptores de CO2/pH localizados nas brânquias, tanto no 1º arco branquial quanto nos demais e as respostas respiratórias são mediadas preferencialmente por quimiorreceptores localizados no 1º arco branquial. Além disso, a hipercarbia também induz a respiração aérea em animais intactos e com o 1º arco branquial denervado. A denervação dos 4 arcos branquiais não aboliu completamente a respiração aérea, o que sugere que os quimiorreceptores envolvidos nesta resposta estão localizados em sítios extrabranquiais. As vii mudanças nas concentrações externas e internas de H+ ou no meio experimental não afetam as respostas cardio-respiratórias e freqüência de respiração aérea a menos que acompanhado pela hipercarbia.

Caracterização dos quimiorreceptores de O2 envolvidos no controle dos reflexos cardio-respiratórios de trairão, Hoplias lacerdae (Teleostei, Erythrinidae) em resposta à hipóxia ambiental

This is even more so in tropical freshwater habitats where environmental hypoxia can occur rapidly in space and time. Recent physiological studies on the cardio-respiratory response of tropical fishes have focused on hypoxia tolerant species. This aim of this study was to examine the neural control and function of the cardio-respiratory system in a fish that is adapted to living at high levels of oxygen. Trairão, Hoplias lacerdae was chosen because it is a member of the Erythrinidae family, in which most of species are hypoxia tolerant, including some where the O2 receptors has already been characterized. The study focused on investigation of the localization, distribution and orientation of chemoreceptors of O2 in trairão. Also how the cardio-respiratory parameters were modified by hypoxia. The fish were submitted to selective denervations of the branchial branches of the cranial nerves IX and X. These were, (1) only IX nerve sectioned at the 1st gill arch (IX; n = 9), (2) first gill arch both IX and branch of X sectioned (G1; n = 9), (3) all four gill arches both IX and X sectioned (G4; n = 8), (4) a control group (n = 8) and (5) a sham-operated group that underwent surgery but no nerves were sectioned (n = 4). A cannula was inserted into the ventral aorta to monitor changes in heart rate and blood pressure. Ventilation rate and amplitude was also monitored with a cannula through the roof of the buccal cavity. The fish were exposed to a gradual hypoxia from 140 to 10 mmHg. Either the internal and external surfaces of the gill were treated with NaCN to determine the receptors orientation. Results showed that the receptors related to the heart rate control and ventilation amplitude were distributed over all gill arches on both the internal and external surfaces, and the branches of the both nerves IX and X were involved in these responses. This pattern differs from that observed for the ventilation rat. The receptors involved in the control of this response are located internally to the first gill arch. Tairão also showed blood pressure control chemorecptors on the first gill arch, involved in a very efficient barostatic control. Extrabranchial O2 chemoreceptors were not identified in trairão. This differs from the hypoxia tolerant fish, where extrabranchial chemoreceptors are important to start the cardio-respiratory responses the most fast and accurate possible / A capacidade de sentir e responder às variações de oxigênio no ambiente é de extrema importância para peixes. Isto é mais acentuado em ambientes dulceaquícolas tropicais onde a hipóxia ambiental ocorre freqüentemente. Estudos recentes sobre respostas cardio-respiratórias em peixes tropicais têm enfocado espécies tolerantes à hipóxia. O foco do presente estudo foi examinar o controle neural e a função cardio-respiratória de um peixe adaptado a ambientes bem oxigenados. O trairão, Hoplias lacerdae foi escolhido por pertencer à família Erythrinidae, que apresenta a maioria das espécies tolerantes à hipóxia, incluindo algumas onde os receptores de O2 já foram caracterizados. O foco do estudo foi investigar a localização, distribuição e orientação dos quimiorreceptores de O2 em trairão, bem como os parâmetros cardio-respiratórios são modificados pela hipóxia. Os peixes foram submetidos à denervações seletivas dos ramos branquiais dos nervos cranianos IX e X. Os grupos foram: (1) apenas nervo IX seccionado do 1o arco branquial (IX; n = 9), (2) primeiro arco, ambos IX e ramo do X seccionados (G1; n = 9), (3) os quatro arcos, ambos IX e X seccionados (G4; n = 8), (4) grupo controle (n = 8) e (5) grupo sham-operated , que passou por cirurgia, mas não teve os nervos seccionados (n = 4). Uma cânula foi inserida na aorta ventral para monitorar freqüência cardíaca e pressão arterial. A freqüência e a amplitude ventilatórias foram monitoradas por uma cânula no palato da cavidade bucal. Os peixes foram expostos à hipóxia gradual de 140 a 10 mmHg. Ambas as superfícies, interna e externa, das brânquias receberam tratamento de NaCN para determinar a orientação dos receptores. Os resultados mostraram que receptores relacionados ao controle da freqüência cardíaca e amplitude ventilatória estão distribuídos por todos os arcos branquiais, orientados tanto interna quanto externamente, e ambos os nervos IX e X estão envolvidos nesta resposta. Este padrão difere do observado para a freqüência respiratória. Os receptores envolvidos no controle dessa resposta localizam-se internamente no 1o arco branquial. O trairão também possiu quimiorreceptores de O2 envolvidos no controle da pressão arterial apenas no primeiro arco branquial, participando de um controle barostático eficiente. Não foram identificados receptores de O2 extrabranquiais em trairão. Este padrão difere da maioria dos peixes tolerantes à hipóxia onde os estes receptores são importantes para iniciar rapidamente as respostas cardio-respiratórias à hipóxia

Estruturas sensoriais tarsais de opiliões (Arachnida, Opiliones): morfologia funcional, evolução e uso em sistemática / Tarsal sensory structures in harvestmen (Arachnida, Opiliones): functional morphology, evolution and their use in systematics

Opiliões (Arachnida, Opiliones) são especialmente dependentes da quimiorecepção de contato, além de serem muito dependentes de altos níveis de umidade e de temperaturas amenas. No entanto, o conhecimento acerca das estruturas que detectam esses estímulos é muito limitado em opiliões, quando comparado ao que se sabe sobre outros aracnídeos. Além disso, são raros os estudos investigando a morfologia interna dessas estruturas - um tipo de informação essencial para a determinação de suas funções- e o potencial das sensilla para a sistemática. Neste trabalho, nós investigamos as sensilla tarsais de todos os pares de perna do opilião Heteromitobates discolor (Laniatores, Gonyleptidae), buscando refinar o conhecimento das sensilla quimioreceptoras e investigar a localização dos receptores de temperatura e umidade, através de microscopia eletrônica de varredura (MEV) e transmissão. Para determinar a abrangência dos resultados obtidos com H. discolor e testar o uso de algumas estruturas para a sistemática do grupo, realizamos uma amostragem com MEV em espécies das quatro subordens de Opiliones (Cyphophthalmi, Eupnoi, Dyspnoi e Laniatores), com foco em Laniatores (subordem com 2/3 das espécies do grupo). Na primeira parte, fornecemos a primeira evidência morfológica de receptores olfativos em Laniatores (em H. discolor), mostrando que há abundância e diversidade de sensilla olfativas. Além disso, fornecemos evidência (com MEV) de cerdas olfativas em 17 famílias de Laniatores, o que sugere que a olfação é mais importante para os Laniatores do que previamente considerado. Na segunda parte, fornecemos a primeira evidência morfológica de detectores de umidade e temperatura em opiliões (em H. discolor), discutindo os mecanismos de funcionamento dessas estruturas e uma associação morfológica entre elas, localizada na parte mais distal das pernas I e II. Por fim, mostramos que essas sensilla candidatas a detectores de umidade e temperatura são extremamente conservadas em Laniatores (28 famílias), e que existem estruturas comparáveis em espécies de Cyphophtalmi, Eupnoi e Dyspnoi. Com uma análise de reconstrução de estado ancestral em uma filogenia de Opiliones compilada da literatura, mostramos que as variações na morfologia externa dessas estruturas fornecem informação filogenética em vários níveis de relacionamento em Opiliones. Esse estudo contribui para o conhecimento de aspectos básicos da anatomia celular de sensilla em Opiliones, refinando o conhecimento sobre a função das sensilla tarsais e forncendo uma base para fomentar o uso de sensilla para a sistemática do grupo / Harvestmen (Arachnida, Opiliones) are especially dependent on contact chemoreception and are dependent on high humidity levels and amenable temperatures. However, knowledge on the sensory structures (sensilla) that detect such stimuli is limited in harvestmen when compared with other arachnid orders. Besides, there are few studies investigating the internal morphology of these structures -which is important for inferring function - and the potential of sensilla for systematics. To refine the knowledge on chemoreceptive sensilla and investigate the identity of hygro- and thermoreceptors, we investigated the tarsal sensilla of all leg pairs of the species Heteromitobates discolor (Laniatores, Gonyleptidae), using scanning electron microscopy (SEM) and transmission electron microscopy. To determine scope of the results obtained with H. discolor and to test the use of some structures for systematics, we surveyed (with SEM) species in all four suborders of Opiliones (Cyphophthalmi, Eupnoi, Dyspnoi and Laniatores), with focus in Laniatores (suborder with 2/3 of harvestmen species). In the first part, we provide the first morphological evidence of olfactory receptors in a species of Laniatores (H. discolor), showing that olfactory sensilla are abundant and diverse. Also, we show evidence (with SEM) of olfactory sensilla on 17 families of Laniatores, which suggests that olfaction is more important for Laniatores than previously considered. In the second part, we provide the first morphological evidence of hygro- and thermoreceptors in harvestmen (in H. discolor) discussing the functioning mechanisms of these structures and their morphological inter-association on the distal-most part of leg pairs I and II. Finally, we show that these putative hygro- thermoreceptive sensilla are widespread in species of Laniatores (28 families) and that comparable structures occur on species of the suborders Cyphophthalmi, Eupnoi and Dyspnoi. With an ancestral state reconstruction on a compiled phylogeny of Opiliones, we show that morphological variations on these sensilla are informative on several levels of phylogenetic relationships in Opiliones. This study constributes for the basic knowledge on the cellular anatomy of Opiliones sensilla, refining the function of tarsal sensillar types and providing a base for their use in systematics

No presente trabalho, avaliamos o envolvimento da região Kölliker Fuse (KF) e núcleo parabraquial lateral (NPBL) nas respostas cardiorrespiratórias induzidas pela ativação dos quimiorreceptores centrais e periféricos em ratos não anestesiados. A injeção bilateral de muscimol (200 pmol/100 nl) no KF reduziu a ventilação basal (978 ± 100, vs. salina: 1436 ± 155 ml/kg/min). Injeção de muscimol no KF reduziu a hiperventilação (1827 ± 61, vs. salina: 3179 ± 325 ml/kg/min) e a taquicardia (380 ± 9, vs. salina: 423 ± 12 bpm), produzidos pela hipóxia (8% O2 - 10 min). Muscimol no KF reduziu a hiperventilação (1488 ± 277, vs. salina: 3539 ± 374 ml/kg/min) produzida por hipercapnia (7% CO2 - 10 min). Injeção de muscimol no NPBL promoveu um aumento de PAM (D = 119 ± 2, vs. salina: 104 ± 2 mmHg), mas não foi capaz de alterar a hiperventilação produzida por hipóxia e hipercapnia. Nossos experimentos mostram a participação da região KF, e não do NPBL, no controle do respiratório durante a ativação do quimiorreflexo central e periférico. / Here we evaluated the involvement of Kölliker-Fuse region (KF) and lateral parabrachial nucleus (LPBN) in the cardiorespiratory responses elicited by chemoreceptor activation in conscious rats. Bilateral injection of muscimol (200 pmol/100 nl) into the KF decreased resting ventilation (978 ± 100, vs. saline: 1436 ± 155 ml/kg/min). Muscimol injection into the KF reduced the increase in ventilation (1827 ± 61, vs. saline: 3179 ± 325 ml/kg/min) produced by hypoxia (8% O2 - 10 min) or hypercapnia (7% CO2 - 10 min) (1488 ± 277, vs. saline: 3539 ± 374 ml/kg/min). The results of the present study suggest that KF region, but not LPBN, have mechanisms to control the ventilation in resting, hypoxic or hypercapnic conditions in conscious rats.
