Eimeria maxima é uma das principais espécies que causam a coccidiose aviária. Visando uma maior compreensão dos mecanismos moleculares envolvidos na esporulação de oocistos de E. maxima, o perfil da expressão gênica em oocistos não esporulados, parcialmente esporulados e esporulados foi avaliado utilizando a técnica ORESTES. As 20.149 leituras geradas foram pré-processadas, agrupadas, anotadas empregando-se a plataforma Egene e em seguida, analisadas estatisticamente. Um total de 1.207 contigs e 2091 singlets foi gerado. Cerca de 72% dos transcritos foram classificados como proteínas hipotéticas, 20% eram similares a proteínas de função conhecida e 8% eram proteínas conservadas. Após a análise estatística, 32% dos transcritos foram classificados como diferencialmente expressos, dos quais, a maioria (65%) era estágio-específica. Para validação dos perfis de expressão obtidos por ORESTES, a expressão diferencial de um pequeno conjunto de genes foi quantificada por RT-qPCR. Os resultados demonstraram boa correlação entre as duas técnicas. / Eimeria maxima is one of the most important causing agents of poultry coccidiosis. Aiming at obtaining a better understanding of the molecular mechanisms involved in oocyst sporulation of E. maxima, the gene expression profiles of unsporulated, sporulating (sporoblast phase) and sporulated oocyst were studied using the ORESTES methodology. The 20,149 reads generated were pre-processed, grouped, annotated employing the platform Egene and statistically analyzed. A total of 1207 contigs and 2091 singlets were generated. About 72% of the transcripts were classified as hypothetical proteins, 20% were similar to proteins of known function and 8% were conserved proteins. After statistical analysis, 32% of the transcripts were classified as differentially expressed of which 65% were stage-specific. Este estudo objetivou estudar os carrapatos e a infecção por patógenos, e sua correlação com os vertebrados desta região entre 10/2007 e 09/2008 foram realizadas mensalmente doze visitas a uma área de floresta ombrófila densa da Amazônica na Estação Ecológica de Samuel. Foram coletados, 60 aves, 36 mamíferos, 14 répteis e 11 anfíbios. Em vida livre foram realizadas 80 coletas de carrapatos em um total de 39.600m em cerca de 2.640min (44h) e coletados 265 carrapatos adultos de quatro espécies, sendo elas: A. scalpturatum (81), A. latepunctatum (84), A. oblongogutattum (18), A. naponense (69) e 597 ninfas de Amblyomma sp. Dos 104 adultos (11,8% do total de carrapatos) foram testados para Anaplasmataceae, obtive-se 27 (25,9%) positivos e mais 36 adultos (total de 140) e 11 carrapatos foram positivos (1) A. scalpturatum, (4) A. latepunctatum, (3) A. naponense. É notável o impacto da hidrelétrica sobre a fauna de pequenos mamíferos. / This study was aimed to analyze the ticks and the pathogenic infections, and their correlation with the vertebrates of such a region from October 2007 to September 2008, 12 surveys were monthly carried out at the Samuel Ecological Station in an Amazonian dense ombrophilous forest. They were collected from 60 birds, 36 mammals, 14 reptiles, and 11 amphibians. Eighty(80) collections of free-living ticks were carried out within an area of about 39.000 meters, during 2.640 minutes (44 hours), and 265 adult ticks or four species were picked up as following: 81 A.scalpturatum, 84 A. latepunctatum, 18 A. oblongogutattum, 69 A. naponense , and 597 Amblyomma sp. nymphs. 104 the adults, which accounted for 11.8% of total of ticks collected, were tested for Anaplasmataceae, and 27 (25.9%) were positive. A esquistossomose é uma doença importante em saúde pública. Dos genes selecionados como diferencialmente expressos em esquistossômulos a partir do transcriptoma do S. mansoni, 56% foram confirmados por RT-PCR em tempo real. Entre eles, a proteína Ly6.5, está presente no tegumento de esquistossômulos e vermes adultos por âncoras de GPI. Não foi detectada a função de inibir o sistema complemento, mas pode estar envolvido na manutenção do tegumento. O gene SmVal7 revelou transcritos nas glândulas esofágicas de vermes adultos por hibridização in situ, enquanto a localização da proteína não está definida. Anexina está associada ao tegumento de esquistossômulos e vermes adultos, de maneira dependente de cálcio. A supressão do gene por RNAi não resultou em alteração fenotípica significativa em esquistossômulos in vitro. Foi observada atividade parcial de inibição de coagulação e potencial atividade de endocitose de anticorpos ligados à superfície. A imunização com rLy6.5, rSmVal7, rAneI-II ou rAneII-III não levou a redução da carga parasitária após desafio. / Schistosomiasis is an important disease in public health. Genes selected from the S. mansoni transcriptome, 56% of them were confirmed as differentially expressed in schistosomula by real time RT-PCR. Among them, the protein Ly6.5 is present in the tegument of schistosomula and adult worms by GPI anchors. The function of inhibiting the complement system was not detected, but it may be involved in maintenance of the tegument. The gene SmVal7 revealed transcripts in the esophageal glands of adult worms by in situ hybridization, while the localization of the protein is not defined. Annexin is associated with the membranes of the schistosomula and adult worms tegument in a calcium-dependent manner. The suppression of the gene by RNAi did not resulted in a significant phenotypic change in schistosomula in vitro. Parcial inhibition of the coagulation activity and potential function of endocytosis of membrane-bound antibodies were observed. Escherichia coli enteropatogênica (EPEC) é uma das principais causas de diarreia em crianças. Na carência de fosfato (Pi), um conjunto de genes conhecido como regulon PHO é induzido. Esse regulon é controlado pelo sistema Pst, que além de ser um transportador de Pi, reprime a expressão de PHO quando Pi é abundante, e pelo sistema de dois componentes PhoB/PhoR. A deleção de pst reduziu a adesão de EPEC à células epiteliais in vitro, pois diminuiu da expressão dos reguladores PerA/PerC, que por sua vez controlam a expressão de genes envolvidos na adesão. Este efeito foi exclusivo de pst e não devido a expressão constitutiva dos genes de PHO causada pela deleção de pst. A expressão da fímbria BFP, PerA e PerC também dependem da síntese de ppGpp, uma molécula de alarme envolvida na regulação de genes relacionados à carência nutricional. ppGpp regula positivamente a expressão de PerA e PerC. Entretanto, RpoS, o fator relacionado à resposta ao estresse, afetou negativamente o nível de adesão de EPEC e a expressão de BFP. / Enteropathogenic E. coli (EPEC) is one of the causes of diarrhea in children. Phosphate (Pi) shortage induces transcription of the genes known as the PHO regulon. These genes are controlled by the Pst system, that is also a high-affinity Pi transporter, and represses PHO expression under Pi-replete conditions. PHO is also controlled by the two-component system PhoB/PhoR. Deletion of the pst operon reduced the adhesion of EPEC to epithelial cells in vitro due to a decrease in the expression of the regulators PerA and PerC that in turn control the expression of genes related to adhesion. The constitutive expression of the PHO genes in the pst mutant was not the cause of adhesion inhibition. Expression of bfp and the regulators PerA and PerC was also dependent on ppGpp, an alarmone involved in the regulation of genes related to nutrient limitation. On the other hand, RpoS, the factor that controls the general stress response, negatively affected EPEC adhesion and bfpA expression.

Nesta tese, demonstramos que o fungo Paracoccidioides brasiliensis promove um aumento na ativação de ERK1/2 e SFKs em células epiteliais A549 (de pulmão humano), de aproximadamente 6 e 7 vezes, respectivamente, em relação aos níveis basais. Utilizando PP2, inibidor da atividade de SFKs, e PD98059, inibidor da ativação da via ERK1/2, verificamos que a ativação de ERK1/2 é parcialmente dependente de SFKs ativadas e que provavelmente outras quinases também estão envolvidas neste evento. Além da modulação da sinalização de células do hospedeiro, diversos estudos têm demonstrado que patógenos seqüestram domínios presentes nas membranas da célula hospedeira denominados “membrane rafts”, os quais são enriquecidos em esfingolipídeos e colesterol e estão envolvidos em diversos eventos celulares, incluindo a sinalização celular. Por diferentes metodologias, como a desorganização de “membrane rafts” por drogas que extraem (metil-β- ciclodextrina, MβCD) ou que se ligam (nistatina) ao colesterol, e a localização do gangliosídeo GM1, um marcador de “membrane rafts”, utilizando a subunidade B da toxina da cólera (CTB), mostramos o envolvimento destes domínios de membranas de células epiteliais na adesão de P. brasiliensis. A partir do isolamento de membranas resistentes a detergente (DRMs) de células epiteliais após incubação com o fungo, mostramos a ativação e o deslocamento de SFKs para as frações que contêm os “membrane rafts”. Além disso, verificamos que a depleção do colesterol com MβCD inibe completamente a ativação de SFKs, corroborando a importância dos domínios de membranas para a ativação destas quinases. Os resultados apresentados nesta tese demonstram, pela primeira vez, que fungos patogênicos modulam a organização e a atividade de “membrane rafts” de células hospedeiras para o estabelecimento da infecção. / Many pathogens are able to manipulate host cell signaling in order to facilitate infection. In this thesis, we demonstrated that the fungus Paracoccidioides brasiliensis promotes an increase of ERK12 and SFK activation in A549 epithelial cells (human lung cells), by approximately 6- and 7-fold over basal levels, respectively. By using PP2, inhibitor for SFK activity, and PD98059, inhibitor for ERK1/2 activation, we verified that ERK1/2 activation is partially dependent of activated SFKs and probably other kinases are involved in this event. Besides modulation of host cell signaling, several studies have been demonstrated that pathogens hijack domains that are present in host cell membranes called membrane rafts, which are cholesterol- and sphingolipidenriched domains, that are involved in several cell events, including cell signaling. By using several approaches, such as membrane rafts disruption with cholesterolextractor (methyl-β-cyclodextrin, MβCD) or –binding (nystatin) drugs, and the localization of ganglioside GM1, a membrane raft marker, by using cholera toxin subunit B (CTB), we showed the involvement of these epithelial cell membrane domains in P. brasiliensis adhesion. By isolating detergent-resistant membrane (DRM) from epithelial cells after incubation with this fungus, we showed the activation and the dislodgment of SFKs to fractions which contain membrane rafts. Moreover, we verified that cholesteroldepletion with MβCD completely inhibits SFK activation, corroborating the importance of membrane domains in activation of these kinases.The results presented in this thesis demonstrate for the first time that pathogenic fungi may modulate the organization and activity of host cell membrane rafts for infection establishment. / TEDE / BV UNIFESP: Teses e dissertações
